Different doses of arsenic (As) were used to investigate comparative toxicity on the liver and brain glycogen content on male and female Indian catfish Clarias batrachus (L.). As-induced effects were associated with gender, dose (5, 10, or 15?mg?L?1), and varying time periods (48, 96, or 144?h). It was noted that As produced dose- and time-dependent liver glycogenolysis and late brain glycogenolysis. Liver glycogenolysis was significantly increased after 48?h at all three As doses. At the highest dose 15?mg?L?1, liver glycogen were markedly diminished at in both male and female fish, but in females more reduction was observed than in males. However, with brain glycogen, the significant decrease was noted at 144?h with all three dose levels in both genders, with male being more susceptible. Thus, this study indicates that As produces glycogenolysis. The reduction in the liver glycogen content was more pronounced in female than in the male fish, whereas brain glycogen content decrease was more prominent in males.  相似文献

Fresh and healthy specimens of Oreochromis niloticus (mean weight 70.5± 1.23 g and mean length, 12.3± 1.5cm) and Clarias gariepinus (mean weight, 41.8±2.3 g and mean length 13.78 ± 1.2 cm) were exposed to heavy metal lead, ranging in concentration from 10 to 18 mg L‐1 for O. niloticus and 20 to 28 mg L‐1 for C. gariepinus respectively. The 96 h LC50 for both the species was computed as 12.45 mg L‐1 (O. niloticus) and 22.65 mg L‐1 (C. gariepinus). The cichlid fish was exposed to sublethal concentration of lead (2, 4 and 6 mgL‐1) and catfish (4, 8 and 12 mgL‐1). The erythrocytes count, haemoglobin concentration, haematocrit and the value of serum glucose increased significantly (P < 0.01), whereas leucocytes count, serum protein and serum triglyceride after intoxication declined in both the species with the same pattern but in different quantity. After 96 h of time a significant depletion of muscle and liver glycogen was noted, being greater in muscle (16.7% in O. niloticus) and in liver (15.9% in C. gariepinus). The metabolism of 14C-labelled lactate was investigated in the horseshoe crab Limulus polyphemus and the stone crab Menippe mercenaria. When a bolus of (14C-U)-D-lactate was injected into L. polyphemus, there was substantial release pf 14CO2 into the medium. In the case of M. mercenaria, 14CO2 release was also observed after injection of (14C-U)-L-lactate into experimental individuals. Analysis of the distribution of radioactivity in whole body extracts of both species revealed easily detectable amounts of radioactivity in the glycogen fraction, although the bulk of the radioactivity was in the cation, anion/neutral and CO2 fractions. To investigate the metabolism of lactate further, 14C-labelled lactate was injected into large individuals of L. polyphemus and M. mercenaria, and the distribution of radioactivity was determined in the hemolymph, muscle and hepatopancreas. Utilization of (14C-U)-D-lactate by L. polyphemus resulted in the accumulation of significant amounts of labelled glucose in all three body compartments as well as the production of labelled glycogen in the telson levator muscle and hepatopancreas. Utilization of (14C-U)-L-lactate resulted in a similar pattern of glucose and glycogen labelling in the hemolymph, cheliped muscle and hepatopancreas of M. mercenaria. These studies demonstrate that both L. polyphemus and M. mercenaria have the capacity for glyco- and gluconeogenesis using lactate as the substrate.  相似文献

Metabolic changes and toxic effects in Crangon crangon (collected during summer 1989 near Helsingør, Denmark and at Kulhuse in the Isefjord) were observed during exposure to different arsenate concentrations. Survival, respiration, and concentrations of hemolymph glucose, muscle glycogen andadenine nucleotides were measured. C. crangon was very tolerant of arsenate; survival was only affected at arsenate concentrations >25 ppm. Small individuals (0.2 to 0.29 g wet weight) were less tolerant than medium-sized and large individuals (0.5 to 0.59 g and 0.8 to 0.89 g wet weight), e.g. at 50 ppm the LT50 for small individuals was 180 h compared to 343 h and 360 h in middle-sized and large individuals. The relation between size and toxicity is probably due to the change in surface/volume ratio. Under clean conditions the respiration of C. crangon showed a clear relation between routine MO2 (O2 in mlg-1 h-1) and wet weight (r2=0.89; P<0.001). During shortterm exposure to arsenate (10 and 50 ppm), the respiratory rates (MO2) seemed to decrease in a size-dependent manner. The concentrations of blood glucose and muscle glycogen did not change in response to arsenate exposure. The concentrations of adenylate phosphates were high and constant throughout the experiments. C. crangon had maximal adenylate energy charge (AEC) values of ca. 0.8. No decrease in AEC due to the exposure to arsenate was observed.  相似文献

Specimens of the mudskipper Boleophthalmus boddaerti, collected along the estuarine canal at Pasir Ris, Singapore between April 1987 and December 1989, were examined in the laboratory. After being exposed to environmental hypoxia for 6 h, no accumulation of lactate, alanine or succinate was observed in the muscle tissue of B. boddaerti. In addition, the blood lactate content and muscle creatine phosphate (CrP), adenosine triphosphate (ATP) and glycogen contents were not significantly different from those of the control. During normoxic recovery, oxygen debt repayment was only a small fraction of the oxygen deficit incurred during the 6 h of hypoxic exposure. Therefore, it would appear that B. boddaerti coped with environmental hypoxia by undergoing metabolic rate reduction. After anoxic exposure, the CrP, ATP and adenosine diphosphate (ADP) contents in the muscle tissue of B. boddaerti were significantly lower than the corresponding control values. Lactate accumulated in the blood and muscle tissue of the anoxic fish though the muscle energy charge and glycogen content remained constant. Hence, B. boddaerti was able to cope with such a period of anoxia without increasing its glycolytic rate. Succinate, which was not detected in the muscle tissue of the control and hypoxia-exposed fish, accumulated in significant quantities in the muscle tissue of B. boddaerti exposed to environmental anoxia and functional hypoxia. In the case of functional hypoxia, there was a significant decrease in the muscle glycogen content, and the muscle lactate content increased 17.3-fold, indicating that glycolysis was activated to provide the source of energy during muscular exercise.  相似文献

InEmerita asiatica, the quantitative fluctuations in the glycogen content of the hepatopancreas were markedly related to the moult cycle. The glycogen content of the hepatopancreas was maximum during premoult stages. It has been suggested that reserve glycogen, in addition to meeting general metabolic needs, may be especially involved in the genesis of the sugar precursors of chitin. The marked fall in the hepatopancreas glycogen values following moulting is probably due to its utilization in chitin synthesis during and immediately following moulting. InE. asiatica, the storage of glycogen was found to be meagre in muscles, and muscle glycogen had no bearing on chitin synthesis during moulting. InLigia exotica, glycogen is stored to a lesser extent than inE. asiatica, and was found in connective tissue and muscles. Significant fluctuations were noted in glycogen values correlated with the phases in the moult cycle when chitin synthesis took place. InL. exotica, the hepatopancreas does not serve as a storage depot for glycogen related to chitin synthesis.  相似文献

Knowledge of the structure and energy reserves, in the liver of commercially important fish species, is important in understanding metabolic processes and in assessing the impact of potential environmental physical and chemical stressors in both wild and cultured stocks. The present study investigated the microscopic morphology and histochemistry (total and neutral lipids, glycogen) of liver tissue of wild (3 +) and cultured (1 +) sexually immature female and male yellowtail flounder ( Limanda ferruginea Storer), sampled in late April 2001. Hepatosomatic indices [HSI: (liver weight/body weight-liver weight)×100] of cultured fish were significantly higher than those of wild fish. Females in the cultured group had significantly lower HSIs than males. The liver of both wild and cultured L. ferruginea was interspersed with pancreatic tissue. The main components of the liver tissue were irregular cords of hepatocytes arranged in tubules which surrounded vascular sinusoids. The hepatocytes contained an abundance of lipid, much of which appeared to be neutral lipids, in both sexes of the cultured fish. Total and neutral lipid droplets were larger, and the area occupied by these droplets was significantly greater in both cultured females and males compared to the wild fish, suggesting lipidosis in the cultured fish. In the cultured fish these differences were sex-dependent, the females having significantly more total and neutral lipids in hepatocytes than the males. This suggests a potentially greater storage capacity in females and/or a higher lipid metabolism in males. There were no statistically significant differences in glycogen content between the cultured and wild fish, or between the sexes in both sampling groups.  相似文献

The carbohydrate and oxidative metabolism of the lateral musculature and of the liver was investigated in two Black Sea fishes. In Trachurus mediterraneus ponticus, muscular work leads to an intensification of glycolysis in the white muscle, and of oxidation in the red muscle as well as in the liver. Extirpation of the lateral red muscle provokes strong disturbances of muscle performance (without affecting the double character of contraction); the glycogen content of the white muscle decreases, while the lactate content of the liver increases. Hypoxic conditions lead, within a short time, to disturbances of the oxidative capacity in the red muscle and in the liver. In Gobius melanostomus, the effects of hypoxy or of muscular work are less pronounced. In isolated mixed muscle strips of Trachurus, contractions lead to a decrease of glycogen content in the red portion only. In isolated red muscle strips, the glycogen content rises during the incubation. These results are interpreted as supporting the hypothesis of a metabolic role of the lateral red muscle in fishes as energy source for the white muscle and as oxidative site for the catabolites of the latter.  相似文献

The effects of deltamethrin at acute and sublethal concentrations on some hematological and biochemical profiles in the Indian major carp Labeo rohita have been examined. The lethal concentrations (LC50) of deltamethrin for 24 and 96 h were found to be 0.44 and 0.38 mg L?1, respectively. During acute treatment (24 h), hemoglobin (Hb), hematocrit (Hct), red blood cell (RBC), white blood cell (WBC), mean cellular volume (MCV), mean cellular hemoglobin (MCH), and mean cellular hemoglobin concentration (MCHC) values were significantly increased compared to control group. Plasma glucose, protein, cholesterol, albumin, globulin, triglyceride, and glycogen in gill and muscle were significantly decreased. Upon sublethal treatment (0.044 mg L?1, 10th of 24 h LC50), RBC and the biochemical parameters, except glucose and cholesterol, decreased significantly, while MCV, MCH, and MCHC values increased up to the 28th day and then declined. Cholesterol level was significantly increased throughout the study period. A biphasic response in the levels of Hb, Hct, WBC, and glucose were observed during sublethal treatment.  相似文献

Nodularin (NODLN) is a cyclic pentapeptide hepatotoxin produced by the cyanobacterium Nodularia spumigena, which forms extensive blooms during the summer in the Baltic Sea. Nodularin was detected in liver, muscle and/or feather samples of several common eiders (Somateria mollissima) from the Gulf of Finland (northern Baltic Sea) in 2002–2005. Published information on the adverse effects of NODLN in marine birds is scarce. The aim of this study was to evaluate the toxicity of NODLN, and determine the concentrations of NODLN in liver and muscle tissue in mallards (Anas platyrhynchos) exposed to N. spumigena. Mallards received a single or multiple exposure via oral gavage with an aqueous slurry containing toxic N. spumigena. Dosages ranged from 200 to 600 µg NODLN per kg body weight (bw). There were minimal histopathological changes in liver tissue, and brain cholinesterase activity did not differ among treatment groups. Concentrations of NODLN measured by LC-MS in liver varied between approximately 3–120 µg kg?1 dry weight (dw) and ducks receiving multiple exposures had significantly greater liver toxin levels than ducks receiving the two lowest single exposures. In muscle, NODLN concentrations were approximately 2–6 µg kg?1 dw, but did not differ significantly among exposure groups. This is the first in vivo lab study examining the effects and bioaccumulation of NODLN from N. spumigena in birds. The mallards in this study were resistant to adverse effects and did not bioaccumulate substantial levels of NODLN at the doses given.  相似文献

In the present study, an attempt has been made to quantify the fenvalerate accumulated in different tissues (gill, muscle and liver) and observe changes involved in the levels of sodium, potassium and calcium ions and Na+–K+, Mg2+ and Ca2+ adenosine triphosphatase (ATPase) activities in the freshwater fish, Cirrhinus mrigala on short-term and long-term exposure to the median lethal and sublethal concentration of fenvalerate. Residue analysis using gas–liquid chromatography (GLC) technique revealed that fenvalerate accumulated in highest quantity in gill followed by liver and muscle under median lethal concentration (6?µg?L?1). Whereas in sublethal concentration (0.6?µg?L?1), muscle accumulated highest quantity followed by gill and liver, which might be due to the fact that fenvalerate is highly lyphophilic. The ion concentration and ATPase activity were found effected in fish exposed to lethal and sublethal concentrations of fenvalerate. Concentration of Na+, K+ and Ca2+ ions decreased in gill, muscle and liver on being exposed to median lethal concentration to a significant level. Whereas the changes were not highly pronounced at sub lethal level indicating low concentration of fenvalerate and its non-toxic effect at chronic exposure. Na+–K+, Mg2+ and Ca2+ ATPases activity were also found decreased in correspondence to the ionic change under median lethal and sub lethal concentrations in target tissues. This might have lead to behavioural changes and create wide-spread disturbance in the normal physiology, ultimately causing the death of the fish. The results suggest that in biomonitoring programmes, ions and associated ATPases can be a good diagnostic tool for fenvalerate toxicity.  相似文献

Leatherback turtles, Dermochelys coriacea, are highly migratory, spending most of their lives submerged or offshore where their feeding habits are difficult to observe. In order to elucidate the foraging ecology of leatherbacks off Massachusetts, USA, stable isotope analyses were performed on leatherback tissues and prey collected from 2005 to 2009. Stable isotope ratios of nitrogen and carbon were determined in whole blood, red blood cells, blood plasma, muscle, liver, and skin from adult male, female, and subadult leatherbacks. Isotopic values were analyzed by body size (curved carapace length) and grouped by sex, and groups were tested for dietary differences. Gelatinous zooplankton samples were collected from leatherback foraging grounds using surface dip nets and stratified net tows, and prey contribution to leatherback diet was estimated using a two-isotope Bayesian mixing model. Skin and whole blood δ13C values and red blood cell δ15N values were correlated with body size, while δ13C values of red blood cells, whole blood, and blood plasma differed by sex. Mixing model results suggest that leatherbacks foraging off Massachusetts primarily consume the scyphozoan jellyfishes, Cyanea capillata and Chrysaora quinquecirrha, and ctenophores, while a smaller proportion of their diet comes from holoplanktonic salps and sea butterflies (Cymbuliidae). Our results are consistent with historical observations of leatherback turtles feeding on scyphozoan prey in this region and offer new insight into size- and sex-related differences in leatherback diet.  相似文献

A study on the toxicokinetic behavior, metabolism of chlorpropham, and its effect on cytochrome P450 from liver microsomes was carried out in albino rats after a single and consecutive oral administration at 500?mg?kg?1 body weight for 10 and 20 days. Chlorpropham was detected in the blood at 0.08?h (11.43?±?1.72?µg?mL?1) reaching a maximum concentration at 2?h (30.90?±?2.55?µg?mL?1) and a minimum at 48?h (1.95?±?0.20?µg?mL?1) after a single oral administration of 500?mg?kg?1. The absorption rate constant (K a) was 0.66?±?0.48?h?1. The Vd area (18.01?±?2.78?L?kg?1) and t 1/2 β (12.23?±?1.96?h) values suggested a wide distribution and long persistence of the compound in the body, respectively. The higher ClR (0.82?±?0.00?L?kg?1?h?1) compared to ClH (0.18?±?0.02?L?kg?1?h?1) value indicated that a major portion of chlorpropham was excreted through the urine (30%) compared to the faeces (2.81%). Chlorpropham residue was detected in all tissues of rat at 0.25?h while its metabolite, meta-chloroaniline was detected in liver, kidney, heart, lung, and spleen tissue at 0.25?h. Meta-chloroaniline was not detected in skeletal muscle, brain, fat, and stomach tissue at any time of the observation period. Maximum concentrations of chlorpropham and meta-chloroaniline were detected at 2?h (except in the spleen), and minimum concentrations of chlorpropham at 24 (heart, lung, spleen, skeletal muscle, and stomach) and 48?h (liver, kidney, brain, and fat tissue) respectively; and meta-chloroaniline at 24?h (except heart and spleen). The tissue half-life of chlorpropham in rat varied from 3.80 to 11.60?h. Repeated oral administration of chlorpropham at 500?mg?kg?1 for 10 and 20 days caused an induction of the liver microsomal pellet of rat.  相似文献

为了解全氟辛烷磺酸盐(perfluorooctane sulfonate, PFOS)暴露对半滑舌鳎(Cynoglossus semilaevis)免疫功能的影响,在实验室条件下,运用RT-PCR方法分析了PFOS暴露对半滑舌鳎热休克蛋白hsp70、hsp90、C型凝集素(c-type lectin)和细胞色素c氧化酶(cytochrome c oxidase, cox)等4种免疫相关基因表达水平的影响。实验测定了上述4种基因在半滑舌鳎肝、鳃、肠及肌肉4种不同组织中随时间(0、24 h、48 h、96 h和7 d)的表达变化情况。结果表明,在4种组织中,hsp70基因的表达与对照相比为上调,其中,肝组织hsp70基因的表达量显著高于其他各组织,且表达高峰值的出现也早于其他各组织;hsp90基因在肝和鳃组织中表达量随时间不同而波动,在肠组织中表达上调,在肌肉中表达显著下调;c-type lectin基因表达量与对照组相比表达显著下调或无明显差异;cox基因在肝组织和肠组织中表达下调,在鳃和肌肉中表达上调。上述研究结果表明,PFOS能引起免疫相关基因的表达变化,对半滑舌鳎具有潜在的免疫毒性。肝组织中各免疫基因对PFOS胁迫的响应高于其他组织。本研究可为阐明全氟辛烷磺酸盐对半滑舌鳎的免疫毒性提供基础数据。  相似文献

Several carbohydrate metabolism pathways were studied in four different tissues (liver, gonad, kidney and white muscle) of female turbot (Scophthalmus maximus) at three different stages of gonad maturation, i.e., previtellogenesis, early vitellogenesis and late vitellogenesis. During carbohydrate metabolism in the liver, the only major change noticed throughout vitellogenesis was increased glycolytic activity at early vitellogenesis. The major changes observed in the gonad during vitellogenesis were an increased use of exogenous glucose and increased gluconeogenesis, which were reflected in increased glucose levels throughout vitellogenesis accompanied by a small increase in glycogen levels. Changes observed in carbohydrate metabolism in the kidney pointed to increased gluconeogenesis during early vitellogenesis. Finally, during ovarian maturation, the muscle mobilized proteins whose constituent amino acids could have been used in other tissues either as substrates for gluconeogenesis or for protein synthesis. As an overall conclusion, changes in the carbohydrate metabolism of female turbot during maturation are quite marked in the gonad, kidney and white muscle, in contrast to other species, in which major changes have been previously reported to occur in the liver.  相似文献

The influence of dietary copper (Cu) exposure on accumulation, growth, and hematological parameters was investigated in Cyprinus carpio after sub-chronic ingestion of 0, 250, 500, 750 or 1000?mg?kg?1 for 60 days. The profile of Cu accumulation among tissues in C. carpio was dependent on the exposure period and Cu concentration. Liver of C. carpio was the predominant storage tissue and the order of Cu accumulation in tissues was liver?>?intestine?> gill?>?kidney?>?muscle. Cu concentration at >125?mg?kg?1 reduced growth rate, and was inversely related to growth. The RNA?:?DNA ratios were not affected by exposure and there was no correlation between growth rate and RNA?:?DNA ratio in liver and muscle. There were no significant effects of exposure on blood parameters except for magnesium. Cu exposure time and dose increased the serum glutamic-oxaloacetic transaminase (SGOT) and serum glutamic-pyruvic transaminase (SGPT) activity levels.  相似文献

A method has been developed to isolate glycogen cells (=vesicular connective tissue cells) from the labial palps of the marine mussel Mytilus edulis L. These cells have a modal density of 1.14 g ml-1 and this property has been exploited to separate them from pronase-dissociated cell dispersions by density-gradient centrifugation. The isolated cells were mainly spherical, with a small peripheral nucleus. The quasitotality of the cell was filled with -like glycogen particles with cellular organelles being limited to the cell periphery. The major recipients of [U-14C] glucose-derived carbon were glycogen and amino acids. Only small quantities of 14C radiolabel were recovered from lipids, protein and CO2. Glucose incorporation into glycogen increased linearly over 6 h and showed saturation kinetics with respect to exogenous glucose concentration.  相似文献

Some experiments have been made on the metabolism of 237Pu — a high specific activity, gamma-emitting isotope of plutonium — by the thornback ray Raja clavata L. Unlike previous experiments with the plaice (Pleuronectes platessa L.) it was found that the thornback ray absorbed plutonium across the gut wall, as readily observed by analysis of the liver. The liver accumulated up to 0.2% of the 237Pu given in a single labelled meal after a period of 12 days. Direct injection of the isotope into the muscle of thornback rays resulted in extremely slow rates of elimination from the fish, the highest internal concentrations being attained by the spleen. The largest fractions of the whole-body burden, however, were those of liver and skeleton.  相似文献

The present study is a comprehensive comparison of the biochemical composition (protein, lipid, glycogen and cholesterol contents, and amino acid and fatty acid profiles) of 14 species of cephalopods with different life strategies (benthic, nektobenthic, benthopelagic and pelagic) in distinct habitats (neritic, demersal, oceanic and deep sea), with special emphasis placed on a male Architeuthis sp. The giant squid showed a significantly lower protein and total amino acid content in the gonad and digestive gland. The major essential amino acids were leucine, lysine and arginine. The major non-essential amino acids were glutamic acid, aspartic acid and proline. The benthopelagic cirrate octopus Opisthoteuthis sp. showed a lower content of these nitrogen compounds in the muscle. In respect to lipid and fatty acid contents, the giant squid showed the highest values in the gonad and digestive gland and the lowest in the muscle. Most of saturated fatty acid content was presented as 16:0; monounsaturated fatty acid content, as 18:1 and 20:1; and polyunsaturated fatty acid content, as 20:4n-6, 20:5n-3 and 22:6n-3. The highest cholesterol content in the gonad was detected in Opisthoteuthis sp. and Architeuthis sp.; in the digestive gland. in Todarodes sagittatus; and in the muscle, in Sepia elegans. The highest glycogen value in the gonad was detected in Octopus vulgaris; in the digestive gland and muscle, the highest values were attained in Opisthoteuthis sp. The relationships between life strategies and biochemical composition were investigated and principal component analysis (PCA) was performed. The different cephalopod life strategies could be well separated on the basis of the first two principal components. The nektobenthic and pelagic strategies were clearly separated from the benthic, suggesting that these groups of species are characterized by lower lipid content and higher protein content in the gonad. A rather similar life-strategy distinction was obtained for the digestive gland. The benthopelagic strategy was also well separated from benthic and pelagic strategies and from Architeuthis sp. In the muscle, the results indicated lower nitrogen and carbon compound contents in Architeuthis sp. and Opisthoteuthis sp. The environmental conditions that possibly explain the Architeuthis sp. stranding, the relationships between biochemical compositions and the life strategies of the different cephalopod species studied are discussed.Communicated by S.A. This study aims to describe the variability of albacore (Thunnus alalunga) diet in the Northeast Atlantic and Mediterranean Sea and to identify possible relationships between this variability and the features of different feeding areas, the behavior, and the energetic needs of albacore. Stomach contents from albacore caught in five zones of the Bay of Biscay and surrounding waters (n = 654) and three zones of the Mediterranean Sea (n = 152) were analyzed in terms of diet composition and stomach fullness. Carbon and nitrogen stable isotope and C/N ratios were measured for white muscle and liver from albacore in the Bay of Biscay (n = 41) and Mediterranean Sea (n = 60). Our results showed a spatial, seasonal, inter-annual, and size-related variability in the diet of albacore. Albacore diet varied by location in the Mediterranean Sea, with a particularly high proportion of cephalopods, and low δ15N values in the Tyrrhenian Sea. In the Northeast Atlantic, albacore consumed a higher proportion of crustaceans and a lower proportion of fishes in the most offshore sampling zone than inshore. The digestion states of the major prey reflected a diurnal feeding activity, indicative of feeding in deeper waters offshore, whereas on the continental slope, feeding probably occurred in surface waters at night. Important seasonal and inter-annual diet variability was observed in the southeast of the Bay of Biscay, where preferred albacore prey appeared to be anchovy (Engraulis encrasicolus). Stomach fullness was inversely related to body size, probably reflecting higher energetic needs for smaller individuals. Albacore from the Bay of Biscay had significantly lower δ13C and higher δ15N values compared with albacore from the Mediterranean Sea, indicative of regional baseline shifts, and trophic position and muscle lipid stores in albacore increased with body size.  相似文献
