Solid-phase microextraction (SPME) is an effective technique for studying frog volatile secretions. Its primary advantage is in its application to sampling live animals. the ability to sample an organism over an extended period allows changes in an individual's chemical signature to be determined. the presence of eucalyptol in the skin secretion of Ewing's tree frog, Litoria ewingi, was used to assess the effectiveness of SPME in sampling frog volatiles. Rapid sample times coupled with the polydimethylsiloxane/divinylbenzene (PDMS/DVB) fibre provided the best signal/noise ratio for the majority of frog volatiles analysed, and importantly resulted in the least amount of stress to the animals involved.  相似文献

We used a prototype GPS logger to track the movements of breeding African Penguins (Spheniscus demersus). The loggers also recorded temperature and water depth, which allowed us to reconstruct foraging tracks in three dimensions, although GPS signals are interrupted when the birds dive. Here we report the loggers performance in the field and assess the effects of GPS error, resolution and sampling rate on estimates of foraging track length and speed. There is a trade-off between sampling rate and battery lifespan. We tested loggers at sampling intervals of 1 s, 10 s, 1 min, 2 min and 10 min. Sampling less frequently increases the chance of tracking an entire foraging trip, but it slows uplink times, slightly decreases the accuracy of positional fixes, and significantly reduces the ability to measure fine-scale aspects of foraging behaviour. Compared with radio or satellite tracking, GPS loggers offer unprecedented detail about animal movements. The results of our analysis suggest that techniques that sample relatively infrequently, such as satellite tracking, underestimate actual track lengths by up to 50%. However, caution is needed when interpreting fine-scale sampling for relatively slow-moving organisms. Re-sampling 1-s tracks suggests that c. 35% of apparent movements at this scale are due to measurement error and, more importantly, the limited spatial resolution of GPS (1.85×1.54 m at the study area). We recommend that researchers use a 1-s sampling rate for fine-scale studies, but resample at less frequent intervals to remove spurious noise for slow-moving animals. At current levels of resolution, animals should move at least 4 m per sampling interval. We provide empirical correction factors to compare inferred track length sampled at different rates, but caution that these are idiosyncratic and strongly dependent on the animals behaviour. Overall, GPS loggers offer a significant advance for studies of fine-scale animal movement patterns.Communicated by O. Kinne, Oldendorf/Luhe  相似文献

The availability of high-quality water plays a pivotal role for the protection of the ecosystem and the quality of human life. An important step in assessing ground and surface water quality is sampling. The time-integrated accumulation of environmental contaminants by passive sampling is an attractive alternative to conventional snap-shot sampling. Thein situ accumulation during passive sampling allows the detection of even lowconcentrated contaminants and reduces cost and time for continuous monitoring. Passive sampling provides information on the whole sampling period and avoids the transport and storage of large sample volumes. Matrix effects are reduced due to the selective enrichment. Various passive samplers have been developed for sampling in aqueous media. Early developments used water filled dialysis tubes for the sampling of trace elements. Later on, solvent filled devices and triolein-filled semipermeable membrane devices (SPMDs) were deployed. More recent developments use a solid rather than a liquid sorbent as the receiving phase. Examples of this are the solid phase microextraction (SPME) and the membrane enclosed sorptive coating (MESCO). In addition to comprising miniature devices, SPMEs as well as MESCO are thermodesorbable and, as such, no longer require solvent extraction. Conventionally the sampled contaminants are removed from the passive sampling devices by solvent extraction or thermodesorption in order to be analyzed chemically. For an in-depth analysis of sampled analytes, however, it would be advantageous to modify passive sampling such that sampled contaminants can also be analyzed biologically. Thus, it is our goal to construct a passive sampling device that serves both as a sampling device as well as an exposure chamber for toxicity testing. The principle underlying this technology is the bioavailability of sorbed contaminants, thereby eliminating the need for solvent extraction.  相似文献

Summary. We adapted a four-arm olfactometer (55 × 55 × 5.5 cm) for bioassays of large insects and its usefulness was evaluated by testing the responses of three beetles, the palm weevil Rhynchophorus palmarum (L.) (Coleoptera: Curculionidea) and two Dynast scarabs Strategus aloeus (L.) and Oryctes rhinoceros (L.) (Coleoptera: Scarabaeidae) to plant odours and aggregation pheromones. This olfactometer was coupled to a volatile delivery system dispensing highly volatile semiochemicals at constant concentrations. We materialized airflows in the olfactometer by discolouring sensitive papers with acid vapours under various conditions (air straighten by stainless steel grids in inputs and output, 500 ml/min per arm) to visualize air turbulence and to test air-tightness. The volatile delivery system can be used to mix and dispense a broad range of concentrations of semiochemicals in air from diluted aqueous solutions. It was calibrated by measuring the release of the pheromone using solid phase-microextraction (SPME). Dose-response curves to synthetic pheromone were obtained for O. rhinoceros of both sexes. Coupling the volatile delivery system to the fourarm olfactometer provided a system with which the responses of R. palmarum and S. aloeus to their synthetic aggregation pheromones and to natural plant volatiles could be readily observed and studied.  相似文献

Summary. It is well known that feeding by Pieris brassicae caterpillars on cabbage leaves triggers the release of volatiles that attract natural antagonists such as the parasitoid Cotesia glomerata. The temporal dynamics in the emissions of parasitoid attracting volatiles has never been elucidated in this system. In a time course experiment, caterpillar infested leaves attracted the parasitoid within one hour after infestation. At such an early stage of infestation, as much as fifty percent of the parasitoids flew towards the infested plant in a wind tunnel bioassay, while only five percent flew towards the non-infested control plant. Three hours after infestation and later, the response to the volatiles from the infested plant reached its maximum and then continued at a constantly high level for the remaining 14 hours of the experiment. Chemical analyses of volatiles collected from infested leaves at short time intervals during the first 24 hours identified a total of ten compounds, comprising green leaf volatiles, terpenoids, and a nitrile. Significant increase of emission within the first 5 hours following initial herbivory was detected for (Z)-3-hexen-1-ol, (Z)-3-hexen-1-yl acetate, cineole and benzylcyanide. Subsequently, a coupled bioassay-chemical analysis procedure was developed allowing for testing and analyzing the same sample for future identification of the bioactive compounds. This was achieved by using stir bar sorptive extraction for the analysis of solvent extracts of caterpillar-damaged leaves.  相似文献

Summary. We describe the use of pieces of silicone tubing to analyse the mandibular gland components of queen and worker honeybees and show that these compounds can be efficiently trapped on bis(trimethylsilyl)trifluoroacetamide (BSTFA) treated pieces of tubing. The use of this technique rather than that of solid phase microextraction (SPME) techniques with commercially available fibres that have been shown to be efficient at sampling secretions from the cuticle of insects, is necessitated by a requirement for collection of large sample numbers in a short space of time or for sampling in the field. The technique may be generalised for use with semiochemicals of low volatility in other insect communciation systems.  相似文献

Summary. Dufour glands of Paratrechina longicornis were analyzed chemically using GC-MS techniques. The glands contained twelve alkanes between C9 and C20 chain length with undecane and tridecane as the main components, six alkenes (1- and 4-undecene, 1-, 4-, and 6- tridecene, and heptadecadiene), two alkyl formates (C11 and C13), and saturated and polyunsaturated acids. Many of the alkanes and alkenes were behaviorally active causing a short lasting attraction of ants with different intensity. Detection of the major gland compound undecane in recruitment trails by in vivo SPME sampling provided evidence for its use as a recruitment pheromone. Both poison gland (formic acid) and Dufour gland (undecane) contents were detected on SPME fibers which had been attacked by the ants.  相似文献

Abstract: Amphibians are declining worldwide, but these declines have been particularly dramatic in tropical mountains, where high endemism and vulnerability to an introduced fungal pathogen, Batrachochytrium dendrobatidis (Bd), is associated with amphibian extinctions. We surveyed frogs in the Peruvian Andes in montane forests along a steep elevational gradient (1200–3700 m). We used visual encounter surveys to sample stream‐dwelling and arboreal species and leaf‐litter plots to sample terrestrial‐breeding species. We compared species richness and abundance among the wet seasons of 1999, 2008, and 2009. Despite similar sampling effort among years, the number of species (46 in 1999) declined by 47% between 1999 and 2008 and by 38% between 1999 and 2009. When we combined the number of species we found in 2008 and 2009, the decline from 1999 was 36%. Declines of stream‐dwelling and arboreal species (a reduction in species richness of 55%) were much greater than declines of terrestrial‐breeding species (reduction of 20% in 2008 and 24% in 2009). Similarly, abundances of stream‐dwelling and arboreal frogs were lower in the combined 2008–2009 period than in 1999, whereas densities of frogs in leaf‐litter plots did not differ among survey years. These declines may be associated with the infection of frogs with Bd. B. dendrobatidis prevalence correlated significantly with the proportion of species that were absent from the 2008 and 2009 surveys along the elevational gradient. Our results suggest Bd may have arrived at the site between 1999 and 2007, which is consistent with the hypothesis that this pathogen is spreading in epidemic waves along the Andean cordilleras. Our results also indicate a rapid decline of frog species richness and abundance in our study area, a national park that contains many endemic amphibian species and is high in amphibian species richness.  相似文献

Summary Gregarious nymphs of the desert locust,Schistocerca gregaria (Forsk.) (Orthoptera: Acrididae) were more attracted to volatiles from mechanically damaged food plants used for rearing than to either the undamaged or damaged food plants not used as diet in Y-tube olfactometer assays. Comparative analysis of the volatile emissions from plants used for rearing and food plants not used for rearing,e.g. Sorghum bicolor, Pennisetum clandestinum, Schouwia thebaica, wheat (Triticum sp., var. Nyangumi),Zygophyllum simplex, Heliotropium undulatum andTribulus terrestris was carried out by GC, GC-EAD and GC-MS. Significant quantitative and qualitative differences were found in the volatile emissions and olfactory responses of nymphs in GC-EAD assays. Up to 33 compounds were identified in volatiles of the plants of which 9 evoked EAGs. EAG-active components included common green leaf compounds (E)-2-pentenal, (E)-2-hexenal, 4-methyl-3-pentenal, (E)-3-hexenyl acetate, (Z)-3-hexenyl acetate, (Z)-2-hexenyl acetate, (Z)-3-hexen-1-ol and (Z)-2-hexen-1-ol. (Z)-3-Hexenyl butyrate and (Z)-3-hexenyl isovalerate were detected in stimulatory amounts only in the volatiles ofS. thebaica. (E, Z)-2,6-Nonadienal was detected as a component in the volatiles ofT. terrestris and was highly stimulatory. In EAG assays with seven common green leaf volatiles, (Z)-3-hexenyl acetate was most stimulatory while hexanal was the least. No significant differences were recorded between antennal responses of males and females to the tested compounds. These results are discussed with regard to current hypotheses on host plant recognition through detection of their airborne volatiles and the learning behaviour by nymphs ofS. gregaria.  相似文献

Adaptive cluster sampling (ACS) is an adaptive sampling scheme which operates under the rule that when the observed value of an initially selected sampling unit satisfies some condition of interest, C, other additional units in some pre-defined accompanying neighborhood are also added to the sample. In turn, if any of these additional units satisfy C, then their corresponding unit neighborhoods are added to the sample as well, and so on. This process stops when no additional units satisfying C are encountered. This paper will provide a review of the major developments and issues in ACS since its introduction by Thompson (1990) [Journal of the American Statistical Association, 85, 1050–1059].  相似文献

The paper is about the accurate (i.e. unbiased and precise) and efficient estimation of structural indices in forest stands. We present SIAFOR, a computer programme for the calculation of four nearest-neighbour indices, which describe the spatial arrangement of tree positions, the distribution pattern of species, and the size differentiation between trees. The study uses SIAFOR as a sampling simulator in eight completely stem-mapped forest stands of varying area and structural complexity. We statistically evaluate two sample types (distance and plot sampling), comparing sampling error, bias and minimum sample size for index estimation. We introduce the concepts of measurement expansion factor (MEF) and design expansion factor (DEF) for the technical evaluation of sample type efficiency (optimal sample type). Results indicate that sampling error can reach high levels and that minimum sample sizes for index estimation often amply exceed the limit of 20% of tree density or 20 trees per species per hectare, that we set as the highest feasible sample size in normal situations. We found clear feasibility limits (in terms of minimal tree densities and reachable accuracy levels) for the estimation of all investigated indices. Generally, equal or higher sample sizes are needed for plot sampling than for distance sampling to reach equal accuracy levels. Nevertheless, plot sampling resulted more efficient for the estimation of tree size differentiation at low to medium accuracy levels. For all other investigated indices distance sampling resulted more efficient than plot sampling. Minimum sample size increases with accuracy and is negatively correlated with tree density. At a given accuracy level minimum sample size is highest for the estimation of relative mingling and lowest for tree size differentiation; furthermore it is generally lower in large stands than in small ones. Because of the consistency of our conclusions in all of the investigated stands, we think they apply in most stands of similar area (between 1 and 10 ha) and species diversity (not more than four species).  相似文献

Some territorial animals exhibit a form of social recognition, commonly termed the "dear enemy effect", in which territory residents display lower levels of aggression toward familiar neighbors compared to unfamiliar individuals who are non-territorial "floaters". Despite the widespread occurrence of territorial social systems and use of acoustic signals for communication in anuran amphibians, only two previous studies have demonstrated vocally mediated dear enemy behavior in a territorial frog. In this study, I conducted neighbor-stranger discrimination playback experiments in a third species of territorial frog, the strawberry dart-poison frog, Dendrobates pumilio (Anura, Dendrobatidae). In the first experiment (n=24), I broadcast the calls of a subject's nearest neighbor and the calls of an unfamiliar individual from the approximate midpoint between the subject's and the neighbor's territories. Although males responded to the stimuli, they did not exhibit differential responses to the calls of neighbors and strangers. In a second experiment (n=22), I broadcast the calls of a neighbor and a stranger to subjects through a speaker located in the approximate center of the neighbor's territory. Males also responded to the playback, although less intensely than in the first experiment, but no discrimination between the calls of neighbors and strangers was found. Thus, territorial males of the strawberry dart-poison frog appear not to discriminate behaviorally between the advertisement calls of neighbors and strangers. Several proximate and ultimate-level hypotheses for this lack of vocally mediated neighbor-stranger discrimination are discussed.Communicated by T. Czeschlik  相似文献

Large-scale patchiness in the distribution of the benthic fauna was investigated in Loch Etive (Scottish west coast) by two series of van Veen grab hauls. Each series was taken along a traverse across the width of the loch, with sampling points about 100 m apart. One sample series was taken on sandy mud and the other, at a greater depth, on soft mud. Two approaches in data processing were applied: (1) The variance: mean ratio and the Morisita I tests for significant aggregation were applied to the species abundances in the sample series; (2) 3 measures of sample homogeneity, each involving the calculation of an index of faunal similarity, were applied to the separate samples. The results, however, could not show significant differences between the two series of samples. These findings were compared to results obtained from (a) a previous study, where differences in patchiness between areas were shown for a smaller scale of sampling design in Loch Etive and neighbouring areas, and to which the I method is also applied here; and (b) the results of applying the variance: mean ratio and I tests to data published by Holme (1953). The present results indicate greater aggregation at the present scale of sampling than for the previous, smaller scale of sampling, or for the comparable scale of Holme's sampling. The degree of concordance shown by the values of I for the circular-design sampling to the values of the 3 measures of patchiness applied previously, and to the means of the species abundances, was measured by Spearman's rank correlation coefficient. The results clearly demonstrated the I values, unlike the others measured, to be almost completely independent of the mean. It was concluded that, for comparing the pattern from benthic samples using standard-size bottom samplers, where the mean may vary widely between each set of samples, the I method is probably most useful.  相似文献

Abstract: The cane toad (Bufo marinus), a large, toxic, American anuran, was introduced to Australia in 1935. Populations of many of Australia's reptiles (snakes, varanid lizards, crocodiles) and carnivorous mammals (dasyurid marsupials) have declined because these predators are killed by the toad's powerful toxins. In contrast to these well‐studied species, little is known about the cane toads impacts on Australian birds. We reviewed published and unpublished data on behavioral interactions between Australian avian predators and cane toads and collated distributional and dietary information to identify avian taxa potentially at risk from cane toad invasion. Cane toads are sympatric with 172 frog‐eating bird species in Australia, and an additional 8 bird species overlap with the predicted future range of the toad. Although many bird species thus are potentially at risk, behavioral observations suggest the risk level is generally low. Despite occasional reports of Australian birds being killed when they ingest cane toads, most birds either ignore toads or survive the predation event. The apparently higher tolerance of Australian birds to toad toxins, compared with Australian reptiles and marsupials, may reflect genetic exchange between Australian birds and Asian populations that encounter other bufonid species regularly and hence have evolved the capacity to recognize or tolerate this toxic prey.  相似文献

Summary. Field tests were performed to evaluate the response of the garden chafer, Phyllopertha horticola (Coleoptera: Scarabaeidae) to known scarab attractants and to investigate the influence of trap colour and height on the number of captured beetles. Both sexes were attracted by a lure mixture composed of geraniol, eugenol, and 2-phenylethyl propionate (PEP) (ratio 3:7:3). When testing floral volatiles individually, only geraniol, eugenol and methyl anthranilate but not PEP was attractive. Response of garden chafer to (Z)- 3-hexen-1-ol was strongly male-biassed. Both the response to floral volatiles and (Z)-3-hexen-1-ol was increased by using funnel traps with yellow instead of grey vanes. Traps positioned at 50 and 125 cm above ground captured significantly more garden chafers than those at 200 cm. The strongly male-biassed response to (Z)-3-hexen-1-ol suggests that in P. horticola (subfamily Rutelinae) orientation towards plant volatiles emitted upon mechanical damage of plants is part of the male mate finding strategy as recently demonstrated for cockchafers of the genus Melolontha (subfamily Melolonthinae). Possible application of plant volatiles for control of P. horticola is discussed.  相似文献

Markov Chain Monte Carlo on optimal adaptive sampling selections   总被引:1,自引:0,他引:1  
Under a Bayesian population model with a given prior distribution, the optimal sampling strategy with a fixed sample size n is an n-phase adaptive one. That is, the selection of the next sampling units should sequentially depend on the information obtained from the previously selected units, including the observed values of interest. Such an optimal strategy is in general not executable in practice due to its intensive computation. In many survey sampling situations, an important problem is that one would like to select a set of units in addition to a certain number of sampling units which have been observed. If the optimal strategy is an adaptive one, the selection of the additional units should take both the labels and the observed values of the already selected units into account. Hence, a simpler optimal two-phase adaptive sampling strategy under a Bayesian population model is proposed in this article for practical interest. A Markov chain Monte Carlo method is used to approximate the posterior joint distribution of the unobserved population units after the first phase sampling, for the optimal selection of the second phase sample. This approximation method is found to be successful to select the optimal second-phase sample. Finally, this optimal strategy is applied to a set of data from a study of geothermal CO2 emissions in Yellowstone National Park as a practical illustrative example.  相似文献

Summary Antennae of femalePapilio butterflies perceive many volatile plant constituents with widely differing, constituent-specific sensitivities. We compared the responses of threePapilio species to volatiles from host and non-host plants to assess species-specificity and the degree of evolutionary conservatism in olfactory responses.Since previous studies had demonstrated that the polar constituents in odor fromDaucus carota stimulate oviposition behavior inPapilio polyxenes, we collected headspace volatiles fromD. carota, Pastinaca sativa (both Apiaceae) andArtemisia dracunculus (Asteraceae) and separated the polar fraction of these volatiles by gas chromatography. GC-coupled electroantennograms (GC-EAG) were recorded from the speciesPapilio polyxenes, P. machaon hippocrates andP. troilus. In addition, the responses of the three species to five compounds known as generally occurring constituents of plant odor were recorded. The relative sensitivities for these compounds were nearly identical in all threePapilio species. The response spectra to the separated plant volatiles also showed considerable similarities among the species.From the limited set of GC peaks evoking a response in one of the species, 64% (D. carota), 44% (P. sativa) and 29% (A. dracunculus) also evoked a response in both of the other species. The responses of the two closely related Apiaceae feeders (P. polyxenes, P. m. hippocrates) to volatiles fromD. carota were more similar to each other than was either to the response ofP. troilus, which feeds on Lauraceae. However, this was not true for the responses to volatiles fromP. sativa. The least congruence among the three species was found in the responses to volatiles fromA. dracunculus, a non-host for all of them. The differences and similarities found in the response profiles of the threePapilio species are discussed with respect to evolutionary adaptation to host odor versus evolutionary conservatism in adaptation of olfactory receptors.  相似文献

Properly sampling soils and mapping soil contamination in urban environments requires that impacts of spatial autocorrelation be taken into account. As spatial autocorrelation increases in an urban landscape, the amount of duplicate information contained in georeferenced data also increases, whether an entire population or some type of random sample drawn from that population is being analyzed, resulting in conventional power and sample size calculation formulae yielding incorrect sample size numbers vis-à-vis model-based inference. Griffith (in Annals, Association of American Geographers, 95, 740–760, 2005) exploits spatial statistical model specifications to formulate equations for estimating the necessary sample size needed to obtain some predetermined level of precision for an analysis of georeferenced data when implementing a tessellation stratified random sampling design, labeling this approach model-informed, since a model of latent spatial autocorrelation is required. This paper addresses issues of efficiency associated with these model-based results. It summarizes findings from a data collection exercise (soil samples collected from across Syracuse, NY), as well as from a set of resampling and from a set of simulation experiments following experimental design principles spelled out by Overton and Stehman (in Communications in Statistics: Theory and Methods, 22, 2641–2660). Guidelines are suggested concerning appropriate sample size (i.e., how many) and sampling network (i.e., where).

Summary. We investigated the volatile emissions of Manchurian ash seedlings, Fraxinus mandshurica, in response to feeding by the emerald ash borer, Agrilus planipennis, and to exogenous application of methyl jasmonate (MeJA). Feeding damage by adult A. planipennis and MeJA treatment increased volatile emissions compared to unexposed controls. Although the same compounds were emitted from plants damaged by beetles and treated with MeJA, quantitative differences were found in the amounts of emissions for individual compounds. Adult virgin female A. planipennis were similarly attracted to volatiles from plants damaged by beetles and those treated with MeJA in olfactometer bioassays; males did not respond significantly to the same volatiles. Coupled gas chromatographic-electroantennogram detection (GC-EAD) revealed at least 16 antennally-active compounds from F. mandshurica, including: hexanal, (E)-2-hexenal, (Z)-3-hexen-1-ol, 3-methyl-butylaldoxime, 2-methyl-butylaldoxime, (Z)-3-hexen-1-yl acetate, hexyl acetate, (E)-β-ocimene, linalool, 4,8-dimethyl-1,3,7-nonatriene, and E,E-α-farnesene. Electroantennogram (EAG) dose–response curves using synthetic compounds revealed that females had a stronger EAG response to linalool than males; and male responses were greater to: hexanal, (E)-2-hexenal, (Z)-3-hexen-1-ol, 3-methyl-butylaldoxime, 2-methyl-butylaldoxime, and hexyl acetate. These results suggest that females may use induced volatiles in long-range host finding, while their role for males is unclear. If attraction of females to these volatiles in an olfactometer is upheld by field experiments, host plant volatiles may find practical application in detection and monitoring of A. planipennis populations.  相似文献
