Abstract

The modification effect of Nafion on the generation performance at different temperatures and methanol concentrations was investigated. The direct methanol fuel cell performances and electrochemical properties of the DMFC system using as-received Nafion117, a modified Nafion membrane, and using the Nafion117 MEA preparation by spray treatment on the surface of each catalyst layer for conductivity improvement. The open circuit voltage using the modified Nafion membrane was higher than that using Nafion117 at the cell temperature of 343–383 K and methanol concentration of 1.5–10 kmol/m3. The spray treatment of MEA was effective and improved the short circuit current up to 461 mA/cm2 at a 5 kmol/m3 methanol concentration in comparison with no treatment of MEA because of the low interfacial resistance. The power density of 75 mW/cm2 (no treatment Nafion117 MEA:40 mW/cm2) was obtained. The method of the spray treatment was found to be very effective for the DMFC system. The cell performance of Nafion117 MEA increased with the methanol concentration because of the reduction of the concentration overvoltage.  相似文献

One of the most practical approaches for establishing a successful microbial fuel cell (MFC) is to fasten the oxidation rate of the substrate by the microorganisms to get quick paced electron transfer between microbes and electrode. A genetically modified Escherichiacoli, overexpressing α-amylase, is constructed and applied as biocatalyst in MFC using starch as substrate. The results are compared with nonrecombinant, native E.coli. The results show better performance for the MFC containing the recombinant strain demonstrated by higher power density (PD), lower resistance, and significant electrochemical activity. Maximum PD has been recorded as 279.04 mW m?2 compared to 120.33 Mw m?2 for the MFC operated with nonrecombinant E.coli. The impedance results also suggest the effectiveness of the recombinant strain by lowering the internal resistance by more than half order as compared to the nonrecombinant one. These results affirm that the engineered strain can be used as a superior biocatalyst in contrast to the native strain and by using the technique of genetic alteration; gene of interest can be inserted based on the substrate to be treated. So, this work gives a useful insight for accomplishing successful MFC operation with the use of bacterial stains engineered at the molecular level.  相似文献

We examined the possibility of harvesting electricity from the surface of a tidal mud flat using a cylindrical-type sediment microbial fuel cell (SMFC), a marine mud battery (MMB), which can be applied in a sea environment where the ebb and flow occur due to tidal difference. In addition, we indirectly investigated the influence of ebb and flow in a lab, using aeration, argon gassing, and by agitating the cathodic solution. The MMBs consisted of cylindrical acrylic compartments containing a nylon membrane, an anode, and a cathode in a single body. The MMBs were stuck vertically into an artificial tidal mud flat such that the anode electrode was in direct contact with the tidal mud surface. As a result, the maximum current and power density generated were 35 mA/m2 and 9 mW/m2, respectively, thus verifying that it is possible to harvest electricity from the surface of a tidal mud flat using an MMB without burying the anode electrode in the tidal mud. Furthermore, the results of tests using an artificial turbulence flow showed the flow induced by the tidal ebb and flow could allow the performance of MMBs to be enhanced.  相似文献

Microbial fuel cells (MFCs) which are operated in continuous mode are more suitable for practical applications than fed batch ones. The aim of the present study was to characterize an air-cathode MFC operating in continuous mode and to determine the intrinsic properties for suitable performance and scalability. Air-cathode MFCs were constructed from plexiglass with a total working volume of 220 mL. Zirfon® separator used in this MFC had cross section area of 100 cm2. The air cathode MFCs were operated in fed-batch mode and then shifted to the continuous mode. To determine the behavior of anode and cathode in long term operation (274 days), their contribution in MFC performance was evaluated over time. Once the active biofilm was formed, power production and substrate consumption rate were significantly higher. The internal resistance increased with the passage of time. After stabilization of biofilm when the MFC was placed in close circuit by connecting an external resistance, the anode-reference and cathode-reference electrode behavior showed that anode potential is near to the bacterial cell inside potential. The maximum open circuit voltage achieved was 623 mV and the highest power and volumetric power density were 38.03 mW/m2 and 1296 mW/m3, respectively.  相似文献

Abstract: Water‐quality standards have been placed on fecal indicator organisms such as Escherichia coli in an attempt to limit the concentrations in water bodies. Cattle can be a significant source of bacteria to water systems, particularly when they are allowed direct access to streams. A flume study was conducted to quantify the effect and understand the transport of E. coli from directly deposited cattle manure. Five steady‐state flows, ranging from 0.00683 to 0.0176 m3/s, were studied and loads from a single cowpie exceeded the U.S. Environmental Protection Agency’s recommended water‐quality standards (235 CFU/100 ml) at each flow over the hour study period. Average E. coli concentrations ranged from 102 to 105 CFU/100 ml over the hour sampling period for all flows. High spatial variations in E. coli concentrations were often seen at each sampling time, with higher concentrations typically at the bottom of the flume. E. coli resuspension was initially greater at 0.5 min after deposition, for the lowest flow (105 CFU/m2/s); however, resuspension rates became similar over time, on the order of 103 CFU/m2/s. This study demonstrates that the concentrations of E. coli can vary over the water column, and therefore grab samples may inaccurately measure bacteria concentrations and loads in streams. In addition, resuspension rates were often high, so the incorporation of this process into water‐quality models is important for bacteria prediction.  相似文献

The photosensitizing perylenequinone toxin elsinochrome A (EA) is produced in culture by the bindweed biocontrol fungus Stagonospora convolvuli LA39 where it apparently plays a pathogenicity related role. We investigated the fate of EA with reference to its stability under different temperature and light conditions. EA remained stable when boiled in water at 100C for 2 h. Similarly, exposing EA to 3–27C in the dark for up to 16 weeks did not affect its stability either in dry or in aqueous form. However, results from irradiation experiments indicate that direct photolysis may be a significant degradation pathway for EA in the environment. EA either in dry form or dissolved in water was degraded by different irradiation wavelengths and intensities, with degradation plots fitting a first order rate kinetics. EA degraded faster if exposed in aqueous form, and at higher quantum flux density (μmol s−1 m−2). Sunlight was more effective in degrading EA than artificial white light and ultraviolet radiations (UV-A or UV-B). Exposing EA to natural sunlight, particularly, during the intense sunshine (1,420– 1,640 μmol s−1 m−2) days of 30 July to 5 August 2004 in Zurich caused the substance to degrade rapidly with half-life under such condition only 14 h. This implies that should EA gets into the environment, particularly on exposed environmental niches, such as on plant surfaces through biocontrol product spray, or released from shed diseased leaves, it may have no chance of accumulating to ‘level of concern’. Furthermore, a toxicity assay using Trichoderma atroviride P1 as biosensor showed that photo-degraded EA was not toxic, indicating that no stable toxic by-products were left.  相似文献

The effects of aspect, altitude, and slope have been observed on the moist temperate forests of Abies pindrow Spach in the Garhwal Himalaya, India. Four aspects, namely, north-east, north-west, south-east and south-west, have been studied to understand the growth behaviour of A. pindrow at varying altitudes. The total basal cover was found to be highest (5099.6 cm2/100 m2) on the north-east facing slopes, and lowest (3092.7 cm2/100 m2) on the south-east facing slopes. The A. pindrow was found to be associated with Quercus semecarpifolia as a main companion species on all the faces, whereas, Lyonia ovalifolia, was associated only on the west facing slopes.  相似文献

DOI: 10.1111/j.1752‐1688.2011.00520.x Abstract: The simulation of the impact of Escherichia coli loads from watersheds is of great interest for assessing estuarine water quality, especially in areas with shellfish aquaculture or bathing activities. For this purpose, this study investigates a model association based on the Soil and Water Assessment Tool (SWAT) coupled with a hydrodynamic model (MARS 2D; IFREMER). Application was performed on the catchment and estuary of Daoulas area (France). The daily E. coli fluxes simulated by SWAT are taken as an input in the MARS 2D model to calculate E. coli concentrations in estuarine water and shellfish. Model validation is based on comparison of frequencies: a strong relationship was found between calculated and measured E. coli concentrations for river quality (r2 = 0.99) and shellfish quality (r2 = 0.89). The important influence of agricultural practices and rainfall events on the rapid and large fluctuations in E. coli fluxes from the watershed (reaching three orders of magnitude in <24 hours) is one main result of the study. Response time in terms of seawater quality degradation ranges from one to two days after any important rainfall event (greater than 10 mm/day) and the time for estuary to recover good water quality also mainly depends on the duration of the rainfall. In the estuary, three effects (rainfall, tidal dilution, and manure spreading) have been identified as important influences.  相似文献

Performance of a full-scale duckweed-covered treatment system in removing faecal bacteria is presented. The system consisted of three ponds in series and received septage from holding tanks. Inflow averaged between 36 m3 d−1 in the cold season and 60 m3 d−1 in the warm season, resulting in a total hydraulic retention time of 88 and 58 days, respectively. Duckweed (Lemna minor) colonized the ponds in the summer and continued to grow in the cold season. Due to the difficult harvesting process of the duckweed biomass, the investigation of the treatment efficiency was carried out without plant harvesting. The system was monitored for temperature, pH, oxygen, chlorophyll-a, Escherichia coli and Enterococci. Duckweed growth resulted in chlorophyll-a concentration reduction from 924 to 13 μg L−1, causing neutral and anoxic conditions in the pond water. A temperature effect was noticed on the E. coli decay coefficient with a decreasing trend along the treatment system. Enterococci always decayed less than E. coli. Differences on decay coefficients and removal efficiencies were not observed between the three ponds for both bacterial types. Effluent quality in terms of E. coli was 489 and 1377 cfu/100 mL, in the warm and the cold seasons, respectively, with average removals of 99.65 ± 1.46% and 99.33 ± 3.03%. Total Enterococci removal was 88.91 ± 23.1% in the warm season and 94.43 ± 24.45% in the cold season, resulting in mean effluent values of 1058 and 1404 cfu/100 mL, respectively. The seasonal differences in total removal efficiencies were insignificant for both bacterial types.  相似文献

The Missouri Department of Natural Resources (MDNR) has closed or posted advisories at public beaches at Lake of the Ozarks State Park in Missouri because of Escherichia coli (E. coli) concentration exceedances in recent years. Spatial and temporal patterns of E. coli concentrations, microbial source tracking, novel sampling techniques, and beach‐use patterns were studied during the 2012 recreational season to identify possible sources, origins, and occurrence of E. coli contamination at Grand Glaize Beach (GGB). Results indicate an important source of E. coli contamination at GGB was E. coli released into the water column by bathers resuspending avian‐contaminated sediments, especially during high‐use days early in the recreational season. Escherichia coli concentrations in water, sediment, and resuspended sediment samples all decreased throughout the recreational season likely because of decreasing lake levels resulting in sampling locations receding away from the initial spring shoreline as well as natural decay and physical transport out of the cove. Weekly MDNR beach monitoring, based solely on E. coli concentrations, at GGB during this study inaccurately predicted E. coli exceedances, especially on weekends and holidays. Interestingly, E. coli of human origin were measured at concentrations indicative of raw sewage in runoff from an excavation of a nearby abandoned septic tank that had not been used for nearly two years.  相似文献

In the present study, bioelectricity generation using a living plant in soil was achieved by converting solar energy into electricity on the principles of plant photosynthesis. The plants Chlorophytum comosum, Chasmanthe floribunda and Papyrus diffuses were used in microbial fuel cells (MFCs) and produced, respectively, a maximum current of 25, 31 and 19 mA/m2, without the use of any nutrient or membrane. In this study, the importance of light was demonstrated by the high current values generated during daytime, and the microorganisms at the anode-rhizosphere interface were examined.  相似文献

Macroinvertebrates were examined on an impounded valley marsh in Stonington, Connecticut, that has changed from aTypha-dominated system to one with typical salt-marsh vegetation during 13 years following the reintroduction of tidal exchange. Animal populations on this restored impounded marsh were evaluated by comparing them with populations on a nearby unimpounded valley marsh of roughly the same size. Populations of the high marsh snail,Melampus bidentatus Say, were quantitatively sampled along transects that extended from the water-marsh edge to the upland; those of the ribbed mussel,Geukensia demissa Dillwyn, were sampled in low marsh areas on transects along the banks of creeks and mosquito ditches. The occurrence of other marsh invertebrates also was documented, but their abundance was not measured. The mean density ofMelampus was 332±39.6 SE/m2 on the restored impounded marsh and 712±56.0 SE/m2 on the unimpounded marsh. However, since snails were larger on the restored impounded marsh, the difference in snail biomass was less pronounced than the difference in snail density. MeanMelampus biomass was 4.96±0.52 SE g dry wt/m2 on the restored impounded marsh and 6.96±0.52 SE g dry wt/m2 on the unimpounded marsh. On the two marshes, snail density and biomass varied in relation to plant cover and other factors. The density and biomass ofGeukensia at the edge of the marsh were comparable on the restored impounded and unimpounded marshes. Mean mussel densities ranged from 80 to 240/m2 and mean mussel biomass varied from 24.8–64.8 g dry wt/m2 in different low marsh areas. In contrast, below the impoundment dike, meanGeukensia density was 1100±96.4 SE/m2 and meanGeukensia biomass was 303.6±33.28 SE g dry wt/m2. A consideration of all available evidence leads to the conclusion that the impounded marsh is in an advanced phase of restoration.  相似文献

Our objective was to determine whether a colony of cliff swallows nesting underneath a bridge would yield a measurable increase in fecal indicator bacteria (specifically Escherichia coli) in the underlying creek. When the swallows were absent, dry‐weather concentrations of E. coli upstream and downstream of the bridge (in Austin, Texas) were below the Texas contact recreation criteria. When the swallows were present, dry‐weather geometric‐mean E. coli concentrations increased significantly from upstream (43 most probable number [MPN]/100 ml) to downstream (106 MPN/100 ml) of the bridge. One exceedance and one near‐exceedance of the Texas single‐sample contact recreation criterion were observed during the swallows’ nesting phase. When the swallows were present, the downstream E. coli geometric‐mean concentration in storm events (875 MPN/100 ml) was significantly higher than the upstream concentration (356 MPN/100 ml), suggesting that runoff flushes swallow feces from the ground into the creek. Although the loading of E. coli from cliff swallows nesting under bridges can be significant (e.g., dry‐weather loading of 3.1 × 108 MPN/day/nest), the zoonotic potential of the cliff swallow must be examined to determine the risk to human health from contact recreation in waters contaminated with cliff swallow feces.  相似文献

Understanding sediment Escherichia coli levels (i.e., pathogen indicators) and their contribution to the water column during resuspension is critical for predicting in‐stream E. coli levels and the potential risk to human health. The U.S. Environmental Protection Agency's current water quality testing strategies, however, rely on water borne E. coli concentrations to assess stream E. coli levels and identify impaired waters. In this work, we conducted a scenario analysis using a range of flows, sediment/water bacteria fractions, and particle sizes to which E. coli attach to assess the impact of E. coli in streambed sediments on water column E. coli levels. We used simple sediment transport theory to calculate the potential total E. coli concentrations in a stream with and without the resuspension process. Results clearly indicate that inclusion of resuspending sediment attached E. coli is necessary for watershed assessments and data on sediment attached E. coli concentrations is much needed. When neglecting the streambed sediment E. coli concentrations, the model predicted average E. coli loads of 107 Colony Forming Units (CFU)/s; however, when streambed sediment E. coli concentrations were included in the model, the predictions ranged from 1010 to 1014 CFU/s. To evaluate the predictions, E. coli data in the streambed sediment and the water column were monitored in Squaw Creek, Iowa. Comparisons between measured and predicted E. coli loads yielded an R2‐value of 0.85.  相似文献

In interior western North America, many riparian forests dominated by cottonwood and willow are failing to reproduce downstream of dams. We tested the hypothesis that establishment is now prevented by absence of the bare, moist substrate formerly provided by floods and channel movement. Along Boulder Creek, a dammed stream in the Colorado plains, we tested the effects of disturbance (sod removal), irrigation, and addition of seed on the establishment of seedings of plains cottonwood (Populus deltoides subsp.monilifera) and peachleaf willow (Salix amygdaloides). In unirrigated, undisturbed plots, mean cottonwood density was 0.03 seedlings/m2. Irrigation or disturbance alone produced mean cottonwood densities of 0.39 and 0.75 seedlings/m2. Plots that were both irrigated and disturbed produced a mean cottonwood density of 10.3 seedlings/m2. The effects of irrigation and disturbance on cottonwood establishment were significant (P<0.005); added seed had no significant effect (P=0.78). The few cottonwood seedlings in unirrigated plots were in low positions susceptible to scour by future moderate flows. We conclude that cottonwood establishment along Boulder Creek is limited by the scarcity of bare, moist sites safe from future scour. Establishment of peachleaf willow was significantly affected only by disturbance; daily sprinkler irrigation did not provide sufficient moisture to increase survival of this species. Our results demonstrate the feasibility of restoring plains cottonwood forests using natural seedfall, even where only widely scattered adult trees are present. Because use of natural seedfall conserves the genetic makeup of the local population, this method may be preferable to the use of imported cuttings.  相似文献

Hobart's bushlands consist of eight distinct vegetation types ranging from open woodland to wet forest. Fine fuel accumulation characteristics are distinctive across this range of vegetation types and mostly seem to conform to the function Wt = Wss(1−ekt)+ l·92(ekt), although there is considerable scatter around the lines of best fit in many types.This information contributes to determining the applicability and appropriate frequency of controlled burning as a means of reducing fuels to protect life and property from wildfire. Regular burning of grassy vegetation should ensure relatively safe zones within the urban/bush matrix. It is likely that burning heathy forest types at frequencies that would ensure safe fuel loads would be ecologically detrimental. It would be difficult and futile to use fuel reduction burning in wet forest types as a means of protecting life and property. The fuel accumulation characteristics and the existing ecological knowledge of Allocasuarina verticillata shrubland suggests that fuel reduction burning would be counter-productive in this vegetation type.  相似文献

The increasing use and subsequent accumulation of polystyrene containers has triggered a substantial environmental problem. This study investigated using varied percentages of solid waste polystyrene disposable food dishes in the production of lightweight concrete samples with 350 kilograms per cubic meter (kg/m3) of cement and a density of 1,300 kg/m3. The polystyrene disposable dishes were ground into beads of 0–3 millimeters (mm) and 3–6 mm in size. First, the characteristics of Type II Portland cement, polystyrene, and aggregates were examined. The following characteristics of concrete using ASTM International and British Standards Institution standards were tested: slump, compressive strength, ability to resist chloride ion penetration, and resistance of concrete to rapid freezing and thawing cycles. Scanning electron microscopy (SEM) and energy dispersive X‐ray spectroscopy analytical techniques were also used. The slump of samples varied between 40 and 70 mm and was not dependent on either the polystyrene percentage or the size of the polystyrene beads in the concrete samples (p‐value > .05). The compressive strength of the concrete samples after 90 days of curing, and using different percentages of polystyrene, varied between 96 and 113 kilograms per square centimeter (kg/cm2). The resistance of the samples to the freezing and thawing cycle and chloride ion penetration were affected unfavorably by the presence of the polystyrene. The SEM technique indicated that concrete samples containing 15% and 25% polystyrene had denser crystals and less void than concrete samples with 40% and 55% polystyrene.  相似文献

This study was aimed to investigate the changes in the human electroencephalographic (EEG) signal caused by modulated low-level microwaves. The 450 MHz microwave exposure modulated at 40 Hz and 70 Hz frequencies was applied to a group of 15 volunteers. The field power density at the scalp was 0.16 mW/cm2. Ten cycles of the exposure (1 min on and 1 min off) at both modulation frequencies were applied. Analysis of the EEG signal was performed using three different methods: nonlinear method of scaling analysis for length distribution of low variability periods (LDLVP), relative changes in EEG energy (S-parameter) and beta ratio (H-parameter). The analysis revealed significant changes caused by microwave for the whole group (H-parameter method). The exposure caused increase of the EEG beta power (S-parameter method). Statistically significant changes in EEG were detected for four subjects (26.7%) at 40 Hz modulation frequency (LDLVP method).  相似文献

Abstract: Escherichia coli was used as a bacterial tracer for the development, calibration, and validation of a watershed scale fate and transport model to be extended to a suite of reference pathogens (Cryptosporidium, Giardia, Campylobacter, E. coli O157:H7). E. coli densities in water and sediments from the Blackstone River Watershed, Massachusetts, were measured at three sites for a total of five wet weather events and three dry weather events covering three seasons. The confirmed E. coli strains were identified by ribotyping for tracking the sources of E. coli and for determining the association of downstream E. coli isolates with isolates from upstream sediments. A large number of downstream samples were associated with upstream sediment sources of E. coli. E. coli densities ranged from 71 to 6,401 MPN/100 ml in water samples and from 2 to 335 MPN/g in sediments. Pearson correlation analysis revealed significant correlations between E. coli and total coliforms in water (r = 0.777, p < 0.01) and sediments (r = 0.728, p < 0.01). In addition, E. coli concentrations in water were weakly correlated with sediment particle size and sediment concentrations (r = 0.298, p < 0.01). A hydrologic model, WATFLOOD/SPL9, was used to predict the temporal and spatial variation of E. coli in the Blackstone River. The rapid rise of stream E. coli densities was more accurately predicted by the model with the inclusion of sediment resuspension, thus demonstrating the importance of the process.  相似文献

Summary This study focuses on discrimination of changes, produced by low-level microwave exposure in intensity and time variability of the human EEG at rest. The power spectral density (PSD) method and nonlinear scaling analysis of the length distribution of low variability periods (LDLVP) were selected for analysis of the EEG signal. During the study, 19 healthy volunteers were exposed to a microwave (450 MHz) of 217 Hz frequency on-off modulation. The field power density at the scalp was 0.16 mW/cm2. The experimental protocol consisted of ten cycles of repetitive microwave exposure. Signals from frontal, temporal, parietal and occipital EEG channels on EEG theta, alpha and beta rhythms were analysed. Exposure to microwave causes average increase of EEG activity. LDLVP analysis discriminated significant effect in time variability for 2 subjects (11%). PSD method detected significant changes in intensity for 4 subjects (21%). The effect of low-level microwave exposure is stronger on EEG beta rhythm in temporal and parietal regions of the human brain.  相似文献
