Fish are often exposed to highly contaminated water, especially in areas where the dilution rate of waste water is low. Environmental pollutants or other stress may affect one or more of the immunological functions in fish. Effective control of toxic substances is essential for the success of fish culture, which requires fish health monitoring using biomarkers. This study evaluated the effects of heavy metal copper (Cu) at environmentally relevant concentrations on the hematopoiesis in Labeo rohita. L. rohita fingerlings were exposed to different Cu concentrations for 6, 24, 48, or 72 hr. Following exposure, percentage blast cells was found to be increased after 6 and decreased after 24, 48, and 72 hr both at sublethal and LC50 concentrations, whereas percentage mature erythrocytes decreased after 6 and increased after 24, 48, and 72 hr. Erythropoietic and leukopoietic efficiencies were also affected upon exposure. Erythropoietic and leucopoietic efficiencies increased markedly after 6, 24, and 48 hr but decreased after 72 hr in sublethal-treated fish. Erythropoietic efficiency decreased significantly after 6, 24 and 48 hr but increased after 72 hr in LC50-treated fish. Leukopoietic efficiency decreased significantly after 6 hr exposure and increased after 24 and 48 hr but decreased after 72 hr. Flow cytometric studies of head kidney cell cycle phase distribution revealed cell death and DNA synthesis. The percentage cell death increased in fish exposed to sublethal concentrations and rose further at LC50 during the earlier hours of exposure. Synthesis of DNA was increased at sublethal concentrations but was significantly reduced at LC50 dose.  相似文献

龟壳攀鲈是一种在极端污染水域中具备生存优势的鱼类,收集重污染区和轻污染区的龟壳攀鲈,并通过比较其体内各组织中的酸性磷酸酶(ACP)及碱性磷酸酶(AKP)活性来揭示其耐污能力。贵屿作为重污染区有4个采样点:大陇村、市上村、联堤村和北林村;潮州作为轻污染区有2个采样点:卧石村和洗马桥。水质结果表明,贵屿采样地水中的Pb、Cd、Cr、As、Hg含量分别是潮州采样点的27.6倍、3.9倍、5.8倍、3.9倍和3.5倍,说明贵屿采样地的污染远高于潮州的。酶活结果表明,在龟壳攀鲈所有组织中,内脏组织(胃肠、性腺、肝脏、脾脏、肾脏)中的ACP和AKP活力均高于外周组织(肌肉、鳞片、鳃);重金属污染均能提高攀鲈体内所有组织的ACP和AKP活力;AKP活性更容易受到重金属的影响,各组织的AKP活力增幅普遍超过ACP活力增幅;其中,受重金属影响最大的组织是脾脏,ACP和AKP活力可增长到2.37倍和6.21倍。这些结果说明了龟壳攀鲈可以通过显著升高内脏组织(如肠胃、肝脏、肾脏等)中ACP和AKP活性,以便更好地应对高污染环境;其中AKP更容易受到重金属的影响,适合用于作为评估污染的指标。  相似文献

The present study was undertaken to assess the toxicity of sublethal concentrations (125, 250, and 375 µg L?1) of chlorpyrifos on Anabas testudineus for 21 days. The morphological changes on the gills, scales, and erythrocytes of A. testudineus were observed using scanning electron microscopy. Gill alterations included highly active mucous cells, epithelial hyperplasia, and fusion of secondary lamellae. The scales showed damaged lepidonts. Oozed out cytoplasmic content and lobopodial projections were observed in the erythrocytes after exposure to chlorpyrifos.  相似文献

Lethal concentrations (LC50) of a synthetic pyrethroid pesticide, fenvalerate, for three species of air breathing fish Clarias batrachus, Channa punctatus and Heteropneustes fossilis were determined under water and acetone soluble condition in the laboratory using the static bioassay procedure of the American Public Health Association (APHA,1995). Acetone soluble fenvalerate was found more toxic than the water-soluble fenvalerate irrespective of species and exposure periods. The LC50 value upon 96 days exposure to acetone soluble fenvalerate for C. batrachus, Channa C. punctatus and Heteropneustes H. fossilis were 1.35, 1.0 and 0.65?µg?L?1, respectively. It is concluded from the present study that fenvalerate is highly toxic even to the hardy air breathing fishes and the pesticide, when dissolved in water, remains photostable and active to render toxicity for long duration.  相似文献

The purpose of this study was to examine the sublethal and/or lethal effects produced by exposure of teleostean fish Solea senegalensis to γ isomer of lindane, γ-hexachlorocyclohexane (γ-HCH). S. senegalensis was exposed to 0.1, 1 or 10 mg L?1 of lindane for 96 h. Samples of different tissues (gills, liver and kidney) were extracted and processed for histopathological observations. The 72 h LC50 value of pesticide was found to be 0.16 ± 0.04 mg L?1 γ-HCH. Histological observations including aneurysms and hypertrophy of lamellar epithelia in gills, blood stagnation and vacuolization in liver and alterations in the hematopoietic tissue in kidney were observed. The results of the study show the sensitivity of juvenile of S. senegalensis to lindane and histology as a reliable tool to detect the sublethal effects of pollutants on the environment and aquatic biota. The toxicological implications arising from these results are the subject for further multi-concentration tests useful in establishing water quality criteria.  相似文献

The population of climbing perch, Anabas testudineus, an important food fish in India, is presently under threat and has been declared endangered. A narrow range of environmental conditions for successful reproduction of the species was previously ascertained in breeding experiments. In this study, the role of physiological parameters including plasma glucose, total protein, cholesterol, and glycogen content in liver and ovary were determined with respect to the reproductive cycling stages. Flow cytometric analysis of cell cycle of ovary and testis in relation to reproductive cycle was also analyzed. In both the sexes, plasma glucose and plasma protein changed significantly during breeding cycle. Plasma cholesterol levels were reduced markedly during spawning. Liver and ovarian glycogen levels showed significant variation during reproductive cycling. Flow cytometric study revealed that the hypoploid and synthesis phase were altered significantly with breeding cycle in ovarian and testicular cells. Data indicate that the biochemical parameters indicative of the fish reproduction capacity are influenced by environmental conditions. Thus, climatic changes may therefore potentially induce biochemical alterations that may exert detrimental effects on fish reproduction and subsequent population decline.  相似文献

The effect of carbofuran, a carbamate pesticide, on the breeding performance of the freshwater fish Labeo rohita (Hamilton) has been investigated in the present study. Breeding of L. rohita was conducted after treatment with three sublethal concentrations, i.e. 0.06, 0.15 and 0.30 mg L?1 of carbofuran for 96 h. The present investigation showed that the number of total eggs and total amount of eggs (litre per kg body weight) decreased significantly (p < 0.01) at all concentrations of carbofuran in comparison to controls, while the reduction in fertilization percentage at all concentrations of carbofuran was not significantly different from controls. The reduction in hatching percentage, the expected number of hatchlings, and the expected number of hatched larvae were significantly (p < 0.01) different between treated and control groups at all carbofuran concentrations. No significant differences for the 96 h survivability of hatched larvae were reported at all concentrations of carbofuran.  相似文献

The objective of this study was to examine the toxicological effect of two major heavy metal pollutants, lead chloride (PbCl2) and cadmium chloride (CdCl2), in the freshwater climbing perch, Anabas testudineus. Fish were exposed to 1.0 and 2.0 mg/L both metals and histopathological changes in gill, kidney and liver tissues were studied. Major changes observed in gill tissue were epithelial lifting, proliferation of epithelial cells, fusion of secondary lamellae, hyperplasia and hypertrophy of mucous cells, and necrosis of epithelial cells. Cell necrosis, degenerated kidney tubules, congestion, lymphocytic infiltration and vacuolation were the major abnormalities observed in kidney tissue. The most conspicuous changes in liver were darker nucleoli, irregularly shaped hepatocytes with dilated blood capillaries, and focal as well as single necrosis. Fish specimens exposed to PbCl2 exhibited pronounced changes in all tissues examined compared with those exposed to CdCl2. It is evident from this study that heavy metals can cause significant histopathological changes in fish tissue.  相似文献

Studies conducted on the distribution, fate and metabolism of DDT in a model ecosystem simulating a tropical marine environment of fish, Gobious nudiceps, Lethrinus harak, Gobious keinesis, Gobious nebulosis and white shrimp (Panaeus setiferus), show that DDT concentration in the water decreases rapidly within the first 24?h. Rapid accumulation of the pesticide in the biota also reaches a maximum level in 24?h before gradually declining. The bioaccumulation factors calculated for the fish species (G. keinesis) and white shrimp (P. Setiferus) were 270 and 351, respectively, after 24?h. There was a steady build up of DDT residues in the sediment during the first 24?h which continued to a maximum concentration of 6.66?ng/g in the seawater/fish/sediment ecosystem after 3 weeks and 5.27?ng/g in the seawater/shrimps/sediment ecosystem after 2.7 days. The depuration of the accumulated pesticide was slow with only 54% lost in G. nudiceps within 3 days of exposure in fresh sea water. By contrast, depuration was fast in the white shrimp, which lost 97% of the accumulated pesticide under the same conditions. DDT was found to be toxic to two of the fish species (G. nebulosis and L. harak) and to white shrimp, and the degree of toxicity was dependent on the particular species. The 24?h LC50 at room temperature for the fish species G. nebulosis and white shrimp was found to be 0.011 and 0.116?mg/kg, respectively. These levels are comparable to the ones recorded for the temperate organisms. Degradation of DDT to its primary metabolites, DDE and DDD, was found in all the compartments of the ecosystem with DDE being the major metabolite in the fish, shrimps and sediment, while in seawater, DDD dominated as the major metabolite.  相似文献

Toxicities of an azo dye methyl red and a heavy metal copper (Cu) were quantified, using growth and mortality as end points, in four plant species and three animal species by subjecting them to short-term (4 days for animals, 10 days for plants) static bioassays. Lemna aequinoctialis Welwitch (EC50: 7–16 ppm) was found to be the most sensitive species for methyl red, Ceratophyllum demersum L. (EC50: 25 ppm) and Lactuca sativa L. (EC50: 56 ppm) were intermediate, while Oryza sativa L. shows reduction in seedling vigor (9–27%) of <50%, being the least sensitive amongst the tested plant species. Methyl red toxicity is almost 3–5-fold higher in growing medium (pH = 5.8–6.0), even at high nutrient levels, while Cu toxicity is higher in nutrient-poor alkaline medium at alkaline pH (8.3–8.7; EC50: Ceratophyllum = 104–200 ppb; Lemna = 100–170 ppb) compared to nutrient-rich acidic medium (pH = 5.4–5.7; EC50: Ceratophyllum = 2600–3175 ppb; Lemna = 4350–4715 ppb). Rice tolerance (EC50: 6500 ppb) was found to be higher than hydrophytes while lettuce was most tolerant to Cu. Fish sensitivity toward the test chemicals was almost parallel to Ceratophyllum and Lemna [Gambusia affinis Baird and Gerard (LC50: 250 ppb for Cu) and Poecilia reticulata Peters (LC50: 24 ppm for methyl red)]. Similar to the plants, dye toxicity increased markedly (LC50: 7 ppm) in the acidic medium (pH = 6.0). Amongst the tested organisms, Daphnia was found to be most sensitive to methyl red (EC50: 6 ppm) while its sensitivity to Cu (EC50: 230 ppb) was similar to hydrophytes and fish. Initially, the combination of dye and Cu (at their sublethal concentrations) had additive effects in duckweed, while dye concentration ruled afterward. These results indicate that hydrophytes and animals are equally sensitive toward the test chemicals. Dye toxicity in hydrophytes and fish was pH dependent, while in the case of Cu, it is related to the nutrient status of the growth medium of plants.  相似文献

The effects of deltamethrin at acute and sublethal concentrations on some hematological and biochemical profiles in the Indian major carp Labeo rohita have been examined. The lethal concentrations (LC50) of deltamethrin for 24 and 96 h were found to be 0.44 and 0.38 mg L?1, respectively. During acute treatment (24 h), hemoglobin (Hb), hematocrit (Hct), red blood cell (RBC), white blood cell (WBC), mean cellular volume (MCV), mean cellular hemoglobin (MCH), and mean cellular hemoglobin concentration (MCHC) values were significantly increased compared to control group. Plasma glucose, protein, cholesterol, albumin, globulin, triglyceride, and glycogen in gill and muscle were significantly decreased. Upon sublethal treatment (0.044 mg L?1, 10th of 24 h LC50), RBC and the biochemical parameters, except glucose and cholesterol, decreased significantly, while MCV, MCH, and MCHC values increased up to the 28th day and then declined. Cholesterol level was significantly increased throughout the study period. A biphasic response in the levels of Hb, Hct, WBC, and glucose were observed during sublethal treatment.  相似文献

The concentrations of Al, As, Cd, Cr, Cu, Fe, Hg, Mn, Ni, Pb, Sn and Zn were analysed in water, sediment, muscle of climbing perch fish (Anabas testudineus), and tissue of water hyacinth plants (Eichhornia crassipes) collected from a former tin-mining pond in Perak, Malaysia. The monitoring was performed during the minimum and maximum rainfall periods. The concentrations of As, Cr, Fe, Ni, Pb and Zn in water exceeded the permissible limits set by the Interim National Water Quality Standards for Malaysia (INWQS). The risk index (RI) values determined for As, Cd, Cr, Cu, Hg, Pb and Zn in sediment presented a low degree of ecological risk. The order of the top three heavy metals measured in fish muscle was Zn?>?Fe?>?Al. The estimated daily intake (EDI) of As and Cr exceeded the allowable limits in both rainfall periods. The bioconcentration factor (BCF) and translocation factor (TF) values of Zn in water hyacinth were 24.865 and 3.214, respectively. The concentrations of Cu and Zn in the plant tissue were significantly correlated (p?<?.05) with the concentrations of heavy metals in water. Overall, climbing perch and water hyacinth are excellent bioindicators of environmental impacts on water bodies.  相似文献

The acute toxicity of three formula grade pesticides namely, triazophos (an organophosphate, OP), deltamethrin (a pyrethroid) and combined pesticide (triazophos?+?deltamethrin) was determined in earthworm, Eudrilus eugeniae. They were exposed to different concentrations of these pesticides for 48 h by paper contact toxicity method. The LC50 values for triazophos, deltamethrin and of combination were determined as 0.076, 0.031 and 0.065?μg/cm2, respectively. To study the sublethal effect of these pesticides, E. eugeniae were exposed to 5% and 10% of LC50 of pesticides for 48 h. Morphological alterations such as coiling, clitellar swelling, mucus release and bleeding followed by body segmentation were observed in exposed earthworm. Acetylcholinesterase (AChE) activity assayed in different regions of body segment exhibited a significant (p?<?0.05) decrease in its activity particularly in the pre-clitellar region as compared to other regions. The altered behavioural responses in pesticides exposed earthworms would have been due to decline in AChE activity of the nervous system.  相似文献

Sulfide tolerance and detoxification in shallow-water marine fishes   总被引:4,自引:0,他引:4  
Hydrogen sulfide is a potent inhibitor of aerobic respiration. Sulfide is produced in sediments, and many species of fish live in association with the bottom. Tolerance tests, enzyme assays, and chromatography of sulfur compounds in thirteen species of shallow-water marine fishes (collected in San Diego, California, USA in 1987–1988) indicate adaptations to sulfide that vary with habitat and lifestyle. Tidal-marsh inhabitants, like Gillichthys mirabilis and Fundulus parvipinnis, have higher tolerance to sulfide (96 h LC50 at 525 to 700 M) relative to outer-bay and open-coast inhabitants (surviving <12 h at much lower concentrations). The cytochrome c oxidase of all species shows high activity and susceptibility to sulfide poisoning, with 50% inhibition at 30 to 500 nM in various tissues. The two marsh species are able to survive at sulfide concentrations already inhibitory to their cytochrome c oxidase and fatal to other species. All species detoxify sulfide by oxidizing it to thiosulfate. All have sulfide-oxidizing activity in the blood, spleen, kidney, liver and gills, which correlates significantly with heme content. Thiosulfate appears in the tissues of sulfide-exposed fish and builds up to high concentrations (up to 2 mM) with stronger and longer exposure. Unexposed fish contain little or no thiosulfate. Sulfide is barely detectable in the tissues, even in high-sulfide exposure tests. We suggest that fish blood, in having high sulfide-oxidizing activity and no cytochrome c oxidase, can act as a short-term first line of defense against sulfide, and thus minimize the amount that reaches the vital organs. The results of this study indicate that sulfide is a significant environmental factor influencing the ecological distribution of marine fishes.  相似文献

Fresh and healthy specimens of Oreochromis niloticus (mean weight 70.5± 1.23 g and mean length, 12.3± 1.5cm) and Clarias gariepinus (mean weight, 41.8±2.3 g and mean length 13.78 ± 1.2 cm) were exposed to heavy metal lead, ranging in concentration from 10 to 18 mg L‐1 for O. niloticus and 20 to 28 mg L‐1 for C. gariepinus respectively. The 96 h LC50 for both the species was computed as 12.45 mg L‐1 (O. niloticus) and 22.65 mg L‐1 (C. gariepinus). The cichlid fish was exposed to sublethal concentration of lead (2, 4 and 6 mgL‐1) and catfish (4, 8 and 12 mgL‐1). The erythrocytes count, haemoglobin concentration, haematocrit and the value of serum glucose increased significantly (P < 0.01), whereas leucocytes count, serum protein and serum triglyceride after intoxication declined in both the species with the same pattern but in different quantity. After 96 h of time a significant depletion of muscle and liver glycogen was noted, being greater in muscle (16.7% in O. niloticus) and in liver (15.9% in C. gariepinus). Sublethal effects of chromium trioxide on mosquito fish, Gambusia affinis, were carried out for 28 days on day 4, 8, 12, 16, 20, 24, and 28. The sublethal effects of chromium (LC10, 77.62?mg?L?1) on antioxidant enzymes, thiobarbituric acid reactive substance (TBARS), locomotor behavior, and gill morphology were studied. The antioxidant enzymes like superoxide dismutase (SOD) and catalase were induced throughout the exposure span of 28 days. Percent induction was also increase with the days of exposure. Lipid peroxidation product, malondialdehyde was enhanced in the viscera tissue of chromium-exposed fish. Perturbation of locomotor behavior like decrease in distance travelled (m?min?1) and in swimming speed (cm?s?1) was observed in exposed fish. Chromium toxicity caused alterations in gill morphology like hypertrophy and hyperplasia in secondary lamellae followed by detached epithelium with severe necrosis. It can be concluded from the results that Cr intoxication in Gambusia fish, caused induction in antioxidant enzymes and lipid peroxidation, plus changes in gill morphology and locomotor behavior.  相似文献

Oxidative stress and antioxidant responses of crucian carp, upon chronic exposure to endosulfan, were evaluated in vivo. The lethal concentration (LC50–96?h) was 70 μg L?1; on its basis, the fish were exposed to endosulfan at 20, 35, and 50 μg L?1 and autopsy was done on days 1, 2, 3, 4, 7, 14, 21, 28, and 35. Lipid peroxidation was induced in a concentration-dependent manner, being highest at 50 μg L?1 (3/4 LC50–96 h, sub-lethal concentration-I, SL-I) on day 4 (720% versus control), followed in its extent (490%) at 30 μg L?1 (1/2 LC50–96 h, sub-lethal concentration-II, SL-II) on day 7 and lowest (260%) at 10 μg L?1 (1/4 LC50–96 h, sub-lethal concentration-III, SL-III) on day 14. Glutathione showed a concentration- and time-dependent elevation in the initial phase, with highest level on day 4 (180%) at SL-I, but showed significant reduction in all test concentrations from day 21 of post-exposure. Superoxide dismutase was decreased significantly throughout the study, with highest reduction (63%) on day 4 at SL-I; catalase increased in all test concentrations up to day 14 but showed a significant decrease from the day 28 of post-exposure. The potential role of these parameters as indicators of pesticide pollution in aquatic systems is discussed.  相似文献

This study was conducted to evaluate the toxicological response of p-nitrotoluene and p-nitroaniline to the key fish species, Cyprinus carpio. A freshwater fish bioassay based on the 96 h LC 50 was used to estimate the single and joint toxicity of the two chemicals. The toxicity of p-nitrotoluene was greater than that of p-nitroaniline based on 96 h LC 50 values of 40.74 mg·L?1 and 48.99 mg·L?1, respectively. Both compounds had moderate toxicity toward Cyprinus carpio, and this toxicity increased with the exposure duration and concentration. Binary mixtures of the compounds were more toxic than the individual compounds at 96 h, and they acted upon partial addition. When the exposure time was longer, the toxicity increased for mixtures of compounds with the same concentration or toxicity. The results of this study suggest that exposure to a combination of these chemicals would result in a higher environmental risk in aquatic systems than exposure to either compound alone. Further research is needed to investigate the combined effects and sublethal toxicity of p-nitrotoluene and p-nitroaniline, since they are both still used in China.  相似文献

Profenofos and trizaophos (organophosphates) carbofuran and carbaryl (carbamates) are among the major toxicants polluting freshwater bodies, as well as exerting a significant effect on the health of fish. In this study, the 96 h LC50 of the most commonly used organophosphates and carbamates was determined in one of the indigenous fish species, Labeo rohita fingerlings. Pesticides were applied to fingerlings that had been grown under optimized standard conditions under a maintained static bioassay system. Probit analysis was used for the determination of LC50 values, which were ascertained as 6.64, 0.32, 1.4, and 8.24 mg/L for triazophos, profenofos, carbofuran, and carbaryl, respectively. Acute toxic stress was noted with fish exhibiting behavioral toxicity including suffocation, lying on the bottom, erratic swimming, lethargy and downward movements, and gulping prior to mortality.  相似文献
