This study examined the adverse effects of N,N,N′,N′-tetramethylethylenediamine (TEMED) on electric eel (Electrophorus electricus) acetylcholinesterase (AChE) and human serum butyrylcholinesterase (BChE). Data show that TEMED inhibited AChE in electric eel as well as human serum BChE. Kinetic studies indicated that the inhibition produced by TEMED in both sources was of mixed type, i.e. Km increased and V max decreased in a concentration-dependent manner. KI (constant of ChE–substrate–TEMED complex into ChE–substrate complex and TEMED) was estimated to be 0.674 mM for electric eel and 0.024 mM for human serum BChE. The γKm (dissociation constant of ChE–substrate–TEMED complex into ChE–TEMED complex and substrate) was 0.083 and 0.2 mM for electric eel AChE and human serum BChE, respectively. The IC50 for electric eel and for human serum ChE was 1.57 and 0.043 mM, respectively. The present results suggest that TEMED produced adverse effects on electric eel and human serum via inhibition of ChE.  相似文献

The Brazilian Amazon is known to be a region with high levels of mercury (Hg) in the environment and studies point to an association between high levels of natural mercury in the mother rock and the vast number of clandestine gold mines. Other studies already report the contamination of fish in this region, as well as high levels of Hg in biological material from environmentally exposed populations. On the other hand, this is one of the least developed regions of the planet and it is necessary to understand the vulnerability factors in these populations that may be intoxicated by this element. The purpose of the present study was to investigate the vulnerability factors in communities from Xingu River—Amazon basin probably exposed to Hg. A cross-selection study in two cities localized in Xingu River was conducted, and the sample contained was 268 individuals. sociodemographic questions, lifestyle, diet habits and health conditions were collated. The majority of the sample was female, between 30 and 59 years old, had less than 3 years of educational level and lived in the local of study more than 240 months. There was regular fish consumption (95.9%), principally carnivorous species (80.5%). The visual problem has a highest prevalence (43.3%) between the health problems and about the symptoms of Hg intoxication, memory loss (42.9%), weakness (35.1%), fatigue (34.3%), mood changes (28.7%) and difficulties in concentration (27.2%) was most reported. The female sex, age over 60, educational level below 3 years of study, did not had flush toilet, smoke and least one chronic non-communicable disease represent higher probability to had symptoms of Hg intoxication. Lack of access to health services, low education level and income evidence the susceptibility of this community to diseases and injuries. The vulnerable groups identified in this study should be a priority in public health and environmental health policies.  相似文献

建立了气相色谱质谱法测定纺织品中N,N-二甲基甲酰胺(DMF)、N,N-二甲基乙酰胺(DMAC)和N-甲基吡咯烷酮(NMP)的方法.纺织品中N,N-二甲基甲酰胺、N,N-二甲基乙酰胺和N-甲基吡咯烷酮经丙酮超声萃取,用0.45μm滤膜过滤后直接进样,将提取物进行GC-MS定性定量分析.结果表明,在0.1—10μg·mL-1线性范围内,DMF、DMAC和NMP线性方程分别是y=75041x+8039,y=71622x+12371,y=96278x+8548,相关系数均大于0.9995;方法检出限DMF和DMAC为5 mg·kg-1,NMP为3 mg·kg-1;平均回收率为86.3%—92.0%;相对标准偏差(n=6)为1.4%—5.0%.该方法简便、快速、灵敏度高、重复性好,可用于测定纺织品中N,N-二甲基甲酰胺、N,N-二甲基乙酰胺和N-甲基吡咯烷酮.  相似文献

汞是一种高毒性且具有持久性的重金属污染物,汞污染的治理与修复在近几十年一直是国内外研究热点.了解微生物对汞赋存形态的转化作用,对汞污染的治理与修复具有重要意义.总结汞的不同赋存形态、毒性及对应的常用分析方法,其中甲基汞(methyl mercury,MeHg)是毒性最强的汞形态之一.环境中汞的化学形态能发生转化,尤其以微生物驱动的汞的甲基化、MeHg的去甲基化和汞的氧化还原最为常见.依据汞转化类型将汞转化相关微生物分为汞甲基化、MeHg去甲基化、汞还原、汞氧化等类群,将对应的汞转化作用机制分为基于hgcAB基因的汞甲基化、基于mer操纵子基因的MeHg去甲基化和Hg2+还原、胞内过氧化氢酶介导的Hg0氧化.微生物汞转化过程不仅受到pH和温度的显著影响,而且还受到汞的赋存形态和游离汞的浓度、微生物种/群结构与功能、矿物种类、中间体和次生产物及其交互作用的影响,基于此,提出正确客观表征汞的微生物转化过程需要综合分析微生物组和矿物组的变化规律及其交互作用的综合效应.针对酸性矿山废水(AMD)极端环境微生物汞转化研究的不足,未来的工作将聚焦结合多组学手段、同步辐射谱学和密度泛函理论(DFT)计算等分析技术研究汞赋存形态的微生物转化过程,分析和阐明汞转化中间体的键合作用方式和转化机制,从而为AMD汞污染的预防、治理和修复提供依据.(图2表2参107)  相似文献

N,N′-bis(trans-cinnamaldehyde)-1,2-diiminoethane and its derivatives were designed and synthesized. Fourier transform infrared spectroscopy (FTIR) and nuclear magnetic resonance (1H-NMR) techniques were applied to characterize chemical structures, and the filter paper disk method was used to test for antifungal effect of the products against Aspergillus niger, Paecilomyces variotii, and Paecilomyces lilacinus. Chemical structure charaterization showed that the products are target compounds. Antimicrobial experiment results indicated that N,N′-bis(trans-cinnamaldehyde)-1,2-diiminoethane displayed reliable antifungal effects against three fungi, and its antifungal effect is close to that of cinnamaldehyde. N,N′-bis(p-methoxy cinnamaldehyde)-1,2-diiminoethane and N,N′-bis(p-chlorocinnamaldehyde)-1,2-diiminoethane also showed antifungal effects at a low concentration. Of the two, N,N′-bis(p-methoxy cinnamaldehyde)-1,2-diiminoethane appeared to be more suitable as it was more effective at higher concentrations against Paecilomyces.  相似文献

Mercury concentrations in hair are typically used as a biomarker to assess exposure to mercury. A total of 108 hair samples were collected from residents (age range 5–73 years) of Wujiazhan town, northeast China, to determine total mercury concentrations. Hair mercury concentrations ranged from 0.16 to 199 mg kg–1 with an average value of 3.41 mg kg−1. The relationships between mercury concentration and gender and between hair mercury concentration and age were not significant. Overall, 16.7% of all samples were above the RfD value published by the United States Environmental Protection agency. The results indicate that there may be some been health risk from mercury exposure to the residents in the study area.  相似文献

The present study reports the isolation and partial purification of a mercury‐binding polypeptide in whole and anucleated cells of Acetabularia calyculus. Sephadex G‐75 Gel filtration showed that cells exposed to Hg exhibit a major peak of protein(s) concomitant with both, a major peak of Hg (revealed by atomic absorption spectroscopy), and the major peak of mercury‐thiolate complex (based on a high 254 nm absorbance) accompanied by high content of SH groups. SDS‐Page electrophoresis showed that this peak contains one major polypeptide of apparent molecular weight 27.000 dalton. These results are in agreement with our previous finding of unusually high bioconcentration factor of Hg in Acetabularia related to the induction of a polypeptide in both, whole and anucleated cells of Acetabularia, in presence of Hg.  相似文献

结合紫外分光光度法研究系列羟基卟啉与汞离子的显色反应,并优化其检测条件.在pH=7.5的条件下,以N,N-二甲基甲酰胺(DMF)作为溶剂,加入0.4 mL表面活性剂Tween-80,系列羟基卟啉与Hg2+发生配位反应,发现其最大吸收峰与卟啉环上(meso)位苯基对位上的羟基数目相关.热力学研究表明,单羟基卟啉化合物对Hg2+检测效果最好,形成1∶1配位化合物,检测限可达0.042μg.L-1,且在0—50 mg.L-1范围内符合比尔定律.  相似文献

论文以血清瓶为间歇反应器,以醋酸钙为基质,研究了厌氧消化过程中甲烷菌所需要的微量金属营养元素Fe、Co、NI(1.0mg/(L.d),0.1mg/(L.d),0.2mg/(L.d)对毒性物质NH4^+-N的拮抗作用,研究结果表明,Fe、Co、Ni对毒性物质NH4^+-N有明显的拮抗作用,而且NH4^+-N浓度越高,Fe、Co、Ni对其毒性的拮抗作用越明显。  相似文献

Mercury (Hg) accumulation in fishes is a serious threat for food security because mercury induces dieseases. Here we studied the total mercury content in four species of Pacific salmon (pink, chum, sockeye and chinook), caught in the Sea of Okhotsk, the Bering Sea, and the Kuril oceanic waters. Results show that the lowest concentration of mercury was found in the pink salmon, and the highest was 5–6 times higher in sockeye. Mercury (Hg) exposure is ubiquitous in modern society via vaccines, fish/crustacea, dental amalgam, food, water, and the atmosphere. This article examines Hg exposure in the context of primary exposure to pregnant women and secondary exposure experienced by their unborn babies. Babies in utero are particularly at risk of higher Hg exposure than adults (on a dose/weight basis through maternal Hg transfer via the placenta), and are more susceptible to adverse effects from mercury and its biologically active compounds. It is, therefore, critical that regulatory advisories around maximum safe Hg exposures account for pregnant women and secondary exposure that children in utero experience. This study focused on standardized embryonic and fetal Hg exposures via primary exposure to the pregnant mother of two common Hg sources (dietary fish and parenteral vaccines). Data demonstrated that Hg exposures, particularly during the first trimester of pregnancy, at well-established dose/weight ratios produced severe damage to humans including death. In light of research suggestive of a mercuric risk factor for childhood conditions such as tic disorders, cerebral palsy, and autism, it is essential that Hg advisories account for secondary prenatal human exposures.  相似文献

The main purpose of this study was to monitor ambient air particles and particulate-bound mercury Hg(p) in total suspended particulate (TSP) concentrations and dry deposition at the Hung Kuang (Traffic), Taichung airport and Westing Park sampling sites during the daytime and nighttime, from 2011 to 2012. In addition, the calculated/measured dry deposition flux ratios of ambient air particles and particulate-bound mercury Hg(p) were also studied with Baklanov & Sorensen and the Williams models. For a particle size of 10 μm, the Baklanov & Sorensen model yielded better predictions of dry deposition of ambient air particulates and particulate-bound mercury Hg(p) at the Hung Kuang (Traffic), Taichung airport and Westing Park sampling site during the daytime and nighttime sampling periods. However, for particulates with sizes 20–23 μm, the results obtained in the study reveal that the Williams model provided better prediction results for ambient air particulates and particulate-bound mercury Hg(p) at all sampling sites in this study.

Two different Calanus finmarchicus (Gunnerus) cohorts originating from 60°N (Bergen) and 69°N (Tromsø) were investigated in equal environmental conditions to study their different physiological responses to the same environment. A two-plus-two-bag mesocosm study was carried out between March and July 1998, in Håkøybotn, Tromsø, in order to determine development and mortality rates of the two parallel cohorts of C. finmarchicus. For practical reasons, the cohort from Bergen was incubated 10 days earlier than the Tromsø cohort. Consequently, they were exposed to elevated food conditions as compared to the Tromsø cohort. A high initial mortality among the Bergen cohort could clearly be ascribed, by genetic discrimination, to "contamination" with C. helgolandicus. After this initial mass mortality, the mortality was constantly 0.03-0.04 dayу. In cohorts starting from naupliar stage I, there was no significant difference in development or growth, the median development time (NI-CIV) differing by only 7 days (~6%). The difference in development time can be explained to a large extent (~4 days) by temperature differences. This is substantiated with model simulations using a physiological model developed for C. finmarchicus. There was a time lag in incubation between the two cohorts, resulting in elevated temperature during incubation of the Tromsø cohort. A fraction of both cohorts differentiated sexually at stage CV, with males differentiating before females. Females from both cohorts produced eggs, but specific egg production differed significantly (P>0.001, t-test). This was supported by elevated RNA:DNA ratio in females from the Bergen cohort. Both cohorts demonstrated quite similar development and physiological growth rates and, consequently, are considered to belong to the same genetic population inhabiting the Norwegian Shelf. The study demonstrates that C. finmarchicus is capable of adaptation to changes in environment and, thereby, demonstrates a significant physiological plasticity.  相似文献
