Enumeration of Legionella pneumophila in cooling tower water systems   总被引:3,自引:0,他引:3  
Legionella pneumophila, the causative agent of Legionnaires disease, is known to colonise and frequently grow in cooling tower waters. Disease is acquired by inhaling aerosol contaminated by legionellae. Determination of the count of Legionella pneumophila in cooling tower waters may, therefore, be useful for risk assessment. In our survey, 103 water samples from 50 cooling towers were examined over a five-year period to indicate the seasonal distribution and the ecology of L. pneumophila, as regards temperature and pH. L. pneumophila serogroup 1 was found in 44% of the isolated strains, which is primarily responsible for the majority of Legionnaires disease. The large majority of examined towers had levels of L. pneumophila in the high-risk category. These cooling towers have been linked to many outbreaks of Legionnaires disease.  相似文献

The resistance pattern and mechanisms of bacterial isolates obtained from clinical origin, soil, industrial effluent, orange juice products and drinking water were studied using commonly used antibiotics. The microbial load of the water samples, industrial effluent and orange juice products were 1.0 × 101–2.25 × 106, 2.15 × 105, and 3.5 × 104–2.15 × 105 cfu mL–1, respectively. The faecal coliform test revealed that only two out of twenty orange juice products had MPN of 2 and 20, the MPN of water ranged from 1–1800, while the effluent had MPN of 1800. The bacterial isolates that were isolated include E. coli, S. aureus, P. vulgaris, S. marcescens, S. pyogenes, B. cereus, B. subtilis, Micrococcus sp., Klebsiella sp., P. aeruginosa, and Enterobacter sp. Also, clinical and soil isolates of P. aeruginosa were used in the study. Among the eight antibiotics tested for resistance on five strains of each bacterium, seven different resistance patterns were observed among the bacterial isolates obtained from water, effluent and orange juice products. Among the clinical and soil isolates of P. aeruginosa, four multiple-drug resistance patterns were obtained. Thirty strains of E. coli and S. aureus were tested for -lactamase production and fourteen strains, seven each of E. coli and S. aureus that had high Minimum Inhibitory Concentration values (MIC) for both Amoxycillin and Cloxacillin were positive.  相似文献

Water quality and bacterial contamination from 18 drinking water municipal plants in three locations at Giza governorate were investigated. The average total count of bacteria detected after four stages of treatments in the investigated plants was 32 CFU/1 mL compared to 2330 cfu/mL for raw water, with a reduction percentage of 98.6. Although there is a relatively high removal percent of bacterial contamination from the water sources, however, several bacterial pathogens were identified in the produced water prepared for drinking including Enterococcus faecalis, Escherichia coli, Pseudomonas aeruginosa, and Shigella spp. After 3 days of water incubation at 30 °C, the amount of bacterial endotoxins ranged from 77 to 137 ng/mL in the water produced from the municipal plants compared to 621–1260 ng/mL for untreated water. The main diseases reported from patients attending different clinics and hospitals during summer 2014 at the surveyed locations and assuredly due to drinking water from these plants indicated that diarrheas and gastroenteritis due to E. coli and Campylobacter jejuni constituted 65.7% of the total patients followed by bacillary dysentery or shigellosis due to Shigella spp. (7.9%) and cholera due to Vibrio cholera (7.2%). There was an increase in serum aspartate aminotransferase (AST), alanine aminotransferase (ALT), and alkaline phosphatase (ALP) as well as urea and creatinine values of guinea pigs consuming water produced from the non-governmental plants for 6 months indicating remarkable liver and kidney damages. Histological sections of liver and kidney from the tested animal revealed liver having ballooning degeneration of hepatocytes and distortion and fragmentation of the nuclei, while the section of the kidney showed irregularly distributed wrinkled cells, degenerated Bowman’s capsule, congested blood vessels, and inflammatory cells.  相似文献

This study investigates the inherent optical properties (IOP) of a Brazilian river during a non-natural, anthropogenically mediated, toxic spill of a wood-pulping factory (the ‘Cataguazes accident’). The results indicated an outstanding transformation in the river water chromophoric dissolved organic matter (CDOM) pools. For instance, increases in CDOM absorption coefficients, a CDOM (λ), which were averaged at specific spectral intervals, , ranged from 58-fold at the UV-B and UV-A ranges to 95-fold at the PAR range. As a result, the water color expressed as CDOM absorption at 440 nm, a CDOM (440), varied from 4.16 to 365.03 m-1. For S-coefficient, the variations ranged from ∼1.1 to 5.6-fold, respectively, at the 300–650 nm and UV-B range. The variability of S as a proxy of dissolved chromophores was thus clearly influenced by the spectral range used. Optical proportions were also investigated through the use of and S ratios at the UV-B, UV-A, and PAR ranges and, in the case of , also at the NIR range. This approach also showed clear variations between the water samples, likely reflecting changes in the composition of optically active substances in the river system. As a whole, the findings obtained here indicated that both the quantity and quality of the chromophoric material dissolved in the river water were greatly altered by the toxic spill. The changes in the optical properties of the river water, although extreme and likely with no parallel in the literature, were quite rapid as indicated by the optical resilience of the system. Overall, this study indicates that IOP might be thought, and possibly used, as a metric tool for monitoring the state of waters and aquatic ecosystems.  相似文献

Cooling towers have the potential to develop infectious concentrations of Legionella pneumophila. Legionella counts increases where biofilm and warm water temperatures are present. In this study, biofilm associated L. pneumophila and heterotrophic bacteria were compared in terms of material dependence. Model cooling tower system was experimentally infected by L. pneumophila standard strain and monthly monitored. Different materials were tested for a period of 180 days. The lowest L. pneumophila and heterotrophic plate counts were measured on plastic polymers, whereas L. pneumophila and heterotrophic bacteria were accumulated rapidly on galvanized steel surfaces. It can be concluded that selection of plastic polymers, as a manufacturing material, are suitable for recirculating water systems.  相似文献

Natural spring water has unique properties, as it is rich in minerals that are considered to be beneficial to human health. A survey of the microbiological quality of natural spring water was conducted to assess possible risks from the consumption of the water by visitors in recreational mountain areas located in Seoul, South Korea. The densities of total coliforms and Escherichia coli were measured during the spring and the summer of 2002 to investigate the presence of coliform bacteria in the drinking spring waters. Total coliforms were detected in all samples and the mean density of total coliforms was up to a maximum of 228 CFU/mL. Detectable E. coli was found in 78% of all samples and the mean densities of E. coli varied from a minimum of 0 CFU/mL to a maximum of 15 CFU/mL in all samples. Malfunctioning septic systems and wildlife population appear to be the main source of E. coli contamination. Presence of E. coli in natural spring water indicates potential adverse health effects for individuals or populations exposed to this water. The fecal contaminated spring water may present an unacceptable risk to humans if it is used as raw drinking water.  相似文献

Although bacterial contamination is widely researched in dental unit water systems, we have been unable to find any published reports to date about the presence and distribution of sulphate-reducing bacteria (SRB) in dental unit waterlines (DUWLs). The aim of this study was to evaluate microbial contamination and to determine the presence and distribution of SRB in DUWLs. One hundred twenty-three water samples were collected from the air–water syringes, high-speed drills and water sources from 41 dental units in Istanbul, Turkey. The counts of aerobic heterotrophic bacteria and SRB were investigated in the water samples. In addition, the presence of free-living amoebae (FLA) was examined. In this work, we evaluated microbial contamination and reported for the first time the distribution of SRB in DUWLs. We determined that only ten out of 123 water samples were able to meet the American Dental Association’s limit (≤200 CFU ml − 1). SRB were observed in 102 out of 123 samples (82.9%). In addition, SRB were detected in all of the air–water syringes and high-speed drills. FLA were established in 103 out of 123 samples (83.7%).  相似文献

The analysis of total organic carbon (TOC) by the American Public Health Association (APHA) closed-tube reflux colorimetric method requires potassium dichromate (K2Cr2O7), silver sulfate (AgSO4), and mercury (HgSO4) sulfate in addition to large volumes of both reagents and samples. The method relies on the release of oxygen from dichromate on heating which is consumed by carbon associated with organic compounds. The method risks environmental pollution by discharging large amounts of chromium (VI) and silver and mercury sulfates. The present method used potassium monochromate (K2CrO4) to generate the K2Cr2O7 on demand in the first phase. In addition, miniaturizing the procedure to semi microanalysis decreased the consumption of reagents and samples. In the second phase, mercury sulfate was eliminated as part of the digestion mixture through the introduction of sodium bismuthate (NaBiO3) for the removal of chlorides from the sample. The modified method, the potassium monochromate closed-tube colorimetry with sodium bismuthate chloride removal (KMCC-Bi), generates the potassium dichromate on demand and eliminates mercury sulfate. The semi microanalysis procedure leads to a 60% reduction in sample volume and ≈?33.33 and 60% reduction in monochromate and silver sulfate consumption respectively. The LOD and LOQ were 10.17 and 33.90 mg L?1 for APHA, and 4.95 and 16.95 mg L?1 for KMCC-Bi. Recovery was between 83 to 98% APHA and 92 to 104% KMCC-Bi, while the RSD (%) ranged between 0.8 to 5.0% APHA and 0.00 to 0.62% KMCC-Bi. The method was applied for the UV-Vis spectrometry determination of COD in water and wastewater. Statistics was done by MINITAB 17 or MS Excel 2016.

Aguelmam Azizgza (LAZ) and Dayet Afourgah (DAF) are two Moroccan natural lakes located in a humid hydrographic basin of the Middle Atlas Mountains. Both are considered important reservoirs of plant and animal biodiversity. In addition, they are extensively used for recreational and fishing activities and as a water source for irrigation of agricultural crops. Recurrent cyanobacteria scum episodes in the two water bodies have been reported, Microcystis being the main genus in the scums. Here, we report on the toxic potential of three Microcystis aeruginosa strains isolated from those lakes: Mic LAZ and Mic B7 from LAZ and Mic DAF isolated from DAF. The toxic potential was checked by their microcystin (MC) content and the presence of mcy genes involved in MC synthesis. The identification and quantification of MC variants were performed by high-performance liquid chromatography-photo-diode array. The detection of mcy genes was achieved by whole-cell multiplex PCR that allowed the simultaneous amplification of DNA sequences corresponding to specific mcy regions. MC content of cultured cells, as MC-LR equivalents per gram cell biomass, was slightly higher in Mic LAZ (ca. 860) than in Mic B7 (ca. 700) and Mic DAF (ca. 690). Four MC variants were identified in the three isolates: MC-WR, MC-RR, MC-DM-WR, and MC-YR. The presence of toxic Microcystis strains in the two studied lakes may be regarded as an environmental and health hazard, especially during periods of bloom proliferation. It would be recommended the use of two complementary techniques, as those utilized herein (HPLC and mcy detection) to alert on highly probable toxicity of such lakes.  相似文献

Cryptosporidium oocysts and Giardia cysts has been isolated from waters worldwide. In Italy, studies on these parasites in the environment are still limited due to absence of epidemiological evidence and difficulty of adequate methodologies of sampling and analysis. The new Drinking Water Directive 98/83/CE states that Cryptosporidium has to be determined in water intended for human consumption if Clostridium perfringens is detected. This investigation contributes to the knowledge of both Cryptosporidium and Giardia occurrences in Italian aquatic environments through a two-year monitoring program. In addition, indicator bacteria and Clostridium perfringens were monitored in sewage, surface waters, drinking water, and swimming pools and possible correlations were calculated among all the selected parameters. Cysts and oocysts were detected in sewage and surface water and Giardia numbers always prevailed over Cryptosporidium. The parasites were not detected in drinking water, while a positive sample was obtained from the analysis of swimming pools. Pearsons correlation coefficients evidenced a reciprocal correlation between both the protozoa and the Enterococci counts.  相似文献

In late August 1994, an outbreak of human pneumonic/bubonic plague was reported in Surat, India. During the epidemic, large amounts of pesticides, such as benzene-hexachloride (BHC) and other organochlorinated compounds were used to control the vector that might have transmitted the plague. In order to evaluate the extent of contamination, both environmental samples (9 water samples and 4 soil samples) and biological samples (5 blood and urine samples) were collected and analyzed for organochlorine residues. Samples were analyzed after hexane extraction by gas chromatography equipped with electron capture detector (GC-ECD), using a PTE-5 capillary column. The results showed that the levels of 1,1-(2,2-dichloroethenylidene)-bis[4-chlorobenzene] (DDE), a major metabolite of DDT, were low in environmental samples (water, soil), while BHC levels were relatively high in one of the water sample as well as two soil samples. The BHC and DDE levels in the biological samples were not higher than those reported earlier from India, except for serum DDE. Further studies are needed to evaluate the effect of long-term exposure as the present study was based on limited samples.  相似文献

A one-year study was conducted on the chloroform concentrationof the municipal drinking water of Hyderabad City. Consumertap water samples were collected from different parts of thewater-distribution system of the city. Duplicate samplescollected from the sites were extracted with organic solvent,pentane and analysed using a gas chromatograph equipped with63Ni electron capture detector. Results indicate avariation in haloform concentration in different seasons ofthe year. A maximum concentration, ranging from 67.0–70.5g/l was observed during the summer. To verify the applicability of identifying Microcystis aeruginosa by matrix-assisted laser desorption-ionization-time-of-flight mass spectrometry (MALDI-TOF MS), mixed and field samples were employed to study the sensitivity and the analysis power, respectively. Series diluted samples and artificially mixed samples by the M. aeruginosa NIES-843 strain were designed to verify the sensitivity. The lowest detection limit was 1.955?×?106 cells in pure samples, while for mixed samples, the lowest detection limit and ratio of NIES-843 strain were 2.88?×?106 cells and 33.7%, respectively. The results provided a reference for the reasonable volume of the water sample in which the M. aeruginosa could be detected. Ribosomal protein biomarkers for identifying M. aeruginosa which were successfully detected from the field samples in Taihu Lake, indicated that the identification of M. aeruginosa by MALDI-TOF MS could be applied in field samples. Furthermore, different genetic types of M. aeruginosa strains were also detected at different locations in Taihu Lake, which revealed the diversity of M. aeruginosa and the detection power of MALDI-TOF MS at the strain level for the field samples. The sensitivity and detection power in the analysis of M. aeruginosa by the MALDI-TOF MS demonstrated the applicability of this method in routine environmental monitoring.  相似文献

The extent of contamination by endosulfans in soil samples collected from the Point Mugu watershed near Oxnard, California was determined using capillary gas chromatography/mass selective detection (GC/MSD). The study was designed to detect three organochlorinated pesticides: endosulfan , endosulfan and endosulfan sulfate. Thirteen sets of two soil samples each were taken from various sites in the region. Our results show that the endosulfan levels in these soils range from trace amounts to nearly 30 ppm, with endosulfan being the most abundant and endosulfan sulfate the least. Two sites of the study, Hueneme and Revolon and Farm and Revolon, showed high amounts of endosulfans and , with concentrations between 20 and 30 ppm. The majority of the other sites studied in this research produced concentrations of less than 10 ppm for each of the three endosulfans monitored. At five areas in particular, Pleasant Valley and Creek, Laguna and Creek, Etting and Creek, Road and Creek and Hueneme and Creek, less than 2 ppm or only trace amounts of endosulfans , and sulfate were detected.  相似文献

The concentrations of cadmium, chromium, copper, iron, lead, manganese, nickel, and zinc in water and bed sediments of river Gomti have been studied in a fairly long stretch of 500 km from Neemsar to Jaunpur. Grab samples of water (October 2002–March 2003) and bed sediments (December 2002 and March 2003) were collected from 10 different locations following the standard methods. The river water and sediment samples were processed and analyzed for heavy metals viz., Cd, Cr, Cu, Fe, Pb, Mn, Ni, and Zn, and using ICP-AES. The heavy metals found in the river water were in the range: Cd (0.0001–0.0005 mg/L); Cr (0.0015–0.0688 mg/L); Cu (0.0013–0.0.0043 mg/L); Fe (0.0791–0.3190 mg/L); Mn (0.0038–0.0.0973 mg/L); Ni (0.0066–0.011 mg/L); Pb (0.0158–0.0276 mg/L); and Zn (0.0144–0.0298 mg/L) respectively. In the sediments the same were found in the range: Cd (0.70–7.90 g/g); Cr (6.105–20.595 g/g); Cu (3.735–35.68 g/g); Fe (5051.485–8291.485 g/g); Mn (134.915–320.45 g/g); Ni (13.905–37.370 g/g); Pb (21.25–92.15 g/g); and Zn (15.72–99.35 g/g) of dry weight respectively. Some physico-chemical parameters viz., pH, total solids, total dissolved solids, total suspended solids, dissolved oxygen, biological oxygen demand, chemical oxygen demand, hardness etc. were estimated as these have direct or indirect influence on the incidence, transport and speciation of the heavy metals. Based on the geoaccumulation indices, the Gomti river sediments from Neemsar to Jaunpur are considered to be unpolluted with respect to Cr, Cu, Fe, Mn, and Zn. It is unpolluted to moderately polluted with Pb. In case of Cd it varies from moderately polluted to highly polluted. As far as Ni is concerned the sediment is very highly polluted at Barabanki and Jaunpur D/s. No correlation was found between enrichment factor and geoaccumulation index.  相似文献

Legionella pneumophila (L. pneumophila) which is also known as etiologic agent Legionnaires Disease lives in natural water and man made water systems. These bacteria belonging to Legionellaceae family are divided 15 serogroups. Phenotypical methods used for the identification of Legionella isolates are not very discriminatory. In this study we investigated genotypic features of eight L. pneumophila serogroup 1 and 18 L. pneumophila serogroup 2-14 strains isolated from different buildings in Istanbul by randomly amplified polymorphic DNA (RAPD) method. Eight L. pneumophila serogroup 1 strains (37.5%) were similar RAPD profile and they were isolated from buildings located in a short distance (about 500 m). Four L. pneumophila serogroup 2-14 strains (22%) were identical genotypically. Three of these strains were isolated from buildings located in a short distance.  相似文献

Atmospheric concentrations of nitric acid (HNO3), sulfur dioxide (SO2), particulate nitrate and particulate sulfate on the urban- and mountain-facing sides of Mt. Gokurakuji were measured from November 2002 to October 2003, in order to evaluate the effects of anthropogenic activity on air quality and dry deposited nitrate and sulfate on the surfaces of pine foliage. The results showed that HNO3, SO2 and concentrations were significantly higher (P < 0.05) on the urban-facing side (1.54, 2.48 and 0.65 μg m−3, respectively) than the mountain-facing side (0.67, 1.19 and 0.37 μg m−3, respectively), while concentrations did not differ significantly between the two sides (urban-facing: 2.80 μg m−3; mountain−facing: 2.05 μg m−3). Indirect estimates of dry deposition rates of nitrate and sulfate to the surfaces of pine foliage based on the measured concentrations approximately agreed with the measured values determined by the foliar rinsing technique in a previous study. It was found that HNO3 was the major source (approximately 80%) of dry deposited nitrate on pine foliage, while the contribution from was about equal to that from SO2. In conclusion, HNO3 and SO2 appear to be dominant species reflecting higher dry deposition rates of nitrate and sulfate on the urban-facing side compared to the mountain-facing side of Mt. In the present study, the influence of aluminium on the regeneration potential of leaf and stem-cuttings of Portulaca oleracea was studied in order to identify a terrestrial plant species as an alternate biomonitoring toolof fresh water environment. The leaves and stem cuttings of theterrestrial plant, P. oleracea grew well in the distilled water producing adventitious and lateral roots. The aluminium treated leaves and stem cuttings showed a reduction in the growthof the adventitious and lateral roots and increase in the decay of leaves and stem cuttings with increasing aluminium concentration. The tolerance index calculated for the leaves and the stem cuttings showed that the leaves were more sensitivethan stems to aluminium. Since all the parameters studied showedconcentration dependent changes, the terrestrial plant, P. oleracea can be considered a suitable biomonitoring tool of fresh water environment besides its usage in the Al toxicity testing.  相似文献

Diarrhoea remains a global public health enigma raising deep concerns for the health planners since contaminated potable water often spoils the community health structure. We hereby report a 6-year odd continuing outbreak surveillance report based on potable water indices, during which 264 water samples were screened from different districts of West Bengal, India. Samples were analysed for the presence of different enteropathogenic bacterial species by conventional molecular tools and their sensitivity to antibiotics. 78.03% samples were positive for enteropathogenic bacterial organisms and 75% samples harbored Coliform. 45.45, 12.12, and 4.16% samples were positive for E.coli, V. cholerae, and V. mimicus, respectively. Diarrhoeagenic E.coli 7 EPEC, 10 ETEC, and 2 EIEC were isolated along with 2 V. cholerae O1 Ogawa (ctxA and tcpA ElTor positive), one each from tube well and pond. Interestingly, 4 V.cholerae non-O1/non-O139 also harbored hlyA gene. The detection of toxin genes among this bacterial pool of sampled water indicates the fallout of the potable water sources, thus enabling us to establish that it is none other than the contaminated potable water system which often wreaks havoc in the south Bengal diarrhoeal menace. The consequences are further complicated by the presence of drug-resistant pathogenic bacterial pool to fluoroquinolone, beta-lactams, and cephalosporins, in the accessible potable water, with threats of outbreaks exploding into an epidemic, given suitable environment, poor sanitation, and unhygienic practices. Therefore, we strongly recommend re-modelling of ‘point-of-use water disinfection’ measures and adequate personal hygiene for healthier community life.  相似文献

Gomti river receives industrial as well as domestic wastes from various drains of Lucknow city. In the process the water and sediment of the river Gomti get contaminated with heavy metals and other pollutants. In the present study, impacts of domestic/industrial wastes on the water and sediment chemistry of river Gomti with special reference to heavy metals have been investigated in different seasons (summer, winter and rainy). For this, seven sampling sites: Gaughat, Mohan Meakin, Martyrs Memorial, Hanuman Setu, Nishatganj bridge, Pipraghat and Malhaur, in the river Gomti in Lucknow region were identified and samples of water and sediments were collected in all the three seasons. In the collected water and sediment samples, six metals (Cd, Cr, Cu, Ni, Pb, and Zn) were analyzed on ICP-AES (Inductively coupled plasma emission spectroscopy) Labtam Plasmalab 8440. High concentrations of all the metals were noticed in water and sediment in rainy season compared to summer and winter. Because in rainy season runoff from open contaminated sites, agricultural field and industries, directly comes into the river without any treatment. In both the cases, the concentration of zinc was maximum (0.091 g/ml in water and 182.13 g/g in sediment) and the concentration of cadmium (0.001 g/ml in water and 17.26 g/g in sediment) was minimum. Higher concentration of metal in water and sediment during rainy season could be due to the industrial/agricultural/domestic runoff coming into the river.  相似文献
