BACKGROUND: Incidence of amphibian deformities have increased in recent years, especially in the northern region of the United States. While many factors have been proposed as being responsible for generating deformities (e.g., contaminants, ultraviolet radiation [UV], parasites), no single cause has been definitively established. METHODS: To determine whether waterborne chemicals are responsible for amphibian deformities in ponds in north-central Minnesota, we deployed semipermeable membrane devices (SPMDs) in an impacted and a reference site to accumulate lipophilic contaminants. We then exposed native tadpoles (northern leopard frogs; Rana pipiens) to the SPMD extracts combined with two agricultural pesticides (atrazine, carbaryl) at two levels of UV radiation. RESULTS AND DISCUSSION: UV radiation alone caused a slight increase in hatching success and tadpole growth rate. Deformity rate among hatchlings was high following exposure to SPMD extracts from the reference site in the absence of UV, suggesting that chemicals present at this site are broken down by UV to less harmful forms, or become less bioavailable. Conversely, impacted site SPMD extracts caused hatchling deformities only in the presence of UV, suggesting that UV potentiates the teratogenicity of the compounds present there. Impacted site SPMD extracts significantly increased the number of bony triangles among metamorphs, a common deformity observed at this site. The incidence of skin webbings increased significantly with SPMD extracts from both sites as well as with our pesticide control containing atrazine and carbaryl alone. CONCLUSIONS: Higher deformity rates among tadpoles reared in the presence of UV radiation and SPMD extracts from sites where deformities are common indicates a chemical compound (or compounds) in the water at this site may be causing the deformities. RECOMMENDATIONS AND OUTLOOK: It is important to examine the effects of chemical stressors in the presence of other natural stressors (e.g., UV radiation) to gain a better understanding of how multiple stressors work to impact amphibians and amphibian populations.  相似文献

Mercury (Hg) causes a range of deleterious effects in wildlife, but little is known about its effects on amphibians. Our objective was to determine whether Hg affects performance and behavior in two-lined salamanders (Eurycea bislineata). We collected salamanders from Hg-contaminated and reference sites and assessed speed, responsiveness, and prey capture ability. Mercury concentrations were >17× higher in salamanders from the contaminated sites and were among the highest documented in amphibians. In the first, but not in the second, locomotion trial, we found a significant effect of Hg on speed and responsiveness. In the prey capture experiment, reference salamanders ate approximately twice as many prey items as the contaminated salamanders. Together, our results suggest that sublethal Hg concentrations may negatively affect salamanders by reducing their ability to successfully execute tasks critical to survival. Future work is warranted to determine whether Hg has other sublethal effects on salamanders and whether other amphibians are similarly affected.  相似文献

Bisphenol A (BPA), which is ubiquitous in the environment, is an example of an endocrine-disrupting compound (EDC). Ammonium assimilation has an important function in plant growth and development. However, insufficient information on the potential effect of BPA on ammonium assimilation in plants is available. In this study, the effects of BPA on ammonium assimilation in roots of soybean seedlings were investigated. During the stress period, 1.5 mg L?1 of BPA improved glutamine synthetase (GS)/glutamate synthase (GOGAT) cycle and glutamate dehydrogenase (GDH) pathway in ammonium assimilation. The amino acid and the soluble protein contents increased in the soybeans. At 17.2 and 50.0 mg L?1 of BPA, the GS/GOGAT cycle was inhibited and the GDH pathway was promoted. The amino acid content increased and the soluble protein content decreased. During the recovery period, the GS/GOGAT cycle and the GDH pathway recovered at 1.5 and 17.2 mg L?1 of BPA but not at 50.0 mg L?1 of BPA. The amino acid content continuously increased and the soluble protein content decreased compared with those in the control treatment. In summary, BPA treatment could affect the contents of soluble protein and amino acid in the soybean roots by regulating ammonium assimilation.  相似文献

The effects of long-term enhanced UV-B radiation on growth and secondary compounds of two conifer species were studied in an outdoor experiment. Scots pine (Pinus sylvestris) seedlings were exposed for two growing seasons and Norway spruce (Picea abies) seedlings for three growing seasons to supplemental UV-B radiation, corresponding to a 30% increase in ambient UV-B radiation. The experiment also included appropriate controls for ambient and increased UV-A radiation. Enhanced UV-B did not affect the growth of the conifer seedlings. In addition, neither the concentrations of terpenes and phenolics in the needles nor the concentrations of terpenes in the wood were affected. However, in the UV-A control treatment the concentrations of diterpenes in the wood of Scots pine decreased significantly compared to the ambient control. Apparently, a small increase in UV-B radiation has no significant effects on the secondary compounds and growth of Scots pine and Norway spruce seedlings.  相似文献

Some environmental contaminants have the potential to affect humans or animals by mimicking the effects of hormones. Bisphenol A (BPA) is a weak estrogen agonist when tested using in vitro or in vivo bioassays. In addition to the well documented effects of estrogens on reproductive functions, ovarian hormones also have salient effects on mammalian energy balance and feeding behavior. In this study, we investigated the effects of BPA on body weight and food intake of ovariectomized adult female rats. Treatment with doses of 4 or 5 mg/day for 15 days resulted in a significant reduction of body weight gain with no reduction in food intake. A dose of 1 mg/day did not affect feeding or weight gain. BPA was detected in the blood, brain and adipose tissues of the BPA-treated animals but not in the vehicle control group. There was a preferential concentration of BPA in brown adipose tissue. These results indicate that BPA can affect energy balance and that brown adipose tissue may be a primary tissue into which BPA accumulates in mammals.  相似文献

When food producing animals are contaminated with PCDD/F congeners, information on the contaminant’s concentration in the bodies of the animals at time of slaughter is needed for risk management purposes. We have developed a mathematical model for the kinetics of PCDD/Fs in growing pigs in case of contaminated feed fed for a limited duration of time. This model allows the prediction of concentrations in body fat. It considers absorption fractions of PCDD/Fs, clearance by metabolism, dilution by growth and excretion through fecal fat. The model parameters were calibrated by fitting the model to experimental data. On the basis of this toxicokinetic model a probabilistic model has been constructed. The probabilistic model handles the parameters with appropriate probability distributions and Monte-Carlo simulation technique, providing for realistic situations with many animals and a range of contaminations and feeding intervals. We applied the new model to describe the German dioxin incident of winter 2010/2011 and discuss its viability as decision tool. The approach demonstrated here is a showcase how a risk assessment in the case of contaminated feeding can be performed.  相似文献

Effects of ultraviolet-B (UV-B) radiation on amphibian embryos have been investigated in a number of studies, but the effects on larvae have received less attention. We investigated the effects of UV-B radiation on the behaviour and growth of larvae of three amphibians (Rana arvalis, Rana temporaria and Bufo bufo) in two different experiments. First, we tested whether larvae of the three species actively avoid UV-B exposure if given a choice. We found no evidence for active avoidance of UV-B or changes in activity in the presence of UV-B in any of the species. Second, we assessed the effects of natural (1.25 kJm(-2)) and enhanced (1.58 kJm(-2)) UV-B radiation on the survival and growth of the three species and found that the exposure to UV-B radiation did not have any effect on survival rates of any of the species. However, UV-B radiation had a positive effect on the growth of R. arvalis and R. temporaria, whereas the growth of B. bufo tadpoles was unaffected by the UV-B treatments. Our results suggest that a short-term exposure to UV-B radiation does not induce any UV-B avoidance behaviour in tadpoles of these three species. Furthermore, unlike some previous studies, the results suggest that the young tadpoles of these species are not negatively affected by UV-B radiation. In fact, our results demonstrate that a moderate amount of UV-B radiation enhance tadpole growth rates in two of the three species.  相似文献

The temperate forests of the southern hemisphere are the most likely forests to be affected by increased levels of ultraviolet-B (UVB) radiation resulting from reduced ozone. The review describes these forests and then discusses the morphological changes, physiological effects, and protection mechanisms, particularly UV absorbing compounds that result from present day and increasing UVB radiation. A number of currently used industrial chemicals are estrogenic, and therefore have potential to disrupt sexual differentiation in vertebrate wildlife during critical developmental windows. We assessed the effect of larval exposure to bisphenol A (BPA) on growth, development and sexual differentiation of the gonad in the African Clawed frog, Xenopus laevis. Larvae were maintained in flow-through conditions at 22 +/- 1 degrees C and exposed to BPA at mean measured concentrations of 0.83, 2.1, 9.5, 23.8, 100, and 497 microg/l, from developmental stages 43/45-66 (completion of metamorphosis). Each test concentration, plus dilution water control (DWC) and positive control (17beta-estradiol (E2), 2.7 microg/l) employed four replicate test vessels with 40 larvae per tank. Individual froglets were removed from test vessels upon reaching stage 66, and the study was terminated at 90 days. Froglets were dissected and sex was determined by inspection of gross gonadal morphology. Test concentrations of BPA had no effect on survival, growth, developmental stage distributions at exposure days 32 and 62, or mean time to completion of metamorphosis, compared to DWC. Analysis of post-metamorphic sex ratio, determined by gross gonadal morphology, indicated no significant deviations from expected (50:50) sex ratio, in DWC or any BPA test concentration. In contrast, exposure of larvae to (E2) resulted in feminisation, with sex ratio deviating significantly (31% male, replicates pooled). Exposure to BPA in the concentration range 0.83-497 microg/l in flow-through conditions had no observable effect on larval growth, development or sexual differentiation (as determined by gross gonadal morphology) in this study.  相似文献

The soil ecotoxicology literature has focused primarily on a few major taxa, to the neglect of other fossorial organisms such as amphibians. We selected cadmium (Cd) and the American toad (Bufo americanus) as a model contaminant and biological species to assess the impact of soil contamination on amphibian hibernation survival and post-hibernation condition. Soil sand composition (50, 70, 90%) and hydration (100, 150% water holding capacity (WHC)) were manipulated in addition to Cd concentration (0, 56, 165, 483 microg/g) to determine whether these soil properties affect toxicity. Soil Cd concentration significantly reduced survival and locomotor performance, and was correlated negatively with percent mass loss and positively with whole body Cd concentration. Higher sand content resulted in less mass loss and greater Cd uptake. Toads that were hibernated in 50% sand hydrated to 100% WHC had higher survival, less mass loss, and better sprint performance than those hibernated in 50% sand, 150% WHC. This study demonstrates that concentrations of Cd found in soil at highly contaminated sites can be bioaccumulated by hibernating amphibians and may reduce fitness. Differences in microhabitat use may cause species to vary in their exposure and susceptibility to soil contamination. The toxicity of Cd to amphibians could be greater in natural systems where there are multiple stressors and fluctuations in environmental variables.  相似文献

We exposed albino and pigmented medakaOryzias latipes to simulated solar ultraviolet-B (UVB) radiation to determine if albino medaka were less tolerant of UVB radiation than medaka pigmented with melanin. There was no difference in the number of albino and pigmented medaka that died during the exposure period. Spectrophotometric analyses of the outer dorsal skin layers from albino and pigmented medaka indicated that, prior to exposure, both groups of fish had similar amounts of an apparent colorless non-melanin photoprotective substance that appears to protect other fish species from UVB radiation. Our results indicate that albino medaka were as tolerant of UVB radiation as pigmented medaka because they had similar amounts of this photoprotective substance in the outer layers of the skin.  相似文献

Background  Exposure to harmful levels of ultraviolet-B radiation (UVB), a component of solar radiation, has been suggested as a potential cause of amphibian declines. Methods  We measured solar radiation (UVB, ultraviolet-A, and visible) wavebands in breeding ponds of Bufo boreas (boreal toad, a montane species that has undergone severe population declines) and Bufo woodhousii (Woodhouse’s toad, a plains toad that has not experienced declines) and examined tolerances of these species to simulated solar UVB exposures in the laboratory. Results  We found larvae of both species to be tolerant of simulated solar UVB in excess of solar UVB levels observed in their breeding ponds. B. boreas tadpoles were more tolerant of simulated solar UVB exposure than B. woodhousii tadpoles, possibly because of greater amounts of photoprotective melanin in B. boreas skin. Conclusions  UVB levels observed in B. boreas habitats do not currently appear to constitute a threat to the survival of these animals; however, long-term (>1 month) exposure to UVB levels comparable to levels associated with the water interface appears to reduce survival in B. woodhousii tadpoles. Therefore, future increases in surface and water column UVB radiation in bufonid habitats might pose significant survival risks to B. boreas or B. woodhousii populations.  相似文献

The aim of this work was the development of monoclonal antibodies (MAbs) and highly sensitive immunoassays (ELISAs) to bisphenol A (BPA), a well-known endocrine disruptor able to migrate from the internal coating of cans to food contained inside, particularly vegetables. To produce MAbs to BPA, four synthetic compounds were conjugated to proteins and used as immunizing haptens in mice. By applying hybridoma technology, several MAbs were produced and selected. These antibodies were characterized in the conjugate-coated and in the antibody-coated formats, using both homologous and heterologous conjugates. Three indirect ELISA based on the MAbs showing the highest affinity to BPA were selected. The limit of detection of the most sensitive ELISA was 0.22 nM (0.05 ng/mL), with an I?? value of around 1 nM (0.23 ng/mL). An homologous ELISA based on the MAb BPAB-11 was applied to the simple, direct determination of BPA in the liquid portion of canned artichoke, peas, and sweet corn. Only sample dilution in an appropriate saline buffer was required to minimize matrix effects and to enter the ELISA working range. Recovery and precision of the method were evaluated by spiking the liquid portion of these cans with BPA at 20, 50, and 100 ng/mL. Coefficients of variation were below 20% in most cases. With regard to recovery, the analytical data obtained were also acceptable. This immunoassay has therefore proved its potential as a new tool for the rapid, sensitive and accurate determination of BPA in canned food.  相似文献

In this study, we tested phenolic compounds such as bisphenol A (BPA), 4-nonylphenol (NP), 4-octylphenol (OP) and 4-propylphenol (PP) by using glucose-6-phosphate dehydrogenase (G6PD) in estrogen sensitive human breast cancer cells (MCF-7 cells) and glutathione peroxidase (GPx) in female immature Sprague-Dawley (SD) rats. This study was designed to investigate whether phenolic compounds have estrogenic effects in these useful screening methods for endocrine disruptors. We chose 6 h as the incubation period for the G6PD assay through a preliminary experiment using 17beta-estradiol (E2). Above the concentration of 1 x 10(-8) M, BPA significantly increased the G6PD activity in a concentration-dependent manner, relative to the control. NP (over the concentration of 1 x 10(-9) M) also enhanced the G6PD activity by about 1.8 times that of the control. OP produced weaker effects on G6PD than NP, and showed a tendency to increase the G6PD activity. PP did not affect the G6PD activity. These results show that BPA and NP have the effect of enhancing G6PD activities in MCF-7 cells. In the in vivo GPx assay, both BPA and E2 significantly increased the uterus wet weights and dramatically enhanced uterine GPx activities in immature female rats in a dose-dependent manner. Treatment with NP (500 mg/kg/day) increased significantly both the uterine GPx activity and the uterus wet weights in immature female rats. OP (500 mg/kg/day) also caused a significant increase in uterine GPx activity, but had no effect on the uterus wet weights. This finding indicates that the change in uterine GPx activities could be a more sensitive parameter than that of uterus wet weights in immature rats. This study implies that phenolic compounds have a weak estrogenic effects.  相似文献

Battery tests serve as integral tools to decide whether a treatment process is ecotoxicologically safe or not. In the present study, a battery of toxicity tests was employed to elucidate the toxicity of the potential endocrine-disrupting pollutant bisphenol A (BPA) and its advanced oxidation products. For this purpose, BPA was subjected to Fenton treatment in the growth medium of the test organisms employed as well as in real lake water. Treatment results indicated that BPA removals were fast and complete within less than a minute, whereas total organic carbon (TOC) removals were rather incomplete, speaking for the accumulation of refractory degradation products. The presence of chloride and/or natural organic matter influenced H2O2 consumption rates and the treatment performance of the Fenton’s reagent as well. The sensitivity of the selected test organisms for BPA and its Fenton treatment products in different water matrices was found in the following decreasing order: the freshwater microalgae (Pseudokirchneriella subcapitata) > the freshwater cladoceran (Daphnia magna) > marine photobacteria (Vibrio fischeri).  相似文献

以太阳能固定膜光催化中试装置,研究了光解、初始浓度和平均光强等对双酚A(BPA)光催化去除的影响及BPA的矿化和在自来水中的处理效果.试验结果表明,BPA在日光照射下很难光解,其光催化降解呈表观一级反应,在平均光强介于5.7~23.5 W/m2时,表观反应速率常数和平均光强呈线性关系.太阳能光催化对BPA具有良好的矿化作用,但其降解与以UV254为光源的降解有不同的机理.太阳能光催化对自来水中BPA也具有较好的处理效果.  相似文献

Bisphenol A (BPA) is a well-known environmental toxic substance, which exerts unfavorable effects through endocrine disruptor (ER)-dependent and ER-independent mechanisms to threaten ecological systems seriously. BPA may also interact with other environmental factors, such as light and heavy metals, to have a synergetic effect in plants. However, there is little data concerning the toxic effect of BPA on the primary producers-plants and its possible interaction with light-dependent response. Here, the effects of BPA on germination, fresh weight, tap root length, and leaf differentiation were studied in Arabidopsis thaliana under different parts of light spectrum (dark, red, yellow, green, blue, and white light). Our results showed that low-dose BPA (1.0, 5.0 μM) caused an increase in the fresh weight, the tap root length and the lateral root formation of A. thaliana seedlings, while high-dose BPA (10.0, 25.0 μM) show an inhibition effect in a dose-dependent manner. Unlike karrikins, the effects of BPA on germination fresh weight and tap roots length under various light conditions are similar, which imply that BPA has no notable role in priming light response in germination and early seedling growth in A. thaliana. Meanwhile, BPA exposure influences the differentiation of A. thaliana leaf blade significantly in a light-dependent manner with little to no effect in dark and clear effect under red illumination.  相似文献

GOAL, SCOPE AND BACKGROUND: Bioaccumulation and biomagnification of organic pollutants have been increasingly assessed and modeled during the last years. Due to the complexity of these processes and the large variability of food webs, setting generic assessments for these parameters is really difficult. Equilibrium models, based on a compound's lipophylicity, are the main tool in regulatory proposals, such as for identifying Persistent, Bioaccumulative and Toxic Substances (PBTs), although a refinement has been claimed by the scientific community. Toxicokinetic studies offer an alternative for these estimations, where biomagnification is modeled as a succession of bioaccumulation processes, each one regulated by toxicokinetic parameters. METHODS: A review of kinetic models covering species belonging to different trophic levels and with different ecological behavior has been conducted. The results were employed for setting a conceptual model for estimating the biomagnification potential in a generic food web, which was mathematically implemented through system dynamic models developed under data sheet software. Crystal Ball was then employed for allowing Monte Carlo based probabilistic calculations. Bioaccumulation laboratory assays have been performed to estimate toxicokinetic parameters in mussels (Mytilus edulis) with two PAHs (chrysene and benzo[a]pyrene). The contamination was delivered via food. The exposure period lasted more than one month followed then by a depuration phase. The contaminant content was determined on an individual basis on five replicates. RESULTS AND DISCUSSION:. The reviewed information suggested the development of a tiered conceptual biomagnification model, starting with a simplified food chain which can be refined to more realistic and complex models in successive levels. CONCLUSIONS: The mathematical implementation of the conceptual model offers tools for estimating the potential for bioaccumulation and biomagnification of chemicals under very different conditions. The versatility of the model can be used for both comparative estimations and for validating the model. RECOMMENDATIONS AND PERSPECTIVES: Since bioaccumulation and biomagnification processes are crucial elements for a proper risk assessment of chemicals, their estimation by mathematical models has been widely tested. However, inregulatory assessments, too simplistic models are still being used quite often. The biomagnification model presented in this study should be amore accurate alternative to these models. In comparison to other previously published biomagnification models, the present one covers the time variation of bioaccumulation using just a few toxicokinetic parameters.  相似文献

It is often argued that the concentration of a pollutant inside an organism is a good indicator of its bioavailability, however, we show that the rate of uptake, not the concentration itself, is the superior predictor. In a study on zinc accumulation and toxicity to isopods (Porcellio scaber) the dietary EC(50) for the effect on body growth was rather constant and reproducible, while the internal EC(50) varied depending on the accumulation history of the animals. From the data a critical value for zinc accumulation in P. scaber was estimated as 53 microg/g/wk. We review toxicokinetic models applicable to time-series measurements of concentrations in invertebrates. The initial slope of the uptake curve is proposed as an indicator of bioavailability. To apply the dynamic concept of bioavailability in risk assessment, a set of representative organisms should be chosen and standardized protocols developed for exposure assays by which suspect soils can be evaluated.  相似文献
