The present investigation examined the detoxifying potential of methanolic herbal extracts, namely the leaf and bark extract of Eucalyptus tereticornis, bark extract of Saraca asoca, Cassia fistula and Withania somnifera in vitro using primary chicken embryo fibroblast (CEF) cells against damaging effects of urea and mercuric chloride (HgCl) (II). The influence of 20?mM urea and 10?µM?HgCl (II) was determined on cell viability or proliferation of cells after treatment with plant extracts. Higher survival rate of primary CEF cells treated with higher concentrations of plant extracts was observed due to their protective ability against urea and HgCl (II). Cassia fistula bark extract (10?mg?mL?1) was found to be most effective against 20?mM urea as it protects 90% of CEF cells whereas W. somnifera protects 86% of the cells within 24?h. After treating cells with10?µM HgCl, W. somnifera and E. tereticornis leaf extracts were found to be more effective among all other extracts as they protect approximately 86% and 70% of CEF cells, respectively, within 24?h. These results indicate that C. fistula and W. somnifera has the highest potential amongst all the five plant extracts for protecting CEF cells against damaging effects of urea and HgCl (II), respectively.  相似文献

Cell integrity is affected by oxidative stress when the production of active oxidants overwhelms antioxidant defense mechanisms. Latex, a natural polymer obtained from Hevea brasiliensis, is used in medical industry for manufacturing surgical gloves, urinary catheters, and dental dams. The aim of this study was to evaluate the effects of latex material on oxidative stress by in vivo and in vitro methods. In addition, the material was screened for its ability to induce any chromosomal aberrations (CAs) by in vitro method. In vivo studies were carried out with implanted latex material onto subcutaneous tissue of various batches of experimental Wistar rats. At the end of experimental period, animals were anesthetized, blood was collected for serum analysis, and sacrificed. Liver was excised for the determination of antioxidant enzymes and lipid peroxidation (LPO). Subcutaneous tissues were obtained for the extraction of genomic DNA from implanted animals and checked for the presence of 8-hydroxy-2-deoxyguanosine (8-OHdG), considered an indicator of DNA damage. Simultaneously, in vitro studies were carried out using fresh liver and subcutaneous tissue obtained from Swiss albino mice treated with physiological saline extract of latex material. For the estimation of both in vitro and in vivo oxidative stress, 10% liver homogenate was assessed for stress indicators like reduced glutathione, glutathione reductase, glutathione peroxidase, LPO and protein content. The results of both in vivo and in vitro studies indicated that the chemical leachents from the latex material did not significantly affect LPO and the levels of antioxidant enzymes. There was also no significant increase in 8-OHdG content due to the presence of implanted latex material. Finally, the results of in vitro CA test and G banding indicated that extracts of test material did not induce any chromosomal abnormalities.  相似文献

The effects of an arbuscular mycorrhizal fungi (AMF) association on the growth, survival capabilities, nutrients and lead (Pb) uptake of Miscanthus sacchariflorus under different Pb concentrations were studied in the form of pot cultures. The treatments comprised inoculation or non-inoculation of the AMF, Gigaspora margarita, and the addition of three Pb concentrations to the soil (0, 100 and 1000?mg?kg?1). The addition of Pb significantly decreased mycorrhizal colonisation. The inoculation of AMF with Pb increased chlorophyll content, Fv/Fm, total dry mass, indole-3-acetic acid (IAA), total nitrogen, and total phosphorus, whereas H2O2 level, indole-3-acetic acid oxidase (IAAO) activity, and peroxidase (POD) activity were low compared to those in the non-inoculated treatments. Moreover, the application of AMF together with Pb doses induces concentrations of Pb in the plant, where the higher dose of Pb (1000?mg?kg?1) induces a lower content of Pb in the aerial part of the plant but a higher content in the root. G. margarita enhanced the tolerance of M. sacchariflorus against Pb toxicity, and facilitated the accumulation of Pb in the plant roots, whereas translocation to the shoots was inhibited at the highest dose Pb (1000?mg?kg?1). However, in contaminated soil, the Pb removal capability of M. sacchariflorus with AMF was remarkable.  相似文献

This study discusses the similarities and differences between the antifungal activity of Chelidonium majus L. extracts and chelerythrine/sanguinarine, the main alkaloids in these extracts, against the plant pathogenic fungus, Botrytis cinerea. The obtained data show that the Ch. majus L. extracts inhibit the growth of B. cinerea very effectively. The extracts activity is significantly greater than that of chelerythrine/sanguinarine standard solutions alone and it does not always reflect the total content of alkaloids in the extracts. It has been proved that B. cinerea exhibits very diverse resistance to chelerythrine and sanguinarine. The IC50 values for chelerythrine and sanguinarine equal 5.41?mg/mL and 1.76?mg/mL, respectively. In the mixture the antifungal activity of chelerythrine and sanguinarine is antagonistic in the concentration dependent way. Based on the presented data it is not clear, however, which alkaloids present in the Ch, majus extracts at significantly lower concentration levels enhance the activity of chelerythrine and sanguinarine so that the extracts activity is considerably greater than that of both main alkaloids mixtures.  相似文献

A new series of 5- or 7-substituted 3-{4-(5-mercapto-1,3,4-oxadiazol-2-yl)phenylimino}-indolin-2-one derivatives were screened for in vitro antioxidant and antimicrobial activities. The results of this study revealed that all synthesized compounds were significantly scavenged DPPH free radicals in a concentration-dependant manner. The IC50 values of all test compounds were found to be between 18.34 and 50.54?µM. All compounds significantly inhibited growth of bacteria and most of the minimum inhibitory concentration values were between 20 and 100?mg?mL?1 indicating potent to moderate antibacterial activity. In conclusion, 3-{4-(5-mercapto-1,3,4-oxadiazole-2-yl)phenylimino)-5 or 7-substituted indolin-2-ones derivatives exhibited moderate antioxidant and antimicrobial activities.  相似文献

The aim of the study was to investigate the antiproliferative properties of extracts of five plants on breast cancer cells (MDA-MB-231) and their effects on antioxidant activities. The total phenol contents of the extracts were determined to establish a correlation to their antiproliferative effects which were evaluated by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide viability assay. Enzymes involved in oxidative stress were also determined. Total polyphenol contents of the five extracts varied from 170 to 440 mg L?1 of gallic acid per mg of extract with the highest value for the extract of Sida cordifolia. Cell growth inhibition was observed within 24 h, with inhibitory concentration values ranging from 7.3 to 25 mg L?1 depending on plant extract. At concentrations between 100 and 200 mg L?1, all the extracts exhibited significant reduction of cell proliferation in time-dependent and linear manner. The activities of superoxide dismutase and catalase of cells treated for 24 h with the extract of S. cordifolia were 3.5 U per mg protein and 8 mole H2O2 consumed per min per mg protein higher than those of cells treated with an extract of Viscum album.  相似文献

This study is the second of two surveys designed to systematically screen extracts from marine plants for antimicrobial effects against ecologically relevant marine microorganisms, and to compare results on a geographical basis. In the preceding survey, extracts from tropical Atlantic marine algae and seagrasses were screened in growth inhibition assays against the pathogenic fungus Lindra thalassiae, the saprophytic fungus Dendryphiella salina, the saprophytic stramenopiles, Halophytophthora spinosa and Schizochytrium aggregatum, and the pathogenic bacterium Pseudoaltermonas bacteriolytica. In this study, the same assay microorganisms were used to examine the antimicrobial effects of lipophilic and hydrophilic extracts from 54 species of marine algae and two species of seagrasses collected from Indo-Pacific reef habitats. Overall, 95% of all species surveyed in this study yielded extracts that were active against one or more, and 77% yielded extracts that were active against two or more assay microorganisms. Broad-spectrum activity against three or four assay microbes was observed in the extracts from 50 to 21% of all species, respectively. Extracts from the green alga Bryopsis pennata and the red alga Portieria hornemannii inhibited the growth of all assay microorganisms. Given that antimicrobial activity was prevalent among extracts of Indo-Pacific marine plants, it is interesting to note that the inhibitory effects of each extract varied considerably between the assay microorganisms. Overall, H. spinosa and D. salina were the most susceptible while L. thalassiae, S. aggregatum, and P. bacteriolytica were the most resistant to the extracts tested. These results provide good evidence that antimicrobial chemical defenses are widespread among Indo-Pacific marine plants. Further, the activity profiles of plant extracts suggest that antimicrobial secondary metabolites can have pathogen-selective or broad-spectrum effects. To confirm these results, chemical studies will be needed to isolate and characterize the compounds responsible for the observed antimicrobial activities.  相似文献

Medicinal herbs are steadily increasing in complementary use for chronic and alterative therapies. The health risks associated with herbal supplements have therefore been identified as a top research priority. Tecoma stans (Tronadora), a Bignoniaceae plant, is a herbal drug traditionally consumed as tea in South America for the control of diabetes. It contains the alkaloids of tecomine and tecostanine which were shown as functional compounds responsible for hypoglycemic activity. However, the side effects of aqueous extracts of this herbal tea have not been reported. In this study, studies sought to evaluate the cytotoxicity of water extracts from T. stans in human hepatoblastoma (HepG2). Toxic effects of T. stans were concentration- and time-dependent in the presence and absence of fetal bovine serum (FBS). Cells were incubated for up to 72-h with varying concentrations of herbal extracts (60–100%), cytotoxicity was determined spectrophotometrically by MTT and reported in terms of % cell viability. For IC50 assay (24 h exposure), cytotoxicity was found at concentration of 60–100%. Evaluation of the effects of T. stans and Brickellia cavanillesi (Asteraceae) in combination revealed that extracts of both herbal extracts' nontoxic levels of T. stans significantly reduced cell viability in a time-dependent manner. Further studies are needed regarding the identification, toxicity, and molecular mechanism of action of active compounds, as well as their cytotoxicity to other cell lines following exposure to both single and combined herbal extracts of these plants with anti-diabetic properties.  相似文献

The aim of this study was to compare the cytotoxicity and the (pro-)inflammatory responses of two-stroke (direct injection and carburetor technology) and four-stroke scooter and diesel car exhaust emissions on lung cells in vitro. This was analyzed by exposing a 3D in vitro model of the epithelial airway (consisting of human bronchial epithelial cells (cell line 16HBE14o?) combined with human whole blood monocyte-derived macrophages and dendritic cells) to physically characterized exhaust emissions. Biological endpoints of cytotoxicity (lactate dehydrogenase release), as well as pro-inflammatory cytokine (tumor necrosis factor (TNF)-α) and inflammatory chemokine (interleukin(IL)-8) stimulation were examined. Two-stroke direct injection scooter exhaust contained the highest particle number concentration and nitrogen oxides (NO x ) concentrations and the emissions from the two-stroke carburetor scooter contained the highest hydrocarbon and lowest NO x concentrations. The four-stroke scooter emitted the highest carbon monoxide concentration whereas the cars emitted the lowest. The combination of various technical optimizations for the two-stroke direct injection scooter (particle filter, oxidative catalyst, better oil and fuel) reduced the total emissions strongly and the TNF-α concentration significantly (p?相似文献

Two wetland plant species, Phragmites australis and Oryza sativa, were grown in a glasshouse under hydroponics conditions. Enzyme extracts from different parts of the plants were used to determine the transformation rate of o,p-DDT, p,p-DDT and PCBs. The organic pollutants were directly spiked into the enzyme extracts, and samples were collected every 30 min and analyzed with a GC-ECD. Root extracts of P. australis readily degraded and transformed DDT and some PCB congeners with a low degree of chlorination. In contrast, crude extracts of O. sativa showed no appreciable degradation or transformation of DDT or PCBs. Inhibition studies indicated that the degradation and transformation of both DDT and PCBs by P. australis enzymes were partly mediated by peroxidase and the plant P-450 system. PCBs with a high degree of chlorination were highly resistant to transformation or degradation by plant enzymes. Both wetland plant species accumulated substantial quantities of the persistent organic chemicals but had different degradation capacities. The enzyme systems in P. australis were much more effective that those in rice in the degradation and transformation of the organic pollutants.  相似文献

Abstract

Fusarubin analogues of Fusarium oxysporum f. sp. ciceris were investigated for antifungal activity in vitro against five soil borne phytopathogenic fungi. 3-O-Methyl-8-O-methyl-fusarubin was inhibitory towards S. sclerotiorum (EC50 0.33?mmol L?1) and Sclerotium rolfsii (EC50 0.38?mmol L?1). A structure–antifungal activity relationship of fusarubin analogues was established from their activity performance. Possible mechanism of action of these compounds was studied using molecular docking and simulations against three target enzymes which revealed receptor ligand binding affinity. Docking of 3-O-methyl-8-O-methyl-fusarubin into the succinate dehydrogenase site revealed formation of salt bridge, hydrogen bond, π–anion, π–alkyl, and Van der Waals interactions.  相似文献

Biological control agents have become a useful alternative for the reduction of the use of chemical insecticides. LABIOFAM (Cuba) is developing a new formulation of a biolarvicide that possesses as active biological agent Bacillus thuringiensis var israelensis serotype H14. In order to evaluate the genotoxicity of this new formulation, an in vivo battery test was used: micronucleus (MN), chromosome aberrations (CAs), and sperm morphology (SM) assays. A dose of 6.45?×?108 spores was administered per animal via oral administration. Bone marrow cells were collected 24?h after a two day treatment for the MN assay, and 24?h after a unique treatment for the CA assay, using cyclophosphamide as the positive control. Sperm cells were collected at 5 weeks from the first of five administrations for the SM test, using acrylamide as positive control. Bacillus thuringiensis var israelensis serotype H14 failed to show either a significative increase of micronucleated polychromatic erythrocytes, chromosomal aberrations, or sperm abnormalities. Acute oral administration of a high dose of Bacillus thuringiensis var israelensis serotype H14 did not produce mutagenic effects in bone marrow or sperm cells.  相似文献

Biology of euphausiids in the subarctic waters north of Iceland   总被引:2,自引:0,他引:2  
The seasonal abundance, maturity, spawning, and population dynamics of Thysanoessa inermis (Krøyer, 1846), T. longicaudata (Krøyer, 1846), and Meganyctiphanes norvegica (M. Sars, 1857) were studied in the subarctic waters north of Iceland from February 1993 to February 1994. The material was sampled at approximately monthly intervals along a transect of eight stations extending from 66°16′ to 68°00′N at 18°50′W. Information on temperature and chlorophyll a concentrations is also presented. Spring warming of the water began in March to April and maximum temperatures were recorded in August (3.8?°C). The spring bloom of the phytoplankton started in late March and highest chlorophyll a concentrations were measured during middle to late April (7.0?mg chlorophyll a m?3). T. inermis was the dominant species in the samples, constituting 77% of juvenile, male and female euphausiids present. The greatest abundance of juvenile, male and female T. inermis and M. norvegica was observed during autumn and winter, with lower abundance in spring and summer. T. longicaudata showed only limited changes in seasonal abundance. Male T. inermis had spermatophores in their ejaculatory ducts from February to May, while mature females had spermatophores attached during April and May. T. longicaudata males bore spermatophores from February to July, whereas females only bore spermatophores in April and May. M. norvegica males had spermatophores from February to April, while the single female with spermatophores was caught in February. Euphausiid eggs were first recorded during the latter part of April; the highest numbers of eggs were observed in the samples taken in late May. Maximum numbers of nauplii of both Thysanoessa spp. and M. norvegica were recorded in late May. The main spawning of the euphausiids coincided with the phytoplankton spring bloom. Most male T. inermis took part in breeding at 1 yr of age while most females appeared not to mature until 2 yr of age. T. inermis has a life span of just over 2 yr, T. longicaudata appears to live just over 1 yr. Limited data did not allow the life span of M. norvegica to be determined.  相似文献

Cultures of the copepod Acartia tonsa are used both in aquaculture and ecotoxicology studies. However, the cultivation of these crustaceans at high densities results in the proliferation of microorganisms that can affect the organisms of interest, leading to illness or death. Antimicrobials inhibit microbial growth and may favour the cultivated species, aiding the development of ecological studies. This study investigated the potential of antimicrobials (antibiotic + antifungal) to inhibit bacteria and fungi when applied to marine zooplankton cultures, using the copepod A. tonsa as a bioindicator of acute toxicity. Treatment with 0.025?g?L?1 of penicillin G potassium + 0.08?g?L?1 of streptomycin sulphate + 0.04?g?L?1 of neomycin sulphate + 0.005?g?L?1 of nystatin resulted in 95% bacterial inhibition (after 12?h of exposure); however, after this time, the inhibitory effect was lost. The antimicrobial combination tested in this study prevented colonisation by fungi until 168?h after exposure, without causing acute toxicity to A. tonsa. Thus, it has potential for use in marine cultures of less sensitive organisms.  相似文献

Particulate organic matter (PM) present in the atmosphere is a complex mixture of chemicals like polycyclic aromatic hydrocarbons (PAH) that may exert adverse health effects including respiratory and cardiovascular disturbances and cancer. In this study, airborne samples from environmental or industrial areas exhibiting different physicochemical composition were compared for their capacities to induce DNA damage in human hepatocytes HepG2. DNA strand breaks and DNA adducts formed by benzo[a]pyrene-7,8-dihydrodiol-9,10-epoxide (BPDE), the most reactive metabolite of the carcinogenic benzo[a]pyrene (B[a]P), were measured with the comet assay and by high-performance liquid chromatography (HPLC)/mass spectrometry, respectively. Cells were exposed to organic matter extracted from PM. Experiments were performed either at a constant concentration of B[a]P or at concentrations corresponding to fixed air volumes. Results show that industrial extracts tend to produce more benzo[a]pyrene diol epoxide-N 2-2′-deoxyguanosine (BPDE-N 2-dGuo) DNA adducts than strand breaks, whereas the opposite was observed with environmental extracts. The chemical composition of the extracts significantly impacts the nature and levels of DNA damage. The amount of B[a]P and interaction with other contaminants in the extracts need to be considered to explain the formation of DNA damage. These results emphasize the use of in vitro tests as promising and complementary tools to widely used toxic equivalent factor (TEF) approach in order to assess health hazards related to chemical exposure of the general population.  相似文献

Aqueous and flavonoid-enriched extract as well as essential oil (EO) obtained from leaves of Pistacia lentiscus were assessed for antibacterial and antimutagenic activities. Antibacterial activity of different extracts and EO were evaluated against six bacterial strains. A marked inhibitory effect was observed against Salmonella typhimurium, whereas lower activity was observed against Staphylococcus aureus, Pseudomonas aeruginosa, and Salmonella enteritidis. EO showed significant inhibitory effects against Salmonella typhimurium, Salmonella enteritidis and Staphylococcus aureus. The antimutagenic activity of the different extracts against Aflatoxin B1 (AFB1) and sodium azide was demonstrated with the Salmonella typhymurium assay. The number of revertants per plate decreased significantly when the plant extracts were added to the assay system using Salmonella typhimurium TA100, TA98 and TA1535.  相似文献

Dry body weights, lipid levels and lipid compositions were measured in I-and II-group Thysanoessa raschi (M. Sars) and T. inermis (Krøyer) collected in April-May, 1980 in Balsfjorden, north Norway. Dry body weights were mininal in late April but had doubled by mid-May for I-group T. raschi and for I- and II-group T. inermis. II-group T. raschi had increased its dry weight by less than 50% by mid-May. Lipid accounted for approximately 10% of the dry body weight throughout the period, with free fatty acids and phospholipids being the dominant classes. An exception occurred for II-group T. inermis in mid-May, when wax esters were present in substantial amounts. Wax esters were present only in small amounts at other times and triacylglycerols were negligible. The free fatty acids in I-group T. raschi and I-group T. inermis in late April were deficient in polyunsaturates; by mid-May the free fatty acids were rich in polyunsaturates. II-group T. raschi and II-group T. inermis had free fatty acids rich in polyunsaturates throughout the period of study. The wax esters present in II-group T. inermis in mid-May consisted mainly of 16:0 and 14:0 alcohols esterified to 18:1 fatty acid. The traces of wax esters present in T. raschi did not contain significant amounts of phytol. Results are discossed with respect to the metabolic activities of the two euphausiids and their trophic positions in terms of different dietary inputs.  相似文献

Summary. Recent economic and social changes in north Mediterranean regions have led to an important rural depopulation. Consequently, meadows developed on abandoned agricultural lands (characterized by high species richness) undergo reforestation. These former fields are mainly colonized by Pinus halepensis Miller, which is known to synthetize a wide range of secondary metabolites, among these, some could influence plant succession through allelopathy. The allelopathic potential of P. halepensis, was tested against two target species (Lactuca sativa L. and Linum strictum L.) with aqueous extracts obtained from different organs (root and needle) taking into account the individual age (±10, ±30 and > 60 years old). Root and needle extracts affected differently germination and growth of the two target species, the responses varying with concentration of extracts, age and organs tested. The strongest inhibitory effect was observed on the germination and growth of L. strictum, exposed to needle extracts of young P. halepensis (±10 years old), and root extracts of older P. halepensis (> 30 years old). These extracts contained several phenolic acids (e.g. 4-hydroxybenzoic acid and p-coumaric acid), which are known as allelochemicals and their concentrations vary with age and organ tested. Hence, P. halepensis could influence secondary succession through the release of potential allelochemicals in the environment by leaf leachates or root exudates.  相似文献

Summary Thirty-one species in twenty genera of the plant family Meliaceae were assayed for the production of growth-inhibiting phytochemicals, using the generalist herbivorePeridroma saucia. Most species were inhibitory when methanolic extracts were incorporated into artificial diets at concentrations at or below those occurring naturally. In general members of the subfamily Melioideae were more inhibitory than members of the Swietenioideae. Extracts of deciduous species with short leaf lifetimes were significantly more inhibitory than those of evergreen species with longer leaf lifetimes. In a smaller sample of species, evergreen species showed a trend towards having tougher leaves than deciduous species. These results support the resource availability hypothesis of Coleyet al. (1985), and suggest that life history attributes may be of some value in selecting plants for phytochemical prospecting.  相似文献
