Two new photoactive materials compatible with environmentally friendly solvents (water and methanol) have been synthesized and characterized. They are comprised of a porous matrix of polystyrene and divinylbenzene with bound Rose Bengal and additional pendant groups added to increase the hydrophilicity (ethylenediamine and γ-gluconolactone). The new polymers are efficient photocatalysts capable of generating singlet oxygen after irradiation with visible light. Photochemical oxygenations of 9,10-anthracenedipropionic acid and 2-furoic acid have been carried out. The measured conversions indicate that the new supported photosensitizers are more effective than the parent hydrophobic polymer. Figure

In this work, the efficiency of electrochemical oxidation (EO) was investigated for removing a dye mixture containing Novacron Yellow (NY) and Remazol Red (RR) in aqueous solutions using platinum supported on titanium (Ti/Pt) as anode. Different current densities (20, 40 and 60 mA cm?2) and temperatures (25, 40 and 60 °C) were studied during electrochemical treatment. After that, the EO of each of these dyes was separately investigated. The EO of each of these dyes was performed, varying only the current density and keeping the same temperature (25 °C). The elimination of colour was monitored by UV-visible spectroscopy, and the degradation of organic compounds was analysed by means of chemical oxygen demand (COD). Data obtained from the analysis of the dye mixture showed that the EO process was effective in colour removal, in which more than 90 % was removed. In the case of COD removal, the application of a current density greater than 40 mA cm?2 favoured the oxygen evolution reaction, and no complete oxidation was achieved. Regarding the analysis of individual anodic oxidation dyes, it was appreciated that the data for the NY were very close to the results obtained for the oxidation of the dye mixture while the RR dye achieved higher colour removal but lower COD elimination. These results suggest that the oxidation efficiency is dependent on the nature of the organic molecule, and it was confirmed by the intermediates identified. Figure

Four series of dicephalic cationic surfactants, considered as new antielectrostatic agents have been investigated in order to establish their toxicity and biodegradability. Among them N,N-bis[3,3′-(dimethylamine)propyl]alkylamides, N,N-bis[3,3′-(dimethylamine)propyl]alkylamide dihydrochlorides, N,N-bis[3,3′-(trimethylammonio)propyl]alkylamide dibromides and N,N-bis[3,3′-(trimethylammonio)propyl]alkylamide dimethylsulphates with different hydrophobic chain length (n-C9H19 to n-C15H31) and type of counterion (chloride, bromide and methylsulfate) have been studied. The inhibitory effect against microorganisms has been examined using model gram-positive and gram-negative bacteria, and yeasts. None of the tested surfactants have shown antimicrobial activity against gram-negative bacteria (Escherichia coli, Pseudomonas putida) and yeasts (Saccharomyces cerevisiae, Rhodotorula glutinis) at a concentration below 1000 μg mL−1, however some of them were moderately active against gram-positive bacteria (Staphylococcus aureus, Bacillus subtilis). The Microtox® test was successfully applied to measure EC50 values of the studied dicephalic cationic surfactants. Their toxicity to Vibrio fischeri depended upon the alkanoyl chain length with the EC50 values in a range of 2.6-980 mg L−1. N,N-bis[3,3′-(dimethylamine)propyl]alkylamide dihydrochlorides 2a-b and N,N-bis[3,3′-(trimethylammonio)propyl]alkylamide dibromides 3a-b comprising n-decanoyl and n-dodecanoyl hydrophobic tails appeared to be the least toxic. Furthermore, the biodegradability under aerobic conditions of 2a-b, 3a-b was evaluated using OECD Method 301F. According to the obtained results 2a, 3a-3b can be considered as almost readily biodegradable and they are not expected to be persistent in the environment. Additionally, partial biodegradation was observed for 2b, indicating its possible biodegradation in wastewater treatment systems.  相似文献

The search for novel microorganisms able to degrade olive mill wastewaters (OMW) and withstand the toxic effects of the initially high phenolic concentrations is of great scientific and industrial interest. In this work, the possibility of reducing the phenolic content of OMW using new isolates of fungal strains (Coriolopsis gallica, Bjerkandera adusta, Trametes versicolor, Trichoderma citrinoviride, Phanerochaete chrysosporium, Gloeophyllum trabeum, Trametes trogii, and Fusarium solani) was investigated. In vitro, all fungal isolates tested caused an outstanding decolorization of OMW. However, C. gallica gave the highest decolorization and dephenolization rates at 30 % v/v OMW dilution in water. Fungal growth in OMW medium was affected by several parameters including phenolic compound concentration, nitrogen source, and inoculum size. The optimal OMW medium for the removal of phenolics and color was with the OMW concentration (in percent)/[(NH4)2SO4]/inoculum ratio of 30:6:3. Under these conditions, 90 and 85 % of the initial phenolic compounds and color were removed, respectively. High-pressure liquid chromatography analysis of extracts from treated and untreated OMW showed a clear and substantial reduction in phenolic compound concentrations. Phytotoxicity, assessed using radish (Raphanus sativus) seeds, indicated an increase in germination index of 23–92 % when a 30 % OMW concentration was treated with C. gallica in different dilutions (1/2, 1/4, and 1/8).

To evaluate the genotoxic risk that contaminated sediment could constitute for benthic organisms, three contaminated (VA, VC and VN) and one uncontaminated (RN) sediment samples were collected in the Berre lagoon (France). Potentially bioavailable contaminants in sediments were obtained using sediment extraction with synthetic seawater adjusted to pH 4 or pH 6, simulating the range of pH prevailing in the digestive tract of benthic organisms. The genotoxic activities of these extracts were evaluated by three short-term bioassays: the Salmonella mutagenicity test using the Salmonella typhimurium strain TA102, the alkaline comet assay and the micronucleus assay on the Chinese Hamster Ovary cells CHO-K1. Results of the Salmonella mutagenicity assay detected a mutagenic response for RN extract at pH 6, and for VA extract at pH 4. Results of the comet and micronucleus assays detected low genotoxic/clastogenic activities for VA and VC extracts at pH 6 and higher activities for RN, VA and VC extracts at pH 4. To identify if metals (Al, Fe, Mn, As, Cd, Co, Cr, Cu, Hg, Ni, Pb and Zn) were involved in these genotoxic activities, their concentrations were determined in the extracts, and their speciation was assessed by thermodynamic calculations. Results showed that extracts from sites VA, VC and VN generally presented the highest trace metal contents for both extractants, while the site RN presented lower trace metal contents but the highest Fe and Mn contents. Thermodynamic calculations indicated that Fe, Mn, As and in a lower extend Co, Ni and Zn were mainly present under free forms in extracts, and were consequently, more likely able to induce a genotoxic effect. Results globally showed no correspondence between free metal contents and genotoxic activities. They suggested that these positive results could be due to uncharacterized compounds, acting as direct genotoxic agents or enhancing the genotoxic properties of analyzed metals.  相似文献

Introduction

Trends in precipitation pH and conductivity during 1992?C2009, and in ionic compositions from January 2007 to June 2009, are reported from Lushan Mountain, one of the highest mountains in mid-east China. Annual mean pH was in the range of 4.35?C5.01 and showed a statistically very significant (P?P
?Results and discussions Over the period of study, Lushan Mountain received more rainfall in spring and summer. The pH values varied seasonally with winter minima. The winter multiyear seasonal mean pH was 4.35. The corresponding summer value was 4.88. SO 4 2? and NO 3 ? were the main anions, and NH 4 + and Ca2+ the main cations. The anion to cation ratio was 0.8?C1.0, and that of [SO 4 2? ] to [NO 3 ? ] was 2.4-3.0, much lower than that of the 1980s. However, sulfuric acid was still the main acid present. The ratio of [NH 4 + ] to [Ca2+] was about 1.0, suggesting that these two alkaline substances provided close acid neutralizing capacity. The ratio of [Cl?] to [Na+] was about 0.67, somewhat lower than that of natural precipitation.

Conclusions

Ionic composition varied seasonally and was closely correlated to the amounts of rainfall and pollution. Trajectory analyses showed that the trajectories to Lushan Mountain could be classified in six clusters and trajectories originating from the South Sea and the areas surrounding Lushan Mountain had the greatest impacts on precipitation chemistry.  相似文献

This work was designed to investigate the removal efficiency as well as the ratios of toluene and xylene transported from air to root zone via the stem and by direct diffusion from the air into the medium. Indoor plants (Schefflera actinophylla and Ficus benghalensis) were placed in a sealed test chamber. Shoot or root zone were sealed with a Teflon bag, and gaseous toluene and xylene were exposed. Removal efficiency of toluene and total xylene (m, p, o) was 13.3 and 7.0 μg·m?3·m?2 leaf area over a 24-h period in S. actinophylla, and was 13.0 and 7.3 μg·m?3·m?2 leaf area in F. benghalensis. Gaseous toluene and xylene in a chamber were absorbed through leaf and transported via the stem, and finally reached to root zone, and also transported by direct diffusion from the air into the medium. Toluene and xylene transported via the stem was decreased with time after exposure. Xylene transported via the stem was higher than that by direct diffusion from the air into the medium over a 24-h period. The ratios of toluene transported via the stem versus direct diffusion from the air into the medium were 46.3 and 53.7 % in S. actinophylla, and 46.9 and 53.1 % in F. benghalensis, for an average of 47 and 53 % for both species. The ratios of m,p-xylene transported over 3 to 9 h via the stem versus direct diffusion from the air into the medium was 58.5 and 41.5 % in S. actinophylla, and 60.7 and 39.3 % in F. benghalensis, for an average of 60 and 40 % for both species, whereas the ratios of o-xylene transported via the stem versus direct diffusion from the air into the medium were 61 and 39 %. Both S. actinophylla and F. benghalensis removed toluene and xylene from the air. The ratios of toluene and xylene transported from air to root zone via the stem were 47 and 60 %, respectively. This result suggests that root zone is a significant contributor to gaseous toluene and xylene removal, and transported via the stem plays an important role in this process.  相似文献

The concentrations of 15 priority PAHs were determined in the atmospheric gaseous and particulate phases from nine sites across Assiut City, Egypt. While naphthalene, acenaphthene, and fluorene were the most abundant in the gaseous phase with average concentrations of 377, 184, and 181 ng/m3, benzo[b]fluoranthene, chrysene, and benzo[g,h,i]perylene showed the highest levels in the particulate phase with average concentrations of 76, 6, and 52 ng/m3. The average total atmospheric concentration of target PAHs (1,590 ng/m3) indicates that Assiut is one of the highest PAH-contaminated areas in the world. Statistical analysis revealed a significant difference between the levels of PAHs in the atmosphere of urban and suburban sites (P?=?0.029 and 0.043 for gaseous and particulate phases, respectively). Investigation of diagnostic PAH concentration ratios revealed vehicular combustion and traffic exhaust emissions as the major sources of PAHs with a higher contribution of gasoline rather than diesel vehicles in the sampled areas. Benzo[a]pyrene has the highest contribution (average?=?32, 4 % for gaseous and particulate phases) to the total carcinogenic activity (TCA) of atmospheric PAHs. While particulate phase PAHs have higher contribution to the TCA, gaseous phase PAHs present at higher concentrations in the atmosphere are more capable of undergoing atmospheric reactions to form more toxic derivatives.  相似文献

Purpose

In this work, short-term effects of diesel fuel on Huangpu?CYangtze estuarine wetland soil microbial community structure were studied under simulated conditions through phospholipid fatty acids (PLFAs) analysis. Four native plant species, bulrush (Scirpus tripueter), galingale (Cyperus rotundus), wildrice (Zizania latifolia), and reed (Phragmites australis) were tested in the experiments.

Method

In the pot experiment, 20?g rhizosphere soils were mixed with 20?g diesel-blended soils. The concentration of total petroleum hydrocarbon was 16,000?mg/kg. All pots were incubated for 14?days in dark at 28°C and watered with 12?mL sterile distilled water to keep a liquid level. Microbial activity of the samples was assessed by hydrolysis of fluorescein diacetate. Measurements of soil PLFAs and analysis on gas chromatography were performed.

Results

The microbial activity in the samples of reed was highest after the exposure. In all samples, the common PLFA was straight-chain saturated fatty acid (SFA) and monounsaturated fatty acid (MUFA). After the exposure the relative abundance of MUFA and polyunsaturated fatty acid decreased by 20%, and the relative abundance of straight-chain SFA increased by 20%. The results of diversity and PCA indicated that the effect of diesel pollutant on the microbial community was far stronger than the root effect and the reed roots enhanced the tolerance of soil microorganisms to diesel significantly.

Conclusions

All results showed that the soil microbial community structure differed significantly with the exposure to diesel. In reed rhizosphere, the soil microorganisms exhibited a strong resistance to diesel fuel. It confirmed that the root of reed improved the biodegradation ability of soil microorganisms for diesel pollutants and they could be reasonably matched to cure and restore the ecological environment of oil-contaminated wetlands.  相似文献

To explain the detailed process involved in phosphorus removal by periphyton, the periphyton dominated by photoautotrophic microorganisms was employed in this study to remove inorganic phosphorus (P i ) from wastewater, and the removal kinetics and isotherms were then evaluated for the P i removal process. Results showed that the periphyton was capable of effectively removing P i that could completely remove the P i in 24 h at an initial P i concentration of 13 mg P L?1. Furthermore, the P i removal process by the periphyton was dominated by adsorption at initial stage (~24 h), which involved physical mechanistic process. However, this P i adsorption process was significantly influenced by environmental conditions. This work provides an insight into the understanding of phosphorus adsorption by periphyton or similar microbial aggregates.

Although the effect of volatile organic compounds (VOCs) on the oxidation of dissolved sulfur dioxide by oxygen has been the subject of many investigations, this is the first study which examines the effect of a large number of precisely 16 hydroxy compounds. The kinetics both in the absence and the presence of VOCs was defined by rate laws (A and B): A $$ \hbox{-} \mathrm{d}\left[\mathrm{S}\left(\mathrm{IV}\right)\right]/\mathrm{dt}={R}_o={k}_o\left[\mathrm{S}\left(\mathrm{IV}\right)\right] $$ B $$ \hbox{-} \mathrm{d}\left[\mathrm{S}\left(\mathrm{IV}\right)\right]/\mathrm{dt}={R}_i={k}_i\left[\mathrm{S}\left(\mathrm{IV}\right)\right] $$ where R o and k o are the initial rate and first-order rate constant, respectively, in the absence of VOCs, R i , and k i are the initial rate and the first-order rate constant, respectively, in the presence of VOCs, and [S(IV)] is the concentration of dissolved sulfur dioxide, sulfur(IV). The nature of the dependence of k i on the concentration of inhibitor, [Inh], was defined by Eq. (C). C $$ {k}_i={k}_0/\left(1+B\left[\mathrm{Inh}\right]\right) $$ where B is an empirical inhibition parameter. The values of B have been determined from the plots of 1/k i versus [Inh]. Among aliphatic and aromatic hydroxy compounds studied, t-butyl alcohol and pinacol were without any inhibition effect due to the absence of secondary or tertiary hydrogen. The values of inhibition parameter, B, were related to k inh , the rate constant for the reaction of SO4 ? radical with the inhibitor, by Eq. (D). D $$ B=\left(9\pm 2\right)\times 1{0}^{-4}\times {k}_{inh} $$ Equation (D) may be used to calculate the values of either of B or k inh provided that the other is known. The extent of inhibition depends on the value of the composite term, B[Inh]. However, in accordance with Eq. (C), the extent of inhibition would be sizeable and measurable when B[Inh]?>?0.1 and oxidation of S(IV) would be almost completely stopped when B[Inh]?≥?10. B[Inh] value can be used as a guide whether the reaction step: SO4 ??+?organics? \( \overset{k_{inh}}{\to } \) ?SO4 2??+?non-chain products: should be included in the multiphase models or not.  相似文献

Background

This study investigated the acute effect of benzo[a]anthracene, a significant compound among polycyclic aromatic hydrocarbons, on the biodegradation of a synthetic organic substrate??a peptone/meat extract mixture??under aerobic conditions.

Methods

A laboratory-scale sequencing batch reactor was sustained at steady state at a sludge age of 10?days with substrate feeding. Inhibition tests involved running a series of batch reactors initially seeded with the biomass obtained from the parent reactor. After the biomass seeding, the reactors were started with the peptone mixture and a range of initial benzo[a]anthracene concentrations between 0.5 and 88?mg/L. Experimental profiles of oxygen uptake rates and polyhydroxyalkanoates were evaluated by calibration of a selected model.

Results

Lower doses of benzo[a]anthracene had no effect on process kinetics. The noticeable acute impact was only observed with the addition of 88?mg/L of benzo[a]anthracene, but it was limited with the storage mechanism: the amount of organic substrate diverted to polyhydroxyalkanoates was significantly reduced with a corresponding decrease in the maximum storage rate, k STO, from 2.7 down to 0.6?day?1. Similarly, the maximum growth rate from internally stored polyhydroxyalkanoates was lowered from 2.3 to 1.0?day?1.

Conclusion

Among the mechanisms for direct substrate utilization, only the hydrolysis rate was slightly reduced, but otherwise, the overall COD removal efficiency was not affected.  相似文献

Oropharyngeal cancer is a significant public health issue in the world. The incidence of oropharyngeal cancer has been increased among people who have habit of chewing smokeless tobacco (SLT) in Pakistan. The aim of present study was to evaluate the concentration of nickel (Ni) in biological samples (whole blood, serum) of oral (n?=?95) and pharyngeal (n?=?84) male cancer patients. For comparison purposes, the biological samples of healthy age-matched referents (n?=?150), who consumed and did not consumed SLT products, were also analyzed for Ni levels. As the Ni level is very low in biological samples, a preconcentration procedure has been developed, prior to analysis of analyte by flame atomic absorption spectrometry (FAAS). The Ni in acid-digested biological samples was complexed with ammonium pyrrolidinedithio carbamate (APDC), and a resulted complex was extracted in a surfactant Triton X-114. Acidic ethanol was added to the surfactant-rich phase prior to its analysis by FAAS. The chemical variables, such as pH, amounts of reagents (APDC, Triton X-114), temperature, incubation time, and sample volume were optimized. The resulted data indicated that concentration of Ni was higher in blood and serum samples of cancer patients as compared to that of referents who have or have not consumed different SLT products (p?=?0.012–0.001). It was also observed that healthy referents who consumed SLT products have two to threefold higher levels of Ni in both biological samples as compared to those who were not chewing SLT products (p? Butyltin (BT) contamination was evaluated in hermit crabs from 25 estuaries and in sediments from 13 of these estuaries along about 2,000 km of the Brazilian coast. BT contamination in hermit crabs ranged from 2.22 to 1,746 ng Sn g-1 of DBT and 1.32 to 318 ng Sn g-1 of TBT. In sediment samples, the concentration also varied widely, from 25 to 1,304 ng Sn g-1 of MBT, from 7 to 158 ng Sn g-1 of DBT, and from 8 to 565 ng Sn g-1 of TBT. BTs are still being found in surface sediments and biota of the estuaries after the international and Brazilian bans, showing heterogeneous distribution among and within estuaries. Although hermit crabs were previously tested as an indicator of recent BT contamination, the results indicate the presence of contamination, probably from resuspension of BTs from deeper water of the estuary.

Background, Aims and Scope

Vallisneria spiralis Linn., a common, submerged macrophyte, is widely available in quiet waters of lakes, ponds, marshes and streams in Southeast Asia. V. spiralis plays a significant role not only in decreasing eutrophication of water body for its productivity, but also in inhibiting the growth of blue-green algae? The aim of the paper involves the isolation and identification of allelochemicals from extracts of V. spiralis by activity-guided fractionation and column chromatography.

Methods

Leaves of V. spiralis was washed free of debris, air-dried and refluxed in 95% EtOH. The extract was isolated using column chromatography and fractionation with antialgal activity. Potential allelochemicals were analyzed by high-resolution gas chromatography-mass spectrometry (HRGC-MS).

Results

Two fractions with strong antialgal activity were isolated using column chromatography and activity-guided fractionation from the extract of V. spiralis. 2-Ethyl-3-methylmaleimide, dihydroactinidiolide and 4-oxo-β-Ionone were identified in the first fraction, and 3-hydroxy-5,6-epoxy-β-ionone, loliolide, 6-hydroxy-3-oxo-α-ionone and an unknown compound in the second fraction. They had strong inhibitory effects on Microcystis aeruginosa Kütz.

Discussion

2-Ethyl-3-methylmaleimide is a byproduct of photooxidation of chlorophyll, and five other compounds identified were derivatives of β-carotene. HRGC-MS and derivatization technology were used to identify and confirm their molecular structures. The formula of the unknown compound was C16H19NO4. Metabolites of plant pigments had strong inhibitory activities on growth of algae.

Conclusions

Six compounds had been identified in V. spiralis, among them, 2-ethyl-3-methylmaleimide was the main allelochemical, and derivatives of ionone were also potential allelochemicals.

Recommendations and Perspective

. The results of our research could help us to study further mechanisms of inhibitory effect on algae and develop new potential antialgal substances.  相似文献

Evaluation of olive oil mill wastewater toxicity on spinach   总被引:1,自引:1,他引:0

Background, aim, and scope

Olive oil mill wastewater (OMW), a by-product of the olive oil extraction process, is annually produced in huge amounts in olive-growing areas and represents a significant environmental problem in Mediterranean areas. We studied the impact of OMW dilutions (1:20 and 1:10) on spinach plants in order to evaluate OMW dilutions as a low-cost alternative method for the disposal of this waste.

Materials and methods

The effects of OMW dilutions were evaluated on seed germination, shoot and root elongation, biomass production, nutrient uptake and translocation, ascorbic acid content, polyphenols, photosynthetic pigments, and photosynthetic performance of spinach.

Results

Plant biomass was more affected than plant height and total chlorophyll; carotenoid and ascorbic acid content progressively decreased with decreasing OMW dilution. Exposure to both OMW dilutions resulted in overaccumulation of total polyphenols, which were negatively correlated to plant biomass and nutrients. Nutrient (Fe, Ca, and Mg) content was insufficient leading to reduced growth. Water use efficiency decreased mainly due to decreased CO2 assimilation rate rather than to a decline of transpiration rate. Disturbances in photosystem II (PSII) photochemical efficiency could be better envisaged by the ratio between variable fluorescence and initial fluorescence (Fv/Fo), which showed much greater amplitude than the maximal photochemical efficiency of PSII photochemistry (Fv/Fm).

Conclusions

From the data obtained, it is suggested that 1:20 OMW dilutions are still phytotoxic and that higher OMW dilutions should be used in order to use this waste for the irrigation of spinach plants.  相似文献

A novel photocatalytic reactor for wastewater treatment was designed and constructed. The main part of the reactor was an aluminum tube in which 12 stainless steel circular baffles and four quartz tube were placed inside of the reactor like shell and tube heat exchangers. Four UV–C lamps were housed within the space of the quartz tubes. Surface of the baffles was coated with TiO2. A simple method was employed for TiO2 immobilization, while the characterization of the supported photocatalyst was based on the results obtained through performing some common analytical methods such as X-ray diffraction (XRD), scanning electron microscope (SEM), and BET. Phenol was selected as a model pollutant. A solution of a known initial concentration (20, 60, and 100 ppmv) was introduced to the reactor. The reactor also has a recycle flow to make turbulent flow inside of the reactor. The selected recycle flow rate was 7?×?10?5 m3.s?1, while the flow rate of feed was 2.53?×?10?7, 7.56?×?10?7, and 1.26?×?10?6 m3.s?1, respectively. To evaluate performance of the reactor, response surface methodology was employed. A four-factor three-level Box–Behnken design was developed to evaluate the reactor performance for degradation of phenol. Effects of phenol inlet concentration (20–100 ppmv), pH (3–9), liquid flow rate (2.53?×?10?7?1.26?×?10?6 m3.s?1), and TiO2 loading (8.8–17.6 g.m?2) were analyzed with this method. The adjusted R 2 value (0.9936) was in close agreement with that of corresponding R 2 value (0.9961). The maximum predicted degradation of phenol was 75.50 % at the optimum processing conditions (initial phenol concentration of 20 ppmv, pH?~?6.41, and flow rate of 2.53?×?10?7 m3.s?1 and catalyst loading of 17.6 g.m?2). Experimental degradation of phenol determined at the optimum conditions was 73.7 %. XRD patterns and SEM images at the optimum conditions revealed that crystal size is approximately 25 nm and TiO2 nanoparticles with visible agglomerates distribute densely and uniformly over the surface of stainless steel substrate. BET specific surface area of immobilized TiO2 was 47.2 and 45.8 m2 g?1 before and after the experiments, respectively. Reduction in TOC content, after steady state condition, showed that maximum phenol decomposition occurred at neutral condition (pH?~?6). Figure

Background, aim and scope

Lead (Pb) accumulation in soils affects plants primarily through their root systems. The aim of this study was to investigate early symptoms of the loss of membrane integrity and lipid peroxidation in root tissues and physiological adaptation mechanism to Pb in accumulating ecotypes (AE) and non-accumulating ecotypes (NAE) of Sedum alfredii under Pb stress in hydroponics.

Methods and results

Histochemical in situ analyses, fluorescence imaging, and normal physiological analysis were used in this study. Pb accumulation in roots of both AE and NAE increased linearly with increasing Pb levels (0?C200???M), and a significant difference between both ecotypes was noted. Both loss of plasma membrane integrity and lipid peroxidation in root tissues became serious with increasing Pb levels, maximum tolerable Pb level was 25 and 100???M for NAE and AE, respectively. Pb supplied at a toxic level caused a burst of reactive oxygen species (ROS) in root cells in both ecotypes. However, the root cells of AE had inherently higher activities of superoxide dismutase (SOD), guaiacol peroxidase (POD), and lipoxygenase (LOX) in control plants, and the induction response of these antioxidant enzymes occurred at lower Pb level in AE than NAE. AE plants maintained higher ascorbic acid and H2O2 concentrations in root cells than NAE when exposed to different Pb levels, and Pb induced more increase in dehydroascorbate (DHA), catalase (CAT), and ascorbate peroxidase (APX) in AE than NAE roots.

Discussion and conclusion

Results indicate that histochemical in situ analyses of root cell death and lipid peroxidation under Pb short-term stress was sensitive, reliable, and fast. Higher tolerance in roots of accumulating ecotype under Pb stress did depend on effective free oxygen scavenging by making complex function of both constitutively higher activities and sensitive induction of key antioxidant enzymes in root cells of S. alfredii.  相似文献

This study aimed to determine the occurrence, abundance, and fate of nine important antimicrobial resistance genes (ARGs) (sul1, sul2, tetB, tetM, ermB, ermF, fexA, cfr, and Intl1) in the simulated soil and pond microcosms following poultry and swine manure application. Absolute quantitative PCR method was used to determine the gene copies. The results were modeled as a logarithmic regression (N?=?mlnt?+?b) to explore the fate of target genes. Genes sul1, Intl1, sul2, and tetM had the highest abundance following the application of the two manure types. The logarithmic regression model fitted the results well (R 2 values up to 0.99). The reduction rate of all genes (except for the genes fexA and cfr) in manure-pond microcosms was faster than those in manure-soil microcosms. Importantly, sul1, intl1, sul2, and tetM had the lowest reduction rates in all the samples and the low reduction rates of tetM was the first time to be reported. These results indicated that ARG management should focus on using technologies for the ARG elimination before the manure applications rather than waiting for subsequent attenuation in soil or water, particularly the ARGs (such as sul1, intl1, sul2, and tetM investigated in this study) that had high abundance and low reduction rate in the soil and water after application of manure.  相似文献

Purpose

Bacterial community structure and the chemical components in aerosols caused by rotating brushes in an Orbal oxidation ditch were assessed in a Beijing municipal wastewater treatment plant.

Methods

Air samples were collected at different distances from the aerosol-generating rotating brushes. Molecular culture-independent methods were used to characterize the community structure of the airborne bacteria in each sample regardless of cell culturability. A clone library of 16S rDNA directly amplified from air DNA of each sample was constructed and sequenced to analyze the community composition and diversity. Insoluble particles and water-soluble ions emitted with microorganisms in aerosols were analysis by a scanning electron microscope together with energy dispersive X-ray spectroscopy and ion chromatogram analyzer.

Results

In total, most of the identified bacteria were Proteobacteria. The majority of sequences near the rotating brushes (the main source of the bioaerosols) were Proteobacteria (62.97 %) with ??-(18.52 %) and ??-(44.45?%) subgroups and Bacteroidetes (29.63 %). Complex patterns were observed for each sampling location, suggesting a highly diverse community structure, comparable to that found in water in the Orbal oxidation ditch. Accompany with microorganisms, 46.36???g/m3 of SO 4 2? , 29.35???g/m3 of Cl?, 21.51???g/m3 of NO 3 ? , 19.76???g/m3 of NH 4 + , 11.42???g/m3 of PO 4 3? , 6.18???g/m3 of NO 2 ? , and elements of Mg, Cl, K, Na, Fe, S, and P were detected from the air near the aerosols source.

Conclusions

Differences in the structure of the bacterial communities and chemical components in the aerosols observed between sampling sites indicated important site-related variability. The composition of microorganisms in water was one of the most important sources of bacterial communities in bioaerosols. Chemical components in bioaerosols may provide a media for airborne microorganism attachment, as well as a suitable microenvironment for their growth and survival in the air. This study will be benefit for the formulation of pollution standards, especially for aerosols, that take into account plant workers?? health.  相似文献
