In this study, Bacillus cereus isolate from tannery effluent was employed for orange II dye decolorization in simulated minimal salt broth and textile effluent. Most of the physicochemical parameters of textile effluent were above the permissible limits. The strain was highly tolerant to dye up to 500 mg l?1. Increasing dye concentration exerted inhibitory effect on the bacterial growth and decolorization. The maximum decolorization of initial 100 mg dye l?1 was achieved at optimum pH 8.0 and 33 °C under static culture conditions during 96-h incubation. Supplementation with optimized glucose (0.4 %, w/v) and ammonium sulfate (0.1 %, w/v) with 3.0 % B. cereus inoculum further enhanced dye decolorization to highest 68.5 % within 96-h incubation. A direct correlation was evident between bacterial growth and dye decolorization. Under above optimized conditions, 24.3 % decolorization of unsterilized real textile effluent by native microflora was achieved. The effluent decolorization enhanced substantially to 37.1 % with B. cereus augmentation and to 40.5 % when supplemented with glucose and ammonium sulfate without augmentation. The maximum decolorization of 52.5 % occurred when textile effluent was supplemented with optimized exogenous carbon and nitrogen sources along with B. cereus augmentation. Gas chromatography–mass spectrometry identified sulfanilic acid as orange II degradation product. Fourier transform infra red spectroscopy of metabolic products indicated the presence of amino and hydroxyl functional groups. This strain may be suitably employed for in situ decolorization of textile industrial effluent under broad environmental conditions. 相似文献

Two novel and facile ligands ammonium piperidine dithiocarbamate (Amm Pip-DTC) and ammonium morpholine dithiocarbamate (Amm Mor-DTC) were synthesized for the development of rapid and cost effective catalytic hydrogen current technique to analyze cobalt(II) in the presence of NH(4)Cl-NH(4)OH at pH 7.8 and 8.4 with Amm Pip-DTC and Amm Mor-DTC. These ligands produce catalytic hydrogen currents with Co(II) at peak potentials -1.24 V and -1.44 V vs. SCE respectively. Quantitative experimental conditions are developed by studying effect of pH, supporting electrolyte (NH(4)Cl), ligand and metal ion concentrations and effect of adverse ions on peak height to improve the sensitivity, selectivity and detection limits of the catalytic hydrogen current technique and compared it in terms of Student's t test and variance ratio f test with differential pulse polarographic (DPP) method. The developed technique was applied for the analysis of cobalt(II) in various water samples, agricultural materials and pharmaceuticals and the results obtained are in good agreement with the DPP data. 相似文献

This work proposes the quantification of Cr (VI) ions in natural waters in trace level, using activated alumina (Al2O3) as preconcentration support, controlled in-line dissolution of the solidified chromophore diphenylcarbazide after heat treatment and spectrophotometric detection. The manifold ensures high sensitivity of analytical response, good repeatability, and stability. In this work, optimization of experimental conditions of a flow injection system was chosen as the parameters for greater sensitivity and better selectivity. The selected optimized conditions were 0.30 mol L?1 for H2SO4 concentration, system flow rate as 0.40 mL min?1, sample injection volume of 192.50 μL, 2 min for preconcentration time, and 0.10 mol L?1 for eluent concentration. The analytical curves obtained for real sample analysis show linear range from 0.192 to 0.961 μM, linear correlation coefficient R?=?0.9997 and LOD?=?0.024 μM. The preconcentration factor of about four times was obtained through the passage of 800 μL of a standard solution containing 0.961 μM of Cr (VI) through mini-column of preconcentration followed by elution at 192.5 μL of NH4OH 0.1 mol L?1 solution. The solid chromogenic reagent presented high durability (weeks in daily use with mass of 0.0993 g) and good reproducibility in analytical signal. The reactivation of the mini-column of alumina should be executed after ten injections of eluent, using 800 μL of HCl 0.02 mol L?1 solution in flow through the same. Each cycle of injection and elution of the sample takes about 5 min on the proposed terms. Despite the length of each cycle still be high, low concentrations can be detected using a technique of relatively low cost. This is due in part, the association dissolution of the chromogenic reagent directly in the line and the preconcentration step. Another important factor is the economy of reagent chromogenic, low generation of reject contributing to better quality of the environment, and the high potential for applications to work in field. 相似文献

Physical, chemical and microbiological efficiencies of Sewage Treatment Plants (STPs) located in Delhi's watershed in context of different treatment technologies employed in these plants have been determined. There were in all seventeen STPs treating domestic wastewater which were studied over a period of 12 months. These STPs were based on Conventional Activated sludge process (ASP), Extended aeration (Ex. Aeration), physical, chemical and biological removal treatment (BIOFORE) and oxidation pond treatment process. Results suggests that except "Mehrauli" STP which was based on Extended aeration process and "Oxidation pond", effluents from all other STPs exceeded FC standard of 10(3) MPN/100 ml for unrestricted irrigation criteria set by National river conservation directorate (NRCD). Actual integrated efficiency (IE(a)) of each STP was evaluated and compared with the standard integrated efficiency (IE(s)) based upon physical, biological and microbiological removal efficiencies depending upon influent sewage characteristics. The best results were obtained for STPs employing extended aeration, BIOFORE and oxidation pond treatment process thus can be safely used for irrigation purposes. 相似文献

Stable colloidal silver nanoparticles (AgNPs) were synthesized using Caulerpa serrulata (green marine algae) aqueous extract as an efficient reducing and stabilizing agent. This method is considered to be a sustainable alternate to the more complicated chemical procedures. To achieve the optimization synthesis of AgNPs, several effects such as extract concentration, contact time, pH values, and temperature were examined. The synthesized AgNPs were characterized by UV–Vis spectroscopy, FT-IR, XRD, and HR-TEM. The synthesized AgNPs showed an intense surface plasmon resonance band at 412 nm at the optimal conditions (20% (v/v) extract and 95 °C). TEM reveal that higher extract concentration and higher temperature leading to the formation of spherical AgNPs with an average particle size of 10 ± 2 nm. The synthesized AgNPs showed excellent catalytic reduction activity of Congo red (CR) dye from aqueous solutions. The degradation percentage of CR with AgNPs accelerated by increasing either NaBH4 concentration or catalytic dosage. The AgNPs synthesized at higher temperature (e.g., 10Ag-95) exhibited the highest catalytic activity. The reaction kinetics was found to be pseudo first order with respect to the dye concentration. Moreover, the AgNPs displayed antibacterial activity at lower concentration against Staphylococcus aureus, Pseudomonas aeruginosa, Shigella sp., Salmonella typhi, and Escherichia coli and may be a good alternative therapeutic approach. The outcomes of the current study confirmed that the synthesized AgNPs had an awesome guarantee for application in catalysis and wastewater treatment. 相似文献

We propose to combine a passive sampler for polar organic compounds (POS) with a specific bioassay for phytotoxicity to assess the hazard of herbicidal compounds in surface waters. The POS consisted of an Empore disk coated with styrenedivinylbenzene deployed in a Teflon housing, which has relatively high sampling rates (e.g., approximately 1 L d(-1) for diuron). POS were deployed for 5 days in a small-scale field study in South East Queensland, Australia, in a relatively pristine environment and an urban environment to explore sensitivity towards herbicides and potential influences of non-herbicidal pollutants. Besides chemical analysis of 8 herbicides, a novel bioassay (Maxi-Imaging-PAM, IPAM) was employed to assess the phytotoxic effects of water samples and POS extracts. The IPAM allows rapid assessment of photosynthetic quantum yields of a large number of samples via chlorophyll-fluorescence imaging and the saturation pulse method. Sampling rates for several herbicides from laboratory calibrations were found to be applicable under field conditions. Toxic equivalent concentrations (with reference to the herbicide diuron) were computed from the concentrations determined by chemical analysis and the relative potency (also termed toxic equivalence factor) of the detected herbicides. There was good agreement between diuron equivalent concentrations from chemical analysis and diuron equivalent concentrations determined with the IPAM. 相似文献

Suitable techniques have been developed for the extraction of arsenic species in a variety of biological and environmental samples from the Pak Pa-Nang Estuary and catchment, located in Southern Thailand, and for their determination using HPLC directly coupled with ICP-MS. The estuary catchment comprises a tin mining area and inhabitants of the region can suffer from various stages of arsenic poisoning. The important arsenic species, AsB, DMA, MMA, and inorganic arsenic (As III and V) have been determined in fish and crustacean samples to provide toxicological information on those fauna which contribute to the local diet. A Hamilton PRP-X100 anion-exchange HPLC system employing a step elution has been used successfully to achieve separation of the arsenic species. A nitric acid microwave digestion procedure, followed by carrier gas nitrogen addition- (N2)-ICP-MS analysis was used to measure total arsenic in sample digests and extracts. The arsenic speciation of the biological samples was preserved using a Trypsin enzymatic extraction procedure. Extraction efficiencies were high, with values of 82-102%(As) for fish and crustacean samples. Validation for these procedures was carried out using certified reference materials. Fish and crustacean samples from the Pak Pa-Nang Estuary showed a range for total arsenic concentration, up to 17 microg g(-1) dry mass. The major species of arsenic in all fauna samples taken was AsB, together with smaller quantities of DMA and, more importantly, inorganic As. For sediment samples, arsenic species were determined following phosphoric acid (1 M H3PO4) extraction in an open focused microwave system. A phosphate-based eluant, pH 6-7.5, with anion exchange HPLC coupled with ICP-MS was used for separation and detection of AsIII, AsV, MMA and DMA. The optimum conditions, identified using an estuarine sediment reference material (LGC), were achieved using 45 W power and a 20 minute heating period for extraction of 0.5 g sediment. The stability and recovery of arsenic species under the extraction conditions were also determined by a spiking procedure which included the estuarine sediment reference material. The results show good stability for all species after extraction with a variability of less than 10%. Total concentrations of arsenic in the sediments from the Pak Pa-Nang river catchment and the estuary covered the ranges 7-269 microg g(-1)and 4-20 [micro sign]g g(-1)(dry weight), respectively. AsV was the major species found in all the sediment samples with smaller quantities of AsIII. The presence of the more toxic inorganic forms of arsenic in both sediments and biota samples has implications for human health, particularly as they are readily 'available'. 相似文献

This study reports a very selective, easy, and precise method for rapid separation of trace amounts of copper in aqueous samples using octadecyl silica-bonded phase membrane disks modified by 2,2'-[ethane-1,2-diylbis(thio)]dianiline (EDTD) combined with flame atomic absorption spectrometric determination. In addition, the synthesis and spectral characterization of EDTD have been described in detail. All the affecting experimental variables such as pH, amount of modifier, eluent type, sample and eluent flow rate, interfering ions, and disk capacity were also investigated. The target analyte (trace copper) was quantitatively retained at pH?=?4 and eluted with 6.0 mL of 0.5 M HNO3 at flow rates of 40 and 10 mL min?1 for analyte passage and elution steps, respectively, through the disks modified with 17.0 mg of EDTD. The proposed method also allows an enrichment factor of about 500 and has a detection limit of 0.005 ng mL?1. The method has been successfully applied for isolation and determination of copper in different water samples, peppers, and standard alloys. 相似文献
