胞外聚合物(extracellular polymeric substance, EPS)是污水处理系统内微生物聚集体的重要组成部分,其组分及含量与微生物聚集体的结构、功能和表面特性息息相关。基于已有研究成果,系统综述了EPS在维持微生物聚集体结构组成、强化其功能活性中发挥的作用,进一步从EPS对微生物聚集体的传质性能、亲/疏水性、物化稳定性、流变性能和脱水性能的调控作用,探究了EPS影响微生物聚集体功能活性的行为机理,旨在为改善污水生物聚集体的聚集性能、提高泥水分离效率、强化水处理能力提供理论基础和科学依据。  相似文献

Humin, the most recalcitrant and abundant organic fraction of soils and of sediments, is a significant contributor to the stable carbon pool in soils and is important for the global carbon budget. It has significant resistance to transformations by microorganisms. Based on the classical operational definition, humin can include any humic-type substance that is not soluble in water at any pH. We demonstrate in this study how sequential exhaustive extractions with 0.1 M sodium hydroxide (NaOH) + 6 M urea, followed by dimethylsulphoxide (DMSO) + 6% (v/v) sulphuric acid (H2SO4) solvent systems, can extract 70–80% of the residual materials remaining after prior exhaustive extractions in neutral and aqueous basic media. Solid-state 13C NMR spectra have shown that the components isolated in the base + urea system were compositionally similar to the humic and fulvic acid fractions isolated at pH 12.6 in the aqueous media. The NMR spectra indicated that the major components isolated in the DMSO + H2SO4 medium had aliphatic hydrocarbon associated with carboxyl functionalities and with lesser amounts of carbohydrate and peptide and minor amounts of lignin-derived components. The major components will have significant contributions from long-chain fatty acids, waxes, to cuticular materials. The isolates in the DMSO + H2SO4 medium were compositionally similar to the organic components that resisted solvation and remained associated with the soil clays. It is concluded that the base + urea system released humic and fulvic acids held by hydrogen bonding or by entrapment within the humin matrix. The recalcitrant humin materials extracted in DMSO + H2SO4 are largely biological molecules (from plants and the soil microbial population) that are likely to be protected from degradation by their hydrophobic moieties and by sorption on the soil clays. Thus, the major components of humin do not satisfy the classical definitions for humic substances which emphasise that these arise from microbial or chemical transformations in soils of the components of organic debris.  相似文献

Immigrant bacteria located on leaf surfaces are important to the health of plants as well as to people who consume fresh fruits and vegetables. However, the spatial distribution and organization of these immigrant bacteria on leaf surfaces are still poorly understood. To examine the spatial organization of these strains, two bacterial strains on tobacco leaves: (1) an indigenous strain, Pseudomonas stutzeri Nov. Y2011 labeled with green fluorescent protein, and (2) an immigrant strain Pantoea agglomerans labeled with cyan fluorescent protein isolated from pear, were studied. Under moist conditions, P. agglomerans cells quickly disappeared from direct observation by laser- scanning confocal microscopy, although elution results indicated that large amounts of live cells were still present on the leaves. Following exposure to desiccation stress, particles of cyan fluorescent protein-labeled P. agglomerans were visible within cracked aggregates of P. stutzeri Nov. Y2011. Detailed observation of sectioned aggregates showed that colonies of immigrant P. agglomerans were embedded within aggregates of P. stutzeri Nov. Y2011. Furthermore, carbon-resource partitioning studies suggested that these two species could coexist without significant nutritional competition. This is the first observation of an immigrant bacterium embedding within aggregates of indigenous bacteria on leaves to evade harsh conditions in the phyllosphere.  相似文献

All known cosmic and geological conditions and laws of chemistry and thermodynamics allow that complex organic matter could have formed spontaneously on pristine planet Earth about 4,000 mya. Simple gasses and minerals on the surface and in oceans of the early Earth reacted and were eventually organized in supramolecular aggregates and enveloped cells that evolved into primitive forms of life. Chemical evolution, which preceded all species of extant organisms, is a fact. In this review, we have concentrated on experimental and theoretical research published over the last two decades, which has added a wealth of new details and helped to close gaps in our previous understanding of this multifaceted field. Recent exciting progress in the molecular and genetic analyses of existing life, in particular microorganisms of ancient origin, even supports the possibility that a cellular, self-reproducing common ancestor might be assembled and resurrected in anaerobic cultures at some time in the future. Charles Darwin did not, and indeed, could not, address and specify the earliest phases of life which preceded the Origin of Species. However, in a famous letter, he sketched “a warm little pond with all sorts of… (chemicals, in which) …a protein was chemically formed.” We try to trace the impact of his charming clear-sighted metaphor up to the present time.  相似文献

The paper describes a model for environmental quality and resource efficiency parameters to be used in evaluation of options for improvement of product systems in a life cycle and system perspective. Four main preventive strategies for product system improvements are described and discussed in the paper. (1) Reformulating user needs and requirements to the system; (2) improvement of product system performance; (3) substitution of the whole product systems or system components on different levels; (4) optimization within and between system units and components. These main strategies are discussed with respect to the structure of the product system (the domain theory), and with respect to efficiency in general. It is concluded that at present, it is not possible to make any definite rank of priorities between the different strategies, but that this should be evaluated when more systematic studies of cases are available.  相似文献

The water quality of the Volga catchment, Russia, is diminished and major nutrients reach critical levels. Proper management strategies depend on process knowledge about nutrient sources and pathways as well as appropriate models. For the 18.8 km2 experimental catchment Lubazhinkha, 100 km south of Moscow, a monitoring scheme was conducted to identify runoff generation and nutrient source areas. The grey forest soils of the catchment are utilized with arable land and pasture (54%) and forest areas. The received results show a high intra-annual and inter-annual dynamic of the snowmelt runoff generation as major hydrological process. Overall sediment export rates during the 3 years of snowmelt investigation from 2003 to 2005 are low with a maximum of 0.33 t ha−1 during snowmelt period 2005 and effectively reduced by a dam at the catchment outlet. Also, the total phosphorus loss from the catchment is low reaching from 0.03 to 0.35 kg ha−1 during the snowmelt periods. However, sediment and total phosphorus concentrations vary within one snowmelt event and can reach high concentrations. Detailed analysis of element concentrations during snowmelt events allows an identification of different flow components and runoff generating mechanisms. During low flow situations phosphorus and dissolved organic carbon are transferred from the arable soils with slow flow components to the channel. In contrast to this situation, the sources of sediment and nutrients split up temporarily and spatially during high flow situations. Aside arable fields that are source of sediment and phosphorus during snowmelt episodes, the forested areas were detected as major source for the export of dissolved organic carbon with surface runoff. In particular these areas are characterized by a late thawing of the topsoil which causes a delayed peak of dissolved organic carbon concentration. The awareness of the dynamic and complex catchment reaction during snowmelt events is of importance for the development of proper management strategies.  相似文献

First, an arriving axonal growth cone identifies its appropriate partner cell, creating an initial contact, and, second, specific axonal and dendritic protein components are recruited to this initial contact site, forming a functional synapse. Three cell adhesion systems have recently been shown to be specifically enriched at synaptic contacts: the cadherin/catenin system, the cadherinlike neuronal receptors, and the β-neurexin/neuroligin system. Components of all three cell adhesion systems have been localized to synaptic contacts using immunogold electron microscopy but are also present outside of synapses. The present short review discusses the possible role of these synaptic cell adhesion molecules in synaptogenesis.  相似文献

   Hopanoids are pentacyclic triterpenoid lipids occurring in bacteria. They are synthesized from isopentenyl units which are formed in a new biosynthetic route leading to isopentenyl diphosphate. Six C5 units are joined to form squalene, the immediate precursor in hopanoid synthesis. In a highly complex cyclization reaction that shares considerable similarities with that of oxidosqualene to sterols, the hopane skeleton is formed from squalene by the squalene-hopene cyclase. Recent elucidation of the X-ray structure of this membrane-bound cyclase has shed some light on the properties of this unusual enzyme. The active site is located in a cavity within the enzyme. The squalene substrate diffuses through a channel structure from the membrane into this cavity and is there transformed into hopene. Polar side chains are attached to hopene resulting in the amphiphilic molecular structure of many hopanoids. These hopanoids are membrane components involved in regulating membrane fluidity and stability. However, the many structural variants of hopanoids indicate that they may have other interesting but as yet unknown functions.  相似文献

为了揭示水盐梯度对河口湿地土壤水稳性团聚体分布及稳定性的影响,对闽江河口不同淹水环境和盐度下短叶茳芏(Cyperus malaccensis)湿地土壤水稳性团聚体进行了测定与分析.结果表明:①闽江河口半咸水湿地和淡水湿地0~30 cm土壤粉+黏团聚体、微团聚体和大团聚体的含量分别为63.12%~77.49%、6.82%~31.64%、4.38%~22.63%.除20~30 cm土层外,高潮滩0~20 cm土壤粉+黏团聚体和大团聚体含量均随盐度的增加而增加,增幅分别为8.74%~9.85%和105.54%~144.40%;0~20 cm土壤微团聚体含量均随盐度的增加而降低,高潮滩降幅为59.56%~65.20%,低潮滩降幅为55.65%~60.92%.②高潮滩土壤团聚体稳定性随盐度的增加而增加,盐度对微团聚体、大团聚体含量(DR_(0.25))和平均重量直径(MWD)的作用力在不同土层均影响显著,盐度和淹水的交互作用对各级土壤水稳性团聚体分布及稳定性的影响均不显著.③土壤团聚体稳定性与土壤TC含量呈倒"U"型关系.综上,淹水环境变化对土壤水稳性团聚体分布及稳定性的影响较小,盐度和有机碳含量是影响闽江河口湿地土壤水稳性团聚体分布及稳定性的重要限制性参数.  相似文献

The ever increasing quantity of construction and demolition waste (C&DW) in Italy is presently challenging public administrators, which strive to ensure that collection and recycling are sustainably managed and need to understand whether and to what extent recycled aggregates can complement natural aggregates in a sustainable supply mix (SSM) for the construction industry. The paper presents a research aimed at analysing energy and environmental implications of the C&DW recycling chain in the administrative territory of Provincia di Torino in Northern Italy, with 2.25 million inhabitants and yearly generation of 1.8 million tons of C&DW. A combined Geographical Information System (GIS) and Life Cycle Assessment (LCA) model was developed using site-specific data and paying particular attention to land use, transportation and avoided landfill: crucial issues for sustainable planning and management. A GIS was used to handle data and information about 89 recycling plants, including technological features, output and physical–mechanical characteristics of recycled aggregate. The LCA methodology was used to identify and quantify energy and environmental loads, under different assumptions relevant to delivery distances, quality of recycled aggregates, local availability of natural aggregates and geographical coverage of market demand. The C&DW recycling chain was proved to be eco-efficient, as avoided impacts were found to be higher than the induced impacts for 13 out of 14 environmental indicators. It was also estimated that the transportation distance of recycled aggregate should increase 2–3 times before the induced impacts outweigh the avoided impacts.  相似文献

Methods for monitoring particle aggregation are briefly reviewed. Most of the techniques are based on some form of light scattering and may be greatly dependent on the optical properties of aggregates, which are not generally known. As fractal aggregates grow larger their density can become very low and this has important practical consequences for light scattering. For instance, the scattering coe cient may be much less than for solid objects, which means that the aggregates can appear much smaller than their actual size by a light transmission method. Also, for low-density objects, a high proportion of the scattered light energy is within a small angle of the incident beam, which may also be relevant for measurements with aggregates. Using the “turbidity fluctuation” technique as an example, it is shown how the apparent size of hydroxide flocs depends mainly on the included impurity particles, rather than the hydroxide precipitate itself. Results using clay suspensions with hydrolyzing coagulants and polymeric flocculants under di erent string conditions are discussed.  相似文献

为了阐明外源有机碳对土壤碳库的作用机制,以餐厨垃圾为原料,制备了4种不同分子量级的外源有机碳,通过室内试验,研究了外源有机碳对土壤有机碳组分及土壤团聚体的影响.结果表明:①以分子量≤1 ku为主的小分子有机碳处理组土壤中w(DOC)(DOC为水溶性有机碳)比CK组增加了0.383 g/kg,CO2累积释放量达到(6.595±0.259)mg/kg(以C计);微生物活性增强,>5 mm水稳大团聚体质量分数相对于CK组提升了25.06%,表明小分子有机碳增强了土壤微生物活性,微生物通过菌根网络缠绕作用促进大团聚体形成,增强土壤团聚体稳定性.②以分子量≥5 ku的大分子有机碳为主的大分子有机碳处理组w(POC)(POC为颗粒态有机碳)为CK组的5.65倍,0.5~1 mm水稳大团聚体质量分数增加了10.84%,有机碳贡献率提升了14.79%,表明大分子有机碳通过增加土壤中w(POC),促进0.5~1 mm水稳大团聚体形成,提高团聚体有机碳贡献率.研究显示,不同分子量级外源有机碳通过不同的方式改善土壤团聚体结构进而促进土壤有机碳固定,餐厨垃圾生物强化有机肥中各量级有机碳比例合理,能够明显提升土壤有机碳水平,增加土壤团聚体稳定性.   相似文献

In the Southern Mediterranean region hilly and remote areas are afflicted with the high cost of diesel fuel, problematic supply, poor or non-existent grid connections and lack of maintenance technicians to service farm grid generation systems and machinery. A prototype Battery Powered Electric Vehicle (BPEV) charged using a solar photovoltaic array (10 kWp) was installed at a monastery situated at Achkout (Lebanon) with a total agricultural area of 50 ha, 10 ha of which was devoted to viticulture. The purpose of this was to investigate the potential for the cleaner production of power hence reducing the use of diesel fuels in agriculture. Data collected from the test site is presented and used to validate a numerical model. The numerical model was then used to extend the study further to the other countries bordering the Mediterranean Sea which have different levels of solar irradiance. Currently the average cost of diesel fuel within the EU is at an all-time high of 1.32 €/L. A life cycle costing determined that if the current trends in inflation in the EU continue and that fuel costs increase by 7.5% per annum, then battery powered electric vehicles charged using solar photovoltaics are economically viable in areas of high solar irradiance. The numerical model was then used to determine the outcome of using lithium titanate batteries instead of conventional lead-acid batteries. Adoption of the described system for a 10 ha vineyard would result in annual fuel saving of 4200–5200 L of diesel depending on which location in the Mediterranean area a system such as this is installed.  相似文献

包括量子点在内的人工纳米颗粒是近年来突出的环境污染物,为确定其在生物膜中的扩散过程和特征,应用TIRF(全内反射荧光)、FRAP(荧光漂白后恢复)等技术,研究了CdTe/CdS/ZnS核壳式量子的微米级的团聚体在细菌Comamonas testoteroni生物膜表面的吸附动力学、生物膜内部的扩散及其在生物膜中的溶解和毒性.结果表明:通过TIRF技术观察到生物膜可快速吸附>1 μm的CdTe/CdS/ZnS核壳式量子点团聚体,而且通过CLSM(激光扫描共聚焦荧光显微镜)进行深度扫描发现,量子点团聚体吸附到生物膜表面后可以进一步扩散到生物膜深层,在25 min内可穿透45 μm的生物膜,并在生物膜中随深度呈线性分布特征.FRAP分析表明,量子点团聚体被生物膜固定后还具有较强的移动性,漂白区的荧光强度在5 min可恢复30%.量子点团聚体在生物膜中会溶解产生Cd2+、Zn2+等重金属离子,从而对生物膜产生毒性并杀死细菌.研究显示,虽然纳米颗粒进入环境中会形成微米级的团聚体,但依然可以进入生物膜,对水生微生物生态系统产生危害.TIRF、CLSM和FRAP技术是研究纳米颗粒物在生物膜表面吸附和内部扩散动力学的有效工具.   相似文献

Life Cycle Assessment (LCA) was applied to two smallholder milk production systems in Peru in order to evaluate the environmental burden of milk produced in each. An Andean highland milk production system where livestock feeding is restricted to permanent pastures supplemented with on farm grown ryegrass-clover was opposed to a coastal system with dairy cows fed a diet consisting of fodder maize and purchased concentrate. Milk production levels (kg/cow day) differed considerably with 2.57 for the highland and 19.54 for the coastal system. A Life Cycle Inventory was calculated for the functional unit of 1 kg energy corrected milk (ECM) and the environmental impacts global warming, acidification and eutrophication were estimated for 1 kg ECM, 1 ha and 1 animal, considering the multi-functionality of the system. The highland system was characterized by a high land use (23.1 m2a/kg ECM vs. 1.71 m2a/kg ECM at the coast). Irrigation water and energy were on the other hand used to a much higher amount at the coast (7291 l/kg ECM and 8791 MJ/kg ECM, respectively) than in the highlands (848 l/kg ECM and 0.20 MJ/kg ECM). Global warming potential, acidification and eutrophication were higher for 1 kg ECM produced in the highlands than at the coast by 10.6 kg CO2 equivalents, 6.58 g sulfur dioxide equivalents and 10.63 g phosphate equivalents, respectively. Nevertheless, 5220 kg CO2 equivalents more were emitted per animal at the coast than in the highlands. Also acidification and eutrophication were estimated to be on average 6 and 4 times higher at the coast compared to the highlands when expressed for the functional units of 1 ha and 1 animal.

Results

Whereas livestock is mainly responsible for impacts on the environment in the highlands, at the coast both livestock related emissions and forage cultivation play an important role. Furthermore CO2 releases from soybean cultivations heavily contribute to total emissions. Sensitivity analysis indicates that for dairy systems relying on crop by-products as feed the choice of the allocation method is a crucial point in a LCA study. Based on the results of this study, strategies in order to reduce the environmental burden of milk production should focus on an increase of production levels and a reduction of methane emissions from enteric fermentation in the highlands and a modification of the concentrate components replacing soya as the protein source at the coast.  相似文献

在85kg/m3的含沙高浊水中投加阳离子高分子聚合物,借助图像分析技术与沉降技术分析探讨了不同絮凝条件下泥沙絮凝形态学参数:絮体粒径、絮体有效质量密度、絮体自由沉速、浑液面沉速与上清液余浊等的变化规律.利用表征参数“分维”定量控制不同絮凝条件(如搅拌速率、搅拌时间、高分子浓度等)对含沙高浊水絮体结构分形特性的影响.实验证明,不合适的絮凝条件将导致絮体分形构造疏松脆弱,分维值低.絮凝条件合适时(快速絮凝强度为:r1=300r/min,t1=10s;慢速絮凝强度:r2=120r/min,t2=180s;CP浓度:0.1%),絮体分形结构处于最佳状态.该状态下的絮体具有粒径较大、沉速快、有效质量密度高、粒度分布均匀,分维值最高(D3=2.16)的特点.而且,由静沉实验测得浑液面沉速高,上清液余浊也低.泥沙絮体分形结构达最佳时的混凝性能、沉降性能与结构密实性均较理想.  相似文献

Mexico City is the world’s largest and most polluted urban center. We examine the distribution of methane and other hydrocarbons within the Valley of Mexico, using it as a model for the role developing megacities will play in the next century of geochemical cycling. Seventy-five whole air samples were analyzed with multivariate statistical techniques, including factor analysis using principal components. Methane concentrations are highly variable in space and time, due to air circulations and source distribution. Landfills and open sewage canals are major inputs. Emissions into and out from the valley are modeled to be ∼515 t per day. Per capita emission is 0.01 t per annum per person, consistent with the global average for human related anaerobic generation. Natural gas leaks are small, and likely to be higher in other developing megacities; Mexican natural gas use has been discouraged out of earthquake safety concerns. In contrast, liquefied petroleum gas loss constitutes the major emission of propane and butane estimated at a leak rate of 5–10%. Kyoto and other environmental conventions have ignored methane as a greenhouse gas. Our analysis underscores the need to consider methane and other hydrocarbons, and the urbanization process, in future emission protocols.  相似文献

为探讨不同腐殖质组分团聚体对苯酚富集能力的影响,应用国际腐殖酸协会(IHSS)推荐方法提取腐殖质,以Ca2+、Al3+、Fe3+为桥键离子,制备了高岭石、蒙脱石不同腐殖质组分团聚体,采用平衡吸附法研究了不同腐殖质组分团聚体对苯酚的富集特征.结果表明:高岭石不同腐殖质组分团聚体对苯酚的饱和吸附量(Qm)大小依次为FA(富里酸)团聚体[(254.11±5.35)mg/kg] > HA(胡敏酸)团聚体[(186.14±1.61)mg/kg] > HM(胡敏素)团聚体[(120.61±1.67)mg/kg];蒙脱石不同腐殖质组分团聚体对苯酚的饱和吸附量大小依次为FA团聚体[(418.72±19.14)mg/kg)] > HA团聚体[(290.00±13.06)mg/kg)] > HM团聚体[(160.46±4.92)mg/kg)];高岭石不同腐殖质组分团聚体对苯酚的富集系数(ER)大小依次为FA团聚体(2.03±0.07)> HA团聚体(1.91±0.04)> HM团聚体(1.85±0.04);蒙脱石不同腐殖质组分团聚体对苯酚的富集系数大小依次为FA团聚体(2.50±0.10)> HA团聚体(2.45±0.11)> HM团聚体(1.36±0.04).研究显示,FA团聚体对苯酚的饱和吸附量和富集系数较大,对苯酚的吸附(富集)作用较强,HM团聚体对苯酚的饱和吸附量和富集系数较小,对苯酚的吸附(富集)作用较弱.不同腐殖质组分团聚体对苯酚的富集能力大小表现为FA团聚体> HA团聚体> HM团聚体.因此,FA团聚体可作为控制受苯酚污染场地土壤的首选修复剂.   相似文献

In the subarctic winter, fungal spores are found in indoor air even when outdoor spore levels are very low. The results of this study support an explanation that some indoor airborne fungal spores are derived from unnoticeable fungal microcolonies, which may develop on temporarily wet surfaces. Laboratory experiments on Penicillium verrucosum indicated that the fungus germinated on new wallpaper very quickly (about half an hour) under moist conditions. Hyphal growth and sporulation of the fungus on moist wallpaper occured within one day of incubation. In gravity-settling tape samples from occasionally wet surfaces in a suburban home, large spore aggregates, hyphal fragments with some spores and spores in the germination stage were found, indicating fungal growth. These experiments showed that fungal microcolonies can develop within a week on occasionally wet indoor surfaces.  相似文献

以藻源有机质(AOM)和腐殖酸(HA)作为典型生物大分子可溶有机质(DOM)和腐殖化DOM,采用荧光猝灭滴定和光谱分析考察磺胺二甲嘧啶(SMZ)与DOM的络合作用.三维荧光光谱结合平行因子分析显示AOM荧光组分主要由类酪氨酸和类色氨酸物质组成,而类富里酸和类腐殖酸物质是HA荧光组分的主要构成.4种荧光组分可与SMZ发生不同程度的静态猝灭,且猝灭过程中DOM分子构象改变.同步荧光光谱结合二维相关图谱进一步发现类酪氨酸优先于类色氨酸组分与SMZ发生络合作用.Ryan-Weber非线性模拟拟合表明AOM中各荧光组分与SMZ的络合稳定常数(logK)大小为:类富里酸(3.33)类酪氨酸(3.12)类色氨酸(2.15)类腐殖酸(1.57);而HA中大小为:类富里酸(3.06)类腐殖酸(2.02).总体上生物大分子DOM对SMZ的亲和力高于腐殖化DOM.DOM与SMZ的相互作用可改变水环境中抗生素的形态和归趋,影响其生物有效性和生态毒性.  相似文献
