The objective of this study was to examine the toxicological effect of two major heavy metal pollutants, lead chloride (PbCl2) and cadmium chloride (CdCl2), in the freshwater climbing perch, Anabas testudineus. Fish were exposed to 1.0 and 2.0 mg/L both metals and histopathological changes in gill, kidney and liver tissues were studied. Major changes observed in gill tissue were epithelial lifting, proliferation of epithelial cells, fusion of secondary lamellae, hyperplasia and hypertrophy of mucous cells, and necrosis of epithelial cells. Cell necrosis, degenerated kidney tubules, congestion, lymphocytic infiltration and vacuolation were the major abnormalities observed in kidney tissue. The most conspicuous changes in liver were darker nucleoli, irregularly shaped hepatocytes with dilated blood capillaries, and focal as well as single necrosis. Fish specimens exposed to PbCl2 exhibited pronounced changes in all tissues examined compared with those exposed to CdCl2. It is evident from this study that heavy metals can cause significant histopathological changes in fish tissue.  相似文献

The influence of dietary copper (Cu) exposure on accumulation, growth, and hematological parameters was investigated in Cyprinus carpio after sub-chronic ingestion of 0, 250, 500, 750 or 1000?mg?kg?1 for 60 days. The profile of Cu accumulation among tissues in C. carpio was dependent on the exposure period and Cu concentration. Liver of C. carpio was the predominant storage tissue and the order of Cu accumulation in tissues was liver?>?intestine?> gill?>?kidney?>?muscle. Cu concentration at >125?mg?kg?1 reduced growth rate, and was inversely related to growth. The RNA?:?DNA ratios were not affected by exposure and there was no correlation between growth rate and RNA?:?DNA ratio in liver and muscle. There were no significant effects of exposure on blood parameters except for magnesium. Cu exposure time and dose increased the serum glutamic-oxaloacetic transaminase (SGOT) and serum glutamic-pyruvic transaminase (SGPT) activity levels.  相似文献

The acute toxicity of paper mill wastewater to Oreochromis mossambicus was investigated with the lethal concentration (LC50) value 6.5% for 96?h exposure. This concentration was used as a baseline to study the effects of paper mill effluent on histopathological changes in gills, liver, kidney, and brain of fish. In the gills, filament cell proliferation, cellular infiltration, hemorrhage, and epithelial lifting were observed. In the liver, vacuolation of hepatocytes and necrosis were noted. In kidney, exfoliation and swollen with pyknotic nuclei were identified. Similarly, the brain also showed enlarged pyramidal cells, binucleated nuclei, vacoulation, and necrosis. These changes occurred predominantly in 21days following exposure of fish to the industrial waste water. Paper mill wastewater was found to be highly toxic to fish.  相似文献

Sublethal effects of chromium trioxide on mosquito fish, Gambusia affinis, were carried out for 28 days on day 4, 8, 12, 16, 20, 24, and 28. The sublethal effects of chromium (LC10, 77.62?mg?L?1) on antioxidant enzymes, thiobarbituric acid reactive substance (TBARS), locomotor behavior, and gill morphology were studied. The antioxidant enzymes like superoxide dismutase (SOD) and catalase were induced throughout the exposure span of 28 days. Percent induction was also increase with the days of exposure. Lipid peroxidation product, malondialdehyde was enhanced in the viscera tissue of chromium-exposed fish. Perturbation of locomotor behavior like decrease in distance travelled (m?min?1) and in swimming speed (cm?s?1) was observed in exposed fish. Chromium toxicity caused alterations in gill morphology like hypertrophy and hyperplasia in secondary lamellae followed by detached epithelium with severe necrosis. It can be concluded from the results that Cr intoxication in Gambusia fish, caused induction in antioxidant enzymes and lipid peroxidation, plus changes in gill morphology and locomotor behavior.  相似文献

In this study, the effects of environmental hypercapnia on hemato-immunological parameters and the activities of respiratory enzymes such as carbonic anhydrase (CA) and Na+, K+-ATPase were investigated in rainbow trout (Oncorhynchus mykiss) tissues (gill, liver and kidney). Batches of 12 fish were exposed to 4.5 mg L?1 (control) and 14 mg L?1 CO2. No mortalities occurred during the 14 days of the experimental period. Red blood cell (RBC), hemoglobin (Hb), and hematocrit (Ht) levels, and innate immune parameters such as nitro blue tetrazolium (NBT), lysozyme, and myeloperoxidase activities, and the melano-macrophage frequency were negatively affected by elevated CO2 levels. Patterns of change in CA activity differed among the gill, liver, and kidney. Compared with the activities of CA in the control group, the CA enzyme was significantly stimulated at day 7 in the gill tissue, whereas it was stimulated at day 14 of the experiment in the liver tissue of fish exposed to 14 mg L?1 CO2 (P < 0.05). In contrast to the pattern of CA enzyme activities, the Na+, K+-ATPase enzymes were stimulated significantly in the liver after day 7 but inhibited in the kidney and gill (P < 0.05). These results suggest that a subchronic exposure to hypercapnia of rainbow trout tissues may lead to adaptive changes in the respiratory enzymes and negatively affects hemato-immunological parameters.  相似文献

The cytogenotoxicity and histopathological alterations induced by xenobiotics in Lekki Lagoon and Ogun River on Synodontis clarias were investigated. Fish from these water bodies and a fish farm (control) were examined for micronucleated, binucleated, and immature erythrocytes in both gill and peripheral blood. Also gill, liver, kidney, and ovary were processed for histopathology using hematoxylin-eosin staining. Concentrations of cadmium, zinc, lead and copper in the water were determined. There was significant (p < 0.05) increase in micronucleated, binucleated, and immature erythrocytes in both gill and peripheral blood of S. clarias from the lagoon and river compared to the reference site. Loss and disorganization of the primary and secondary lamellae, multifocal degeneration, hemorrhages, cellular infiltration, congestions, vacuolations, atresia, and necrosis were common lesions in the examined tissues of fish from the lagoon and river. Cd, Zn, Pb, and Cu in water samples from the lagoon and river were higher than the reference site. Xenobiotics in Lekki Lagoon and Ogun River, mostly metals, induced deoxyribonucleic acid (DNA) and pathological damage in S. clarias.  相似文献

In current research, the combined effects of copper oxide nanoparticles (CuO NPs) and titanium dioxide nanoparticles (TiO2 NPs) on the histopathological anomalies of gill and intestine tissues in common carp (Cyprinus carpio) were studied. Common carp were exposed to TiO2 NPs (10.0?mg L?1), CuO NPs (2.5 and 5.0?mg L?1), and mixture of TiO2 NPs (10.0?mg L?1)?+?CuO NPs (2.5 and 5.0 mg?L?1) for two periods of exposure (10 and 20 days) and recovery (30 and 40 days). The most common histopathological anomalies in the gill of common carp such as hyperplasia, oedema, curvature, fusion, aneurism, and necrosis were observed. The synergistic effect of co-existing TiO2 NPs and CuO NPs reduced the length of secondary lamella and increased the diameters of the gill filaments and secondary lamellae. Moreover, the presence of TiO2 NPs increased the CuO NPs effects on the histopathological anomalies of intestine tissue and the synergistic effect of TiO2 NPs and CuO mixture leads to an increase in the severity of histopathological lesions such as degeneration, swelling of goblet cells, and necrosis - erosion in the intestine tissue. In conclusion, the presence of TiO2 NPs increased the toxicity of CuO NPs.  相似文献

With juvenile fish as the subject, the effects of low concentration ammonia on antioxidant system were studied using Mugil cephalus. Samples of gill and liver tissue were obtained from 0.35, 0.70, 1.5 and 3?mg/L ammonia groups at 0, 5, 10, 15 and 20 days of exposure, at which times the biomarkers were measured. Results showed that gill malondialdehyde (MDA) content exhibited an initial significant increase (p?≤?0.05) at unionised ammonia concentrations of 0.70, 1.5 and 3.0?mg/L on day 5, followed by subsequent declines, while liver MDA levels exhibited significant increases (p?≤?0.05) at unionised ammonia concentrations of 1.5?mg/L starting on day 10 and at 3.0?mg/L starting on day 5. With exposure to ammonia at different concentrations, Na+-K+-ATPase activity in liver and gill decreased over time. The Na+-K+-ATPase activity was negatively related to ammonia concentration from 0.70 to 3.0?mg/L. Overall, our results show that MDA and Na+-K+-ATPase, evaluated here as potential biomarkers of ammonia exposure, exhibited responses to sublethal concentrations of ammonia that were concentration dependent.  相似文献

Concentrations of organochlorine (OC) contaminants and histomorphology of liver, kidney, and thyroid tissues were studied in nine adult and one subadult glaucous gulls (Larus hyperboreus) collected at Svalbard on 2 August 2011. Concentrations of liver polychlorinated biphenyls (PCB; range: 150–2820?ng?g?1 ww), dichlorodiphenyltrichloroethane (DDT; range: 58–724?ng?g?1 ww), and chlordanes (CHL; range: 11–126?ng?g?1 ww) dominated the OC profile followed by hexachlorobenzene (HCB; range: 11–42?ng?g?1 ww), mirex (range: 2–52?ng?g?1 ww), and β-hexachlorocyclohexane (β-HCH; range: 1–7?ng?g?1 ww). Histological examination of the liver showed mononuclear cell infiltrations and granulomas in 10 and 6 gulls, respectively, while intense intrahepatic lipid accumulation (steatosis) was found in two and focal necrosis in one gull. In kidney, glomerular sclerosis and adhesions was found in five and one gull, respectively. Thickening of the glomerular basement membranes and tubular necrosis was found in four and seven gulls, respectively, while mononuclear cell infiltrations were found in two individuals. In the thyroid gland, a high density of small follicles accompanied by follicular epithelial cell proliferation was observed in five glaucous gulls. Gulls with hepatic steatosis had significantly higher ΣDDT levels than the gulls without hepatic steatosis and a similar trend was found for ΣPCB. When normalizing OC concentrations for lipid content in liver, gulls with lipid granulomas had significantly lower β-HCH and significantly higher mirex levels, respectively, than gulls without lipid granulomas. Also; gulls with thickening of the glomerular basement membranes had non-significantly higher ΣPCB levels than gulls without. The histological findings were similar to those of controlled laboratory studies and OC-contaminated wildlife (e.g., polar bears; Ursus maritimus) and the data of this study therefore suggest that OC exposure may be a co-factor in the development of organ alterations in glaucous gulls. However, other environmental factors such as age, element exposure, and infectious micropathogens cannot be ruled out as co-factors, and it is uncertain if the tissue changes found exert adverse health effects on the population of Svalbard glaucous gulls.  相似文献

Abstract

For many years, the sculpin has been utilized as a sentinel monitoring species for anthropogenic impacts on the marine environment. To further develop its potential as a screening model body burden of several trace elements, including cadmium (Cd), copper (Cu), mercury (Hg), lead (Pb) and selenium (Se), were investigated. In addition, histopathology in shorthorn sculpins (Myoxocephalus scorpius; n?=?20) and fourhorn sculpins (Myoxocephalus quadricornis; n?=?10) were examined at three sites around the Scoresby Sound settlement in Central East Greenland. Fourhorn sculpins caught at the most distant site from the settlement, contained significantly higher hepatic Cu and Cd concentrations as well as significantly elevated gill Pb levels compared to the shorthorn sculpins collected at two other sites closer to the settlement. Histological examinations showed that fish with significantly higher concentrations of hepatic Cd and Se exhibited greater nuclear alterations, interstitial hyperplasia/hypertrophy, interstitial mononuclear infiltrations and granulomas. Further, fish with higher Cd and Cu gill tissue levels displayed a significantly higher number of cytoplasmic alterations and lamellar epithelium lifting, hypertrophic and hyperplastic epithelium along with mucus cell hyperplasia. While the presence of liver lesions were not species or sex-specific, the presence of gill lesions decreased in the order female fourhorn sculpins?>?female shorthorn sculpins?>?male fourhorn sculpins?>?male shorthorn sculpins. Hepatic Hg concentrations exceeded known lowest observable effect doses (LOED) for fish (0.1–0.5?μg g?1 ww) in 27% of fish, while liver Cd residues in 80% exceeded LOED (0.42–1.8?μg g?1 ww). Based upon these results, data suggest that using the sculpin as a valuable sentinel fish species histopathology may serve as a reliable tool for assessing marine ecosystem exposure to trace metals. However, confounding physiological and ecological factors also need to be considered.  相似文献

Responses of gill ATPase and liver esterase of topmouth gudgeon, Pseudorasobora parva under sublethal exposure to glyphosate (WSC 41% as isopropyl amine salt) at 1.0, 5.0 and 25?mg/L and metsulfuron methyl (technical 98.2%) at 0.0095, 0.085 and 0.85?mg/L of water were measured at 8th, 16th, 24th and 65th days of exposure. Two way analysis of variance (ANOVA) followed by least significant difference (LSD) test and parameter estimates indicated a significant inhibitory effect on gill ATPase activity (max. 57%) by metsulfuron methyl and liver esterase activity by glyphosate (max. 43%) as compared to the control, but the differences in the residual activities among the concentration levels of the herbicides were not significant. The activities changed significantly with the sampling times except gill ATPase activity under metsulfuron methyl exposure. In most of the cases, the maximum inhibitory effect on the enzymes was recorded on the 8th day and over-recovering appeared with time.  相似文献

为探究褐牙鲆(Paralichthys olivaceus)幼鱼抗氧化系统酶活力及相关基因表达在悬浮物胁迫下的变化情况,设计了浓度为5 000、10 000 mg·L-1悬浮物水体对褐牙鲆(14.53 cm±1.58 cm)肌肉、肝脏、鳃及血液总超氧化物歧化酶(T-SOD)、谷胱甘肽转移酶(GST)、过氧化氢酶(CAT)酶活力及SOD2、GST、CAT的mRNA转录水平表达变化情况。结果显示:在悬浮物胁迫24 h或者48 h时,4种组织3种酶活力具有升高的趋势(P0.05);4种组织的T-SOD酶活力在96 h均高于对照组(P0.05),肌肉、鳃丝及血液中SOD2基因相对表达量96 h实验组低于对照组(P0.05);肌肉中CAT酶活力呈现先升高再降低趋势,鳃中CAT基因相对表达量呈现先降低、再升高、再降低的趋势;肝脏、鳃、血液中GST酶活力在悬浮物胁迫下呈现升高趋势,4种组织GST基因相对表达量在12~24 h时呈现升高趋势。研究表明:悬浮物对褐牙鲆抗氧化酶活力及相关基因的表达具有一定的影响,血液中3种酶活力变化幅度最大,鳃中3种基因相对表达水平变化幅度最大,这与血液及鳃参与呼吸作用过程有关;抗氧化酶活性及基因相对表达变化趋势并不完全一致。本研究可为揭示褐牙鲆应对悬浮物胁迫的耐受机制及褐牙鲆耐悬浮物品种选育提供基础数据。  相似文献

Adult males of the quail Coturnix coturnix were exposed to lead acetate trihydrate in drinking water (0.1, 0.25, 0.5, and 1% for one–six months) to investigate histological and histochemical alterations induced by lead intoxication in the liver. Chronic exposure to subtoxic concentrations of lead produced changes in hepatocytes, portal veins, Kupffer cells, and blood sinusoids. Alterations in hepatocytes mainly involved cytoplasmic vacuolation, necrosis, cytolysis, and glycogen accumulation. Kupffer cell hyperplasia, hemosiderosis, blood sinusoidal dilatation, portal vein congestion, and edema were also observed. No fibrosis or cirrhosis in the liver of any member of the dose groups over the entire period of the study was noted. The findings revealed that chronic exposure to lead produced mild histological and histochemical changes in the liver of the quail Coturnix coturnix.  相似文献

采用不同质量浓度的溴氰菊酯(0.0070 mg·L~(-1)、0.014 mg·L~(-1)、0.020 mg·L~(-1)、0.027 mg·L~(-1))对菲律宾蛤仔进行20 d半静置染毒,测定不同时间淋巴液中乙酰胆碱酯酶(ACh E)和钠离子-钾离子-三磷酸腺苷酶(Na~+-K~+-ATPase)活性、鳃和肝脏中谷胱甘肽转硫酶(GST)活性的变化,并观察染毒20 d后鳃丝组织和消化盲囊组织的损伤情况。酶活性分析结果显示,与对照组相比,低浓度组(0.0070 mg·L~(-1))试验期间酶活性均无显著差异(P0.05);中浓度组(0.014 mg·L~(-1)、0.020 mg·L~(-1))淋巴液中ACh E和Na~+-K~+-ATPase均呈先激活后抑制的变化规律(P0.05),鳃和肝脏中GST活性均呈上升趋势(P0.05);高浓度组(0.027mg·L~(-1))淋巴液中ACh E和Na~+-K~+-ATPase、肝脏中GST活性在试验期间持续下降(P0.01),而鳃中GST活性呈先抑制后升高的趋势(P0.05)。研究表明,低中浓度的溴氰菊酯对菲律宾蛤仔体内的酶活性表现为先诱导后抑制,具有明显的时间、剂量效应;高浓度的溴氰菊酯对菲律宾蛤仔体内酶活持续抑制,且染毒浓度越高,组织细胞变异越显著,表现为鳃丝上皮细胞纤毛层萎缩、纤毛脱落,消化盲囊上皮细胞膨胀,出现包涵体样结构。  相似文献

This study elucidated the protective role of alga against mercury (Hg)-induced toxicity in marine fish. Bath immersion with mercury chloride (HgCl) [0.125 and 0.25?ppm] in combination with the algal extract (3?ppm) to Therapon jarbua offered a significant protection against Hg only induced disturbed liver function, damaged histoarchitecture, elevated oxidative stress, and DNA fragmentation of tissues. Mercury exposure decreased hepatic superoxide dismutase (SOD), catalase (CAT) activities, and the level of nonprotein-soluble thiol (NPSH, GSH), with a concomitant increase in thiobarbituric acid reactive substances (TBARS) in the gill, kidneys, liver, and blood. Algal supplementation diminished the rise in TBARS restoring CAT, SOD, and GSH levels to control. Reduced generation of free radicals may be correlated to protect DNA stability and morphology. This study indicates the ability of alga to ameliorate Hg-mediated injuries. In conclusion, Kappaphycus alvarezii extract did not show any toxicity and its safety is suggestive for using as a supplement in fish food.  相似文献

In the present study, an attempt has been made to quantify the fenvalerate accumulated in different tissues (gill, muscle and liver) and observe changes involved in the levels of sodium, potassium and calcium ions and Na+–K+, Mg2+ and Ca2+ adenosine triphosphatase (ATPase) activities in the freshwater fish, Cirrhinus mrigala on short-term and long-term exposure to the median lethal and sublethal concentration of fenvalerate. Residue analysis using gas–liquid chromatography (GLC) technique revealed that fenvalerate accumulated in highest quantity in gill followed by liver and muscle under median lethal concentration (6?µg?L?1). Whereas in sublethal concentration (0.6?µg?L?1), muscle accumulated highest quantity followed by gill and liver, which might be due to the fact that fenvalerate is highly lyphophilic. The ion concentration and ATPase activity were found effected in fish exposed to lethal and sublethal concentrations of fenvalerate. Concentration of Na+, K+ and Ca2+ ions decreased in gill, muscle and liver on being exposed to median lethal concentration to a significant level. Whereas the changes were not highly pronounced at sub lethal level indicating low concentration of fenvalerate and its non-toxic effect at chronic exposure. Na+–K+, Mg2+ and Ca2+ ATPases activity were also found decreased in correspondence to the ionic change under median lethal and sub lethal concentrations in target tissues. This might have lead to behavioural changes and create wide-spread disturbance in the normal physiology, ultimately causing the death of the fish. The results suggest that in biomonitoring programmes, ions and associated ATPases can be a good diagnostic tool for fenvalerate toxicity.  相似文献

Scanning electron microscopic (SEM) study of gills of Catla catla catla (17-day-old) exposed to UV-B radiation (145?µW?cm?2 at the water surface) for three different exposure times: 5, 10 and 15?min was conducted. Fish without UV-B exposure served as control. UV-B radiation damaged both gill filaments and lamellae. The intensity of damage was minimal in 5?min exposed fish, followed by 10?min exposed fish and maximal in 15?min. The gill epithelium was severely damaged in 15?min irradiated fish compared to control. Pavement cells (PVCs) were damaged and the numbers of microridges within PVCs decreased. The deep boundary of PVC was not clear. In some area of gill epithelium, PVCs were destroyed and mitochondrion-rich cells (MRCs) were exposed. The 5?min exposure reduced the number of microridges in the PVCs, but the boundary of PVCs was still visible. MRCs in the gill epithelium were not exposed in 5?min exposed fish. The damage to PVCs and subsequent exposure of MRCs in UV-B irradiated fish may hamper respiratory functions and disturb osmoregulation in catla.  相似文献

The aim of this study was to determine the bioavailability and adverse effects of cadmium (Cd) and copper (Cu) on hibernating Egyptian toads and whether ascorbic acid (vitamin C) blocked Cd- and Cu-induced effects during hibernation. The oxidative status of liver, kidney, and intestine of Bufo regularis to Cd, Cu, and/or a combination of both metals administered orally for 2 weeks was determined. In the protection studies, vitamin C was given for 1?h prior to administration of Cu, Cd, and/or metal combination for 2 weeks. Treatment with Cu, Cd, and a combination of both metals produced a reduction in red blood count cells and hemoglobin content, while white blood count cells showed an increase in numbers during these treatments. After 2 weeks exposure, Cd and Cu increased significantly in all the tissues studied. Cu storage presented the following sequence: liver?>?intestine?>?kidney. Cd storage presented the following sequence: intestine?>?kidney?>?liver. When exposed to both metals, Cu and Cd storage presented the following sequence: liver?>?intestine?>?kidney. Histopathological examination of the liver revealed marked alterations including loss of hepatic cell architecture, and some cells exhibited distinct cytoplasmic vacuoles. The majority of blood vessels exhibited a marked dilatation and congestion with infiltration of blood cells, prominent hyperemia of hepatic veins, and significant proliferation of bile ductules. Histopathological changes in the kidney showed destruction and degeneration of both renal tubule cells and glomerular with infiltration of leukocytes and inflammatory cells. Histopathological alterations in the intestine were restricted to the innermost mucosal epithelium with marked degeneration of the villi and submucosa and an extensive fragmentation of mucosal epithelium as well as atrophy of goblet cells. The administration of vitamin C 1?h prior to administration of Cd, Cu, and metal combination did not protect against hepatic, renal, and intestinal damage. However, parental vitamin C given alone increased tissue toxicity.  相似文献

The objective of the present study was to investigate the levels of Cd, Pb, Co, and Cu, in A. chukar, A. griseogularis, and Columba livia, in order to (1) examine the age- and gender-related variation in trace metal accumulation and (2) to determine the significance between metal concentrations in the kidney, liver, and pectoral muscle. Mean concentrations of Cd and Pb in the kidney of A. chukar, A. griseogularis, and C. livia were 3.7, 4.1, and 3.9?µg/g and 15.9, 13.6, and 15.5?µg/g, respectively. In the liver, they were 4.8, 4.3, and 3.9?µg/g and 21.4, 21.3, and 21.1?µg/g, and in the pectoral muscle, 2.3, 2.3, and 2.2?µg/g and 7.1, 7.1, and 7.8?µg/g, respectively. Metal concentrations in three bird species were decreased in the sequence of liver?>?kidney?>?pectoral muscle. Trace metal concentrations in the three species were higher in females than in males. The mean concentrations of Cd in the kidney and liver were higher than the background levels, as well as Pb concentrations in the liver were higher than the toxic level.  相似文献

The activity of alanine aminotransferase (ALAT) and aspartate aminotransferase (AAT) of different tissues of fingerlings of Labeo rohita under the influence of two effluents has been studied. The alanine aminotransferase activity was increased over the control in different exposed periods of tannery and distillery effluent treatments. The alanine aminotransferase in the liver showed increased activity at different periods than that of the muscle, kidney, gill and brain (p < 0.001) (60.09%) over the control during the 40 days exposure in both the effluents treatments. The increased activity of alanine aminotransferase was highly significant (p > 0.001) in all the tissue in tannery and distillery effluents treatments. Similarly aspartate aminotransferase activity was increased over the control in all the treated tissues from 10 to 40 days exposure. But this increase, was not significant in the muscle tissue in distillery and tannery treatements after 10 days exposure.
