The aim of the present study was to evaluate the possible protective effects of thymoquinone (TQ), an antioxidant agent, against imidacloprid (IMI)-induced oxidative stress in male and female mice. In total, 48 Swiss Albino male and female mice were fed a standard rodent diet and divided into 3 equal groups: the animals in the control group (vehicle treated) were given corn oil, the second group were orally administered 15 mg/kg/day IMI alone, and the third group were orally administered 15 mg/kg/day IMI and with TQ at 10 mg/kg/day for 21 days. During the experimental period, there were no significant changes between initial body weights and final body weights of IMI treated male and female mice. IMI produced significant increase in blood, liver, kidney, and heart malondialdehyde (MDA) levels and decrease in blood and liver glutathione (GSH) levels. In addition, IMI treatment decreased erythrocyte, liver, and kidney superoxide dismutase (SOD) activity in male mice and decreased erythrocyte and liver SOD activity in female mice. Erythrocyte catalase (CAT) activities were found to be low in male and female mice. However, treatment with TQ reversed IMI-induced oxidative stress, lipid peroxidation, and activities of antioxidant enzymes. Moreover, TQ exhibited protective action against the IMI-induced histopathological changes in tissues of male and female mice. In conclusion, TQ was found to be effective in protecting mice against IMI-induced oxidative stress by enhancing antioxidant defense mechanisms.  相似文献

Silver nanoparticles (AgNPs) possess properties that are important for industrial and medical applications. This study is aimed to investigate intra-peritoneal toxicity of AgNPs at 26, 52 or 78 mg/kg/day for 5 days in mice Swiss albino mice. The effects on oxidative stress markers activities of serum superoxide dismutase (SOD) and catalase (CAT), levels of serum glutathione (GSH), apoptosis (TUNEL assay), DNA strand breaks (comet assay) in lymphocytes, as well as histopathological of kidney tissue were determined. AgNPs significantly increased SOD and CAT activities reduced GSH levels. In kidney apoptosis (TUNEL assay) while DNA strand breaks (comet assay) in lymphocytes revealed that AgNPs at concentration 78 mg/kg produced significant apoptosis and DNA damage. AgNPs also produced associated histological renal tissue damage. Evidence suggests that AgNPs-mediated alterations may be attributed to oxidative stress.  相似文献

四环素类抗生素(TCs)是目前我国应用广泛、用量最大的一类抗生素,畜禽粪便和土壤中存在TCs残留,影响蔬菜作物的生长发育. TCs因水溶性较高更容易被植物转运和积累,植物对TCs耐受性机理研究仍不足.为更全面探究土壤TCs对蔬菜的毒性作用,研究不同浓度四环素(TC)和土霉素(OTC)分别对生菜的抗生素残留、生长特征及抗氧化酶系统的影响.结果显示,在0(对照)、2、10、50、250 mg/kg 5个施用水平下,生菜叶片抗生素含量逐渐增加,且土霉素含量始终大于四环素含量.与对照相比,抗生素浓度在50 mg/kg以上时对生菜生长具有显著抑制作用,其中,株高、根长、地上部和地下部鲜重与叶片TC残留量具显著负相关.生菜叶片的脯氨酸含量随浓度增加呈先增加后降低的趋势,在10 mg/kg时达到最大.并且低浓度(2 mg/kg)促进抗氧化酶基因SOD、POD21和CAT的表达,高浓度抗生素(50、250 mg/kg)对其产生抑制作用,10 mg/kg的抗生素处理对SOD、POD21和CAT基因表达的影响在抗生素种类上存在差异.本研究表明抗生素浓度超过50 mg/kg对生菜生长产生抑制作用,脯氨酸和抗氧化酶SOD、POD、CAT的转录水平及其酶活性能快速响应抗生素胁迫,可作为生菜对抗生素抗性的辅助评价指标.(图8表3参19)  相似文献

Cadmium (Cd) is a toxic-heavy metal that induces a wide range of behavioural, biochemical and physiological effects in aquatic organisms. Oxidative damage has been proposed as a possible mechanism involved in cadmium toxicity. The current study was carried out to evaluate the antioxidant activity of Spirulina as feed additive (1?mg/L) against the toxicity of cadmium (Cd) 0.5?mg/L in freshwater mussel Unio ravoisieri. At the end of the exposed period of 4 days, digestive gland antioxidant status Superoxide dismutase, Catalase, Glutathione-S-transferase and damage markers such as Malondialdehyde and Protein carbonyl were determined. Associations between biomarkers were assessed by a multivariate analysis technique, principal component analysis (PCA). The results of this study revealed that digestive gland antioxidant status showed a significant decrease when mussels were exposed to Cd. 氟虫双酰胺对蚯蚓的生化毒性与细胞毒性研究   总被引:1,自引:0,他引:1  
双酰胺类杀虫剂已成为全世界第4大类最常用的杀虫剂,具有非常广阔的应用前景。然而,目前关于双酰胺类杀虫剂生态毒性评估方面的研究还比较少。为探究双酰胺类杀虫剂对非靶标生物的毒性作用,选取赤子爱胜蚓(Eisenia fetida)为受试生物,研究了典型双酰胺类杀虫剂氟虫双酰胺对非靶标动物蚯蚓的生化毒性和细胞毒性以及其在人工土和蚯蚓体内的浓度变化情况。结果表明,氟虫双酰胺在人工土壤中十分稳定,在整个暴露期间氟虫双酰胺的浓度变化不超过20%。氟虫双酰胺在蚯蚓体内的含量随染毒浓度的升高和暴露时间的推移而增加,呈明显的时间和剂量-效应关系;在染毒浓度为0.1和1.0 mg·kg-1的处理组中,氟虫双酰胺未对蚯蚓产生明显的氧化胁迫效应。在染毒浓度为5.0和10.0 mg·kg-1的处理组中,蚯蚓体内活性氧(ROS)含量显著高于其他处理组,过量的ROS诱导蚯蚓体内各种抗氧化酶活性发生异常变化,并在蚯蚓体内造成了脂质过氧化、蛋白质羰基化和DNA损伤。研究表明,当土壤中氟虫双酰胺的浓度为5.0和10.0 mg·kg-1时可能会对蚯蚓产生很高的风险。此外,彗星实验对氟虫双酰胺诱导的氧化胁迫较为敏感,可以作为敏感生物标志物对氟虫双酰胺造成的土壤污染进行预警。  相似文献

采用急性和28 d慢性暴露试验研究了海洋典型危险化学品对二甲苯对褐牙鲆幼鱼的致死效应和生长抑制效应,并测定了遗传毒性、神经毒性以及免疫毒性效应相关的毒理学评价指标。研究结果表明:对二甲苯对褐牙鲆幼鱼的96 h-LC50为45.7 mg·L~(-1),根据国家环保局水和废水监测分析方法编委会规定的化学物质对鱼类毒性分级标准,属于中等毒性,浓度高于2.3 mg·L~(-1)的暴露能显著抑制褐牙鲆幼鱼的生长;4.6和9.2 mg·L~(-1)浓度组观察到对二甲苯暴露28 d后褐牙鲆肝脏中的丙二醛含量与DNA损伤程度显著升高,脑组织中乙酰胆碱酯酶(Ach E)活力被明显抑制;9.2 mg·L~(-1)浓度组暴露28 d后,褐牙鲆幼鱼体内总血细胞数量和溶菌酶活力显著降低。这些结果表明较高浓度的对二甲苯长期作用可导致褐牙鲆幼鱼的氧化胁迫,造成其肝脏毒性损伤并产生神经毒性与免疫毒性。本实验结果为了解对二甲苯对鱼类的毒性作用机制以及对二甲苯的海洋生态风险评估提供了科学依据。  相似文献

The protective effect of Spirulina platensis (SP) powder against aluminium-induced nephrotoxicity and DNA damage in rats was studied. Male rats receiving daily 40 mg/kg b.wt. aluminium chloride (AlCl3) orally had increased serum levels of urea and creatinine, up regulated kidney injury molecule-1 and tissue inhibitor of metalloproteinase-1 genes, down regulated catalase and glutathione peroxidase genes, and increased all parameters of kidney DNA damage using comet assay. Treatment with SP alleviated all AlCl3-induced effects of toxicity, especially when the animals were pre-treated.  相似文献

Feeding deltamethrin-contaminated grains to domestic poultry, such as quails may result in toxic effects in these birds. This study was done to investigate the effects of recommended doses of deltamethrin, sometimes used in grain storage silos, on Japanese quail (Coturnix japonica). Quails were fed grains contaminated with 0.25 and 0.50 mg deltamethrin per kg diet for 21 days and the effects on survival and blood biochemical parameters were studied. Plasma uric acid, creatinine levels, and creatinine phosphokinase activity in the blood were increased. Aspartate aminotransferase and lactate dehydrogenase activities and glucose levels significantly increased in birds treated with the high dose of deltamethrin. Alanine aminotransferase activity and albumin or cholesterol levels were not changed, and acetylcholinesterase and alkaline phosphatase activities, total protein and globulin in plasma were decreased. Administration of 0.25 mg/kg deltamethrin caused increased blood triglyceride levels, 0.50 mg/kg deltamethrin decreased triglyceride levels.  相似文献

Radical scavenging activity of ethanolic extract of Trianthema triquetra root was investigated against CCl4 in rats. The rats were treated with T. triquetra (100 mg, 200 mg/kg b.w.) for a period of 7 days. Antihepatotoxic effect was studied by assaying the activities of thiobarbituric acid (TBARS), reduced glutathione (GSH), glutathione peroxidase (GPx),catalase (CAT), super oxide dismutase (SOD) and vitamin C (Vit. C). Lipid peroxidation is evidenced by an increase in the value of TBARS and also a distinct diminution in the level of GSH, Vit. C at 200 mg/kg b.w. The activity of antioxidant enzymes, such as GPx, CAT SOD and Vit. E is significantly recovered towards an almost normal level in animals coadministrated with T. triquetra. The maximum protection against CCl4 induced hepatic injury was afforded by the dose of 200 mg/kg b.w. of Trianthema triquetra.  相似文献

The aim of the present study was to evaluate the hepatoprotective activity of 1-(4-(dimethylamino)benzylidene)-5-(2-oxoindolin-3-ylidene) thiocarbohydrazone, a novel isatin derivative against carbon tetrachloride (CCl4)-induced hepatotoxicity in rats. Hepatic damage was induced by administration of CCl4 (1 ml/kg, b.w., p.o.) in combination with liquid paraffin (1:1) as a single dose. The hepatotoxic rats were treated with test compound at doses of 50 or 100 mg/kg for three days and liver damage biomarkers, including activities of serum glutamate pyruvate transaminase (SGPT), serum glutamate oxaloacetate transaminase (SGOT), serum alkaline phosphatase (ALP), and levels of total serum bilirubin (TB) measured in blood samples. Results demonstrated that treatment with test compound at doses of 50 or 100 mg/kg to hepatotoxic rats produced a significant dose-dependent reduction of elevated SGOT, SGPT, ALP activities and TB levels indicating a hepatoprotective effect that was confirmed by histopathological examination of liver tissues. The study results confirmed the hepatoprotective activity of 1-(4-(dimethylamino)benzylidene)-5-(2-oxoindolin-3-ylidene) thiocarbohydrazone in rats.  相似文献

Abstract

The purpose of this study was to investigate the protective effects of zinc nanoparticles against cisplatin-induced testicular toxicity in mice. Zinc nanoparticles were produced by microwave-assisted synthesis using Lavandula vera extract as reducing agent. Single doses of cisplatin (7?mg/kg, intraperitoneally) and ZnSO4 (10?mg/kg, orally), and various doses of zinc nanoparticles (10???50?mg/kg, orally) and vitamin E (100?mg/kg, interaperitoneally) were administered. The protective role of zinc nanoparticles was determined biochemically and histologically. Gradual reduction in malondialdehyde levels and elevation in glutathione levels and in the activities of superoxide dismutase and catalase upon administration of zinc nanoparticles were observed. The pathology of mice treated with cisplatin/vitamin E and cisplatin/zinc nanoparticles were apparently equal, but vitamin E treatment was more effective in lowering oxidative stress markers than zinc nanoparticles. These findings suggest that co-administration of zinc nanoparticles with cisplatin could prevent adverse effects on the male reproductive system via their potential antioxidant properties.  相似文献

Abstract

The in vivo genotoxic potential of bisphenol A using the comet assay in mice and in human sperm cells in vitro without metabolizing enzymes was studied. Male mice were exposed by oral gavage to the following doses of bisphenol A (0 125, 250 and 500?mg/kg body weight). DNA damage was investigated in liver, kidney, testes, urinary bladder, colon and lungs cells. In testicular cells, a significant increase in DNA strand breaks was observed in the lowest, but not in the medium or highest dose groups. Histopathological investigation of the testicular samples did not show any treatment dose-related effects. No DNA strand breaks were observed in any of the other investigated tissues. In human sperm cells in vitro, bisphenol A did not induce DNA strand breaks.  相似文献

Metal pollution produces damage to marine organisms at the cellular level possibly leading to ecological imbalance. The present investigation focused on the acute and chronic toxicity of lead (Pb) and zinc (Zn) by examining the effects of biomarker enzymes in post-larvae of Penaeus monodon (Tiger prawn). Antioxidant biomarker responses such as lipid peroxidation (LPO) and catalase (CAT) activity for Pb and Zn were determined following chronic exposure. Acute Lethal Concentration 50 (LC50) values observed in the study at 96?h for Pb and Zn at 5.77?±?0.32?mg?L?1 and 3.02?±?0.82?mg?L?1, respectively. The estimated No Observed Effect Concentration and Lowest Observed Effect Concentration values for Pb were 0.014 and 0.029?mg?L?1 and that recorded for Zn was 0.011 and 0.022?mg?L?1, respectively. Among the two metals studied, toxicity of Zn was found to be greater to P. monodon than Pb. The activities of antioxidant defense enzymes and total protein content differed significantly from control following exposure to both metals. Overall, the biomarker studies demonstrated that alterations in antioxidant enzymes and induction of LPO reflect the consequences of heavy metal exposure in P. monodon.  相似文献

Hydrogen sulfide (H2S) is an endogenous gaseous molecule, functioning as a physiological regulator. This study aimed to investigate the role of H2S in atherosclerosis. Rabbits receiving high cholesterol were given atorvastatin (10 mg/kg/day, orally) or sodium sulfide (1.1 mg/kg/day, intramuscular) 2 weeks after commencement of high-cholesterol diet and continued for 4 weeks. Serum lipid profiles, lipid peroxidation indexed as malondialdehyde, glutathione, superoxide dismutase, the vascular adhesion molecule-1, and aortic vascular endothelial growth factor were determined. Rabbits receiving a high-cholesterol diet showed elevated levels of total serum cholesterol, triglycerides, low-density lipoprotein cholesterol, malondialdehyde and vascular adhesion molecule-1, and aortic vascular endothelial growth factor expression and lower levels of high-density lipoprotein cholesterol, glutathione, and superoxide dismutase. Treatment with atorvastatin and sodium sulfide has beneficial effects on serum lipid profile, oxidative damage, vascular adhesion molecule-1, and aortic expression of vascular endothelial growth factor.  相似文献

以紫背浮萍为实验对象,探究双酚A(bsiphenol A, BPA)对其生长、光合色素含量、抗氧化系统和渗透压调节物质的影响。结果表明,不同浓度(0、1、5、20和50 mg·L~(-1))BPA连续暴露7 d后,与对照组相比,随暴露浓度升高,紫背浮萍湿重和叶绿素含量呈现先显著增加后降低的趋势;暴露组过氧化氢(H_2O_2)含量和抗超氧阴离子自由基活性显著降低;丙二醛(MDA)含量在20和50 mg·L~(-1)组显著升高;暴露组抗氧化酶活性包括超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)、抗坏血酸过氧化物酶(APX)、谷胱甘肽还原酶(GR)和总抗氧化能力(T-AOC)均低于对照组;谷胱甘肽(GSH)含量在高浓度组显著升高,20 mg·L~(-1)组可溶性糖含量显著升高,可溶性蛋白含量在中高浓度组显著增加。研究表明,中低浓度BPA会提高紫背浮萍光合色素含量,有机物积累增多,促进其生长;但随BPA浓度不断升高,紫背浮萍体内产生氧化损伤,抗氧化酶活性及叶绿素含量降低,生长受到抑制。本研究结果将为BPA污染控制及对水生植物生态毒性效应评价提供理论基础。  相似文献

Permethrin is a common insecticide that does not show genotoxic potential in standard in vitro and in vivo assays. To investigate the genotoxic potential of permethrin in more detail, two in vivo studies were conducted on female mice to assess DNA damage in tumor target organs by the comet assay and micronucleus test. For this, mice were administered permethrin at doses of 150, 300, or 600 mg/kg/day by gavage for 2 days, and their lung, liver, glandular stomach, peripheral blood, and bone marrow cells were examined for DNA damage. There were no significant increases in % tail DNA in the organs examined and no increase in micronuclei in peripheral blood by flow cytometry. Taken together, the present findings provide evidence that permethrin has no genotoxic, aneugenic, or clastogenic potential.  相似文献

Myocardial infarction was produced by subcutaneous administration of isoproterenol hydrochloride (85 mg/kg b.w. for two consecutive days). The myocardial damage was proved by observing increase in the activity of SGOT and SGPT in serum whereas AChE activity was inhibited by increasing Km, without affecting Vmax. The inhibition of AChE and inhibitory kinetic may be useful in the diagnosis and management of salvage of myocardium.  相似文献

The effects of different pesticides (propiconazole, profenofos, pretilachlor) on vermicomposting were evaluated for the adverse effects on soil enzyme activities (dehydrogenase, phosphatase, urease) and total microbial counts (TMC). There were remarkable increase in enzyme activities and TMC in presence of earthworm compared to control (earthworm absent). In comparison to control, the activities of phosphatase, dehydrogenase and urease in presence of vermicompost increased upto 30, 128 and 31.3% respectively; whereas increase of TMC was 71.9%. But, in presence of each of the pesticide, said activities decreased. Maximum inhibition of soil phosphatase activity (46.6%) was observed in presence of propiconazole (100 mg kg(-1)) after 120 days. Profenofos affected the soil dehydrogenase activity in the tune of 47% at 1000 mg kg(-1) concentration after 80 days and thereafter, the extent of toxicity decreased little. Soil urease activity was affected markedly in presence of profenofos and was 62% at 1000 mg kg(-1) level after 80 days. TMC also declined in presence of profenofos and pretilachlor. Increase in TMC was about 71.9% compared to control and the inhibition was more or less 60% when profenofos (1000 mg kg(-1)) was present even after 120 days of treatment.  相似文献
