Biomax® is an aliphatic-aromatic polyester. The biodegradability of Biomax® was studied at 58 °C using a laboratory scale bioreactor. The bioreactor was inoculated with bacteria derived from compost and supplemented with powdered Biomax® and an additional energy source. After a period of acclimation, the microorganisms in the bioreactor were capable of metabolizing the major components of the polymer, i.e., TPA and ethylene glycol. TPA and ethylene glycol were detected in the bioreactor only when they were added. Degradation and disintegration of the powdered Biomax® was monitored by laser diffraction. The particle size distribution of the powdered polymer progressively shifted toward smaller sizes until the diameters of the polymer particles were indistinguishable from bacteria. The types of microbes in the bioreactor were determined by analyzing 16S rRNA gene sequences. The bacteria belonged to 35 different groups, and the majority of the bacteria appeared to represent new species.  相似文献

The distribution of degading microorganisms of high molecular weight poly(-propiolactone) (PPL), whose individual structural units are similar to those of poly(-hydroxybutyrate) (PHB) and poly(€-caprolactone) (PCL), was examined. Despite the fact that PPL is a chemosynthetic polymer, many kinds of PPL-degrading microorganisms were found to be distributed as resident populations widely in natural environments. A total of 77 strains of PPL-degrading microorganisms was isolated. From standard physiological and biochemical tests, at least 41 strains were referred to as Bacillus species. Microbial degradation of fibrous PPL proceeded rapidly in some enrichment cultures but was not as complete as that of PHB. Most of the isolated PPL-degrading microorganisms were determined to be PCL degraders and/or PHB degraders. Therefore, it can be assumed that mostly PPL is recognized by the microorganisms as PHB or another natural substrate of the same type as which PCL is regarded. Microbial degradation of PPL was confirmed by some Bacillus strains from type culture collections. The similarity of microbial degradation between PPL and PCL was found to be very close.  相似文献

Two pilot tests of an aerobic in situ bioreactor (ISBR) have been conducted at field sites contaminated with petroleum hydrocarbons. The two sites differed with respect to hydrocarbon concentrations. At one site, concentrations were low but persistent, and at the other site concentrations were high enough to be inhibitory to biodegradation. The ISBR unit is designed to enhance biodegradation of hydrocarbons by stimulating indigenous microorganisms. This approach builds on existing Bio‐Sep® bead technology, which provides a matrix that can be rapidly colonized by the active members of the microbial community and serves to concentrate indigenous degraders. Oxygen and nutrients are delivered to the bioreactor to maintain conditions favorable for growth and reproduction, and contaminated groundwater is treated as it is circulated through the bed of Bio‐Sep® beads. Groundwater moving through the system also transports degraders released from Bio‐Sep® beads away from the bioreactor, potentially increasing biodegradation rates throughout the aquifer. Groundwater sampling, Bio‐Traps, and molecular biological tools were used to assess ISBR performance during the two pilot tests. Groundwater monitoring indicated that contaminant concentrations decreased at both sites, and the microbial data suggested that these decreases were due to degradation by indigenous microorganisms rather than dilution or dispersion mechanisms. Taken together, these lines of evidence showed that the ISBR system effectively increased the number and activity of indigenous microbial degraders and enhanced bioremediation at the test sites. © 2013 Wiley Periodicals, Inc.  相似文献

The fertilizer properties of anaerobic digestate depend on the feedstock and operating conditions of digestion. In this study, the comparative fertilizer properties of mesophilic and thermophilic digestates from dairy manure were evaluated for plant nutrient contents, and special attention was paid to plant growth promoting bacteria (PGPB). Two digestates contained similar plant nutrient contents, while the thermophilic digestate contained higher contents of NH4+–N. The quantity of Bacillus and Pseudomonas in the mesophilic digestate was significantly higher than in the thermophilic digestate. Furthermore, Bacillus showed siderophore production and antifungal activity (43.5–75.3%), and Pseudomonas showed siderophore and phytohormone production (4.2–75.2 µg ml?1). One phosphate solubilizing isolate was also detected in the mesophilic digestate. These results indicated that two digestates showed different fertilizer properties with respect to nutrient contents and PGPB, and digestates had the potential to increase the availability of phosphorus and iron in the soil, both to provide phytohormones to plant roots and protect plants from fungal phytopathogens. The contents of indicator bacteria and heavy metals were analyzed to determine their environmental risk, and the results showed a high reduction in indicator bacteria and lower levels of heavy metals than in other feedstocks.  相似文献

The application of renewable raw materials in the automotive sector became more and more interesting in the last years. For the first time, a material system based on natural fibers and PTP®, a vegetable based thermoset resin, was produced and manufactured into a bus body component by Sheet-Moulding-Compound-technology. The Life Cycle Assessment on this component based on renewable raw materials shows advantages especially in the categories fossil resources and global warming. It can be expected that further concepts of development and optimization lead to a more efficient use of materials and so to an additional reduction of environmental impacts.  相似文献

The current study is interested in evaluating the decay of cotton, Whatman and chemical pulp caused by Trichoderma harzianum and Paecilomyces variotii. The structural changes of the paper were evaluated by Infrared Spectroscopy (FTIR) and Scanning Electron Microscope (SEM). The SEM results show differences in hyphae colonization and paper decay patterns between studied species under the current study; P. variotii caused an eroded structure in the cotton (cavity forming), whereas the initial T. harzianum colonization produced rupture and erosion (soft-rot decay type II) for the three types of paper ,the gaps were elongated with sharp pointed ends, which consisted either of individual cavities or in chains. Moreover, FTIR results confirmed that there a relationship could be observed between fungal decay and crystalline cellulose content because the intensity of peaks at 1335 and 1111 cm?1 significantly decreased due to the fungal decay. Furthermore, the intensity of O–H stretching absorption slightly decreased, and this may be attributed to hydrolysis of cellulose molecules.  相似文献

Over a hundred of halophilic/halotolerant microorganisms were screened for alkaline protease production. The bacterium showing the highest enzyme production was characterized and identified as Bacillus halodurans US193 on the basis of 16S rRNA gene analysis. It was alkalophilic, thermophilic and halotolerant since it grew optimally at pH 9.7 and 50?°C with tolerance of up to 125 g NaCl l?1. The alkaline protease was purified 4.9 times with about 40186.1 U/mg as specific activity. It exhibited optimal activity at pH 10, 70?°C and 0.25 M NaCl with perfect stability at wide ranges of pH (6–12), temperatures (30–60?°C) and NaCl concentrations (0–2 M). The serine alkaline protease maintained high stability in the presence of Cu2+, Mg2+, Ba2+ and Ca2+ ions, various organic solvents [50% (v/v)] and ionic and non ionic detergent additives. In addition, it was more compatible with various commercialized detergents than other reported detergent proteases, and was very efficient in blood stain removal. These findings let B. halodurans US193 alkaline protease be an ideal candidate for many industrial processes at harsh conditions, especially as a bio-additive in detergent industry.  相似文献

The effects of adding biosolids to a green waste feedstock (100% green waste, 25% v/v biosolids or 50% biosolids) on the properties of composted products were investigated. Following initial composting, 20% soil or 20% fly ash/river sand mix was added to the composts as would be carried out commercially to produce manufactured soil. Temperatures during composting reached 50 °C, or above, for 23 days when biosolids were included as a composting feedstock but temperatures barely reached 40 °C when green waste alone was composted. Addition of biosolids to the feedstock increased total N, EC, extractable NH4, NO3 and P but lowered pH, macroporosity, water holding capacity, microbial biomass C and basal respiration in composts. Additions of soil or ash/sand to the composts greatly increased the available water holding capacity of the materials. Principal component analysis (PCA) of PCR-DGGE 16S rDNA amplicons separated bacterial communities according to addition of soil to the compost. For fungal ITS-RNA amplicons, PCA separated communities based on the addition of biosolids. Bacterial species richness and Shannon’s diversity index were greatest for composts where soil had been added but for fungal communities these parameters were greatest in the treatments where 50% biosolids had been included. These results were interpreted in relation to soil having an inoculation effect and biosolids having an acidifying effect thereby favouring a fungal community.  相似文献

Hydrolytic, enzymatic degradation and composting under controlled conditions of series of triblock PCL/PEO copolymers, PCEC, with central short PEO block (M n 400 g/mol) are presented and compared with homopolymer (PCL). The PCEC copolymers, synthesized via ring-opening polymerization of ε-caprolactone, were characterized by 1H NMR, quantitative 13C NMR, GPC, DSC and WAXS. The introduction of the PEO central segment (<?2 wt%) in PCL chains significantly affected thermal degradation and crystallization behavior, while the hydrophobicity was slightly reduced as confirmed by water absorption and moisture uptake experiments. Hydrolytic degradation studies in phosphate buffer after 8 weeks indicated a small weight loss, while FTIR analysis detected changes in crystallinity indexes and GPC measurements revealed bulk degradation. Enzymatic degradation tested by cell-free extracts containing Pseudomonas aeruginosa PAO1 confirmed high enzyme activity throughout the surface causing morphological changes detected by optical microscopy and AFM analysis. The changes in roughness of polymer films revealed surface erosion mechanism of enzymatic degradation. Copolymer with the highest content of PEO segment and the lowest molecular weight showed better degradation ability compared to PCL and other copolymers. Furthermore, composting of polymer films in a model compost system at 37 °C resulted in significant degradation of the all synthesized block copolymers.  相似文献

Channa argus, a type of snakehead fish native to China, is a popular food fish in certain Asian countries but is a known destructive invasive species in the US. In this study, the two collagens, i.e. acid-soluble collagen (ASC) and pepsin-solubilized collagen (PSC), were obtained from C. argus skin. The yield of ASC was 28.0% and that of PSC was 16.8% on the dry bases. The collagens were identified as the collagen of type I by SDS–PAGE patterns. The Tds were approximately 27.0?°C. Similar ultraviolet spectra of both collagens were observed. Fourier Transform infrared spectra indicated PSC structure had a little change due to the loss of terminal domains by pepsin digestion. The results of XRD proved that the two collagens retained their helical structures. The results suggest that the collagens isolated from C. argus can potentially be alternative sources of vertebrate collagens for use in the food and other industries.  相似文献

The global demand of bioplastics has lead to an exponential increase in their production commercially. Hence, biodegradable nature needs to be evaluated in various ecosystems viz. air, water, soil and other environmental conditions to avoid the polymeric waste accumulation in the nature. In this paper, we investigated the progressive response of two indigenously developed bacterial consortia, i.e., consortium-I (C-I: Pseudomonas sp. strain Rb10, Pseudomonas sp. strain Rb11 and Bacillus sp. strain Rb18), and consortium-II (C-II: Lysinibacillus sp. strain Rb1, Pseudomonas sp. strain Rb13 and Pseudomonas sp. strain Rb19), against biodegradation behavior of polyhydroxybutyrate (PHB) film composites, under natural soil ecosystem (in net house). The biodegraded films recovered after 6 and 9 months of incubation were analyzed through Fourier transform infrared spectroscopy and scanning electron microscopy to determine the variations in chemical and morphological parameters (before and after incubation). Noticeable changes in the bond intensity, surface morphology and conductivity were found when PHB composites were treated with C-II. These changes were drastic in case of blends in comparison to copolymer. The potential isolates not only survived, but, also, there was a significant increase in bacterial diversity during whole period of incubation. To the best of our knowledge, it is the first report which described the biodegradation potential of Lysinibacillus sp. as a part of C-II with Pseudomonas sp. against PHB film composites.  相似文献

Cellulose gel films were prepared by regeneration process using pre-cooled aq.(8 wt% LiOH + 15 wt% urea) mixture as solvent and ethyl alcohol as non solvent. The Terminus cattapa leaf extract diffused wet cellulose films were then dipped in 1–5 mM aq.AgNO3 solutions to allow in situ generation of silver nanoparticles (AgNPs). Besides the in situ generation, some AgNPs were also formed outside the wet films in the solution. The AgNPs formed outside the films were observed under transmission electron microscope and scanning electron microscope. The nanocomposite films were also characterized by Fourier transform infrared spectroscopy, X-ray diffraction and thermogravimetric analysis and tensile test. The thermal stability of the composite films was lower than that of the matrix up to a temperature of ~300 °C and afterwards showed a reverse trend. The tensile strength of the nanocomposite films was found to be higher than the matrix but decreased with increasing concentration of aq.AgNO3. The cellulose/AgNPs composite films showed good antibacterial activity against E. coli (gram positive) and Bacillus sp. (gram negative). Based on the aforementioned properties, the cellulose/AgNPs composite films can be considered for antibacterial packaging and medical applications.  相似文献

Two thermophilic digesters were inoculated with manure and started-up under mixed and stagnant conditions. The Archaea in the mixed digester (A) were dominated by hydrogenotrophic Methanobateriaceae (61%) with most of the methane being produced via syntrophic pathways. Methanosarcinales (35%) were the only acetoclastic methanogens present. Acetate dissipation seems to depend on balanced hydrogenotrophic-to-acetotrophic abundance, which in turn was statistically correlated to free ammonia levels. Relative abundance of bacterial community was associated with the loading rate. However, in the absence of mixing (digester B), the relationship between microbial composition and operating parameters was not discernible. This was attributed to the development of microenvironments where environmental conditions are significantly different from average measured parameters. The impact of microenvironments was accentuated by the use of a non-acclimated seed that lacks adequate propionate degraders. Failure to disperse the accumulated propionate, and other organics, created high concentration niches where competitive and inhibiting conditions developed and favored undesired genera, such as Halobacteria (65% in B). As a result, digester B experienced higher acid levels and lower allowable loading rate. Mixing was found necessary to dissipate potential inhibitors, and improve stability and loading capacity, particularly when a non-acclimated seed, often lacking balanced thermophilic microflora, is used.  相似文献

The effect of phenol concentration on phenol biodegradation at an industrial site in the south of Wales, United Kingdom, was investigated using standard Bio‐Sep® Bio‐Traps® and Bio‐Traps® coupled with stable isotope probing (SIP). Unlike many 13C‐amendments used in SIP studies (such as hydrocarbons) that physically and reversibly adsorb to the activated carbon component of the Bio‐Sep® beads, phenol is known to irreversibly chemisorb to activated carbon. Bio‐Traps® were deployed for 32 days in nine site groundwater monitoring wells representing a wide range of phenol concentrations. Bio‐Traps® amended with 13C‐phenol were deployed together with non‐amended Bio‐Traps® in three wells. Quantitative polymerase chain reaction (qPCR) analysis of Bio‐Traps® post‐deployment indicated an inhibitory effect of increasing phenol concentration on both total eubacteria and aerobic phenol‐utilizing bacteria as represented by the concentration of phenol hydroxylase gene. Despite the chemisorption of phenol to the Bio‐Sep® beads, activated carbon stable isotope analysis showed incorporation of 13C into biomass and dissolved inorganic carbon (DIC) in each SIP Bio‐Trap® indicating that chemisorbed amendments are bioavailable. However, there was a clear effect of phenol concentration on 13C incorporation in both biomass and DIC confirming phenol inhibition. These results suggest that physical reductions of the phenol concentrations in some areas of the plume will be required before biodegradation of phenol can proceed at a reasonable rate. © 2013 Wiley Periodicals, Inc.  相似文献

Partly because of the low bioavailability of metals, the soil cleaning-up using phytoremediation is usually time-consuming. In order to enhance the amount of metals at the plant's disposal, the soil bioaugmentation coupled together with phytoextraction is an emerging technology. In this preliminary work, two agricultural soils which mainly differed in their Cr, Hg and Pb contents (LC, low-contaminated soil; HC, high-contaminated soil) were bioaugmented in laboratory conditions by either bacterial (Bacillus subtilis, Pseudomonas aeruginosa, Pseudomonas fluorescens or Ralstonia metallidurans) or fungal inocula (Aspergillus niger or Penicillium simplicissimum) and incubated during three weeks. The LC soil pots bioaugmented with A. niger and P. aeruginosa contained higher concentrations of Cr (0.08 and 0.25 mg.kg−1 dw soil) and Pb (0.25 and 0.3 mg.kg−1 dw soil) in the exchangeable fraction F1 (extraction with MgCl2) by comparison with the non-bioaugmented soil where neither Cr nor Pb was detected. Conversely, immobilization of Cr and Pb in the soil were observed with the other microorganisms. The soil bioaugmentation not only modified the metal speciation for the most easily extractable fractions but also modified the distribution of metals in the other fractions, to a lesser extent nevertheless. The difference in microbial concentrations between the bioaugmented or not HC soils reached up to 1.8 log units. Thus the microorganisms that we chose for the soil bioaugmentation were competitive towards the indigenous microflora. The PCA analysis showed close positive relationships between the microorganisms which potentially produced siderophores in the soil and the amount of Cr and Pb in the fraction F1.  相似文献

15N-labeling and solid-state 13C and 15N nuclear magnetic resonance (NMR) spectroscopy was applied to study the immobilization of 2,4,6 trinitrotoluene (TNT) into soil organic matter (SOM). Uncontaminated soil from the Ap horizon of a Luvisol was mixed with 15N-TNT (enrichment: 99 atm%) and laid over an unspiked layer of the same material. The latter covered soil from the Bt horizon. The microcosms were aerobically incubated under laboratory conditions for up to 11 months. After 1 week, within the total microcosm approximately 90% of the added 15N (15Nadd) were recovered, mostly in the top layer (87%). After 11 months, this amount decreased to 71%, indicating losses due to denitration or transamination. Within two months, half of 15Nadd had been immobilized in the residues not extractable with organic solvents and water. The amount of the sequestered 15Nadd remained fairly constant until the end of the experiment pointing towards a high stability of TNT-SOM associates. Solid-state 15N NMR revealed their formation by covalent binding, most tentatively as amides. Complete reduction of TNT to triaminotoluene (TAT) was not prerequisite. The most pronounced downwards movement of 15N-TNT occurred during the first two months. The major part of it, however, experienced quick immobilization, leaving approximately 10% of 15Nadd recovered in the leachate at the end of the experiment. Calculations indicated contributions of inorganic 15Nadd. Approximately 25% of its organic 15Nadd originated from condensed N, suggesting that in soils the transport of partly reduced TNT is in close association with the organic matter of the soil solution to which they are covalently bound.  相似文献

The size and isotopic behavior of sulfur pools in210Pb-dated peat cores were investigated to obtain aninsight into retention mechanisms of pollutant S in twomountain-top peatlands of the Northern Czech Republic, CentralEurope. The bogs were situated 40 km apart in an area whichbetween the years 1985 and 1995 received as much as 130 kg Sha-1 yr-1 from the atmosphere. Vertical peataccretion was faster at Pod Jelení horou (JH) than atVelký moál (VM). Organic carbon-bonded S was themost abundant sulfur pool, constituting 77 and 65 wt. % at JHand VM, respectively. At JH both the S concentration maximumand the highest annual S deposition rate were displaceddownward by more than 20 years (from 1987 to the 1960s)indicating that the buried S is vertically mobile. At VM the Sconcentration was the highest in the topmost 2-cm section eventhough atmospheric S deposition peaked in 1987. Differentmechanisms of S isotope redistribution prevailed in thetopmost peat layers at JH, where a negative 34Sshift occurred, and at VM, where a positive 34Sshift occurred. Bacterial sulfate reduction was responsiblefor the negative 34S shift at JH. One possibleexplanation of the positive 34S shift at VM isrelease of 32S-enriched products of mineralization duringpeat diagenesis. There was a strong positive correlationbetween the abundance of total and pyrite S along the profiles.The presence of pyrite S at VM (526 ± 60 ppm) suggestedthat even at VM bacterial sulfate reduction occurred. Ananaerobic incubation of JH peat indicated sulfate reductionrate of 600 nmol g-1 day-1. The turnover times forinorganic S pools were shorter than for the organic S pools.Cumulative S contents in the Czech peat bogs were found to besignificantly lower than in similar sites in the NortheasternU.S., even though the atmospheric S inputs were more thanthree times higher at the Czech sites. Possible causes of suchdiscrepancy are discussed.  相似文献

The polyester amide BAK 1095 is fully degraded by microorganisms. Bacteria that can grow on this polymer were isolated from various environments, such as freshwater, saltwater, arable land, woodland and compost. The mechanism of polymer degradation was examined using selected isolates. It was discovered that the bacteria selectively cleave the polymer at its ester bonds, releasing low-molecular water-soluble oligoamides. These can then be metabolised by other microorganisms [1, 2, 3]. The isolates were also tested for their ability to break down other ester-containing polymers such as Degranil W 50, a linear polyester urethane urea. Eight of the 12 strains examined were also able to break down this polymer.The isolated microorganisms predominantly belong to the genus Bacillus. Mesophilic, halophilic and thermophilic species were isolated. The studies show that polyester amide-degrading bacteria occur in numerous ecosystems and provide data on the microbial breakdown of random copolymers. Information was also obtained on the mechanism involved in the microbial breakdown of polyester amides.  相似文献

Point Pelee National Park (PPNP) is highly contaminated with dichlorodiphenyltrichloroethane (DDT) and dieldrin due to the historical use of these two persistent organochlorine pesticides. Zero‐valent iron (ZVI) technology with and without amendments has been successfully used in the past to promote organochlorine pesticides degradation in several locations in North America and Europe. In this study, the use of two commercially available ZVI products, DARAMEND® and EHC®, to promote DDT and dieldrin degradation in PPNP's soil and groundwater were investigated. DARAMEND® was applied to PPNP's soil in a laboratory experiment and in an in situ pilot‐scale plot. In both cases, DARAMEND® did not significantly increase DDT or dieldrin degradation in treated soils. The effectiveness of EHC® was tested in a laboratory experiment that simulated the park's groundwater environment using PPNP's pesticide contaminated soil. The result was consistent with the one reported for DARAMEND®, in that there was no significant increase in DDT or dieldrin degradation in any of the samples treated with EHC®. These results demonstrate that both of these ZVI commercially available products are not suitable for in situ remediation at PPNP.  ©2017 Wiley Periodicals, Inc.  相似文献

Composting has been recognized as one of the most cost effective and environmentally sound alternatives for organic wastes recycling from long and composted wastes have a potential to substitute inorganic fertilizers. We investigated the potential of composted tannery sludge for ornamental purposes and to examine the effects of two different composts and concentrations on ornamental Capsicum growth. The two composts were produced with tannery sludge and the composition of each compost was: compost1 of tannery sludge (C1TS) – tannery sludge + sugarcane straw and cattle manure mixed in the ratio 1:3:1 (v:v:v); compost2 of tannery sludge (C2TS) – tannery sludge + “carnauba” straw and cattle manure in the ratio 1:3:1 (v:v:v). Each compost was amended with soil at rates (% v:v) of 0%, 25%, 50%, 75% and 100% (designation hereafter as T1T5, respectively). The number of leaves and fruits were counted, and the stem length was also measured. Chlorophyll content was recorded on three leaves of each harvested plant prior to harvest. Number of leaves and fruits, stem length, dry weight of shoot and roots did not vary significantly between the plants grown in two tannery composts. All the treatments with composted tannery sludge application (T2T5) significantly increased the number of leaves and fruits, stem length and chlorophyll content compared with the control (T1). The chlorophyll content was higher in plants growing in the C1TS compared to C2TS. The results of the present study further suggest that Capsicum may be a good option to be grown on composted tannery amended soil.  相似文献
