Introduction
Triclosan (TCS) is a multi-purpose biocide. Its wide use in personal care products (PCPs) fosters its dispersal in the aquatic environment. A review was undertaken on the occurrence, toxicity, and degradation of triclosan (TCS; 5-chloro-2,4-dichlorophenoxy)phenol) in the environment. Purpose
The interaction between triclosan (TCS) and human serum albumin (HSA) was investigated in order to obtain the binding mechanism, binding constant, the type of binding force, the binding distance between the donor and acceptor, and the effect of TCS on the conformation change of HSA.Methods
A HSA solution was added to the quartz cell and then titrated by successive addition of TCS. The fluorescence quenching spectra and synchronous spectra were recorded with the excitation and emission slits of the passage of band set at 10 and 20 nm. Three-dimensional fluorescence spectra of HSA were recorded before and after the addition of TCS. The capillary electrophoresis was conducted with the pressure injection mode at 0.5 psi for 5 s, separation under 25 kV, and detection at 214 nm.Results
Fluorescence data indicated the fluorescence quenching of HSA by TCS was static quenching, and the quenching constants (K a ) were 1.14?×?105, 8.75?×?104, 6.67?×?104, and 5.00?×?104 at 293, 298, 303, and 309 K, respectively. The thermodynamic parameters, enthalpy change (??H) and entropy change (??S) for the interaction were calculated to be ?37.9 kJ mol?1 and 32.6 J?mol?1 K?1. The binding distance between TCS and tryptophan residues of HSA was obtained to be 1.81 nm according to F??rster nonradioactive energy transfer theory. The UV-Vis absorption spectroscopy, the synchronous fluorescence spectroscopy, three-dimensional fluorescence spectroscopy, and circular dichroism spectroscopy revealed the alterations of HSA secondary structure in the presence of TCS. Finally, the interaction between TCS and HSA was further confirmed by capillary electrophoresis.Conclusions
TCS was bound to HSA to form the TCS-HSA complex, with the binding distance of 1.81 nm. Hydrophobic interaction and hydrogen bond were dominated in the binding. TCS could change the secondary conformation of HSA. This work provides an insight into noncovalent interaction between emerging pollutants and protein, helping to elucidate the toxic mechanism of such pollutants. 相似文献4.
Ecotoxicological effects of typical personal care products on seed germination and seedling development of wheat (Triticum aestivum L.) 总被引:1,自引:0,他引:1
Biochemical responses of wheat (Triticum aestivum L.) seedlings stressed by two typical personal care products (PCPs) – triclosan (TCS) and galaxolide (HHCB) were experimentally investigated to assess their ecological risks. The results showed that wheat shoot and root elongation was significantly inhibited by 50–250 mg L−1 TCS and HHCB. Wheat roots were sensitive to TCS, while shoots were sensitive to HHCB. The median effect concentration (EC50) of TCS and HHCB based on the inhibition of their sensitive sites were 147.8 and 143.4 mg L−1, respectively. Moreover, the damage of wheat seedlings treated by low concentration of TCS and HHCB during a long period cannot be neglected. After a 21-d exposure, 0.2–3.0 mg L−1 TCS and HHCB treatment caused the damage to the accumulation of chlorophyll (CHL), the synthesis of soluble protein (SP), and the activity of peroxidase (POD) and superoxide dismutases (SOD) in different degree. However, different changing trends of these physiological indexes treated by different PCPs were observed after 7-d to 14-d exposures, especially the activity of POD and SOD. The activity of POD and SOD in wheat leaves and roots decreased with an increase in the concentration of TCS and the exposure time. However, the enzyme activities in wheat leaves treated by 0.2–3.0 mg L−1 HHCB increased after a 14-d exposure, and with the prolongation of exposure time, the enzyme activities significantly decreased. The variations in these physiological indexes of wheat could be considered as good biomarkers of serious stress by TCS and HHCB in the environment. 相似文献

The present study focuses on the temporal concentration changes of four common organic pollutants in small freshwater streams of Hesse, Germany. The substances (tris(2-chloroethyl)phosphate (TCEP), the technical isomer mixture of 4-nonylphenol (NP), 2-(t-butylamino)-4-(ethylamino)-6-(methylthio)-s-triazine (terbutryn), and N,N-diethyl-m-toluamide (DEET)) are subject to differing regulations. Whereas the use of NP and the related nonylphenolethoxylates (NPEOs) are almost completely banned under EU directive 2003/53/EC, the herbicide terbutryn is only restricted for use as a herbicide in the majority of member states of the European Union (EU). In contrast, TCEP and DEET are not regulated by legislation, but have been replaced in some products through consumer pressure.
