The following areas are discussed in this paper: immobilisation of bacterial consortium in sol-gel; methyl parathion degradation and bioremediation applications; evaluation of indigenous bacterial isolates of contaminated soils. Bacterial strains were isolated from agricultural areas of Pakistan which were contaminated with methyl parathion. A bacterial consortium of seven (out of 64) Enterobacteriaceae isolates including Citrobacter, Enterobacter and Proteus vulgaris capable of degrading methyl parathion (enzyme activity ranging 410–675 mU mL?1 for individual isolates and 982 mU/mL for consortium) was selected and subsequently immobilised in tetraethyl orthosilicate (TEOS) and sodium-silicate-based sol-gel matrices. Cell viability of suspended and immobilised bacterial consortium was monitored using a minimal salt medium supplemented with methyl parathion. The results indicate that sol-gel immobilisation can be helpful to increase the shelf life of methyl parathion degrading bacterial strains along with preservation of biological activity for bioremediation applications in field.  相似文献

研究了假单胞菌 (DLL - 1)在水溶液介质中降解甲基对硫磷的性能与影响因素及其降解机理。结果表明 ,当菌体浓度为 10 5个·ml-1时 ,即发生快速的降解作用 ,3h时 ,降解率达 88.5 %。在pH为 5 .0、7.0和 9.0条件下 ,DLL - 1菌均产生对甲基对硫磷农药的高效降解作用。采用气相色谱 -质谱联用技术与离子色谱法 ,测定了DLL - 1菌 -甲基对硫磷作用过程产生的中间产物和最终产物 ,表明对硝基苯酚为主要中间产物 ,且DLL - 1菌能将其进一步降解为NO2 -和NO3 -。  相似文献

Adult pre-spawning fish Labeo rohita were sublethally (1/5th 96h LC50) exposed to mercuric chloride and metacid-50 (methyl parathion). Accumulation of mercury and methyl parathion was studied and it was found that pre-spawning ovary appears as a potent organ for deposition of both the pollutants. RNA/DNA ratio of the control and treated fish were studied. It was found that the signifcant decrease in RNA/DNA ratio occurs after 9 and 30 days of exposure for mercury and 30 days for methyl parathion. Fluorescence microscopic studies by acridine orange staining method were also performed to show how much it is related to biochemical alterations. In some cases loss of metachromasia is correlated with the fall in RNA/DNA ratio. Some other abnormalities like fall in stage II: stage I oocyte ratio and necrosis was also observed.  相似文献

从长期施用甲基对硫磷的污染土壤中分离到一株能以甲基对硫磷为唯一碳源生长且能将其完全矿化的细菌 D L L- 1 ,经鉴定,为邻单胞菌( Plesiomonas sp .) .该菌株3 h 内对50 mg/ L甲基对硫磷的降解率为93 % ,24h 内对50 mg/ L甲基对硫磷的降解率为95 % 以上.在葡萄糖铵盐培养基中, D L L1 对甲基对硫磷的耐受浓度和降解速度均增大.降解曲线表明延滞期内,菌体依靠上一生长阶段分泌的酶类对甲基对硫磷进行降解,一旦菌体开始生长,则检测不到中间代谢产物对硝基苯酚的存在.生长情况和粗酶液试验均显示 D L1 优良的降解性状  相似文献

Toxicity of methyl parathion was studied on survival of Rana tigrina tadpoles. The LC50 values for short-term (upto 96 h) and long term studies (hatching to froglet) were determined. The LC50 values for 96 h and the total larval period were 4.36 and 1.30 ppm respectively. The toxicity of methyl parathion enchanced in relation to increase in concentration and duration. Teratological defects such as scoliosis and failure in emergence of fore limb were noticed. Safe levels were calculated for short-term (0.1 ppm) and long-term (0.03 ppm) studies. The present work stresses the neccessity of long term study especially for the developing amphibians.  相似文献

Methyl parathion is an organophosphate insecticide that has been used in agriculture and the domestic sector for several years. This pesticide and others, arriving through different processes, exert significant effects on water quality with serious consequences for environmental and human health. The main objective of this study was to investigate the changes of Glutathione S-transferase enzyme activity in methyl parathion exposed rat tissues. For this purpose, wistar rats (Rattus norvegicus) were injected intraperitoneally with 7 mg kg?1 dose of methyl parathion, while corn oil was applied to control groups in the same way. The liver, kidneys, brain and small intestine were quickly removed after 0, 2, 4, 8, 16, 32, 64, and 72 hours of injection of methyl parathion and the glutathione S-transferase activity was determined in these tissues. As a result it was seen that glutathione S-transferase activity increases in all tissues in the group of male and female rats to which methyl parathion was given. The increase of glutathione S-transferase activity may be a result of methyl parathion's toxic effect because it is one of the most important enzymes of detoxification metabolism.  相似文献

This study provides an electrocoagulation process for the removal of metals such as cobalt, copper, and chromium from water using magnesium as anode and galvanized iron as cathode. The various parameters like pH, current density, temperature, and inter electrode distance on the removal efficiency of metals were studied. The results showed that maximum removal efficiency was achieved for cobalt, copper, and chromium with magnesium as anode and galvanized iron as cathode at a current density of 0.025?A?dm?2 at pH 7.0. First- and second-order rate equations were applied to study adsorption kinetics. The adsorption process follows second-order kinetics model with good correlation. The Langmuir and Freundlich adsorption isotherm models were studied using the experimental data. The Langmuir adsorption isotherm favors monolayer coverage of adsorbed molecules for the adsorption of cobalt, copper, and chromium. Temperature studies showed that adsorption was endothermic and spontaneous in nature.  相似文献

Electrocoagulation (EC) technique was adopted for the treatment of fluoride-containing drinking water in a 3?L capacity batch reactor. Experimental investigation was carried out to observe the effect of different operating parameters, such as initial fluoride concentration, current density, inter-electrode distance, on the removal of fluoride from the fluoride-rich drinking water. Aluminum flat sheet was used as the electrode material. It was observed that with an increase in current density, percentage removal of fluoride increased. It was seen that at electrode distance of 0.005?m, removal of fluoride attained its maximum compared to the other performances obtained at different inter-electrode distances. Life time of the aluminum electrodes was estimated from the electrode corrosion observed during the experiment. Total operating cost was estimated as a combination of energy cost and electrode cost. The total operating cost was evaluated as 0.38?US$?m?3 to remove fluoride from the solution with an initial fluoride concentration 10?mg?L?1. Characterizations of the electrochemically generated by-products were carried out by powder X-ray diffraction, Fourier transform infra-red Raman spectroscopy, scanning electron microscopy image and corresponding elemental analysis.  相似文献

Trace amounts of heavy metals have been analysed by electrochemical techniques in ecstasy tablets obtained from different police seizures in Spain. Lead, cadmium, copper and zinc were determined by differential-pulse anodic stripping voltammetry at a hanging mercury drop electrode, whereas nickel and cobalt were determined by adsorptive differential-pulse cathodic stripping voltammetry from their dimethylglyoxime complexes, M(DMG)2. The performance of the procedure was compared with electrothermal atomic absorption spectrometry. The procedure was applied to the determination of these elements in nine ecstasy samples, finding that Zn is the element present in the highest concentration, ranging from 0.3 to 200?mg?kg?1, Ni, Cu appear below 15?mg?kg?1 and Pb below 8?mg?kg?1, while Cd and Co levels were always lower than 0.51?mg?kg?1.  相似文献

从农药厂污泥中分离得到一个降解甲基一六○五的混合菌群,该菌群由两种菌 M6 和 P3 组成,初步鉴定均为假单胞菌属( pseudmonas .sp) . M6 菌具有一硫代磷酸酯键水解酶,能够催化甲基一六○五水解为对硝基酚, P3 具有对硝基酚降解能力. M6 和 P3 各自均不能彻底矿化甲基一六○五. M6 与 P3 混和可以彻底降解甲基一六○五.质粒消除实验表明, S D S、吖啶橙( O A) 、丝裂霉素及几种消除剂混和处理均不能使 M6 和 P3 丧失一六○五水解和对硝基酚降解能力,质粒检测发现在 M6 中检测到质粒条带,而 P3 中未检测到质粒条带  相似文献

用GC-MS-MS同时测定蔬菜中的甲拌磷、二嗪农、甲基对硫磷、毒死蜱、倍硫磷、喹硫磷、杀扑磷和亚胺硫磷.对各个化合物的特征离子对、碰撞电压进行了优化,并阐述了优化反应监测模式(MRM)条件的过程和方法,同时分析了各个化合物的线性范围和最低检出限.将该方法用于蔬菜样品的检测,测得加标回收率在90.4%-105.0%之间,RSD在3.2%-9.3%之间.  相似文献

ZnO/TiO2 composites were synthesized by using the solvothermal method and ultrasonic precipitation followed by heat treatment in order to investigate their photocatalytic degradation of methyl orange (MO) in aqueous suspension under UV irradiation. The composition and surface structure of the catalyst were characterized by X-ray diffraction (XRD), field emission scanning electron microscope (FE-SEM), and transmission electron microscopy (TEM). The degradation efficiencies of MO at various pH values were obtained. The highest degradation efficiencies were obtained before 30 min and after 60 min at pH 11.0 and pH 2.0, respectively. A sample analysis was conducted using liquid chromatography coupled with electrospray ionization ion-trap mass spectrometry. Six intermediates were found during the photocatalytic degradation process of quinonoid MO. The degradation pathway of quinonoid MO was also proposed.  相似文献

The photodegradation of aqueous solutions containing 0.2 mM methyl parathion has been studied through the optimization of the [H2O2]/[Fe3+] ratio in a Fe3+/H2O2/UV-C flow system of 1.3 L capacity. The decay kinetics and TOC abatement have been analyzed for the experiments performed at pH 3.0 and room temperature. All experiments lead to the total methyl parathion destruction after a few minutes, following a pseudo-first-order decay kinetics. Total mineralization can be reached after 120 min at the optimum ratio found, due to the synergistic effect of the very oxidizing hydroxyl radical (·OH) produced via the Fenton reagent and the effective photodecarboxylation at 253.7 nm.  相似文献

通过筛选敌敌畏、马拉硫磷和对硫磷3种有机磷农药对水生生物的急性毒性数据和乙酰胆碱酯酶抑制效应数据,构建物种敏感度分布曲线进行了比较分析。结果表明,敌敌畏对水生生物的急性毒性和乙酰胆碱酯酶抑制效应的大小顺序为:酶体内抑制效应酶体外抑制效应急性毒性;马拉硫磷和对硫磷的乙酰胆碱酯酶抑制效应数据不足但趋势相似,顺序为:酶体内抑制效应急性毒性酶体外抑制效应。敌敌畏的急性毒性和酶体外抑制效应的5%危害浓度(HC5)分别为2.07μg·L~(-1)和1.53μg·L~(-1),两者相差1.4倍。在水质基准推导中,乙酰胆碱酯酶抑制效应数据对有机磷农药的水生生物基准具有重要的参考价值。  相似文献

In this study, the adsorption characteristics of As(III) and As(V) from water and wastewater using polyacrylamide-grafted banana stem with quaternary ammonium functionality (PGBS-AE) were investigated. Infrared spectroscopic, and thermogravimetric analyses were performed to affirm the polymer grafting, functionality, morphology, and thermal stability. Batch experiments were carried out to understand the effect of contact time, concentration, pH, adsorbent dose, and temperature of the solution for the adsorption of As(III) and As(V) onto PGBS-AE. Equilibrium was achieved within 1 h and the optimum pH was found to be 9.0 and 3.0 for As(III) and As(V), respectively. Isotherm studies showed that the Langmuir equation fits best. Maximum adsorption capacities of 50 and 5.5?g?kg?1 were obtained for As(III) and As(V) at 30°C. The endothermic nature of adsorption was evident as the adsorption efficiency increased with temperature. The thermodynamic parameters were evaluated to explain the feasibility of adsorption and to predict the nature of adsorption. The competence of the adsorbent for practical purposes was also analyzed by treating with a fertilizer industry effluent sample. Studies pertaining to adsorbent regeneration and readsorption of As(III) and As(V) were carried out for four consecutive cycles.  相似文献

Carbon capture and storage has become a new mitigation option to reduce anthropogenic CO2 emissions. The effects of the CO2-related acidification, associated with unpredictable leaks of CO2 during the operational phases were studied using the Paracentrotus lividus sea urchin-liquid-phase-assays (fertilisation and embryo-larval development tests). Besides to the direct effects of the CO2-associated pH decrease, the possible effects on marine sediment toxicity were studied by exposing the urchin larvae to elutriate of sediments with different metals concentration, which were subjected to various pH treatments. The resulted median toxic effect pH ranged from 6.33?±?0.02 and 6.91?±?0.01 for the egg fertilisation, and between 6.66?±?0.03 and 7.16?±?0.01 for the larval development assays. Concentrations of Co, Zn, As, Cu, and Fe were detected in the sediment elutriates. For all the sediment elutriates tested the amount of the dissolved Zn increased in parallel with the pH reductions. Correlated effects of acidification, biological response, and the presence of dissolved metals were observed in this work evidencing that the most important factor controlling the reduction of egg fertilisation and larval development success was the combined effects of the reduction of pH and the presence of the contaminants (mainly Zn, Co, and As).  相似文献

A chemically modified carbon paste electrode for chromium (III) based on the formation of ion-association complex of chromium with 5-ethyl-6-oxo-5-pentan-2-yl-sulfanyl-pyrimidin-4-olate (thiopental, THP) as electroactive ion-exchanger (Cr-THP) was prepared and investigated. The electrode has a linear dynamic range of 1.7?×?10?6–1.3?×?10?2?mol?L?1, with a Nernstian slope of 19.6?±?0.5?mV per decade and a detection limit of 9.0?×?10?7?mol?L?1. It has a fast response time of <15?s and can be used for at least 7 weeks without any considerable divergence in potential. The proposed sensor revealed good selectivity for Cr(III) over a wide variety of mono-, di-, and trivalent cations and could be used in the pH range of 2.3–6.5. The proposed electrode was used in batch measurements and in flow injection analysis. The Cr-CMCPE was successfully applied for the determination of Cr(III) in real samples (wastewater of hot dip galvanizing unit and alloys). The present electrode was also used as an indicator electrode in potentiometric titration of Cr(III). The results were highly satisfactory.  相似文献

The extensive extraction of arsenic (As)-contaminated groundwaters for drinking, household and agricultural purposes represents a serious health concern in many districts of Bangladesh. This laboratory-based incubation study investigated the sources and mechanisms of As mobilization in these groundwaters. Several incubation studies were carried out using sediments collected from the Bangladesh aquifer that were supplemented, or not, with different nutrients, followed by an analysis of the sediment suspensions for pH, ORP (oxidation-reduction potential), EC (electrical conductivity) and As and Fe(ΙΙ) concentrations. In the substrate-amended sediment suspensions incubated under anaerobic environment, there was a mobilization of As (maximum: 50–67 μg/l) and Fe(ΙΙ) (maximum: 182 μg/l), while the ORP value decreased immediately and drastically (as much as −468 mV to −560 mV) within 5–6 days. In the sediment suspensions incubated under control and aerobic conditions, no significant As mobilization occurred. The simultaneous mobilization of As and Fe(ΙΙ) from sediments is a strong indication that their mobilization resulted from the reduction of Fe oxyhydroxide by the enhanced activity of indigenous bacteria present in the sediments; this phenomenon also provides insights on the mobilization mechanism of As in groundwater. The concentrations of As in the sediments used in the incubation studies were strongly linked to the gradients of redox potential development that was stimulated by the quantity of organic nutrient (glucose) used. The penetration of surface-derived organic matter into the shallow aquifer may stimulate the activity of microbial communities, thereby leading to a reduction of iron oxyhydroxide and As release.  相似文献
