过程监测及调控是废水生物除磷工艺得以长期稳定运行的基础。为了获取氧利用速率(OUR)在生物除磷过程的潜在应用及该过程氧利用特性,开展聚磷菌好氧吸磷过程的呼吸测量实验,分析了OUR曲线变化与磷酸盐浓度变化、胞内聚合物(PHAs)氧化、外源COD存在与否与浓度变化及pH变化之间的响应关系。研究得到如下结果:富集污泥无磷酸盐存在时,体系依然存在明显的氧利用行为,在不吸磷的情况下聚磷菌胞内的PHAs依然会好氧氧化;外源COD的存在将改变PAOs吸磷速率并延长好氧吸磷的结束时间;体系初始pH的不同导致好氧吸磷效率、吸磷过程pH变化趋势及变化程度的不同;不同的初始条件得到不同的OUR。分析证实,利用测量OUR来监测聚磷菌好氧吸磷过程的运行状态是可行的。  相似文献

The aim of this study was to characterize the labile part of dissolved organic carbon (DOC) present in groundwater by identification of natural organic carbon substrates and to assess their microbial utilization during aeration of the groundwater. The studied chlorophenol (CP) contaminated groundwater contained 60-2650 micromoll(-1) of DOC of which up to 98.0% were CPs; 1.7% were low-molecular weight organic acids and 0.2% were dissolved free amino acids. Traces of following natural organic carbon substrates were identified: L-alanine, L-isoleucine, L-leucine, L-serine, L-threonine, L-tyrosine, L-valine, L-aspartic, acetic, citric, formic, lactic, malic and oxalic acid. Dissolved oxygen concentration inside the CP-plume was lower (mean 25 micromoll(-1)) than outside of the plume (mean 102 micromoll(-1)). Over a monitoring period of four years the concentrations of CPs, Fe(II) and NH4+ were higher inside than outside of the CP-plume. Oxygen availability within the CP-plume limits in situ biological oxidation of CPs, DOC, NH4+ and Fe(II). The microbial enzymatic hydrolysis rates of 4-methylumbelliferyl and 7-amino-4-methylcoumarin-linked substrates varied from 0.01 to 52 micromoll(-1)h(-1) and was slightly higher inside than outside the plume. Microbial uptake rates of 14C-acetate, 14C-glucose and 14C-leucine were on average 28, 4 and 4 pmoll(-1)h(-1) outside and 17, 25 and 8 pmoll(-1)h(-1) inside the plume, respectively. The indigenous microorganisms were shown able of hydrolysis of dissolved organic matter, uptake and utilization of natural organic carbon substrates. Therefore, the labile part of DOC serves as a pool of secondary substrates beside the CP-contaminants in the groundwater and possibly help in sustaining the growth of CP-degrading bacteria.  相似文献

Uptake and fate of TNT and RDX by three aquatic and four wetland plants were studied using hydroponic, batch, incubations in explosives-contaminated groundwater amended with [U-14C]-TNT or [U-14C]-RDX in the laboratory. Substrates in which the plants were rooted were also tested. Plants and substrates were collected from a small-scale wetland constructed for explosives removal, and groundwater originated from a local aquifer at the Milan Army Ammunition Plant. This study demonstrated rapid uptake of [U-14C]-TNT derived 14C, concentration at the uptake sites and limited transport in all plants. Per unit of mass, uptake was higher in submersed than in emergent species. Biotransformation of TNT had occurred in all plant treatments after 7-day incubation in 1.6 to 3.4 mg TNT L-i, with labeled amino-dinitrotoluenes (ADNTs), three unidentified compounds unique for plants, and mostly polar products as results. Biotransformation occurred also in the substrates, yielding labeled ADNT, one unidentified compound unique for substrates, and polar products. TNT was not recovered by HPLC in plants and substrates after incubation. Uptake of [U-14C]-RDX derived 14C in plants was slower than that of TNT, transport was substantial, and concentration occurred at sites where new plant material was synthesized. As for TNT, uptake per unit of mass was higher in submersed than in emergent species. Biotransformation of RDX had occurred in all plant treatments after 13-day incubation in 1.5 mg RDX L-1, with one unidentified compound unique for plants, and mostly polar products as results. Biotransformation had occurred also in the substrates, but to a far lower extent than in plants. Substrates and plants had one unidentified 14C-RDX metabolite in common. HPLC analysis confirmed the presence of RDX in most plants and in three out of four substrates at the end of the incubation period.  相似文献

Wastewater quality characteristics in terms of biomass, its substrates, and the corresponding kinetic and stoichiometric parameters were determined based on 109 wastewater samples originating from five different campaigns in four different sewer networks. Quality parameters were determined by model calibration of measured wastewater oxygen uptake rates applying a model that describes the aerobic breakdown of wastewater organic matter. Thereafter, the distributions of the parameters were analyzed. Two of the five datasets were obtained at the upstream end of a five-km-long, intercepting gravity sewer. For each of these upstream wastewater samples, downstream samples were collected with a delay corresponding to the residence time. The upstream distributions of the wastewater composition were used as boundary conditions for a Monte Carlo simulation. The calculated downstream distributions were compared to the measured downstream distributions and good agreement was observed.  相似文献

研究了两套不同基质复合垂直流人工湿地小试装置在不同季节对富营养化景观水的净化效果,分析了温度变化对污染物去除效果的影响,考察了微生物的硝化强度和反硝化强度以及基质理化性质对植物生长状况的影响,并探讨了氮、磷的去除途径.结果表明:不同基质复合垂直流人工湿地随季节变化对浊度、COD和磷的去除效果差异不明显;温度降低对脱氮效果影响显著,对磷的去除影响不大.沸石-页岩湿地硝化强度和反硝化强度优于砾石湿地.采用沸石作为基质可以提高系统对氮的去除效率,促进湿地植物的生长.对氮、磷去除途径的分析表明:微生物的硝化反硝化作用以及基质对磷的吸附沉淀作用是复合垂直流人工湿地去除氮、磷的主要途径;植物吸收分别占湿地TN、TP去除量的16%和35%左右,也是个重要途径.  相似文献

Abstract

The biological effects of clofibrate (ethyl p‐chlorophenoxy‐isobutyric acid) on the growth and metabolism of the soil‐borne wheat pathogen Fusarium culmorum, were examined.

In mid log phase (16 hr) cultures both phenylalanine uptake and secondary spore production were stimulated at 0.1 μM concentration; the net sterol content was reduced 50% at 0.35 μM; oxygen uptake was stimulated at 0.1 mM; growth was inhibited 50% at 0.1 mM concentration. Both phenylalanine and oxygen uptake were inhibited at 1.0 mM and pyruvate dehydrogenase activity was reduced 50% at 50 mM concentration of clofibrate.

The data indicate that clofibrate affects a number of biological and enzyme systems. The inhibitory effect on the growth of the pathogen suggest a potential use of hypolipidemic agents like clofibrate as an antifungal agent for seed protection.  相似文献

Diffusion of dissolved oxygen through activated sludge flocs was studied, as it represents a potential mechanism for simultaneous nitrification and denitrification in activated sludge systems. Dissolved oxygen profiles through six floc particles collected at different times from a full-scale activated sludge plant demonstrated that that the dissolved oxygen concentration declines through all floc particles. For larger floc particles (2-mm diameter and greater), the dissolved oxygen concentration reached near-zero values at depths depending on process operating conditions. A mathematical model based on diffusion of dissolved oxygen, organic substrate (methanol), ammonia, nitrite, and nitrate through a spherical floc and consumption of dissolved oxygen by heterotrophs and autotrophs accurately predicted the dissolved oxygen profile and required adjustment of only one model parameter--the concentration of heterotrophs. A different dissolved oxygen decline pattern was exhibited for the smaller floc particles characterized, with the dissolved oxygen reaching a non-zero plateau toward the center of the floc. This pattern was not reproduced with the mathematical model developed and suggests that additional mechanisms are responsible for the transport of dissolved oxygen into the center of these flocs. Implications of these results regarding the occurrence of simultaneous nitrification and denitrification include consideration of the factors that affect floc size and distribution (simultaneous nitrification and denitrification is maximized with larger floc particles), coupling of the International Water Association (London) activated models to predict activated sludge composition with diffusion models to consider intrafloc effects, and the effects of substrate diffusion on the apparent half-saturation constant for various substrates in activated sludge systems.  相似文献

The effect of compost quality on performance of biofiltration media was investigated. Three types of compost media were characterized in terms of their key properties, including pH, total organic carbon (TOC) and nitrogen, moisture content, oxygen uptake rates and heterotrophic and fungal plate counts. Laboratory-scale biofilter columns were each filled with a different compost medium mixed with buffer and a bulking agent. The columns were operated with a volatile organic compound (VOC) mixture consisting of ethyl alcohol, butyraldehyde, ethyl acetate, and 1,1-diethoxybutane and subjected to various operating conditions. After 13 months, the columns were sampled and post-characterization of the media was performed. A mixture of biosolids/horse manure compost, which had a high initial microbial activity, had the shortest acclimation period. Best overall performance, however, was observed with yard waste compost, which was the most stable medium. Media hydrophobicity was a key factor in long-term performance. Significant changes in compost structure, moisture content, pH, volatile solids, oxygen uptake rates, and microbial population densities were observed over the course of the study. Analyses of organic nitrogen content showed that significant amounts of nitrogen were not depleted from the media.  相似文献

三氯乙烯(trichloroethylene,TCE)是土壤和地下水中广泛存在的有机污染物,好氧生物降解因可将污染物彻底转化成无毒的终产物,一直受到广泛关注,但是TCE好氧降解需要共代谢底物。首次提出以汽油为底物,选取真养产碱杆菌作为活性降解菌株,对地下水中三氯乙烯的好氧共代谢降解进行了初步研究。分别优化了共代谢底物、底物与TCE浓度比、培养基、pH值、盐度、溶解氧等条件,确定了最佳降解条件。当水中TCE的浓度为1 mg/L时,通过对体系预曝氧气,调节汽油浓度为10 mg/L,pH值为5,降解24 h,TCE的降解率可达66.8%。为修复同时被汽油和TCE污染的场地提供了一个新的研究方向。  相似文献

This study investigated the anoxic and aerobic phosphate uptake and release reactions and the fraction of denitrifying phosphate-accumulating organisms (DPAOs) under various initial chemical oxygen demand (COD) and residual COD conditions. The results showed that DPAOs and non-DPAOs could release phosphate when high soluble COD was present. Consequently, the phosphate-uptake potential was dynamic and increased when the initial COD increased, the initial polyhydroxyalkanoates (PHA) increased, and the residual COD decreased. Furthermore, the electron acceptor (oxygen of nitrate) has more significant influence on the phosphate uptake/release characteristics, while the residual COD concentrations have little influence on that. The fraction of DPAOs to phosphate-accumulating organisms was 42% when the initial PHA storage was enough by both DPAOs and non-DPAOs. This was closely related to the relative phosphate uptake (47%) in the anoxic zone of the process.  相似文献

聚-β-羟基丁酸酯(polyhydroxybutyrate,PHB)是一种可完全生物降解和具有良好生物相容性的高分子材料,许多情况下可作为传统塑料的替代品。以活性污泥微生物作为PHB合成菌,研究了动态底物投加方式下,溶解氧对污泥贮存PHB过程的影响。结果表明,PHB的贮存过程相当程度上取决于氧的提供。溶解氧浓度的提高明显加快了底物乙酸的吸收,进而导致更大比例的乙酸被活性污泥贮存为PHB。试验中,溶解氧浓度由饱和浓度的10%提高到70%,PHB的产率由0.36 C mmol/C mmol 提高到0.56 C mmol/C mmol,PHB的胞内含量也由26%提高到37%。试验证明溶解氧是控制活性污泥合成PHB工艺的重要因素。  相似文献

Contamination of soil with pesticides can be evaluated using toxicity tests with worms because their ecological niche makes them good bioindicators. Bioaccumulation in compost worms of [methyl-14C] paraquat (1,1'-dimethyl-4,4'-bipyridinium dichloride) was measured after three-month exposure in two substrates with differing physicochemical characteristics, in particular their organic matter and clay contents. The treatments were 1.2, 12, and 120 microg paraquat g(-1) substrate. The action of the worms did not influence the loss of 14C from the substrates, as the 14C-recovered was essentially quantitative at the end of the study in both the presence and absence of the worms. The organic matter and clay contents of the substrates determined the extent of the paraquat uptake by the worms; worms from the substrate with smaller amounts of clay and organic matter had the higher values of the bioconcentration factor (BCF), these being about 5 (fresh-weight basis) and independent of the application rate. The BCF values in the substrate containing more organic matter and clay were smaller but increased from 1.1 to 3.8 with the increasing rates of application. However, in both substrates the amounts of paraquat bioaccumulated in the worms was always less than 1% of that applied, indicating the very strong binding of paraquat to the substrates and hence low availability to the worms.  相似文献

基于硝化过程中合成代谢和能量代谢耦合原理,分析硝化过程中的电子流,建立自养硝化菌细胞生长与氨氮氧化耗氧量之间的关系。采用间歇实验方法,通过电解质呼吸仪测定硝化过程的累积耗氧量和耗氧速率,使用M ann-Ken-dell趋势检验方法确定实验中硝化菌进入内源呼吸的时刻以及内源呼吸过程中的耗氧速率,估算出氨氮氧化的耗氧量。计算结果表明本实验中硝化菌产率系数为0.2345±0.0115 mg COD/mg NH4+-N。  相似文献

The industrial solvent trichloroethylene (TCE) is among the most ubiquitous chlorinated compounds found in groundwater contamination. The objective of this study was to develop a biobarrier system containing oxygen-organic releasing material to enhance the aerobic cometabolism of TCE in situ. The oxygen-organic material, which contains calcium peroxide and peat, is able to release oxygen and primary substrates continuously upon contact with water. Batch experiments were conducted to design and identify the components of the oxygen-organic releasing material, and evaluate the oxygen and organic substrate (presented as COD equivalent) release from the designed oxygen-organic material. The observed oxygen and chemical oxygen demand (COD) release rates were approximately 0.0246 and 0.052 mg/d/g of material, respectively. A laboratory-scale column experiment was then conducted to evaluate the feasibility of this proposed system for the bioremediation of TCE-contaminated groundwater. This system was performed using a series of continuous-flow glass columns including a soil column, an oxygen-organic material column, followed by two consecutive soil columns. Aerobic acclimated sludges were inoculated in all three soil columns to provide microbial consortia for TCE biodegradation. Simulated TCE-contaminated groundwater with a flow rate of 0.25 l/day was pumped into this system. Effluent samples from each column were analyzed for TCE and other indicating parameters (e.g., pH, dissolved oxygen). Results show that the decreases in TCE concentrations were observed over a 4-month operating period. Up to 99% of TCE removal efficiency was obtained in this passive system. Results indicate that the continuously released oxygen and organic substrates from the oxygen-organic materials enhanced TCE biotransformation. Thus, the biobarrier treatment scheme has the potential to be developed into an environmentally and economically acceptable remediation technology.  相似文献

以TiCl4和钠基蒙脱土为原料,采用水解法制备了TiO2-蒙脱土复合材料,采用XRD、IR和TEM对其结构和性质进行表征;通过藻细胞的内含物含量、生长代谢活性和形态的变化以及藻细胞膜脂质过氧化分析,研究了TiO2-蒙脱土复合材料在光辅助下去除颤藻的性能.结果表明, TiO2-蒙脱土复合材料中一部分TiO2插入蒙脱层间,增加了蒙脱土的层间距,另一部分以锐钛矿相均匀分散在蒙脱土上;在紫外光辅助下,TiO2-蒙脱土的用量为 50.0 mg/L,1 h后颤藻的除藻率可达94.58%;TiO2-蒙脱土通过光催化作用破坏藻细胞膜、降解藻细胞内含物、降低藻细胞代谢活性和破坏藻细胞形态,从而产生抑藻作用.  相似文献

活性污泥的比耗氧速率(SOUR)是表征污泥生物活性的重要参数之一,从微生物呼吸速率角度反映了活性污泥生理状态和基质代谢状况.通过监测SBR工艺活性污泥的SOUR,考察了SBR工艺中活性污泥SOUR的变化规律,研究了活性污泥的SOUR与各项控制参数之间的相关关系.试验结果表明,活性污泥的SOUR可以有效地表征SBR工艺的生化反应进程,并与DO、氧化还原电位(ORP)和pH之间存在良好的相关关系.  相似文献

Purpose

In order to better understand if the metabolic responses of echinoids could be related to their acid?Cbase status in an ocean acidification context, we studied the response of an intertidal sea urchin species, Paracentrotus lividus, submitted to low pH at two different temperatures.

Methods

Individuals were submitted to control (8.0) and low pH (7.7 and 7.4) at 10°C and 16°C (19?days). The relation between the coelomic fluid acid?Cbase status, the RNA/DNA ratio of gonads and the individual oxygen uptake were studied.

Results

The coelomic fluid pH decreased with the aquarium seawater, independently of temperature, but this explained only 13% of the pH variation. The coelomic fluid showed though a partial buffer capacity that was not related to skeleton dissolution ([Mg2+] and [Ca2+] did not differ between pH treatments). There was an interaction between temperature and pH on the oxygen uptake (V O2) which was increased at pH?7.7 and 7.4 at 10°C in comparison with controls, but not at 16°C, indicating an upregulation of the metabolism at low temperature and pH. However, gonad RNA/DNA ratios did not differ according to pH and temperature treatments, indicating that even if maintenance of physiological activities has an elevated metabolic cost when individuals are exposed to stress, they are not directly affected during short-term exposure. Long-term studies are needed in order to verify if gonad production/growth will be affected by low pH seawaters exposure.  相似文献
