Kitchen wastes containing high amounts of carbohydrates have potential as low-cost substrates for fermentable sugar production. In this study, enzymatic saccharification of kitchen waste was carried out. Response surface methodology (RSM) was applied to optimize the enzymatic saccharification conditions of kitchen waste. This paper presents analysis of RSM in a predictive model of the combined effects of independent variables (pH, temperature, glucoamylase activity, kitchen waste loading, and hydrolysis time) as the most significant parameters for fermentable sugar production and degree of saccharification. A 100 mL of kitchen waste was hydrolyzed in 250 mL of shake flasks. Quadratic RSM predicted maximum fermentable sugar production of 62.79 g/L and degree of saccharification (59.90%) at the following optimal conditions: pH 5, temperature 60°C, glucoamylase activity of 85 U/mL, and utilized 60 g/L of kitchen waste as a substrate at 10 h hydrolysis time. The verification experiments successfully produced 62.71 ± 0.7 g/L of fermentable sugar with 54.93 ± 0.4% degree of saccharification within 10 h of incubation, indicating that the developed model was successfully used to predict fermentable sugar production at more than 90% accuracy. The sugars produced after hydrolysis of kitchen waste were mainly attributed to monosaccharide: glucose (80%) and fructose (20%). The fermentable sugars obtained were subsequently used as carbon source for bioethanol production by locally isolated yeasts: Saccharomyces cerevisiae, Candida parasilosis, and Lanchancea fermentati. The yeasts were successfully consumed as sugars hydrolysate, and produced the highest ethanol yield ranging from 0.45 to 0.5 g/g and productivity between 0.44 g L–1 h–1 and 0.47 g L–1 h–1 after 24-h incubation, which was equivalent to 82.06–98.19% of conversion based on theoretical yield.  相似文献

In the present work, characteristics of ethanol production from a single particle composing of pretreated rice straw, cellulase, and β-glucosidase were invesitgated by simultaneous saccharification and fermentation (SSF). The experiment experienced a start-up stage for S. cerevisiae biofilm formation, which was operated at an initial pH value of 4.8 for yeast solution, culture temperature of 30°C, flow rate of 0.8 mL/min for yeast solution, and stably operating stage for ethanol production at a culture temperature of 30°C. Investigations found that the maximal ethanol yield of 9.7 mg/g and the biofilm thickness of 0.37 mm were obtained at 30 mL/min of carrier gas flow rate. Also, the optimal interval time of water supplement was 4 h for SSF. The results show that the appropriate gas flow rate and the interval time of water supplement can keep the high activities of biofilm and enzymes during SSF and result in a high ethanol yield.  相似文献

Fuel ethanol was produced using rice straw with the simultaneous saccharification and fermentation (SSF) method. The influence of cellulose liquefaction pretreatment and Fe2+ quantity on ethanol productivity was investigated in detail. At the same time, the optimized conditions including fermentation temperature, Fe2+ amount, yeast inoculation quantity, and the inoculated cellulose enzyme dosage in the SSF process were systematically investigated by analyzing fuel ethanol yield. The result indicated that fuel ethanol yield was 0.319 g per gram rice straw by SSF approach when appropriate amount of Fe2+ was added into the reaction system. The optimal technology parameters were: fermenting temperature of 36°C, Fe2+ amount of 4 mg · g?1, inoculating proportion of 20%, cellulose enzyme of 20 IU · g?1, and Pachysolen tannophilu/saecharomyces cerevisiae of 1:2 ratio. The ethanol yield under the best conditions was larger than that of the control group. We hope that this research can facilitate to achieve large-scale comprehensive utilization for rice straw.  相似文献

An investigation was carried out on the performance of an anaerobic sequencing batch biofilm reactor (ASBBR) treating diluted cheese whey when submitted to different feed strategies and volumetric organic loads (VOL). Polyurethane foam cubes were used as support for biomass immobilization and stirring was provided by helix impellers. The reactor with a working volume of 3 L treated 2 L of wastewater in 8-h cycles at 500 rpm and 30 degrees C. The organic loads applied were 2, 4, 8 and 12 g COD L(-1) d(-1), obtained by increasing the feed concentration. Alkalinity was supplemented at a ratio of 50% NaHCO(3)/COD. For each organic load applied three feed strategies were tested: (a) batch operation with 8-h cycle; (b) 2-h fed-batch operation followed by 6-h batch; and (c) 4-h fed-batch followed by 4-h batch. The 2-h fed-batch operation followed by 6-h batch presented the best results for the organic loads of 2 and 4 g COD L(-1) d(-1), whereas the 4-h fed-batch operation followed by 4-h batch presented results slightly inferior for the same organic loads and the best results at organic loads of 8 and 12 g COD L(-1) d(-1). The concentration of total volatile acids varied with fill time. For the higher fill times maximum concentrations were obtained at the end of the cycle. Moreover, no significant difference was detected in the maximum concentration of total volatile acids for any of the investigated conditions. However, the maximum values of propionic acid tended to decrease with increasing fill time considering the same organic load. Microbiological analyses revealed the presence of Methanosaeta-like structures and methanogenic hydrogenotrophic-like fluorescent bacilli. No Methanosarcina-like structures were observed in the samples.  相似文献

Dilute acid pretreatment and steam pretreatment were evaluated for maximum sugars release and ethanol production from sweet sorghum bagasse (SSB). The fermentation potential of the condensate and hydrolysate obtained from steam pretreatment (10 kg/cm2, 10 minutes) and dilute acid hydrolysis (1% (w/w) sulphuric acid, 25% substrate loading) respectively, was checked with Pichia stipitis NCIM 3497 and Debaryomyces hansenii sp. Ethanol production and yield using acid hydrolysate was higher with Debaryomyces hansenii sp. (28.4 g/L and 0.37 g/g respectively) as compared with Pichia stipitis NCIM 3497 (21.9 g/L and 0.29 g/g respectively).  相似文献

This work presents an analysis of a stirred anaerobic sequencing discontinuous reactor with different substrate feeding strategies resulting in batch, fed-batch/batch and fed-batch operating modes. The reactor, containing granulated biomass, was fed with approximately 2.0L of synthetic domestic wastewater with Chemical Oxygen Demand of nearly 500 mg/L per cycle and operated at 30 degrees C and 50 rpm. Three feeding strategies with a total cycle time of 6 h, including 30-min settling, were adopted: batch mode with a fill cycle of 6 min, a fed-batch/batch mode with fill cycles of 60, 120 and 240 min and fed-batch mode with a fill cycle of 320 min. The system attained average non-filtered and filtered substrate removal efficiency of 78 and 84%, respectively, for all operating conditions, presenting good stability, solid retention and no granule break-up. A first order kinetic model with a residual organic matter concentration was proposed to analyze the influence of the feeding strategy on the performance during a cycle and bicarbonate alkalinity and total volatile acids concentration profiles were also quantified in order to verify the transient stability behavior.  相似文献

In current work, an optimum solid loading (solid: liquid = 1:20), pH (4.8), temperature (50°C), and enzyme dosing of 20 filter paper unit (amount of enzyme required to release 1 µmol of glucose as reducing sugar from filter paper in per mL per minute) were enumerated for enzymatic hydrolysis of banana stem using cellulase from Trichoderma reesei. Further, inhibition study on enzymatic hydrolysis of banana stem was investigated by the supplementation of monosaccharides (glucose, galactose, mannose, xylose, and arabinose), disaccharide (cellobiose), and inhibitors (acetic acid and furfural obtained from pre-enzymatic hydrolysis steps). Glucose and cellobiose showed inhibitory effect on enzymatic hydrolysis of pretreated banana stem at or above 8 g/L while galactose, mannose, and xylose showed a significant inhibitory effect at or above 4 g/L. Instead of inhibition, arabinose enhanced the enzymatic hydrolysis with increase in total reducing sugars. Acetic acid did not show any significant inhibition while furfural inhibited the system at a comparative low concentration of 2 g/L. Further, scanning electron microscopy analysis was performed to investigate the difference in ultra-structural morphology of raw biomass, pretreated biomass, and biomass obtained after enzymatic hydrolysis.  相似文献

In this paper the cost of producing the enzyme laccase by the white-rot fungus Trametes pubescens under both submerged (SmF) and solid-state fermentation (SSF) conditions was studied. The fungus was cultured using more than 45 culture medium compositions. The cost of production was estimated by analyzing the cost of the culture medium, the cost of equipment and the operating costs. The cost of the culture medium represented, in all cases, the highest contribution to the total cost, while, the cost of equipment was significantly low, representing less than 2% of the total costs. The cultivation under SSF conditions presented a final cost 50-fold lower than the one obtained when culturing under SmF conditions at flask scale. In addition, the laccase production under SSF conditions in tray bioreactors reduced the final cost 4-fold compared to the one obtained under SSF conditions at flask scale, obtaining a final price of 0.04 cent €/U.  相似文献

The influence of temperature (175 to 195°C), residence time (5 to 15 min), and sulfuric acid concentration in high (2 to 4% w/w) and low (0.5 to 1.5% w/w) levels in dilute acid pretreatment of Moringa oleifera Lam is studied. Glucose recoveries in the liquid fraction and in the hydrolyzed insoluble fraction as well as the presence of inhibitors in the liquid fraction are determined. Best experimental results are achieved at 185°C, 2% w/w acid concentration, and 5 min reaction time obtaining a glucose recovery of 83.68%. An increment in 48.81% in glucose yield compared with the one of not pretreated Moringa is obtained. 0.13 g ethanol/g Moringa from fermentation of pre-hydrolysate and hydrolysate obtained at the optimal pre-treatment conditions are obtained.  相似文献

The objective of this work was to analyze the interaction effects between temperature, feed strategy and COD/[SO(4)(2-)] levels, maintaining the same ratio, on sulfate and organic matter removal efficiency from a synthetic wastewater. This work is thus a continuation of Archilha et al. (2010) who studied the effect of feed strategy at 30 °C using different COD/[SO(4)(2-)] ratios and levels. A 3.7-L anaerobic sequencing batch reactor with recirculation of the liquid phase and which contained immobilized biomass on polyurethane foam (AnSBBR) was used to treat 2.0 L synthetic wastewater in 8 h cycles. The temperatures of 15, 22.5 and 30 °C with two feed strategies were assessed: (a) batch and (b) batch followed by fed-batch. In strategy (a) the reactor was fed in 10 min with 2 L wastewater containing sulfate and carbon sources. In strategy (b) 1.2 L wastewater (containing only the sulfate source) was fed during the first 10 min of the cycle and the remaining 0.8 L (containing only the carbon source) in 240 min. Based on COD/[SO(4)(2-)] = 1 and on the organic matter (0.5 and 1.5 gCOD/L) and sulfate (0.5 and 1.5 gSO(4)(2-)/L) concentrations, the sulfate and organic matter loading rates applied were 1.5 and 4.5 g/L.d, i.e., same COD/[SO(4)(2-)] ratio (=1) but different levels (1.5/1.5 and 4.5/4.5 gCOD/gSO(4)(2-)). When reactor feed was 1.5 gCOD/L.d and 1.5 gSO(4)(2-)/L.d, gradual feeding (strategy b) showed to favor sulfate and organic matter removal in the investigated temperature range, indicating improved utilization of the electron donor for sulfate reduction. Sulfate removal efficiencies were 87.9; 86.3 and 84.4%, and organic matter removal efficiencies 95.2; 86.5 and 80.8% at operation temperatures of 30; 22.5 and 15 °C, respectively. On the other hand, when feeding was 4.5 gCOD/L.d and 4.5 gSO(4)(2-)/L.d, gradual feeding did not favor sulfate removal, indicating that gradual feeding of the electron donor did not improve sulfate reduction.  相似文献

In order to obtain the characteristics of anaerobic fermentation with different parts of corn stalks at low concentrations, air-dried corn stalks stem bark (SB), stem pith (SP), leaves (L), and corn stalks (CS) were, respectively, mixed with cow dung to perform fermentation at the temperature of 35 oC and carbon-to-nitrogen ratio (C/N) of 25. Mixed with cow dung compost, the fermentation broths were adjusted to a neutral pH value. Along with the enhancing of the total solid (TS) content of SB, SP, L, and CS fermentation broths, both of the daily biogas yields and methane contents increased under the same fermentation condition, except for Sample TS 6% of L. The optimal TS content of SB, SP, L, and CS broth is 8%, 5%, 10%, and 8%, separately. In 35 days, the highest methane yield of SB, SP, L, and CS broth was 125.0 mL/(volatile solid) VS g, 115.3 mL/VS g, 109.7 mL/VS g, and 80.0 mL/VS g, respectively, and the potential of methane transformation production of broth ranks as: SB> L> SP> CS. Daily methane producing rate of SB, SP, and L broth are faster than that of CS. It is necessary to separate the corn stalks into different parts to ferment because the optimal fermentation concentrations for the different parts are different. Additionally, the Gompertz equation was also adopted to simulate the anaerobic digestion process of different materials. The Gompertz equation fitting parameters show that the biodegradation (from easy to difficult) was: L < SP < SB < CS.  相似文献

Biological sulfate reduction was studied in a laboratory-scale anaerobic sequential batch reactor (14 L) containing mineral coal for biomass attachment. The reactor was fed industrial wastewater with increasingly high sulfate concentrations to establish its application limits. Special attention was paid to the use of butanol in the sulfate reduction that originated from melamine resin production. This product was used as the main organic amendment to support the biological process. The reactor was operated for 65 cycles (48 h each) at sulfate loading rates ranging from 2.2 to 23.8 g SO(4)(2-)/cycle, which corresponds to sulfate concentrations of 0.25, 0.5, 1.0, 2.0 and 3.0 g SO(4)(2-) L(-1). The sulfate removal efficiency reached 99% at concentrations of 0.25, 0.5 and 1.0 g SO(4)(2-) L(-1). At higher sulfate concentrations (2.0 and 3.0 g SO(4)(2-) L(-1)), the sulfate conversion remained in the range of 71-95%. The results demonstrate the potential applicability of butanol as the carbon source for the biological treatment of sulfate in an anaerobic batch reactor.  相似文献

Using dog food as a model of the organic waste that comprises composting raw material, the degradation pattern of organic materials was examined by continuously measuring the quantity of CO2 evolved during the composting process in both batch and fed-batch operations. A simple numerical model was made on the basis of three suppositions for describing the organic matter decomposition in the batch operation. First, a certain quantity of carbon in the dog food was assumed to be recalcitrant to degradation in the composting reactor within the retention time allowed. Second, it was assumed that the decomposition rate of carbon is proportional to the quantity of easily degradable carbon, that is, the carbon recalcitrant to degradation was subtracted from the total carbon remaining in the dog food. Third, a certain lag time is assumed to occur before the start of active decomposition of organic matter in the dog food; this lag corresponds to the time required for microorganisms to proliferate and become active. It was then ascertained that the decomposition pattern for the organic matter in the dog food during the fed-batch operation could be predicted by the numerical model with the parameters obtained from the batch operation. This numerical model was modified so that the change in dry weight of composting materials could be obtained. The modified model was found suitable for describing the organic matter decomposition pattern in an actual fed-batch composting operation of the garbage obtained from a restaurant, approximately 10 kg d(-1) loading for 60 d.  相似文献

In this research study, biodiesel has been successfully produced from vegetable seed oil of an indigenous plant Salvadora persica L. that meets the international biodiesel standard (ASTM D6751). The biodiesel yield was 1.57 g/5 g (31.4% by weight) and the in-situ transesterification ester content conversion was 97.7%. The produced biodiesel density was 0.894 g/mL, its kinematic viscosity 5.51 mm2/s, HHV 35.26 MJ/kg, flash point 210°C, cetane no. 61, and sulfur content 0.0844%. Thermal analysis of the biodiesel showed that 97% weight loss was achieved at 595°C with total oxidation of the biodiesel. The production energy efficiency was 0.46% with a lab scale setup, assuming the volume fraction ratio (volume of the sample/total volume of the equipment used). The results revealed that single-step in-situ transesterification method is suitable for the production of biodiesel from S. persica seed oil.  相似文献

Waste cooking oil is a potential substitution of refined vegetable oil for the production of biodiesel due to the low cost of raw material and for solving their disposal problem. In this study, optimization of esterification process of free fatty acids in artificially acidified soybean oil with oleic acid has been carried out using methanol as an agent and ion exchange resin as a heterogeneous catalyst. The esterification reaction has been investigated based on the mass balance of the developed model. The model has been validated against experimental data and effects of temperature and catalyst weight have been analyzed. Thereafter, optimization process has been fulfilled for two different objective functions as conversion of acid oil and benefit. Optimization results indicated that the maximum conversion of acid is 95.95%, which is achievable at 4.48-g catalyst loading and reaction temperature of 120°C. Maximum benefit was obtained as US$0.057 per batch of reaction at a catalyst amount of 1 g and temperature of 120°C.  相似文献

The aim of this investigation was to evaluate the influence of batch versus continuous flow on the removal efficiencies of chemical oxygen demand (COD), nitrogen (N) and total phosphorus (TP) in tropical subsurface flow constructed wetlands (SSF CW). The quantitative role of the higher aquatic plants in nutrient removal in these two operational modes was also investigated. Results indicated no significant difference (p > 0.05) in COD removal between batch and continuous flow modes for either the planted or unplanted treatments. Furthermore, the batch-loaded planted wetlands showed significantly (p < 0.05) higher ammonium removal efficiencies (95.2%) compared with the continuously fed systems (80.4%), most probably because the drain and fill batch mode presented systematically more oxidized environmental conditions. With respect to TP removal, for both planted and unplanted beds, there was significant enhancement (p < 0.05) in batch flow operation (69.6% for planted beds; 39.1% for unplanted beds) as compared to continuous flow operation (46.8% for planted beds; 25.5% for unplanted beds). In addition, at a 4-day hydraulic retention time (HRT), the presence of plants significantly enhanced both ammonia oxidation and TP removal in both batch and continuous modes of operation as compared to that for unplanted beds. An estimation of the quantitative role of aeration from drain and fill operation at a 4-day HRT, as compared to rhizosphere aeration by the higher aquatic plant, indicated that drain and fill operation might account for only less than half of the higher aquatic plant's quantitative contribution of oxygen (1.55 g O2 per m2 per day for batch flow versus 1.13 g O2 per m2 per day for continuous flow).  相似文献

The olive mill waste (OMW) generated from olive oil extraction process constitutes a major environmental concern owing to its high organic and mineral matters and acidic pH. Anaerobic digestion (AD) is a main treatment for reducing the organic matter and toxic substances contained in OMW and generating at the same time, energy in the form of biogas. AD of OMW that contains lignocellulose is limited by the rate of hydrolysis due to their recalcitrant structure. This study is devoted to the effect of Fenton process (FP) pretreatment on olive mill wastewater (OMSW) /olive mill solid waste (OMWW) co-digestion to improve their digestibility and in this way the biogas production. The FP pretreatment was performed in batch mode at 25°C, various H2O2/[Fe2+] ratios (100–1200), catalyst concentration ([Fe2+]) ranging from 0.25 to 2 mM, reaction time varying from 30 to150 min, and different pH (3–11). The best performance was obtained with H2O2/[Fe2+] = 1000, [Fe2+] = 1.5 mM, 120 min, and pH 3. Biochemical methane potential (BMP) tests conducted in batch wise digester and at mesophilic conditions (37 °C) showed that cumulative biogas and methane production were higher without FP treatment, and correspond to 699 and 416 mL/g VS, respectively. However, pre-treated OMSW results into an increase of 24% of methane yield. After 30 days of AD, the methane yield was 63%, 54%, and 48%, respectively, for OMSW treated without iron precipitation, with iron precipitation and untreated OMSW sample.  相似文献

The LCA emissions from four renewable energy routes that convert straw/corn stover into usable energy are examined. The conversion options studied are ethanol by fermentation, syndiesel by oxygen gasification followed by Fischer Tropsch synthesis, and electricity by either direct combustion or biomass integrated gasification and combined cycle (BIGCC). The greenhouse gas (GHG) emissions of these four options are evaluated, drawing on a range of studies, and compared to the conventional technology they would replace in a western North American setting. The net avoided GHG emissions for the four energy conversion processes calculated relative to a “business as usual” case are 830 g CO2e/kWh for direct combustion, 839 g CO2e/kWh for BIGCC, 2,060 g CO2e/L for ethanol production, and 2,440 g CO2e/L for FT synthesis of syndiesel. The largest impact on avoided emissions arises from substitution of biomass for fossil fuel. Relative to this, the impact of emissions from processing of fossil fuel, e.g., refining of oil to produce gasoline or diesel, and processing of biomass to produce electricity or transportation fuels, is minor.  相似文献

Biosorption of zinc (II) ions onto pre-treated powdered waste sludge (PWS) was investigated using a completely mixed tank operating in fed-batch mode instead of an adsorption column. Experiments with variable feed flow rate (0.05-0.5 L h(-1)), feed Zn(II) ion concentrations (37.5-275 mg L(-1)) and amount of adsorbent (1-6 g PWS) were performed using fed-batch operation at pH 5 and room temperature (20-25 degrees C). Break-through curves describing variations of aqueous (effluent) zinc ion concentrations with time were determined for different operating conditions. Percent zinc removal from the aqueous phase decreased, but the biosorbed (solid phase) zinc ion concentration increased with increasing feed flow rate and zinc concentration. A modified Bohart-Adams equation was used to determine the biosorption capacity of PWS (q'(s)) and the rate constant (K) for zinc ion biosorption. Biosorption capacity (q'(s)=57.7 g Zn kg(-1) PWS) of PWS in fed-batch operation was found to be comparable with powdered activated carbon (PAC) in column operations. However, the adsorption rate constant (K=9.17 m(3) kg(-1) h(-1)) in fed-batch operation was an order of magnitude larger than those obtained in adsorption columns because of elimination of mass transfer limitations encountered in the column operations. Therefore, a completely mixed tank operated in fed-batch mode was proven to be more advantageous as compared to adsorption columns due to better contact between the phases yielding faster adsorption rates.  相似文献
