The reef coral Pocillopora damicornis (Linnaeus) was grown for 8 wk in four nutrient treatments: control, consisting of ambient, unfiltered Kaneohe Bay seawater [dissolved inorganic nitrogen (DIN, 1.0 M) and dissolved inorganic phosphate (DIP, 0.3 M)]; nitrogen enrichment (15 M DIN as ammonium); phosphorus enrichment (1.2 M DIP as inorganic phosphate); and 15 M DIN+1.2 M DIP. Analyses of zooxanthellae for C, N, P and chlorophyll a after the 8 wk experiment indicated that DIN enrichment increased the cellular chlorophyll a and excess nitrogen fraction of the algae, but did not affect C cell-1. DIP enrichment decreased both C and P cell-1, but the decrease was proportionally less for C cell-1. the response of cellular P to both DIN and DIP enrichment appeared to be in the same direction and could not be explained as a primary effect of external nutrient enrichment. The observed response of cellular P might be a consequence of in situ CO2 limitation. DIN enrichment could increase the CO2 (aq) demand by increasing the net production per unit area. Growth characteristics and nutrient uptake kinetics were determined for zooxanthellae (Gymnodinium microadriaticum) in laboratory culture. The maximum specific growth rate (max) was 0.35 d-1 at 27 °C, 12 hL:12 hD cycle, 45 E m-2 s-1. Anmmonium and nitrate uptake by G. microadriaticum in distinct growth phases exhibited Michaelis-Menten kinetics. Ammonium half-saturation constants (Ks) ranged from 0.4 to 2.0 M; those for nitrate ranged from 0.5 to 0.8 M. Ammonium maximum specific uptake rates (Vmax) (0.75 to 1.74 d-1) exceeded those for nitrate (0.14 to 0.39 d-1) and were much greater than the maximum specific growth rate (0.35 d-1), suggesting that ammonium is the more significant N source for cultured zooxanthellae. Ammonium and nitrate Vmax values compare with those reported from freshly isolated zooxanthellae. Light enhanced ammonium and nitrate uptake; ammonium inhibited nitrate uptake which was not reported for freshly isolated zooxanthellae, suggesting that physiological differences exist between the two. The separate and combined effects of ammonium (10M) and phosphate (2M) on the ultrastructure of zooxanthellae (Symbiodinium sp.) from giant clams, Tridacna maxima, were examined in the field. Nitrogen addition significantly changed the ultrastructure of the zooxanthellae inhabiting the clams. After 9 mo exposure, the cross-sectional area of zooxanthellae from N-treated clams was significantly lower than that from other treatments [N=39.3 m2; C=47.9 m2; P=43.2m2; N+P=44.5 m2; (P=0.001)]. There was also a significant decrease in the size of starch bodies, especially around the pyrenoid of the zooxanthellae from N and N+P treatments [N=1.2 m2; C=2.0 m2; P=1.8 m2; N+P=1.2 m2; (P=2.08E-11)]. This presumably occurs as a result of the mobilization of organic carbon stores in response to stimulated amino acid synthesis under enriched nutrient conditions. These data strongly suggest that the symbiotic zooxanthellae of clams are limited to some extent by the availability of inorganic nitrogen, and that relatively minor changes to the nutrient loading of the water column can have substantial effects on the biochemistry of symbioses such as that which exists between clams and zooxanthellae.  相似文献

Sea anemones (Aiptasia pulchella) containing zooxanthellae (Symbiodinium microadriaticum) were maintained in a long-term laboratory culture on a 12 h light (100 E m-2 s-1):12 h dark cycle. Photosynthetic oxygen production was measured for the symbiotic association and for freshlyisolated zooxanthellae. Light utilization efficiencies () were similar for both sets of zooxanthellae, suggesting negligible shading of zooxanthellae by animal tissue in this association. Whereas freshly-isolated zooxanthellae were photoinhibited at high irradiances (800 to 1 800 E m-2 s-1), zooxanthellae in the host continued to function at photosynthetic capacity. Time of day may influence photosynthetic measurements in symbiotic organisms, as it was found that photosynthesis in A. pulchella followed a diel periodicity at both light-saturating (1 200 E m-2 s-1) and subsaturating (150 E m-2 s-1) irradiances. There was a peak period of photosynthesis between 12.00 and 14.00 hrs. Light stimulated dark respiration rates of A. pulchella. Dark respiration of sea anemones increased somewhat towards the end of the light cycle and was always greater after exposure to high irradiances.  相似文献

Michaelis-Menten uptake kinetics were observed at all light intensities. With constant illumination, the Vmax and K1 in nitrate uptake over the natural light intensity range of 0 to 2000 E were 0.343 g-at NO3–N(g)-1 at protein-N h-1 and 26 E, respectively. Nitrate uptake was inhibited at higher light intensities. The Ks for nitrate uptake did not vary as a function of light intensity remaining relatively constant at 0.62 g-at NO3–N 1-1. With intermittent illumination, the Vmzx for light intensity in nitrate uptake over a light intensity range of 0 to 5000 E was 0.341 g-at NO3–N(g)-1-at protein-N h-1. No inhibition of nitrate uptake was observed at higher than natural light intensities. Chaetoceros curvisetus will probably never experience light inhibition of nitrate uptake under natural conditions.  相似文献

The locus of symbiotic dinoflagellates within host cells provides a habitat which could potentially be exploited by the alga through heterotrophic uptake of host-derived organic substrates. Using zooxanthellae (Symbiodinium sp.) isolated from the tropical sea anemone Aiptasia pulchella collected from Kaneohe Bay, Hawaii, the effect of various potential organic substrates on growth in vitro was assessed in Erdschreiber seawater medium (ES) supplemented with organic compounds. Zooxanthellae maintained at 5 to 7 E m-2 s-1 (below compensation irradiance) grew heterotrophically when supplied with 100 M glycerol, glycolate, acetate, malate, or propionate, and grew in darkness on 100 M propionate. Zooxanthellae exposed to irradiance below compensation were able to utilize carbon sources in the unsupplemented ES medium for slow growth, but generally the growth rate of cultured zooxanthellae was a function of incubation irradiance. Zooxanthellae incubated for 10 wk in unsupplemented ES at 5 to 7 E m-2 s-1 were capable of growth at this low irradiance, but were also capable of net photosynthetic oxygen production at higher irradiances. This suggests that zooxanthellae can be photoautotrophic or facultatively heterotrophic. An estimate for the duration of mitosis (t d ) is made on the basis of growth rate of cultured zooxanthellae in log-phase; this estimate of t d =4.88 h is less than half the estimated t d for zooxanthellae in situ.  相似文献

Feeding, growth and bioluminescence of the thecate heterotrophic dinoflagellate Protoperidinium huberi were measured as a function of food concentration for laboratory cultures grown on the diatom Ditylum brightwellii. Ingestion of food increased with food concentration. Maximum ingestion rates were measured at food concentrations of 600 g C l-1 and were 0.7 g C individual-1 h-1 (1.8 D. brightwelli cells individual-1 h-1). Clearance rates decreased asymptotically with increasing food concentration. Maximum clearance rates at low food concentration were ca. 23 l ind-1 h-1, which corresponds to a volume-specific clearance rate of 5.9x105 h-1. Cell size of P huberi was highly variable, with a mean diameter of 42 m, but no clear relationship between cell size and food concentration was evident. Specific growth rates increased with food concentration until maximum growth rates of 0.7 d-1 were reached at a food concentration of 400 g C l-1 (1000 cells ml-1). Food concentrations as low as 10 g C l-1 of D. brightwellii (25 cells ml-1) were able to support growth of P. huberi. The bioluminescence of P. huberi varied with its nutritional condition and growth rate. Cells held without food lost their bioluminescence capacity in a matter of days. P. huberi raised at different food concentrations showed increased bioluminescence capacity, up to food concentration that supported maximum growth rates. The bioluminescence of P. huberi varied over a diel cycle, and these rhythmic changes persisted during 48 h of continuous darkness, indicating that the rhythm was under endogenous control.  相似文献

The effect of phosphate on the giant clam Tridacna gigas and on its symbiotic dinoflagellate Symbiodinium sp. was compared with that on cultured Symbiodinium sp. originally isolated from the same clarn species. Incubation of whole clams in elevated phosphate (10 M) reduced their capacity for phosphate uptake, but the uptake capacity of the clam's zooxanthellae population was not influenced. In addition, there was no change in the zooxanthellae density and the N:P ratio, of these algae., On the other hand, cultured zooxanthellae were influenced by the phosphate regimen of their culture medium. Compared with controls (0 M P), addition of 10 M phosphate to the culture medium caused an increase of 100% in cell density and decreases of 50% in the N:P ratio, and 80% in the phosphate-uptake capacity of the zooxanthellae. Zooxanthellae freshly isolated from the clams exhibited properties similar to those of zooxanthellae cultured in the absence of phosphate. These results demonstrate that the zooxanthellae population of T. gigas have limited access to the inorganic phosphate in sea water and the phosphate reserves within the animal host.  相似文献

The feeding behaviors of Acartia clausi and A. tonsa were measured in samples of water containing low levels of a water-accommodated fraction of No. 2 fuel oil. The copepods fed normally at a hydrocarbon concentration of 70 g l-1, but their feeding behavior was altered both quantitatively and qualitatively at a concentration of 250 g l-1. Three types of response to the higher oil level were found. The first was total suppression of feeding. Both other types involved suppression of feeding on particles between 7 and 15 m diameter, but one showed no change in the ingestion of larger particles, whereas the other displayed increased feeding on particles larger than 15 m diameter. These results suggest that the species of Acartia studied use three different modes of feeding, each on a different size range of particulate material. Experiments were conducted to develop a sensitive sublethal toxicity test protocol to determine the toxicity of municipal wastewater effluents to larvae of the red abalone Haliotis rufescens. In multiple tests, fertilized abalone embryos were exposed for 48 h to dilutions of a reference toxicant, zinc sulfate, and to dilutions of primary-and secondary-treated effluents. The resulting veliger larvae were examined microscopically for larval shell abnormalities. In a longer flowthrough experiment, abalone were exposed for the entire larval phase, from the two-cell stage through metamorphosis, to compare zinc effects on metamorphosis with zinc effects on short-term larval shell development. Dissolved oxygen, pH, salinity and temperature were measured daily in test solutions, and zinc concentrations were verified by chemical analysis. No observed effect concentrations (NOECs) for zinc were 39±2.1 g l-1 in three 48 h exposures, and 19 g l-1 for the 9 d exposure through metamorphosis. Median effect concentrations (EC50s) were 68±6.9 g l-1 in 48 h tests and 50 g l-1 in the 9 d test. Abalone larvae were affected at lower concentrations of primary than of secondary effluent.  相似文献

The distribution of cyanobacteria in the surface waters of the North Sea was measured during July 1987. Numbers of cyanobacteria ranged from 2.5x106 to 1.7x108 cells 1-1. In the majority of stations, cyanobacterial numbers were highest in the near-surface water and a subsurface maximum was found at only one station. The distribution of 14C among the end-products of photosynthesis was determined for picoplankton (<1 m) and other phytoplankton >1 m throughout the North Sea. The majority of label was found in the protein fraction of both picoplankton and >1 m phytoplankton; incorporation into lipids and polysaccharides plus nucleic acids was much lower. We interpret the large incorporation into protein to be a consequence of nutrient limitation of these natural assemblages. Photosynthetic parameters of the two size fractions were also determined. Assimilation number (P m B ) and initial slope were greater for the picoplankton fraction than for phytoplankton >1 m but there was no evidence of significant photoinhibition of either fraction at irradiances up to 1 000 E m-2 s-1.  相似文献

A method is described for the incubation of undisturbed sediment cores under in situ conditions with the addition of low concentrations of 14C-glucose. Data are presented for respiration, gross uptake and actual uptake rate of glucose by bacteria in sandy, wave-washed beaches of the Baltic Sea. On the average, the bacteria respired 8% of the total glucose taken up. The gross uptake measured was between 2.3×10-3 and 6.8×10-3 g 14C-glucose g sediment-1 (dry weight) h-1 (average 4.7×10-3 g g-1 h-1). Minima in the gross uptake rate corresponded with maxima in the concentration of natural free dissolved glucose. For the actual uptake rate, however, very similar uptake rates were calculated for the sediments examined (between 1.4×10-1 and 1.9×10-1 g glucose g-1 h-1, average 1.7×10-1 g g-1 h-1).Publication No. 183 of the Joint Research Program at Kiel University (Sonderforschungsbereich 95 der Deutschen Forschungsgemeinschaft).  相似文献

The uptake of nitrate and ammonium was investigated experimentally during early spring 1989 in the Greenland Sea, with particular attention placed on the roles of irradiance, nitrogen concentrations and nitrateammonium interactions. The phytoplankton assemblage was dominated by the colonial prymnesiophyte Phaeocystis pouchetii. Nitrate concentrations ranged from undetectable at the end of the cruise to greater than 10 M, and ammonium levels ranged from less than 0.1 to 1.9M. The uptake of both nitrate and ammonium as a function of irradiance was found to be a saturation response. Photoinhibition occurred and was found to be greater for ammonium uptake. Ammonium uptake also saturated at irradiance levels five times lower than those needed to saturate nitrate uptake. Nitrate and ammonium uptake as a function of nitrogen concentration also was characterized by a saturation response, with the estimated half-saturation constant (K s) value for nitrate uptake being 0.29 M. Elevated ammonium concentrations inhibited nitrate uptake, and the response appeared to be one of exponential decrease with increasing concentrations of ammonium. The most important factor in the Greenland Sea influencing ammonium uptake during the spring was irradiace, while both irradiance and ammonium concentrations played major roles in regulating nitrate uptake and new production.  相似文献

Adult Elasmopus rapax, collected from the eastern coast of Venezuela in 1990, were exposed to seawater containing various CdCl2 concentrations ranging from 0.25 to 5.5 mol l-1. The 48-h and 96-h LC50 values obtained were 4.0 and 1.6 mol Cd l-1, respectively. In amphipods exposed to 1 mol Cd l-1 for up to 240 h, the apparent rate of cadmium uptake was higher in dead animals (most of which had molted during the preceding 24 to 48 h) than in those which survived throughout the treatments without molting. Thus, whole-body cadmium content reached 1.74 mol g-1 dry weight (dw) in the former and only 0.85 mol g-1 dw in the latter; the higher body Cd-load may have caused the increased mortality observed in molters. On exposure to cadmium levels above 0.5 mol l-1 the oxygen consumption rate of non-molters decreased from 2.2 to about 1.5 ml O2 g-1 dw h-1 over the first 24 h, remaining unchanged thereafter. The results place E. rapax among the most sensitive marine organisms yet studied concerning cadmium toxicity, and emphasize the usefulness of the Amphipoda as bioindicators and research tools for bioassays.  相似文献

We estimated primary productivity and distributions of carbon in the phytoplankton, micro-zooplankton, and suspended and dissolved matter in various areas of the World Ocean to increase our information about the organic carbon cycle in the surface layer of the sea. Primary productivity ranged from about 0.1 gC m–2 day–1 in the Gulf of Mexico to 9 gC m–2 day–1 in nutrient-rich water off Peru. Phytoplankton carbon ranged from less than 10 g/l in the former to 750 g/l in the latter and in nutrient-rich water off southwest Africa. Micro-zooplankton carbon usually was less than 50 g/l in all waters, and was dominated by ciliates, copepodids, and copepod nauplii in all areas. Concentrations of particulate carbon ranged from 12 g/l off the east coast of South America to 850 g/l off southwest Africa. Concentrations of dissolved organic carbon varied between 0.5 and 1.5 mg/l in all areas except off Peru, where maximum values of 4.5 mg/l were observed. Turnover rates of carbon by small standing crops of micro-flagellates (1 to 5 longest dimension) and dinoflagellates in nutrient-poor waters were lower than those by large standing crops of diatoms and micro-flagellates in nutrient-rich waters. Concentrations of phytoplankton usually accounted for 20 to 55% and micro-zooplankton for 2 to 30% of the particulate carbon in the surface layer of the sea. Concentrations of dissolved organic carbon were not related to concentrations of particulate carbon in most waters except off Peru, where they appear to be directly related.  相似文献

Specimens of the oceanic decapod Systellaspis debilis were collected from six sites in the East Atlantic Ocean between 1970 and 1984, and were analysed for Mn, Fe, Cu, Zn and Cd. The data confirm that there are small but significant differences in mean metal concentrations from some sites which showed no obvious pattern in relation to geographic location of the samples. As a result, ranges of site means are quoted as baseline levels for each metal (g g-1 dry wt): 2.3 to 2.9 g Mn g-1, 31.2 to 77.8 g Fe g-1, 25.9 to 83.4 g Cu g-1, 41.9 to 92.9 gZn g-1, 11.1 to 31.8 g Cd g-1. The concentration of cadmium in S. debilis from all sites was raised relative to cadmium concentrations reported for coastal decapods, perhaps as a result of dietary enrichment. Metal accumulation may provide useful information for understanding the complex feeding behaviour of many oceanic animals.  相似文献

Rainwater concentrations of either ammonium or nitrate were sufficient to stimulate chlorophyll a (chl a) production in bioassay experiments using Gulf Stream surface water collected off North Carolina during the summer of 1991. Previous studies primarily examined inshore waters and did not address the impact of rainwater ammonium. An increase in chl a occurred within 1 d of the addition of synthetic rainwater (2 or 5% rainwater, 98 or 95% seawater) containing up to 10 M ammonium; this increase was followed by a decrease in chl a the following day. A similar response to nitrate addition (5% addition of 20 M nitrate rain) was observed. In separate experiments, natural rainwater having nitrate and ammonium concentrations less than those in the experimental synthetic rain yielded a greater chl a response than synthetic rain when added at similar dilutions (0.5 to 5.0% rain). The maximum dissolved inorganic nitrogen concentration in the enriched seawater in these bioassays was 1.8 M; prior to enrichment the maximum was < 0.4 M. Bioassay experiments begun 2 d after a major storm event (sustained NE winds with gusts to 13 m s-1 and ca. 390 mol m-2 inorganic nitrogen deposition from rain) showed a chl a increase in response to addition of natural rainwater, but not to synthetic rainwater with similar dissolved inorganic nitrogen concentration. These results suggest that phytoplankton stimulants, in addition to nitrate and ammonium, exist in natural rain but not in the synthetic rain used in these experiments.  相似文献

Orthophosphate uptake by a natural estuarine phytoplankton population was estimated using two methods: (1) 32P uptake experiments in which filters of different pore sizes were used to separate plankton size-fractions; (2) 33P autoradiography of phytoplankton cells. Results of the first method showed that plankton cells larger than 5 m were responsible for 2% of the total orthophosphate uptake rate. 98% of the total uptake rate occurred in plankton composed mostly of bacteria, which passed the 5 m screen and were retained by the 0.45 m pore-size filter. There was no orthophosphate absorption by particulates in a biologically inhibited control containing iodoacetic acid. Orthophosphate uptake rates of individual phytoplankton species were obtained using 33P autoradiography. The sum of these individual rates was very close to the estimated rate of uptake by particulates larger than 5 m in the 32P labelling experiment. Generally, smaller cells were found to have a faster uptake rate per m3 biomass than larger cells. Although the nannoplankton constituted only about 21% of the total algal biomass, the rate of phosphate uptake by the nannoplankton was 75% of the total phytoplankton uptake rate. Results of the plankton autoradiography showed that the phosphate uptake rate per unit biomass is a power function of the surface: volume ratio of a cell; the relationship is expressed by the equation Y=2x10-11 X 1.7, where Y is gP m-3 h-1 and X is the surface: volume ratio. These results lend support to the hypothesis that smaller cells have a competitive advantage by having faster nutrient uptake rates.  相似文献

The blacksmith Chromis punctipinnis, an abundant planktivorous damselfish off southern California, USA, shelters along rocky reefs at night. While sheltered, blacksmiths excrete ammonium that could, in turn, be utilized by nearby benthic macrophytes. Laboratory experiments during the summer and fall of 1983 and 1984 indicate that ammonium excretion at night ranged from 18.1 mol h-1 by a 8.5 g (dry) fish, to 89.1 mol h-1 by a 27.3 g fish; excretion rates generally declined throughout the night. Field measurements at night indicate that ammonium concentrations were significantly higher in rocky crevices occupied by blacksmiths than in unoccupied shelters, and the ammonium level in one shelter dropped after a blacksmith was experimentally removed. Young kelp plants (Macrocystis pyrifera) are capable of taking up ammonium at night. Ammonium levels in chambers containing both a blacksmith and a young kelp plant were significantly lower than in chambers containing only a fish, and ammonium levels dropped in ammoniumspiked chambers that contained kelp plants. Nighttime ammonium uptake rates by young kelp plants, which averaged 1.6 mol g-1 (dry) h-1, were only slightly lower than those during the day. Daytime excretion by blacksmiths occasionally results in elevated ammonium levels in the water column. On two of six days, ammonium concentrations in midwater foraging aggregations were slightly but significantly higher than in upcurrent controls; since blacksmiths typically aggregate at the incurrent margin of kelp beds, the ammonium is swept downcurrent and may be utilized by large M. pyrifera that extend through the water column. Thus, the activities of blacksmiths may results in the importation of extrinsic, inorganic nitrogen to primary producers on temperate reefs.  相似文献

Nauplii of Calanus pacificus were raised on a mixture of algae. Details of the mouth-parts, such as denticles, labial palps and lobes, setations and structure of the masticatory teeth were examined by scanning electron microscopy (SEM). Under the experimental conditions (15°C and 300 gC l-1), exponential growth coefficients for the period Nauplii II–VI were 0.179 for carbon and 0.228 for nitrogen. C:N ratios dropping from 5.1 to 4.7. Growth was isochronous, each stage lasting 1.5 days. Respiratory losses were 15 to 19.6% of body carbon daily. Nauplii raised on a given alga showed higher rates of ingestion in the presence of this food, compared to nauplii switched to other algae. Minimal threshold concentrations for feeding were found, depending on the size of the food offered and ranging from 5.8 gC I-1 for Lauderia borealis (28.7 m spherical diameter) to 47.1 gC 1-1 for Chlamydomonas sp. (11.0 m). Unlike the Copepodite I stage, Nauplii II–VI larvae were not able to ingest small cells such as Isochrysis galbana (4.3 m), or very large ones such as Ditylum brightwellii (47.5 m) at more than maintenance rations. Below the critical concentration for maximal feeding, ingestion was clearly dependent on size of the cells offered, but the size-dependent relationship was different for diatoms and non-diatoms. Filtering rates increased from a threshold concentration to a maximal rate at about 50 gC 1-1, and decreased at higher concentrations. Critical concentrations ranged from 125 gC 1-1 for L. borealis to 1000 gC 1-1 for Chlamydomonas sp. Maximal daily rations ranged between 100 and 150% of body carbon.  相似文献
