除草剂西玛津对非洲爪蟾生存和性腺发育的影响   总被引:1,自引:1,他引:1  
为了研究三嗪类除草剂西玛津对两栖动物非洲爪蟾(Xenopus laevis)生存和性腺发育的毒性作用,并且与另一种三嗪类除草剂阿特拉津的毒性进行比较,将非洲爪蟾从46/47阶段开始暴露西玛津和阿特拉津到变态1个月后停止,再饲养2个月后将其解剖,取性腺做形态学和组织学观察.暴露期间,每天记录蝌蚪的生长发育情况和存活率.结果显示,在暴露的第1周内(蝌蚪处于46~50阶段),西玛津可导致蝌蚪死亡率明显升高,随后的时间内西玛津对非洲爪蟾的生存不再有明显影响,但却使发育阶段明显的不整齐.阿特拉津对非洲爪蟾的生存和发育则没有明显影响.西玛津和阿特拉津对非洲爪蟾性腺的总体形态和性别比没有明显影响,然而两种除草剂均在一定程度上导致了睾丸组织学的改变.西玛津可能与阿特拉津一样能够通过雌性化/去雄性化作用影响非洲爪蟾睾丸的发育.  相似文献

在美国材料与测试协会(ASTM)的非洲爪蟾胚胎致畸试验(FETAX)的基础上,以已知具有发育神经毒性的氯化甲基汞为模式化合物,探索一种以体征、运动神经元形态和运动行为参数为终点指标的研究发育神经毒性的方法.非洲爪蟾胚胎暴露氯化甲基汞3 d时,观察到暴露组胚胎的运动能力随暴露浓度(100—400 nmol·L-1)的增加而减弱.暴露4 d发现300 nmol·L-1和400 nmol·L-1暴露组胚胎体长和运动神经元明显短于对照组.暴露持续7 d,通过行为分析软件对蝌蚪运动行为定量,发现暴露处理的蝌蚪的游泳速率明显小于对照组.以上结果显示,非洲爪蟾胚胎可用来研究化学品的发育神经毒性,胚胎的体征、运动神经元形态和运动行为可以作为相对敏感的评价指标.  相似文献

研究了不同条件下阿特拉津溶液中 (ρ1~ 4/mgL-1:5 ,10 ,15 ,2 0 )弹琴蛙 (Ranaadenopleura)蝌蚪的行为 ,运用方差分析 (ANOVA)检验了蝌蚪在不同浓度的阿特拉津、时间段、pH值、溶氧量等条件下的行为差异性 .根据实验结果 ,确立了正常及异常行为模式 .结果表明 :在水温 (15 .0± 0 .5 )℃的条件下 ,弹琴蛙蝌蚪异常行为的出现主要与阿特拉津的浓度有关 ,同时与实验时间的长短也有一定的关系 ,而与 pH值、溶氧量关系不大 .实验组与对照组相比 ,蝌蚪出现异常行为的频次差异极显著 (P <0 .0 1) ;其中在实验组中 ,蝌蚪在 5mg/L的阿特拉津溶液中与 10mg/L、15mg/L、2 0mg/L中出现异常行为的频次差异极显著 (P <0 .0 1) ,而在 10mg/L、15mg/L、2 0mg/L阿特拉津溶液中出现异常行为的频次差异不显著 (P >0 .0 5 ) ,相关性分析表明 ,蝌蚪异常行为的出现与阿特拉津的浓度没有剂量反应关系 (r2 =0 .4 94 ) ;在对照组中 ,0 .5mL/L丙酮对照和空白对照中蝌蚪出现异常行为的频次差异不显著 (P >0 .0 5 ) .图 3表 2参 17  相似文献

体外培养的神经细胞是神经毒性机制研究和神经毒物筛查的重要材料.目前鼠体外神经细胞的应用最为广泛.鉴于近年来发育生物学的模型动物非洲爪蟾越来越多地应用于毒理学研究,论文建立了一种体外培养非洲爪蟾脑神经细胞的方法.该方法取52~53阶段的非洲爪蟾蝌蚪的脑组织,在L-15培养液中直接吹打获得分散细胞,接种于包被多聚赖氨酸的培养板中,22℃培养.培养细胞状态良好,72小时后神经元细胞初步建立神经网络.用β2tubulin7B9可对脑神经元细胞进行荧光染色,鉴别神经突的生长和神经网络的形成.与鼠脑神经元细胞体外培养方法比较,该方法具有无需胰蛋白酶分离、无需胶质细胞共培养、可同时操作多只脑、能获得大量细胞等简单快捷的特点.因此,体外培养的非洲爪蟾蝌蚪脑神经细胞可作为目前体外神经毒理研究中鼠脑神经元的补充材料,用于神经毒性机制的研究和神经毒物的筛查.  相似文献

PFOS及其4种替代品对两栖动物非洲爪蟾和黑斑蛙蝌蚪的急性毒性结果为:用调聚法合成的织物三防整理剂对非洲爪蟾蝌蚪和黑斑蛙蝌蚪的96h-LC50分别为8和21mg·L-1,而PFOS对两种蝌蚪的96h-LC50分别为92和81mg·L-1。此实验结果说明织物三防整理剂的急性毒性高于PFOS。用电解氟化法合成的C4、C6织物三防整理剂和50%的全氟丁基有机铵盐阳离子表面活性剂浓度在100mg·L-1时,对两种蝌蚪都没有毒性。这说明,从急性毒性的角度,C4、C6织物三防整理剂和表面活性剂可作为PFOS的替代品使用,但是织物三防整理剂的急性毒性比PFOS大,作为替代品使用应慎重考虑。另外,PFOS和织物三防整理剂对黑斑蛙蝌蚪的急性毒性与对非洲爪蟾蝌蚪的急性毒性存在差异。出于保护我国本土两栖动物的目的,使用黑斑蛙开展毒性评价比使用非洲爪蟾更有现实意义。  相似文献

两栖动物蝌蚪急性毒性试验是评价化学品急性毒性的一种方法。以毒死蜱、乙草胺、重铬酸钾和全氟辛烷磺酸盐(PFOS)为测试物,比较了我国本土黑斑蛙(Rana nigromaculata)与国际通用种非洲爪蟾(Xenopus laevis)在蝌蚪急性毒性试验中的敏感性。结果发现:2类蝌蚪分别进行的11次试验中,空白对照组黑斑蛙蝌蚪死亡率(0.9%)远低于非洲爪蟾蝌蚪的死亡率(5.8%);重铬酸钾和PFOS对黑斑蛙蝌蚪的96 h-LC50分别为34.0 mg·L-1和81.0 mg·L-1,而对非洲爪蟾蝌蚪的96 h-LC50分别为51.6 mg·L-1和92.1 mg·L-1,显示黑斑蛙蝌蚪对这2种化学品的敏感性略高于非洲爪蟾蝌蚪;毒死蜱和乙草胺对黑斑蛙蝌蚪的96 h-LC50分别为0.41 mg·L-1和4.1 mg·L-1,而对非洲爪蟾蝌蚪的96 h-LC50分别为0.12 mg·L-1和3.1 mg·L-1,显示黑斑蛙蝌蚪对这2种化学品的敏感性略低于非洲爪蟾。鉴于2类蝌蚪对化学品的敏感性存在差异,且黑斑蛙蝌蚪的自然死亡率低,材料更易获得,笔者认为黑斑蛙蝌蚪比非洲爪蟾更适合作为蝌蚪急性毒性试验的材料,用于我国化学品环境管理中的毒性评价。  相似文献

采用扩散系数C等5种分布型指标测定阿特拉津溶液中(ρ1-3/mg L-1:10,20,30)弹琴蛙(Rana adenopleura)蝌蚪(均处于25期和26期,体长范围26-60 mm)在水族箱(l×b×h=40 cm×20 cm×25 cm)中的空间分布模式,结果表明蝌蚪在水族箱中为聚集分布.经多次适合性检验,证明弹琴蛙蝌蚪水族箱中的水平分布呈现负二项分布状态.随着阿特拉津浓度的升高,蝌蚪在水族箱内的聚集度(由负二项分布的k值来表明)逐渐减小(kρ1相似文献

本文旨在探讨低剂量PCBs和PBDEs单一暴露和复合暴露的甲状腺干扰作用.非洲爪蟾46期蝌蚪单独或共暴露于100 ng·L-1Aroclor 1254和BDE-209至62期.暴露结束后检测变态时间、甲状腺组织学结构、甲状腺相关基因表达水平等指标.结果发现,Aroclor 1254和BDE-209单独暴露使蝌蚪变态发育呈现一定的延迟趋势,而复合暴露却显著抑制蝌蚪变态发育;所有的暴露处理均导致蝌蚪甲状腺组织代偿性改变,表现为胶质面积减少,甲状腺滤泡上皮细胞高度显著增加;Aroclor 1254单独暴露显著抑制甲状腺激素受体(TRA)、Ⅱ和Ⅲ型脱碘酶(DI-2,DI-3)的表达,BDE-209单独暴露仅抑制DI-2的表达,但BDE-209协同促进Aroclor1254对肝脏内TRA表达的抑制作用.综上,低剂量Aroclor 1254和BDE-209单独暴露和复合暴露对非洲爪蟾变态发育具有一定的甲状腺抑制作用,复合暴露的抑制作用明显高于单一暴露的作用.鉴于甲状腺系统在脊椎动物生长发育过程中的重要作用,低剂量PCBs和PBDEs复合暴露的甲状腺干扰效应应该受到格外关注.  相似文献

为了研究多氯联苯(Aroclor 1254)对非洲爪蟾变态发育的影响,将非洲爪蟾受精卵暴露在一定浓度(0、400ng·L-1、2μg·L-1、10μg·L-1、20μg·L-1)的Aroclor 1254中,直至完全变态.结果表明,20μg·L-1Aroclor1254暴露可使变态所需时间显著延长(p<0.05),低于20μg·L-1的各组变态所需时间与对照组无显著差异.2μg·L-1以上的Aroclor1254暴露可使蝌蚪甲状腺发生明显的组织学改变,包括甲状腺组织滤泡扩张、胶体面积相对缩小,滤泡细胞增殖、高度增加,并且这种变化随暴露剂量的增加而愈发明显.提示多氯联苯可干扰非洲爪蟾的变态发育,引起甲状腺组织学改变.采用定量组织病理学技术检测甲状腺激素干扰物较为灵敏,方法简单易行,是一种切实可行的筛选和确证甲状腺干扰物的体内方法.  相似文献

A bioblitz inexpensively and quickly generates biodiversity data, but bioblitzes are often conducted with haphazard, unreplicated sampling. Results tend to be taxonomically, geographically, or temporally biased, lack metadata, and consist of lists of observed taxa that do not enable further analyses or correction for imperfect detection. A rapid, recurring, structured survey (RRSS) uses a structured sampling design and temporal and spatial replication to survey randomly selected sites on a conservation property. We participated in a loosely structured bioblitz and a subsequent RRSS at Big Canoe Creek Nature Preserve in Springville (St. Clair County), Alabama (USA) to compare observed richness derived from the 2 survey approaches. The RRSS data structure enabled us to fit models that accounted for imperfect detection to estimate abundances, occupancy probabilities, and habitat associations. The loosely structured bioblitz data could not be used in such models. We present a new integrated multispecies abundance model that we applied to avian RRSS data. Our model extension enables estimation for the community, employs data augmentation to estimate the number of undetected species, and incorporates covariates. The RRSS generated a more comprehensive and less biased list of observed taxonomic richness than the loosely structured bioblitz (e.g., 73 vs. 45 bird species and 104 vs. 63 insect families from the RRSS vs. loosely structured bioblitz, respectively). Models fit to the RRSS data identified seasonal patterns in avian community composition and allowed for estimation of habitat–occupancy relationships for insect taxa. The RRSS protocol has potential for broad transferability as a standardized, quick, and inexpensive way to inventory biodiversity and estimate ecological parameters while providing an outreach opportunity.  相似文献

Land-use change via human development is a major driver of biodiversity loss. To reduce these impacts, billions of dollars are spent on biodiversity offsets. However, studies evaluating offset project effectiveness that examine components such as the overall compliance and function of projects remain rare. We reviewed 577 offsetting projects in freshwater ecosystems that included the metrics project size, type of aquatic system (e.g., wetland and creek), offsetting measure (e.g., enhancement, restoration, and creation), and an assessment of the projects’ compliance and functional success. Project information was obtained from scientific and government databases and gray literature. Despite considerable investment in offsetting projects, crucial problems persisted. Although compliance and function were related to each other, a high level of compliance did not guarantee a high degree of function. However, large projects relative to area had better function than small projects. Function improved when projects targeted productivity or specific ecosystem features and when multiple complementary management targets were in place. Restorative measures were more likely to achieve targets than creating entirely new ecosystems. Altogether the relationships we found highlight specific ecological processes that may help improve offsetting outcomes.  相似文献

An argument is presented in which areas of natural arsenic contamination of modern groundwaters throughout Asia have a common origin. Arsenic originally accumulated in oceanic ferro-manganoan sediments of the eastern Palaeo-Tethys. This was further concentrated through oceanic crustal extinction in what later became the south-east Chinese accreted mineralised terrain. Proto-Himalayan uplift of this area created the palaeo-drainage systems of the Ganges – Brahmaputra, Irrawaddy, Mekong, and Red Rivers, with consequent headwater erosion of arsenic-rich sediments. Their downstream deposition as immature and easily redistributed Neogene sandstones, silts, and iron-rich clays has created secondary and tertiary reservoirs of adsorbed and authigenic arsenic, from which the current arsenic-rich groundwaters have evolved. Considering river basins within the above palaeo-hydrogeological framework provides a basis for assessing the risk of arsenic in groundwater basins of south and south-eastern Asia.  相似文献

How should managers choose among conservation options when resources are scarce and there is uncertainty regarding the effectiveness of actions? Well‐developed tools exist for prioritizing areas for one‐time and binary actions (e.g., protect vs. not protect), but methods for prioritizing incremental or ongoing actions (such as habitat creation and maintenance) remain uncommon. We devised an approach that combines metapopulation viability and cost‐effectiveness analyses to select among alternative conservation actions while accounting for uncertainty. In our study, cost‐effectiveness is the ratio between the benefit of an action and its economic cost, where benefit is the change in metapopulation viability. We applied the approach to the case of the endangered growling grass frog (Litoria raniformis), which is threatened by urban development. We extended a Bayesian model to predict metapopulation viability under 9 urbanization and management scenarios and incorporated the full probability distribution of possible outcomes for each scenario into the cost‐effectiveness analysis. This allowed us to discern between cost‐effective alternatives that were robust to uncertainty and those with a relatively high risk of failure. We found a relatively high risk of extinction following urbanization if the only action was reservation of core habitat; habitat creation actions performed better than enhancement actions; and cost‐effectiveness ranking changed depending on the consideration of uncertainty. Our results suggest that creation and maintenance of wetlands dedicated to L. raniformis is the only cost‐effective action likely to result in a sufficiently low risk of extinction. To our knowledge we are the first study to use Bayesian metapopulation viability analysis to explicitly incorporate parametric and demographic uncertainty into a cost‐effective evaluation of conservation actions. The approach offers guidance to decision makers aiming to achieve cost‐effective conservation under uncertainty.  相似文献

Large, intact areas of tropical peatland are highly threatened at a global scale by the expansion of commercial agriculture and other forms of economic development. Conserving peatlands on a landscape scale, with their hydrology intact, is of international conservation importance to preserve their distinctive biodiversity and ecosystem services and maintain their resilience to future environmental change. We explored threats to and opportunities for conserving remaining intact tropical peatlands; thus, we excluded peatlands of Indonesia and Malaysia, where extensive deforestation, drainage, and conversion to plantations means conservation in this region can protect only small fragments of the original ecosystem. We focused on a case study, the Pastaza‐Marañón Foreland Basin (PMFB) in Peru, which is among the largest known intact tropical peatland landscapes in the world and is representative of peatland vulnerability. Maintenance of the hydrological conditions critical for carbon storage and ecosystem function of peatlands is, in the PMFB, primarily threatened by expansion of commercial agriculture linked to new transport infrastructure that is facilitating access to remote areas. There remain opportunities in the PMFB and elsewhere to develop alternative, more sustainable land‐use practices. Although some of the peatlands in the PMFB fall within existing legally protected areas, this protection does not include the most carbon‐dense (domed pole forest) areas. New carbon‐based conservation instruments (e.g., REDD+, Green Climate Fund), developing markets for sustainable peatland products, transferring land title to local communities, and expanding protected areas offer pathways to increased protection for intact tropical peatlands in Amazonia and elsewhere, such as those in New Guinea and Central Africa which remain, for the moment, broadly beyond the frontier of commercial development.  相似文献

Summary Undamaged plants emit low levels of green leaf volatiles (GLVs), while caterpillar-damaged and artificially damaged plants emit relatively higher levels of certain GLVs. Female braconid parasitoids,Microplitis croceipes, oriented to both damaged plants and to individual GLVs in no-choice tests in a wind tunnel, but seldom oriented to undamaged plants. Female ichneumonid parasitoids,Netelia heroica, also oriented to individual GLVs in a wind tunnel. Males of both wasp species failed to orient to the GLVs. These data show that leaf-feeding caterpillars can cause the release of GLVs, and that parasitic wasps can respond to these odors by flying upwind (chemoanemotactic response), which brings the wasps to their caterpillar hosts. This supports the hypothesis that plants communicate with members of the third trophic level,i.e., plants under herbivore attack emit chemical signals that guide natural enemies of herbivores to sites of plant damage. In this interaction, the GLVs serve as tritrophic plant-to-parasitoid synomones. That parasitoids from two different wasp families oriented to GLVs suggests that the response may be widespread among the Hymenoptera.Mention of a commercial or proprietary product does not constitute an endorsement by the U.S. Department of Agriculture  相似文献

Biogeographic theory predicts that rare species occur more often in larger, less‐isolated habitat patches and suggests that patch size and connectivity are positive predictors of patch quality for conservation. However, in areas substantially modified by humans, rare species may be relegated to the most isolated patches. We used data from plant surveys of 81 meadow patches in the Georgia Basin of Canada and the United States to show that presence of threatened and endangered plants was positively predicted for patches that were isolated on small islands surrounded by ocean and for patches that were isolated by surrounding forest. Neither patch size nor connectivity were positive predictors of rare species occurrence. Thus, in our study area, human influence, presumably due to disturbance or introduction of competitive non‐native species, appears to have overwhelmed classical predictors of rare species distribution, such that greater patch isolation appeared to favor presence of rare species. We suggest conservation planners consider the potential advantages of protecting geographically isolated patches in human‐modified landscapes because such patches may represent the only habitats in which rare species are likely to persist. The macro-algae communities observed in the south lake of Tunis are characterized by the predominance of nitrophilous algae which are in the order of biomass importance:Ulva, Cladophora andEnteromorpha. We have noted seasonal changes of alga distribution. The wind appears to be one of the most important factors influencing this distribution. The total biomass reaches a maximum in the spring. Rapid decomposition of the biomass leads to a severe ecological imbalance, resulting in crises of anoxia and fish death. A restoration project has already started. It aims at removal of contaminated muds and the introduction of a new circulation system. The main objectives of this work were to collect information on the distribution and biomass of the phytobenthic communities as a first step in the constitution of a database for further comparison.  相似文献

International Union for Conservation of Nature (IUCN) Red List assessments rely on published data and expert inputs, and biases can be introduced where underlying definitions and concepts are ambiguous. Consideration of climate change threat is no exception, and recently numerous approaches to assessing the threat of climate change to species have been developed. We explored IUCN Red List assessments of amphibians and birds to determine whether species listed as threatened by climate change display distinct patterns in terms of habitat occupied and additional nonclimatic threats faced. We compared IUCN Red List data with a published data set of species’ biological and ecological traits believed to infer high vulnerability to climate change and determined whether distributions of climate change‐threatened species on the IUCN Red List concur with those of climate change‐threatened species identified with the trait‐based approach and whether species possessing these traits are more likely to have climate change listed as a threat on the IUCN Red List. Species in some ecosystems (e.g., grassland, shrubland) and subject to particular threats (e.g., invasive species) were more likely to have climate change as a listed threat. Geographical patterns of climate change‐threatened amphibians and birds on the IUCN Red List were incongruent with patterns of global species richness and patterns identified using trait‐based approaches. Certain traits were linked to increases or decreases in the likelihood of a species being threatened by climate change. Broad temperature tolerance of a species was consistently related to an increased likelihood of climate change threat, indicating counterintuitive relationships in IUCN assessments. To improve the robustness of species assessments of the vulnerability or extinction risk associated with climate change, we suggest IUCN adopt a more cohesive approach whereby specific traits highlighted by our results are considered in Red List assessments. To achieve this and to strengthen the climate change‐vulnerability assessments approach, it is necessary to identify and implement logical avenues for further research into traits that make species vulnerable to climate change (including population‐level threats).  相似文献
