Assessment of oxidative stress levels and tissue concentrations of elements in plants growing wild on fly ash basins is critical for realistic hazard identification of fly ash disposal areas. Hitherto, levels of oxidative stress markers in plants growing wild on fly ash basins have not been adequately investigated. We report here concentrations of selected metal and metalloid elements and levels of oxidative stress markers in leaves of Cassia occidentalis growing wild on a fly ash basin (Badarpur Thermal Power Station site) and a reference site (Garhi Mandu Van site). Plants growing on the fly ash basin had significantly high foliar concentration of As, Ni, Pb and Se and low foliar concentration of Mn and Fe compared to the plants growing on the reference site. The plants inhabiting the fly ash basin showed signs of oxidative stress and had elevated levels of lipid peroxidation, electrolyte leakage from cells and low levels of chlorophyll a and total carotenoids compared to plants growing at the reference site. The levels of both protein thiols and nonprotein thiols were elevated in plants growing on the fly ash basin compared to plants growing on the reference site. However, no differences were observed in the levels of cysteine, reduced glutathione and oxidized glutathione in plants growing at both the sites. Our study suggests that: (1) fly ash triggers oxidative stress responses in plants growing wild on fly ash basin, and (2) elevated levels of protein thiols and nonprotein thiols may have a role in protecting the plants from environmental stress.  相似文献

Use of lignite in power generation has led to increasing environmental problems associated not only with gaseous emissions but also with the disposal of ash residues. In particular, use of low quality coal with high ash content results in huge quantities of fly ash to be disposed of. The main problem related to fly ash disposal is the heavy metal content of the residue. In this regard, experimental results of numerous studies indicate that toxic trace metals may leach when fly ash contacts water. In this study, fly ash samples obtained from thermal power plants, namely Soma and Tunçbilek, located at the west part of Turkey, were subjected to toxicity tests such as European Committee for standardization (CEN) and toxicity characteristic leaching (TCLP) procedures of the U.S. Environmental Protection Agency (U.S. EPA). The geochemical composition of the tested ash samples from the power plant show variations depending on the coal burned in the plants. Furthermore, the CEN and TCLP extraction results showed variations such that the ash samples were classified as `toxic waste' based on TCLP result whereas they were classified as `non-toxic' wastes based on CEN results, indicating test results are pH dependent.  相似文献

Proper disposal and/or recycling of different industrial waste materials have long been recognized as a prime environmental concern throughout the world, and fly ash is major amongst them. In the present study, we tried to assess the feasibilities of possible effective and safe utilization of fly ash as soil amendment in Indian paddy field and its impact on rice plants, especially at growth and yield level. Our results showed that certain doses of fly ash amendments have significantly improved the physico-chemical and mineralogical properties of paddy field soil, and at lower level of amendments, fly ash induced the growth performances of three rice cultivars too. Grain yield and grain quality also responded similarly as per the growth responses. However, differential cultivar response was observed accordingly, and cultivar Sugandha-3 showed higher yield as compared with cultivars Sambha and Saryu-52. Based on the observed results, it was concluded that up to a certain level, fly ash amendments could be beneficial for Indian paddy field and can be utilized as feasible management strategy for the disposal of this major industrial waste.  相似文献

A cost-effective method was developed to map fire scars on Quicklooks of Landat TM imagery. The method was compared with a full resolution Landsat image using visual interpretation and supervised classification using the Maximum Likelihood procedure, resulting in a high degree of agreement between methods. A long time series of fire scars was developed using all available Landsat Quicklooks between 1989 and 2001 for an area of 63000 sq km in north-east Namibia. Between 27 and 51% of the study area burned annually, while only 10% of the area did not burn between 1989 and 2001. Not-burned areas were mainly settled areas and permanent wetlands. 33% of the area burned between 5 and 7 times during the 13 years indicating a high frequency overall. Rainfall and livestock had little influence on burned areas. In 1996 formal fire management started in a portion of the study area consisting of building firebreaks and holding awareness programs. A comparison of burned areas before and after the intervention started allowed evaluating its effectiveness. The area where the formal fire management program was undertaken showed a significant decrease in burned area. It is suggested that awareness campaigns rather than firebreaks contributed to this decrease. Selected tree population data were compared with fire frequencies. Differences in tree occurrence, regeneration, and stem diameter distributions between low and high fire frequencies could be detected and explained with known responses of the species to fire. This suggests that the observed time series is representative of a long-term fire regime in the area.  相似文献

Fly ash is a by-product of coal-fired electricity generation plants. Its utilization and disposal is of utmost importance. Using onion (Allium cepa) root tip system, the present study was carried out to evaluate the potential toxic and genotoxic effects of fly ash, collected from a thermal power plant in West Bengal, India. Prior to testing, the collected fly ash sample was mixed with sand in different proportions. Allium bulbs were allowed to germinate directly in fly ash and after five days the germinating roots were processed for the Allium test. Additionally, the Allium test was adapted for detecting DNA damage through comet assay. The results from the Allium test indicate that fly ash at 100% concentration inhibits root growth and mitotic indices; induces binucleated cells as a function of the proportion, but is not toxic at very low concentration. In the comet assay, a statistical increase for DNA strand breaks was found only at higher concentrations. The sample was analyzed by flame atomic absorption spectrometer for Zn, Pb, Cu, Ni, Cd and As, whose presence could partly be responsible for the toxicity of fly ash. The study concludes that the classical Allium test can give a more comprehensive data when done in combination with the comet assay, which is faster, simpler and independent of mitosis. Also when fly ash is used for other purposes in combination with soils, it should be judiciously used at very low concentrations in order to protect the ecosystem health from any potential adverse effects.  相似文献

Indirect competitive immunoassays were developed on protein microarrays for the sensitive and simultaneous detection of multiple environmental chemicals in one sample. In this assay, a DNA/SYTOX Orange conjugate was employed as an antibody label to increase the fluorescence signal and sensitivity of the immunoassays. Epoxy-modified glass slides were selected as the substrate for the production of 4 × 4 coating antigen microarrays. With this signal-enhancing system, competition curves for 17β-estradiol (E2), benzo[a]pyrene (BaP) and 2,2',4,4'-tetrabromodiphenyl ether (BDE-47) were obtained individually on the protein microarray. The IC(50) and calculated limit of detection (LOD) are 0.32 μg L(-1) and 0.022 μg L(-1) for E2, 37.2 μg L(-1) and 24.5 μg L(-1) for BaP, and 31.6 μg L(-1) and 2.8 μg L(-1) for BDE-47, respectively. LOD of E2 is 14-fold lower than the value reported in a previous study using Cy3 labeled antibody (Du et al., Clin. Chem, 2005, 51, 368-375). The results of the microarray immunoassay were within 15% of chromatographic analysis for all three pollutants in spiked river water samples, thus verifying the immunoassay. Simultaneous detection of E2, BaP and BDE-47 in one sample was demonstrated. There was no cross-reaction in the immunoassay between these three environmental chemicals. These results suggest that microarray-based immunoassays with DNA/dye conjugate labels are useful tools for the rapid, sensitive, and high throughput screening of multiple environmental contaminants.  相似文献

Sensitive analytical methods indicate the presence of hundreds of chemical contaminants in our environment. However, concentration of these pollutants is usually at the low parts per million or parts per billion level. At such low levels, toxicants induce long-term (chronic) rather than short-term (acute) toxicities. Experiments are designed to evaluate chronic toxicity using early bioindicators. Recently, fish have been used as experimental animals because some species show early (weeks as opposed to years in other research animals), sensitive responses. Thus, medaka (Oryzias latipes), following exposure to diethylnitrosamine (DEN), exhibited liver tumors in several weeks; more interestingly its DNA was modified (to a DNA-adduct) after only 24–48 hours exposure. Such adducts show promise as early bioindicators because they are formed within hours of exposure. The formation of DNA-adducts were monitored in medaka and rainbow trout (Salmo gairdneri) exposed to DEN by aqueous or intra-peritoneal routes. Using HPLC-Fluorimetry, O 6-ethylguanine (O 6-EtGua) was detected and monitored in acid thermal hydrolysates of DNA isolates. (Detection limit for O 6-EtGua was as low as 3 ng, 1.7×10-11 moles). Fourier transform cyclotron resonance mass spectrometry with IR laser desorption/ionization was used advantageously to establish the structure of nucleotides, bases and nucleosides directly, without further cumbersome derivatizations. Discharges of coloured effluents into surface waters provide conspicuous evidence of the impact of industry on the environment. The textile industry is an obvious candidate for sources of such discharges. Conventional treatment methods appear to alleviate this situation by removing colour, however the affect on toxicity is less obvious. The objective of this study was to examine the changes in effluent toxicity during the course of two alternative wastewater treatment methods, ozonation and electrochemical oxidation, using a novel toxicity biosensor, GreenScreen EM. The biosensor is capable of measuring both general acute toxicity (cytotoxicity), and more specifically genotoxicity, that is damage to a cell's DNA structure, replication or distribution, caused by substances that may be mutagenic and/or carcinogenic. The biosensor utilises a modified strain of the brewers yeast Saccharomyces cerevisiae, incorporating a gene encoding green fluorescent protein (GFP) linked to the inducible promoter of the DNA damage responsive RAD54 gene. Upon exposure to a genotoxin, the production of GFP is up-regulated in parallel with RAD54, and the resulting cellular fluorescence provides a measure of genotoxicity. Acute toxicity is simultaneously determined by monitoring relative total growth of the cell culture during incubation. The results presented in this paper show that a reduction in colouration does not necessarily correspond to a reduction in effluent toxicity.  相似文献

Smoke from residential wood burning has been identified as a major contributor to air pollution, motivating detailed emission measurements under controlled conditions. A series of experiments were performed to compare the emission levels from two types of wood-stoves to those of fireplaces. Eight types of biomass were burned in the laboratory: wood from seven species of trees grown in the Portuguese forest (Pinus pinaster, Eucalyptus globulus, Quercus suber, Acacia longifolia, Quercus faginea, Olea europaea and Quercus ilex rotundifolia) and briquettes produced from forest biomass waste. Average emission factors were in the ranges 27.5-99.2 g CO kg(-1), 552-1660 g CO(2) kg(-1), 0.66-1.34 g NO kg(-1), and 0.82-4.94 g hydrocarbons kg(-1) of biomass burned (dry basis). Average particle emission factors varied between 1.12 and 20.06 g kg(-1) biomass burned (dry basis), with higher burn rates producing significantly less particle mass per kg wood burned than the low burn rates. Particle mass emission factors from wood-stoves were lower than those from the fireplace. The average emission factors for organic and elemental carbon were in the intervals 0.24-10.1 and 0.18-0.68 g kg(-1) biomass burned (dry basis), respectively. The elemental carbon content of particles emitted from the energy-efficient "chimney type" logwood stove was substantially higher than in the conventional cast iron stove and fireplace, whereas the opposite was observed for the organic carbon fraction. Pinus pinaster, the only softwood species among all, was the biofuel with the lowest emissions of particles, CO, NO and hydrocarbons.  相似文献

There is little information on the indoor environment in hotels. Analysis of fungal DNA by quantitative PCR (qPCR) is a new method which can detect general and specific sequences. Dust was collected through swab sampling of door frames in 69 hotel rooms in 20 countries in Europe and Asia (2007-2009). Five sequences were detected by qPCR: total fungal DNA, Aspergillus and Penicillium DNA (Asp/Pen DNA), Aspergillus versicolor (A. versicolor DNA), Stachybotrys chartarum (S. chartarum DNA) and Streptomyces spp. (Streptomyces DNA). Associations were analysed by multiple linear regression. Total fungal DNA (GM = 1.08 × 10(8) cell equivalents m(-2); GSD = 6.36) and Asp/Pen DNA (GM = 1.79 × 10(7) cell equivalents m(-2); GSD = 10.12) were detected in all rooms. A. versicolor DNA, S. chartarum DNA and Streptomyces DNA were detected in 84%, 28% and 47% of the samples. In total, 20% of the rooms had observed dampness/mould, and 30% had odour. Low latitude (range 1.5-64.2 degrees) was a predictor of Asp/Pen DNA. Seaside location, lack of mechanical ventilation, and dampness or mould were other predictors of total fungal DNA and Asp/Pen DNA. Hotel ranking (Trip Advisor) or self-rated quality of the interior of the hotel room was a predictor of total fungal DNA, A. versicolor DNA and Streptomyces DNA. Odour was a predictor of S. chartarum DNA. In conclusion, fungal DNA in swab samples from hotel rooms was related to latitude, seaside location, ventilation, visible dampness and indoor mould growth. Hotels in tropical areas may have 10-100 times higher levels of common moulds such as Aspergillus and Penicillium species, as compared to a temperate climate zone.  相似文献

The chromium species leaching from a coal combustion fly ash landfill has been characterized as well as a novel approach to treat leachates rich in hexavalent chromium, Cr(VI), by using another natural waste by-product, acid mine drainage (AMD), has been investigated during this study. It is observed that as much as 8% (approximately 10 microg g(-1) in fly ash) of total chromium is converted to the Cr(VI) species during oxidative combustion of coal and remains in the resulting ash as a stable species, however, it is significantly mobile in water based leaching. Approximately 1.23 +/- 0.01 microg g(-1) of Cr(VI) was found in the landfill leachate from permanent deposits of aged fly ash. This study also confirmed the use of AMD, which often is in close proximity to coal combustion by-product landfills, is an extremely effective and economical remediation option for the elimination of hexavalent chromium in fly ash generated leachate. Speciated isotope dilution mass spectrometry (SIDMS), as described in EPA Method 6800, was used to analytically evaluate and validate the field application of the ferrous iron and chromate chemistry in the remediation of Cr(VI) runoff.  相似文献

The relative abundance of ant species was measured by pit-fall trapping at 44 sites in southern New Mexico and southeastern Arizona, U.S.A.. Sites were selected for study based on documentation of a history of disturbance or protection from disturbance, exposure to varying intensities of livestock grazing, dominance by an exotic species of plant and vegetation change resulting from disturbance or restoration efforts. Ant community composition, relative abundances of species, and species richness were the same on disturbed and undisturbed sites. None of the metrics based on hypothesized responses of ants to disturbance clearly distinguished between disturbed and undisturbed sites. Ant communities on sites where restoration efforts have resulted in distinct differences in vegetative cover and composition were similar to the ant communities on degraded unrehabilitated sites on the same soil type. Ant communities in riparian cottonwood gallery forests in Arizona and New Mexico were similar but differed from the assemblages in exotic salt cedar and native ash riparian woodlands. Ant species exhibited remarkable resistance to human-induced disturbances in these rangeland areas. In grasslands dominated by the South African grass, Eragrostis lehmanniana Nees, large seed harvesting ants, Pogonomyrmex spp., were greatly reduced in abundance compared to native grasslands. Other ant metrics were not different in E. lehmanniana grasslands and native grasslands. We conclude that ants cannot be used as indicators of exposure to stress, ecosystem health or of rehabilitation success on rangeland ecosystems. Ants are also not useful indicators of faunal biodiversity in rangeland ecosystems.  相似文献

Monitoring drinking water quality is an important public health issue. Two objectives from the 4 years, six nations, EU Project μAqua were to develop hierarchically specific probes to detect and quantify pathogens in drinking water using a PCR-free microarray platform and to design a standardised water sampling program from different sources in Europe to obtain sufficient material for downstream analysis. Our phylochip contains barcodes (probes) that specifically identify freshwater pathogens that are human health risks in a taxonomic hierarchical fashion such that if species is present, the entire taxonomic hierarchy (genus, family, order, phylum, kingdom) leading to it must also be present, which avoids false positives. Molecular tools are more rapid, accurate and reliable than traditional methods, which means faster mitigation strategies with less harm to humans and the community. We present microarray results for the presence of freshwater pathogens from a Turkish lake used drinking water and inferred cyanobacterial cell equivalents from samples concentrated from 40 into 1 L in 45 min using hollow fibre filters. In two companion studies from the same samples, cyanobacterial toxins were analysed using chemical methods and those dates with highest toxin values also had highest cell equivalents as inferred from this microarray study.  相似文献

This study utilized telemetric systems to sample microbes and pathogens in forest, burned forest, rangeland, and urban watersheds to assess surface water quality in northern New Mexico. Four sites included remote mountainous watersheds, prairie rangelands, and a small urban area. The telemetric system was linked to dataloggers with automated event monitoring equipment to monitor discharge, turbidity, electrical conductivity, water temperature, and rainfall during base flow and storm events. Site data stored in dataloggers was uploaded to one of three types of telemetry: 1) radio in rangeland and urban settings; 2) a conventional phone/modem system with a modem positioned at the urban/forest interface; and 3) a satellite system used in a remote mountainous burned forest watershed. The major variables affecting selection of each system were site access, distance, technology, and cost. The systems were compared based on operation and cost. Utilization of telecommunications systems in this varied geographic area facilitated the gathering of hydrologic and water quality data on a timely basis.  相似文献

Cattle grazing nearby coal-fired power stations are exposed to fly ash. The present investigation aims to assess the environmental and health impacts of fly ash containing mercury emitted from thermal power plant. The health effect of fly ash were studied using 20 lactating cattle reared within a 5-km radius of s thermal power plant for the possible effect of fly ash such as the alterations in hematological and biochemical parameters of blood, milk, and urine. Results indicated that the hemoglobin levels (6.65?±?0.40?g/dl) were significantly reduced in all the exposed animals. Biochemical parameters viz., blood urea nitrogen (27.35?±?1.19?mg/dl), serum glutamate oxaloacetate transaminase (43.39?±?3.08?IU/l), albumin, and creatinine were found to be increased, whereas serum glutamate pyruvic transaminase (29.26?±?2.02) and Ca2+ were observed to be statistically insignificant in exposed animals. Mercury concentrations estimated in the blood, milk, and urine of exposed (n?=?20) and control (n?=?20) animals were 7.41?±?0.86, 4.75?±?0.57, 2.08?±?0.18, and 1.05?±?0.07, 0.54?±?0.03, 0.20?±?0.02?μg/kg, respectively. The significant increase (P?<?0.01) in the levels of mercury in blood, milk, and urine of exposed animals in comparison to control indicated that the alterations of biochemical parameters in exposed cattle could be due to their long term exposure to fly ash mercury which may have direct or indirect impact on human populations via food chain.  相似文献

The attenuation of arsenic in groundwater near alkaline coal fly ash disposal facilities was evaluated by determining the uptake of arsenic from ash leachates by surrounding alkaline soils. Ten different alkaline soils near a retired coal fly ash impoundment were used in this study with pH ranging from 7.6 to 9.0, while representative coal fly ash samples from two different locations in the coal fly ash impoundment were used to produce two alkaline ash leachates with pH 7.4 and 8.2. The arsenic found in the ash leachates was present as arsenate [As(V)]. Adsorption isotherm experiments were carried out to determine the adsorption parameters required for predicting the uptake of arsenic from the ash leachates. For all soils and leachates, the adsorption of arsenic followed the Langmuir and Freundlich equations, indicative of the favorable adsorption of arsenic from leachates onto all soils. The uptake of arsenic was evaluated as a function of ash leachate characteristics and the soil components. The uptake of arsenic from alkaline ash leachates, which occurred mainly as calcium hydrogen arsenate, increased with increasing clay fraction of soil and with increasing soil organic matter of the alkaline soils. Appreciable uptake of arsenic from alkaline ash leachates with different pH and arsenic concentration was observed for the alkaline soils, thus attenuating the contamination of groundwater downstream of the retired coal fly ash impoundment.  相似文献

‘Metal ash’ presents a waste disposal problem in most of the developing countries as the industries employ obsolete technologies. In this paper we describe analysis of tin ash, zinc ash and aluminium ash by means of optical methods, such as X-ray diffraction (XRD), inductively coupled plasma mass spectrometry (ICP-MS), electron probe micro analysis (EPMA), scanning electron microscopy (SEM) and chemical methods. The results of tin ash obtained by XRD method matched well with the cassiterite, a naturally occurring mineral of tin. ICP-MS studies reveal the presence of a large number of tracer metals, which may cause pollution by tertiary dispersion and this aspect is discussed. Conversely, the data generated by chemical methods are limited. However, the methods are simple and cost-effective. Then, they can easily be adopted by low-budget industries. Simple and cost-effective process to recover tin from tin ash is described. It is based on heating tin ash with sodium cyanide to about 900°C to separate tin component from the metal ash. The process recovers good quality tin and offers a very high yield. The process can be scaled up to small pilot plant.  相似文献

Endosulfan, malathion, and phosphamidon are widely used pesticides. Subchronic exposure to these contaminants commonly affects the central nervous system, immune, gastrointestinal, renal, and reproductive system. There effects have been attributed to increased oxidative stress. This study was conducted to examine the role of oxidative stress in genotoxicity following pesticide exposure using peripheral blood mononuclear cells (PBMC) in vitro. Further possible attenuation of genotoxicity was studied using N-acetylcysteine (NAC) and curcumin as known modulators of oxidative stress. Cultured mononuclear cells was isolated from peripheral blood of healthy volunteers, and exposed to varying concentrations of different pesticides: endosulfan, malathion, and phosphamidon for 6, 12, and 24 h. Lipid peroxidation was assessed by cellular malondialdehyde (MDA) level and DNA damage was quantified by measuring 8-hydroxy-2'-deoxyguanosine (8-OH-dG) using ELISA. Both MDA and 8-OH-dG were significantly increased in a dose-dependent manner following treatment with these pesticides. There was a significant decrease in MDA and 8-OH-dG levels in PBMC when co-treated with NAC or/and curcumin as compared to pesticide alone. These results indicate that pesticide-induced oxidative stress is probably responsible for the DNA damage, and NAC or curcumin attenuate this effect by counteracting the oxidative stress.  相似文献

Biomarker responses and histopathological lesions have been documented in laboratory mammals exposed to elevated concentrations of lead and cadmium. The exposure of white-footed mice (Peromyscus leucopus) to these metals and the potential associated toxic effects were examined at three contaminated sites in the Southeast Missouri Lead Mining District and at a reference site in MO, USA. Mice from the contaminated sites showed evidence of oxidative stress and reduced activity of red blood cell δ-aminolevulinic acid dehydratase (ALAD). Histological examinations of the liver and kidney, cytologic examination of blood smears, and biomarkers of lipid peroxidation and DNA damage failed to show indications of toxic effects from lead. The biomagnification factor of cadmium (hepatic concentration/soil concentration) at a site with a strongly acid soil was 44 times the average of the biomagnification factors at two sites with slightly alkaline soils. The elevated concentrations of cadmium in the mice did not cause observable toxicity, but were associated with about a 50% decrease in expected tissue lead concentrations and greater ALAD activity compared to the activity at the reference site. Lead was associated with a decrease in concentrations of hepatic glutathione and thiols, whereas cadmium was associated with an increase. In addition, to support risk assessment efforts, we developed linear regression models relating both tissue lead dosages (based on a previously published a laboratory study) and tissue lead concentrations in Peromyscus to soil lead concentrations.  相似文献
