Recharge of waste water in an unconsolidated poorly sorted alluvial aquifer is a complex process, both physically and hydrochemically. The aim of this paper is to analyse and conceptualise vertical transport mechanisms taking place in an urban area of extensive wastewater infiltration by analysing and combining the water balance, the microbial (Escherichia coli) mass balance, and the mass balance for dissolved solutes. For this, data on sediment characteristics (grain size, organic carbon, reactive iron, and calcite), groundwater levels, and concentrations of E. coli in groundwater and waste water were collected. In the laboratory, data on E. coli decay rate coefficients, and on bacteria retention characteristics of the sediment were collected via column experiments. The results indicated that shallow groundwater, at depths of 50 m below the surface, was contaminated with E. coli concentrations as high as 10(6) CFU/100 mL. In general, E. coli concentrations decreased only 3 log units from the point of infiltration to shallow groundwater. Concentrations were lower at greater depths in the aquifer. In laboratory columns of disturbed sediments, bacteria removal was 2-5 log units/0.5 cm column sediment. Because of the relatively high E. coli concentrations in the shallow aquifer, transport had likely taken place via a connected network of pores with a diameter large enough to allow bacterial transport instead of via the sediment matrix, which was inaccessible for bacteria, as was clear from the column experiments. The decay rate coefficient was determined from laboratory microcosms to be 0.15 d(-1). Assuming that decay in the aquifer was similar to decay in the laboratory, then the pore water flow velocity between the point of infiltration and shallow groundwater, coinciding with a concentration decrease of 3 log units, was 0.38 m/d, and therefore, transport in this connected network of pores was fast. According to the water balance of the alluvial aquifer, determined from transient groundwater modelling, groundwater flow in the aquifer was mainly in vertical downward direction, and therefore, the mass balance for dissolved solutes was simulated using a 1D transport model of a 200 m column of the Quaternary Alluvium aquifer. The model, constructed with PHREEQC, included dual porosity, and was able to adequately simulate removal of E. coli, cation-exchange, and nitrification. The added value of the use of E. coli in this study was the recognition of relatively fast transport velocities occurring in the aquifer, and the necessity to use the dual porosity concept to investigate vertical transport mechanisms. Therefore, in general and if possible, microbial mass balances should be considered more systematically as an integral part of transport studies.  相似文献

Changes in aerobic bacteria and autotrophic nitrifier numbers, and in respiration and nitrification in two soils treated with cinosulfuron at 42 (field rate) and 4200 microg/kg were studied after 1 and 4 weeks of incubation under laboratory conditions. Only nitrification at 1 week was slightly inhibited by the cinosulfuron treatment, even at the field rate. In vitro toxicity tests carried out in agar media on representative aerobic bacteria, fungi and Azotobacter strains isolated from the two soils, as well as on nine collection soil bacteria, showed that only a very high cinosulfuron concentration (100 mg/l) can have negative effects on the growth of a limited number of soil heterotrophic microorganisms, under conditions similar to those of soil environment. The absence of three branched-chain amino acids increased bacterial sensitivity, thus showing the importance of the chemical conditions and suggesting acetolactate synthase enzyme blockage as the toxicity mechanism. It is concluded that cinosulfuron has a negative effect on only a few aspects of the microbial community in soil ecosystems, even at concentrations higher that those currently in use.  相似文献

Tree nurseries and greenhouses within the USDA red imported fire ant (RIFA) quarantine zone are required to incorporate insecticides into their potting media to prevent artificial spread of RIFA. Bifenthrin and fipronil are two common insecticides that are incorporated into potting media. During irrigation and stormwater events, there is potential for insecticides to leach from nursery pots, resulting in the contamination of nearby surface waters. In this study, occurrences of insecticides in simulated nursery runoff were compared with two irrigation strategies and two types of containers in single pot leaching and field runoff simulations. In addition, toxicity of pot leachate to the aquatic invertebrate, Hyallela azteca, was measured, and removal efficiencies of insecticides from bioretention cell media were evaluated. Overhead irrigation resulted in significantly higher concentrations than drip irrigation, and RootMaker® pots allowed more leaching as compared to standard slick-wall pots. However, in all tests, the average concentration of bifenthrin during 15 days of leaching in both pot and field simulations was greater than 200 ng/L; more than 100-fold greater than the LC50 for H. azteca. Toxicity studies confirmed this level of toxicity. Higher amounts of compost, 20 and 40 %, in bioretention cell media resulted in greater percent reduction of both bifenthrin and fipronil. This study determined that management techniques may be able to limit the amount of insecticide that leaches from pots and runs off to receiving water bodies. Specifically, the selection of appropriate pot types, irrigation strategies, or filtering runoff through bioretention cells may reduce contamination loads. Thus, further best management strategies such as the use of bioretention cells are needed in nursery and greenhouse facilities to prevent surface water runoff from transporting toxic insecticides.  相似文献

The type of bacteria, bacterial cell concentration, initial urea concentration, reaction temperature, the initial Ca(2+) concentration, ionic strength, and the pH of the media are some factors that control the activity of the urease enzyme, and may have a significant impact on microbial carbonate precipitation (MCP). Factorial experiments were designed based on these factors to determine the optimum conditions that take into consideration economic advantage while at the same time giving quality results. Sporosarcina pasteurii strain ATCC 11859 was used at constant temperature (25°C) and ionic strength with varying amounts of urea, Ca(2+), and bacterial cell concentration. The results indicate that the rate of ureolysis (k(urea)) increases with bacterial cell concentration, and the bacterial cell concentration had a greater influence on k(urea) than initial urea concentration. At 25 mM Ca(2+) concentration, increasing bacterial cell concentration from 10(6) to 10(8)cells mL?1 increased the CaCO(3) precipitated and CO(2) sequestrated by over 30%. However, when the Ca(2+) concentration was increased 10-fold to 250 mM Ca(2+), the amount of CaCO(3) precipitated and CO(2) sequestrated increased by over 100% irrespective of initial urea concentration. Consequently, the optimum conditions for MCP under our experimental conditions were 666 mM urea and 250 mM Ca(2+) at 2.3×10? cells mL?1 bacterial cell concentration. However, a greater CaCO(3) deposition is achievable with higher concentrations of urea, Ca(2+), and bacterial cells so long as the respective quantities are within their economic advantage. X-ray Diffraction, Scanning Electron Microscopy and Energy Dispersive X-ray analyzes confirmed that the precipitate formed was CaCO(3) and composed of predominantly calcite crystals with little vaterite crystals.  相似文献

Bioretention systems have been implemented as stormwater best management practices (BMPs) worldwide to treat non-point sources pollution. Due to insufficient research, the design guidelines for bioretention systems in tropical countries are modeled after those of temperate countries. However, climatic factors and stormwater runoff characteristics are the two key factors affecting the capacity of bioretention system. This paper reviews and compares the stormwater runoff characteristics, bioretention components, pollutant removal requirements, and applications of bioretention systems in temperate and tropical countries. Suggestions are given for bioretention components in the tropics, including elimination of mulch layer and submerged zone. More research is required to identify suitable additives for filter media, study tropical shrubs application while avoiding using grass and sedges, explore function of soil faunas, and adopt final discharged pollutants concentration (mg/L) on top of percentage removal (%) in bioretention design guidelines.

通过序批实验和土柱淋洗实验研究了由生物表面活性剂鼠李糖脂(RL)与非离子化学表面活性剂十二烷基醇聚氧乙烯(6)醚(POE(6))混合得到的复配试剂洗脱污染土壤中多氯联苯(PCBs)的作用效果及其作用机理。结果表明,同一土样中,从批实验得到的PCBs洗脱率略高于土柱淋洗实验。RL与POE(6)两种单一试剂对人工污染土样中的PCBs洗脱率均大于60%,而对陈化土样中的洗脱率均不到20%,且土壤中的TOC含量越高,PCBs的洗脱率越低。在质量浓度为301 mg/L(10 CMC)及1 505 mg/L(50 CMC)的RL-POE(6)复配试剂中,RL与POE(6)对人工污染土壤中PCBs的洗脱具有一定的协同作用。当复配试剂的浓度为301 mg/L时,RL与POE(6)对陈化土样中PCBs的洗脱没有协同作用;但当RL-POE(6)的浓度增加到1 505 mg/L时,RL与POE(6)对陈化土样中PCBs的洗脱具有明显的协同作用。  相似文献

In bioretention systems used for stormwater treatment, runoff interception improves with increased infiltration rates. However, nitrogen retention improves with increased retention time or decreasing infiltration rates. These contrasting responses were analyzed in 240-L experimental mesocosms using a variety of media treatments. The mesocosms were vegetated, except for one barren control. Dual-stage outlets were installed to extend retention time and equalize hydraulic responses. One unregulated treatment was free-draining. This part 1 paper presents the media properties and hydraulic responses. The highly aggregated media had saturated hydraulic conductivities ranging from 20.7 to 59.6 cm/h in August 2008 (austral winter), which increased to 42.8 to 110.6 cm/h in March 2009 (austral summer). The outlet regulated mesocosms provided retention over 8 times longer than the free-draining mesocosms, while still being able to capture large events. The outlets provide adaptive management for bioretention design to improve both runoff capture and nitrogen retention.  相似文献

This study investigated the effectiveness of bioretention as a stormwater management practice using repetitive bioretention columns for phosphorus removal. Bioretention media, with a higher short-term phosphorus sorption capacity, retained more phosphorus from infiltrating runoff after 3 mg/L phosphorus loading. A surface mulch layer prevented clogging after repetitive total suspended solids input. Evidence suggests that long-term phosphorus reactions will regenerate active short-term phosphorus adsorption sites. A high hydraulic conductivity media overlaying one with low hydraulic conductivity resulted in a higher runoff infiltration rate, from 0.51 to 0.16 cm/min at a fixed 15-cm head, and was more efficient in phosphorus removal (85% mass removal) than a profile with low conductivity media over high (63% mass removal). Media extractions suggest that most of the retained phosphorus in the media layers is available for vegetative uptake and that environmental risk thresholds were not exceeded.  相似文献

Twenty-eight bacterial and Br transport experiments were performed in the field to determine the effects of physical and chemical heterogeneity of the aquifer sediment. The experiments were performed using groundwater from two field locations to examine the effects of groundwater chemistry on transport. Groundwater was extracted from multilevel samplers and pumped through 7-cm-long columns of intact sediment or repacked sieved and coated or uncoated sediment from the underlying aquifer. Two bacterial strains, Comamonas sp. DA001 and Paenibacillus polymyxa FER-02, were injected along with Br into the influent end of columns to examine the effect of cell morphology and cell surface properties on bacterial transport. The effects of column sediment grain size and mineral coatings coupled with groundwater geochemistry were also investigated. Significant irreversible attachment of DA001 was observed in the Fe oxyhydroxide-coated columns, but only in the suboxic groundwater where the concentrations of dissolved organic carbon (DOC) were ca. 1 ppm. In the oxic groundwater where DOC was ca. 8 ppm, little attachment of DA001 to the Fe oxyhydroxide-coated columns was observed. This indicates that DOC can significantly reduce bacterial attachment due electrostatic interactions. The larger and more negatively charged FER-02 displayed increasing attachment with decreasing grain size regardless of DOC concentration, and modeling of FER-02 attachment revealed that the presence of Fe and Al coatings on the sediment also promoted attachment. Finally, the presence of Al coatings and Al containing minerals appeared to significantly retard the Br tracer regardless of the concentration of DOC. These findings suggest that DOC in shallow oxic groundwater aquifers can significantly enhance the transport of bacteria by reducing attachment to Fe, Mn and Al oxyhydroxides. This effect appears to be profound for weakly and strongly charged hydrophilic bacteria and may contribute to differences in observations between laboratory experiments versus field-scale investigations particularly if the groundwater pH remains subneutral and Fe oxyhydroxide phases exist. These observation validate the novel approach taken in the experiments outlined here of performing laboratory-scale experiments on site to facilitate the use of fresh groundwater and thus be more representative of in situ groundwater conditions.  相似文献

Key to managing heavy metals in bioretention is to understand their fates in bioretention facilities. In this study, pot prototypes filled with bioretention media were built to simulate the conditions of natural growth of plants. Synthetic runoff with different heavy metal loadings (copper, cadmium, lead, and zinc) was periodically applied. Metal accumulations in tissues of grasses -Panicum virgatum, Kentucky-31, and Bromus ciliatus, were investigated after 230d of growth and multiple runoff treatment events. After 183d of periodic runoff application, the concentrations of Zn, Cu, Pb and Cd with low and high loadings had the same trends in the plant tissues, Zn>Cu>Pb>Cd, following the trend of the input metal concentrations. The fates of input metals were 88-97% captured in soil media, 2.0-11.6% not captured by bioretention media, and 0.5-3.3% accumulated in plants. Compared to the metals retained by the soil, the percentages of input metals taken up by plants were relatively low due to the low plant biomass produced in this study. Greater biomass density would be required for the vegetation to have a valuable impact in prolonging the lifetime of a bioretention cell.  相似文献

There have been concerted efforts to find alternative sources of water for small rural communities in the Southern Africa region, and one such initiative is Sand Abstraction Systems (SAS) or infiltration galleries. The sand formation of the riverbed of an ephemeral river is utilised as a natural filter giving potable water of high quality, but is prone to pathogenic contamination. The main objective of our study was to test the suitability of Harvey and Garabedian model for describing bacterial removal in SAS. The Harvey and Garabedian model with some modifications was adopted due to the few parameters required in applying the model. The coefficients and media properties were established from laboratory experiments and field studies. The implicit finite difference method was applied to solve the numerical model equation and to simulate the aquifer response to Escherichia coli for Nkayi SAS. The results of the bacterial simulation from the model fitted well with measured field data and with a small margin of error between the field results and the model (+/-5 coliform count/100 ml).  相似文献

A sand column leaching system with well-controlled suction and flow rate was built to investigate the effects on bacterial transport of air-water interface effects (AWI) correlated to water content, particle size, and column length. Adsorption of Escherichia coli strain D to silica sands was measured in batch tests. The average % adsorption for coarse and fine sands was 45.9+/-7.8% and 96.9+/-3.2%, respectively. However, results from static batch adsorption experiments have limited applicability to dynamic bacterial transport in columns. The early breakthrough of E. coli relative to bromide was clear for all columns, namely c. 0.15 to 0.3 pore volume earlier. Column length had no significant effects on the E. coli peak concentration or on total recovery in leachate, indicating retention in the top layer of sands. Tailing of breakthrough curves was more prominent for all fine sand columns than their coarse sand counterparts. Bacterial recovery in leachate from coarse and saturated sand columns was significantly higher than from fine and unsaturated columns. Observed data were fitted by the convection-dispersion model, amended for one-site and two-site adsorption to particles, and for air-water interface (AWI) adsorption. Among all models, the two-site+AWI model achieved consistently high model efficiency for all experiments. Thus it is evident from experimental and modeling results that AWI adsorption plays an important role in E. coli transport in sand columns.  相似文献

Five bacterial strains were compared for halogenated compounds conversion in aqueous media. Depending on the strain, the optimal temperature for dehalogenase activity of resting cells varied from 30 to 45 degrees C, while optimal pH raised from 8.4 to 9.0. The most effective dehalogenase activity for 1-chlorobutane conversion was detected with Rhodococcus erythropolis NCIMB13064 and Escherichia coli BL21 (DE3) (DhaA). The presence of 2-chlorobutane or propanal in the aqueous media could inhibit the 1-chlorobutane transformation.  相似文献

This study is the first to be conducted in Lebanon on the isolation and molecular characterization and the antimicrobial resistance profile of environmental pathogenic bacterial strains. Fifty-seven samples of seawater, sediment, crab, and fresh water were collected during the spring and summer seasons of 2003. The isolation of Escherichia coli and Salmonella using appropriate selective media revealed that 94.7% of the tested samples were contaminated with one or both of the tested bacteria. The polymerase chain reaction (PCR) was then used to identify the species of both bacteria using various sets of primers. Many pathogenic E. coli isolates were detected by PCR out of which two were identified as O157:H7 E. coli. Similarly, the species of many of the Salmonella isolates was molecularly identified. The confirmed isolates of Salmonella and E. coli were then tested using the disk diffusion method for their susceptibility to four different antimicrobials revealing high rates of antimicrobial resistance.  相似文献

Enhanced biodegradation of carbofuran (2, 3-dihydro-2, 2 dimethyl-7-benzofuranyl methyl carbamate) is an economically significant, but poorly understood, microbial phenomenon in soil. A series of experiments was conducted to examine short term changes in soil bacterial populations stimulated by carbofuran application at field rates. In the field experiment, commercially formulated carbofuran and butylate (S-ethyl diisobutyl carbamothioate) were applied at 5.6 kg ai ha-1 and 8.4 kg ai ha-1, respectively, on a soil (Putnam silt loam) exhibiting enhanced degradation of carbofuran. In laboratory studies, technical grade carbofuran (20 mg kg-1 soil) was applied to samples of the field soil. Bacterial populations were estimated using non-selective (tryptic soy agar) and selective media containing carbofuran or butylate. Largest population increases in pesticide-treated soil were observed between 7 and 15 days after treatment (DAT) compared to populations in non-treated soil. Significant increases (P less than 0.05) in total bacterial populations and presumed carbofuran-degraders due to carbofuran application were associated with increased populations of Pseudomonas spp. and Flavobacterium spp. Application of carbofuran appeared to provide a competitive advantage to these species over actinomycetes persisting beyond 20 DAT. Growth responses of bacteria to carbofuran in the Putnam soil were compared to those in a native prairie soil (Mexico silt loam), which exhibited a much slower rate of carbofuran degradation. Bacterial population response to carbofuran was measurable, but small and short-lived. Perpetuation of the enhanced degradation phenomenon may lie in a persistent pesticide-induced competitive advantage given to a very small segment of the microbial population. This advantage may not be detectable after 20 days using conventional plating techniques.  相似文献

A soil column experiment was carried out to investigate the effects of inoculation of bacteria on metal bioavailability, mobility and potential leachability through single chemical extraction, consequential extraction and in situ soil solution extraction technologies. Results showed that bacteria inoculated, including Azotobacter chroococcum, Bacillus megaterium and Bacillus mucilaginosus, may pose both positive and negative impacts on bioavailability and mobility of heavy metals in soil, depending on the chemical nature of the metals. The activities of bacteria led to an increase of water dissolved organic carbon (DOC) concentration and a decrease of pH value, which enhanced metal mobility and bioavailability (e.g. an increase of water-soluble and HOAc-soluble Zn). On the other hand, bacteria could immobilize metals (e.g. a great reduction of water-soluble Pb) due to the adsorption by bacterial cell walls and possible sedimentation reactions with phosphate or other anions produced through bacterial metabolism.  相似文献

Studies on the role of individual species of bacteria in wastewater treatment methods, especially the effect of environmental factors and enzyme activities, are limited. In the present investigation Escherichia coli, isolated from a stabilisation pond, was grown in sterile sewage at various temperatures, ranging from 10 degrees to 50 degrees C, and its growth and associated biochemical activities were studied. A temperature of 30 degrees C was found to be optimum for the growth, BOD removal, NH(3)-N release and the activities of protease and catalase. E. coli which was once considered to be of little value in wastewater treatment, also accounted for a considerable reduction in BOD and NH(3)-N release. At optimum and lower temperatures, the catalase activity was proportional to the viable cell count of the bacterium. Dissolved oxygen exhibited an inverse relationship with bacterial growth and protease activity was more pronounced during the declining phase of bacterial growth.  相似文献

The effects of bacteria acting as immersion ice nuclei were investigated in numerical sensitivity studies and compared to the efforts of other ice nuclei such as mineral dust and soot particles. An adiabatic air parcel model was employed simulating convective situations with different initial aerosol particle distributions. The maximum fractions of active ice nuclei were based on field measurements of the proportioning of atmospheric aerosol particle types in continental and marine air masses. Recent field measurements of bacteria concentrations in cloud water and in snow samples were used. From the concentrations in bulk samples the concentration in mean sized cloud droplets was estimated. Immersion freezing was described based on laboratory measurements to constrain the freezing fraction versus temperature. The results indicated that the effects of diminutive amounts of bacteria on ice formation in convective clouds, while being significantly less than the effects of mineral dust particles, might be comparable to the expected effects of soot particles acting as ice nuclei. It can be predicted that bacterial ice nuclei would have to be enriched by at least 104 times reported concentrations in cloud water in order to equate to the impact of mineral dust ice nuclei present in 20–25% of all cloud droplets.  相似文献

The activity response of the antioxidant enzymes superoxide dismutase, catalase, ATP enzyme activities of Escherichia coli (G-), Bacillus subtilis (G+), and Burkholderia cepacia WZ1 (G-) following exposure to quinclorac was investigated. The bacterial strains were treated with the different concentrations of quinclorac (1.65, 16.5, 33.0, 165.0, 330.0, and 500.0 microg L(-1)). Results obtained indicated that SOD and CAT activities of these bacteria were induced positively and obviously by quinclorac, especially to gram-positive (G+) bacteria treated by lower than 330 microg L(-1) of quinclorac. The inhibition of ATPase in E. coli K12, B. subtilis, and B. cepacia WZ1 appeared stronger with the increase of quinclorac concentration, showing a striking dose response relationship, which can, therefore, be used as an available bioindicator for quinclorac pollution. The concentration of quinclorac applied in this research had significant effects on these three bacteria at the early stage of incubation, but none of which was persistent. Native polyacrylamide gel electrophoresis and activity staining of SOD revealed that quinclorac had effects on isoforms of E. coli and B. subtilis, and on the staining intensities of the isoforms of B. cepacia WZ1. When E. coli K12 was incubated with 330 microg L(-1) of quinclorac, the upper band of the isoforms of SOD tended to become slightly more apparent at 1 h after the quinclorac treatment, but the staining activity was slightly reduced after the prolonged treatment of quinclorac. No such changes of the isoforms of B. cepacia WZ1 was observed.  相似文献
