INTENTION, GOAL, SCOPE, BACKGROUND: Cyanobacteria have the natural ability to degrade moderate amounts of organic pollutants. However, when pollutant concentration exceeds the level of tolerance, bleaching of the cells and death occur within 24 hours. Under stress conditions, cyanobacterial response includes the short-term adaptation of the photosynthetic apparatus to light quality, named state transitions. Moreover, prolonged stresses produce changes in the functional organization of phycobilisomes and in the core-complexes of both photosystems, which can result in large changes in the PS II fluorescence yield. The localization of ferredoxin-NADP+ reductase (FNR) at the ends of some peripheral rods of the cyanobacterial phycobilisomes, makes this protein a useful marker to check phycobilisome integrity. OBJECTIVE: The goal of this work is to improve the knowledge of the mechanism of action of a very potent pesticide, lindane (gamma-hexaclorociclohexane), in the cyanobacterium Anabaena sp., which can be considered a potential candidate for bioremediation of pesticides. We have studied the effect of lindane on the photosynthetic apparatus of Anabaena using fluorescence induction studies. As ferredoxin-NADP+ reductase plays a key role in the response to oxidative stress in several systems, changes in synthesis, degradation and activity of FNR were analyzed. Immunolocalization of this enzyme was used as a marker of phycobilisome integrity. The knowledge of the changes caused by lindane in the photosynthetic apparatus is essential for rational further design of genetically-modified cyanobacteria with improved biorremediation abilities. METHODS: Polyphasic chlorophyll a fluorescence rise measurements (OJIP) have been used to evaluate the vitality and stress adaptation of the nitrogen-fixing cyanobacterium Anabaena PCC 7119 in the presence of increasing concentrations of lindane. Effects of the pesticide on the ultrastructure have been investigated by electron microscopy, and FNR has been used as a marker of phycobilisome integrity. RESULTS AND DISCUSSION: Cultures of Anabaena sp. treated with moderate amounts of lindane showed a decrease in growth rate followed by a recovery after 72 hours of pesticide treatment. Concentrations of lindane below 5 ppm increased the photosynthetic performance and activity of the cells. Higher amounts of pesticide caused a decrease in these activities which seems to be due to a non-competitive inhibition of PS II. Active PS II units are converted into non-QA reducing, so called heat sink centers. Specific activity and amount of FNR in lindane-treated cells were similar to the values measured in control cultures. Release of FNR from the thylakoid after 48 hours of exposure to 5 ppm of lindane towards the cytoplasm was detected by immunogold labeling and electron microscopy. CONCLUSIONS: From these results, we conclude that the photosynthetic performance and activity of the cells are slightly increased in the presence of lindane up to 5 ppm. Moreover, in those conditions, lindane did not produce significant changes in the synthesis, degradation or activity of FNR. The high capability of Anabaena to tolerate lindane makes this cyanobacterium a good candidate for phytoremediation of polluted areas. RECOMMENDATION AND OUTLOOK: The results of this study show that cultures of Anabaena PCC 7119 tolerate lindane up to 5 ppm, without significant changes in the photosynthetic vitality index of the cells. However, a slight increase in phycobiliprotein synthesis is observed, which is related to total protein content. This change might be due to degradation of proteins less stable than phycobiliproteins. An identification of the proteins with altered expression pattern in the presence of the pesticide remains the subject of further work and will provide valuable information for the preparation of strains which are highly tolerant to lindane.  相似文献

The immunosuppressive effect of lindane was assessed in weanling rats over a period of 5 weeks. Agglutinin titres against S. typhi 'O' and 'H', S. paratyphi 'AH' and 'BR' antigens were determined following TAB vaccination in control and lindane-fed animals. The titres attained in non-treated controls were higher significantly than those in treated animals indicating a suppressive effect of lindane on humoral immune responses.  相似文献

The leachability of lindane from different biosolid amended soils was determined and compared to its bioavailability. Sand, soil, and a mixture of soil-sand (1:1 w/w) were spiked with lindane, blended with different amounts of biosolids, and subjected to a leaching process with water that lasted for 1-28 d. This procedure is in accordance with ISO/TS 21268-1: 2007. After these batch tests, lindane was extracted from the leachates using three different solvent-free microextraction techniques, including solid phase microextraction (SPME), stir-bar sorptive extraction (SBSE), and silicone rod extraction (SRE). The amount of lindane was determined with thermal desorption and gas chromatography coupled to mass spectrometry (GC-MS). The efficiencies of the three microextraction techniques were statistically different, and the efficiency could be related to the amount of polydimethylsiloxane (PDMS) in each extraction device. However, all of the techniques provide data that shows that the leachability of lindane is dependent on the amount of organic matter contained in the matrix. The results of the lindane leachability assay were compared to the bioavailability of lindane, which was determined by measuring the amount of lindane that accumulated in the roots of wheat plants grown in similar soil-biosolid systems. It was confirmed that the amount of organic matter in the matrix is a determining factor for lindane immobilization. The presence of biosolids decreases the mobility of lindane in all of the systems under study. Similarly, increasing biosolid concentrations in the soil significantly decreased the bioavailability of lindane and, consequently, plant absorption. The good correlation (R2 = 0.997) between the leachability of lindane from the matrix and plant absorption of lindane indicates that the proposed biomimetic methodology can predict the bioavailability of lindane in a time period as short as 7 d.The results of this work confirm that amending contaminated soils with biosolids is beneficial for immobilizing lindane and helps prevent the percolation of lindane through the soil profile and into groundwater.  相似文献

Effects of chronic application of a mixture of the herbicide atrazine and the insecticide lindane were studied in indoor freshwater plankton-dominated microcosms. The macroinvertebrate community was seriously affected at all but the lowest treatment levels, the zooplankton community at the three highest treatment levels, with crustaceans, caddisflies and dipterans being the most sensitive groups. Increased abundance of the phytoplankton taxa Cyclotella sp. was found at the highest treatment level. Threshold levels for lindane, both at population and community level, corresponded well with those reported in the literature. Atrazine produced fewer effects than expected, probably due to decreased grazer stress on the algae as a result of the lindane application. The safety factors set by the Uniform Principles for individual compounds were also found to ensure protection against chronic exposure to a mixture of a herbicide and insecticide at community level, though not always at the population level.  相似文献

Rice is mostly cultivated on soil held under flooded conditions during the growing season, where the plow layer differentiates into a thin oxidized surface layer and an underlying reduced layer. Under these conditions, certain pesticides undergo reductive transformations which are characteristic to rice fields and other anaerobic systems. Thus, lindane and pentachlorophenol are reductively dechlorinated into less toxic products, whereas, in highly reduced soils, thiobencarb is dechlorinated into a substance phytotoxic to rice plants. Nitrophenyl compounds, such as parathion and chlornitrofen, are converted into respective amino analogs, which are apt to remain as soil-bound residues. Transformation processes of these and several other environmentally important pesticides used in rice fields in the past and/or at present are reviewed.  相似文献

Displacement of lindane presorbed on the pristine and OH-functionalized multiwalled carbon nanotubes (MWCNTs) by phenanthrene, naphthalene, and atrazine, and competition of these compounds with lindane on the aforementioned sorbents were investigated. Displacement of lindane presorbed on MWCNTs by atrazine, naphthalene, and phenanthrene, and competitive sorption effect of these chemicals with lindane on MWCNTs followed the same order: atrazine > naphthalene > phenanthrene. The lowest competition and displacement of lindane by phenanthrene were mainly because of the strong interactions between these two chemicals, whereas interaction of lindane with atrazine and naphthalene was quite low. The more pronounced displacement of lindane by atrazine than naphthalene and higher competitive sorption of lindane with atrazine than with naphthalene can be ascribed to the larger molecular volume of atrazine; thus, the steric hindrance effect is higher relative to naphthalene. This study is valuable for evaluating influence of the coexisting organic compounds on sorption of primary solute towards MWCNTs in the environment.  相似文献

Impacts of diazinon (O,O-diethyl O-2-isopropyl-6-methylpyrimidin-4-yl phosphorothioate), imidacloprid [1-(6-chloro-3-pyridylmethyl)-N-nitroimidazolidin-2-ylideneamine] and lindane (1,2,3,4,5.6-hexachlorocyclohexane) treatments on ammonium, nitrate, and nitrite nitrogen and nitrate reductase enzyme activities were determined in groundnut (Arachis hypogaea L.) field for three consecutive years (1997 to 1999). Diazinon was applied for both seed- and soil-treatments but imidacloprid and lindane were used for seed treatments only at recommended rates. Diazinon residues persisted for 60 days in both the cases. Average half-lives (t1/2) of diazinon were found 29.3 and 34.8 days respectively in seed and soil treatments. In diazinon seed treatment, NH4(+), NO3(-), and NO2(-) nitrogen and nitrate reductase activity were not affected. Whereas, diazinon soil treatment indicated significant increase in NH4(+)-N in a 1-day sample, which continued until 90 days. Some declines in NO3(-)N were found from 15 to 60 days. Along with this decline, significant increases in NO2(-)N and nitrate reductase activity were found between 1 and 30 days. Imidacloprid and lindane persisted for 90 and 120 days with average half-lives (t1/2) of 40.9 and 53.3 days, respectively. Within 90 days, imidacloprid residues lost by 73.17% to 82.49% while such losses for lindane residues were found 78.19% to 79.86 % within 120 days. In imidacloprid seed-treated field, stimulation of NO3(-)N and the decline in NH4+NO2(-)-N and nitrate reductase enzyme activity were observed between 15 to 90 days. However, lindane seed treatment indicated significant increases in NH4(+)-N, NO2(-)-N and nitrate reductase activity and some adverse effects on NO3(-)N between 15 and 90 days.  相似文献

The dynamics of the disappearance of lindane, HCH isomers and HCB in soil after lindane application were studied, as well as the phenomenon of lindane bioisomerization to HCH isomers. The disappearance of the compounds studied depended on their volatilization into the atmosphere, plant absorption and degradation. During the experiment, lindane was bioisomerized in very small amounts to alpha-, beta-, delta-HCH and HCB, but not to epsilon-HCH. The limited magnitude of this phenomenon indicates that bioisomerization does not contribute to the contamination of food and the environment with HCH isomers that has been observed.  相似文献

The toxic effects of lindane on the zooplankton communities may be strongly related to the population fitness, which is highly dependent on food availability. In order to test this hypothesis, acute (immobilisation) and chronic (life-history) responses of Daphnia longispina and Daphnia magna, reared at different food levels (low, normal, and high), were assessed in laboratorial exposures to several concentrations of lindane. A bifactorial design was employed (food level versus lindane concentration) for both species. Results showed that lindane was toxic to both D. magna and D. longispina, within a similar range. However, lindane toxicity to daphnids was dependent on food level, suggesting that the latter is an important factor to take into account when assessing the toxic effects of lindane on zooplankton communities.  相似文献

Spatial distribution of lindane in topsoil of Northern France   总被引:1,自引:0,他引:1
Lindane is a persistent organochlorine insecticide and the use of this insecticide in agriculture was banned in France in 1998. In this study we investigated the concentrations of lindane in topsoil in Northern France and used robust geostatistics to map the geographical distribution of lindane. The study was based on a 16 km × 16 km grid covering an area of ca 25 000 km2. Lindane was found in all soils, even those from non-agricultural-application areas. Very low ratios of α-/γ-HCH and δ-/γ-HCH suggested that a long time had passed since technical HCH was used in the studied area, or that emission sources of lindane were still present. A strong gradient in lindane concentration was observed, with the highest lindane concentrations in an area located in the northern region. Results suggested that some of the lindane observed in the high concentration area may have come from volatilization of old lindane applied to intensively cultivated areas, which was then transported by prevailing winds coming from the south-west and deposited in a densely inhabited depression.  相似文献

Mussels (Mytilus galloprovincialis), collected from the Bizerta lagoon, were acclimated for four days to various conditions of temperature, salinity, photoperiod and food supply and then exposed to lindane at a concentration of 40 microg l(-1). Catalase activity, which is a biomarker of exposure to an oxidative stress, was measured in the whole soft tissues of control and assay groups. In control mussels, high temperature, high salinity and light duration significantly increased catalase activity whereas this activity decreased when food, composed of freeze-dried, algae was available. When mussels were treated with lindane, catalase activities were higher than in controls. This increase was significant with temperature, salinity and light duration. The food supply did not change catalase activity, which was always higher compared to controls. Oxidative stress was shown in mussels exposed to lindane. The results highlight the need of considering abiotic parameters in biomonitoring studies, and especially when using catalase as a biomarker.  相似文献

Chironomus riparius Meigen were exposed to 0, 0.01, 0.1, 0.5, 0.75 and 1.0 ppm lindane for 48 h as fourth instar larvae. Exposure had no effect on glutathione-S-transferase (GST) activity in larvae snap-frozen immediately following exposure. In contrast, exposure had longer-term consequences affecting developmental parameters. Concentrations above 0.5 ppm lindane affected larval behaviour, reduced adult body size and fecundity and delayed emergence times. The lack of significant change in GST activity when life history characters were affected by high concentrations of lindane, suggests that in C. riparius, GST is not a sensitive biomarker of pesticide exposure or effect.  相似文献

In vitro investigations of the influence of lindane and its metabolites were performed on microsomal and mitochondrial ATPases from liver, kidney and brain of rat and mouse. The microsomal Na+-K+-ATPases in rat liver were inhibited by the tested substances. An increase of activity was observed only with 2.5 X 10(-5) M gamma-HCH. Effects on the microsomal Na+-K+-ATPase from kidney and brain of rat were also indicated. The mitochondrial enzyme in rat liver was stimulated by all the compounds tested at concentrations of 10(-4) M - 10(-2) M. The effects on mitochondrial enzymes from kidney and brain varied in dependence on the tested substances. In the microsomes and mitochondria of mouse an influence on the Na+-K+-ATPases similar to the effects on the preparations from organs of rat was evident.  相似文献

Effects of organochlorine xenobiotics on human spermatozoa.   总被引:4,自引:0,他引:4
The organochlorine insecticide lindane is a widely distributed environmental pollutant belonging to the growing family of endocrine disrupting chemicals (EDCs). Lindane intercalates into the sperm membrane and alters the molecular dynamics of the bilayer. In the present paper, preliminary data are reported showing that doses of lindane as low as those found in the female genital tract secretions inhibit the sperm cytological responsiveness to progesterone, the physiological agonist which stimulates the onset of acrosome reactions at the site of fertilization. The hypothesis is put forth that even background levels of lindane may exert antifertility effects independently on the health status of either the male and female reproductive organs.  相似文献

We assessed the threshold concentrations for toxic effects by lindane on a zooplankton community using mesocosms in a natural pond. Enclosures (1000 litres) were treated to give mean concentrations of 0, 2, 6, 12, 18, 24, and 50 microg litre(-1) over a test period of 39 days. Larvae of the phantom midge Chaoborus flavicans showed a high susceptibility to lindane and were virtually eliminated from enclosures with > or =12 microg litre(-1) 4 days after application. Populations of cyclopoid nauplii (Eucyclops serrulatus) were reduced at a lindane concentration of 2 microg litre(-1) and showed high mortality after 11 days exposure to > or =6 g litre(-1). No lindane-induced effects could be seen in the cladoceran species Daphnia longispina and Chydorus sphaericus. Rotifer species were probably not directly affected by lindane; however loss of the principal predator Chaoborus was accompanied by an increase in the numbers of Asplanchna priodonta. A simultaneous decrease in Keratella quadrata possibly reflected predation since its decline showed no dose-response relationship with lindane and it is among the major prey of Asplanchna. Such complex secondary effects, occurring through trophic interactions between a predator and two co-occurring prey, would not have been predicted from single-species toxicity tests. These, and effects on cyclopoid populations that occurred predominantly through the mortality of nauplii, demonstrate the value of toxicological studies on natural communities. We conclude that a combination of laboratory and field investigations, coupled with mono-species and community assessments, provide the best route for understanding the effects of toxicants such as lindane for regulatory purposes.  相似文献

Abstract

Acute and chronic toxicity tests with lindane were conducted on Daphnia magna. The 24‐hr static LC50 was 1.64 mgL‐1. The sublethal effects of 0.16, 0.25, 0.32, 0.60 and 0.80 mgL‐1 lindane on the survival, reproduction and growth of D. magna were monitored for 21 days. The algae Nannochloris oculata (5 × 105 cellsmL‐1) was used to feed the daphnids. The parameters used to determined the effect of the pesticide on D. magna were:mean total young per female, mean brood size, days to first brood, intrinsic rate of natural increase (r), growth, and survival. Reproduction as well as survival was significantly reduced at lindane concentrations of 0.25 mgL‐1 and higher. The intrinsic rate of natural increase (r) decreased with increasing concentrations of lindane. Growth, as measured by body length, was depressed significantly at 0.25 mgL‐1 lindane and higher. The chronic data was used to formulate an acute/chronic ratio.  相似文献

Abstract

The wood charcoal treated by 1N HNO3 (WCT) was used to remove toxic chlorinated pesticide lindane (y‐HCH) residue from water by the sorption process. Using a simple first order reversible kinetics constants and half time equations, the film and pore diffusion coefficients were determined. Film diffusion was found to be rate limiting step in sorbing lindane by WCT. This was further authenticated by kinetics studies at different initial sorbate concentrations, different sorbent sizes, and different agitation speeds besides interruption test. A pore diffusion model was used to fit the data of kinetics in continuously mixed batch reactors (CMBR), and the tortuosity, external resistance, and surface diffusion effects on lindane sorption by WCT were noticed. The tortuosity values of 15 to 28 were obtained for WCT‐lindane system.  相似文献

Previously unreported metabolites from the urine of rats treated with lindane have been identified as configurational isomers of 2,4,5,6- and 2,3,4,6-tetrachloro-2-cyclohexen-1-ol. In addition, an intermediate metabolite from the incubation of lindane with liver preparations, under N2, has been identified as the configurational isomer γ-3,4,5,6-tetrachlorocyclohex-1-ene. The pathways leading to these metabolites appear to have an important role in the metabolism of lindane by mammals.  相似文献

A Mg0/Pd(+4) bimetallic system was evaluated to dechlorinate endosulfan and lindane in the aqueous phase. Studies were conducted with endosulfan and lindane separately, with or without acid in a 1:1 (v/v) water:acetone phase. In the absence of any acid, higher degradation of endosulfan and lindane was observed using Mg0/Pd(+4) doses of 10/0.5 and 4/0.1 mg/mL, respectively. Acetone plays an important role in facilitating the dechlorination reaction by increasing the solubilities of pesticides. Dechlorination kinetics for endosulfan and lindane (30 and 50 mg/L [30 and 50 ppm] concentration of each pesticide) were conducted with varying Mg0/Pd(+4) doses, and the time-course profiles were well-fitted into exponential curves. The optimum observed rate constants (k(obs)) for endosulfan and lindane were obtained with Mg0/Pd(+4) doses of 5/0.5 and 4/0.1 mg/mL, respectively. Gas chromatography-mass spectrometry analyses revealed that endosulfan and lindane were dechlorinated completely into their hydrocarbon skeletons-Bicyclo [2,2,1] hepta 2-5 diene and benzene, respectively.  相似文献
