The main objective of this work was to investigate the kinetic characteristics of acid and alkaline phosphatases isolated from different sources and to study the effects of the herbicide atrazine and insecticide methomyl on the activity and kinetic properties of the enzymes. Acid phosphatase (ACP) was isolated from the tomato plant (Solanum lycopersicum L. var. lycopersicum); alkaline phosphatase (ALP) was isolated from two sources, including mature earthworms (Aporrectodea caliginosa) and larvae of the Egyptian cotton leafworm (Spodoptera littoralis). The specific activities of the enzymes were 33.31, 5.56 and 0.72 mmol substrate hydrolyzed per minute per milligram protein for plant ACP, earthworms ALP and cotton leafworm ALP, respectively. The inhibition kinetics indicated that atrazine and methomyl caused competitive–non-competitive inhibition of the enzymes. The relationships between estimates of Km and Vmax calculated from the Michaelis–Menten equation have been explored. The extent of the inhibition was different, as estimated by the values of the inhibition constant Ki that were found to be 3.34 × 10?3, 1.12 × 10?2 and 1.07 × 10?2 mM for plant ACP, earthworms ALP and cotton leafworm ALP, respectively, with methomyl. In the case of atrazine, Ki were found to be 8.99 × 10?3, 3.55 × 10?2 and 1.36 × 10?2 mM for plant ACP, earthworms ALP and cotton leafworm ALP, respectively.  相似文献

Gossypol occurs naturally in the pigment glands in cotton. It has a role in protecting cotton plants from insect pests such as bollworms, yet it was not toxic to the cotton leafworm larvae Spodoptera littoralis up to 2-5% concentration in artificial diet or at 125 micrograms/ larva by topical application. The compound inhibited protease and lipid peroxidase activities in larvae with in vitro I50 values of 1.5 X 10(-3)M, and 4.4 X 10(-4)M respectively. When gossypol was fed to Spodoptera larvae, it stimulated the microsomal N-demethylase in vitro. This inductive effect was time-dependent similar to that of phenobarbital. Gossypol stimulates ATPase at lower concentrations and inhibited it at higher concentrations. The I50 for mitochondrial ATPase was 1.7 X 10(-4)M, while the corresponding values for DDT and fenvalerate were 1.1 X 10(-4)M and 7.0 X 10(-4)M respectively. Gossypol at 1.5% concentration in the diet reduced the larval weight to 50% of the control within two days, and increased the duration of each larval stage. The number of eggs and their hatchability was seriously decreased in larvae treated for three consecutive generations. Such an effect can be attributed to the ability of gossypol to interfere with protein bio-synthesis.  相似文献

Resistant biotypes have a slightly higher activity than the susceptible biotype. Tolerance of cotton to MSMA may be related to glutathione synthetase activity and possibly to the presence of phytochelatins. Also, increased glutathione levels in the resistant biotype may implicate phytochelatin involvement in the resistance mechanism.  相似文献

Abstract

Adult and larval insects from the terrestrial and aquatic environments were exposed to acephate. The chemical was more toxic to adult insects than to larvae, and was a poor insect cholinesterase inhibitor in vitro compared to methamidophos which was a much stronger inhibitor. Both acephate and methamidophos inhibited the adult cholinesterase in vitro much more strongly than they did the larval enzymes. Acephate was metabolized by the insects to methamidophos which did not appear to be the only metabolite, although no other metabolites were looked for. The cholinesterase of insects exposed to sublethal levels of acephate was inhibited, but this inhibition appeared to be due to the combined effect of acephate and methamidophos and not to any hypothetical substance with greater anticholinesterase activity. This was bourne out when acephate was incubated with mixed function oxidases (MFO). No activated product with potent anticholinesterase activity was identified. Methamidophos was not produced by the MFO system but by some other unidentified mechanism.  相似文献

Insecticides have long been used as the main method in limiting agricultural pests, but their widespread use has resulted in environmental pollution, development of resistances, and biodiversity reduction. The effects of insecticides at low residual doses on both the targeted crop pest species and beneficial insects have become a major concern. In particular, these low doses can induce unexpected positive (hormetic) effects on pest insects, such as surges in population growth exceeding what would have been observed without pesticide application. Methomyl and chlorpyrifos are two insecticides commonly used to control the population levels of the cotton leafworm Spodoptera littoralis, a major pest moth. The aim of the present study was to examine the effects of sublethal doses of these two pesticides, known to present a residual activity and persistence in the environment, on the moth physiology. Using a metabolomic approach, we showed that sublethal doses of methomyl and chlorpyrifos have a systemic effect on the treated insects. We also demonstrated a behavioral disruption of S. littoralis larvae exposed to sublethal doses of methomyl, whereas no effects were observed for the same doses of chlorpyrifos. Interestingly, we highlighted that sublethal doses of both pesticides did not induce a change in acetylcholinesterase activity in head of exposed larvae.  相似文献

Pseudomonas fluorescens SM1 isolate was found to be resistant to some major water pollutants namely Cd2+, Cr6+, Cu2+, Ni2+, Pb2+, BHC, 2,4-D, mancozeb and phenols up to a concentration four times to the normal levels occurring in the highly pollulated regions. Curing experiment brought about the loss of one or more resistance markers indicating the plasmid born resistance. Plasmid profile of SM1 strain showed the presence of one DNA band of 43.6 kb. This Plasmid was isolated from SM1 strain and introduced into Escherichia coli DH5 alpha with a transformation frequency of 6.7 x 10(-4)transformants/recipient cell. The test SM1 strain was also capable of biotransforming Cr(VI) to Cr(III) which is less toxic compounds. Present studies further indicated that the test SM1 strain was not only resistant to 2,4-D, phenols and catechol but also capable of bioremediating these toxicants quite efficiently. Moreover, studies with inhibitors like sodium azide, 2,4-DNP and chloramphenicol suggested that the major mechanism for the bioremediation of the heavy metals other than Cr6+ would be the biosorption process.  相似文献

Effects of the botanical extract (myrrh), Commiphora molmol, and the sublethal treatments of profenofos/chlorofluazuron, fenvalerate and pyriproxyfen on the larvae of S. littoralis were investigated. The results showed that myrrh induced the highest activity after 7 days of treatment with concentration of 10000 ppm and the percentage of mortality reached 44.4%. The chemical insecticide profenofos/chlorofluazuron appeared to be the most effective treatment, giving a mortality of 54% after one day of treatment with a concentration of 100 ppm. Different treatments showed adverse effects on pupation, emerging adults and larval duration. The highest concentration of myrrh (10000 ppm) induced 35% pupation. No pupa was obtained with profenofos/chlorofluazuron while fenvalerate and pyriproxyfen gave 13.3% at 50 ppm. At 10000 ppm of myrrh, 25% adult was emerged, also fenvalerate and pyriproxyfen decreased the emerging moth to 13.3% at 50 ppm. For control there was no larval mortality at 7 days and the percentages of pupation and adult emergence were 90%. Bioassays were also conducted to test the joint action of myrrh applied in binary mixtures with each chemical insecticide on Spodoptera larvae. Results of the bioassay of mixtures indicated antagonistic effects on larval mortality. However, combination of myrrh with profenofos/chlorofluazuron showed additive effect. Also, the degree of pupation and emerging moth varied considerably when myrrh combined with the chemical insecticides. The present work shows the strong efficacy of the botanical extract (myrrh) which could be used alone or in combination with sublethal doses of certain insecticides to control the cotton leafworm.  相似文献

The house fly, Musca domestica L., a relevant sanitary pest, is mainly controlled by insecticides. However, an inappropriate use of chemicals may induce resistance, treatment efficacy decline and environmental damages. We evaluated toxicity of some organophosphates, pyrethroids, spinosad and neonicotinoids by topical applications on adults of a M. domestica strain (OcRo) collected from an intensive chicken farm in Northern Italy, in comparison to a susceptible strain (s-DBF). The OcRo strain exhibited higher levels of resistance (RR??) to four pesticides in comparison to s-DBF. Spinosad and imidacloprid had very low RR??, thus were still efficient in OcRo control. We also tested resistance to pesticides in OcRo after topical applications of synergistic compounds. Hydrolases were involved in phosphorganic detoxification and cytochrome P450 monoxygenases in that of pyrethroids. These results indicate that OcRo strain is now multiresistant to organophosphates and pyrethroids, and this should be considered for an environmentally safe pest management.  相似文献

Resistance to acetolactate synthase (ALS)-inhibiting herbicides in Brazil has been documented for six species. The probability to select biotypes of Euphorbia heterophylla (EPPHL) with multiple resistance increases in the same order of magnitude as the use of other herbicides belonging to only one mechanism of action. The objectives of this work were to evaluate the distribution of resistant populations (R) in the states of the Parana and Santa Catarina; to determine the existence of populations of EPHHL with multiple resistance to ALS and PROTOX inhibitors, and to confirm the occurrence of cross resistance to compounds of these mechanisms of action. Seeds of EPHHL of areas with suspected resistance had been sampled in 97 places during 2003. In the greenhouse experiment samples of each population were sprayed with imazethapyr or fomesafen, at only one rate. To identify the resistant ones they were sprayed with different levels of the herbicides imazethapyr and fomesafen. Later they were sprayed with diverse herbicides of the same mechanisms of action to confirm the multiple/cross resistance. There is widespread distribution in the region of populations with resistance to ALS inhibitors. Some biotypes demonstrated resistance to herbicides from the two mechanisms of action. The resistance factor (FR), or the relation of resistance between R and susceptible biotypes, confirms the existence of two biotypes of EPHHL with cross resistance to several herbicides inhibitors of ALS and PROTOX.  相似文献

Relative sensitivity of five common Egyptian plant species namely, Senecio vulgaris, Malva parviflora, Sonchus oleraceus, Medicago sativa and Melilotus indicus to elevated levels of ozone has been studied. The plants were exposed to charcoal filtered air (CFA) and different levels of O3 (50 and 100 ppb) for 5 h per day. The studied parameters were recorded for five consecutive days after fumigation. The foliar injury varied significantly among species in a dose-dependent manner. Severe injury symptoms were recorded on the leaves of M. sativa. With the exception of M. parviflora, all species exhibited significant increases in the percentage reduction of the above-ground dry weight as a result of reductions in both leaf and stem dry weights. M. sativa showed a marked reduction in its relative growth rate at elevated levels of O3. The extent of chlorophyll a destruction was higher in both M. sativa and S. oleraceus than in the other species tested. No differences in the sensitivity of chlorophylls a+b and carotenoids to ozone levels were recorded in this work. Percentage reduction of ascorbic acid was higher in M. sativa and S. oleraceus, compared with the other species studied. With respect to relative percentages of proline, there was a significant difference in the responses of plants to ozone. According to the ozone resistance (R%), measured as relative growth rate, the test species were arranged in the descending order: M. parviflora>M. Indicus>S. Vulgaris>S. Oleraceus>M. sativa. In M. sativa, both determinant and correlation coefficients are well reflected in the relationship between its physiological response, its performance and ozone levels, supporting its recommendation as a candidate for biomonitoring in Egypt.  相似文献

Field populations of daphnids historically-stressed by metal contamination may show increased resistance to those contaminants. This study was undertaken aiming to confirm/infirm three main hypotheses: (1) field populations living in historically-impacted environments are more tolerant to metal stress than populations from reference sites; (2) resistance differences are genetically-determined, i.e., differences persist after controlling for environmental and maternal effects, by acclimating cloned lineages to similar conditions; and (3) resistance to stress in field populations living in historically-impacted environments is due to the disappearance of sensitive individuals rather than the appearance of highly resistant ones, i.e., the shift in the central tendency of resistance is linked to a decrease in the range of population resistance and not to an increased upper limit of the population resistance. Three populations of the cladoceran Ceriodaphnia pulchella Sars in Southern Portugal were sampled; one of which has been historically-stressed by acid mine drainage (AMD) from an abandoned cupric-pyrite mine and two from reference sites within the same watershed. To assess if resistance differences were genetically-determined, the three populations were acclimated for at least five generations under the same controlled conditions. Assays with AMD contaminated water samples were performed with both non-acclimated and acclimated individuals from all studied populations. Reproduction results in sub-lethal assays revealed significant differences between the reference and stressed populations. Significant differences in resistance to lethal levels of toxicity were observed for both non-acclimated and acclimated populations, individuals from population I being more resistant than those from reference populations. The existence of genetically-determined sensitivity differences was attested by the presence of significant differences in resistance to lethal levels of toxicity in acclimated individuals from reference and stressed populations. Results from cumulative mortality assays revealed that sensitive individuals were most probably present in the original population, but no conclusion could be draw about the presence of extreme resistant individuals in the historically-stressed population. Finally, it was shown that responses among populations converged from high to low levels of contamination.  相似文献

Azinphos-methyl is an organophosphate insecticide used for pest control on a number of food crops in many parts of the world. The oligochaete Lumbriculus variegatus and pigmented and non-pigmented specimens of the gastropod Biomphalaria glabrata are freshwater invertebrates that have been recommended for contamination studies. Recently, it has been shown that L. variegatus worms exhibit a higher cholinesterase (ChE) activity and a greater sensitivity to in vivo ChE inhibition by azinphos-methyl than pigmented B. glabrata snails. The aims of the present study were (1) to investigate if, in addition to its anticholinesterase action, azinphos-methyl has also pro-oxidant activity in L. variegatus and B. glabrata, and (2) to examine if species that are highly susceptible to the neurotoxic effects of organophosphates also suffer a greater degree of oxidative stress. Therefore, total glutathione (t-GSH) levels and activities of cholinesterase (ChE), superoxide dismutase (SOD), catalase (CAT), glutathione S-transferase (GST), and glucose 6-phosphate dehydrogenase (G6PDH) were measured in the whole body soft tissue of organisms exposed for 48 and 96 h to a level of azinphos-methyl that produces 50% of inhibition on ChE. Results showed different patterns of antioxidant responses between the gastropods and the oligochaetes, and even between the two phenotypes of gastropods: (1) in exposed L. variegatus t-GSH levels increased and CAT and SOD activities decreased with respect to control organisms, (2) in pigmented gastropods, SOD decreased while CAT transiently diminished, and (3) in non-pigmented gastropods, SOD activity showed a biphasic response. GST and G6PDH were not altered by azinphos-methyl exposure. Of note, t-GSH levels were 4-fold times higher in L. variegatus than in both phenotypes of B. glabrata. This may suggest that GSH could play a more important role in antioxidant defense in L. variegatus than in B. glabrata.  相似文献

Resistance to fenitrothion and enzyme activities associated with the toxicity and metabolism of organophosphorus insecticides were measured in three genetically unique Daphnia magna clones collected from rice fields of Delta del Ebro (NE Spain) during the growing season and a lab sensitive clone. The studied clones showed up to sixfold differences in resistance to fenitrothion. The lack of correlation between in vitro sensitivity of acetylcholinesterase (AChE) to fenitrooxon and resistance to fenitrothion indicated that insensitivity of AChE to the most active oxon metabolite was not involved in the observed differences in resistance. Inhibition of mixed- function oxidases (MFOs) by piperonyl butoxide (PBO) increased the tolerance to fenitrothion by almost 20-fold in all clones without altering their relative ranking of resistance. Conversely, when exposed to fenitrooxon, the studied clones showed similar levels of tolerance, thus indicating that clonal differences in the conversion of fenitrothion to fenitrooxon by MFOs were involved in the observed resistance patterns. Despite that resistant clones showed over 1.5 higher activities of carboxilesterase (CbE) than sensitive ones, toxicity tests with 2-(O-cresyl)-4H-1,3,2-benzodioxaphosphorin-2 oxide, which is a specific inhibitor of these enzymes, evidenced that this system only contributed marginally to the observed clonal differences in tolerance. Glutathione-S-transferases activity (GST) varied across clones but not under exposure to fenitrothion, and was only related with tolerance levels in the field clones. In summary, our results indicate that MFO mediated differences on the bio-activation of the phosphorotionate OP pesticide to its active oxon metabolite contributed mostly in explaining the observed moderate levels of resistance, whereas the activities of CbE and GST had only a marginal role.  相似文献

Agrobacterium tumefaciens-mediated transformation (ATMT) was used to generate an insertional mutant library of the mycelial fungus Trichoderma harzianum. From a total of 450 mutants, six mutants that showed significant influence on maize resistance to C. lunata were analyzed in detail. Maize coated with these mutants was more susceptible to C. lunata compared with those coated with a wild-type (WT) strain. Similar to other fungal ATMT libraries, all six mutants were single copy integrations, which occurred preferentially in noncoding regions (except two mutants) and were frequently accompanied by the loss of border sequences. Two mutants (T66 and T312) that were linked to resistance were characterized further. Maize seeds coated with T66 and T312 were more susceptible to C. lunata than those treated with WT. Moreover, the mutants affected the resistance of maize to C. lunata by enhancing jasmonate-responsive gene expression. T66 and T312 induced maize resistance to C. lunata infection through a jasmonic acid-dependent pathway.  相似文献

The goal of this study was to contribute to understanding of the mechanisms behind sensitivity differences between early and late instar larvae of Chironomus riparius and to address the influence of the differences in standard testing approaches on the toxicity evaluation. A 10-day contact sediment toxicity test was carried out to assess sensitivity to cadmium exposure in relation to different age and laboratory culture line origin of test organisms. Chironomid larvae of early (OECD 218 method) and late instar (US-EPA600/R-99/064 method) differed substantially in sensitivity of traditional endpoints (OECD: LOEC 50 and 10 μg Cd/g dry weight (dw); US-EPA: LOEC?>?1000 and 100 μg Cd/g dw for survival and growth, respectively). Bioaccumulated cadmium and metallothioneins (MTs) concentrations were analyzed to investigate the role of MTs in reduced sensitivity to cadmium in late instar larvae. Metallothioneins were induced after treatment to greater Cd concentrations, but their levels in relation to cadmium body burdens did not fully explain low sensitivity of late instars to cadmium, which indicates some other effective way of detoxification in late instars. This study brings new information related to the role of MTs in age-dependent toxicant sensitivity and discusses the implications of divergence in data generated by chironomid sediment toxicity tests by standardized methods using different instars.  相似文献

Invasive mosquitoes are economic and sanitary concerns especially in Europe and America. Most work has emphasized the role of resistance [Berrada, S., Fournier, D., Cuany, A., Nguyen, T.X., 1994. Insect Biochemistry and Molecular Biology 34, 653-665] to insecticides. Compounds acting on larval sensitivity to insecticides are not well studied and their action remains poorly understood. Among several residual chemicals in ecosystems, particularly in wetlands, we identified a possible interaction of an herbicide on larval resistance to an insecticide. Our work contributes to the global control of mosquito populations by identifying possible pathways of resistance to insecticides of these vectors. Resistance or tolerance to insecticide treatments might contribute to successful invasion by mosquitoes. Here we report an ecotoxicological approach to test the hypothesis of an indirect effect of atrazine on mortality of an invasive vector. A brief contact (48h) between Aedes aegypti mosquito larvae and atrazine led to a modification of larval sensitivity to an insecticide: using atrazine as an inducer led to a decrease in the mortality of larvae treated with Bacillus thuringiensis var. israelensis (Bti).  相似文献

The aim of this study was to test and select one or more highly sensitive, specific and environmentally successful Egyptian bioindicator plants for ozone (O3). For that purpose more than 30 Egyptian species and cultivars were subjected to extensive screening studies under controlled environmental and pollutant exposure conditions to mimic the Egyptian environmental conditions and O3 levels in urban and rural sites. Four plant species were found to be more sensitive to O3 than the universally used O3-bioindicator, tobacco Bel W3, under the Egyptian environmental conditions used. These plant species, jute (Corchorus olitorius c.v. local), clover (Trifolium alexandrinum L. c.v. Masry), garden rocket (Eruca sativa c.v. local) and alfalfa (Medicago sativa L. c.v. local), ranked in order of decreasing sensitivity, exhibited typical O3 injury symptoms faster and at lower 03 concentrations than Bel W3. Three variables were tested in search of a reliable tool for the diagnosis and prediction of O3 response prior to the appearance of visible foliar symptoms: pigment degradation, stomatal conductance (g(s)) and net photosynthetic CO2 assimilation (Pnet). Pigment degradation was found to be unreliable in predicting species sensitivity to O3. Evidence supporting stomatal conductance involvement in 03 tolerance was found only in tolerant species. A good correlation was found between g(s), restriction of O3 and CO2 influx into the mesophyll tissues, and Pnet. Changes in Pnet seemed to depend largely on fluctuations in g(s).  相似文献

Thiram-induced toxic liver injury in male Sprague-Dawley rats   总被引:1,自引:0,他引:1  
A single i.p. dose (120 mg/kg) of thiram given to male Sprague-Dawley rats caused a significant increase in the activity of SGOT and SGPT 24 hr post-treatment indicating liver damage. A considerable diminution in the serum cholinesterase activity was also noted in the treated rats as against the control animals. Additional evidence for thiram-induced liver toxicity is provided by the observation that there was approximately 50% inhibition of the activity of hepatic microsomal benzphetamine N-demethylase with a concomitant decrease in the concentration of cytochrome P-450, an important component of the mixed-function oxidase system. Although not significant, hepatic glutathione levels were also depleted by thiram, probably making the liver susceptible to toxic injury.  相似文献
