Allozyme and mitochondrial DNA (mtDNA) genetic variation was compared in orange roughy (Hoplostethus atlanticus Collett) collected from waters off southern Australia and from waters about 22 000 km away in the North Atlantic west of Scotland. Samples were screened for 11 polymorphic allozyme loci and with 9 restriction enzymes. Significant heterogeneity between the two areas was detected for three allozyme loci (ADA *, CK * and GPI-1 *), and the overall G ST (gene-diversity statistic) value of 1% was small but significant. Significant mtDNA haplotype heterogeneity was observed after 2- of haplotype frequencies but not after a G ST analysis. Nucleotide sequence-diversity analysis showed very low net divergence (0.0023%) between the two samples. The Australian orange roughy had a lower allozyme heterozygosity and a lower mitochondrial DNA nucleon diversity than the North Atlantic sample. The very limited, although significant, allozyme and mitochondrial DNA heterogeneity between these areas suggests that there is some gene flow between these two populations. The species appears to be widespread, with its presence reported from the southern Pacific, southern Indian, and northern and southern Atlantic Oceans, and it is likely that gene flow between the antipodes is mediated by stepping-stone exchange between adjacent populations rather than by direct migration.  相似文献

The variability of 14 enzyme-coding genes has been analysed in samples from 19 populations of the oyster Ostrea edulis L., collected along the Atlantic and Mediterranean coasts of Europe. We found an abundance of clines, which appeared at 8 loci, including the most polymorphic (AP-2 *, ARK *, EST-4 *, MDH-2 *, ME-1 *, 6PGH *, PGI * and PGM *). Another 6 loci (ALDH *, EST-3 *, EST-5 *, IDH-2 *, MDH-1 *, ME-2 *) exhibited V-shaped patterns of gene-frequency variation, with clines at one or both sides of the Straits of Gibraltar. The observation of coincident clines at many loci can be explained by a model of secondary intergradation. The geographical location of the midpoints of the clines and V-shaped patterns suggests the existence of two ancient Atlantic and Mediterranean oyster stocks which became differentiated in allopatry and subsequently merged. Clines observed along Atlantic and/or Mediterranean coasts at the loci with V-shaped patterns must have arisen independently. The large heterogeneity observed in the levels of gene differentiation (G ST ) across loci (G ST ranged from 0.008 to 0.290) and important differences in estimates of gene flow obtained by different methods suggest that the populations of O. edulis are not in genetic equilibrium. Lack of population equilibrium can be due to natural selection and/or restrictions to gene flow. The average among-population variability was higher than in other oyster species that do not show incubatory habits, and represented 8.8% of the total heterozygosity. Levels of intrapopulation variability were lowest in populations from the North Atlantic, suggesting low population sizes in that area.  相似文献

Samples representing the damselfishes (Perciformes: Pomacentridae) Stegastes nigricans, Chrysiptera biocellata, C. glauca, and C. leucopoma from North Male Atoll, Republic of Maldives, and Guam, Southern Marianas, were surveyed electrophoretically. A sample representing C. leucopoma from Ngemelis Island, Republic of Palau, was included in the analysis to provide a within-Micronesia comparison. On average, products of 23 presumptive protein-coding loci were examined in each sample. In comparisons of Maldivian and Guamanian samples, absolute or virtually fixed allelic differences were detected at: (1) ADA * and PEPS * for Stegastes nigricans; (2) ADA *, AAT-1 *, AAT-2 *, and mMDH * for C. biocellata, and; (3) ADA *, AAT-1 *, and AAT-2 * for C. glauca. At three polymorphic loci, ADA *, sMDH-1 *, and sMDH-2 *, similar allele frequencies were found in Palauan and Guamanian samples representing C. leucopoma, suggesting that these samples share a gene pool. Significantly different allele frequencies at ADA * and sMDH-1 * and a virtually fixed allelic difference at sMDH-2 * were detected between the Maldivian samples and each of the Micronesian samples representing C. leucopoma. The observed patterns of allele frequency differentiation suggest that Maldivian and Micronesian samples of each of the four study species represent separate demes.  相似文献

A survey of intraspecific allozymic variation among samples of the damselfishes Chrysiptera cyanea and Pomacentrus coelestis collected from Palau and Okinawa revealed levels of genetic divergence far in excess of estimates reported previously for populations of coral reef fishes. Absolute or nearly fixed differences in allele frequencies were detected at ADA *, sMDH-2 *, MEP-1 *, PEPB *, PEP-LT * and sSOD *, and at sAAT *, GPI-A *, LDH-1 *, and PEPB *, among the geographic samples of C. cyanea and P. coelestis, respectively. Examination of allele frequencies at most other loci (26 for C. cyanea; 24 for P. coelestis) revealed slight differentiation or identical fixation among the geographic samples. The observed patterns of allele frequency distribution suggest the existence of localized demes of these fishes: these demes may be cryptic and/or incipient species. Whether or not speciation is incipient, the observed patterns of allozymic and isozymic variability suggest that natural selection is a factor in the maintenance of population substructuring of the study species. Pronounced allelic differences were highly concentrated at a small number of loci: strongly bimodal frequency distributions of loci relative to their genetic identities (I) were observed for among-population comparisons in both study species. Allozymes encoded by the PEPB * locus in samples of the noncongeners from Palau exhibited identical electrophoretic mobilities. In Palauan P. coelestis, mSOD * is expressed but sSOD * apparently is not, whereas in Okinawan P. coelestis, both mSOD * and sSOD * are expressed.  相似文献

Dascyllus aruanus were collected from 13 different locations in French Polynesia between 15 November 1990 and 15 February 1991 in order to examine larval dispersal on four spatial scales: within-feef, within-island, within-archipelago, among-archipelagoes. Average polymorphism was analysed by protein electrophoresis at two levels (P95=0.285 and P90=0.107) from 12 and 3 polymorphic loci, respectively. Spatial genetic variation displayed a low level of differentiation between populations among archipelagoes, and homogeneity at lower spatial scales. Two hypotheses are proposed to explain the genetic structure observed. The first suggests substantial gene flow between the islands during the pelagic larval phase, the second that the absence of differentiation is the result of recent colonisation. Genetic variation amongst size classes showed a significant heterozygote deficiencies at two loci (PGM * and EST-2 *) in the smallest size class. This suggests a cyclic selection which affects larvae and adults differentially. The data revealed little differentiation among populations at the different localities, despite the short larval duration of D. aruanus; this suggests that larval duration is not the main factor presently affecting genetic structure in an insular model. The possible modification of mercury toxicity by selenium in embryos of the Pacific oyster Crassostrea gigas and the larvae of the crab Cancer magister was investigated. Mercury concentration eliciting abnormal development in 50% of the oyster embryos (EC50) was 5.7 g l-1 (48 h) and mortality in 50% of the crab larvae (LC50) occurred with 6.6 g l-1 (96 h). The 48 h EC50 for selenium was greater than 10,000 g l-1 for oyster embryos and the 96 h LC50 for crab zoeae was 1040 g l-1. The response from each species, when exposed to both toxicants, revealed, that a high level of selenium (5,000 g l-1) increased mercury toxicity. Moderate selenium concentrations (10 to 1,000 g l-1) tended to decrease mercury toxicity, although no statistical verification could be made. The order of administration of toxicants had no effect on the response of Crassostrea gigas embryos. Early developmental stages (8 h) of C. gigas embryos were most sensitive to dissolved Hg; toxicant administration 24 h after fertilization resulted in no apparent abnormalities in development.  相似文献

Esterase and glucosephosphate isomerase loci were examined in different year classes of the snapper Chrysophrys auratus Forster from the Hauraki Gulf in New Zealand and compared with temperatures during the immediate post-spawning period, the time when snapper larvae suffer high mortality. Year classes spawned in warm years have a low frequency of the Est-42 allele and year classes from cold years a high frequency of Est-42. Gpi-12 frequencies showed no significant change between warm and cold years. Annual fluctuations in allele frequencies within stocks may account for the observed excess of esterase homozygotes in other marine teleosts.  相似文献

Samples of southern bluefin tuna, Thunnus maccoyii (Castelnau), taken from off the coasts of South Africa, Western Australia, South Australia and Tasmania from 1992 to 1994 were analysed for six polymorphic allozyme loci (ADA *, GDA *, GPI-A *, MPI *, PGDH * and PGM-1 *, n = 595 to 733 per locus) and for mitochondrial DNA variants revealed by three restriction enzymes (Bam HI, Bcl I and Eco RI) detecting polymorphic cut sites (n = 555). No significant spatial heterogeneity was detected. There were no sex-related differences in allele or mtDNA haplotype frequencies. Juveniles (30 to 35 cm and 46 to 54 cm) from what are thought to be two temporally-separated spawning peaks showed no significant genetic differentiation. There were also no significant differences in allele or haplotype frequencies between fish smaller than 70 cm and those larger than 70 cm. These data are consistent with the null hypothesis of a single unit stock of southern bluefin tuna, with a single spawning area. Nine polymorphic loci were found among 42 presumptive protein-coding gene loci surveyed among 474 red drum (Sciaenops ocellatus) sampled in 1987 from 13 nearshore and 1 offshore localities from the Atlantic coast of the southeastern USA and the northern Gulf of Mexico. The mean number of alleles over the polymorphic loci was 3.8, and the average heterozygosity over all loci examined was estimated as 0.047. These data indicate that red drum have normal levels of genetic variability. Wright'sF1 ST values (the standardized variance of allele frequencies between samples) over all polymorphic loci ranged from 0.009 to 0.027 (meanF1 ST =0.019), and estimates of the effective number of migrants (N e m) per generation using Wright's island model ranged from 9.0 to 27.5. High levels of gene flow among the red drum samples were also indicated by Slatkin's qualitative analysis using conditional average allele frequencies. Nei's estimates of genetic distance between pairs of samples ranged from 0.000 to 0.009, indicating a high degree of nuclear gene similarity among all samples. Highly significant heterogeneity in allele frequencies at the locus for adenosine deaminase was detected between red drum sampled from the Atlantic and those sampled from the Gulf and among red drum sampled from the Gulf.  相似文献

The northern Pacific seastar Asterias amurensis Lütken was recently introduced to Tasmanian waters, possibly through ballast water discharged from ocean-going vessels. Three Tasmanian populations and seven native populations from Japan and eastern Russia were examined in 1994 for variation at 22 allozyme loci. There was significant spatial heterogeneity in gene frequencies among the native populations, especially for the ocus APK *, where G ST was a very high 0.47. The Tasmanian populations could not be separated genetically from one another, suggesting they have a common origin from a single introduction. The average heterozygosity per locus of the Tasmanian populations (0.116 to 0.127, mean 0.123) was 30 to 40% less than that of the native populations (0.177 to 0.216, mean 0.192), suggesting that colonisation of Tasmanian waters was accompanied by a population-size bottle-neck. The Tasmanian seastars were genetically more closely related to the two populations from central Japan (Suruga and Tokyo Bays) than to populations from Vladivostok, northern Japan (Yoichi, Nemuro and Mutsu Bays) or southern Japan (Ariake Sea). However, there were significant differences between the populations from Tasmania and central Japan, which may be attributable to founder effects or may mean that the true source of the Tasmanian introduction has yet to be identified.  相似文献

To determine the genetic population structure of blue crabs (Callinectes sapidus Rathbun), electrophoretic allozyme analysis was performed on 750 individuals collected from 16 nearshore locations ranging from New York to Texas, USA. Twenty enzymes and non-enzymatic proteins coded by 31 presumptive loci were examined. Twenty-two loci were either monomorphic or polymorphic at less than theP 95 level; alleles for these polymorphic loci were geographically dispersed. Allele frequencies for three of the remaining polymorphic loci were homogeneous over all populations, as were levels of polymorphism and heterozygosity. Phenograms generated by the UPGMA (unweighted pair-group method using arithmetic averages) and distance Wagner methods exhibited no geographic pattern in the clustering of populations. Estimates ofN em (effective number of migrants per generation between populations) indicated substantial gene flow, with aalues sufficiently high to infer panmixia between all blue crab populations from New York to Texas. However, despite this high level of gene flow, two striking patterns of geographic differentiation occurred: genetic patchiness and clinal variation. Allele frequencies atEST-2, GP-1, IDHP-2, DPEP-1, DPEP-2, andTPEP exhibited genetic patchiness on local and range-wide geographic scales, and allele frequencies atEST-2 varied temporally. Genetic patchiness in blue crabs is likely to be the result of the pre-settlement formation and subsequent settlement of genetically heterogeneous patches of larvae; allele frequencies of those larval patches may then be further modified through ontogeny by localized selection. In the Atlantic Ocean, a regional latitudinal cline ofEST-2 allele frequencies was superimposed on the range-wide genetic patchiness exhibited by that locus. This pattern against a background of high gene flow is highly likely to be maintained by selection. In estuaries along the Atlantic Ocean coast, a combination of low adult long-distance migration and a high retention rate of locally spawned larvae could serve to segregate populations and allow for the development of the geographic cline inEST-2. The Gulf of Mexico showed no apparent cline, perhaps due to long-distance migration of females in some regions of the Gulf, or to masking by genetic patchiness. These results emphasize the importance of both ecological and evolutionary time scales and structuring mechanisms in determining genetic population structure.  相似文献

Trochus niloticus L. and Tectus coerulescens Lmk., two coral reef trochid gastropods that have similar life-histories including a lecithotrophic larval stage, were sampled from reefs in the northern, central and southern sections of the Great Barrier Reef (GBR) in 1991. Significant sex-ratio biases were noted, and these varied among reefs, apparently with latitude. Demographic data suggested that highly discontinuous and localized recruitment occurs. Surveys of allozyme frequencies at 12 loci revealed no significant genetic differences among populations of Trochus niloticus at any geographical scale. High gene flow between zones of the GBR was inferred, with the number of migrants per generation (N m ) of the order of 100. Directional selection was thought to occur at one locus (GDH *). Genetic variability in T. niloticus (H=0.069 to 0.110, and only three loci polymorphic) was low compared with other trochids. This was thought to be due to smaller effective population size, resulting from an unbalanced sexratio, aggregative spatial distribution of adults, high variance in reproductive success, and/or the occurrence of population extinctions and recolonisations. In contrast, Tectus coerulescens exhibited significant genetic differences between zones, indicating a much lower rate of migration between populations (N m 1 to 10), and displayed high genetic diversity (H=0.225 to 0.279). A trend for increasing genetic diversity from the northern to southern GBR was found in both species. It is not clear whether the occurrence of two contrasted population genetic structures in species with apparently similar life-histories is due to ecological or historical factors.  相似文献

In the period from 1980 to 1984 organic phosphorus, nutrients, primary production rates (14C), chlorophyll a (chl a) standing crops, and basic oceanographic parameters were measured during 23 cruises at six stations in the open waters of the northern Adriatic Sea. These waters are significantly influenced by polluted Po River discharge. Organic phosphorus was correlated with several parameters which characterize phytoplankton activity and organic matter decomposition processes. In the late winter-spring period, organic phosphorus is produced during phytoplankton blooms. It is hypothesized that microzooplankton grazing is the main factor increasing the organic phosphorus concentrations in summer (up to 1.1 mol 1-1). Fall and winter had much lower values (below 0.3 mol 1-1) due to remineralization processes and an increased water mass exchange between the northern and central Adriatic regions. The direct contribution of organic phosphorus by freshwater discharge was not found to be significant. The higher organic phosphorus concentrations that can occur in low salinity waters are most likely due to their increased capability to support primary production.  相似文献

Samples of yellowfin tuna (Thunnus albacares) collected in 1991 and 1992 from the western, central and eastern regions of the Pacific Ocean were examined for genetic variability. Four polymorphic allozyme loci (ADA *, FH *, GPI-S * and GPI-F *) were examined in all samples and a fifth polymorphism (GDA *) was examined in western and central samples only. Samples were also screened for mitochondrial DNA variation following restriction analysis by two enzymes (BcII and EcoRI) detecting polymorphic cut sites. Eighteen mtDNA haplotypes were revealed, with an overall nucleon diversity of 0.678. A subset of individuals screened for eight restriction enzymes had an overall nucleon diversity of 0.724 and a mean nucleotide diversity per sample of 0.359%. No significant spatial heterogeneity was detected for alleles at the ADA *, FH *, GPI-S * and GDA * loci nor for the mtDNA haplotypes. Significant heterogeneity was detected for GPI-F *. At this locus, the two eastern samples (southern California and northern Mexico) were not significantly different from each other but were significantly different (P<0.001) from the five western/central samples (Philippines, Coral Sea, Kiribati, Hawaii-91 and Hawaii-92). GPI-F * 100 was the most common allele in western and central regions, GPI-F * 75 the most common in eastern samples.  相似文献

The Patagonian squid, Loligo gahi DOrbigny, has been described as having temporally and spatially identifiable spawning aggregations. Variation at six microsatellite loci was used to assess whether seasonal and geographical spawning groups around the Falkland Islands represent distinct sub-populations. Genetic variation at these loci is high in this species (mean expected heterozygosity=0.87; mean number of alleles=14.7). No evidence of significantly different allele frequencies was found, either between samples from putative spawning cohorts or geographical areas, indicating that L.gahi around the Falkland Islands comprise a single genetically homogeneous population. Age structure analysis of samples (from statolith growth increments) indicated substantial spread in hatching dates among individuals of similar size and maturity status, suggesting the potential for extensive interbreeding between seasonal cohorts. A sample of L.gahi from the SE Pacific (Peru) displayed distinctly different gene frequencies (and allele size distribution at one locus) from SW Atlantic samples, supporting the suggestion that SE Pacific and SW Atlantic populations may represent distinct subspecies.Contributed by J.P. Growth rates of anchovy larvae, Engraulis mordax, reared for 19 days under constant environmental conditions on a diet of laboratory-cultured organisms, exceeded the growth rates of anchovies fed on a diet of wild plankton. The rotifer Brachionus plicatilis was found to be a nutritous food source when fed to the larvae in concentrations of 10 to 20/ml and in combination with the dinoflagellate Gymnodinium splendens (100/ml). Optimum conditions were determined for mass culture of the rotifer. A high food concentration was the most important parameter needed to assure a high yield of rotifers. Large volumes (464 I) of the unicellular flagellate Dunaliella sp. were cultured for feeding the rotifers. The rotifer culture technique described produces approximately 2.5×106 organisms/day, providing a reliable food source for rearing studies. The lengths of B. plicatilis (without eggs) ranged between 99 and 281 , most rotifers being larger than 164 and less than 231 . Individuals weighed 0.16 g and contained 8×10-4 cal.  相似文献

Variations at 22 enzyme coding loci were surveyed in 11 populations of the oyster Ostrea edulis L., which were sampled between 1988 and 1990 along the Atlantic and Mediterranean coasts of Europe. Atlantic oyster beds suffered a steady decline during the last century, and restocking of beds with oysters of foreign origin has probably resulted in a high degree of interbreeding of natural oyster stocks from all Atlantic Europe. Our study confirms the low levels of genetic variability previously reported for the oyster populations from the Atlantic coasts, and extends it to the Mediterranean coasts. The locus arginine-kinase (ARK *) exhibited a high degree of interpopulation differentiation (F ST=0.289), resulting from extensive variation in gene frequencies along a geographical cline. However, the overall genetic differentiation between populations was slight, and similar to that reported for other local populations of bivalves (mean genetic distance between populations is 0.010, mean F ST=0.062). A general pattern of increasing differentiation along the coastline in an Atlantic-mediterranean direction emerged; but genetic differentiation among the Atlantic populations was not significantly lower than that observed among the Mediterranean populations. This and other results suggest that the effects of extensive transplantation of oysters among various areas in Europe are detectable only in some particular localities. The geographical distribution of low-frequency alleles suggests a restriction to gene flow outwards from the Mediterranean Sea, across the Straits of Gibraltar.  相似文献

Electrophoretic variation in proteins encoded by 23 loci revealed substantial genetic differentiation among populations of bicolor damselfish (Eupomacentrus partitus) collected from four coral reefs in the Florida Keys, USA, during 1986–1988. Genetic differentiation was concentrated between a sample collected from Little Grecian Rocks Reef (LGR) and the remaining samples, including fish from a reef only 600 m distant (Grecian Rocks Reef). Genetic distinction of the LGR sample derived from significantly heterogenous allelic frequencies at six of eight polymorphic loci. Aco-1 (aconitase); Ada (adenosine deaminase); Gpi-2 (glucosephosphate isomerase); Ldh-2 and Ldh-3 (lactate dehydrogenase); and Me-1 (malic enzyme); nevertheless, differentiation at cytosolic aconitase (Aco-1) far exceeded that observed for other loci (fixation index, F ST=0.482), and differences in Aco-1 allele frequencies were largely responsible for large genetic distances (0.20) between LGR and the other reefs. Paradoxically, estimates of numbers of migrants exchanged between reefs per generation (mN e=17.47) indicated the potential for extensive gene flow. The extent of genetic differentiation among these populations is evaluated relative to models of population genetic structure based on equilibrium between gene flow and natural selection or genetic drift.  相似文献

Adenosine triphosphate (ATP) measurements are used to determine vertical and seasonal distributions of microorganisms and meiofauna in sediments from a 14 m-deep mud bottom in central Long Island Sound on 12 sampling dates from April 1975 to October 1976. Below the topmost 1 cm of sediment, ATP measurements can be useful in estimating and comparing standing stocks of microorganisms and meiofauna. In the top 1 cm, however, large quantities of newly settled bivalves (Yoldia limatula, Nucula annulata, and Mulinia lateralis) and juvenile polychaetes (Owenia fusiformis) in summer and fall months account for total ATP concentrations. The ATP content of individual meiofauna ranges from 1.97 ng individual copepod nauplius-1 to 190.7±60 ng individual M. lateralis -1. In general, the total ATP content of individual polychaetes and bivalves is much higher than that of individuals of other groups. However, on a mg ATP per g wet or dry tissue basis, the ATP content of micro- and meiofaunal taxa are not significantly different. In addition to providing a means for comparing micro- and meiofaunal standing stocks, ATP measurements permit examination of the relative contribution of different meiofauna to the total living biomass of meiofauna in sediments. Total sediment ATP concentrations are greatest in the top 1 cm at all seasons, and decrease with increasing depth in the sediment. Annual concentrations in the topmost centimeter average 4.22 g g dry sediment-1 and range seasonally from 1.09 to 7.64 g g dry sediment-1. At a depth of 10 cm, values average 0.16 and range from 0.019 to 0.35 g g dry sediment-1. High ATP concentrations in surface sediment reflect high concentrations of microorganisms and meiofauna at the sediment-water interface. The top 2 cm of sediment contain 71% of all meiofauna, with 41% occurring in the topmost cm. In general, densities are lowest in the winter and highest during the spring and summer, averaging 490 individuals 10 cm-2, and varying from 87 to 1366 individuals 10 cm-2. Because of wide variation in recruitment patterns of the benthos in Long Island Sound, the extremes of the range in meiofaunal densities can be observed in the same month in two different years. In order to monitor and compare standing stocks of organisms less than 1 mm in size in sediments, the ATP assay can save hours of processing time compared with alternate methods such as direct counts.  相似文献

Squids of the genus Illex are representative of the family Ommastrephidae and account for 65% of the world’s cephalopod captures. Illex is formed by four taxa distributed throughout the Atlantic Ocean (I. argentinus, I. coindetii, I. illecebrosus and I. oxygonius), whose identification and phylogenetic relationships based on morphological characters remain controversial. Thirty-seven enzyme-coding loci were analysed in 230 individuals from seven populations of Illex and ten specimens of Todaropsis eblanae, which were used as the outgroup. Two to four enzyme loci (ALPDH*, IDHP-1*, MEP* and SOD*) were diagnostic among Illex species depending on the species-pair comparison. Individuals morphologically identified as I. oxygonius were also found genetically distinct, which proves the taxonomic validity of this species. No significant intraspecific genetic heterogeneity was detected within Illex argentinus, I. coindetii and I. illecebrosus (Mean G ST= 0.011, 0.003, 0.017, respectively). I. illecebrosus and I. oxygonius were shown as sister species with a close relationship to I. argentinus, whereas I. coindetii formed a different lineage.  相似文献
