Nephropsnorvegicus (L.) were subjected to 8 h of emersion, either between layers of seawater-soaked hessian with periodical (20 min) flushes of seawater (high humidity, HH) or to unprotected emersion (low humidity, LH). Blood ammonia levels rose during emersion in both groups but reached higher levels under LH conditions. Ammonia efflux rates after re-immersion were higher than those of control prawns, and amounts of ammonia excreted at such times were considerably higher than those calculated to have accumulated in the blood during emersion. Possible explanations for such differences are discussed. C aO2 and C vO2 decreased rapidly to ca. 10% normoxia values within 2 h of HH and LH emersion and remained low throughout the remaining emersion time. Emersion-induced tissue hypoxia increased blood concentrations of glucose and lactate. Lactate accumulation was higher during LH emersion, compared with HH emersion. Blood pH dropped ca. 0.40 units but increased again after 2 h of re-immersion. Acidosis was probably related more to respiratory difficulties (CO2 accumulation) than to lactate accumulation, as blood lactate values remained high after 2 h of re-immersion. The ability of N. norvegicus to cope with emersion appears to be little influenced by high humidity conditions. Received: 26 June 1996 / Accepted: 5 August 1996  相似文献

During commercial handling of Nephropsnorvegicus (L.) there are a number of situations when the prawns may be exposed to very high ambient ammonia levels. These experiments evaluated the effects of increased levels of ambient total ammonia (TA = NH3 + NH4 +) on␣blood ammonia, ammonia efflux rates and on the cardio-ventilatory performance of N. norvegicus. When prawns were taken from <1 to 2000 μmol TA l−1 medium, blood TA concentrations increased rapidly for the first 2 h but tended to drop thereafter. Original blood TA levels were restored 6 h after the prawns were transferred back from seawater containing 2000 to <1 μmol TA l−1. Sudden exposure to 500, 1000, 2000 or 4000 μmol TA l−1 medium induced blood TA concentrations to increase respectively to 50, 30, 33 and 36% of external concentrations (normally, internal TA values are much higher than external levels). Immediately after transfer back to seawater with low ammonia concentration ( <1 μmol TA l−1), excretion rates were higher than those of control prawns, and the absolute amounts of TA excreted were considerably higher than those calculated to have accumulated in the haemolymph. Heart rate (HR) and scaphognathite rate (SR) were not altered when prawns were subjected to sudden alterations in ambient ammonia ( <1 to 2000 to <1 μmol TA l−1). When water ammonia concentrations were altered more gradually, both rates increased, but only at 4000 μmol TA l−1. These results show that N. norvegicus is able to remove ammonia from the haemolymph and/or transform ammonia into some other substance when subjected to increased levels of ambient ammonia. Possible mechanisms involved (e.g. active transport across the gills; storage in some other tissue; glutamate synthe sis) are discussed. Received: 20 May 1996 / Accepted: 1 July 1996  相似文献

The subtidal crab Cancer pagurus (L.) experiences involuntary periods of emersion associated with practices used in their marketing and distribution. During 24 h emersion, impaired gill function caused an increase of circulating total ammonia (TA=NH3+NH4+) of 0.35 mmol TA l-1 (167%). The oxygen-binding characteristics of the haemocyanin of C. pagurus were examined at 10°C in the presence of total ammonia (0.2-1.0 mmol TA l-1). The haemocyanin-oxygen affinity was decreased in the presence of TA ((logP50/(log[TA]=0.16). Emersion induced significant acidosis and elevated circulating levels of haemolymph TA, lactate and urate, but all had returned to normal levels within 24 h of re-immersion. The accumulation of haemocyanin-modulating substances during 24 h emersion compensated partially (40%) for the effect of the acidosis, but the net effect of the emersion period was a significant decrease in oxygen affinity, corresponding to an increase of P50 (10°C ) from 1.24 kPa (immersed) to 1.96 kPa (24 h emersion). The implications of the findings are considered in terms of the effects and adaptations to emersion.  相似文献

The concentration of glutamine in Tridacna gigas haemolymph increased >35-fold following exposure to sea water supplemented with ammonium (20 μM), but no increase was observed with nitrate (20 μM). Lack of a diel cycle, no decrease in haemolymph glucose levels, the expression patterns of glutamine synthetase in zooxanthellae and host, and the lack of glutamine release in response to nitrate supplementation all support the proposition that the increase in haemolymph glutamine is a product of the host and not the zooxanthellae. Unlike ammonium, nitrate accumulates rapidly in the haemolymph. It has no effect on the concentration of glutamine in the haemolymph, but there is an increase in arginine, histidine and lysine in the haemolymph, suggesting the release of these essential amino acids from zooxanthellae. Glutamine synthetase (GS) activity decreased markedly in the gill and less so in the mantle over a period of 6 d exposure to elevated ammonium (20 μM). In contrast, GS activity in zooxan- thellae doubled. The response of zooxanthellae in situ was confirmed by incubating freshly isolated zooxanthellae for 4 d in ammonium, which resulted in a ten-fold increase in GS activity. Comparison of the in situ response of zooxanthellae with that obtained in vitro indicates that the symbionts are likely to be exposed to ammonium concentrations lower than that found in the haemolymph.  The effects of cadmium exposure and dietary status on cadmium accumulation, fatty acid (FA) content and profiles were investigated in two colour forms of the shore crab Carcinus maenas. Groups of shore crabs were either starved or fed with blue mussels, Mytilus edulis, during a 40 d exposure period to 2 or 6 μM Cd2+ (as CdCl2). Starved green individuals accumulated more cadmium in haemolymph and hepatopancreas than did red crabs and green crabs fed during the experiments. In the red colour form, no difference in cadmium accumulation was observed between starved and fed individuals. In both colour forms, hepatopancreas contained more FA than gills and muscle. The FAs often present in the largest amounts in the tissues were 16:0, 16:1ω7, 18:1ω7, 18:1ω9, 20:4ω6, 20:5ω3 and 22:6ω3. However, saturated (SAFA) and mono-unsaturated fatty acids (MUFAs) were dominant in hepatopancreas, whereas poly-unsaturated fatty acids (PUFAs) were dominant in gills and muscles. At the beginning of the experiment, the total FA content in the hepatopancreas was 111.6 mg g−1 (dry weight) for red crabs and 78.4 mg g−1 for green shore crabs. During the experiment, however, the FA content decreased in red crabs. This decrease was more pronounced for starved individuals than for fed individuals. Also, the decrease in FA content was more pronounced in crabs exposed to 6 μM cadmium compared to crabs exposed to 2 μM or crabs not exposed to cadmium. No change in FA content was observed in green shore crabs, irrespective of diet and cadmium exposure. For both colour forms, no change in FA content was observed for gills and muscle. In red crabs, a decrease was observed for all FAs in the hepatopancreas. This decrease, however, was more pronounced for SAFAs and MUFAs than for PUFAs, indicating that the metabolism of FAs during starvation and cadmium exposure is selective. The experiments indicate that green colour forms of shore crabs are more tolerant of natural stress such as starvation and anthropogenic stress, e.g. cadmium exposure, than are red colour forms of shore crabs. Oxygen levels were monitored within the mantle cavities of three barnacle species ( Chthamalus stellatus, Semibalanus balanoides, Elminius modestus), using optode microsensors. Conditions were always hypoxic, even when barnacles were actively using the prosoma and cirri to pump aerated seawater into the mantle cavity. Mantle fluid oxygen concentrations were extremely variable; behaviour and oxygen concentrations were not closely coupled. Ventilation of the mantle cavity depended partially on external water flow, with higher and more stable mantle fluid oxygen concentrations being sustained when the water around barnacles was agitated. During emersion, barnacles initially pumped seawater between the mantle cavity and the cone above the opercular plates to achieve ventilation. As water was lost it was replaced by air bubbles, eventually resulting in an air-filled mantle cavity. In S. balanoides and E. modestus, once the mantle cavity was filled with air, the barnacle usually used up the oxygen within the bubble within 2–3 h and did not regain oxic conditions until the barnacle was reimmersed in seawater. In C. stellatus, the air bubble was repeatedly refreshed for many hours by pneumostome formation. In response to low environmental salinity, all three species closed the opercular plates firmly and rapidly used up oxygen within the mantle fluid.  相似文献

Synthesis of the heat shock protein Hsp70 is one of the most important physiological mechanisms that intertidal organisms possess to counteract damage to macromolecules caused by stressors associated with the tidal cycle. However, the synthesis and activity of Hsp70 involves an elevated energetic cost. We evaluated the effect of the nutritional status (fed vs. starved for 2 weeks) of juvenile Concholepas concholepas mollusc on their capacity to synthesize Hsp70 during emersion (i.e. low tide) and immersion (i.e. high tide) at high temperatures (24 °C, e.g. summer conditions) and at low temperatures (7 °C, e.g. winter conditions). In addition, we evaluated whether Hsp70 is induced directly upon exposure to stress (emersion) or during recovery (re-immersion). Starvation decreased the content of stored energy substrates of juveniles as well as their ability to synthesize Hsp70 during emersion under thermal stress, especially at high temperatures. Additionally, analysis of environmental factors associated with laboratory simulation of tidal regimes indicated that juveniles in starvation, in contrast to fed juveniles, did not significantly increase their levels of Hsp70 during cold emersion (7 °C) or warm emersion (24 °C) or upon re-immersion. Induction of Hsp70 occurred during exposure to stress (low-tide conditions) and not when juveniles returned to “normal” conditions (high-tide conditions). Thus, the synthesis of Hsp70 for the juveniles of this intertidal snail species was coordinated and adapted to the tidal cycle, and the species responds in a similar way to hot and cold emersion conditions. The observed levels of Hsp70 reflect the ability of the individual to synthesize these proteins, which is dependent on the nutritional status of the individual.  相似文献

Survival, developmental and consumption rate (Artemia nauplii ingested per day) as well as predation efficiency (ingested per available Artemia nauplii) were studied during the larval development of the shallow-water burrowing thalassinid Callianassa tyrrhena (Petagna, 1792), which exhibits an abbreviated type of development with only two zoeal stages and a megalopa. The larvae, hatched from berried females from S. Euboikos Bay (Aegean Sea, Greece), were reared at 10 temperature–food density combinations (19 and 24 °C; 0, 2, 4, 8 and 16 Artemia nauplii d−1). Enhanced starvation resistance was evident: 92 and 58% of starved zoeas I molted to zoea II, while metamorphosis to megalopa was achieved by 76 and 42% of the hatched zoeas at 19 and 24 °C, respectively. The duration of both zoeal stages was affected by temperature, food density and their interaction. Nevertheless, starvation showed different effects at the two temperatures: compared to the fed shrimp, the starved zoeae exhibited accelerated development at 19 °C (8.4 d) but delayed metamorphosis at 24 °C (5.9 d). On the other hand, both zoeal stages were able to consume food at an increased rate as food density and temperature increased. Predation efficiency also increased with temperature, but never exceeded 0.6. Facultative lecithotrophy, more pronounced during the first zoeal stage of C.tyrrhena, can be regarded as an adaptation of a species whose larvae can respond physiologically to the different temperature–food density combinations encountered in the wide geographical range of their natural habitat. The population dynamics of zooxanthellae living in the mantle of a giant clam, Tridacna derasa, was studied. The giant clams with shell lengths of 5 to 6 cm which had been reared in the Palau Mariculture Demonstration Center, in the Republic of Palau, were transferred to aquaria on deck of the R.V. “Sohgen-maru” and kept in running sea water at 29 to 30 °C. Two clams were removed from the aquaria, and zooxanthellae in the mantle were isolated every 2 h for 24 h. Numbers of the zooxanthellae in or not in the cell division stage were counted for calculations of the zooxanthellae population in the mantle and their mitotic index (MI). The MI increased after sunset and reached the maximum values of 6.1 to 11.5% at 03:00 to 05:00 hrs. The specific growth rate, μ, estimated from the MI was 0.083 to 0.14 d−1. Five clams were kept in each of 2 Plexiglas containers in the aquarium for collection of the discharged feces every 3 to 4 h. The discharged zooxanthellae in the feces were counted. The zooxanthellae discharged in 24 h were 0.38 to 1.46% of the total zooxanthella population in the mantle, and 2.7 to 16.9% of the newly formed zooxanthella population in a day. Increase of zooxanthella population in the mantle was estimated from clam shell growth rate and from the correlation between zooxanthella population and clam shell size. Daily increase of zooxanthella population in the mantle was estimated to be approximately 7.6 to 19% of the newly formed zooxanthella population. Therefore, the sum of zooxanthellae populations accounting for daily increase in the mantle and discharge in the feces was 11 to 36% of the newly formed population. About 64 to 89% of the newly formed cells were missing; some of these may have been digested by the clam. Received: 14 July 1996 / Accepted: 19 August 1996  相似文献

Marine invertebrates are thought to accumulate 210Po primarily from their food. In this study, a pulse-chase methodology was used to examine the assimilation and depuration of 210Po by Mytilus edulis from the common marine alga Isochrysis galbana. The digestion of 210Po from I. galbana occurred via a biphasic process, characteristic of a rapid (extracellular) and slow (intracellular) digestion typical of marine bivalves. The mantle/gill and foot have no known digestive role, yet their 210Po specific activities increased after 24 h. It is proposed that this increase in 210Po specific activity was related to 210Po being incorporated into these tissues from 210Po assimilated from I. galbana during extracellular digestion. It is proposed that the linear loss of 210Po previously accumulated by control mussels was related to the continual state of renewal and replacement of cellular proteins, with 210Po turnover and metabolism governed by protein turnover and metabolism. M. edulis' assimilation efficiency of 210Po from the 210Po-labelled alga was calculated to be 17.2 ± 2.1%, and thus similar to that of Ag, Cd, Co, Se and Zn by bivalves from other marine algae species. It is proposed that the assimilation efficiency of 210Po is a function of protein assimilation. Received: 27 August 1998 / Accepted: 3 September 1999  相似文献

Mytilus edulis L., Ruditapes philippinarum (Adams & Reeve) and Tapes decussatus L. were fed particles of the same shape (spherical), the same density (2.1 g cm−3) and the same chemical composition (SiO2), but which varied in diameter from 5 to 37 μm. Findings obtained at different particle concentrations (mean ± SD) of 51 ± 2, 105 ± 18 and 171 ± 17 mg l−1 invariably indicate that significant proportions of all particles with diameters larger than from between 7.5 and 22.5 μm were preferentially rejected as pseudofaeces. We define the preferential ingestion index (PII) as the ratio between average particle volume in pseudofaeces and average particle volume in food. Whatever the particle concentration or the species, this PII was always statistically higher than 1. Irrespective of particle concentration, PII values in M. edulis were lower than in T. decussatus (averages of 1.2 and 2, respectively). PII values in M. edulis were also lower than in R. philippinarum maintained at particle concentrations above 171 ± 17 mg l−1. We suggest that preferential size-dependent rejection of larger particles could be of significant adaptive value in the natural environment, either if there are large inorganic particles, or if the average organic content of smaller particles is higher. We determined the temporal evolution of amylase, cellulase, laminarinase and protease in the digestive gland and crystalline style of cockles Cerastoderma edule held over 9 to 12 d in the presence and absence of food. Cockles were fed a constant diet of 1.5 mm3 l−1 of Tetraselmis suecica for 9 to 12 d and were then starved for 6 to 8 d in late summer (September 1992) and in winter (January 1993). Feeding increased the dry weight and total cellulase, laminarinase and protease activities of the digestive gland irrespective of season, whereas amylase activity remained unchanged. In winter (i.e. when cockles are metabolically weak) the response was faster and stronger, especially for protease. An additional experiment in September starved cockles for 20 d before resuming feeding. In agreement with the seasonal differences, the presence of food after prolonged starvation induced a rapid and marked increase in protease in the digestive gland of the cockles. In winter, the possible effects of the biochemical composition of food on their enzymatic response were tested by feeding two groups of cockles with the same ration of T. suecica but harvested at different growth phases. A compensatory induction of cellulases occurred in cockles fed on T. suecica with a lower carbohydrate content. In the crystalline style, the protein level and carbohydrase fell during the first day of feeding and increased during the first day of subsequent starvation. These results indicate that the release of enzymes from the style prevails over the incorporation of enzymes during the early stages of feeding, whereas the opposite occurs during starvation. We tested the influence of limiting access to prey on larval development of the crabs Cancer magister and Hemigrapsus oregonensis by raising their Stage 1 larvae in the laboratory on different prey densities and with various periods of access to prey. Experiments were conducted in 1995 and 1996 at the Shannon Point Marine Center in Anacortes, Washington, USA. Our results show that crab larvae do not require continuous access to prey for optimal development nor do they appear to require light for prey capture. Survival and duration of Stage 1 C. magister fed continuously on only one-fourth the amount of the control density of prey and those fed at the control density for only 6 h per day were the same as for larvae fed continuously at the control density (20 ml−1). Larvae with cyclic access to prey at the control density for 24 h and then starved for 72 h showed significantly lower survival and longer instar duration to Stage 2. Experiments on Stage 1 H. oregonensis which investigated a combination of prey density, period of access to prey and light/dark conditions during feeding revealed that survival decreased with decreasing prey density or with decreasing feeding period, but no differences were observed during periods of limited prey availability as a function of light or dark conditions. Stage duration was not affected by reduced prey density nor by the light/dark condition at the time of feeding, but it was prolonged when the period of access to prey was limited. The period of access to prey did not affect the weight of Day 1 Stage 2 larvae. Larvae fed high densities of prey for 4 h followed by 20 h of reduced-density diet exhibited the same survival and stage duration as controls that were continuously fed high-density prey. Our results define sub-optimal diets that can be used experimentally to determine the nutritional contributions made by naturally-occurring prey organisms during larval development in the two species. In nature, larvae may satisfy nutritional requirements through periodic encounters with dense prey patches during vertical migrations by day or night. The pattern and characteristics of diving in 14 female northern rockhopper penguins, Eudyptes chrysocome moseleyi, were studied at Amsterdam Island (37°50′S; 77°31′E) during the guard stage, using electronic time–depth recorders. Twenty-nine foraging trips (27 daily foraging trips and two longer trips including one night) with a total of 16 572 dives of ≥3 m were recorded. Females typically left the colony at dawn and returned in the late afternoon, spending an average of 12 h at sea, during which they performed ∼550 dives. They were essentially inshore foragers (mean estimated foraging range 6 km), and mainly preyed upon the pelagic euphausiid Thysanoessa gregaria, fishes and squid being only minor components of the diet. Mean dive depth, dive duration, and post-dive intervals were 18.4 m (max. depth 109 m), 57 s (max. dive duration 168 s), and 21 s (37% of dive duration), respectively. Descent and ascent rates averaged 1.2 and 1.0 ms−1 and were, together with dive duration, significantly correlated with dive depth. Birds spent 18% of their total diving time in dives reaching 15 to 20 m, and the mean maximum diving efficiency (bottom time:dive cycle duration) occurred for dives reaching 15 to 35 m. The most remarkable feature of diving behaviour in northern rockhopper penguins was the high percentage of time spent diving during daily foraging trips (on average, 69% of their time at sea); this was mainly due to a high dive frequency (∼44 dives per hour), which explained the high total vertical distance travelled during one trip (18 km on average). Diving activity at night was greatly reduced, suggesting that, as other penguins, E. chrysocome moseleyi are essentially diurnal, and locate prey using visual cues. Assimilation efficiencies (AEs) and physiological turnover-rate constants (k) of six trace elements (Ag, Am, Cd, Co, Se, Zn) in four marine bivalves (Crassostrea virginica Gmelin, Macoma balthica Linnaeus, Mercenaria mercenaria Linnaeus, and Mytilus edulis Linnaeus) were measured in radiotracer-depuration experiments. Egestion rates of unassimilated elements were highest during the first 24 h of depuration and declined thereafter. Significant egestion of unassimilated Co, however, continued for up to 5 d in Macoma balthica, Mercenaria mercenaria and Mytilus edulis. With the exception of the extremely low values for 110 mAg, 109Cd, and 65Zn in C. virginica, physiological turnover-rate constants (k) showed no general pattern of variation among elements, bivalve species or food types, and were relatively invariant. Values from  ≤0.001 to 0.1 d−1 were observed, but excluding those for Co, most values were  ≤0.04 d−1. In all four species, the AEs of Ag, Am, and Co were generally lower than those of Cd, Se, and Zn. The AEs of Ag, Cd, Se, and Zn in these bivalves are directly related to the proportion of each element in the cytoplasmic fraction of ingested phytoplankton, indicating that >80% of elements in a prey alga's cytoplasm was assimilated. C. virginica, Macoma balthica, and Mercenaria mercenaria assimilated ∼36% of the Ag and Cd associated with the non-cytoplasmic (membrane/organelle) fraction of ingested cells in addition to the cytoplasmic fraction. The ratio of AE:k, which is proportional to the consumer–prey trace-element bioaccumulation factor (concentration in consumer:concentration in prey) was generally greater for Cd, Se, and Zn than for Ag, Am, and Co. This ratio was lowest in Mytilus edulis, suggesting that this bivalve, the most widely employed organism in global biomonitoring, is relatively inefficient at accumulating important elements such as Ag, Cd, and Zn from ingested phytoplankton. Contents of free amino acids (FAA), protein and ammonium ions together with rates of ammonia excretion and oxygen consumption were measured in order to study the role of FAA as an energy substrate in developing eggs and larvae of seabass (Lates calcarifer) maintained in seawater (30 ppt) at 28 °C without feeding. Initially eggs contained 25.3 nmol ind−1 of FAA of which 21.5 nmol was rapidly utilised by the developing eggs and larvae during the period up to 40 h post spawning (PS) when nearly all the yolk had been resorbed. During the same period, a net increase in protein content of 1.7 μg ind−1 was observed, indicating that the major part of the amino acids lost from the free pool had been polymerised into body proteins. Assuming that the balance of the FAA after protein synthesis was used entirely for energy metabolism, FAA appeared to be an important energy substrate during the embryonic stages (2 to 16 h PS); after hatching, the contribution of FAA to energy metabolism was less significant. From 50 h PS until the end of the study period at 100 h PS, amino acids derived from somatic protein were used for energy metabolism. For the overall period from just after spawning up to 100 h PS, the data indicate that ca. 14% of the total aerobic energy metabolism was derived from amino acid catabolism.  The influence of moulting and ovarian maturation on cadmium accumulation in the tissues of female shore crabs Carcinus maenas exposed to 1 mg Cd l−1 in the water was investigated. Cadmium accumulation in all tissues examined was markedly increased in crabs in the postmoult stages (A and B) compared to crabs in all other moult stages. During the moult cycle, average cadmium accumulation in the midgut gland ranged from 29 μg Cd g−1 dw at premoult stage (D2) to 589 μg Cd g−1 dw at postmoult stage (A). Average cadmium concentrations in the haemolymph ranged from 0.56 μg Cd ml−1 at intermoult stage (C4) to 4.6 μg Cd ml−1 at postmoult stage (A), while the gills accumulated from 103 μg Cd g−1 dw in intermoult stage (C3) to 352 μg Cd g−1 dw in postmoult stage (A). Cadmium concentration in gills and haemolymph was also significantly higher in crabs in late premoult stage (D3) compared to C4-crabs, while midgut gland cadmium concentration remained elevated in C1- and C3- intermoult stages relative to C4. During ovarian maturation the cadmium accumulation in midgut gland, gills, ovaries and haemolymph decreased. Average cadmium concentration in the midgut gland decreased from 63 μg g−1 dw in ovarian Stage I to 19 μg g−1 dw in ovarian Stage VI. The same pattern was observed for gills, haemolymph and ovaries. The present study demonstrates that cadmium accumulation in the female shore crab strongly depends on the physiological status of the animal. A possible association between physiological calcium requirements and cadmium accumulation during moulting is discussed. Received: 20 January 2000 / Accepted: 20 July 2000  相似文献

The effects of acute (24 h) and chronic (4␣weeks) copper exposure and subsequent recovery in “clean” conditions on cardiac activity were examined in shore crabs ( Carcinus maenas). Cardiac activity was monitored by automated interpulse duration assessment (AIDA) which permits detection of irregularities in cardiac rhythms in selected invertebrates. Mean interpulse durations, maximum interpulse durations and coefficients of variation were significantly affected by exposure to copper for 24 h and 4 weeks. Mean interpulse durations decreased reflecting an increase in heart rate, whereas maximum interpulse durations and coefficients of variation increased with increasing exposure concentration. Recovery of normal cardiac activity was evident following a 6-week recovery period in “clean” conditions. However, high levels of mortality were seen among crabs exposed to the two highest copper concentrations (0.6 and 0.8 mg l−1 Cu). The extent of recovery was assessed by subjecting the surviving crabs to a physiological challenge. There were no apparent differences in circulatory response to physiological challenge between crabs previously exposed to copper and control crabs. Gill copper concentrations were significantly elevated in crabs that had previously been exposed to copper, despite their return to “clean” sea water for 6 weeks. Midgut gland wet weight/dry weight ratios, haemolymph total protein and midgut gland metallothionein concentrations did not reflect the previous copper exposure. The findings of the present study indicate that AIDA may be a valuable tool for use in toxicity testing. Received: 24 March 1997 / Accepted: 21 October 1997  相似文献

Cements from two species of barnacles, Balanuseburneus Gould and Balanus crenatus Bruguiére, were analyzed to identify the number and nature of proteins present. B. crenatus cement was composed mainly of a small peptide cross-linked into aggregates ranging in size from ˜ 3 kD to 40–50 kD. These aggregates could be reduced with 2-mercaptoethanol (2-ME) in the presence of sodium dodecyl sulfate (SDS) but only after incubation at 40 °C for 12 to 24 h and only when the cement was recently formed. Reductive alkylation of cysteine residues with 4-vinylpyridine produced a water-soluble peptide of less than 5 kD. By comparison, the cement of B. eburneus could be dissolved only partially in SDS and 2-ME when heated at 100 °C for 10 min. Five major proteins were identified by SDS-PAGE: 7, 22, 36 and 58 kD bands for which N-terminal sequence and amino acid compositions are presented; and a 52 kD band for which sequence data are given. A minor protein band of ˜ 80 kD has the same N-terminus as the 36 kD band. CNBr digests of individual proteins produced peptides for which sequence and composition data are also presented. The study was conducted during 1993 to 1995. In general, the proteins identified from B. crenatus cement are similar to those characterized from B. eburneus, and they are different in composition and sequence than those previously reported from Mytilus edulis. We examined natural selection within a population of marine mussels, sampled in southwestern England in June 1991, containing a high frequency of hybrids between Mytilus edulis L. and M. galloprovincialis Lmk. This system is particularly tractable for the assessment of natural selection because hybridization is common and individual mussels can be aged, allowing changes in the frequency of hybrid genotypes among age classes to be determined. We show that strong viability selection occurs among hybrid genotypes which results in the virtual elimination of M. edulis–like genotypes from the population over a period of 3 years. Recombinant hybrid genotypes are intermediate in fitness, with M. edulis–like genotypes having a lower survival rate and M. galloprovincialis–like genotypes having a higher survival rate than genotypes of mixed ancestry. Since intermediate fitness for hybrid genotypes is inconsistent with endogenous selection models we conclude that the structure and position of this hybrid zone is probably generated by exogenous selection. This pattern of selection is a recurring feature of this hybrid population and likely occurs elsewhere in the hybrid zone. Selection against M. edulis–like genotypes appears to be offset by extensive immigration of larvae dispersed from pure populations of M. edulis.
