海洋菌株y3的分离鉴定及其异养硝化-好氧反硝化特性   总被引:5,自引:4,他引:5  
从胶州湾海底沉积物中筛选出1株高效的海洋异养硝化-好氧反硝化细菌y3,经形态学观察、生理生化实验和16S rRNA基因序列分析,确定该菌株为假单胞菌属(Pseudomonas sp.).对其在实际含氮海水中的脱氮实验结果表明,菌株y3的最佳碳源为柠檬酸三钠,最适p H为7.0,最适C/N为13;菌株均能以NH4Cl、Na NO_2和KNO_3为唯一氮源进行反应,20 h后其去除率分别为98.69%、78.38%和72.95%,在硝化过程中几乎没有亚硝酸盐和硝酸盐的积累.以不同比例混合两种氮源反应20 h,当NO~-_3-N∶NO~-_2-N分别为2∶1、1∶1和1∶2时,脱氮率分别为99.56%、99.75%和99.41%;当NH~+_4-N∶NO~-_3-N分别为2∶1、1∶1、1∶2时,脱氮率均为100%;当NH~+_4-N∶NO~-_2-N分别为2∶1、1∶1、1∶2时,脱氮率分别为90.43%、92.79%和99.96%,多高于单一氮源的情况.该菌株具有较好的高盐废水脱氮处理效能.  相似文献

为探究进水流量分配比对低温城市污水脱氮效率的影响,并解析多级AO耦合流离生化工艺脱氮规律,实验采用三级AO耦合流离生化工艺,在温度为(10±1)℃、水力停留时间8 h、气水比恒定的条件下处理模拟低C/N值城市污水.系统依次在进水比5∶4∶4(等容积负荷)、3∶2∶1(等停留时间)和25∶15∶6(等污泥负荷)这3种工况下运行.结果表明,该工艺对低温低C/N值污水的处理效果较好,其中在进水比为3∶2∶1的工况下脱氮效率最高,COD、NH+4-N和TN平均去除率分别为87. 44%、96. 63%和76. 81%.进一步对氮的迁移转化规律进行研究发现,制约工艺低温脱氮的主要因素为各级硝化效率,3∶2∶1的进水比合理地分配了进水负荷,各级硝化率均超过85%,为反硝化创造了有利条件,最终获得了较高的脱氮效率,此时系统也具有最高的总生物量.研究结果丰富了多级AO耦合工艺低温脱氮理论,同时为工程设计应用提供参考.  相似文献

为了考察硫铁比对反硝化脱氮同步除磷效果的影响,进行了不同硫铁比的反硝化脱氮除磷静态实验,并对复合填料系统反硝化脱氮同步除磷作用进行了分析.结果表明,硫铁复合填料的脱氮除磷效果均显著高于单一填料;硫铁比是影响复合填料反硝化脱氮除磷效果的一个关键因素,当硫铁比(体积比)大于等于1∶1时,TN、TP去除率分别达到了85%和97%以上.复合填料脱氮除磷过程均满足二级动力学方程,系统脱氮作用主要依赖于异养反硝化和硫自养反硝化过程,而除磷主要由于海绵铁腐蚀产生的化学除磷作用.  相似文献

针对低碳氮比猪场废水传统脱氮法碳源不足的问题,采用SBBR反应器进行短程硝化反硝化-厌氧氨氧化联合脱氮.实验表明,短程硝化反硝化预处理可为厌氧氨氧化创造良好的进水条件;经预处理的猪场废水厌氧氨氧化脱氮效果显著,氨氮、亚硝态氮和总氮的平均去除率分别为91.8%、 99.3%、 84.1%,废水中残留有机物未对厌氧氨氧化效果产生明显影响,氨氮、亚硝态氮、硝态氮平均变化量之比为 1∶1.21∶0.24.色质联用分析结果显示,猪场废水中有机物成分在厌氧氨氧化反应前后未发生明显变化,主要化合物为酯类和烷烃类物质;特殊功能菌种检测结果表明,实验条件下的微生物系统是一个厌氧氨氧化菌与硝化菌、亚硝化菌和反硝化菌共存的系统,厌氧氨氧化菌是该系统主要脱氮功能菌.  相似文献

化学沉淀法去除稀土湿法冶炼废水中钙与高浓度氨氮研究   总被引:1,自引:0,他引:1  
离子型稀土湿法冶炼过程中会产生大量氨氮废水,由于废水中含有大量Ca2+,而Ca2+是影响磷酸铵镁沉淀法脱氮效率的重要因素.向废水中投入Na2CO3固体生成CaCO3沉淀物,去除废水中的Ca2+,再利用磷酸铵镁(MAP)沉淀法去除废水中的氨氮.实验采用响应面实验设计方法中的中心复合设计法,利用响应面分析法对磷酸铵镁沉淀法反应参数进行优化,得到最优反应条件及最优反应条件下沉淀产物.利用扫描电子显微镜(SEM)及X射线衍射(XRD)对最优反应条件下两种沉淀物进行分析.结果表明,当n(Ca2+)∶n(CO23-)=1∶1.05,搅拌速率为1 500 r.min-1,反应时间为30 min时,Ca2+去除率接近100%;对除钙后废水进行磷酸铵镁法脱氮处理的最优反应条件为:pH=9.03,n(Mg)∶n(N)=1.20,n(P)∶n(N)=1.1,反应时间为30 min,搅拌速率为1 000 r.min-1,氨氮去除率达到95.40%,剩余总磷浓度为5.65 mg.L-1;沉淀物分别为纯净的CaCO3及MgNH4PO4.6H2O.  相似文献

污水深度脱氮问题日益突出,在实现污水深度脱氮的过程中尽可能降低运行成本更是符合目前我国的发展目标,因此,开发经济绿色的污水脱氮技术对可持续发展具有重大意义.本试验提出自养反硝化脱氮耦合沼气同步脱硫工艺,具有成本低,资源利用率高等优势.以沼气中的硫化氢作为电子供体,实现了污水中同步脱氮及沼气脱硫净化的耦合,并探究了上升流速、硫氮比对该工艺运行效能的影响.实验结果显示,以硫化氢代替硫化物作为电子供体参与反硝化,对工艺脱氮效能无明显影响,在低硝酸盐负荷条件下运行时,污水脱氮效能不受气体上升流速及硫氮比的影响,均能达到100%.而本工艺的脱硫效能受上升流速影响较小,受硫氮比影响较大.在不同上升流速下,硫化氢去除率均为100%.在硫氮比为5∶8时,硫化氢100%转化为硫酸盐;硫氮比为5∶5时,硫化氢去除率为99.1%,单质硫产率约为30%;硫氮比为5∶2,回流比为1∶1时,硫化氢去除率最高可达91%,单质硫产率为77%.本试验可为后续自养反硝脱氮同步沼气脱硫工艺参数优化及应用的拓展提供理论依据和参考.  相似文献

利用从南四湖人工湿地中分离的一株具有良好脱氮作用的反硝化细菌XP1,分别接种于湿地植物芦竹、芦苇和香蒲的根际土壤,考察菌株XP1对3种植物的强化脱氮作用,并利用PCR-DGGE(变性梯度凝胶电泳)考察强化脱氮过程中菌株XP1在3种植物根际土壤微生物中的变化. 结果表明:模拟湿地在未强化脱氮时,3种植物湿地在相同条件下脱氮能力大小顺序为芦竹>芦苇>香蒲;3种植物的根际土壤所含微生物种类基本相同,所含微生物总量顺序为芦竹>芦苇>香蒲,其中土著XP1菌株在芦竹、芦苇和香蒲根际土壤微生物中的含量比值约为1.5∶1.3∶1. 加入菌株XP1强化脱氮后,香蒲、芦苇、芦竹3种植物湿地总氮去除率分别由14%,56%和56%提高至98%以上,强化脱氮作用明显;含氮废水ρ(TN)降至《地表水环境质量标准》(GB 3838—2002)Ⅲ类水质要求〔ρ(TN)≤1 mg/L〕时,3种植物根际土壤中的微生物种群数及微生物总量都有所下降,其中菌株XP1在3种植物根际土壤中的含量分别下降了40%,53%和67%,但根际细菌菌群结构未发生较大变化.   相似文献

研究了聚氨酯生物膜反应器在短程硝化反硝化工艺中的应用,考察碳氮比(15∶1、10∶1、5∶1和1.8∶1)对聚氨酯脱氮系统脱氮性能和微生物群落结构的影响,以及微生物群落结构与其处理效果的对应关系.结果表明,经过100 d的运行,当进水碳氮比从15依次下降到10、5和1.8,亚硝酸氮累积率由56.1%逐次上升到62.3%、72.3%和83.2%.在进水碳氮比为10时,系统取得最佳处理效果,氨氮和总氮去除率分别为99.1%和91.0%.进水碳氮比在15、10、5和1.8时,硝化反应和反硝化反应均同时发生在聚氨酯生物膜系统内,随着进水碳氮比的降低,同时硝化反硝化效率逐渐降低.生物膜的功能微生物分析表明,在碳氮比15时,生物膜的微生物多样性要显著高于其他工况.生物膜上的优势亚硝酸菌和硝酸菌分别以亚硝化单胞菌(Nitrosospira sp.)和硝化螺旋菌(Nitrospira sp.)为主,而反硝化细菌则以假单胞菌(Pseudomonas sp.)占据优势.  相似文献

在长期运行的处理污泥压滤液的气提亚硝化反应器中发现了单级自养脱氮反应,并形成了具有自养脱氮性能的原生颗粒污泥.以原生颗粒污泥启动并运行了单级自养脱氮反应器,对污泥压滤液进行脱氮处理取得了良好效果.进水总氮浓度为350 mg.L-1左右时,总氮平均去除率为74.81%,最高达86.92%,总氮平均去除负荷(以N计,下同)为0.68 kg.(m3.d)-1,最高达0.90 kg.(m3.d)-1.投加粉末活性炭后单级自养脱氮反应得到强化,运行稳定性得到提高.进水基质浓度、氮负荷及曝气量对自养脱氮反应影响较大.污泥压滤液中的有机物、pH值和碱度对单级自养脱氮反应影响较小.曝气量/ΔTN、曝气量/ΔNH4+-N及ΔALK/ΔTN比值可作为单级自养脱氮反应重要的运行指标.  相似文献

以高浓度氮磷模拟废水为处理对象,通过静态烧杯试验研究了磷酸铵镁法(MAP法)与沸石吸附组合工艺的脱氮除磷效果.分析了单独使用浙江缙云天然斜发沸石吸附氮磷受吸附时间、沸石粒径和沸石用量等因素的影响,所得最佳吸附条件为:反应时间80 min、粒径180~200目(0.076~0.088 mm).用量12 g/100 mL模拟废水.以MAP法除磷脱氮后的出水作为沸石吸附过程的进水,最终出水的氮、磷去除率可达86.69%和99.9%,且在MAP反应过程中采取较高的pH值和Mg2+浓度有利于后期沸石对氮、磷的吸附去除.研究表明,采取MAP法与沸石吸附组合工艺去除氮磷是可行的,可同时达到较高的除磷脱氮效果.  相似文献

Toxic effects of two agrochemicals on nifH gene in agricultural black soil were investigated using denaturing gradient gel electrophoresis (DGGE) and sequencing approaches in a microcosm experiment. Changes of soil nifH gene diversity and composition were examined following the application of acetochlor, methamidophos and their combination. Acetochlor reduced the nifH gene diversity (both in gene richness and diversity index values) and caused changes in the nifH gene composition. The effects of acetochlor on nifH gene were strengthened as the concentration of acetochlor increased. Cluster analysis of DGGE banding patterns showed that nifH gene composition which had been affected by low concentration of acetochlor (50 mg/kg) recovered firstly. Methamidophos reduced nifH gene richness that except at 4 weeks. The medium concentration of methamidophos (150 mg/kg) caused the most apparent changes in nifH gene diversity at the first week while the high concentration of methamidophos (250 mg/kg) produced prominent effects on nifH gene diversity in the following weeks. Cluster analysis showed that minimal changes of nifH gene composition were found at 1 week and maximal changes at 4 weeks. Toxic effects of acetochlor and methamidophos combination on nifH gene were also apparent. Different nifH genes (bands) responded differently to the impact of agrochemicals: four individual bands were eliminated by the application of the agrochemicals, five bands became predominant by the stimulation of the agrochemicals, and four bands showed strong resistance to the influence of the agrochemicals. Fifteen prominent bands were partially sequenced, yielding 15 different nifH sequences, which were used for phylogenetic reconstructions. All sequences were affiliated with the alpha- and beta-proteobacteria, showing higher similarity to eight different diazotrophic genera.  相似文献

The effects of arbuscular mycorrhizal (AM) fungus (Glomus mosseae) and phosphorus (P) addition (100 mg/kg soil) on arsenic (As) uptake by maize plants (Zea mays L.) from an As-contaminated soil were examined in a glasshouse experiment.Non-mycorrhizal and zero-P addition controls were included.Plant biomass and concentrations and uptake of As,P,and other nutrients,AM colonization,root lengths,and hyphal length densities were determined.The results indicated that addition of P significantly inhibited root colonization and development of extraradical mycelium.Root length and dry weight both increased markedly with mycorrhizal colonization under the zero-P treatments,but shoot and root biomass of AM plants was depressed by P application.AM fungal inoculation decreased shoot As concentrations when no P was added,and shoot and root As concentrations of AM plants increased 2.6 and 1.4 times with P addition,respectively.Shoot and root uptake of P,Mn,Cu,and Zn increased,but shoot Fe uptake decreased by 44.6%,with inoculation, when P was added.P addition reduced shoot P,Fe,Mn,Cu,and Zn uptake of AM plants,but increased root Fe and Mn uptake of the nonmycorrhizal ones.AM colonization therefore appeared to enhance plant tolerance to As in low P soil,and have some potential for the phytostabilization of As-contaminated soil,however,P application may introduce additional environmental risk by increasing soil As mobility.  相似文献

Several main metabolites of benzo[a]pyrene (BaP) formed by Penicillium chrysogenum, Benzo[a]pyrene-1,6-quinone (BP 1,6- quinone), trans-7,8-dihydroxy-7,8-dihydrobenzo[a]pyrene (BP 7,8-diol), 3-hydroxybenzo[a]pyrene (3-OHBP), were identified by high-performance liquid chromatography (HPLC). The three metabolites were liable to be accumulated and were hardly further metabolized because of their toxicity to microorganisms. However, their further degradation was essential for the complete degradation of BaP. To enhance their degradation, two methods, degradation by coupling Penicillium chrysogenum with KMnO4 and degradation only by Penicillium chrysogenum, were compared; Meanwhile, the parameters of degradation in the superior method were optimized. The results showed that (1) the method of coupling Penicillium chrysogenum with KMnO4 was better and was the first method to be used in the degradation of BaP and its metabolites; (2) the metabolite, BP 1,6-quinone was the most liable to be accumulated in pure cultures; (3) the effect of degradation was the best when the concentration of KMnO4 in the cultures was 0.01% (w/v), concentration of the three compounds was 5 mg/L and pH was 6.2. Based on the experimental results, a novel concept with regard to the bioremediation of BaP-contaminated environment was discussed, considering the influence on environmental toxicity of the accumulated metabolites.  相似文献

Sorption of chlorotoluron in ammonium sulfate, urea and atrazine multi-solutes system was investigated by batch experiments. The results showed application of nitrogen fertilizers to the soil could affect the behavior of chlorotoluron. At the same concentration of N, sorption of chlorotoluron decreased as the concentration of atrazine increased on the day 0 and 6 in soil, respectively. The sorption of chlorotoluron increased from 0 to 6 d when soils were preincubated with deionized water, ammonium sulfate and urea solution for 6 d. That indicated incubation time was one of the most important factors for the sorption of chlorotoluron in nitrogen fertilizers treatments. The individual sorption isotherms of chlorotoluron in rubbery polymer and silica were strictly linear in single solute system, but there were competition sorption between pesticides or between pesticides and nitrogen fertilizers. That indicated the sorption taken place by concurrent solid-phase dissolution mechanism and sorption on the interface of water-organic matter or water-mineral matter.  相似文献

In this study an effort has been made to use plant polyphenol oxidases; potato (Solanum tuberosum) and brinjal (Solanum melongena), for the treatment of various important dyes used in textile and other industries. The ammonium sulphate fractionated enzyme preparations were used to treat a number of dyes under various experimental conditions. Majority of the treated dyes were maximally decolorized at pH 3.0. Some of the dyes were quickly decolorized whereas others were marginally decolorized. The initial first hour was sufficient for the maximum decolorization of dyes. The rate of decolorization was quite slow on long treatment of dyes. Enhancement in the dye decolorization was noticed on increasing the concentration of enzymes. The complex mixtures of dyes were treated with both preparations of polyphenol oxidases in the buffers of varying pH values. Potato polyphenol oxidase was significantly more effective in decolorizing the dyes to higher extent as compared to the enzyme obtained from brinjal polyphenol oxidase. Decolorization of dyes and their mixtures, followed by the formation of an insoluble precipitate, which could be easily removed simply by centrifugation.  相似文献

A study was conducted to compare the diversity of 2-, 3-, and 4-chlorobenzoate degraders in two pristine soils and one contaminated sewage sludge. These samples contained strikingly different populations of mono-chlorobenzoate degraders. Although fewer cultures were isolated in the uncontaminated soils than contaminated one, the ability of microbial populations to mineralize chlorobenzoate was widespread. The 3- and 4-chlorobenzoate degraders were more diverse than the 2-chlorobenzoate degraders. One of the strains isolated from the sewage sludge was obtained. Based on its phenotype, chemotaxonomic properties and 16S rRNA gene, the organism S-7 was classified as Rhodococcus erythropolis. The strain can grow at temperature from 4 to 37℃. It can utilize several (halo)aromatic compounds. Moreover, strain S-7 can grow and use 3-chlorobenzoate as sole carbon source in a temperatures range of 10-30℃ with stoichiometric release of chloride ions. The psychrotolerant ability was significant for bioremediation in low temperature regions. Catechol and chlorocatechol 1,2-dioxygenase activities were present in cell free extracts of the strain, but no (chloro)catechol 2,3- dioxygenase activities was detected. Spectral conversion assays with extracts from R. erythropolis S-7 showed accumulation of a compound with a similar UV spectrum as chloro-cis,cis-muconate from 3-chlorobenzoate. On the basis of these results, we proposed that S-7 degraded 3-chlorobenzoate through the modified ortho-cleave pathway.  相似文献

Single and joint effects of pesticides and mercury on soil urease   总被引:3,自引:3,他引:3  
The influence of two pesticides including chlorimuron-ethyl and furadan and mercury (Hg) on urease activity in 4 soils (meadow burozem and phaeozem) was investigated. The soils were exposed to various concentrations of the two pesticides and Hg individually and simultaneously. Results showed that there was a close relationship between urease activity and organic matter content in soil. Chlorimuron-ethyl and furadan could both activate urease in the 4 soils. The maximum increment of urease activity by chlorimuronethyl was up to 14%-18%. There was almost an equal increase (up to 13%-21%) in the urease activity by furadan. On the contrary, Hg markedly inhibited soil urease activity. A logarithmic equation was used to describe the relationship (P〈0.05) between the concentration of Hg and the activity of soil urease in the 4 tested soils. Semi-effect dose (ED50) values by the stress of Hg based on the inhibition of soil urease in the 4 soils were 88, 5.5, 24 and 20 mg/kg, respectively, according to the calculation of the corresponding equations. The interactive effect of chlorimuron-ethyl or furadan with metal Hg on soil urease was mainly synergic at the highest tested concentrations.  相似文献

A field study was conducted in the Taihu Lake region, China in 2004 to reveal the organochlorine pesticide concentrations in soils after the ban of these substances in the year 1983. Thirteen organochlorine pesticides (OCPs) were analyzed in soils from paddy field, tree land and fallow land. Total organochlorine pesticide residues were higher in agricultural soils than in uncultivated fallow land soils. Among all the pesticides, ΣDDX (DDD, DDE and DDT) had the highest concentration for all the soil samples, ranging from 3.10 ng/g to 166.55 ng/g with a mean value of 57.04 ng/g and followed by ΣHCH, ranging from 0.73 ng/g to 60.97 ng/g with a mean value of 24.06 ng/g. Dieldrin, endrin, HCB and α-endosulfan were also found in soils with less than 15 ng/g. Ratios of p,p'-(DDD DDE)/DDT in soils under three land usages were: paddy field > tree land > fallow land, indicating that land usage inlfuenced the degradation of DDT in soils. Ratios of p,p'-(DDD DDE)/DDT >1, showing aged residues of DDTs in soils of the Taihu Lake region. The results were discussed with data from a former study that showed very low actual concentrations of HCH and DDT in soils in the Taihu Lake region, but according to the chemical half-lives and their concentrations in soils in 1980s, the concentration of DDT in soils seemed to be underestimated. In any case our data show that the ban on the use of HCH and DDT resulted in a tremendous reduction of these pesticide residues in soils, but there are still high amounts of pesticide residues in soils, which need more remediation processes.  相似文献

Common silver barb,Puntius gonionotus,exposed to the nominal concentration of 0.06 mg/L Cd for 60 d,were assessed for histopathological alterations(gills,liver and kidney),metal accumulation,and metallothionein(MT)mRNA expression.Fish exhibited pathological symptoms such as hypertrophy and hyperplasia of primary and secondary gill lamellae,vacuolization in hepatocytes,and prominent tubular and glomerular damage in the kidney.In addition,kidney accumulated the highest content of cadmium,more than gills and liver.Expression of MT mRNA was increased in both liver and kidney of treated fish.Hepatic MT levels remained high after fish were removed to Cd-free water.In contrast,MT expression in kidney was peaked after 28 d of treatment and drastically dropped when fish were removed to Cd-free water.The high concentrations of Cd in hepatic tissues indicated an accumulation site or permanent damage on this tissue.  相似文献
