对福州产的原尾蜥虎(Hemidactylusbowringii) 雄性生殖周期的研究结果表明:10 月份精子发生开始新的周期性复生,精原细胞发育进入增殖期,中旬曲细精管除精原细胞外尚有较多的初级精母细胞;11 月初已有较多的精细胞,曲细精管管腔中开始出现少量的精子;12 月份,生精活动非常活跃,输精管始见有成熟的精子,12 月下旬进入冬眠期,成熟的精子释放到输精管中过冬,冬眠期间生精活动趋于停滞,表现为曲细精管管径和生精上皮厚度都有所下降;次年3 月底4 月初陆续出蛰,生精活动又恢复并逐渐增强,4、5 月份精子发生相当活跃,6 月份是精子发生的高峰期,也是交配繁殖的高峰期;原尾蜥虎一年只繁殖一次,繁殖期过后,7 ~9 月份,生精活动逐渐减弱,曲细精管的管腔逐渐消失,初级、次级精母细胞、精细胞逐渐减少、退化,精子逐渐退化、消失,到9 月下旬曲细精管和输精管都见不到精子,此时的曲细精管主要由精原细胞组成,一个周期到此结束.  相似文献

对栉孔扇贝Chlamysfarreri 的卵原细胞和卵母细胞发育过程的超微结构和细胞化学研究结果表明:卵原细胞的胞质中含大量的游离核糖体,细胞器不发达,蛋白质和RNA丰富而糖元贫乏;卵黄生成之前,卵母细胞中的核质膨大为生发泡;核仁发达,胞质中线粒体大为增多;卵黄体和皮层颗粒首先出现于核周,接近线粒体聚集处;卵黄生成盛期,卵母细胞的胞质内具平行片层粗面内质网和同心圆状粗面内质网,它们是生成卵黄体和皮层颗粒的主要细胞器,线粒体和高尔基器也参与了这两种内含物的生成;微胞饮作用也为细胞的发育提供某些物质;卵黄体含蛋白质和糖元,皮层颗粒富含多糖类;卵黄膜形成始于卵黄生成初期,先是细胞游离端的胞质出现一个富含多糖的突起,突起部的质膜微绒毛化,微绒毛分泌产生由中性多糖构成的卵黄膜,继而向四周扩展,并于卵母细胞成熟时覆盖整个细胞;RNA 合成贯穿于卵母细胞的整个发育期,延续至卵黄生成结束之后,成熟分裂临近之时.  相似文献

对龟足(Pollicipes metella)精子发生和成熟精子的超微结构进行了研究.精子发生属于变态型.细胞由多边形变为椭圆形到长梨形直至弹开成线形;胞核由椭圆形逐渐变长弯曲成U形,最后变为细长形;中期精子细胞出现的椭圆形附属小滴也弯曲成U形,最终成长柱状.早期、中期、后期精子细胞出现核周微管及核囊,核周微管及核囊在成熟精子中消失.精子为前中部带有一个长柱状附属小滴的线状结构(长约36μm),可分为顶体、颈部、中段和尾部四部分.顶体为锥状(约0.6μm),顶体后的基体发出轴丝,轴丝纵贯末端.核细长线状(约16μm).附属小滴长柱状(约8μm),内有棒状致密斑.核、附属小滴与轴丝并行.鞭毛为典型的“9 2”结构。  相似文献

运用透射电镜研究中国淡水蛏Novaculina chinensis卵子发生过程中细胞和细胞器的超微结构变化,结果表明,卵原细胞的胞质含有丰富的核糖体及少量线粒体和内质网,其它细胞器不发达,卵黄合成前期,卵母细胞胞体增大,线粒体较多,量小泡装和条索状的内质网;同时随着发育的进行,出现卵黄膜、合成少量卵黄粒及产生大量脂滴,卵黄全成期卵母细胞胞体迅速增大,细胞核伸出伪足状突起;胞 内出现丰富的线粒体、溶酶体及结构独特的内质网、卵黄粒越来越多,而脂滴越来越少,成熟期卵母细胞胞体达到最大,细胞器不发达,胞质中充满大量卵黄粒,仅少量脂滴。首次观察到卵母细胞表面伸出伪足状突起,这也是外源卵黄发生的证据。讨论了中国淡水蛏卵子发生的特点.  相似文献

除草剂乙草胺对非洲爪蟾性腺发育的影响   总被引:4,自引:2,他引:4  
使用性腺发育对内分泌干扰作用敏感的两栖动物非洲爪蟾作为模型动物,通过考察性别比、性腺整体形态和组织学形态来揭示乙草胺对其性腺发育的影响,从而确定乙草胺是否具有内分泌干扰活性.46/47期蝌蚪经不同浓度(5μg·L-1、10μg·L-1、20μg·L-1)的乙草胺暴露处理至变态后1个月,在变态结束后3个月时,解剖性腺、鉴别雌雄并固定性腺做组织切片.实验中对照组所有性腺都为典型的卵巢或睾丸,雌性百分率为48.78%(20/41).而乙草胺暴露组却出现了明显的异常睾丸,其总体上是睾丸,但又带有部分卵巢的特征,在统计性别时将具有异常睾丸的爪蟾归为雄性.5μg·L-1、10μg·L-1、20μg·L-1暴露组的雌性百分率分别为:57.14%(24/42)、66.67%(22/36)、55.26%(21/38).虽经统计分析无显著性差异,但乙草胺暴露组中雌性多于雄性的趋势和出现带有雌性特征的雄性的现象,一定程度上却暗示了乙草胺可能对非洲爪蟾的性腺发育有雌性化作用.正常睾丸的组织学结构应有发育成熟的生精小管,其内有各个时期的精子囊和生精细胞及精子.但经乙草胺处理的爪蟾睾丸却表现出几种明显的异常:没有生精小管,相反却像发育早期的睾丸一样,充满大量的精原细胞,完全没有生精细胞和精子;有生精小管结构,但其内精子囊结构不明显,生精细胞少并且排列混乱,其间有很大的空隙;睾丸出现腔隙,并有大量体细胞存在,这种结构与57期的卵巢相似;睾丸中出现异常的生精细胞,与58期卵巢的卵细胞相似;睾丸组织中有明显的卵细胞散布.组织学研究发现的乙草胺导致的睾丸结构证明了乙草胺对非洲爪蟾的睾丸发育有抑制雄性化作用和雌性化作用,即乙草胺对非洲爪蟾的性腺发育有内分泌干扰作用.  相似文献

扫描电镜观察表明,渔游蛇(Natrix Piscater)精子总长度约110μm,分为头和尾两部分;头部呈前稍尖的细长圆柱形,略变曲,约长8.5μm,直径以经约0.6μm。尾部又分4段:颈段、中段、主段和末段;其与从不同之处是中段较长,约73μm,占精子总长度的2/3多;线料体型鞘由近500圈超过5000个线粒体构成;透射电镜揭示了头部顶体体覆疬于核前约1/2;尾部颈段的结构较复杂,包括连接片的踝  相似文献

越来越多的研究表明,环境污染是多种生物生殖力下降的主要原因之一,而精子作为雄性生物的生殖细胞,因高度分化丧失了自我修复能力,极易受环境因素的胁迫。但环境污染物致生物体精子损伤机制的研究目前尚缺乏完整、系统的体系。本文在总结近年来国内外相关研究的基础上,从环境污染物诱导精子产生氧化应激、改变生精细胞的细胞周期、诱导正常生精细胞发生细胞凋亡和扰乱生物体雄激素水平4个方面综合探讨了环境污染物损害精子形态结构、功能以及降低精子数量的潜在作用模式,为开发精子损伤的早期特异性分子标记物奠定了理论基础,同时也为污染物致精子损伤的机制研究提供新的思路。  相似文献

应用透射电镜观察比较雨生红球藻不同形态细胞的超微结构,结果表明,游动细胞除具有典型的绿藻门团藻目的特征性结构外,细胞壁与原生质体分离形成周质空间,细胞具有发达的可进一步发育的网状叶绿体,不动细胞除了无鞭毛,具厚壁外,它与游动细胞的主要不同是:细胞内的大部分区域被色素沉积物占据,叶绿体等细胞器被包裹,光合片层紧贴,早期的不动细胞还具有较大的线粒体,根据形态及超微结构上的差异,对红球藻游动细胞与不动细胞间的生理差异进行了解释。  相似文献

为探讨有机磷农药混合物对生物生殖毒性的影响,以模式动物Sprague- Dawley (SD)大鼠为实验材料,采用亚慢性染毒动物实验,利用3×3析因实验设计方法研究了常见有机磷农药——氧化乐果和毒死蜱的联合雄性生殖毒性效应及机理。结果表明:氧化乐果和毒死蜱联合染毒60 d后,附睾系数增加,精子数量和精子活力降低,联合作用呈现协同效应(p<0.05);混配农药能降低睾丸标志酶乳酸脱氢酶(LDH)、酸性磷酸酶(ACP)、碱性磷酸酶(AKP)、γ-谷氨酰转移酶(γ-GT)的活力,对LDH、ACP、γ-GT的联合作用表现为协同作用(p<0.05);混配农药能增加睾丸中促卵泡生成激素(FSH)含量,降低睾酮(T)含量,联合作用表现为协同作用(p<0.05)。通过组织病理学观察,氧化乐果和毒死蜱混合物能引起曲细精管的萎缩、畸形精子增多、生精细胞坏死、胞浆溶解和线粒体空泡化;支持细胞数量减少,出现坏死,精子从支持细胞上脱落,基底膜脱落等损伤。氧化乐果和毒死蜱对大鼠的联合生殖毒性为协同效应,混合后有明显的增毒作用;对睾丸标志酶(LDH、ACP、γ-GT)活力及睾丸性激素水平(FSH、T)的协同影响,是产生联合生殖毒性的机理之一。以上结果提示,对氧化乐果和毒死蜱进行环境健康风险评价时应考虑这种协同作用。  相似文献

Ultrastructural changes occurring during the course of development in oocytes of Mytilus edulis are described for mussels collected at monthly intervals over a period of one year (September 1981 to October 1982) from a site in Cornwall, England. During early stages of oogenesis the oocyte is surrounded by a small number of follicle cells but, as development proceeds, the follicle cells are restricted to the stalk region which attaches the oocyte to the acinar wall. Contact between the follicle cells and the developing oocyte is maintained by means of desmosomelike gap junctions. Organelles and inclusion bodies present in the ooplasm during oogenesis include rough endoplasmic reticulum (RER), Golgi bodies, mitochondria, free ribosomes, Balbiani's vitelline body, annulate lamellae and yolk and cortical granules. The RER, in particular, varies considerably throughout the course of development. Evidence for uptake of exogenous macromolecules into oocytes by pinocytosis is presented; it occurs in the basal region of previtellogenic oocytes prior to the formation of the vittelline coat. Lipid-yolk granules invariably have mitochondria in close association and, during the winter months, develop in close proximity to small, apparently glycogen-rich vesicles possibly suggesting that conversion of glycogen to lipid takes place in developing oocytes. Oocyte degeneration was commonly observed and involves initial breakdown of the plasma membrane followed by rupture of the vitelline coat. The oocyte contents once released into the acinar lumen are resorbed by the epithelial cells of the gonoducts, which are prevalent throughout the mantle of ripe individuals.  相似文献

In the present study, specimens of catfish (Clariidae) were collected from a polluted location (Main Basin) and a relatively clean area (East Basin) in Lake Mariut, one of the Nile Delta Lakes in Egypt. Fifteen fish were taken from each site. Liver preparations of fish from the two sources were comparatively examined for cellular changes using transmission electron microscopy. Fish hepatocytes from the polluted area showed accumulation of the heterochromatin, enlarged nucleoli, and an extremely folded nuclear envelope. Perichromatin granules were increased and progressively formed small clusters closely associated with patches of heterochromatin. In the cytoplasm, fractionation, dilation, and vesiculation of rough endoplasmic reticulum (RER), and elevated amounts of smooth endoplasmic reticulum (SER) tubules were noted. The most frequent pathological modifications were the swelling of mitochondria, cristae regression and changes in the electron-transparency of the matrix. Lysosomes showing myelin-like stacks of membraneous material (phospholipidosis), glycogenosomes (i.e., glycogen rosettes enclosed by membranes) and cytoplasmic myelinated bodies were strongly developed. Furthermore, increasing numbers of secondary lysosomes with degraded cell organelles were found. With reference to the storage vesicles, there appeared to be an increase in the lipid droplets (lipidosis) within many hepatocytes. This study reinforces the need to select representative sentinel species from different habitats for biomonitoring purposes and it provides further support for the use of biomarkers in assessing the health of aquatic ecosystems.  相似文献

The ultrastructural morphology of storage-cell breakdown and replenishment in Mytilus edulis is described for mussels collected at monthly intervals over a period of one year (September 1981 to October 1982) from a site in Cornwall, England. In addition cytochemical staining reactions at light and electron microscope levels are described for the storage cells and the Sertoli cells. The breakdown of adipogranular (ADG) cell components is believed to be a lysosomally-mediated process of controlled autolysis involving fusion of primary lysosomes with the protein granules. Increasing activity for lysosomal enzymes during ADG cell breakdown is demonstrated using quantitative cytochemistry. The presence of glycogen in vesicular connective tissue (VCT) cells is demonstrated using freeze-dried, formaldehyde-vapour-fixed tissue. The breakdown of VCT cells involves sequestration of glycogen from the central reserve into small cytoplasmic vesicles; subsequent release appears to be mediated by eccrine secretion. The presence of arylsulphatase activity in lysosomes within the peripheral cytoplasm of the VCT cells during the winter months indicates that glycogen breakdown may also be lysosomally mediated. The morphology of the Sertoli cells is described; during the early stages of spermatogenesis they are characterized by the presence of numerous lipid droplets which are depleted as development proceeds. Contact between Sertoli cells and developing spermatogonia is maintained by means of long septate junctions. Following spawning, the well developed lysosomal system is evident in the Sertoli cells and appears to be engaged in intracellular digestion of phagocytosed waste sperm and residual cytoplasm. Resorption of the products of gamete degeneration by the gonoduct epithelial cells is described. During regeneration, VCT cells appear to endocytose material; the ADG cells demonstrate extensive arrays of rough endoplasmic reticulum and are occasionally seen undergoing mitosis. A process involving loss of glycogen vesicles from the mantle epithelial cells by means of apocrine sections is described.  相似文献

Liver alterations were stereologically determined in five groups, each of 10 male and female Sprague–Dawley weanling rats fed PCB 105 (2,3,3′,4,4′-pentachlorobiphenyl) for 13 weeks at concentrations of 0.05, 0.5, 5, and 50?ppm mixed with corn oil. Animals from two groups receiving the oil in diets served as controls for each gender. A significant increase in mean volume fraction (VF) of smooth endoplasmic reticulum (SER) profiles was estimated in hepatocytes of female rats in 0.5, 5, and 50?ppm groups. A significant decrease in VF of mitochondria in female rats at 50?ppm was detected. In addition, a decrease in VF of lipid droplets was measured in female rats at 0.5, 5, and 50?ppm groups. No difference was found in VF of rough endoplasmic reticulum (RER) cisternae, and peroxisome profiles at any concentration of the PCB in either male or female animals as compared to baseline values from controls. A significant elevation of baseline VF for RER in female rats was detected in comparison with males. In the present study, no observable adverse effect level (NOAEL) for PCB 105 was estimated at <0.5?ppm for hepatocyte alterations in female Sprague–Dawley rats.  相似文献

The cytoplasm of Crassostrea angulata (collected from the Atlantic coast of Northern Spain, Galiza, between 1992 and 1993) spermatogonia contains a few proacrosomal dense vesicles and intermitochondrial round granulo-fibrillar dense bodies. In primary spermatocytes, large cytoplasmic spaces appear occupied by large round and more scattered granulo-fibrillar dense bodies which are intermingled with cord-like dense structures and are in close association with mitochondria and the rough endoplasmic reticulum. As a result of Golgian activity, the number of dense proacrosomal vesicles increases and a flagellum arises from the centrioles. During the second meiotic division, several round dense bodies surrounding cord-like dense structures as well as dense proacrosomal vesicles, were observed between chromosomes. During spermiogenesis, no Golgian activity was noticed, and only the rough endoplasmic reticulum was observed in close association with proacrosomal vesicles. Similar results were observed for Ostrea edulis (collected from the Atlantic coast of southern Portugal, Algarve, between 1992 and 1993). Spermatozoa of both species are of the aquasperm type, and although their ultrastructure is quite similar, silver and phosphotungstic acid-chromic acid stainings enabled us to clearly distinguish both species at the ultrastructural level. The different morphological aspects of spermatogenesis in the Pteriomorphia and the use of silver and PTA stainings in the Mollusca are reviewed and discussed.  相似文献

The present study was conducted to establish the relationship between selected oxidative stress parameters and ultrastructural responses in liver tissue of Labeo rohita fingerlings exposed to cypermethrin. Fish were exposed to lethal (4.0 μg L?1) and sublethal (0.4 μg L?1) concentrations of cypermethrin for a period of 24, 48, 72 and 96 h for acute studies and 1, 5, 10 and 15 days for subacute studies, respectively. Results showed increased catalase (CAT) and protease activity, hydrogen peroxide (H2O2), malondialdehyde (MDA), protein carbonyls and free amino acid (FAA) levels at both concentrations. This suggests participation of free-radical-induced oxidative cell injury in mediating the hepatotoxicity of cypermethrin. In corroboration of this, ultrastructural lesions witnessed a reduction in the number of cell organelles, swollen, vacuolated and condensed mitochondria, dilated rough endoplasmic reticulum, and reduced numbers of smooth enodplasmic reticulum, peroxisomes and lysosomes at the lethal (4.0 μg L?1) concentration. At the sublethal (0.4 μg L?1) concentration, cytoplasmic vacuolation, condensed, vacuolated and swollen mitochondria, dilated rough endoplasmic reticulum and an absence of hepatocyte microvilli were prominent. Ultrastructural changes were exhibited as subcellular responses due to the imbalance in cellular oxidative status by means of oxidative damage.  相似文献

In March, 1983 changes in epidermal ultrastructure were examined in Clupea harengus L. larvae hatched from eggs incubated in four zinc concentrations (0.5 2.0, 6.0 and 12.0 ppm). In addition to the outer and inner epidermal cell types described previously, a third type of cell is present. Ultrastructurally this resembles the epidermal chloride cells of Sardinops caerulea Girard, characterised by numerous mitochondria, extensive smooth endoplasmic reticulum and a free surface exposed to the environment. These cells occur on the yolk sac, head and in the regions of the trunk just above the yolk sac. In larvae treated in 12.0 ppm zinc, the cells contain fewer mitochondria with fewer cristae than those in the controls. Smooth endoplasmic reticulum is much reduced forming nodular masses and contains granular osmiophilic inclusions. In larvae hatching from eggs previously incubated in 6.0 and 12.0 ppm zinc, the epidermal cells contain more vesicles, intracellular spaces, and swollen mitochondria and show signs of necrosis. The viability of these larvae is discussed.  相似文献

Intramuscular administration of two doses: 0.50 LD50 (14.70 mg/kg b w) and 0.75 LD50 (22.30 mg/kg b w) of heptachlor in Rattus norvegicus for 14 days induced significant hypocalcemia without altering serum inorganic phosphate value. Parathyroid chief cells of the experimental rats exhibited degranulation, vacuolation, loss of secretory granules and lipid droplets, reduction in chromatin, and degenerative changes in endoplasmic reticulum and cristae of the mitochondria. Not much of histological and ultrastructural changes could be seen in C cells of the heptachlor treated rats.  相似文献
