Microbial biomass carbon and enzyme activities of urban soils in Beijing

Introduction

To promote rational and sustainable use of soil resources and to maintain the urban soil quality, it is essential to assess urban ecosystem health. In this study, the microbiological properties of urban soils in Beijing and their spatial distribution patterns across the city were evaluated based on measurements of microbial biomass carbon and urease and invertase activities of the soils for the purpose of assessing the urban ecosystem health of Beijing.

This paper reports the influences of the herbicide butachlor (n-butoxymethlchloro -2', 6'-diethylacetnilide) on microbial populations, respiration, nitrogen fixation and nitrification, and on the activities of dehydrogenase and hydrogen peroxidase in paddy soil. The results showed that the number of actinomycetes declined significantly after the application of butachlor at different concentrations ranging from 5.5 microg g(-1) to 22.0 microg g(-1) dried soil, while that of bacteria and fungi increased. Fungi were easily affected by butachlor compared to the bacteria. The growth of fungi was retarded by butachlor at higher concentrations. Butachlor however, stimulated the growth of anaerobic hydrolytic fermentative bacteria, sulfate-reducing bacteria (SRB) and denitrifying bacteria. The increased concentration of butachlor applied resulted in the higher number of SRB. Butachlor inhibited the growth of hydrogen-producing acetogenic bacteria. The effect of butachlor varied on methane-producing bacteria (MPB) at different concentrations. Butachlor at the concentration of 1.0 microg g(-1) dried soil or less than this concentration accelerated the growth of MPB, while at 22.0 microg g(-1) dried soil showed an inhibition. Butachlor enhanced the activity of dehydrogenase at increasing concentrations. The soil dehydrogenase showed the highest activity on the 16th day after application of 22.0 microg g(-1) dried soil of butachlor. The hydrogen peroxidase could be stimulated by butachlor. The soil respiration was depressed during the period from several days to more than 20 days, depending on concentrations of butachlor applied. Both the nitrogen fixation and nitrification were stimulated in the beginning but reduced greatly afterwards in paddy soil.

The effects of metsulfuron-methyl, a sulfonylurea herbicide, on the wheat soil microorganisms were evaluated by the methods of microbial inoculation culture, and the activities of three enzymes were measured using the colorimetric method. The tolerant microorganisms that can resist 500 microg x g(-1) metsulfuron-methyl in the counting culture medium were studied specially. Metsulfuron-methyl distinctly inhibited the common aerobic heterotriphic bacteria, but the effects on common fungi and common actinomycete were not evident. In the meantime, the number of tolerant fungi increased greatly in the rhizosphere after the application of metsulfuron-methyl in contrast to the significant decrease of the amount of tolerant actinomycete. It indicates that fungi might turn into the dominant microbial type and actinomycete is the sensitive factor in the soil polluted by sulfonylurea residues. The population of aromatic compounds-decomposing bacteria, aerobic azotobacter, and nitrite bacteria all increased in the earlier period, but the aerobic azotobacter decreased rapidly in number 30 days later, and the amount of nitrite bacteria also showed a temporary decrease with time 15 days later. However, the denitrifying bacteria just began to increase significantly after the crops had grown for 50 days. The amount of sulfur-oxidizing bacteria gradually decreased with the growth of crops, and so were the sulfate-reducing bacteria after metsulfuron-methyl application. To all types of microorganisms, there were more microbes in rhizosphere samples than those in nonrhizosphere except aerobic azotobacter. It means the growth of wheat root system can stimulate the growth of most microorganisms. The activities of hydrogen peroxidase and polyphenol oxidase in soil samples after metsulfuron-methyl application were notably lower than those in the control, and the difference of the activities between the samples of rhizosphere and nonrhizosphere was evident. On the contrary, the activity of dehydrogenase was not inhibited by the application of metsulfuron-methyl, and the rhizosphere effect was not obvious either.

Increasingly often soil residual concentrations of pharmaceutical antibiotics are detected, while their ecotoxic relevance is scarcely known. Thus, dose related effects of two antibiotics, sulfapyridine and oxytetracycline, on microorganisms of two different topsoils were investigated. The fumigation-extracted microbial C (E(C)) and ergosterol were determined to indicate soil microbial and fungal biomass, respectively. Microbial activity was tested as basal respiration (BR), dehydrogenase activity (DHA), substrate-induced respiration (SIR), and Fe(III) reduction. The BR and DHA were uninfluenced even at antibiotic concentrations of 1000 microg g(-1). This revealed that an activation of microbial growth through nutrient substrate addition is required to test possible effects of the bacteriostatic antibiotics. In addition, the effects of both antibiotics were time dependent, showing that short-term tests were not suitable. Clear dose-response relations were determined with SIR when the short-term incubation of 4h was extended into the growth phase of the microorganisms (24 and 48 h). The Fe(III) reduction test, with a 7-d incubation, was also found to be suitable for toxicity testing of antibiotics in soils. Effective doses inhibiting the microbial activity by 10% (ED(10)) ranged from total antibiotic concentrations of 0.003-7.35 microg g(-1), depending on the antibiotic compound and its soil adsorption. Effective solution concentrations (EC(10)), calculated from distribution coefficients, ranged from 0.2 to 160 ng g(-1). The antibiotics significantly (p<0.05) reduced numbers of soil bacteria, resulting in dose related shifts in the fungal:bacterial ratio, which increased during 14 d, as determined from analysis of ergosterol and E(C). It was concluded that pharmaceutical antibiotics can exert a temporary selective pressure on soil microorganisms even at environmentally relevant concentrations.

A field plot study was conducted on two types of paddy soils in the Taihu Lake Region, during the rice season of year 2000 in order to assess phosphorus (P) losses by runoff and vertical leaching, which are considered the two main pathways of P movement from paddy soil into its surrounding water course. Commercial NPK compound fertilizer and single superphosphate fertilizer were applied to furnish 0, 30, 150, and 300 kg applied P ha m(-2). The experiments consisted of three replicates of each treatment in Changshu site and four replicates in Anzhen site, with a plot size of 5 x 6 m2 in a randomized block. Results revealed that the average concentration range for total P (TP) in runoff was 1.857-7.883, 1.038-5.209, 0.783-1.255 and 0.572-0.691 mg P l(-1) respectively for P300, P150, P30 and P0 in Anzhen, while it was 2.431-2.449, 1.578-1.890, 1.050-1.315 and 0.749-0.941 mg P l(-1) respectively in Changshu. In all treatments, particulate P (PP) represented a major portion of the TP lost in runoff, it was 80% in Anzhen, and it was even more (>90%) in Changshu. Phosphate fertilizer treatments significantly affected P concentrations and P loads in the runoff. The mean concentration and average seasonal TP load from the P150 plots were 1.809 mg P l(-1) and 395 g P ha m(-2) season(-1) respectively, and lower than that from the P300 plots (2.957 mg P l(-1) and 652 g P ha m(-2) season(-1)). These were obviously higher than from the P30 (0.761 mg P l(-1) and 221 g P ha m(-2) season(-1)) and P0 (0.484 mg P l(-1) and 146 g P ha m(-2) season(-1)) respectively. There was no significant difference found between the P30 and the P0 in both sites. Under usual P application rate, there were total 31.7 and 20.6 tones P removed by runoff from permeable (Anzhen site) and waterlogged (Changshu site) paddy soils in the southern Jiangsu region (major part of the TLR) in the rice season of the year 2000. But if the P application rate is unusual high, or the Olsen P in soil accumulates to above a certain level, then this could sharply increase in the future. The average concentration of molybdate reactive phosphorus (MRP) in the vertical leachate from the four different P treatments ranged from 0.058 to 0.304 mg P l(-1) in Anzhen and from 0.048 to 0.394 mg P l(-1) in Changshu. P application rate significantly affected the MRP concentration at each depth in both sites, except for the 90 cm in Anzhen. The average MRP loads during the rice season moved by vertical leaching from the four treatments ranged from 163 to 855 g P ha m(-2) season(-1) in Anzhen and 208-1,825 g P ha m(-2) season(-1) in Changshu. Vertical leachate movement does not necessarily mean that it moves towards surface water and contaminate the watercourses in this flat plain paddy soil region, it does, however, imply that P can move down from surface layers of soil to deeper levels.

Bacterial communities and enzyme activities of PAHs polluted soils
Three soils (i.e. a Belgian soil, B-BT, a German soil, G, and an Italian agricultural soil, I-BT) with different properties and hydrocarbon-pollution history with regard to their potential to degrade phenanthrene were investigated. A chemical and microbiological evaluation of soils was done using measurements of routine chemical properties, bacterial counts and several enzyme activities. The three soils showed different levels of polycyclic aromatic hydrocarbons (PAHs), being their contamination strictly associated to their pollution history. High values of enzyme activities and culturable heterotrophic bacteria were detected in the soil with no or negligible presence of organic pollutants. Genetic diversity of soil samples and enrichment cultures was measured as bands on denaturing gradient gel electrophoresis (DGGE) of amplified 16S rDNA sequences from the soil and enrichment community DNAs. When analysed by Shannon index (H'), the highest genetic biodiversity (H'=2.87) was found in the Belgian soil B-BT with a medium-term exposition to PAHs and the poorest biodiversity (H'=0.85) in the German soil with a long-term exposition to alkanes and PAHs and where absence, or lower levels of enzyme activities were measured. For the Italian agricultural soil I-BT, containing negligible amounts of organic pollutants but the highest Cu content, a Shannon index=2.13 was found. The enrichment of four mixed cultures capable of degrading solid phenanthrene in batch liquid systems was also studied. Phenanthrene degradation rates in batch systems were culture-dependent, and simple (one-slope) and complex (two-slope) kinetic behaviours were observed. The presence of common bands of microbial species in the cultures and in the native soil DNA indicated that those strains could be potential in situ phenanthrene degraders. Consistent with this assumption are the decrease of PAH and phenanthrene contents of Belgian soil B-BT and the isolation of phenanthrene-degrading bacteria. From the fastest phenanthrene-degrading culture C(B-BT), representative strains were identified as Achromobacter xylosoxidans (100%), Methylobacterium sp. (99%), Rhizobium galegae (99%), Rhodococcus aetherovorans (100%), Stenotrophomonas acidaminiphila (100%), Alcaligenes sp. (99%) and Aquamicrobium defluvium (100%). DGGE-profiles of culture C(B-BT) showed bands attributable to Rhodococcus, Achromobacter, Methylobacterium rhizobium, Alcaligenes and Aquamicrobium. The isolation of Rhodococcus aetherovorans and Methylobacterium sp. can be consistent with the hypothesis that different phenanthrene-degrading strategies, cell surface properties, or the presence of xenobiotic-specific membrane carriers could play a role in the uptake/degradation of solid phenanthrene.

We studied the effectiveness of remediation on microbial endpoints, namely microbial biomass and activity, microbial and plant species richness, of an As-contaminated mine spoil, amended with compost (C) alone and in combination with beringite (B) or zerovalent iron grit (Z), to increase organic matter content and reduce trace elements mobility, and to allow Holcus lanatus and Pinus pinaster growth. Untreated spoil showed the lowest microbial biomass and activity and hydrolase activities, and H. lanatus as sole plant species, whereas the presented aided phytostabilisation option, especially CBZ treatment, significantly increased microbial biomass and activity and allowed colonisation by several plant species, comparable to those of an uncontaminated sandy soil. Microbial species richness was only increased in spoils amended with C alone. No clear correlation occurred between trace element mobility and microbial parameters and plant species richness. Our results indicate that the choice of indicators of soil remediation practices is a bottleneck.

Leaching of arsenic (As) from chromated copper arsenate (CCA)-treated wood may elevate soil arsenic levels. Thus, an environmental concern arises regarding accumulation of As in vegetables grown in these soils. In this study, a greenhouse experiment was conducted to evaluate As accumulation by vegetables from the soils adjacent to the CCA-treated utility poles and fences and examine the effects of soil amendments on plant As accumulation. Carrot (Daucus carota L.) and lettuce (Lactuca sativa L.) were grown for ten weeks in the soil with or without compost and phosphate amendments. As expected, elevated As concentrations were observed in the pole soil (43 mg kg(-1)) and in the fence soil (27 mg kg(-1)), resulting in enhanced As accumulation of 44 mg kg(-1) in carrot and 32 mg kg(-1) in lettuce. Addition of phosphate to soils increased As accumulation by 4.56-9.3 times for carrot and 2.45-10.1 for lettuce due to increased soil water-soluble As via replacement of arsenate by phosphate in soil. However, biosolid compost application significantly reduced plant As uptake by 79-86%, relative to the untreated soils. This suppression is possibly because of As adsorbed by biosolid organic mater, which reduced As phytoavailability. Fractionation analysis showed that biosolid decreased As in soil water-soluble, exchangeable, and carbonate fraction by 45%, whereas phosphate increased it up to 2.61 times, compared to the untreated soils. Our results indicate that growing vegetables in soils near CCA-treated wood may pose a risk of As exposure for humans. Compost amendment can reduce such a risk by reducing As accumulation by vegetables and can be an important strategy for remediating CCA-contaminated soils. Caution should be taken for phosphate application since it enhances As accumulation.

In this study, we evaluated the effect of the application by two agrochemicals, methamidophos (O,S-dimethyl phosphoroamidothioate) and urea, on microbial diversity in soil, using the combined approaches of soil microbial biomass analysis and community level physiological profiles (CLPPs). The results showed that both a low and a high level of methamidophos application (CS2 and CS3) and urea application (CS4) significantly decreased microbial biomass C (Cmic) by 41-83% compared with the control (CS1). The soil organic C (Corg) values of CS3 and CS4 were significantly higher and lower by 24% and 14%, respectively, than that of CS1. Similarly to Cmic, the values of Cmic/Corg of the three applied soils which decreased were lower by 31-84% than that of CS1. In contrast, the respiration activity of the three applied soils were significantly higher than the control. Agrochemical application also significantly increased the soil total of N and P (Ntol and Ptol) and decreased the Corg/Ntol and Corg/Ptol values. The CLPPs results showed that the AWCD (average well color development) of the three applied soils were significantly higher than that of CS1 during the incubation period. Substrate richness, Shannon and Simpson indices of microbial communities under chemical stresses, increased significantly. In addition, the CFU (colony-forming unit) numbers of methamidophos metabolized bacteria in CS2 and CS3 also increased significantly by 86.1% and 188.9% compared with that of CS1. The combined results suggest that agrochemicals reduce microbial biomass and enhance functional diversities of soil microbial communities; meanwhile, some species of bacteria may be enriched in soils under methamidophos stress.

The natural microbial activity in the unsaturated soil is vital for protecting groundwater in areas where high loads of biodegradable contaminants are supplied to the surface, which usually is the case for airports using aircraft de-icing fluids (ADF) in the cold season. Horizontal and vertical distributions of microbial abundance were assessed along the western runway of Oslo Airport (Gardermoen, Norway) to monitor the effect of ADF dispersion with special reference to the component with the highest chemical oxygen demand (COD), propylene glycol (PG). Microbial abundance was evaluated by several biondicators: colony-forming units (CFU) of some physiological groups (aerobic and anaerobic heterotrophs and microscopic fungi), most probable numbers (MPN) of PG degraders, selected catabolic enzymatic activities (fluorescein diacetate (FDA) hydrolase, dehydrogenase, and β-glucosidase). High correlations were found between the enzymatic activities and microbial counts in vertical soil profiles. All microbial abundance indicators showed a steep drop in the first meter of soil depth. The vertical distribution of microbial abundance can be correlated by a decreasing exponential function of depth. The horizontal trend of microbial abundance (evaluated as total aerobic CFU, MPN of PG-degraders, and FDA hydrolase activity) assessed in the surface soil at an increasing distance from the runway is correlated negatively with the PG and COD loads, suggesting the relevance of other chemicals in the modulation of microbial growth. The possible role of potassium formate, component of runway de-icers, has been tested in the laboratory by using mixed cultures of Pseudomonas spp., obtained by enrichment with a selective PG medium from soil samples taken at the most contaminated area near the runway. The inhibitory effect of formate on the growth of PG degraders is proven by the reduction of biomass yield on PG in the presence of formate.

Abstract

Laboratory tests were conducted with 14 insecticides applied to a sandy loam to determine whether these materials caused any effects on microbial activities related to soil fertility. The ammonification of soil native organic nitrogen was not inhibited by any of the insecticide treatments. Some insecticides showed an effect on nitrification during the second week of incubation which subsequently recovered to levels similar to those obtained in the controls. There was a significant effect on denitrification in a number of treated samples. However, recovery of denitrifying capacity was rapid. This recovery indicated that the indigenous soil microorganisms can tolerate the chemicals used for control of insect pests. No significant inhibition of sulfur oxidation was observed. Results indicated that the insecticide treatments at the level tested were not drastic enough to be considered deleterious to soil microbial activities important to soil fertility.

Oxalate is exuded by plants in the rhizosphere and plays an important role in the soil/root interactions. Phosphate fertilizer is widely used all over the world and may influence the behavior of arsenic (As) in soils. In this study oxalate and phosphate were used as extractants to investigate their effects on the release of As from three As-contaminated soils and the chemical speciation of As. Concentrations of arsenite (As(III)) and arsenate (As(V)) released progressively increased by increasing the concentrations of oxalate or phosphate. The released As(V) content was higher than that of As(III) and the differences between As(V) and As(III) released by oxalate was more obvious than by phosphate. Greenhouse experiment was conducted to evaluate the effects of oxalate and phosphate on As uptake by wheat (Triticum vulgare L.). Addition of oxalate or phosphate resulted in the increase of As accumulation in both wheat root and shoot and the effect of phosphate was more obvious than that of oxalate.

Effects of Cd and Pb on soil microbial community structure and activities

Background, aim, and scope

Soil contamination with heavy metals occurs as a result of both anthropogenic and natural activities. Heavy metals could have long-term hazardous impacts on the health of soil ecosystems and adverse influences on soil biological processes. Soil enzymatic activities are recognized as sensors towards any natural and anthropogenic disturbance occurring in the soil ecosystem. Similarly, microbial biomass carbon (MBC) is also considered as one of the important soil biological activities frequently influenced by heavy metal contamination. The polymerase chain reaction–denaturing gradient gel electrophoresis (DGGE) has recently been used to investigate changes in soil microbial community composition in response to environmental stresses. Soil microbial community structure and activities are difficult to elucidate using single monitoring approach; therefore, for a better insight and complete depiction of the soil microbial situation, different approaches need to be used. This study was conducted in a greenhouse for a period of 12 weeks to evaluate the changes in indigenous microbial community structure and activities in the soil amended with different application rates of Cd, Pb, and Cd/Pb mix. In a field environment, soil is contaminated with single or mixed heavy metals; so that, in this research, we used the selected metals in both single and mixed forms at different application rates and investigated their toxic effects on microbial community structure and activities, using soil enzyme assays, plate counting, and advanced molecular DGGE technique. Soil microbial activities, including acid phosphatase (ACP), urease (URE), and MBC, and microbial community structure were studied.

In the application of biosolids on agricultural lands, 4-nonylphenol (4-NP) in soils is an important environmental concern because of its associated estrogenic risk to animals and human beings. Incubation experiments that involved the mixing of two contrasting soils (A: calcareous sandy soil; B: acidic clayey soil) and biosolids in 4-NP were performed to examine the effect of 4-NP on the rate of production of CO2, the mineralization of N and the microbial biomass, by considering the biodegradation of 4-NP for the evaluation of soil health. The experimental results indicated that the half-life (t1/2) of 4-NP increased with the supplementary concentration of 4-NP (80, 160 and 240 mg kg(-1)) in the two soils, and the t1/2 values in the soil A are always lower than that in soil B. The 4-NP supplement in the biosolids reverses C mineralization in soil B more than it does in soil A, but it reverses N mineralization in soil A more than in soil B. The aeration status and microbial population of the biosolids treated soils are key factors in determining the time course of 4-NP degradation associated with the microbial activities. The 4-NP was biodegraded mainly by bacteria, and the effect on C and N mineralization of 4-NP input is determined by a balance of the reductions in microbial biomass C (MBC) and N (MBN). After destruction in microbial cell membrane and protein structures by the 4-NP, C and N mineralization, MBC and MBN were subsequently followed by a final decline phase for the later period of incubation.

The use of copper-based fungicides leads to an accumulation of copper (Cu) in vineyard soils, potentially causing adverse effects to the microbial function and fertility of the soil. This study used a soil microcosm approach to assess the effects of Cu accumulation on microbial function in vineyard soils. Surface soil samples were collected from 10 vineyards and a number of un-impacted reference sites in each of three different viticultural regions of Australia. The field-collected soils were transferred to microcosms and maintained for up to 93 days in the laboratory at 20–22 °C and 60 % of their maximum water-holding capacity. The microbial function of the soils was indicated by measuring phosphomonoesterase, arylsulfatase, urease, and phenol oxidase activities. In general, the vineyard soils had greater concentrations of Cu and lower enzyme activities than in the reference soils, although a weak negative relationship between Cu and enzyme activity could only be found for phosphomonoesterase activity. The results show that soil physical–chemical properties (i.e., organic carbon, pH) are greater determinants of soil enzyme activity than increased soil Cu concentration at the Cu concentrations present in vineyard soils.

Effects of trifluralin on soil microbial populations and the nitrogen fixation activity of nitrogen-fixing bacteria Azotobacter chroococcum and Bradyrhizobium japonicum and the decomposition of trifluralin by soil microorganisms were studied. Trifluralin at lower concentrations from 0.5 mg microg(-1) dry soil to lower than 10.0 mg microg(-1) dry soil appeared to stimulate the growth of soil bacteria, actinomycetes, mould, and the pure cultures of Br. japonicum and A. chroococcum. Not only the colony amounts of these two species of nitrogen-fixing bacteria increased, grown on agar medium containing lower concentrations of trifluralin, but also these colonies also enlarged in size and appeared obviously in shorter formation time. However, trifluralin at higher concentrations would inhibit the development of microbial colonies both in amount and size. Trifluralin inhibited the activity of acetylene reduction of A. chroococcum when it was added at the same time of inoculation with A. chroococcum, but it showed a noteworthy stimulation to nitrogen fixation of A.chroococcum when it was put into culture after the cells of the nitrogen-fixing bacterium had grown well. The observation that soil microorganisms could use trifluralin as sole carbon and nitrogen resources for their growth, indicated that microorganisms could decompose trifluralin well.
