Platymonas subcordiformis (UTEX 171) was cultured axenically for 4 d in constant light in a nitrate-containing medium and harvested in the log-phase of cell division. Cells were resuspended in artificial sea water without nutrients and either kept in constant light or placed in constant darkness. High-performance liquid chromatography was used to measure the free amino acid pools of the cells and to determine rates of net entry of each of a mixture of 18 amino acids at daily intervals for 5 d. Free amino acid pools decreased both in light and darkness in the absence of a nutrient sypply. The influx of amino acids in cells maintained in the light increased selectively. Comparison of the rate of entry of 14C-labeled glycine and net disappearance of glycine from the medium indicated extrusion of non-volatile labeled carbon that did not interact with reagents specific for amine groups. Light was required for synthesis of additional transporter protein which was apparently responsible for increased influx in cells maintained in the light. This response was blocked in the presence of cycloheximide. Cells maintained in the dark for prolonged periods retained the capacity to respond to light by synthesis of new transporter protein. Analysis of incorporation of amino acids into macromolecules indicated that both the overall rate and the pattern of amino acid incorporation were modified in the light. Analysis of the kinetics of glycine entry at a series of temperatures indicated that the concentration of glycine at which entry is half the maximum rate is approximately 2.7 M at the cell surface.  相似文献

Larvae of the red abalone (Haliotis rufescens Swainson) are functionally incapable of capturing particulate foods. The aim of this study was to determine whether these larvae could acquire energy from seawater in the form of dissolved organic material. Trochophore and veliger larvae were shown to acquire energy by transporting dissolved organic material from seawater. Both larval stages took up all classes of amino acids tested. The influx of radiolabeled alanine represented the net substrate flux, as determined by direct chemical measurement for both trochophore and veliger larvae. Although veliger larvae have a transport system to take up taurine from seawater, a net efflux was observed for this amino acid. The release of taurine occurred independently of the presence of either taurine or other amino acids in the medium. Transported alanine was used in both anabolic and catabolic pathways. The percent of 14C-alanine in the trichloroacetic acid-insoluble fraction (macromolecules) of veliger larvae ranged from 21 to 56% of the total radioactivity in the larvae. No lipid biosynthesis was detected from 14C-labeled alanine. Veliger larvae catabolized 15 to 19% of the total alanine taken up and released it as 14CO2. The metabolic rate (oxygen consumption) and the rate of amino acid uptake were both determined for the same group of veliger larvae. The percent contribution that the uptake of amino acids, from a total concentration of 1.6 M, made to the metabolic demand of abalone larvae ranged from 39 to 70%. The nutritional pattern for heterotrophic growth of Nitzschia angularis var. affinis (Grun.) Perag. is more complex than for other diatom species studied previously. This species grew slowly in the dark in the presence of single amino acids, either glutamate or alanine; other amino acids when supplied singly were not used as substrates. Carbon from glutamate was converted to cell carbon with an efficiency of 43%. Glutamine was inhibitory both in the light and in the dark, and aspartate inhibited heterotrophic growth on glutamate. Glucose and tryptone supplied singly did not support heterotrophic growth, but when combined, together they allowed for rapid growth of N. angularis (generation time of 16 h). Glucose in combination with glutamate, alanine, aspartate, or asparagine (but not with any other amino acids) also supported growth in the dark, at a rate considerably more rapid than with glutamate alone. In the presence of excess glucose and limiting concentrations of glutamate, approximately 50% of the cell carbon for heterotrophic growth came from glucose, while in combination with tryptone about 25% of the cell carbon came from glucose. Amino acids were taken up by cells grown either photoautrophically or in the dark in the presence or absence of organic substrates; uptake rates were some-what higher for dark-grown than for light-grown cells. Glucose was taken up only by dark-grown cells; induction of a glucose uptake system in the dark required the presence of glutamate but not of glucose. The rates of uptake of glutamate and glucose by cells incubated in the dark with glutamate were sufficiently high to account for the observed rates of growth on these substrates in the dark. Kinetics of glucose and amino acid uptake by attached and free-living bacteria were compared in the upper 70 m of the oligotrophic north-western Mediterranean Sea. Potential uptake rates of amino acids were higher than those of glucose in all the samples analysed. Cell-specific potential uptake rates of attached bacteria were up to two orders of magnitude higher than those of total bacteria, both for amino acids and glucose (0.72–153 amol amino acids cell−1 h−1 and 0.05–58.42 amol glucose cell−1 h−1 for attached bacteria and 0.34–1.37 amol amino acids cell−1 h−1 and 0.07–0.22 amol glucose cell−1 h−1 for total bacteria). The apparent K m values were also higher in attached bacteria than in total bacteria, both for amino acids and glucose. These results would reflect the presence of different uptake systems in attached and free-living bacteria, which is in accordance with the different nutrient characteristics of their microenvironments, ambient water and particles. Attached bacteria show transport systems with low affinity, which characterise a bacterial community adapted to high concentration of substrates. The vertical distribution of chlorophylla, copepods, dissolved free amino acid concentration and the fixation of14C by phytoplankton were monitored in the springs of 1983, 1987 and 1988 in the Ushant front region, shelf edge of the Celtic Sea and central Irish Sea, respectively. In each area, two stations characterized by mixed and stratified water conditions were compared. Vertical distributions of amino acids coincided with the distribution of copepods. A positive and significant correlation was found between the abudance of copepods and the concentration of amino acids dissolved in seawater. A negative and significant correlation was found between chlorophylla and the concentration of amino acids. Enrichment of amino acids ( 20 to 500 nM l–1 at specific depths) due to aspartic and glutamic acids, glutamine and ornithine, was assumed to reflect copepod feeding activity and faecal production. At these depths, the natural concentration and diversity of amino acids, including aspartic acid, glutamic acid, asparagine, serine, histidine, glutamine, arginine, threonine, glycine, alanine, tyrosine, valine, phenylalanine, ornithine and lysine, were high enough and in the correct proportions for triggering feeding and swimming and swarming behavior of copepods, as well as their remote detection of food at the micro- and meso-scales (1 to 10 m). This accumulation of amino acids also constitutes a potential additional source of organic nitrogen for bacteria and phytoplankton.  相似文献

The anaerobic microbial uptake of alanine and aspartic acid was determined in 4 diverse salt-marsh soils (tall and short Spartina alterniflora, creek bank, and mud flat). Uptake in soil slurries was determined by the radioisotopic tracer technique at one substrate concentration (<250 pmoles cm-3). Dissolved free alanine and aspartic acid concentrations in the interstitial nutrient pool ranged from approximately 1 to 500 pmoles cm-3. In the short S. alterniflora soil, maximum microbial uptake of alanine was found at a depth of 10 cm (8.32 pmoles cm-3 h-1); in the tall S. alterniflora soil maximum uptake was at 20 cm (23.4 pmoles cm-3 h-1). The utilization of aspartic acid appeared constant over the depth interval investigated (0 to 60 cm). The turnover times of alanine and aspartic acid in the tall and short S. alterniflora soils ranged from 5 to 25 h and 40 to 100 h, respectively. The percent of the labeled alanine and aspartic acid taken up that was mineralized by tall and short s. alterniflora microbenthos ranged from 20 to 50% and 5 to 20%, respectively.  相似文献

The effects of larval diet on the nutritional preferences of butterflies has rarely been examined. This study investigates whether alterations in the larval diet result in changes in adult preferences for nectar amino acids. Larvae of Coenonympha pamphilus were raised on fertilized or unfertilized Festuca rubra, grown under ambient (350 ppm) or elevated (750 ppm) atmospheric CO 2environments. Fertilization led to marked increases in leaf nitrogen concentration. In plants grown under elevated CO 2conditions, leaf water and nitrogen concentrations were significantly lower, and the C/N-ratio increased significantly. Fertilization of the host plant shortened the development time of C. pamphilus larvae, and pupal weight increased. In contrast, larvae of C. pamphilus developed significantly slower on F. rubra grown under elevated CO 2, but adult emergence weight was not affected by CO 2treatment of the plant. C. pamphilus females showed a clear preference for nectar mimics containing amino acids, whereas males, regardless of treatment, either preferred the nectar mimic void of amino acids or showed no preference for the different solutions. Female butterflies raised on fertilized plants showed a significant decline in their preference for nectar mimics containing amino acids. A slight, but not significant, trend towards increased nectar amino acid preference was found in females raised on plants grown under elevated CO 2. We clearly demonstrate that alterations in larval host quality led to changes in butterfly nectar preferences. The ability of the butterfly to either rely less on nectar uptake or compensate for poor larval conditions represents a trade-off between larval and adult butterfly feeding.  相似文献

The concentration and turnover of dissolved free amino acids were measured in samples from 25 and 100 m on three occasions at a station 6 miles off the California (USA) coast. Individual amino acid concentrations varied from undetectable (<0.05 g/l) to 3 g/l, the total amino acid concentration from 1.8 to 8.5 g/l. The greater concentration of total amino acids was always found at 25 m. The predominant amino acids were serine, lysine, aspartate, glutamate and alanine; reliable analyses could not be made for glycine because of a high blank. For the 10 individual amino acids studied, the rate of heterotrophic turnover ranged from undetectable to 1.2 g/l day-1; serine, aspartate, alanine and glutamate showed the highest rates. In samples from 25 m, the rates were 15 to 20 times higher than those taken from 100 m. The total calculated flux of the amino acids studied varied from 0.015 to 3.2 g/l day-1 and amounted to 1–10% of photosynthetic carbon dioxide fixation.  相似文献

Effects of the heavy metals zinc and cadmium on the free amino acid (FAA) pool of Gammarus pulex were studied at different metal concentrations and combinations as well as different exposure times. The dominant effect of these two metals was the reduction of most free amino acids and the whole FAA pool, except in the 10‐day low zinc and cadmium concentration exposures which resulted in a rise of free amino acid pool.

Among the free amino acids, the most sensitive to zinc exposure, were alanine, glutamic acid, arginine, and taurine; valine, leucine, asparagine, and isoleucine were among the most sensitive to cadmium. No predictable changes of individual free amino acids were shown in the mixed metals exposures. Elevation of taurine concentration was constant in seven of the eight treatments, it is suggested that this elevation may be related to the hepatopancreatic damage observed and induced synthesis of metallothioneins.  相似文献

We examined the impact of exposing natural populations of marine bacteria (from seawater collected near Woods Hole, Massachusetts, USA) to multiple nitrogen and carbon sources in a series of batch growth experiments conducted from 1989 through 1990. The substrate C:N ratio (C:Ns) was varied from 1.5:1 to 10:1 either with equal amounts of NH 4 + and different amino acids or an amino acid mixture, all supplemented with glucose to maintain the C:Ns ratio equal to that of the respective amino acid, or with combinations of glucose and NH 4 + alone. A common feature of the experiments involving amino acids was the concurrent uptake of NH 4 + and amino acids that persisted as long as a readily assimilable carbon source (glucose in our case) was taken up. There was no net regeneration of NH 4 + , even though catabolism of amino acids occurred. Regeneration of NH 4 + was evident only after glucose was completely utilized, which usually occurred at the end of exponential growth. The contribution of15NH 4 + to total nitrogen uptake by the end of exponential growth varied from ~60 to 80% when individual amino acids were present and down to ~24% when the amino acid mixture was added. These estimates are conservative because we did not account for possible isotope dilution effects resulting from amino acid catabolism. When NH 4 + and glucose were the sole nitrogen and carbon sources, there was a stoichiometric balance between glucose and NH 4 + uptake over a wide range of C:Ns ratios, leading to a constant bacterial biomass C:N ratio (C:NB) of ~4.5:1. As a result NH 4 + usage varied from 50% when the C:Ns ratio was 3.6:1, to 100% when the C:Ns ratio was 10:1. Gross growth efficiency varied from ~60% when NH 4 + plus glucose were added alone or with the amino acid mixture, to 47% when the individual amino acids were used in place of the mixture. It is thus evident that actively growing bacteria will act as sinks for nitrogen when a carbon source that can be assimilated easily is available to balance NH 4 + uptake, even when amino acids are available and are being co-metabolized.  相似文献

Porphyra perforata J. Ag. was collected from a rocky land-fill site near Kitsilano Beach, Vancouver, British Columbia, Canada and was grown for 4 d in media with one of the following forms of inorganic nitrogen: NO 3 - , NH 4 + and NO 3 - plus NH 4 + and for 10 d in nitrogen-free media. Internal nitrogen accumulation (nitrate, ammonium, amino acids and soluble protein), nitrate and ammonium uptake rates, and nitrate reductase activity were measured daily. Short initial periods (10 to 20 min) of rapid ammonium uptake were common in nitrogen-deficient plants. In the case of nitrate uptake, initial uptake rates were low, increasing after 10 to 20 min. Ammonium inhibited nitrate uptake for only the first 10 to 20 min and then nitrate uptake rates were independent of ammonium concentration. Nitrogen starvation for 8 d overcame this initial suppression of nitrate uptake by ammonium. Nitrogen starvation also resulted in a decrease in soluble internal nitrate content and a transient increase in nitrate reductase activity. Little or no decrease was observed in internal ammonium, total amino acids and soluble protein. The cultures grown on nitrate only, maintained high ammonium uptake rates also. The rate of nitrate reduction may have limited the supply of nitrogen available for further assimilation. Internal nitrate concentrations were inversely correlated with nitrate uptake rates. Except for ammonium-grown cultures, internal total amino acids and soluble protein showed no correlation with uptake rates. Both internal pool concentrations and enzyme activities are required to interpret changes in uptake rate during growth.  相似文献

Photosynthetic assimilation of 14CO2 by the symbiotic green alga Platymonas convolutae Parke et Manton in the marine flatworm Convoluta roscoffensis Graff has been investigated and compared with that in free-living P. subcordiformis and P. tetrathele. All Platymonas species investigated rapidly incorporate 14CO2 into a complex variety of soluble and insoluble assimilates. The rate of dark fixation is considerably lower in P. convolutae. Typical 14C-assimilate patterns are rather uniform in all Platymonas species, but the time courses of 14C-labelling of several compounds are very different. The percentage of 14C-aspartate and 14C-malate is significantly higher in P. convolutae after short-term-photosynthesis, whereas 14C-labelled phosphate esters predominate in the free-living Platymonas species. A comparison of the kinetics of 14C-labelling and of the distribution of 14C-activity between soluble and insoluble fractions suggests that glucose and fructose, not mannitol, as well as several amino acids (especially alanine) move from the algal partner to the tissue of the animal host. For valine uptake by the polychaete Nereis virens Sars, the kinetic constants were: V max=355 nmol g-1 fresh weight h-1, K m=20 M. Leucine and some other amino acids acted as partial inhibitors of valine uptake. Valine uptake rate was 78% higher at 21.5 S than at 14 S. The major portion of valine absorbed by the polychaete could be extracted as free valine, with 6.5 to 15.6% being respired, and 3.6 to 9.5% incorporated into proteins. Calculations indicate that 7 to 12% of the metabolism of N. virens may be sustained by uptake of glycine and aspartic acid from natural concentrations. It is suggested that uptake of amino acids by this worm is important in the nitrogen cycling of marine sediments.  相似文献

Changes in abiotic environmental factors do not adversely influence the capacity of Anemonia sulcata (Coelenterata, Anthozoa) to take up dissolved amino acids from sea water; e. g., A. sulcata resorbs also under anaerobic conditions. The influence of temperature on amino acid uptake (Q10=2) indicates that the translocation of the organic molecules through membranes of the ectoderm depends on energy. Uptake of amino acids is possible against a gradient of up to 9×106:1. It can be postulated that L-amino acids (also D-isomers) and glucose are resorbed by different “carrier systems”, since glucose does not influence the uptake of amino acids; however, amino acids can interfere with each other, e. g. phenylalanine/glycine. Blocking of anaerobic glycolysis results in uptake reduction; at the same concentrations “oxidative” blockers have a much smaller effect on amino acid uptake. The possibility of exploiting the surrounding water as an energy source by taking up dissolved organic material is discussed with reference to ecological and evolutionary aspects.  相似文献

The nutritional history of corals is known to affect metabolic processes such as inorganic nutrient uptake and photosynthesis, but little is known about how it affects assimilation efficiency of ingested prey items or the partitioning of prey nitrogen between the host and symbiont. The temperate scleractinian coral Oculina arbuscula and its tropical congener Oculina diffusa were acclimated to three nutritional regimes (fed twice weekly, starved, starved with an inorganic nutrient supplement), then fed Artemia nauplii labeled with the stable isotope tracer 15N. Fed corals of both species had the lowest assimilation efficiencies (36–51% for O. arbuscula, 38–57% for O. diffusa), but were not statistically different from the other nutritional regimes. Fed and starved corals also had similar NH4+ excretion rates. This is inconsistent with decreased nitrogen excretion and reduced amino acid catabolism predicted by both the nitrogen recycling and conservation paradigms. In coral host tissue, ~90% of the ingested 15N was in the TCA-insoluble (protein and nucleic acids) and ethanol-soluble (amino acids/low molecular weight compounds) within 4 h of feeding. The TCA-insoluble pool was also the dominant repository of the label in zooxanthellae of both species (40–53% in O. arbuscula, 50–60% in O. diffusa). However, nutritional history had no effect on the distribution of prey 15N within the biochemical pools of the host or the zooxanthellae for either species. This result is consistent with the nitrogen conservation hypothesis, as preferential carbon metabolism would minimize the effects of starvation on nitrogen-containing biochemical pools.Communicated by P.W. Absorption of volatile fatty acids (propionate, acetate, butyrate, and formiate) from the ambient water by the lugwormarenicola marina was demonstrated using14C-labelled compounds and by gas-liquid chromatography. Uptake was investigated in the presence of antibiotics. The rate of uptake was highest with propionate (vmax 0,58 mol g-1 w wt · h, Kt 0.75 mol l-1). Butyrate, acetate, and formiate were absorbed at lower rates in that order. Absorption of propionate, butyrate, and acetate showed saturation kinetics. In the presence of butyrate and acetate, propionate absorption was inhibited (maximum inhibition 78%). These results indicate that a carrier is involved in the absorption of propionate. Following absorption, the volatile fatty acids were rapidly metabolized. The largest proportion of label was incorporated into amino acids, with the pattern of distribution of radioactivity being specific for each volatile fatty acid.  相似文献

The phosphorus metabolism of Pyrocystis noctiluca Murray (Schuett) 1886 has characteristics which may enhance its potential for success in orthophosphate impoverished waters. The steady-state phosphate uptake rates were equal in the light and dark, and were directly proportional to both the phosphorus cell quota and the cell division rate. In contrast, nutrient-saturated uptake rates were multiphasic, faster in the light than the dark, 2 to 4 orders of magnitude greater than steady-state rates, and were inversely proportional to both the phosphorus cell quota and the cell division rate. These uptake characteristics suggest that P. noctiluca may take up phosphate coincidently at their typically low ambient concentrations as well as to exploit episodic nutrient events in nature. Cell division rates were a hyperbolic function of the ambient orthophosphate concentration. The shortest doubling time was 8.7 d, the phosphate concentration at half the maximum division rate was 0.15 M and the threshold, concentration for cell division was ca 0.05 M PO 4 3- . Division rates of P. noctiluca in the ocean are much faster than predicted from the measured ambient orthophosphate concentrations. Since this dinoflagellate has high naturally occurring alkaline phosphatase activities, and can utilize organic-P compounds, we suggest that organic-P can be as important as orthophosphate in supporting the observed division rates of P. noctiluca in the sea.  相似文献

During early development in fish, phospholipase A2 (EC 3.1.1.4) regulates membrane lipid modifications, which relates to changes in environmental conditions and provision of fatty acids required for metabolic energy substrates and prostaglandin biosynthesis. A method to analyze phospholipase A2 in rat tissues has been modified to measure its activity in embryonic Atlantic halibut (Hippoglossus hippoglossus L.). Egg and embryo samples were collected during the 1994 spawning season. Enzyme activity was undetectable at fertilization but in 10-d embryos was 230 pmol mg−1 h−1 (at 20 °C) and increased by ∼120% at hatch (17-d). Significant alterations in the fatty acid composition of important phospholipids, phosphatidylcholine (PC) and phosphatidylethanolamine (PE), were also observed. The content of some critical polyunsaturated fatty acids, and the ratio of unsaturated/saturated fatty acids, declined significantly over development. Acyl-chain restructuring mediated through the activity of phospholipase A2, coupled with other observed lipid changes (significant increases in the PC/PE ratio and cholesterol content), would produce a decreased fluidity of membranes during embryonic development, coinciding with the predicted upward movement of larvae in the water column. Arachidonic acid (20:4n-6) removed from PE could serve as a precursor for biosynthesis of 2-series prostaglandins, and eicosapentaenoic acid (20:5n-3) from PC is a likely source for other prostaglandin types. Despite removal of polyunsaturated fatty acids, there was an overall increase in lipid and fatty acid concentration, which can be attributed to amino acid catabolism during early developmental stages. The present study examines for the first time the effects of increased salinity on water relations and osmolyte (carbohydrates and amino acids) concentrations in two Mediterranean seagrass species, Posidonia oceanica and Cymodocea nodosa, which are adapted to growth in environments with contrasting salinity and have a known differential sensitivity to alterations in ambient salinity. The specific aim was to obtain insights into their respective capacities to cope with natural or anthropogenically induced (e.g. desalination plants) hypersaline stress and its ecological implications. To this end, large plant fragments of both seagrass species were maintained for 47 days in a laboratory mesocosm system under ambient salinity (37 psu; control) and three chronic hypersaline conditions (39, 41 and 43 psu). Analyses of leaf-tissue osmolality indicated that both species followed a dehydration avoidance strategy, decreasing their leaf water potential (Ψw) as the external salinity increased, but using different physiological mechanisms: whereas P. oceanica leaves exhibited a reduction in osmotic potential (Ψπ), C. nodosa leaves maintained osmotic stability through a decrease in turgor pressure (Ψp) probably mediated through cell-hardening processes. Accordingly, the concentrations of soluble sugars and some amino acids (mainly Pro and Gly) suggested the activation of osmoregulatory processes in P. oceanica leaves, but not in C. nodosa leaves. Osmotic adjustments probably interfered with leaf growth and shoot survival of P. oceanica under hypersaline stress, whereas C. nodosa showed a more efficient physiological capacity to maintain plant performance under the same experimental conditions. These results are consistent with the more euryhaline ecological behaviour of C. nodosa and contribute to understanding the high vulnerability shown by P. oceanica to even mild increments in seawater salinity.  相似文献
