The development of swimming (myotomal) muscles was studied in herring larvae (Clupea harengus L.) caught in the Firth of Clyde, Scotland, in spring 1990 and reared at either 5, 10 or 15°C. Two muscle-fibre types can be distinguished in the myotomes of herring larvae using ultrastructural criteria. A single layer of small-diameter muscle fibres, packed with mitochondria, is present beneath the entire surface of the skin (superficial muscle fibres). The remaining bulk of the muscle is composed of larger diameter fibres (inner muscle fibres) containing significantly more myofibrils than the superficial fibres. In 1 d-old larvae, the number of inner muscle fibres in myotomes immediately posterior to the yolk-sac was 311±41 at 15°C, 257±22 at 10°C and 187±22 at 5°C (mean±SD,n=6). The average diameter of inner muscle fibres increased with decreasing temperature, so that the total cross-sectional area of muscle was similar at each temperature. After 6 to 7 d, the number of muscle fibres had significantly increased at 15°C (383±25), but not at 10°C (281±32) or 5°C (192±17). In contrast, the average cross-sectional area of inner muscle fibres had increased by 19% at 15°C, 34% at 10°C, and 26% at 5°C. Temperature also influenced the relative proportions and spatial distributions of muscle-fibre organelles. For example, in 1 d-old larvae, the fraction of muscle-fibre volume (volume density) occupied by mitochondria in the superficial fibres was significantly higher at 15°C (46.0%) than at either 5°C (37.6%) or 10°C (38.8%). In the inner muscle fibres, the volume density of mitochondria was 26.1% at 15°C, 20.5% at 10°C and 15.9% at 5°C, whereas the volume density of myofibrils was similar at the three temperatures (33 to 38%). Typically, inner muscle fibres from 10°C larvae, but not from 5 or 15°C larvae contained a large central mitochondrion.  相似文献

Eggs of the plaice Pleuronectes platessa L. were incubated at temperatures of 5, 8, 10, 12 and 15°C in March 1990, 1991 and 1992. The myotomes of yolk-sac larvae contain a single superficial layer of small-diameter muscle fibres which stain intensely for succinic dehydrogenase activity, surrounding 390 to 500 weakly staining inner-muscle fibres of larger diameter. Larvae reared at 15°C only survived for a few days and had significantly more inner-muscle fibres of larger average cross-sectional area than those hatching at 5 to 10°C. Myofibrils occupied 61% of the volume of inner-muscle fibres in 15°C larvae compared with 35 and 36% in larvae hatching at 5 and 10°C, respectively (P(0.01). Following metamorphosis, which occurs between 7 and 10 wk, the myotomes retain the single layer of superficial-muscle fibres characteristic of larvae. A thickening of the superficial-muscle layer is first evident in 4 to 5 mo-old laboratory-reared fish of 20 mm total length (TL) and in 0-group fish caught in June and July. On the basis of the histochemical staining reactions for myofibrillar ATPase and succinic dehydrogenase activities, the myotomes of 1-group (104 mm TL) and adult (280 mm TL) plaice were found to contain a minimum of six distinct muscle-fibre types. Two-dimensional gel electrophoresis and peptide mapping were used to investigate changes in myosin subunit composition during development. Myosin from the inner muscle of larvae contains two isoforms of the phosphorylatable light-chain 2 (LC2L1 and LC2L2). Following metamorphosis and during the first year, inner-muscle fibres co-express LC2 isoforms characteristic of the superficial fast-muscle fibres of adult plaice (LC2F1 and LC2F2) in addition to the larval isoforms. Fast-muscle fibres isolated from deep layers of the myotomes in adult plaice only contain LC2F2. In contrast, myosin from larval muscle and adult fast muscle contain apparently identical alkali light chains (LC1 and LC3). Peptide maps of myosin heavy chains (MHCs) from 6 wk-old larvae and 10 wk-old fish that had completed metamorphosis are similar, but distinct from those of 1-group plaice. Further changes in white-muscle MHC composition are evident between 1-group fish of 104 mm TL and adults of 280 mm TL.  相似文献

Eggs from spring spawning stocks of herring (Clupea harengus L.) were fertilized and reared at either 5, 8 or 12°C in 1991 and 1992. The differentiation of myotomal muscle fibres was investigated in relation to the development of other organs and tissues using light and electron microscopy. The gut, notochord, eyes and haemocoel appeared at the same relative point in development between fertilization and hatching at all temperatures. In contrast, the formation of the spinal cord, pronephros, pectoral fin buds and muscle fibres was relatively retarded at 5°C compared with 8 and 12°C. Myogenesis in the presumptive inner muscle mass occurred after 12 to 16 d at 5°C, 7 to 10 d at 8°C and 3.5 to 6 d at 12°C. Myoblasts aligned in orderly rows running from myosept to myosept prior to fusion to form myotubes. Actin and myosin filaments were synthesised throughout the cytoplasm in associated with presumptive Z-lines at the periphery of myotubes and immature muscle fibres. Differentiation of the superficial and inner muscle fibres types of larvae occurred at around the same time. Following this initial period of myogenesis, the number of myotomal muscle fibres remained constant until after hatching, so that increases in muscle bulk in the late embryo were entirely due to fibre hypertrophy. At hatching, the number of superficial muscle fibres present in myotomes just posterior to the yolk-sac was significantly less at 5°C (108±12) than at either 8°C (132±10) or 12°C (140±10) (mean±SD, 12 fish/temperature). In contrast, there were around 280 inner muscle fibres/myotome, comprising 90% of the trunk cross-sectional area, at all three temperatures. Myofibrillargenesis occurred relatively slowly at low temperatures, so that the volume density of myofibrils in the inner muscle fibres of larvae at hatching was significantly less at 5°C (39.2±9.0) than at either 8°C (49.6±8.8) or 12°C (50.2±9.8) (mean ±SD, 20 fibres/temperature from total of 5 fish). Undifferentiated myoblasts remained at hatching to form a population of presumptive myosatellite cells. The number of presumptive myosatellite cells per mm2 cross-sectional area of muscle fibre was more than two times higher at 8°C (1493±335) than at either 5°C (478±102) or 12°C (924±233) (mean±SD, 5 fish/temperature). The results suggest that temperature can influence the commitment of myoblasts to differentiation at a critical stage in embryogenesis, thereby providing a potential mechanism for influencing future growth characteristics. The projection of Octopus vulgaris motor chromatophore fibres on the body surface is revealed by sectioning the brain nerves. The denervated areas remain pale and, therefore, allow to determine the pattern of nerve projection. Ten of the approximately 30 nerves leaving the O. vulgaris brain contain chromatophore fibres. Histological sections of starved cod (Gadus morhua Linnaeus, 1758) and jelly cats (Lycichthys denticulatus Kröyer, 1844) with similar water contents are compared. Starvation causes the white muscle fibres of cod to shrink into irregular shapes inside their connective tissue matrix, while those of jelly cats of similar water content are seen to be rounded. It appears to be less important for jelly cat muscle to be packed tightly with contractile fibres than for the muscle of cod, so the former may be adapted to a permanent state of unusually high hydration.  相似文献

The bivalve osphradium is a band of putatively sensory tissue located in the gill axis, whose function is uncertain. In the present study, extending from 1987 to 1994, anatomical, histological, and electron microscopical techniques were used to elucidate the structure and ultrastructure of the osphradium in hatchery Pecten maximus L. and Placopecten magellanicus (Gmelin) (collected from Passamaquoddy Bay, New Brunswick, Canada). The osphradium consists of two distinct regions which run longitudinally on both sides of each gill axis: the osphradial ridge, and the dorsal tuft cilia region. The osphradial ridge was largely devoid of cilia other than those of the few free nerve fibres. The dorsal tuft cilia region contained free nerve fibres and ciliary tufts, separated by undifferentiated epithelial cells. No paddle cilia were observed under isosmotic fixation conditions, although under hypotonic conditions such cilia were quite common, suggesting an artefactual nature. Most of the cells of the osphradial ridge were highly secretory, the principal products being large pigment granules (in Pecten maximus) directly secreted by the Golgi bodies, and numerous small, electron-dense vesicles. These vesicles were arranged along extensive microtubule arrays in the basal region, indicative of axonal transport. These data support and extend Haszprunar's hypothesis of the role of the osphradium in the reception of chemical spawning cues and in the synchronization of gamete emission. Together with independent data on nerve pathways, osphradial sensory modalities, and monoamine localisation, an anatomical pathway and neurophysiological mediator are postulated.  相似文献

The distribution and fine structure of muscle-fibre types has been determined for the pelagic fingerling and demersal adult stages of the antarctic teleost Notothenia neglecta Nybelin, collected from Signy Island, Antarctica, between January and February 1984. In both stages, the pectoral fin adductor muscle (m.ad.p) is largely composed of slow fibres which contain abundant mitochondria (34 to 36%). During development, the ratio of capillaries to fibres increases less than does fibre diameter, so that capillary density is significantly lower in the m.ad.p of adults (498 mm-2) than fingerlings (1 727 mm-2). The secondary metamorphosis from a pelagic to a demersal mode of life is associated with the disappearance of subcutaneous lipid sacs and major changes in the distribution and structure of muscle fibres in the myotomes. The trunk cross-section of adult fish is almost entirely composed of poorly vascularised fast-muscle fibres (100 capillaries mm-2), which contain densely packed myofibrils (86.3%), and have few mitochondria (1.4%). Slow-muscle fibres in adults are restricted to a thin wedge adjacent to the lateral line canal. In contrast, slow fibres occur around the entire circumference of the trunk in fingerlings representing 24% of the total cross-sectional area at the post-anal level. Volume densities (%) of mitochondria, intracellular lipid and myofibrils in this tissue are respectively 37.0, 7.9, 38.6 for fingerlings and 13.1, 0, 70.3 for adults. Slow-muscle fibres in adult fish are of unusually large diameter (50 to 120 m) and have relatively low capillary densities (266 mm-2). These morphological changes reflect a general decrease in activity and a shift from a sub-carangiform to a labriform mode of swimming following transition from the fingerling to adult stage of the life cycle. The fine structure of the opercula of Spirobis borealis Daudin and S. pusilloides Bush, and the primary and secondary operculum of S. granulatus (L) have been examined by electron microscopy. In different regions, the epithelium of the operculum exhibits variations in thickness and fine structure which correspond to transitions from peduncular to opercular wall to opercular plate epithelia. Externally, the peduncle and walls of the operculum are covered by a cuticle which is composed of several layers of fibrils/fibres embedded in an amorphous matrix. It is regularly traversed by microvilli from underlying cells. At the operculum rim, the cuticle changes so that its appearance is thick and solid. It continues across the roof of the operculum as a thin layer overlying a calcareous plate. The secondary operculum of S. granulatus differs from those of the other species in that an internal calcareous plate and solid cuticle line the roof of the operculum. It differs further in possessing an opercular pore. The possibilities are discussed of the cuticle fibrils/fibres of the peduncle and the opercular wall being collagenous in nature, and later being transformed to a solid tanned structure overlying a calcareous plate. The modes of secretion of the cuticle and calcareous plate by underlying cells are also discussed.  相似文献

Turbot (Scophthalmus maximus L.) were reared at 12 and 16°C until 26 d after hatching. At both temperatures, starting at the neural plate stage, somites were initially formed every 75 min. Expressed as a percentage of development time (DT, fertilisation to 90% larvae hatching) somite formation occurred relatively earlier during embryogenesis at 12°C (45% DT) than at 16°C (55% DT). At 12°C, after the 32-somite stage the rate of somite formation decreased to one every 300 min. The larvae hatched after 6 d at 12°C and 3 d at 16°C at a relatively primitive stage of development, prior to the opening of the mouth and anus, with unpigmented eyes, and a straight gut. Temperature altered the relative timing of organogenesis in the larval stages. At 12°C, the following characters appeared (in this order): swimbladder>loop in the gut (at the time of yolk exhaustion)>caudal fin. In contrast, at 16°C, the caudal fin appeared at the same time as the loop in the gut. At 16°C, spines formed on the head in the region of the otic capsule at the time the swimbladder formed and the yolk was exhausted, but were absent in 12°C larvae. At both temperatures, in 1 d-old larvae the myotomes just behind the yolk-sac contained 200 inner muscle fibres (presumptive white muscle). The initial growth of inner muscle was largely due to hypertrophy, but by 26 d at 12°C and 11 d at 16°C hyperplastic growth became important, as evidenced by a significant increase in the number of small fibres (<10 m2). By 26 d the average number of inner muscle fibres had increased to 341 at 12°C and 988 at 16°C. New muscle fibres were added in distinct germinal zones at the dorsal and ventral apices of the myotomes. Metamorphosis was associated with a thickening of the superficial (presumptive red) muscle layer and the appearance of tonic muscle fibres.  相似文献

Cod (Gadus morhua L.) eggs may develop and hatch within temperatures of −1.5 to 12 °C, but little is known about the effects of very low temperatures on larval characteristics. Eggs of the Northeast Arctic cod (Gadus morhua) were incubated at 1, 5 or 8 °C from Day 1 after fertilisation until hatching, and transferred to 5 °C after hatching. Histological samples of the axial musculature were taken at hatching and 5 d after hatching, and the data on muscle cellularity from these samples were related to survival and hatching, size, developmental data and viability of the yolk sac larvae. All larvae hatched at the same developmental stage. Incubation of eggs at 1 °C produced shorter larvae with a larger yolk sac and more, small deep fibres at hatching than larvae from eggs incubated at 5 or 8 °C. The larval size difference was still present 5 d after hatching, a time at which the larvae from 1 °C-incubated eggs were less developed and less resistant to an acute viability stress test (65 ppt salinity). Although there were no differences between temperature groups in number and size of muscle fibres 5 d after hatching, the deep fibres of the 1 °C-group contained less myofibrils than the two other groups. The phenotype of the larvae at hatching was thus affected within these incubation temperatures. Although all groups were transferred to the same temperature after hatching, the lowest egg incubation temperature (1 °C) still had a negative effect 5 d after hatching, as these larvae were both smaller, less resistant to stress and had less functional muscles at the time of first feeding. Our conclusion is therefore that 1 °C is close to, or below, the lower thermal tolerance limit for normal functional development of Northeast Arctic cod. Three marine diatoms, Skeletonema costatum, Chaetoceros debilis, and Thalassiosira gravida were grown under no limitation and ammonium or silicate limitation or starvation. Changes in cell morphology were documented with photomicrographs of ammonium and silicate-limited and non-limited cells, and correlated with observed changes in chemical composition. Cultures grown under silicate starvation or limitation showed an increase in particulate carbon, nitrogen and phosporus and chlorophyll a per unit cell volume compared to non-limited cells; particulate silica per cell volume decreased. Si-starved cells were different from Si-limited cells in that the former contained more particulate carbon and silica per cell volume. The most sensitive indicator of silicate limitation or starvation was the ratio C:Si, being 3 to 5 times higher than the values for non-limited cells. The ratios Si:chlorophyll a and S:P were lower and N:Si was higher than non-limited cells by a factor of 2 to 3. The other ratios, C:N, C:P, C:chlorophyll a, N:chlorophyll a, P:chlorophyll a and N:P were considered not to be sensitive indicators of silicate limitation or starvation. Chlorophyll a, and particulate nitrogen per unit cell volume decreased under ammonium limitation and starvation. NH4-starved cells contained more chlorophyll a, carbon, nitrogen, silica, and phosphorus per cell volume than NH4-limited cells. N:Si was the most sensitive ratio to ammonium limitation or starvation, being 2 to 3 times lower than non-limited cells. Si:chlorophyll a, P:chlorophyll a and N:P were less sensitive, while the ratios C:N, C:chlorophyll a, N:chlorophyll a, C:Si, C:P and Si:P were the least sensitive. Limited cells had less of the limiting nutrient per unit cell volume than starved cells and more of the non-limiting nutrients (i.e., silica and phosphorus for NH4-limited cells). Given the excessive use pressure on forests, characterisation of the major users of forest resources can help guide conservation efforts. This study examines whether socio-economic variables explain domestic wood use in an area of Atlantic forest in the community of Três Ladeiras (Pernambuco, north-east Brazil). Socio-economic data were collected from 62 households, and geometric measurements of wood products in each household were taken to calculate the wood volume. An informant in each household indicated the average replacement time for each type of wood use. Data were analysed for static volume (wood volume found at the time of the visit) and dynamic volume (rate of wood consumption, calculated as the ratio between volume and replacement time). Multiple regression analyses were performed to determine the relationships between wood volume (static and dynamic) and the number of species used; independent variables including monthly family income, number of residents, average age of heads of the household, age of oldest household member, average residence time of heads of the household, average education level of heads of the household and education level of the most educated household member. Monthly income was inversely related to the static wood volume (adjusted R 2 = 17.6%, p < 0.001), rate of wood consumption (adjusted R 2 = 23.0%, p < 0.001) and number of species used (adjusted R 2 = 22.3%, p < 0.001) and was the most important predictive variable for these three outcomes. Other factors such as average education level and average age of heads of the household were also explanatory variables of the total rate of wood consumption, as these two variables, together with monthly family income, explain 31.3% of the total dynamic volume. Given the high importance of income in our study, we suggest that future conservation strategies give special attention to the poorest members of the community.  相似文献

Dittoral and sublittoral hermit crabs (Coenobita clypeatus, Clibanarius tricolor, Calcinus dibicen, Petrochirus diogenes, Paguristes grayi, Pagurus miamensis, Paguristes tortugae, Paguristes cadenati and Pagurus sp.) were studied on Lighthouse Atoll, Belize. The Clibanarius tricolor population differed markedly in mean clutch size from congeneric populations at temperate latitudes and from conspecific populations in the Florida Keys. These differences can be attributed primarily to variation in shell utilization pattern, overlap with competitors, and breeding season. Regression analyses indicate that shell weight but not internal shell volume affects the clutch size of C. tricolor in Belize, probably due to selection for optimum volume resulting in the use of broken and encrusted shells with high weight: volume ratios. Both egg size and clutch size differed significantly among egg developmental stages in C. tricolor, Calcinus tibicen, and Pagurus miamensis.  相似文献

Variations in the absolute and relative swimbladder volume of sardine (Sardina pilchardus W.), were examined in relation to fish length, gonads and stomach volume, and lipid content. Fifty specimens were frozen in liquid nitrogen and sliced with a cryo-microtome. From the slices, volume estimates of the organs were obtained through computer reconstruction. Variations in absolute and relative swimbladder volume in relation to fish length and the aforementioned organs' volume and lipid contents were estimated by means of multiple regression analysis. The relative importance of these variables in the calculations of the swimbladder volume for different physiological conditions of the fish was also assessed. The results revealed that the volume can be estimated from two distinct models: the first for the reproductive and the second for the non-reproductive period. The non-reproductive period model is in agreement with the well-known positive correlation of target strength with fish log length. However, during the reproductive period the relative size of gonads and relative lipid content gradually become more important than length.  相似文献

A southern Italian area that is characterized by large outcrops of rocks that are rich in titanium oxide (TiO2) phases were investigated to determine the mineralogical risk induced by the natural dispersion of TiO2 minerals. Rock, sediment and surface water samples were collected to determine the physicochemical and mineralogical factors (i.e., size distribution, morphology and alteration) indicative of potential TiO2 toxicity. X-ray diffraction data suggested that titanium oxides were present as rutile and anatase. Scanning electron microscopy images showed elongated TiO2 morphologies; fibres were found as either isolated or embedded/enclosed in flake-like phyllosilicates. The concentration of fibres in stream water ranged from 1.7 to 4.6 million fibres per litre. The highest fibre amounts in the sediments were in the <8-µm fraction, while single fibres were primarily concentrated in the <2-µm fraction. The results indicate that titanium oxide minerals represent a natural source of environmental risk and that the geomineralogical characterization of rich TiO2 areas is indispensable for understanding their geoavailability, dispersion and distribution.

Tethys fimbria is a poorly studied nudibranch that, recently, has been shown to possess large amounts of unusual prostaglandin (PG) derivatives. The cerata ofT. fimbria collected in the Bay of Naples (Italy) between November 1990 and January 1991 were submitted to histological analysis. This revealed the presence of both longitudinal and transverse smooth muscle fibres together with multicellular defensive glands, responsible for the typical spontaneous contractions of the cerata and for the extrusion of a hyaline secretion, respectively. A role forT. fimbria cerata in mollusc defense behaviour is suggested. Chemical analysis of the cerata led to the isolation of high levels of prostaglandin free acids (PGs) and of PG-1,15-lactones of the E, A and F series, whereas the secretion contained only PG-1,15-lactones of the E and A series. The latter were found to be toxic to mosquito fish at concentrations ranging between 1 and 10 µg ml–1, whereas PG free acids and PG-1,15-lactones of the F series were not toxic. The possibility that PGE- and PGA-1.15-lactones areT. fimbria defense allomones is discussed.  相似文献

The optimal parameters for the degradation of pure terephthalic acid (PTA) wastewater by the white rot fungus Phanerochaete chrysosporium were investigated. The relative mathematical model, objective function equation, constraint equations, constraint conditions and computer program were established in the optimal method. The results showed that the optimal value of the reactor volume needed was 9798 m3. It could save 83.82% of the reactor volume, which was needed for the native bacterium YZ1. This optimal method is simple, useful and economical for average treatment plants. Using the optimal parameters and P. chrysosporium to design and operate the treatment process of PTA wastewater would increase the treatment efficiency and decrease the cost.  相似文献

After acclimation, the copepod Acartia clausi was allowed to graze for 5 days in a mixed suspension of two discrete size classes (species) of the chaining diatoms Thalassiosira spp. derived from continuous culture. Total particle numbers and particle size distributions of Thalassiosira spp. were stable throughout the 5 days, indicating that the effects of algal removal and modification due to grazing were balanced by algal growth. Grazer ingestion is the predominant process affecting all size classes of the smaller diatom population (T. nordenskioldii); however, both ingestion and chain modification are observed with the larger diatom (T. gravida). Although the greatest percentage removal of algal volume occurs in the largest algal size classes for each algal species, the greatest volume removal occurs at the modal peak (T. nordenskioldii) or just to the right of the modal peak (T. gravida). Flask-to-flask replicability of experiments was tested with the single-celled T. fluviatilis, and these tests were compared to the long-term experiments with T. nordenskioldii and T. gravida. Net particle removal occurs on both the large and small algal species in mixed suspension, not just on the larger-cell-side of the T. gravida distribution. Although 80% of the particles ingested are from the T. nordenskioldii distribution, 80% of the volume ingested is from the T. gravida distribution. The apparent multi-peak selection observed in our data is discussed in reference to two separate hypotheses and in light of other recent work pertaining to selective grazing by copepods.  相似文献

The brain of a cod, Gadus morhua L., caught with damaged lateral lines, has been investigated histologically. Injured ganglion cells in the tuberculum acusticum and atrophic Mauthner-cells are demonstrated by staining with gallocyanin, lateral nerve fibres in the mesencephalon by employing Gleessilver impregnation. Terminals of degenerating lateral nerve fibres could be identified as “boutons terminaux”. The course of the afferent fibres from the lateral line through the metencephalon into the mesencephalon is as follows: Fibres of the lateral-line nerve, directly injured by an inflammation, run from the damaged lateral line to their ganglion cells in the tuberculum acusticum in the medulla; the ganglion cells are degenerated to the limits of visibility; from these cells fibres run to the Mauthner-cells, which became atrophic due to lack of afferent impulses. Other fibres run from the tuberculum acusticum into two regions of the subtectum in the mesencephalon, where they terminate in axo-somatic synapses.  相似文献

Marine polychaetes Nereis virens (Sars) were exposed to 9 ppb Hg as mercuric chloride in the aquarium water. Concentrations of mercury in the water and in the worms were monitored using radiolabelled mercury. After 11 d, the average mercury concentration in the nereids was 8.41 ppm Hg and the bioconcentration factor was calculated to be 930. Accumulations of mercury in the tissues were made visible for light and electron microscopy by autometallographic silver enhancement. Mercury was demonstrated in the intestine, nephridia, epidermis and cuticula. In the intestine, mercury was found to be located predominantly in the apical part of the epithelial cells. In the nephridia, mercury deposits were apparent in the peritoneal cells of the nephridial tubules. Silver-enhanced mercury was also observed at the epicuticula, in the collagen fibres of the endocuticula and in the epidermal basal cells. Intracellularly, the observed mercury accumulations were localised predominantly in lysosomes. Extracellularly, mercury was observed in the basal laminae of the intestine and the epidermis, and also in the intestinal peritrophic membrane. How N. virens copes with mercury toxicity is discussed.  相似文献
