The effect of sediment sources on the selection of polychlorinated biphenyl (PCB) dechlorinating competence was investigated using sediments from two different locations, the Grasse River and Owasco Lake. These two sediments had a similar organic carbon content but different particle size distribution. The two PCB-free sediments were spiked with Aroclor 1248 and inoculated with microorganisms from the Reynolds and General Motors sites in the St. Lawrence River, which exhibited different dechlorination patterns. When each inoculum was serially transferred into fresh sediments four times (every 8-10 weeks), they still maintained the initial dechlorination patterns regardless, the source of sediments and the number of transfers, and dechlorination patterns of the two inocula in the same sediments did not converge. In a parallel approach, when the acclimated microorganisms from the Reynolds site were inoculated into fresh sediments from both sources as well as sediments enriched with organic carbon (2%, w/v), the dechlorination pattern remained unchanged after a 40-week incubation. These results suggest that the sediment characteristics or organic carbon content did not play a role in the selection of dechlorinating populations.  相似文献

Siderophores are small organic molecules produced by microorganisms under iron-limiting conditions which enhance the uptake of iron to the microorganisms. In environment, the ferric form of iron is insoluble and inaccessible at physiological pH (7.35–7.40). Under this condition, microorganisms synthesize siderophores which have high affinity for ferric iron. These ferric iron-siderophore complexes are then transported to cytosol. In cytosol, the ferric iron gets reduced into ferrous iron and becomes accessible to microorganism. In recent times, siderophores have drawn much attention due to its potential roles in different fields. Siderophores have application in microbial ecology to enhance the growth of several unculturable microorganisms and can alter the microbial communities. In the field of agriculture, different types of siderophores promote the growth of several plant species and increase their yield by enhancing the Fe uptake to plants. Siderophores acts as a potential biocontrol agent against harmful phyto-pathogens and holds the ability to substitute hazardous pesticides. Heavy-metal-contaminated samples can be detoxified by applying siderophores, which explicate its role in bioremediation. Siderophores can detect the iron content in different environments, exhibiting its role as a biosensor. In the medical field, siderophore uses the “Trojan horse strategy” to form complexes with antibiotics and helps in the selective delivery of antibiotics to the antibiotic-resistant bacteria. Certain iron overload diseases for example sickle cell anemia can be treated with the help of siderophores. Other medical applications of siderophores include antimalarial activity, removal of transuranic elements from the body, and anticancer activity. The aim of this review is to discuss the important roles and applications of siderophores in different sectors including ecology, agriculture, bioremediation, biosensor, and medicine.  相似文献

It is well known that microorganisms can dissolve different minerals and use them as sources of nutrients and energy. The majority of rock minerals are rich in vital elements (e.g., P, Fe, S, Mg and Mo), but some may also contain toxic metals or metalloids, like arsenic. The toxicity of arsenic is disclosed after the dissolution of the mineral, which raises two important questions: (1) why do microorganisms dissolve arsenic-bearing minerals and release this metal into the environment in a toxic (also for themselves) form, and (2) How do these microorganisms cope with this toxic element? In this review, we summarize current knowledge about arsenic-transforming microbes and their role in biomining processes. Special consideration is given to studies that have increased our understanding of how microbial activities are linked to the biogeochemistry of arsenic, by examining (1) where and in which forms arsenic occurs in the mining environment, (2) microbial activity in the context of arsenic mineral dissolution and the mechanisms of arsenic resistance, (3) the minerals used and technologies applied in the biomining of arsenic, and (4) how microbes can be used to clean up post-mining environments.  相似文献

Certain organic compounds, including biphenyl and salicylic acid, stimulate polychlorinated biphenyl (PCB) degradation by microorganisms in some environments. However, the usefulness of these amendments for improving PCB removal by microorganisms from diverse habitats has not been extensively explored. This study evaluated the effects of biphenyl, salicylic acid, and glucose on changes in aerobic PCB removal and bacterial communities from an agricultural soil, a wetland peat soil, a river sediment, and a mixture of these samples. PCB removal patterns were significantly different between soils and sediments amended with carbon compounds: (i) terrestrial soil microorganisms removed more PCBs than river sediment microorganisms, particularly with regard to PCBs with >4 chlorine substituents, (ii) glucose-supplemented, agricultural soil microorganisms removed more hexachlorobiphenyl than unsupplemented samples, (iii) biphenyl-supplemented, river sediment microorganisms removed more di- and tri-chlorobiphenyls than unamended samples. Carbon amendments also caused unique shifts in soil and sediment bacterial communities, as determined by specific changes in bacterial 16S rRNA denaturing gradient gel electrophoresis banding patterns. These results indicate that organic carbon amendments had site-specific effects on bacterial populations and PCB removal. Further work is needed to more accurately characterize PCB degrading communities and functional gene expression in diverse types of environments to better understand how they respond to bioremediation treatments.  相似文献

Phytoremediation of polyaromatic hydrocarbons, anilines and phenols   总被引:12,自引:0,他引:12  
Phytoremediation technologies based on the combined action of plants and the microbial communities that they support within the rhizosphere hold promise in the remediation of land and waterways contaminated with hydrocarbons but they have not yet been adopted in large-scale remediation strategies. In this review plant and microbial degradative capacities, viewed as a continuum, have been dissected in order to identify where bottle-necks and limitations exist. Phenols, anilines and polyaromatic hydrocarbons (PAHs) were selected as the target classes of molecule for consideration, in part because of their common patterns of distribution, but also because of the urgent need to develop techniques to overcome their toxicity to human health. Depending on the chemical and physical properties of the pollutant, the emerging picture suggests that plants will draw pollutants including PAHs into the plant rhizosphere to varying extents via the transpiration stream. Mycorrhizosphere-bacteria and -fungi may play a crucial role in establishing plants in degraded ecosystems. Within the rhizosphere, microbial degradative activities prevail in order to extract energy and carbon skeletons from the pollutants for microbial cell growth. There has been little systematic analysis of the changing dynamics of pollutant degradation within the rhizosphere; however, the importance of plants in supplying oxygen and nutrients to the rhizosphere via fine roots, and of the beneficial effect of microorganisms on plant root growth is stressed. In addition to their role in supporting rhizospheric degradative activities, plants may possess a limited capacity to transport some of the more mobile pollutants into roots and shoots via fine roots. In those situations where uptake does occur (i.e. only limited microbial activity in the rhizosphere) there is good evidence that the pollutant may be metabolised. However, plant uptake is frequently associated with the inhibition of plant growth and an increasing tendency to oxidant stress. Pollutant tolerance seems to correlate with the ability to deposit large quantities of pollutant metabolites in the 'bound' residue fraction of plant cell walls compared to the vacuole. In this regard, particular attention is paid to the activities of peroxidases, laccases, cytochromes P450, glucosyltransferases and ABC transporters. However, despite the seemingly large diversity of these proteins, direct proof of their participation in the metabolism of industrial aromatic pollutants is surprisingly scarce and little is known about their control in the overall metabolic scheme. Little is known about the bioavailability of bound metabolites; however, there may be a need to prevent their movement into wildlife food chains. In this regard, the application to harvested plants of composting techniques based on the degradative capacity of white-rot fungi merits attention.  相似文献

Anthropogenic contaminants as tracers in an urbanizing karst aquifer   总被引:2,自引:0,他引:2  
Karst aquifers are uniquely vulnerable to contamination. In the Barton Springs segment of the karstic Edwards aquifer (Texas, U.S.A.), urban contaminants such as pesticides and volatile organic compounds frequently are detected in spring base flow. To determine whether contaminant concentrations change in response to storms, and if they therefore might act as tracers of focused recharge, samples were collected from Barton Springs at closely spaced intervals following three storms. Two herbicides (atrazine and simazine), two insecticides (carbaryl and diazinon), and a solvent (tetrachloroethene) described breakthrough curves over a 1-week period following one or more storms. The breakthrough curves were decomposed into two to five log-normal subcurves, which were interpreted as representing pulses of contaminants moving through the aquifer. Each subcurve could be used in the same way as an artificial tracer to determine travel time to and recovery at the spring. The contaminants have several advantages over artificial tracers: they represent the actual compounds of interest, they are injected essentially simultaneously at several points, and they are injected under those conditions when transport is of the most interest, i.e., following storms. The response of storm discharge, specific conductance, and contaminant loading at the spring depended on initial aquifer flow conditions, which varied from very low (spring discharge of 0.48 m3/s) to high (spring discharge of 2.7 m3/s): concentrations and recovery were the highest when initial aquifer flow conditions were low. This behavior provides information about aquifer structure and the influence of aquifer flow condition on transport properties.  相似文献

Previous studies have used particle mass and size as metrics to link airborne particles with deleterious health effects. Recent evidence suggests that particle composition can play an important role in PM-toxicity; however, little is known about the specific participation of components (individually or acting in groups) present in such a complex mixture that accounts for toxicity. This work explores relationships among PM(10) components in order to identify their covariant structure and how they vary in three sites in Mexico City. Relationships between PM(10) with cell toxicity and geographical location were also explored. PM(10) was analyzed for elemental composition, organic and elemental carbon, endotoxins and the induction of inhibition of cell proliferation, IL-6, TNFalpha and p53. PM(10) variables were evaluated with principal component analysis and one-way ANOVA. The inhibition of cell proliferation, IL-6 and TNFalpha were evaluated with factorial ANOVA and p53 with the Welch test. The results indicate that there is heterogeneity in particle mass, composition and toxicity in samples collected at different sites. Multivariate analysis identified three major groups: (1) S/K/Ca/Ti/Mn/Fe/Zn/Pb; (2) Cl/Cr/Ni/Cu; and (3) endotoxins, organic and elemental carbon. Groups 1 and 3 showed significant differences among sites. Factorial ANOVA modeling indicated that cell proliferation was affected by PM concentration; TNFalpha and IL-6 by the interaction of concentration and site, and p53 was different by site. Radial plots suggest the existence of complex interactions between components, resulting in characteristic patterns of toxicity by site. We conclude that interactions of PM(10) components determine specific cellular outcomes.  相似文献

The concentrations of three different size fractions of particulate matter (PM) including PM2.5, PM10, and TSP were determined continuously at hourly intervals from four different sites in Seoul, Korea during the spring of 2001. To learn the effects of wind speed change on PM fractionation, the entire data sets were initially sorted into three particle fractions such as: fine (F: PM2.5), coarse (C: PM10-PM2.5), and giant (G: TSP-PM10). The inter-fraction relationships of PM were then explored by linear regression analysis of the data divided into four wind speed regimes. The results of this analysis, when examined in terms of either relative dominance between different PM fractions (i.e., in terms of their slope values) or strength of correlations, indicate the existence of diverse inter-fraction patterns. Most importantly, the physical influence of wind speed is seen to be reflected most efficiently between fine and coarse particle fractions, as the relative contribution of coarse fraction to the mass concentration of total particles (e.g., PM10) changes proportionally with changes in wind speed. However, such systematic patterns decrease noticeably between fine and giant fractions, as they can be affected more sensitively by such factors as the nature of their sources or the surrounding environmental conditions. The results of our comparative analysis thus confirm that wind speed is a useful barometer to distinguish and predict the behavior of different particle fractions in relation to each other.  相似文献

The aim of this paper is to examine the patterns of polychlorinated biphenyls (PCB) in the grey seals (Halichoerus grypus) from the Baltic, Northeast and Eastern England, and the St. Lawrence Estuary (Canada). In fact, the feeding habits of the ringed seal (Phoca hispida) include ingestion of major quantities of benthic crustaceans that might cause observed differences obtained in PCBs, whereas the grey seal feed mainly on fish. The profile (percent in mixture) of polychlorinated biphenyls (PCB) 101, 118, 138, 153, and 180, and the total of their concentrations in mg/kg lipid in grey seals from the Baltic, from Northeast and Eastern England, and from the St. Lawrence estuary (Canada), were examined by principal component analysis (PCA). When considering the possible effects of consuming seafood by the grey seal, it is necessary to characterize populations and individuals according to the amounts they consume, since populations in different parts of the world are likely to show big differences in their consumption of seafood. The patterns differ between juveniles and adult animals, but the gender of adults and geography do not appear to play a role.  相似文献

Conservation of the marine environment mainly focuses on threatened elements and more precisely on vulnerable and endangered species like birds and mammals. When dealing with the conservation of marine habitats, the scientific community is mainly interested in hot spots of diversity, like seagrass beds in Europe, or hot spots of endemism, like coral reefs in tropical areas. Nevertheless, using the example of a common and widespread marine invertebrate, the sandmason worm (Lanice conchilega, Polychaeta, Terebellidae), we show that vulnerability and rarity are not the only criteria to take into account in order to select the best natural element for conservation. This species can form dense beds that increase biodiversity, are attractive feeding grounds for birds and fishes, and have a high socioeconomic value. In consequence, they have a high functional value that should be considered as an important conservation stake. Through the example of the Chausey archipelago and the Bay of the Mont Saint-Michel (France), we propose a synthetic interdisciplinary approach to evaluate the conservation needs of these beds. The issue is even more pressing when one considers that these natural elements and many similar ones still do not benefit from any legal protection in Europe despite their high heritage value.  相似文献

The degradation of the (14)C-labelled fungicide dithianon in an orthic luvisol was investigated under standardized conditions in comparison to stimulated microbial activity by an amendment of maize straw. The compound is characterized by mineralization losses of approximately 33% and the formation of non-extractable bound residues of approximately 63% in 64 days. Despite the major role of microorganisms in mineralizing this compound, the formation of bound residues is not biotically induced. Gel permeation chromatography and polyacrylamide gel electrophoresis, as different size separation techniques of the humic acids fractions, showed differences in the distribution patterns of non-extractable residues depending on the addition of straw material. The results presented support the existence of humic substances in soil as a micellar system rather than as a biopolymer.  相似文献

Background, Aim and Scope Current scientific studies and evaluations clearly show that an increase of urban dust loads, alone or combined with other pollutants und certain meteorological conditions lead to different significant health effects. Premature death, increased hospital admissions and increased respiratory symptoms and diseases as well as decreased lung function can be observed in combination with high pollutant levels. Sensitive groups like elderly people or children and persons with cardiopulmonary diseases such as asthma are more strongly affected. Because of the direct contact between fine particles and lung tissue more information concerning the surface structure (mapping of toxic elements) is required. Materials and Methods: The NanoSims50 ion microprobe images the element composition at the surface of sub-micrometer air dust particles and documents hot spots of toxic elements as a possible threat for human health. Results: The atmospheric fine dust consists of a complex mixture of organic and inorganic compounds. Heavy metals are fixed on airborn particles in the form of hot spots in a nanometer scale. From a sanitary point of view, the hot spots consisting of toxic elements are particularly relevant as they react directly with the lung tissues. Discussion: To what extent particles can penetrate the various areas of the lungs and be deposited there depends on the one hand on their physical characteristics and on the other on breathing patterns and the anatomy of the lung, which is subject to change as the result of growth, ageing or illness. Once inhaled, some particles can reach the pulmonary alveoli and thus directly expose the lung tissues to toxic elements. Conclusions: Especially the mapping of toxic arsenic or heavy metals like copper on the dust particles shows local hot spots of pollution in the dimension of only 50 nanometers. Recommendations and Perspectives: Imaging of elements in atmospheric particles with NanoSIMS will help to identify the material sources.  相似文献

Evidence of reductive dechlorination of polychlorinated biphenyls (PCBs) in sediments was investigated in Hudson River sediments dredged and encapsulated in 1978 at Moreau, NY. The effect of different moisture contents in dredged sediments on dechlorination and dechlorinating microorganisms was also determined using PCB-spiked sediments in which the moisture level was adjusted by simulating a dewatering process. The congener pattern of PCBs indicated that the dechlorination in the dredged sediments was far less advanced than that in the river sediments collected from the general area of the dredged site (Ft. Edward site). Dechlorination in encapsulated sediments at the Moreau site appeared to have stopped soon after dredging. When microorganisms eluted from the encapsulated sediments were inoculated in clean sediments spiked with Aroclor 1242, an extensive dechlorination was observed, indicating that the encapsulated sediments still harbored dechlorinating microorganisms. However, the same inoculum failed to further dechlorinate residual congeners in the dredged sediments. On the other hand, an inoculum obtained in 1990 from the dredged site in the Hudson River dechlorinated the residual congeners further. In simulated dredged sediments, the maximum level of dechlorination was lower at reduced moisture contents. The population size of dechlorinating microorganisms, as determined by the most probable number (MPN) technique, was also smaller at the lower moisture levels. There was a significant correlation between the maximum extent of dechlorination and the specific death rate of dechlorinating populations. These results indicate that the underlying mechanism of the moisture-dependent maximum dechlorination is the moisture-dependence of the death rate of dechlorinating microorganisms.  相似文献

We collected rainwater samples from every rainfall in Matsue, Japan in order to study variations of nitrogen and phosphorus concentrations over time. The seasonal average concentration by magnitude order of Total Nitrogen (here after T-N) was highest in winter, then in spring, fall, and summer and that of Total Phosphorus (here after T-P) was highest in spring, then in winter, fall, and summer. These seasonal variations were examined in relation to the transportation paths of arrived air masses by using a backward trajectory and rainfall patterns from a surface synoptic weather chart. In winter, continental air masses frequently flow from China or Siberia and the resultant winter rainfall is on many occasions of a continental type. In summer, maritime air masses frequently arrive from the Pacific Ocean and this resultant rainfall therefore was often of maritime type. Looking at average concentrations of T-N and T-P for each rainfall type, continental types were high range and maritime types were low. It was therefore concluded that the monthly average concentration of T-N was affected by continental air masses from northern China in winter and by maritime ones from the Pacific Ocean in summer. The maximum deposition of T-N was caused by this concentration in winter and rainfall depth in summer. Seasonal variation of T-P showed a different fluctuation tendency from T-N, with a maximum concentration in spring, and minimum in summer and fall. T-P was susceptible to the yellow sand phenomenon which maximised T-P deposition in spring.  相似文献

Depending on the final application, several methodologies for traffic emission estimation have been developed. Emission estimation based on total miles traveled or other average factors is a sufficient approach only for extended areas such as national or worldwide areas. For road emission control and strategies design, microscale analysis based on real-world emission estimations is often required. This involves actual driving behavior and emission factors of the local vehicle fleet under study. This paper reports on a microscale model for hot road emissions and its application to the metropolitan region of the city of Santiago, Chile. The methodology considers the street-by-street hot emission estimation with its temporal and spatial distribution. The input data come from experimental emission factors based on local driving patterns and traffic surveys of traffic flows for different vehicle categories. The methodology developed is able to estimate hourly hot road CO, total unburned hydrocarbons (THCs), particulate matter (PM), and NO(x) emissions for predefined day types and vehicle categories.  相似文献

This paper investigated the degradation kinetics of norfloxacin in a soil, and its effects on soil respiration and nitrogen transformation under different conditions. Compared to the sterile control, the degradation rates of norfloxacin in the non-sterile soil were greatly enhanced, suggesting that microorganisms played a major role in the degradation. Accelerated degradation for norfloxacin in the soil was observed with decreasing concentrations (30 mg/kg to 5 mg/kg) with its half-life decreasing from 62 days to 31 days. Amending swine manure into the soil and increasing the soil moisture level enhanced the biological degradation of norfloxacin. No obvious inhibition of norfloxacin on soil respiration was observed in the soil, while only slight effect on nitrogen transformation was found. The results suggested that norfloxacin at the reported environmental concentrations (<100 mg/kg) would have little effect on microbial activity and functions in the soils.  相似文献

The purpose of this study is to demonstrate a methodology for quantification of high emissions hot spots along roadways based upon real-world, on-road vehicle emissions measurements. An emissions hot spot is defined as a fixed location along a corridor in which the peak emissions are statistically significantly greater by more than a factor of 2 than the average emissions for free-flow or near free-flow conditions on the corridor. A portable instrument was used to measure on-road tailpipe emissions of carbon monoxide, nitric oxide, hydrocarbons, and carbon dioxide on a second-by-second basis during actual driving. Measurements were made for seven vehicles deployed on two primary arterial corridors. The ratio of average emissions at hot spots to the average emissions observed during a trip was as high as 25 for carbon monoxide, 5 for nitric oxide, and 3 for hydrocarbons. The relationships between hot spots and explanatory variables were investigated using graphical and statistical methods. Average speed, average acceleration, standard deviation of speed, percent of time spent in cruise mode, minimum speed, maximum acceleration, and maximum power have statistically significant associations with vehicle emissions and influence emissions hot spots. For example, stop-and-go traffic conditions that result in sudden changes in speed, and traffic patterns with high accelerations, are shown to generate hot spots. The implications of this work for future model development and applications to environmental management are discussed.  相似文献

Arsenic pollution in aquatic environments is a worldwide concern due to its toxicity and chronic effects on human health. This concern has generated increasing interest in the use of different treatment technologies to remove arsenic from contaminated water. Constructed wetlands are a cost-effective natural system successfully used for removing various pollutants, and they have shown capability for removing arsenic. This paper reviews current understanding of the removal processes for arsenic, discusses implications for treatment wetlands, and identifies critical knowledge gaps and areas worthy of future research. The reactivity of arsenic means that different arsenic species may be found in wetlands, influenced by vegetation, supporting medium and microorganisms. Despite the fact that sorption, precipitation and coprecipitation are the principal processes responsible for the removal of arsenic, bacteria can mediate these processes and can play a significant role under favourable environmental conditions. The most important factors affecting the speciation of arsenic are pH, alkalinity, temperature, dissolved oxygen, the presence of other chemical species - iron, sulphur, phosphate -, a source of carbon, and the wetland substrate. Studies of the microbial communities and the speciation of arsenic in the solid phase using advanced techniques could provide further insights on the removal of arsenic. Limited data and understanding of the interaction of the different processes involved in the removal of arsenic explain the rudimentary guidelines available for the design of wetlands systems.  相似文献

Materials based on polystyrene and starch copolymers are used in food packaging, water pollution treatment, and textile industry, and their biodegradability is a desired characteristic. In order to examine the degradation patterns of modified, biodegradable derivates of polystyrene, which may keep its excellent technical features but be more environmentally friendly at the same time, polystyrene-graft-starch biomaterials obtained by emulsion polymerization in the presence of new type of initiator/activator pair (potassium persulfate/different amines) were subjected to 6-month biodegradation by burial method in three different types of commercially available soils: soil rich in humus and soil for cactus and orchid growing. Biodegradation was monitored by mass decrease, and the highest degradation rate was achieved in soil for cactus growing (81.30 %). Statistical analysis proved that microorganisms in different soil samples have different ability of biodegradation, and there is a significant negative correlation between the share of polystyrene in copolymer and degree of biodegradation. Grafting of polystyrene on starch on one hand prevents complete degradation of starch that is present (with maximal percentage of degraded starch ranging from 55 to 93 %), while on the other hand there is an upper limit of share of polystyrene in the copolymer (ranging from 37 to 77 %) that is preventing biodegradation of degradable part of copolymers.  相似文献
