In this study, we investigated the toxic effects of water-soluble elements from a contaminated soil via gavage in a single dose, simulating a geophagy event. The contaminated soil was collected in a field located in an industrial complex, and the control soil was collected in a reference area. Metabolic and behavioral parameters in Wistar male rats were measured after 24 and 96 h of gavage. After 96 h, the major organs were weighed, blood was collected to check hematological parameters, the bone marrow was taken for the micronucleus test, and the liver was used for evaluating the total antioxidant capacity, lipoperoxidation and protein carbonylation. Animals exposed to contaminated soil presented a few significant alterations by comparison with control animals: TBARS and protein carbonyl levels increased, the relative weight of the kidneys increased, metabolic parameters (body weight gain, food intake, water consumption, urine and feces production) depressed and there was behavioral alteration. These findings suggest that soils impacted by atmospheric contaminants can affect the organism physiological status jeopardizing the health of populations living in industrial areas. Finally, this study reassures that ingestion of potentially contaminated soils, even for short periods of time, can cause health risks.  相似文献

Persistent organic pollutants such as Polychlorinated biphenyls (PCB) and dichloro diphenyl trichloroethane (DDT) are known to exert various adverse health effects in wildlife mammals. The impact from dietary intake of minke whale (Balaenoptera acutorostrata) blubber high in organohalogen and other chemical contaminants on renal morphology and function was investigated via a controlled study on West Greenland sledge dogs (Canis familiaris). Our results showed significantly higher frequencies of glomerular, tubular, and interstitial lesions in the exposed group. Furthermore, higher urine protein?:?creatinine ratio and plasma urea levels were found in the exposed group, which indicated a negative impact on kidney function via tubular and glomerular dysfunctions. The lesions were similar to those observed in top predator marine species–such as seals and polar bears–and humans exposed via diet to persistent environmental pollutants. The lesions may have impacts on the overall health of these animals. It is reasonable to suggest that chronic exposure to organic pollutants via a country diet of high trophic level marine organisms may have an impact on renal morphology and function in Arctic apex predators in general as well as Inuit people.  相似文献

A comparison of PCB congener profiles and limited biological effects was made between direct exposure to PCB-contaminated soil and vapor phase PCBs from that soil to determine congener patterns useful for identifying exposure sources in humans and wildlife. Weanling female Sprague–Dawley rats were exposed to either control or PCB-contaminated soil (from a landfill in Southern Illinois) for 1 and 2 weeks. The exposures were via direct contact with the soil or via airborne exposure with the rats isolated from the soil by a wire screen. Total PCB of 25% contaminated soil used in the study was 13?500?ppm. No PCBs were detectable in control rats. In direct-exposed rats, total PCB residues in fat pad, ear skin, serum, liver, and inguinal lymph nodes after the 1-week exposure were 6256, 185, 3.2, 149, and 41?ppm, respectively, but decreased to 465, 72, 1.7, 106, and 32.4?ppm after the 2-week exposure. In airborne-exposed rats, total PCB residues were 7.8, 1.6, 0.03, 0.2, and 0.6 in the same manner and slightly increased in fat pad and ear skin to 11.6 and 2.14, respectively. Decreases in both the concentrations and percentages of “episodic” PCBs (those congeners rapidly metabolized) in the fat pad were apparent following the 2-week exposure compared to the 1-week exposure by both routes. Both EROD and BROD activities were significantly increased in the direct-exposed rats, whereas only BROD activity increased in airborne-exposed rats. Serum T 4 levels were depleted in the direct-exposed rats regardless of time of exposure but were increased insignificantly after 1-week and significantly after 2 weeks in the airborne-exposed rats. No significant changes in serum insulin levels were apparent in any of the treated groups. The results suggested that exposure of animals to PCBs via different routes could result in different PCB profiles, which could cause different biological effects.  相似文献

The relationship between the chemical con- taminants and soil microbial toxicity of waste foundry sand (WFS) was investigated. Five different types of WFS from typical ferrous, aluminum, and steel foundries in China were examined for total metals, leachable metals, and organic contaminants. The soil microbial toxicity of each WFS was evaluated by measuring the dehydrogenase activity (DHA) of a blended soil and WFS mixture and then comparing it to that of unblended soil. The results show that the five WFSs had very different compositions of metal and organic contaminants and thus exhibited very different levels of soil microbial inhibition when blended with soil. For a given WFS blended with soil in the range of 10wt.%-50wt.% WFS, the DHA decreased almost linearly with increased blending ratio. Furthermore, for a given blending ratio, the WFSs with higher concentrations of metal and organic contaminants exhibited greater microbial toxicity. Correlation analysis shows that the relationship between ecotoxicity and metal and organic contaminants of WFSs can be described by an empirical logarithmic linear model. This model may be used to control WFS blending ratios in soil-related applications based on chemical analysis results to prevent significant inhibition of soil microbial activity.  相似文献

Tobacco exposure may alter homeostasis of iron (Fe), one of the most abundant and essential transition metals in the body. In this study, the effect of aqueous extract of smokeless tobacco was evaluated on Fe homeostasis in rats and human hepatoma, HepG2 cells. Our findings suggested that tobacco consumption even at low doses impairs Fe homeostasis leading to Fe deficiency anemia. Significant alterations were noted with respect to hematological parameters and expression patterns of selected intestinal Fe-transporters, Fe-binding proteins, and Fe-regulatory hormone, hepcidin. Impairment in the hepatic and renal antioxidant defense system was also observed in the treated rats. Histopathological studies revealed cirrhosis of liver and goblet cell hyperplasia of small intestine. Further, analysis of hepcidin promoter and its expression along with ferritin (expression and ELISA) in HepG2 cells demonstrated an enhanced expression of both the genes resulting in sequestration of Fe in treated cells, thus indicating systemic Fe deficiency.  相似文献

Cypermethrin is a synthetic pyrethroid insecticide used for pest control in agriculture and as an acaricide in man and animals. This study was undertaken with the objective to investigate the propensity of cypermethrin to induce oxidative stress in rats following repetitive dermal exposure and its possible attenuation by L-ascorbic acid. Results obtained showed that cypermethrin significantly (p < 0.05) increased malonaldehyde levels, activity of catalase in rat erythrocytes and plasma protein levels. Whereas, activities of glutathione (GSH), glutathione peroxidase (GSH-Px), and superoxide dismutase (SOD) were significantly (p < 0.05) reduced in the cypermethrin exposed rats as compared to the control. Supplementation of L-ascorbic acid in cypermethrin-exposed rats decreased lipid peroxidation of erythrocytes, total plasma protein and catalase activity significantly (p < 0.05) compared to non-cypermethrin-exposed group. However, L-ascorbic acid did not alleviate the negative effects of cypermethrin on the activities of SOD and GSH. This study revealed that the presence of L-ascorbic acid diminishes the adverse effects of cypermethrin on some oxidative stress parameters.  相似文献

The potential risks from oral intake of soil antimony (Sb) depends mainly on the amount of metal ingested and its bioavailability. Relative bioavailability may be determined by comparing Sb present in soil to a reference compound, taking into account accumulation in different target tissues or excretion. However, due to the lack of scientific knowledge concerning the fate of Sb in the organism, there is a need to study the absorption and distribution of Sb in order to select target tissues for assessment of bioavailability of Sb in soils. Thus, 45 piglets were exposed to a soluble pentavalent antimony salt (KSb(OH)6), for 15 days at concentrations ranging from 0–1600 µg Sb/kg body weight (BW) per day. Following the exposure period, blood, plasma, liver, spleen, kidneys, hair, bone, bile and urine were obtained to measure Sb concentrations by ICP-MS. Results showed that tissue Sb levels were dose-related. Higher Sb concentrations were found in urine, kidneys, hair, bone and liver. Sb(V) was not detectable in blood and plasma. In the case of highly contaminated soil with soluble forms of Sb in concentrations ranging from 200–1600 µg Sb/kg BW, kidneys, liver and spleen are the most reliable compartments to determine Sb bioavailability from soil. However, for the soils with lower levels of contamination and a low Sb bioaccessibility, urine may serve as a relevant compartment.  相似文献

There is increasing concern for the disruptive effects seen in aquatic species exposed to environmental contaminants. However, few studies have investigated the impact of such contaminants on the behavior of individuals living in exposed waters. Contaminant exposure can affect animal populations by disrupting behaviors including feeding, locomotion, and mating. In this study, we examined how living in an ecosystem polluted by combinations of polycyclic aromatic hydrocarbons, polychlorinated biphenyls, and heavy metals (arsenic, cadmium, iron, lead, zinc) impacts contest behavior in the round goby (Neogobius melanostomus). Fish collected from heavily contaminated and cleaner sites in Lake Ontario were subjected to a resource contest to determine the effect of these contaminants on aggression and the establishment of dominance hierarchies, which in turn influence access to food, shelter, and mating opportunities. Dominance establishment (a clear resource winner) was less obvious among fish from the contaminated site compared to the more stable hierarchies that formed between pairs of fish from the clean site. Pairs of fish from the contaminated site performed more assessment displays compared to fish from clean sites. These results suggest that the costs of living in an environment under exposure can shape behavioral repertoires. The altered conflict resolution strategies of contaminated fish may reflect impaired cognitive function, sensory perception, and/or higher metabolic load associated with aggression. This study provides support for the utilization of quantifiable behavioral differences as ecologically relevant measures of contaminant exposure.  相似文献

为研究沙尘暴源区土壤盐碱扬尘对大鼠呼吸系统的免疫损伤,采集距北京200多公里的张家口张北地区的安固里诺尔干盐湖(N41.316°,E114.350°)0~5 cm的盐碱表土,过200目(75μm)筛,测试土壤盐碱扬尘中水溶性离子。将32只雄性7周龄Wistar大鼠随机分为4组,即生理盐水对照组、低浓度组(1.5 mg·kg~(-1))、中浓度组(7.5 mg·kg~(-1))、高浓度组(37.5 mg·kg~(-1)),配制颗粒物悬液,采用气管滴注急性染毒方式,24 h后处死动物,检测血常规及血浆和肺泡灌洗液(BALF)的总抗氧化能力(T-AOC)、白介素-6(IL-6)、肿瘤坏死因子(TNF-α)、乳酸脱氢酶(LDH)、转化生长因子(TGF-β1)等氧化炎症因子,并制作肺组织和气管病理切片。结果表明,沙尘暴源区土壤盐碱扬尘中含有大量的SO_4~(2-)、NO_3~-和NH_4~+等离子;土壤盐碱扬尘急性染毒会对大鼠气管和肺组织造成明显的炎症病变,并随着染毒试剂浓度的升高而加重;但对大鼠血常规的影响较小。中、高浓度染毒使血浆中T-AOC显著下降(P0.05);染毒促使血浆TGF-β1分泌显著增加(P0.05),但不存在剂量-效应关系,其他氧化炎症指标分泌无显著变化。中、高浓度组BALF的TGF-β1因子分泌量显著高于生理盐水对照组(P0.05),高浓度组BALF中IL-6含量显著低于生理盐水对照组(P0.05),其他氧化炎症指标分泌无显著变化。这些研究显示土壤盐碱扬尘粒径大小及化学成分可能是造成大鼠氧化炎症差异的原因。  相似文献

The potential risk of ground-water contamination through the infiltration of contaminants originating from suspended matter and sediments deposited within flood retention areas in the Upper Rhine valley, as well as subsequent problems for drinking water production, were analyzed by means of the bacterial dehydrogenase activity (DHA; sediment contact test) assay with Arthrobacter globiformis and in a prolonged embryo toxicity assay using zebrafish (Danio rerio). Since the direct uptake of contaminants from particle-bound sources has usually been assumed to play only a minor role in bioaccumulation, the availability of organic extracts to biota has generally been regarded as being overestimated. In order to confirm or discard this hypothesis, organic (acetone) extracts of recently suspended, freeze-dried matter, as well as freeze-dried sediments collected from Rhine flood plains (i.e. soil samples) were compared to corresponding native samples in order to screen for the relative availability of particle-bound contaminants. Whereas a significantly higher ecotoxicological burden was observed for some native soil samples, organic extracts of suspended matter proved to be more toxic than corresponding native samples when compared to organic extracts using each test system. With respect to organic extracts, the suspended matter samples were more toxic than soil samples taken from a rarely flooded site. In contrast to the native samples, the rarely flooded soils revealed a higher toxicity than suspended matter. Therefore, organic extracts did not necessarily overestimate the toxicity of soil and suspended matter samples. Moreover, considerable toxicity of particle-bound contaminants to both bacteria and fish embryos, even in native samples, corroborate the hypothesis that particle-bound contaminants may be more readily available than is generally assumed.  相似文献

Heavy metal in soil samples and in washed and unwashed samples of Telfaria occidentalis (ugwu) and Talinum triangulare (waterleaf) cultivated on the bank of river Ribila in Odo-nla village were determined. The soil was moderately polluted with cadmium when compared with Federal Environmental Protection Agency standards. The difference between the unwashed and washed plant samples revealed that metal pollutants exist as superficial contaminants on the foliage surface which is the edible portion and if the foliage portion is washed thoroughly it may be safe for dietary consumption. There is no doubt that continuous discharge of effluent and gaseous emissions from the industries located in this area and dumping of domestic wastes into the river may lead to higher concentrations of these heavy metals in the soil and in the tissue of the leafy vegetables cultivated on the river bank over time. This can eventually lead to pollution of the soil and the cultivated plants, which are ready source of food for the people and other organisms in the food chain.  相似文献

Changes occurring in concentrations of certain trace metals and electrolytes viz. chromium, copper, zinc, sodium, potassium, calcium, magenesium and chloride in plasma of rats exposed to intermittent hypobaric hypoxia were evaluated. Batches of Sprague-Dowley rats (12 in each group) were exposed for 1, 7,14 and 21 days to a simulated altitude 7,620 m for 6 h per day and one group of unexposed animals was kept as control. There was a significant rise of 153% in plasma chromium levels of 1 day exposed group in comparison to the unexposed group which tends to normalise on subsequent exposure. There was a gradual increase in plasma copper levels of 9.0, 28.2, 62.6 and 65.6% respectively in 1,7,14 and 21 days exposed rats in comparison to unexposed rats. On the other hand plasma zinc levels were seen to be decreasing during entire exposure. Plasma sodium levels decreased initially in 1 and 7 day exposed rats and increased in later groups whereas plasma potassium levels of exposed groups remained low in comparison to unexposed group. Chloride levels were found to be elevated in 14 and 21 day exposed groups. The plasma calcium and magnesium levels were higher in all exposed groups over unexposed groups. Changes in chromium, copper and zinc observed in the present study during exposure to hypoxic stress may be responsible for the hyperglycemia and anorexia encountered during intial phase of high altitude acclimatisation.  相似文献

Sparingly soluble contaminants are less likely to affect human health through food chain transfers, such as plant uptake or passage through animal-based foods, because mobility in these pathways is limited by solubility. Direct ingestion or inhalation of contaminated soil becomes the dominant pathway. However, both of these can be selective processes. Clay-sized particles carry the bulk of the sparingly soluble contaminants, and mechanisms that selectively remove and accumulate clay from the bulk soil also concentrate the contaminants. Erosion is another process that selectively removes clays. This project examined the degree of clay and contaminant-concentration enrichment that could occur by these processes, using U, Th and Pb as representative contaminants and using a clay and a loam soil. Erosion by water in natural rainfall events caused concentration enrichments up to 7 fold, and enrichments varied with characteristics of the erosion events. Enrichments were higher for the coarser, loam soil. Adhesion to skin gave modest enrichments of 1.3 fold in these soils, but up to 10 fold in sandy soils studied subsequently. Adhesion to plant leaves, where there was no root contact with contaminated soil, gave leaf concentrations comparable to situations where the roots contacted the contaminated soil. Clearly, adhesion to leaves is an important component of plant accumulation of sparingly soluble contaminants.  相似文献

Previous environmental biomonitoring studies indicated higher environmental lead (Pb) pollution levels at the districts of Aveiro and Leiria (Portugal). In evaluating the risk for human health, which is associated with contaminated soils after oral uptake, total soil concentrations have generally been held against criteria established from toxicological studies based upon the assumption that the uptake of the contaminant is similar in the toxicological studies and from the soils assessed. This assumption is not always valid, as most toxicological studies are carried out with soluble forms of the contaminants, whereas many soil contaminants are or become embedded in the soil matrix and thus exhibit limited availability. This study intends to estimate the soluble fraction of Pb in the soils from central Portugal, and to assess the bioaccessibility of Pb and, hence, infer exposure and risk for human health. Yet, as the physical–chemical properties of the soil exert some control over the solubility of Pb in the surface environment, the relation between such soil properties and the estimated soluble and/or bioaccessible fractions of Pb is also investigated. Other objective, with a more practical nature, was to give some contribution to find a suitable in vitro mimetic of the gastrointestinal tract environment. The results indicate relatively low total metal concentrations in the soils, even if differences between regions were observed. The Aveiro district has the higher total Pb concentration and the metal is in more soluble forms, that is, geoavailable. Soils with higher concentrations of soluble Pb show higher estimates of bioaccessible Pb. Soil pH seems to influence human bioaccessibility of Pb.  相似文献

Ovarian hormones affected some enzymes in reproductive tissues. Previous studies revealed Anethum graveolens (dill) seed extract elevated ovarian hormones. In this study, we compared the effects of dill seed and leaf extracts on some enzymes activities and infertility potential. Female Wistar rats were divided into three groups of control, dill seed and dill leaf aqueous extracts treated groups. Experimental groups received 1 mL of 0.5 g kg?1 of each extract orally for 25 days (five estrous cycles). Estradiol, progesterone, luteinizing hormone, and follicle stimulating hormone concentrations were determined. Uterus and ovaries were removed and their glucose-6-phosphate dehydrogenase, lactate dehydrogenase, and alkaline phosphatase activities were measured. Female rats of three groups were mated with untreated males. Fetuses were extracted in the 20th gestational day and their weights and crown rump lengths were measured. Anti-fertility and anti-implantation activities and fertility indices were calculated. Glucose-6-phosphate activity of dill seed treated group increased 1.6 times in the endometrium and 1.7 times in the myometrium. Progesterone concentration and lactate dehydrogenase activity of serum increased in the dill seed treated group compared to the control and dill leaf treated group. A significant decrease was observed in weights and crown rump lengths of fetuses, weights of placenta, and fertility index of the dill seed treated group compared to the control and dill leaf treated group, but its anti-implantation and anti-fertility indices increased. We suggested that high level of glucose-6-phosphate could entail increased oxidative stress in uterus and ovary. High lactate dehydrogenase activity in serum may affect cell membranes. Higher secretion of progesterone caused hormonal imbalance and produced estrous cycle irregularity. It was concluded that dill seed but not dill leaf extract could induce infertility in an animal model.  相似文献

The investigations of four Cases involving mold-contaminated buildings and human reaction to exposure, documents tests of extracted urine containing trichothecene mycotoxins confirming exposure and the diagnosis of mycotoxicosis in humans. In each of four Cases, the urine demonstrated antibiotic activity, sulfuric acid charring, and protein release. Urine was extracted using ethyl acetate 40V/60V[EA]. Extracted mycotoxin spotted on (TLC) displayed color and a range of (rf) between 0.2-0.6 using various solvents. Extract was re-suspended using 50% ethanol V/V to inject mycotoxins into weanling female Sprague-Dawley rats. Degeneration and necrosis of the rat's tissue followed. Koch's Postulates conditions were fulfilled by isolation of the causative agent, the trichothecene mycotoxins and the reproduction of disease. Examination of human tissue within the urine extraction group confirms Koch's Postulates and comparative pathology confirms inhalation Mycotoxicosis, with severe necrosis of the central nervous system and severe scarring within the lungs. Extraction of mycotoxins from human patient urine is a very useful confirmatory test to demonstrate exposure and identify mycotoxicosis. Low concentrations (6%) of sodium hypochlorite were ineffective against the activity of trichothecene mycotoxin. Cadmium (Cd) in coal, fly ash, slag, atmospheric deposition, soils and sediments collected from Tianjin, northern China, were measured to provide baseline information and determine possible Cd sources and potential risk. The concentrations of Cd in coal, fly ash and atmospheric deposition were much higher than the soil background values. Fallout from coal-fired thermal power plants, heating boilers and industrial furnaces has increased the Cd concentration in soils and sediments in Tianjin. The concentrations of Cd in soils of suburban areas were significantly higher than in rural areas, suggesting that coal burning in Tianjin may have an important impact on the local physical environment. Cd from coal combustion is readily mobilized in soils. It is soluble and can form aqueous complexes and permeate river sediments. The high proportion of mobile Cd affects the migration of Cd in soils and sediments, which may pose an environmental threat in Tianjin due to the exposure to Cd and Cd compounds via the food chain. This study may provide a window for understanding and tracing sources of Cd in the local environment and the risk associated with Cd bioaccessibility.  相似文献

赤霉酸是目前国内外使用极其广泛的一种植物生长调节剂,但是针对其发育毒性的数据依然较少。本文探讨了赤霉酸暴露对SD大鼠青春期发育的影响。参考国内外环境内分泌干扰物危害的评价方法,将144只初断乳SD大鼠按体重随机分为4组,分别为对照组和1、10、100 mg·kg-1bw剂量组,采用经口灌胃方式对雄鼠连续染毒28 d,雌鼠连续染毒21 d。暴露结束后检测大鼠的体重、食物利用率、雄鼠包皮分离时间/雌鼠阴道开口时间、血清生化指标、脏器系数及组织病理学的变化。研究结果表明,与对照组相比,仅10和100 mg·kg-1bw剂量组雄鼠的肌酐水平显著升高(P<0.01),100 mg·kg-1bw剂量组雌鼠谷丙转氨酶水平有显著升高(P<0.01)。而与对照组相比,所有剂量组均未观察到大鼠的体重、食物利用率、雄鼠包皮分离时间/雌鼠阴道开口时间、脏器系数等指标有显著性差异的改变(P>0.05),组织病理学结果亦显示大鼠重要器官无损害性改变。因此,在本试验给予的剂量范围内赤霉酸染毒不会对SD大鼠青春期发育产生显著影响。  相似文献
