A series of mesocosms was exposed to a suite of light treatments and nutrient enrichment in order to generate algal communities of varying biomass. the influence of this biomass on the speciation of copper (II) was studied. Distribution coefficients (Kd,Lkg-1) were relatively high (logKd = 5 to 7), indicative of robust trace metal sequestration, and were likely controlled by the particulate organic carbon content (foc). Differences in Kd over time and among treatments were significant, as was the relationship between Kd and foc. Fluorescence quenching was used to determine binding capacities (Lt, M) and their associated binding constants (Kcond,M-1) in order to model the solid phase copper speciation. the Kcond ranged between 2.1 and 5.2 × 1012M-1, indicating a very strong copper-ligand complex, and was higher in mesocosms that received more light. the light Lt increased over time, dramatically after the nutrient enrichment, but did not vary systematically among light treatments. Lt ranged from 7.2 × 10- 7 to 4.9 × 10- 5 M. the large magnitudes of Kd, Kcond and Lt ensured that greater than 97% of total copper in the mesocosms was complexed by organic matter. the total copper concentration ([Cu]T, M) needed to reach a target dissolved copper concentration of 10-12.5 M (pCu = 12.5) was determined for each mesocosm over time. [Cu]T was between 8.02 × 10-5 and 3.41 × 10-2 M, and increased over time. the [Cu]T normalized to the target pCu (Effective Dose Ratio, EDR) increased directly with increases in algal biomass, indicating a direct link between system productivity and copper exposure. Approximately 45% of the variance in EDR was explained by variance in total biomass, while the residual variance in EDR was due likely to differences in the strengths of particle associations and magnitude of binding capacities.  相似文献

Algal populations, either suspended in the water column (planktonic) or present on the walls of an enclosure (periphytic), develop differently depending upon the physical scale of the system. This study determined whether these variations altered the speciation and therefore exposure of polychlorinated biphenyls (PCBs) in estuarine mesocosms. Exposure was defined as the fraction of applied contaminant taken up by plankton after two hours. Using a three phase equilibrium model, the partitioning of a suite of PCBs within variously-sized mesocosms was predicted using laboratory derived distribution coefficients and measured levels of planktonic and periphytic algal biomass. in mesocosms having large wall surface area to volume ratios, sorption of hydrophobic PCBs to periphyton significantly decreased contaminant exposure. However, within the range of planktonic algal biomasses observed in this study, the regulation of PCB exposure was relatively invariant between variously-sized mesocosms. to minimize sorption of hydrophobic organic contaminants (HOCs) to periphyton and reduce artefacts inherent with this partitioning, we suggest using mesocosms with low wall surface area to volume ratios (less than or equal to 1). in addition, periphytic biomass should be quantified regularly and a three-phase equilibrium approach used to predict the actual exposure concentrations.  相似文献

The effects of cement consolidated phosphogypsum (PG) on marine organisms was investigated under natural conditions in four 1000 m2 estuarine ponds. Two ponds were seeded with 160 kg of PG arranged in aggregations of blocks and two ponds received similar mass of sand/cement blocks. Meiofauna were sampled quarterly and PG did not affect total meiofauna or major taxa (nematodes and copepods) density. Abundant species of copepods either were slightly increased in ponds with PG or were inconsistently affected.

All ponds were drained after one year. Three species of macroinvertebrates and 15 species of fishes were collected. Diversity indices showed modest but inconsistent variation among ponds. Only Pond 1 (control) and Pond 4 (experimental) had similar species abundances and all ponds showed unique distributions of biomasses among species. Thus, no differences in community structure attributable to the presence of PG could be detected among benthic invertebrates, natant invertebrates, or fishes.  相似文献

通过碱性单细胞凝胶电泳法研究了Cu2+暴露剂量对赤子爱胜蚓(Eiseniafoetida)活体基因损伤的动态影响.结果显示:不添加Cu2+的对照组和添加Cu2+的处理组蚯蚓体腔细胞尾部DNA含量和尾长均呈非正态分布(p<0.05);在暴露72h时,125mg·L-1Cu2+处理组尾部DNA含量值最大,为41.44%,100mg·L-1Cu2+处理组尾长值最大,为33.79μm;随着Cu2+暴露剂量的增加,尾部DNA含量和尾长损伤频率增加;对照组和处理组的尾部DNA含量和尾长之间均存在显著性差异(p<0.05),Spearman非参数相关分析表明,尾部DNA百分含量和尾长之间呈显著相关(p<0.01,n=21),Cu2+暴露浓度与尾部DNA百分含量、尾长具有良好的剂量-构效关系(p<0.01).在125mg·L-1Cu2+浓度下暴露72h时蚯蚓的基因损伤程度达到最大,损伤程度为3级.可见,蚯蚓DNA生物标志物是重金属污染基因毒理诊断的重要指标,碱性SCGE试验是检测Cu2+暴露对赤子爱胜蚓活体基因损伤的有效手段.  相似文献

In sludge incineration, the thermal behavior of heavy metal is a growing concern. In this work, the combined analysis of metal partitioning behavior between vapor phase and condensed phase, speciation redistribu- tion in condensed phase and the difference of metal species in binding energy was carried out to investigate the possible volatilization-condensation mechanism of heavy metals in high-temperature sludge incineration. It was found that there were two steps in metal volatilization. The initial volatilization of heavy metal originated from their exchangeable (EXC), carbonate bound (CAR) and iron- manganese bound (FM) fractions, which is primarily composed of simple substance, chlorides, oxides and sulfides. With the increase of chlorine and sulfur in sludge, the inner speciation redistribution of heavy metals occurred in condensed phase, which was an important factor affecting the potential volatility of heavy metals. A partial of metal species with complexed (COM) and residual (RES) fractions gradually decomposed into simple substance or ions, oxides and carbonates, which signifi- cantly strengthened the second volatility. In presence of chlorine, about 46% of Cd with the RES fraction disappeared when the volatility rate of Cd increased by 44.89%. Moreover, about 9% of Pb with COM fraction disappeared when there was an increase of nearly 10% in the volatilization rate. Thus, the second volatilization was mainly controlled by the decomposition of metal species with COM and RES fractions. By virtue of XRD analysis and the binding energy calculation, it was found that metal complex and silicates were inclined to decompose under high temperature due to poor thermo stability as compared with sulfates.  相似文献

The interactions of 10 different chromium(III) complexes with isolated calf thymus DNA have been analysed by studying the electronic and fluoresence spectra of intercalated ethidiumbromide. Triply charged cationic complexes including: [Cr(urea)6]Cl3.3H2O, [Cr(1,10‐phenanthroline)3](ClO4)3.2H2O, [Cr(2,2'‐bipyridyl)3] (ClO4)3.2H2O, [Cr(ethylendiamine)3]Cl3.3.5H2O and [Cr(NH3)6](NO3)3 displaced the dye from DNA. Similar effects were observed in experiments using the non‐intercalating dye bisbenzimidazole ("Hoechst 33258"). However, singly charged cationic, anionic and uncharged chromium(III) complexes such as: cis‐[Cr(1,10‐phenanthroline)2Cl2]Cl.2H2O, cis‐[Cr(2,2'‐bipyridyl)2Cl2]Cl.2H2O, [Cr(glutathione)2]Na2, [Cr(cysteine)2]Na.2H2O and [Cr(glycine)3] were unable to displace both ethidiumbromide and bisbenzimidazole from DNA. There was no evidence for the formation of co‐ordinate bonds between chromium(III) and DNA for any of the above complexes. The charge and type of ligand are important in controlling the interaction of chromium(III) with isolated DNA in vitro. Our findings indicate that the outer sphere interaction of a chromium(III) complex with DNA is weak and unlikely to be the mechanism by which chromate causes DNA impairments in vivo and in vitro.  相似文献

Dissolved organic matter (DOM) plays an important role in heavy metal speciation and distribution in the aquatic environment especially for eutrophic lakes which have higher DOM concentration. Taihu Lake is the third largest freshwater and a high eutrophic lake in the downstream of the Yangtze River, China. In the lake, frequent breakout of algae blooms greatly increased the concentration of different organic matters in the lake sediment. In this study, sediment samples were collected from various part of Taihu Lake to explore the spatial difference in the binding potential of DOM with Cu. The titration experiment was adopted to quantitatively characterize the interaction between Cu(II) and DOM extracted from Taihu Lake sediments using ion selective electrode (ISE) and fluorescence quenching technology. The ISE results showed that the exogenous DOM had higher binding ability than endogenous DOM, and DOM derived from aquatic macrophytes had a higher binding ability than that derived from algae. The fluorescence quenching results indicated that humic substances played a key role in the complexation between DOM and Cu(II) in the lake. However, because of the frequent breakout of algae blooms, protein-like matters are also main component like hnmic matters in Taihu Lake. Therefore, the metals bound by protein-like substances should be caused concern as protein-like substances in DOM were unstable and they will release bound metal when decomposed.  相似文献

The marine bivalve, Perna viridis was exposed to sublethal concentration (one third of 24 h LC50) of either cadmium or copper for 1, 3 and 7 days and alterations in lipid peroxidation in the gill and muscle were investigated. Significant increase of the level of malondialdehyde, which is indicative of the peroxidative process, and a decrease of activity levels of glutathione peroxidase and glutathione transferase were observed in both the tissues of copper exposed mussels. the exposure of mussels to cadmium did not elicit any of the above changes.  相似文献

为了研究饲料中铜暴露对吉富罗非鱼幼鱼的毒性作用,将1 080条罗非鱼幼鱼暴露于6个浓度梯度的高铜日粮中,通过60 d的暴露试验,测定罗非鱼血红细胞微核变化和组织中铜的蓄积.结果表明,铜在各组织中积累量的顺序依次为:肝脏>脾脏>肠道>鳃>肌肉,与对照组相比,其中肝脏、脾脏、肠道中铜的含量随着时间和日粮中铜浓度的升高而逐渐升高,而鳃和肌肉中铜含量无显著差异;罗非鱼幼鱼血红细胞微核率、核异常率、总核异常率与日粮中铜的浓度存在一定的剂量—效应关系,并且核异常率高于微核率.  相似文献

为利用和人类更为接近的动物模型来研究在高脂血症条件下,BPA暴露是否能促进脂类和糖类代谢异常,并探索潜在的毒性效应机制,选取渡边可遗传高脂血症家兔(WHHL)模型,通过灌胃的方式将其暴露于400μg·kg-1体重的BPA溶液长达12周。在持续暴露第8周,检测了在空腹状态下,家兔血浆中葡萄糖、胰岛素和脂肪含量的变化,并根据所得结果进行了静脉注射胰岛素耐受试验(IVTT)。在暴露第12周,同样对空腹状态下,家兔血浆中的葡萄糖,胰岛素和脂肪含量进行了检测,并测量了血压和心率。然后,将所有的家兔进行解剖,通过苏木精-伊红(HE)及糖原(PAS)染色分别对心脏和肝脏部位的脂肪及糖原蓄积情况进行了病理学切片分析。同时,检测了肝脏中与脂类和糖类代谢相关基因在m RNA水平上的表达变化。结果显示,BPA暴露8周后促使WHHL家兔发生了胰岛素抵抗现象,导致第12周血糖及胰岛素含量升高,同时也促进了高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)及游离脂肪酸(NEFA)含量的上升。BPA暴露第12周,暴露组WHHL家兔的冠状动脉粥样硬化损伤程度未发生明显增加,但心肌细胞发生了肿胀,胞质中出现了脂肪的蓄积,同时伴随着心律失常。肝脏重量发生增加,肝细胞中同时出现了脂肪和糖原的蓄积现象,相关基因的表达发生了显著上调。研究结果表明,BPA持续暴露可促进WHHL家兔发生脂类和糖类代谢异常,其毒性效应机制可能和胰岛素抵抗及相关基因的异常表达有关。  相似文献

选择江西赣县大田稀土矿区,研究了矿区耕作农田土壤中稀土元素的含量、分布及赋存形态特征。结果表明,该区土壤中稀土元素含量明显高于对照区和全国土壤平均含量,且相对富含重稀土元素。土壤中稀土元素的分布模式受成土母岩及元素性质控制,呈现Eu和Ce负异常。稀土元素主要以可交换态和有机结合态(特别是松结有机态)形式存在,具有较高的活性和生物有效性,可能影响生态环境安全。  相似文献

运用多级连续提取法,对广州市不同季节不同采样高度大气PM2.5中重金属的化学形态和生物有效性进行分析。研究表明,PM2.5中各重金属元素之间的化学形态分布差异较大,Zn、Cd、As、Mn主要分布在F1(可溶态与可交换态)和F2(碳酸盐态、可氧化态与可还原态),绝大部分的Pb以F2存在,Ni和Mo主要分布在F1和F3(有机质、氧化物与硫化物结合态),Cu主要以F2和F3存在,Cr主要分布在F3和F4(残渣态),Co则4种形态平均分布。采样高度对重金属的化学形态分布影响不大,同一采样期内楼顶与地面样品中同一元素的化学形态分布结果比较一致。两个采样季节重金属的形态百分比存在不同程度的变化,2007年春重金属的不稳定态(F1)比例比2006年秋普遍增加,次稳定态(F2、F3)比例减少。在10种重金属中,Cd、Zn、Pb和As的生物有效性系数高(>0.7),属于生物可利用性元素,在环境中的活动性要明显高于其它元素;Mn、Cu、Mo、Co、Ni和Cr元素的生物有效性系数值在0.2~0.6之间,属于潜在生物有效性元素,在环境中比较稳定。  相似文献

Cypermethrin and carbaryl are both used to control pests of rice paddy crops but the two insecticides have different modes of action. In some areas or climates, the breeding of frogs living in rice paddy fields and the application of the insecticides coincide. The effects of the acetylcholinesterase (AChE) inhibitor carbaryl and the sodium channel blocker cypermethrin, both individually and in combination, were investigated on the survival and emergence of froglets of Fejervarya limnocharis using a 3?×?3 concentration factorial design (0, 25, and 50?μg L?1). Compared to controls, carbaryl showed no effect on survival and froglet emergence. Exposure to cypermethrin reduced the survival and froglet emergence in a concentration-dependent manner. When tadpoles were exposed to combinations of carbaryl and cypermethrin, the influence of carbaryl on survival and froglet emergence is less significant than that of cypermethrin.  相似文献

以天然河流中的日本青鳉(Oryzias latipes)行为变化规律为基础,采用水质安全在线生物预警系统(BEWs-1.1)对氰戊菊酯和氯化镉暴露下日本青鳉的行为反应进行了探讨.结果表明:1)在源水中,日本青鳉行为反应具有明显规律性,并表现出生物钟现象;2)氰戊菊酯和氯化镉连续暴露和间断暴露中,日本青鳉行为反应与污染物浓度和暴露时间均直接相关,并表现出相似的行为变化规律性,行为变化均符合生物行为的环境胁迫阈模型;3)高浓度(10mg·L-1)氯化镉间断暴露下的日本青鳉行为强度变化与低浓度(2mg·L-1)连续暴露下的行为强度变化差别不明显,而高浓度(10mg·L-1)氰戊菊酯间断暴露下的日本青鳉行为强度变化与低浓度(2mg·L-1)连续暴露时具有明显差异,高浓度暴露时产生了明显的行为毒性效应;4)器官损伤性污染物(如镉)引起的行为毒性效应在重新加入源水后可以得到部分恢复,而神经损伤性污染物(如氰戊菊酯)不可恢复,污染物不同的作用机理会影响水生生物的行为反应.  相似文献

We assessed the use of anodic stripping voltammetry (ASV) for in-situ determinations of both total concentration and speciation of dissolved heavy metals (Cd, Cu, Pb and Zn) in acid mine drainage (AMD). In the Kwangyang Au–Ag mine area of South Korea, different sites with varying water chemistry within an AMD were studied with a field portable anodic stripping voltammeter. Anodic stripping voltammetry after wet oxidation (acidification) was very sensitive enough to determine total concentration of dissolved Cd because Cd was dominantly present as ‘labile’ species, whilst the technique was not so effective for determining total Cu especially in the downstream sites from the retention pond, due to its complexation with organic matter. For dissolved Pb, the concentrations determined by ASV after wet oxidation generally agreed with those by ICP-AES. In the downstream samples (pH>5), however, ASV data after wet oxidation were lower than ICP-AES data because a significant fraction of dissolved Pb was present in those samples as ‘inert’ species associated with colloidal iron oxide particles. The determination of total dissolved Zn by ASV after wet oxidation appeared to be unsatisfactory for the samples with high Cu content, possibly due to the interference by the formation of Zn–Cu intermetallic compounds on the mercury coated electrode. In AMD samples with high dissolved iron, use of ultraviolet irradiation was not effective for determining total concentrations because humate destruction by UV irradiation possibly resulted in the removal of a part of dissolved heavy metals from waters through the precipitation of iron hydroxides.  相似文献

土壤活性有机质在土壤碳、氮、磷等养分元素的牛物地球化学循环方面起着十分重要的作用.森林土壤活性有机碳(SAOC)库是极易变动的,其组分动态与周转速率主要受森林植被类型、演替过程以及由此造成的凋落物输人类型、数量、质量和季节与温湿度、pH等变化的控制.尽管在过去几十年中,关于土壤可溶性有机碳(DOC)已开展了大量的研究,但关于其形成、转移与转化速率,以及对环境变化的反应等结论仍然零散,甚至相互矛盾,因此,研究土壤DOC动态及持续管理,必需从探讨其影响因子出发,找出其关键限制凶子进行调控.在将来的研究中,主要应集中在:(i)各种可溶性有机质(DOM)源和汇的大小与DOC定量变化关系,如何进行调控?(ii)在高度人为化管理的森林生态系统中,土壤DOC如何改变?(iii)DOC动态及周转速率的现代研究方法探索等.参110  相似文献

研究孕哺期BDE209暴露对母鼠胎盘和子代脑组织甲状腺激素脱碘酶(deiodinase,DI)基因表达的影响,及其在子鼠神经发育毒性效应中的作用。将75只雌性昆明小鼠随机分为对照组、低剂量组和高剂量组,暴露BDE20910 d后,与雄鼠合笼,每组选取怀孕时间相近(相差不过2 d)的8只母鼠孕期持续染毒至子鼠断乳。采用实时荧光定量PCR检测孕17~18 d胎盘、出生后60 d子鼠脑组织3种类型脱碘酶基因相对表达;利用Morris水迷宫评价出生后60 d子鼠学习记忆能力;测量第2、16、30和60天子鼠体重,观察孕哺期BDE 209暴露对子代生长发育的作用。结果显示,BDE 209对子鼠出生时体重未见明显影响;出生后30 d,高剂量BDE 209暴露组雌性子鼠体重显著低于对照组子鼠体重(p0.05),而低、高剂量暴露组雄性子鼠体重均显著低于对照组子鼠体重(p0.05,p0.01);出生后60 d,BDE 209对子鼠体重影响不明显。BDE 209暴露能够显著延长出生后60 d雌、雄性子鼠逃避潜伏期(p0.05或p0.01)。BDE209暴露显著降低母鼠胎盘中3种类型脱碘酶(主要是DI-3)基因表达(p0.05或p0.01);同时,BDE 209暴露可诱导出生60 d后雄性子鼠脑组织中DI-1基因表达(p0.05或p0.01),抑制雄性子鼠脑组织中DI-3基因表达(p0.05);BDE 209暴露对出生后60 d雌鼠脑组织脱碘酶未见明显影响(p0.05)。研究结果表明,孕哺期BDE209暴露可能通过影响母鼠胎盘组织脱碘酶(特别是DI-3)基因表达,导致子鼠神经发育毒性效应-学习记忆能力障碍。  相似文献
