The kinetic parameters and the purification rates of the hybrid cell Foaz in soybean processing wastewater (SPW) were measured through a shaking reaction and in two automatic regulated control stable fermentation systems (ARCSFS). The maximum specific growth rate of Foaz was 0.576 h‐1, higher than that of one of its parental strains Rhodobacter sphaeroides P9479 and lower than that of another parental strain Saccharomyces cerevisiae Y9407. The BOD5 removal rate of Foaz in the No. 1 system was 61.3%, higher than those of both its parental strains when the influent BOD5 concentration was 4600 mg/L. The results of this study suggest that the hybrid Foaz has a better capacity of the degradation of organic pollutants in SPW than its parental strains and it may be applicable to the treatment of high concentration organic wastewater.  相似文献

为探明铀矿山对周围环境的联合毒性机制,本研究以常用生物毒性测试菌种——发光细菌青海弧菌Q67和费氏弧菌以及禾花鲤为受试生物代表,实地采集的铀矿山废水为目标废水,研究铀矿山废水对发光细菌和禾花鲤的急性毒性。实验结果表明,矿山废水对3种生物的急性毒性存在显著的剂量-效应关系,毒性效应浓度EC_(50)(LC50)的大小顺序为禾花鲤幼鱼费氏弧菌青海弧菌Q67,Pb~(2+)/U~(6+)浓度分别为6.052/3.026 mg·L~(-1)、2.284/1.142 mg·L~(-1)、1.339/0.669 mg·L~(-1),均可有效指示矿山废水的毒性水平,其中发光细菌更为灵敏、快速;且青海弧菌Q67的EC_(50)值最小,灵敏度最高,可作为表征矿山废水毒性风险的首选指示物。研究结果能够为放射性矿区废水生态风险预警、安全处理处置、水质基准制定及流域水环境管理提供依据。  相似文献

为了阐明不同补给水源的城市人工湖中异养菌耐药状况和耐药菌种属分布特征,选取分别以地表水和再生水为补水来源的XQ湖和FQ湖为代表进行研究。从2018年4—11月逐月采集水样,考察了各水样中对氨苄西林(AMP)、磺胺甲恶唑(SMZ)和四环素(TET)这3种不同种类抗生素具有耐药性的异养菌含量,并对分离菌株的耐药表型、耐药菌株的种属分布以及水质理化指标进行了分析。结果表明,2处人工湖中的AMP耐药菌和SMZ耐药菌含量约为10~2~10~4CFU·(100 m L)~(-1),而TET耐药菌含量则约10~1~10~3CFU·(100 m L)~(-1)。分离出的84株耐药菌归属于19个种,其中蜡状芽孢杆菌、大肠埃希氏菌、维氏气单胞菌、豚鼠气单胞菌和鸟氨酸拉乌尔菌为2个湖共有耐药菌。71.4%的耐药菌都是对AMP单一耐药,以蜡状芽孢杆菌为主。由于具有固有耐药性的细菌在分离出的耐药菌中占比很低,获得性耐药很可能在城市人工湖中异养菌耐药性的发展上发挥了主要贡献作用。地表水补水的XQ湖和再生水补水的FQ湖在总异养菌含量、耐药菌含量和检出率上均无显著差异。分离来源对16株耐药性气单胞菌的聚类无明显影响。  相似文献

抗生素废水含有大量的抗生素耐药菌(antibiotic resistant bacteria,ARB)与抗性基因(antibiotic resistance genes,ARGs),处理排放后可能增强受纳环境的微生物抗性,因此有必要深入研究抗生素废水处理过程中ARB与ARGs的削减效果及其影响因素。本研究采用膜生物反应器(membrane bioreactor,MBR)工艺处理螺旋霉素制药废水,考察了不同水力停留时间(hydraulic retention time,HRT)对ARB与ARGs削减效果的影响。结果表明,虽然在HRT=30 h时MBR对COD与氨氮的去除率略高于HRT=40 h,但HRT=40 h时,不仅总异养菌与肠球菌的去除效果更佳,出水肠球菌耐药率及携带的抗性基因检出率也更低,而且MBR对废水中erm B、erm F、erm X、mef A、ere A、mph B和转移元件ISCR 1、Tn 916/1545相对丰度的削减效果更好。这表明长HRT更有利于MBR工艺削减螺旋霉素废水的耐药菌与抗性基因。  相似文献

The occurrence of antibiotic-resistant bacteria and antibiotic resistance genes (ARGs) has been intensively investigated for wastewater treatment systems treating single class of antibiotic in recent years. However, the impacts of alternately occurring antibiotics in antibiotic production wastewater on the behavior of ARGs in biological treatment systems were not well understood yet. Herein, techniques including high-capacity quantitative PCR and quantitative PCR (qPCR) were used to investigate the behavior of ARGs in an anaerobic–aerobic full-scale system. The system alternately treated three kinds of antibiotic production wastewater including ribostamycin, spiramycin and paromomycin, which referred to stages 1, 2 and 3. The aminoglycoside ARGs (52.1–79.3%) determined using high-capacity quantitative PCR were the most abundant species in all sludge samples of the three stages. The total relative abundances of macrolide–lincosamide–streptogramin (MLS) resistance genes and aminoglycoside resistance genes measured using qPCR were significantly higher (P < 0.05) in aerobic sludge than in sewage sludge. However, the comparison of ARGs acquired from three alternate stages revealed that MLS genes and the aminoglycoside ARGs did not vary significantly (P > 0.05) in both aerobic and anaerobic sludge samples. In aerobic sludge, one acetyltransferase gene (aacA4) and the other three nucleotidyltransferase genes (aadB, aadA and aadE) exhibited positive correlations with intI1 (r 2 = 0.83–0.94; P < 0.05), implying the significance of horizontal transfer in their proliferation. These results and facts will be helpful to understand the abundance and distribution of ARGs from antibiotic production wastewater treatment systems.

The present study investigated the influence of 241Am on microbial growth and the degradation of organic waste. Leachate samples collected in a lysimeter were periodically analyzed for bacterial growth, under both aerobic and anaerobic conditions. 241Am inhibited bacterial growth, and the degradation of organic matter was delayed in comparison with the control. Minimal inhibitory concentration assays and survival curves revealed that it inhibits the growth of Pseudomonas putida F1. The assay also revealed that 241Am is more toxic than 238U, Zn2+ and Cd2+. This study further led to the finding of four new radionuclide-tolerant bacterial strains: Flavobacterium spp., Pseudomonas gladioli, Chryseobacterium indologenes and Ochrobactrum anthropi. The survival curves of P. gladioli, C. indologenes revealed that these bacteria are resistant to metal as consortia.  相似文献

A potential method to detoxify pesticides in aquatic system is using bioremediation. In this study, four microorganisms (Pseudomonas sp (EB11), Streptomyces sp. (EB12), Aspergillus niger (EB13) and Trichoderma viride (EB14) were isolated from cucumber leaves previously treated with cymoxanil using enrichment technique. These strains were evaluated for their potential to detoxify cymoxanil in aquatic system at the concentration level of 5×10?4M. The effect of pH and temperature on the growth ability of the tested strains was also investigated by measuring the intracellular protein and mycelia dry weight for bacterial and fungal strains, respectively. Moreover, the remaining toxicity of cymoxanil after 28 days of incubation with tested strains was evaluated to confirm the complete removal of any toxic materials (cymoxanil and its metabolites). The results showed that the optimum pH for the growth of cymoxanil degrading strains (bacteria and fungi) was 7. A temperature of 30°C appears to be the optimum for the growth of either fungal or bacterial strains. Pseudomonas sp. (EB11) was the most effective strain in cymoxanil degradation followed Streptomyces sp (EB12), Trichoderma viride (EB14) and Aspergillus niger (EB13), with half-lives of 4.33, 9.5, 17.3 and 24.7 days, respectively. The degradation of cymoxanil by bacterial strains was much faster than fungal one. There is no remaining toxicity of cymoxanil detected in aqueous media previously treated with Pseudomonas sp. (EB11) for 28 days. The results suggest that bioremediation by Pseudomonas sp. (EB11) are promising for the detoxification of cymoxanil in aqueous media.  相似文献

Following enrichment in its presence, two strains of bacteria, isolated from marine sediments, were shown to degrade the quaternary ammonium surfactant benzyldimethyl hexadecylammonium chloride (BDHAC) in a minimal salts medium. The bacteria identified by 16S ribosomal deoxyribonucleic acid sequencing were shown to belong to several genera and determined to be Bacillus niabensis and Thalassospira sp. Initial investigations demonstrated that the bacteria were capable of degrading BDHAC when it was present at concentrations in the range 2–4 mg mL?1. In media containing BDHAC, up to 90% was degraded within 7 days, but limited growth of the strain was observed at 2 and 4 mg mL?1 BDHAC. Preliminary analysis of samples after degradation experiment by electrospray ionization mass spectrometry/mass spectrometry produced a peak with a parent–daughter ion transition of 136 → 91, corresponding to N,N-dimethylbenzylamine. The presence of this potential metabolite suggests the cleavage of the C-alkyl-N bond as a step in BDHAC catabolism.  相似文献

The uptake of several species of bacteria by the common mussel Mytilus edulis (L.) and the subsequent fate of some polymers of the bacteria have been investigated in a study carried out during 1981. Bacteria (Escherichia coli, Micrococcus luteus, M. roseus, Bacillus cereus, Staphylococcus aureus and a marine pseudomonad, 1-1-1) were radiolabelled by growth in medium containing 3H-thymidine and the uptake of bacteria by Mytilus edulis was monitored. Labelled and unlabelled bacteria, at initial concentrations of 0.5 to 1x107 bacteria ml-1, were cleared at similar, exponential rates with no significant difference in the rates for different bacteria: 90% of bacteria were cleared in a mean time of 1.93±0.12 h (SEM, n=63). Those bacteria with cell walls which were sensitive to M. edulis lysozyme were rapidly degraded by the mussel and 3H-labelled DNA was released in a form not precipitable by 10% trichloroacetic acid. Lysozyme-resistant bacteria (Micrococcus roseus and S. aureus) were cleared from suspension by Mytilus edulis but most were rejected intact. By measuring the rate of release of 3H-thymidine-labelled material from the mussel the rate of degradation of lysozyme-sensitive bacteria by M. edulis was found. For different bacteria the degradation rate varied from approx 2x108 to 27x108 bacteria h-1 with an overall mean of 10x108 bacteria h-1. A thymidine- and diaminopimelicacid-requiring auxotroph of E. coli was radiolabelled with 3H-thymidine, 3H-diaminopimelic acid or 14C-glucose and fed to M. edulis. Bacteria were cleared and degraded by the mussel; 3H-diaminopimelic acid-labelled or 14C-glucose-labelled polymers were retained, whereas 3H-thymidine-labelled polymers were released into the surrounding water. Extracts of the digestive gland of M. edulis degraded lysozyme-sensitive bacteria to release 3H-thymidine-labelled material, but did not release 3H-thymidine-labelled material from lysozyme-resistant bacteria. It is concluded that M. edulis can select lysozymesensitive bacteria for subsequent processing and discriminate between bacterial polymers to reject DNA. Also, bacteria could provide a substantial fraction of the carbon requirement of the mussel.  相似文献

Kinetics of glucose and amino acid uptake by attached and free-living bacteria were compared in the upper 70 m of the oligotrophic north-western Mediterranean Sea. Potential uptake rates of amino acids were higher than those of glucose in all the samples analysed. Cell-specific potential uptake rates of attached bacteria were up to two orders of magnitude higher than those of total bacteria, both for amino acids and glucose (0.72–153 amol amino acids cell−1 h−1 and 0.05–58.42 amol glucose cell−1 h−1 for attached bacteria and 0.34–1.37 amol amino acids cell−1 h−1 and 0.07–0.22 amol glucose cell−1 h−1 for total bacteria). The apparent K m values were also higher in attached bacteria than in total bacteria, both for amino acids and glucose. These results would reflect the presence of different uptake systems in attached and free-living bacteria, which is in accordance with the different nutrient characteristics of their microenvironments, ambient water and particles. Attached bacteria show transport systems with low affinity, which characterise a bacterial community adapted to high concentration of substrates. Marine bdellovibrios have not previously been reported from the southern hemisphere, and knowledge of their occurrence in marine ecosystems is rudimentary. This study examined quantitative and qualitative aspects of bdellovibrios parasitic to the bacterium Vibrio alginolyticus at each of three representative tropical marine habitats of the Great Barrier Reef. Bdellovibrios were found in the water column throughout a 12 mo period from May 1992 at a sandy beach, a mangrove and a fringing coral reef. Their abundance was correlated with water temperature (P<0.001) and was highest in summer, lowest in winter and intermediate in spring and autumn. Over the sampling period, bdellovibrios were most abundant at the mangrove habitat (36.6 ml-1) and least abundant at the reef (9.5 ml-1), but there was substantial variability in numbers at all habitats among seasons and months of the year. On some occasions no bdellovibrios were found in replicate samples from the beach and reef habitats, while on others the maximum detectable by the method used (180 ml-1) was sometimes found at the beach and mangrove habitats. Bdellovibrios within each habitat were uniformly distributed among sampling sites (P>0.05). They were more abundant in sub-surface than bottom waters in summer, but the reverse occurred in winter. Midwater samples usually had least bdellovibrios. Bdellovibrio numbers were significantly correlated with those of potential host bacteria—colony-forming bacteria at all habitats and total bacteria at the beach and reef habitats. Strain characteristics, primarily based on host range, indicated qualitative differences in bdellovibrio populations among habitats. Pseudomonas atlantica, P. aeruginosa, P. marina, Cytophaga marinoflava, Vibrio gazogenes, V. mimicus and a Spirillum-like bacterium were not parasitised by bdellovibrios from any habitat. Of the other 25 Vibrio spp. tested, most were parasitised by the majority of bdellovibrio strains from each habitat. Strain differences were principally with respect to parasitism of non-Vibrio bacteria. All strains required Na+ and grew at 35°C, but some failed to grow at 15°C.  相似文献

Biodegradation experiments of various polycyclic aromatic hydrocarbons were studied with mixed bacteria culture under aerobic conditions. An easy‐to‐handle clean‐up procedure was developed for PAH and their metabolites simultaneously as well as a gc‐ms‐method to identify and quantify these compounds.

Anthracene and dibenzothiophene are completely degradable in an aqeous system, whereas biodegradation of benzo(k)fluoranthene and benzo(h)quinoline is possible only in an oil‐in‐water‐system with dodecane as cosubstrate. No degradation of nitronaphthalene was observed in aqueous systems. New metabolites are 2,3‐dihydroxybenzothiophene, hydroxybenzothiophenecarbonic acid and benzothiophenequinone for dibenzothiophene and hydroxyfluoranthenic acid for benzo(k)flouranthene. Whereas the former metabolites are degradable under the experimental conditions, the latter accumulates during the degradation experiment.

The results are important for microbiological wastewater treatment, since knowledge of biodegradation processes is indespensable for the successful treatment of PAH‐containing wastewater.  相似文献

Feeding experiments were carried out on the harpacticoid copepods Tisbe holothuriae and Paramphiascella vararensis, using various species of marine and brackish water bacteria as food. Short-term experiments with 3H-labelled bacteria are described, as well as experiments covering longer periods of time using non-labelled, dried bacteria. Results show that for both T. holothuriae and P. vararensis, the amount of dry bacteria consumed ranged from 2.06 to 7.07 g copepod-1 day-1, indicating a carbon requirement of 1 to 3.5 g copepod-1 day-1. The food value of the bacteria strains tested is given in terms of the generation time of the copepods raised on these bacteria.  相似文献

ABSTRACT

Two microalgal strains (Chlorella sorokiniana, A&B,) grown optimally at pH 9 (A) and 7 (B) were isolated from a municipal wastewater treatment plant. These strains were used to check their efficacy for nutrients and organic carbon removal capacities from the tertiary wastewater. The strains were characterised using 18S rDNA sequencing method and both the strains closely related to Chlorella sorokiniana. Different inoculum doses (IDs) of both the strains were tried to check their efficacy on wastewater treatment. Due to the increase in the IDs, hydraulic retention time decreased from 6 d to 1?d. On an average, 12–100% of total kjeldahl nitrogen, 53–96% NO3–N, and 59–92% PO4–P, were removed. Despite the same growth environment, strain A with the highest ID (700?mg/L) showed the best performance in terms of overall organic carbon removal, nutrient removal, and value-added products recovery. Strain A showed an increase in carbohydrate and protein content by 42% and 13%, respectively. Both the strains showed luxury phosphorus uptake and found suitable for advanced wastewater treatment.  相似文献

The fate of the benthic bacterial biomass is a topic of major importance in understanding how soft-bottom environments function. Because of their high abundance, production and nutritional value, benthic bacteria may constitute an important food resource for benthic fauna. The trophic role of bacteria for a nematode community on the Brouage mudflat (Marennes-Oléron-France), dominated by three species: Chromadora macrolaima (64% of the abundance), Daptonema oxycerca (15%) and Ptycholaimellus jacobi (8%), was determined in grazing experiments using 15N pre-enriched bacteria. On intertidal flats, seasonal, tidal and circadian cycles induce strong variations in environmental conditions. Grazing experiments were performed in order to measure the effects of abiotic (temperature, salinity and luminosity) and biotic (bacterial and algal abundances) factors on assimilation rates of bacteria by nematodes. In order to assess simultaneously bacteria and algal assimilation rates, algal abundances were modified adding 13C pre-enriched Navicula phyllepta. Assimilation rate was significantly lower at 5°C; moreover, general trend shows a prominent temperature effect with an optimum around 30°C. Assimilation at salinity 18 was not significantly different from the assimilation at salinity 31. Assimilation was higher under light conditions than in the dark. Above 109 bacteria ml−1, assimilation of bacteria remained unaffected by bacterial abundance. However, assimilation of algae increased with the algal concentration. Nematode kept feeding under conditions of stress, which are typical of the surficial sediment habitat and they appeared to be principally dependent on the algal resource.  相似文献

Abstract

A metal-organic framework of iron-doped copper 1,4-benzenedicarboxylate was synthesized and, for the first time, utilized as a heterogeneous photo-Fenton catalyst for degradation of methylene blue dye in aqueous solution under visible light irradiation. The synthesized materials were characterized by scanning electron microscopy, Fourier-transform infrared spectroscopy, X-ray diffraction and energy-dispersive X-ray spectroscopy. The influence factors, kinetics, and stability of the synthesized catalysts were investigated in detail. Iron-doped copper 1,4-benzenedicarboxylate showed higher degradation efficiency than pure copper 1,4-benzenedicarboxylate. An almost complete degradation was achieved within 70?min under visible light irradiation at a solution pH of 6, a catalyst loading of 1?g?L?1, a H2O2 dosage of 0.05?mol L?1 and methylene blue concentration of 50?mg?L?1. Recycling studies demonstrated that the iron-doped copper 1,4-benzenedicarboxylate is a promising heterogeneous photo-Fenton catalyst for long-term removal of methylene blue dye from industrial wastewater.  相似文献

The optimal parameters for the degradation of pure terephthalic acid (PTA) wastewater by the white rot fungus Phanerochaete chrysosporium were investigated. The relative mathematical model, objective function equation, constraint equations, constraint conditions and computer program were established in the optimal method. The results showed that the optimal value of the reactor volume needed was 9798 m3. It could save 83.82% of the reactor volume, which was needed for the native bacterium YZ1. This optimal method is simple, useful and economical for average treatment plants. Using the optimal parameters and P. chrysosporium to design and operate the treatment process of PTA wastewater would increase the treatment efficiency and decrease the cost.  相似文献

The culturability of heterotrophic marine bacteria obtained from the surfaces of two species of marine algae (Lobophora variegata andHalimeda copiosa) was assessed by comparing total DAPI-stained cell counts to colony-forming bacterial counts on two agar media. The colony-forming bacterial counts on a low-nutrient medium (LN) consisting of seawater and agar were significantly greater for both algal species than counts obtained on a high-nutrient medium (HN) similar in composition to that typically used for the isolation of heterotrophic marine bacteria. On average, 14 and 58%, respectively, of the total bacteria fromL. variegata andH. copiosa were culturable on LN. These recovery rates far exceed those typically reported for marine bacteria. Of 119 LN strains obtained in pure culture, 55% failed to grow on HN. The yeast extract component of HN (1.5 gl-1) was responsible for the majority of the observed inhibition, suggesting that this nutrient can be highly toxic to marine bacteria. Eighty-nine percent of the strains inhibited by HN were capable of growth when the nutrients in this medium were diluted by a factor of 100 with seawater. Of 66 epiphytic strains, 30 (45%) initially inhibited by HN showed the ability to adapt to this medium after a period of laboratory handling. The initial inability of low-nutrient-adapted bacteria to grow on high-nutrient media may be due to nutrient shock. The results presented here indicate that the culturability of specific populations of marine bacteria can be dramatically improved by the use of low-nutrient media. Further, the importance of developing new medium formulations that eliminate traditional nutrients, some of which are clearly toxic to bacteria, is demonstrated.  相似文献

The bacterium Serratia marsescens strain JAS16 was isolated from agricultural soil which had prior exposure to monocrotophos for three years. The strain JAS16 tolerated up to 1200 mg L–1 monocrotophos and degraded the insecticide (1000 mg L–1) at a degradation rate constant of 136 d?1 (DT50 = 3.7 d). In soil, the degradation rate constant was 105 d?1 (DT50 = 4.8 d). A schematic pathway is being proposed from the degraded products derived from gas chromatography--mass spectrometry (GC-MS). The phytotoxicity of degradation products to Vigna radiata, Vigna unguiculata, and Macrotyloma uniflorum and the genotoxicity to Allium cepa roots were found to be low. A cost-effective powder-based formulation was achieved with the isolate. The isolate remained viable during the storage and also multiplied with a higher colony forming units (CFU) load g–1 for over a period of seven weeks of storage.  相似文献

Freshly collected faecal pellets of the common intertidal isopodIdotea granulosa Rathke, 1843, were exposed to natural communities of bacteria + protozoa, and bacteria + protozoa + metazoa for 16 d in running North Sea nearshore water during September 1989. All treatments showed quite identical changes in chlorophylla, phaeopigments, carbon (C) to nitrogen (N) ratio, heterotrophic bacteria and protozoa. Plant pigment, C and N content of faeces decreased by about half in 94 h. Interestingly, although the C/N ratio changed considerably during the study, the values at the end of 16 d were similar to ones at the beginning, suggesting that the C/N value per se of particles in the sea may not reflect their age or nutritive value to consumers. Further, dry weight of pellets which remained unchanged in appearance, decreased by half in 4 d and by two thirds in 16 d, indicating loss of substance but not bulk. The development and abundance of free bacteria in the water closely matched that in pellets, supporting the idea that faeces export bacteria or stimulate bacterial growth in the water through release of dissolved nutrients. There were 103 times more bacteria on faeces than free in the water of the same volume. However, given the relative volumes of the pellet (ca 0.1 mm3) and the free water in the microcosms (ca 107 × pellet volume), the degradation of faecal matter may support the production of the bulk of free-living bacteria in water. When organisms of larger sizes were allowed access to the pellets, several metazoans (e.g. nematodes ca 4 per pellet) were observed, but did not significantly accelerate faeces degradation.
