Chlordecone is an organochlorine insecticide that has been widely used to control banana weevil in the French West Indies. As a result of this intense use, up to 20,000 ha are contaminated by this insecticide in the French West Indies, and this causes environmental damage and health problems. A scenario of exposure was drawn by French authorities, based on land usage records. Many efforts have been made to monitor the occurrence of chlordecone and its main metabolites using different analytical methods, including GC, GC/MS, LC/MS, and NIRS. Although these different methods allow for the detection and quantification of chlordecone from soils, none of them estimate the bottleneck caused by extraction of this organochlorine from soils with high adsorption ability. In this study, we used 13C10-chlordecone as a tracer to estimate chlordecone extraction yield and to quantify chlordecone in soil extracts based on the 13C/12C isotope dilution. We report the optimization of 13C10-chlordecone extraction from an Andosol. The method was found to be linear from 0.118 to 43 mg kg?1 in the Andosol, with an instrumental detection limit estimated at 8.84 μg kg?1. This method showed that chlordecone ranged from 35.4 down to 0.18 mg kg?1 in Andosol, Nitisol, Ferralsol, and Fluvisol soil types. Traces of the metabolite β-monohydrochlordecone were detected in the Andosol, Nitisol, and Ferralsol soil samples. This last result indicates that this method could be useful to monitor the fate of chlordecone in soils of the French West Indies.  相似文献

The capacity of two soil fungi, Trichoderma koningii and Penicillium janthinellum, to oxidize n-C10:0 and n-C11:0 fatty acids to CO2 and store intracellular lipids during growth is unknown. This article reports for the first time the metabolism of decanoic acid (DA, C10:0), undecanoic acid (UDA, n-C11:0), a mixture of the acids (UDA+DA) and a mixture of UDA+ potato dextrose broth (PDB) by T. koningii and P. janthinellum and their mixed culture. A control PDB complex substrate was used as a substrate control treatment. The fungal cultures were assayed for their capacity to: (1) oxidize n-C10:0 and n-C11:0 fatty acids to CO2 and (2) store lipids intracellularly during growth. On all four fatty acid substrates, the mixed T. koningii and P. janthinellum culture produced more biomass and CO2 than the individual fungal cultures. Per 150 mL culture, the mixed species culture grown on UDA+PDB and on PDB alone produced the most biomass (7,567 mg and 11,425 mg, respectively). When grown in DA, the mixed species culture produced the least amount of biomass (6,400 mg), a quantity that was lower than those obtained in UDA (7,550 mg) or UDA+DA (7,270 mg). Amounts of CO2 produced ranged from 210 mg under DA to 618 mg under PDB, and these amounts were highly correlated with biomass (r2 = 0.99). Fluorescence microscopy of stained lipids in the mixed fungal cell cultures growing during the exponential phase demonstrated larger fungal cells and higher accumulation of lipids in membranes and storage bodies than those observed during the lag and stationary phases. T. koningii and P. janthinellum grown on n-C10:0 and n-C11:0 fatty acids produced lower amounts of biomass and CO2, but stored higher amounts of intracellular lipids, than when grown on PDB alone.  相似文献

A highly tolerant phenol-degrading yeast strain PHB5 was isolated from wastewater effluent of a coke oven plant and identified as Candida tropicalis based on phylogenetic analysis. Biodegradation experiments with C. tropicalis PHB5 showed that the strain was able to utilize 99.4 % of 2,400 mg l?1 phenol as sole source of carbon and energy within 48 h. Strain PHB5 was also observed to grow on 18 various aromatic hydrocarbons. Haldane model was used to fit the exponential growth data and the following kinetic parameters were obtained: μ max?=?0.3407 h?1, K S?=?15.81 mg l?1, K i?=?169.0 mg l?1 (R 2?=?0.9886). The true specific growth rate, calculated from μ max, was 0.2113. A volumetric phenol degradation rate (V max) was calculated by fitting the phenol consumption data with Gompertz model and specific degradation rate (q) was calculated from V max. The q values were fitted with Haldane model, yielding following parameters: q max?=?0.2766 g g?1 h?1, K S ?=?2.819 mg l?1, K i ?=?2,093 (R 2?=?0.8176). The yield factor (Y X/S ) varied between 0.185 to 0.96 g g?1 for different initial phenol concentrations. Phenol degradation by the strain proceeded through a pathway involving production of intermediates such as catechol and cis,cis-muconic acid which were identified by enzymatic assays and HPLC analysis.  相似文献

The insecticide chlordecone applied for decades in banana plantations currently contaminates 20,000 ha of arable land in the French West Indies. Although the impact of various pesticides on soil microorganisms has been studied, chlordecone toxicity to the soil microbial community has never been assessed. We investigated in two different soils (sandy loam and silty loam) exposed to different concentrations of CLD (D0, control; D1 and D10, 1 and 10 times the agronomical dose) over different periods of time (3, 7, and 32 days): (i) the fate of chlordecone by measuring 14C-chlordecone mass balance and (ii) the impact of chlordecone on microbial community structure, abundance, and function, using standardized methods (-A-RISA, taxon-specific quantitative PCR (qPCR), and 14C-compounds mineralizing activity). Mineralization of 14C-chlordecone was inferior below 1 % of initial 14C-activity. Less than 2 % of 14C-activity was retrieved from the water-soluble fraction, while most of it remained in the organic-solvent-extractable fraction (75 % of initial 14C-activity). Only 23 % of the remaining 14C-activity was measured in nonextractable fraction. The fate of chlordecone significantly differed between the two soils. The soluble and nonextractable fractions were significantly higher in sandy loam soil than in silty loam soil. All the measured microbiological parameters allowed discriminating statistically the two soils and showed a variation over time. The genetic structure of the bacterial community remained insensitive to chlordecone exposure in silty loam soil. In response to chlordecone exposure, the abundance of Gram-negative bacterial groups (β-, γ-Proteobacteria, Planctomycetes, and Bacteroidetes) was significantly modified only in sandy loam soil. The mineralization of 14C-sodium acetate and 14C-2,4-d was insensitive to chlordecone exposure in silty loam soil. However, mineralization of 14C-sodium acetate was significantly reduced in soil microcosms of sandy loam soil exposed to chlordecone as compared to the control (D0). These data show that chlordecone exposure induced changes in microbial community taxonomic composition and function in one of the two soils, suggesting microbial toxicity of this organochlorine.  相似文献

The re-emergence of schistosomiasis has given rise to ubiquitous concentrations of the primary control agent pentachlorophenol (PCP) in the environment, especially in the surface waters of China. In this study, the effects of environmentally relevant concentrations of PCP, namely, 0.0002, 0.002, 0.02, 0.2, and 2 μmol/L on survival, age at first reproduction, fecundity, length of mothers, and number of molts of Daphnia magna were studied over three generations. The survival of D. magna exposed to 2 μmol/L was significantly affected in the three generations. Toxic effects were enhanced in later generations. Age at first reproduction of F 1 and F 2 D. magna was significantly slower than that of the controls. The total number of offspring per female exposed to concentrations of 0.002 μmol/L or greater was less (23.5 to 67.6, 9.4 to 73.7, and 3.6 to 83.7 %) than that of the controls in the F 0, F 1, and F 2 generations, respectively. The body length of mothers significantly decreased (4.7 to 6.8, 9.6 to 15.1, and 13.3 to 23.2 %) after exposure to 0.002 μmol/L or greater than those of unexposed individuals in the F 0, F 1, and F 2 generations, respectively. Dose–response relationships between concentrations of PCP and length and number of molts of D. magna were observed in the F 0 to F 2 generations. PCP concentrations on the surface waters of China caused adverse effects to D. magna, which increased over successive generations. Significant effects were observed in the third generation. The multigenerational studies were more sensitive than the single-generation experiments. Thus, multigenerational exposure may be more predictive of chronic exposure under field conditions.  相似文献

Laboratory experiments were conducted to investigate copper (Cu) subcellular distribution and toxicity in Hydrilla verticillata. Fronds were subjected to different concentrations (15, 75, and 150 μM) of Cu for 7 days. Cu grains were found in cell walls, plasmodesmata, and within the nuclei and chloroplasts using the autometallographic technique. Subcellular fractionation of Cu-containing tissues indicated that in leaves subjected to high Cu concentrations, 59–65 % of the element was located in the cell wall fraction, followed by cell organelles (21–30 %) and the soluble fraction (10–14 %). The levels of K, P, Zn, and Mg declined under all Cu concentrations, but Ca, Mn, and Fe contents reached their peak at 15 μM Cu and decreased thereafter. F v/F m, F 0, and F m fell significantly in line with the decrease in pigment content. Cu exposure also caused significant damage to the chloroplasts, mitochondria, and nuclei, including disintegration of the chloroplasts and vacuolization of the mitochondria and nuclei, all of which suggested that Cu hastened plant senescence. The Cu maximum permissible concentration for H. verticillata was 10 μM, which was less than the existing general water quality standard. This suggested that H. verticillata could be used to assess Cu phytotoxicity.  相似文献

The increasing use of nanoparticles (NPs) worldwide has raised some concerns about their impact on the environment. The aim of the study was to assess the toxicity of metal oxide nanoparticles, singly or combined, in a freshwater fish (Carassius auratus). The fish were exposed for 7, 14, and 21 days to different concentrations of NPs (10 μg Al2O3.L?1, 10 μg ZnO.L?1, 10 μg Al2O3.L?1 plus 10 μg ZnO.L?1, 100 μg Al2O3.L?1, 100 μg ZnO.L?1, and 100 μg Al2O3.L?1 plus 100 μg ZnO.L?1). At the end of each exposure period, antioxidant enzyme activity (catalase, glutathione-S-transferase, and superoxide dismutase), lipid peroxidation, and histopathology were assessed in the gills and livers of C. auratus. The results show an increase in catalase (CAT) and superoxide dismutase (SOD) activity in the gills and livers of fish, especially after 14 days of exposure to single and combined NPs, followed by a reduction at 21 days. An increase in glutathione S-transferase (GST) was observed in gills after 7 days for all tested NP concentrations (single and combined); while in livers, a significant increase was determined after 14 days of exposure to 100 μg.L?1 of both single ZnO and Al2O3 NPs. Lipid peroxidation (LPO) significantly increased in gills after 7 days of exposure to 100 μg.L?1 Al2O3 NPs (single or combined). In livers, LPO increased significantly after 7 days of exposure to all tested concentrations of both single ZnO and Al2O3 (except for 10 μg Al2O3.L?1), and after 14 days of exposure to ZnO (10 and 100 μg.L?1) and Al2O3 (100 μg.L?1). The results from histological observations suggest that exposure to metal oxide NPs affected both livers and gills, presenting alterations such as gill hyperplasia and liver degeneration. However, the most pronounced effects were found in gills. In general, this study shows that the tested NPs, single or combined, are capable of causing sub-lethal effects on C. auratus, but when combined, NPs seem to be slightly more toxic than when added alone.  相似文献

This paper analyzes the effect of exogenous urea in increased concentration gradient (0, 100, 500 and 1,000 mg L?1) on photosynthetic pigments (measured spectrophotometrically), uptake of 14CO2 (using radioisotope), and urease activity (by measuring ammonia with Nessler’s reagent) in leaves of Elodea densa Planch. We have observed that low concentration of urea (100 mg L?1) stimulates the accumulation of photosynthetic pigments and intensifies photosynthesis in E. densa, whereas high concentration (1,000 mg L?1) suppresses these processes. Urease activity increased by approximately 2.7 and 8 fold when exogenous urea concentrations were 100 and 500 mg L?1, respectively. However, exogenous urea in high concentration (1,000 mg L?1) decreased urease activity by 1.5 fold compared to the control. The necessity of mitigating urea and other nitrogen-containing compounds (NH3 from urea) in water bodies has been discussed with emphasis on the potential for phytoremediation of urea using common water weed viz. Fusarium wilt of cucumber caused by the fungus, Fusarium oxysporum, is one of the most destructive soilborne diseases and can result in serious economic loss. No efficient fungicide is currently available to control the disease. The aim of this study was to examine the disease suppression ability of pig manure and sludge composts in peat-based container media and explore the possible disease suppression mechanisms. Pig manure and sewage sludge compost were made in laboratory-scale tanks. Plant growth media were formulated with peat mixture and compost (or 60?°C heated compost) in a 4:1 ratio (v/v). Cucumber seedlings were artificially inoculated with F. oxysporum conidia (5?×?105 conidia mL?1) by the root-dip method. Cucumber Fusarium wilt was effectively suppressed in sludge compost-amended media, while the disease suppression effect of pig manure compost was limited. The ammonia levels in the manure compost-amended media were significantly higher than those of sludge compost-amended media, which could explain its lower disease suppression ability. Heated composts behaved similarly with respect to disease suppression. Adding composts increased microbial biomass, microbial activity, and the microbial diversity of the growth media. PCR-DGGE results indicated that the fungal community had a significant correlation to the disease severity. The artificially inoculated pathogen was retrieved in all treatments and one possible biocontrol agent was identified as a strain of F. oxysporum by phylogenetic analyses. The results indicated that the sludge compost used in this study could be applied as a method for biocontrol of cucumber Fusarium wilt.  相似文献

The full-length cDNA of catalase (EfCAT) from Eisenia fetida was cloned (GenBank accession no. JN617999). Sequence characterization revealed that EfCAT protein sequence contained proximal heme-ligand signature sequence (351RLFSYSDTH359), two glycosylation sites (N145 and N436), the proximal active site signature (61FDRERIPERVVHAKGAGA78), and 12 amino acids (N145, H191, F195, S198, R200, N210, Y212, K234, I299, W300, Q302, and Y355), which were identified as putative residues involved in NADPH binding. These conserved motifs and catalase signature sequences were essential for the structure and function of EfCAT. The present study also investigated the effect of the veterinary food additive zinc oxide on antioxidant processes in E. fetida, at different concentrations and exposure durations. A significant increase (by 106.0 % compared to controls) in CAT activity at 500 mg/kg was registered at day 15. The superoxide dismutase (SOD) activity at 500 mg/kg increased to the maximum value (by 44.0 %) measured at day 15. There was a significant increase in glutathione peroxidase (GPx) activity for all concentrations after 5 days. The results showed that dietary Zn (500 mg/kg) causes oxidative damage to earthworms. At early stages of earthworms exposed to ZnO, GPx is the main enzyme to impair the oxidative status; while at later stages the enzymes CAT and SOD were the main indicators of oxidative stress. The antioxidant enzymatic variations may be an adaptive response of earthworms to survive in contaminated soils.  相似文献

Based on environmental monitoring data in 93 major cities and meteorological records at 398 weather stations in China from 1981 to 2007, total suspended particle (TSP) concentration, the intensity of dustfall, and sand and dust storm frequency (Fd) were analysed. During the past 27 years, the annual average TSP concentration (CTSP) in 93 cities was 402 μg m?3. Annual average CTSP decreased from the north to the south and from inland to the coast areas with a peak value of 628.8 μg m?3 in Lanzhou. In the 1980s, 1990s and 2000s, annual average CTSP was 628.7, 319.2, and 250.1 μg m?3, respectively. Annual average intensity of dustfall (Id) was 240.5 t km?2 a?1, decreased from northern to southern China and from inland to the coast areas with the maximum value of 717.2 t km?2 a?1 in Baotou. In the 1980s, 1990s and 2000s, annual average Id was 334.8, 220.9, 146 t km?2 a?1 respectively. Annual average Id in the Loess Plateau region was commonly higher than 200 t km?2 a?1. The annual average Fd decreased from arid regions in northwestern China to humid areas in southeastern China with two sand and sand storm centers existing in Xinjiang Taklamakan Desert and western Inner Mongolia. The annual average Fd in the 1980s, 1990s, 2000s was 16, 8, 6 days respectively, decreased steadily from 18 days in 1981–5 days in 2007. Annual average Id had a positive linear relation to annual average CTSP (R2 = 0.96). Annual average Fd had a positive relation with annual average CTSP (R2 = 0.97) as well as annual average Id (R2 = 0.94). TSP was the chief pollutant influencing Air Pollution Index (API) in northern China in spring and winter seasons. Sand and dust storm might be a major factor affecting the temporal variability and spatial distribution of TSP and dustfall in China.  相似文献

Abstract

Little is known about the fungal metabolism of nC10 and nC11 fatty acids and their conversion into lipids. A mixed batch culture of soil fungi, T. koningii and P. janthinellum, was grown on undecanoic acid (UDA), a mixture of UDA and potato dextrose broth (UDA+PDB), and PDB alone to examine their metabolic conversion during growth. We quantified seven intracellular and extracellular lipid classes using Iatroscan thin-layer chromatography with flame ionization detection (TLC-FID). Gas chromatography with flame ionization detection (GC-FID) was used to quantify 42 individual fatty acids. Per 150 mL culture, the mixed fungal culture grown on UDA+PDB produced the highest amount of intracellular (531 mg) and extracellular (14.7 mg) lipids during the exponential phase. The content of total intracellular lipids represented 25% of the total biomass-carbon, or 10% of the total biomass dry weight produced. Fatty acids made up the largest class of intracellular lipids (457 mg/150 mL culture) and they were synthesized at a rate of 2.4 mg/h during the exponential phase, and decomposed at a rate of 1.8 mg/h during the stationary phase, when UDA+PDB was the carbon source. Palmitic acid (C16:0), stearic acid (C18:0), oleic acid (C18:1), linoleic acid (C18:2) and vaccenic acid (C18:1) accounted for >80% of the total intracellular fatty acids. During exponential growth on UDA+PDB, hydrocarbons were the largest pool of all extracellular lipids (6.5 mg), and intracellularly they were synthesized at a rate of 64 μg/h. The mixed fungal species culture of T. koningii and P. janthinellum produced many lipids for potential use as industrial feedstocks or bioproducts in biorefineries.  相似文献

The insecticide 14C-chlorpyrifos was found mineralized in a Tunisian soil with repeated exposure to it. From this soil, a bacterial strain was isolated that was able to grow in a minimal salt medium (MSM) supplemented with 25 mg L?1 of chlorpyrifos. It was characterized as Serratia rubidaea strain ABS 10 using morphological and biochemical analyses, as well as 16S rRNA sequencing. In a liquid culture, the S. rubidaea strain ABS 10 was able to dissipate chlorpyrifos almost entirely within 48 h of incubation. Although the S. rubidaea strain ABS 10 was able to grow in an MSM supplemented with chlorpyrifos and dissipate it in a liquid culture, it was not able to mineralize 14C-chlorpyrifos. Therefore, it can be concluded that the dissipation capability of this bacteria might be attributed to its capacity to adsorb CHL. It can also be ascribed to other reasons such as the formation of biogenic non-extractable residues. In both non-sterile and sterile soil inoculated with S. rubidaea strain ABS 10, chlorpyrifos was more rapidly dissipated than in controls with DT50 of 1.38 and 1.05 days, respectively.

The present study attempts to analyze the biosorption trend of biosorbent Caulerpa fastigiata (macroalgae) biomass for removal of toxic heavy metal ion Pb (II) from solution as a function of initial metal ion concentration, pH, temperature, sorbent dosage, and biomass particle size. The sorption data fitted with various isotherm models and Freundlich model was the best one with correlation coefficient of 0.999. Kinetic study results revealed that the sorption data on Pb (II) with correlation coefficient of 0.999 can best be represented by pseudo-second-order. The biosorption capacity (q e ) of Pb (II) is 16.11?±?0.32 mg g?1 on C. fastigiata biomass. Thermodynamic studies showed that the process is exothermic (ΔH° negative). Free energy change (ΔG°) with negative sign reflected the feasibility and spontaneous nature of the process. The SEM studies showed Pb (II) biosorption on selective grains of the biosorbent. The FTIR spectra indicated bands corresponding to –OH, COO?, –CH, C?=?C, C?=?S, and –C–C– groups were involved in the biosorption process. The XRD pattern of the C. fastigiata was found to be mostly amorphous in nature.  相似文献

In a hydroponic culture, experiments were performed to study the influence of potassium (K) supplementation (0, 20, 40, 60, 80, and 100 mg L?1) on the arsenic (As; 0, 8, and 10 mg L?1)-accrued changes in growth traits (plant biomass, root–shoot length) and the contents of lepidine, As and K, in garden cress (Lepidium sativum Linn.) at 10 days after treatment. The changes in these traits were correlated with shoot proline content, protein profile, and the activities of antioxidant enzymes namely superoxide dismutase (SOD, EC 1.15.1.1), catalase (CAT, EC 1.11.1.6), glutathione reductase (GR, EC 1.8.1.7), and ascorbate peroxidase (APX, EC 1.11.1.11). In general, As-alone treatments significantly decreased the growth traits but lead to significant enhancements in shoot proline and enzyme activities. K-supplementation to As-treated L. sativum seedlings decreased shoot-As content, reduced As-induced decreases in growth traits but enhanced the content of shoot proline, and the activities of the studied enzymes maximally with K100 + As8 and As10 mg L?1. Both 8 and 10 mg L?1 of As drastically downregulated the shoot proteins ranging from 43–65 kDa. With As10 mg L?1, there was a total depletion of protein bands below 23 kDa; however, K80 mg L?1 maximally recovered and upregulated the protein bands. Additionally, protein bands were downregulated (at par with As-alone treatment) above K80 mg L?1 level. Interestingly, As-stress increased lepidine content in a dose-dependent manner which was further augmented with the K-supplementation. It is suggested that K protects L. sativum against As-toxicity by decreasing its accumulation and strengthening antioxidant defense system and protein stability.  相似文献

Five studies of the background level of several perfluorocarbon compounds in Europe are here presented together with measurements from the European Tracer Experiment (ETEX). The tracers used during the two ETEX tracer releases were the perfluorocarbons (PFCs); perfluoromethylcyclohexane (C7F14, PMCH) and perfluoromethylcyclopentane (C6F12, PMCP). Their background concentrations were detected by using both passive and active sampling techniques, to define the spatial and temporal variation of the PFCs over Europe. Also the background variations of four isomers of the PFC compound perfluorodimethylcyclohexane (C8F16, PDCH) were studied. The results were compared to other PFC tracer studies in the U.S.A. and Europe. The mean and median values of the measured PFCs were found to vary slightly and randomly in space and time. They were found to be higher and to have a larger standard deviation than the measurements from the American studies. The background concentrations were still found to be low and stable enough for PFCs to be highly suitable for use in tracer studies. The following concentrations were found: PMCP; 4.6±0.3 fl ℓ-1, PMCH: 4.6±0.8 fl ℓ-1, ocPDCH: 0.96±0.33 fl ℓ-1, mtPDCH: 9.3±0.8 fl ℓ-1, mcPDCH: 8.8±0.8 fl ℓ-1, ptPDCH: 6.1±0.8 fl ℓ-1. A study of the correlation between the measured PFC compounds showed a significant correlation between most of the compounds, which indicate that there are no major PFC sources in Europe.  相似文献

The object of this study was to assess the capacity of Populus alba L. var. pyramidalis Bunge for phytoremediation of heavy metals on calcareous soils contaminated with multiple metals. In a pot culture experiment, a multi-metal-contaminated calcareous soil was mixed at different ratios with an uncontaminated, but otherwise similar soil, to establish a gradient of soil metal contamination levels. In a field experiment, poplars with different stand ages (3, 5, and 7 years) were sampled randomly in a wastewater-irrigated field. The concentrations of cadmium (Cd), Cu, lead (Pb), and zinc (Zn) in the poplar tissues and soil were determined. The accumulation of Cd and Zn was greatest in the leaves of P. pyramidalis, while Cu and Pb mainly accumulated in the roots. In the pot experiment, the highest tissue concentrations of Cd (40.76 mg kg?1), Cu (8.21 mg kg?1), Pb (41.62 mg kg?1), and Zn (696 mg kg?1) were all noted in the multi-metal-contaminated soil. Although extremely high levels of Cd and Zn accumulated in the leaves, phytoextraction using P. pyramidalis may take at least 24 and 16 years for Cd and Zn, respectively. The foliar concentrations of Cu and Pb were always within the normal ranges and were never higher than 8 and 5 mg kg?1, respectively. The field experiment also revealed that the concentrations of all four metals in the bark were significantly higher than that in the wood. In addition, the tissue metal concentrations, together with the NH4NO3-extractable concentrations of metals in the root zone, decreased as the stand age increased. P. pyramidalis is suitable for phytostabilization of calcareous soils contaminated with multiple metals, but collection of the litter fall would be necessary due to the relatively high foliar concentrations of Cd and Zn.  相似文献

The Freundlich equation, X/m = K Ce N, is frequently used to express the partitioning of pesticides between solid (soil, sediment) and aqueous phases. When designing adsorption‐desorption experiments, it would be useful to be able to estimate the equilibrium concentration, Ce, in terms of the initial concentration, Co, if reasonable estimates of K and N can be made. Because of the usual non‐integer nature of N, only a numerical solution of the Freundlich equation is possible for Ce in terms of Co. A simple computer program is presented which produces almost an exact solution for the value of Ce.  相似文献

A gas chromatography–mass spectrometry method has been proposed for the determination of low-level mutagenic and carcinogenic nitrosamines in particulate matter. The method includes the collection of particulate matters (PM2.5 and PM10) using a dichotomous Partisol 2025 sampler and extraction of the compounds from aqueous solution with dichloromethane/2-propanol after sonication with a slightly basic water solution prior to their GC-MS analysis in electron impact mode. The obtained recoveries of nitrosamines ranged from 92.4 to 99.2 %, and the precision of this method, as indicated by the relative standard deviations, was within the range of 0.95–2.46?%. The detection limits obtained from calculations using the GC-MS results based on S/N?=?3 were found within the range from 4 to 22 pg/m3. The predominant nitrosamines determined in particulate matter were N-nitrosodimethylamine, N-nitrosodiethylamine, N-nitrosodibutylamine and N-nitrosomorpholine. Furthermore, N-mono- and dinitrosopiperazine and N-nitrosoethylbutylamine were also determined. N-dinitrosopiperazine was detected in PM2.5 samples at the highest concentrations of up to 22.85 ng/m3 and in PM2.5–10 samples at concentrations up to 7.60 ng/m3 in winter, whereas it was found in PM2.5 samples up to 5.15 ng/m3 and in PM2.5–10 samples up to 3.12 ng/m3 in summer. The total concentrations of nitrosamines were up to 161.4 ng/m3 in fine and 53.90 ng/m3 in coarse fractions in winter, whereas in summer were up to 35.24 and 12.60 ng/m3, respectively. The concentration levels of nitrosamines fluctuated significantly within a year, with higher means and peak concentrations in the winter compared to that in the summertime. The seasonal variations of particle-associated nitrosamine concentrations were investigated together with their relationships with meteorological parameters using Pearson’s correlation analysis in the winter and summer periods. Analysis of variance was used to determine which concentrations of nitrosamines were statistically different from one another and, together with meteorological parameters and discriminant analysis, was used to classify the particle samples by particle size according to seasons. The classification results of the particle samples in different seasons were very satisfactory, allowing 99.5 % of cases to be correctly grouped.  相似文献

Sawdust, one of the materials used as sorbent for removing spilled oil from polluted environments was naturally colonized by hydrocarbon-utilizing fungi, 1 × 105-2 × 105 colony forming units (CFU) g−1, depending on the hydrocarbon substrate. This sorbent was initially free of hydrocarbon-utilizing bacteria. Incubating wet sawdust at 30 °C resulted in gradually increasing the fungal counts to reach after 6 months between 5 × 106 and 7 × 106 CFU g−1, and the appearance of hydrocarbon-utilizing bacteria in numbers between 8 × 104 and 3 × 105 cells g−1. The fungi belonged to the genera Candida (32% of the total), Penicillium (21%), Aspergillus (15%), Rhizopus (12%), Cladosporium (9%), Mucor (7%) and Fusarium (4%). Based on their 16S rRNA gene sequences the bacteria were affiliated to Actinobacterium sp. (38%), Micrococcus luteus (30%), Rhodococcus erythropolis, (19%) and Rhodococcus opacus (13%). Individual pure fungal and bacterial isolates grew on a wide range of individual pure aliphatic (n-alkanes with chain lengths between C9 and C40) and aromatic (benzene, biphenyl, anthracene, naphthalene and phenanthrene) hydrocarbons as sole sources of carbon and energy. Quantitative determinations revealed that all fungal and bacterial isolates could consume considerable proportions of crude oil, phenanthrene (an aromatic hydrocarbon) and n-hexadecane (an aliphatic hydrocarbon) in batch cultures. It was concluded that when sawdust is used as a sorbent, the associated microorganisms probably contribute to the bioremediation of oil and hydrocarbon pollutants in the environment.  相似文献
