Membrane damage is one of the important consequences of chromium (Cr), an environmental toxicant, induced cytotoxicity. Reduced glutathione (GSH), a membrane protectant may be used to reduce the Cr-induced membrane damage. In the present study, the impact of Cr in presence and absence of GSH was studied on plasma membrane of the liver and kidneys in male Wistar rats. Significant increases in membrane cholesterol levels as well as significant decreases in membrane phospholipid levels in Cr exposed (0.8 mg per 100 g body weight, i.p., for 28 days) animals suggest structural alterations in both the liver and kidney plasma membranes. Alkaline phosphatase (ALP), total ATPase, and Na+–K+–ATPase activities of plasma membrane were significantly decreased in both the liver and kidneys after Cr treatment. This treatment also produced significant weight loss and increased Cr content in the liver and kidneys. However, GSH (8 mg per 100g body weight, i.p., daily at an interval of 6 h after injection of Cr for a period of 28 days) supplementation restored alterations induced by Cr in plasma membrane of both the liver and kidneys but was not able to eliminate the deposited Cr from the liver and kidney tissues.  相似文献

The therapeutic effect of melatonin (MEL) against aluminum (Al)-induced neurotoxicity was investigated in mouse cerebellum. Two groups of male albino mice were intraperitoneally injected with Al acetate or MEL alone, at doses of 3.5 or 7?mg?kg?1?day?1, respectively, for 6 weeks. During this period, another group of animals received a combination of both Al and MEL (3.5?+?7?mg?kg?1?day?1). At the end of the treatment cerebellum was removed and processed to examine the oxidative stress markers: superoxide dismutase (SOD), catalase (CAT), and thiobarbituric acid-reactive substances (TBA-RS). Oxidative stress increased significantly with administration of Al which was estimated by increased TBA-RS and reduction in the activities of SOD and CAT. However, these alterations were significantly reversed significantly following MEL treatment which was observed in co-administered group. Protective effects of MEL were also observed at electron microscopic level. Ultrastructural studies revealed an increase in vacuolization, chromatin condensation within the nucleus, degenerated purkinje cell, degenerated axon and degenerated granule cells in the cerebellum of Al-treated mice group whereas concurrent administration of MEL with Al reduced these changes. The results of the present investigation emphasize the potential use of MEL as a supplement in therapy of free radical based neurological disorders in which oxidative stress is involved.  相似文献

Aluminum (Al) is a neurotoxicant potentially affecting ionic, cholinergic, and dopaminergic neurotransmission in the central nervous system. These alterations are known to be associated with learning ability, adaptive responses, and other aspects of behavior. The present experiment was designed to study the neurotoxic consequences of Al exposure on neurotransmitters like dopamine (DA), serotonin (5-HT), norepinephrine (NE) along with the activity of acetylcholinesterase (AChE). Furthermore, Centrophenoxine (CpH) was administered as a post treatment to evaluate its potential in Al-induced neurotoxicity. The cognitive functions and memory loss were also studied after both Al and CpH administration. Al was administered orally at a dose of 40?mg?kg?1?day?1 for a period of 8 weeks, whereas CpH was administered intraperitoneally at a dose of 100?1?mg?kg?1?day?1 for a period of 6 weeks. The study was carried out in four regions of the brain, namely cerebrum, cerebellum, medulla oblongata, and hypothalamus. A significant reduction in AChE activity and different neurotransmitters was observed after Al exposure in the regions. CpH as a post treatment proved beneficial in restoring these alterations. Al exposure also affected the cognitive functions and short-term memory, which were significantly improved following CpH post treatment.  相似文献

The present study was carried out to observe the possible beneficial effects of Vitamin E, a natural antioxidant on methomyl-induced biochemical and histological alterations in rat liver. To carry out the investigations, animals were segregated in four different groups. Animals in Group I served as normal controls. Animals in Group II were given single methomyl dose orally in water (9 mg kg?1 b.wt). Animals in Group III were injected intraperitoneally with Vitamin E (50 mg kg?1 b.wt) for 1 week on alternate days. Animals in Group IV were administered Vitamin E 1 week before subjecting them to methomyl treatment. Animals in all the groups were sacrificed 24 h after the end of treatments. Different biochemical estimations were carried out, which included estimation of aspartate aminotransaminase (AST), alanine aminotransaminase (ALT), alkaline phosphatase (ALP) and acetylcholinesterase (AChE). Further, to examine the oxidative damage lipid peroxidation (LPO) and glutathione (GSH) levels as well as antioxidant enzymes such as superoxide dismutase (SOD), catalase, glutathione-S-transferase (GST), glutathione reductase (GR), glutathione peroxidase (GSHPx), and glutathione-6-phosphate dehydrogenase were estimated in liver samples. AchE activity was inhibited significantly both in serum and liver following methomyl treatment. Administration of methomyl caused a significant increase in serum AST, ALT and ALP which indicated hepatic damage. LPO was found to be significantly increased, whereas GSH levels were decreased in the liver of methomyl-treated animals. The activities of SOD and catalase were significantly decreased whereas GST and GSHPx activities were found to be elevated significantly following methomyl treatment. No significant change in the enzyme activity of GR and glutathione-6-phosphatase dehydrogenase was observed after methomyl treatment. Vitamin E supplementation was able to attenuate appreciably the methomyl-induced changes in LPO levels along with SOD and GST activities. Histopathological studies following methomyl treatment revealed that hepatocytes, were not very well delineated and nuclei showed degenerative changes. Whereas, following Vitamin E supplementation in combined treatment group nuclei showing degenerative changes become less in number. The study, therefore, concludes that Vitamin E has a potential in mitigating most of the adverse effects induced by methomyl acute toxicity.  相似文献

The effects of deltamethrin at acute and sublethal concentrations on some hematological and biochemical profiles in the Indian major carp Labeo rohita have been examined. The lethal concentrations (LC50) of deltamethrin for 24 and 96 h were found to be 0.44 and 0.38 mg L?1, respectively. During acute treatment (24 h), hemoglobin (Hb), hematocrit (Hct), red blood cell (RBC), white blood cell (WBC), mean cellular volume (MCV), mean cellular hemoglobin (MCH), and mean cellular hemoglobin concentration (MCHC) values were significantly increased compared to control group. Plasma glucose, protein, cholesterol, albumin, globulin, triglyceride, and glycogen in gill and muscle were significantly decreased. Upon sublethal treatment (0.044 mg L?1, 10th of 24 h LC50), RBC and the biochemical parameters, except glucose and cholesterol, decreased significantly, while MCV, MCH, and MCHC values increased up to the 28th day and then declined. Cholesterol level was significantly increased throughout the study period. A biphasic response in the levels of Hb, Hct, WBC, and glucose were observed during sublethal treatment.  相似文献

The absence of soy phosphatidylcholine (PC) in purified diets fed to juvenile lobstrs causes a significant decrease in the concentrations of total cholesterol and phospholipids in the serum. When egg PC, bovine PC, phosphatidylinositol, and ovine cephalin are used as substitutes for the soy PC, cholesterol and phospholipid levels in the serum are also significantly reduced. Cholesterol and phospholipid concentrations in the serum are highly correlated. Analysis of levels of hepatopancreatic cholesterol indicate that intestinal absorption of cholesterol is not impaired by the absence of dietary PC or by the presence of a phospholipid substitute. We suggest that the PC molecule is an important component of a lipoprotein complex that effectively transfers cholesterol, an essential nutrient for lobsters, from the hepatopancreas to the hemolymph.This work is a result of research sponsored in part by NOAA, Office of Sea Grant, Department of Commerce, under Grant No. 04-8-MO1-189 R/A 28. The levels of extractable aluminum (Al) in soils of tea plantations, Al concentrations in tea leaves and the impact of nitrogen fertilization on these two parameters were investigated. In addition, the properties of soils from tea plantations were compared to those from soils of adjacent non-tea fields to evaluate the effect of land use conversion (from non-tea soils to tea soils). Exchangeable Al (extracted in 1 mol l−1 KCl) ranged from 0.03 to 7.32 cmolc kg−1 in 94 tea fields and decreased rapidly with increasing soil pH. In comparison with non-tea soils, tea soils had a significantly lower pH and exchangeable Mg2+ concentration but higher organic matter contents and exchangeable K+ concentration. Contents of extractable Al were not different (P > 0.05) between these two soils. The concentrations of Al in mature tea leaves correlated significantly with exchangeable Al in soil samples taken at a depth of 20–40 cm and with exchangeable Al saturations in soil sampled at␣depths of 0–20 and 20–40 cm. In the pot experiment, nitrogen fertilization significantly increased extractable Al levels but decreased soil pH and the levels of exchangeable base cations. Nevertheless, the levels of Al in mature leaves and young shoots were significantly reduced by the application of large amounts of N fertilizer.  相似文献

Alloxan-induced diabetic rats were treated with multiple herbal preparation besides a control group receiving distilled water. The levels of glucose and alkaline phosphatase (ALP) increased abnormally in the alloxan treated group and the same were normalized upon treatment with the herbal preparation. The levels of blood urea nitrogen (BUN), alanine aminotransferase (ALT), protein and albumin in all groups remained unaltered. However, weekly body weight gain which got significantly altered in the alloxan-treated group was normalized by treatment with the herbal preparation. On the whole, a profound hypoglycemic effect was observed by the multiple herbal treatment in the diabetic rats.  相似文献

Coriandrum sativum (Coriander) has been documented as a traditional treatment for cholesterol and diabetes patients. In the present study, coriander seeds incorporated into diet and the effect of the administration of coriander seeds on the metabolism of lipids was studied in rats, fed with high fat diet and added cholesterol. The seeds had a significant hypolipidemic action. In the experimental group of rats (tissue) the level of total cholesterol and triglycerides increased significantly There was significant increase in beta-hydroxy, beta-methyl glutaryl CoA reductase and plasma lecithin cholesterol acyl transferase activity were noted in the experimental group. The level of low density lipoprotein (LDL) + very low density lipoprotein (VLDL) cholesterol decreased while that of high density lipoprotein (HDL) cholesterol increased in the experimental group compared to the control group. The increased activity of plasma LCAT enhanced degradation of cholesterol to fecal bile acids and neutral sterols appeared to account for its hypocholesterolemic effect.  相似文献

Feeding deltamethrin-contaminated grains to domestic poultry, such as quails may result in toxic effects in these birds. This study was done to investigate the effects of recommended doses of deltamethrin, sometimes used in grain storage silos, on Japanese quail (Coturnix japonica). Quails were fed grains contaminated with 0.25 and 0.50 mg deltamethrin per kg diet for 21 days and the effects on survival and blood biochemical parameters were studied. Plasma uric acid, creatinine levels, and creatinine phosphokinase activity in the blood were increased. Aspartate aminotransferase and lactate dehydrogenase activities and glucose levels significantly increased in birds treated with the high dose of deltamethrin. Alanine aminotransferase activity and albumin or cholesterol levels were not changed, and acetylcholinesterase and alkaline phosphatase activities, total protein and globulin in plasma were decreased. Administration of 0.25 mg/kg deltamethrin caused increased blood triglyceride levels, 0.50 mg/kg deltamethrin decreased triglyceride levels.  相似文献

This study reports the effects of low level developmental Pb exposure on specific brain regions like hippocampus, cerebellum and cerebral hemispheres of antioxidant enzyme activities. Wistar dams were exposed to 50 ppm, 100 ppm and 500 ppm of Pb acetate in drinking water during pregnancy and lactation (gestation day 6 through PND 21 (post natal day) and activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and glutathione reductase (GR) were determined in the hippocampus, cerebellum and cerebral hemispheres of pups during treatment period (PND 7, 14, and 21 days) and also during withdrawl period (PND 35, 45, 60 and 90 days). During treatment period, SOD activity significantly (p < 0.05) decreased in all regions of all the treated groups with maximum decrease in 500 ppm treated group of 21 days, while GSH-Px and GR activities increased with maximum increase in 21 days aged 500 ppm group. During withdrawl period, the activities of all enzymes were significantly (p < 0.05) reversed. Thus the perinatal exposure of dams to variable dosages of low level lead results in characteristic neurochemical alterations in rat brain regions due to impaired antioxidants function.  相似文献

Neutral lipid and phospholipid fractions and their component lipid classes in the digestive gland, abdominal muscle, epidermis and cuticle ofPenaeus esculentus Haswell were analysed and compared during the moulting cycle and during starvation. The prawns were collected from Moreton Bay, Queensland, Australia, by trawling during 1985–1987, and were fed on a standard, semi-purified diet. The digestive gland appears to be a major site of lipid synthesis, storage and mobilisation in preparation for moulting. Neutral lipid, 59 to 80% of which was triacylglycerol, was the larger fraction. It accumulated during early premoult, mainly due to the increase in triacylglycerol. The digestive gland contained only 18% of the total body lipid, or 8% of body lipid as triacylglycerol. Thus, the reserve lipid available for energy production is very small. Digestive gland triacylglycerol was markedly depleted after 4 d starvation and was almost completely absent after 8 d. In the other tissues, the major fraction was phospholipid, of which over 50% was phosphatidylcholine and up to 20% phoshatidylethanolamine; cholesterol was the major class in the neutral lipid fraction and appeared to be very stable. Most of this lipid was probably a component of cellular membranes. The lipid composition of muscle changed very little during the moulting cycle: total lipid levels in the epidermis were high in late premoult and early postmoult, when new cuticle is being secreted, but the proportions of the component lipids were closely similar. Cuticle lipid, together with other major components, was resorbed from the old cuticle prior to ecdysis, but the cuticle phospholipids appeared to be labile at all moult stages. The total of all lipids in fedP. esculentus was about 3.6% dry weight, of which about 70% was phospholipid. Earlier research had shown that when digestive gland lipid is exhausted after a short period of starvation, muscle is metabolised for energy. The present research showed that in the remaining muscle only about 13% of lipid was lost after 21 d starvation, mostly as phosphatidylcholine. This is in keeping with the need to maintain this tissue in a functional state. In contrast, epidermal lipid levels were markedly reduced after only 4 d starvation and the proportions of phospholipids changed significantly. This sensitivity of the cuticle lipids to starvation may be the cause of delayed moulting, which is characteristic of poor nutrition.  相似文献

Organotin compounds, widely used as antifouling agents, are known to bioaccumulate in the food chain. Among organotin compounds, tributyltin (TBT), a toxic and widespread contaminant, has become a serious factor in environmental pollution and is suspected of being immunotoxic in animals. The purpose of this study was to assess the effects of 80 µg of TBT per kilogram of body weight (kg bw), provided in food, on the immune and neurological systems of C57Bl/6 mice. Data showed that TBT increased the proliferation of T and B lymphocytes in both adult and juvenile female and juvenile male mice, but that it decreased the proliferation of both T and B lymphocytes in adult male mice. The macrophages activity in female and male juvenile mice was higher than in adults of both sexes. The natural killer cytotoxic activity was also increased in juvenile and adult males and females compared to the control groups. In the brain, we observed the presence of TBT in the hippocampus, the striatum, the cortex, and the cerebellum in both the male and female groups. The highest levels were observed in the cortex of females and males, while the lowest levels were found in the cerebellum. TBT also induced an increase in the levels of the antioxidant glutathione (GSH) in the striatum, the hippocampus and in cortical structures but not in the cerebellum where the levels of TBT are lower. Our findings indicate that TBT modulated the immune and nervous systems causing endocrine and nervous perturbations.  相似文献

Free radical production and oxidative stress are known to increase in liver during diabetes and may contribute to oxidative damage. In this study, the effect of oral feeding of Trigonella foenum-graecum seed powder (TSP) has been studied on blood glucose, membrane-linked ATPases, antioxidant enzymes, lipid peroxidation, lipofuscin content and membrane fluidity in livers of a diabetic rat. Diabetes was induced by administration of alloxan monohydrate. Hyperglycemia during diabetes has been shown to entail free radical generation, with a concomitant increase in lipid peroxidation and lipofuscin content, inhibition of membrane-linked ATPases, increased antioxidant enzyme activities and decreased membrane fluidity. TSP treatment reverses the changes to near normal levels thereby ameliorating the management of diabetic complications.  相似文献

The pesticides are one of the most potentially harmful chemicals liberated in the environment in an unplanned manner Malathion is widely used as a potent pesticide in many countries and has been shown to produce some adverse health effects. A study was conducted to asses the effects of malathion on the male reproductive system of wistar rats. The pesticide was administered to rats orally at dose levels of 50, 150 and 250 mg/kg/body wt/day for 60 days. In comparison to the control rats, there was a significant reduction in the weight of testes, epididymis, seminal vesicle and ventral prostate. Testicular and epididymal sperm density were decreased in the animals treated with malathion. Pre and post fertility test showed 80% negative results after treatment Biochemical profile of the testis revealed a significant decline in the contents of sialic acid and glycogen. Whereas a significant increase in the protein content of testis and testicular cholesterol was observed. The activity of testicular enzyme acid phosphatase increased significantly while decreased alkaline phosphatase activity was found. Malathion also suppressed the level of testosterone significantly Results of the present study clearly suggest that malathion induce toxic effects on the male reproductive system of rats.  相似文献

The molecular basis of male reproduction for cross-regulation between androgen and thyroid hormone axes is still rudimentary. This study aims to define a possible mechanism of hypothyroidism-induced reproductive influence with respect to sex hormone, mineral, sperm motility, oxidative stress, c-Fos expression, cell cycle, and apoptosis in rat testes. The Wistar rats were randomly divided into control group (NS) and hypothyroidism group [1 ml/100g BW/day, 0.1% propylthiouracil (PTU)] by intragastric gavage for 60 days. Blood samples were collected to measure the serum levels. The epididymis was excised to measure sperm motility and testes were excised to measure mineral, oxidative stress, c-Fos expression, cell cycle, and apoptosis. After 60 days, body weight, relative testes weight, triiodothyronine, and total thyroxine were all significantly decreased, whereas thyroid stimulating hormone was increased in the hypothyroidism group. A significant increase in sex hormone level of estradiol (E2) and significant decreases in testosterone (T) and T/E2 ratio were observed following PTU treatment. And sperm quality was also significantly changed. There were significant decreases in the contents of calcium (Ca2+) and zinc (Zn2+). On the other hand, malondialdehyde and hydrogen peroxide contents significantly increased, whereas the activity of superoxide dismutase, catalase and nitric oxide synthase and nitric oxide content significantly decreased in hypothyroid rats. The mRNA and protein expressions of c-Fos decreased significantly. The cell percentage in G0/G1 phase increased significantly, whereas decreased significantly in S and G2/M phases. Also, a significant increase in testicular cell apoptosis was observed in hypothyroid-treated rats. These results suggested that hypothyroidism could affect reproductive function in the form of changed sex hormone levels, sperm motility and testicular Ca2+ and Zn2+, and enhanced oxidative stress leading to c-Fos abnormal expression and increased apoptosis.  相似文献

Cypermethrin and chlorpyrifos pesticide- (5, 15 and 30 ppm) induced effects on growth, metabolic and antioxidative behaviour of Azolla pinnata were analysed after 96 h of treatment. Growth of A. pinnata was increased by 72–76% over the initial mass under control condition. Further, the growth was decreased by 26 and 30% following treatment with 5 ppm of cypermethrin and chlorpyrifos, respectively, compared to control. Whereas, 30 ppm of pesticide treatment caused substantial decrease in biomass accumulation, even less than the initial mass. The Chlorophyll a content was declined at higher concentrations of chlorpyrifos, while carotenoids content increased significantly in the presence of both the pesticides. Photosynthetic rate showed declining trend with rising concentration of pesticides, while an increasing trend was noticed in the case of respiratory O2 consumption. The oxidative biomarkers malondialdehyde, superoxide radicals and hydrogen peroxide increased with increasing concentration of both the pesticides. The activity of enzymatic antioxidants enzyme superoxide dismutase, glutathione-S-transferase and ascorbate peroxidase (APX) was enhanced with increasing concentrations of both the pesticides; however, a declining trend was noticed in the case of APX at 30 ppm chlorpyrifos treatment. The study concludes that chlorpyrifos appeared to be more toxic than cypermethrin; however, both the pesticides significantly reduced the growth performance.  相似文献

以鸡脾脏淋巴细胞为试验对象,通过短期体外培养的方法研究了铝暴露对鸡免疫细胞的毒性作用.将不同浓度的AlCl3添加到培养液中(终浓度分别为0、0.2、0.4、0.6、0.8mg·mL-1),对鸡脾淋巴细胞原代培养24h,检测培养液上清中乳酸脱氢酶(LDH)活性,淋巴细胞增殖、白细胞介素-2(IL-2)和肿瘤坏死因子-α(TNF-α)含量.结果表明,与对照组相比,各暴露组培养液上清中LDH活性均显著升高(p<0.05,p<0.01),且随铝暴露浓度的增加,LDH活性逐渐升高,呈明显的剂量-反应关系,表明铝暴露能够破坏鸡脾淋巴细胞膜,导致细胞活性下降;与对照组相比,各暴露组淋巴细胞增殖能力、培养液上清中IL-2和TNF-α含量均显著降低(p<0.05,p<0.01),且随铝暴露浓度的增加,3者均逐渐降低,呈明显的剂量-反应关系,表明铝对体外培养鸡脾淋巴细胞具有一定的免疫毒性作用。  相似文献
