This investigation was undertaken to survey the fungal and mycotoxin contamination of South African wheat ranging from that growing in the field to processed wheat products. Samples of wheat were taken from various growing areas in South Africa and screened for fungi and mycotoxins, using a range of methodologies, including chromatography, immunoaffinity/fluorimetry, and cytotoxicity testing. Similar samples were taken from supermarkets and retail outlets in South Africa and analyzed in a similar manner. The result showed that a range of fungi and mycotoxins could be detected in wheat in all these sample types. The major fungal contaminants were Fusarium spp. and their attendant mycotoxins, in particular deoxynivalenol, which is in keeping with the observations made in the rest of the world. An interesting observation was that samples of wheat taken from the field with heavy Fusarium contamination were contaminated with fumonisin B1, which is not normally associated with this crop. Of more concern were the low but persistent levels of mycotoxins and fungi in wheat-based products sold directly to the public.  相似文献

The variation in grain cadmium (Cd) concentrations was evaluated among 600 barley genotypes grown in the same field condition to select low Cd accumulating genotypes. The results showed that there is considerable genotypic variation in grain Cd concentrations in barley grain samples, with the mean concentration of 0.16 mg kg(-1) DW and the variation of 0 (not detected) to 1.21 mg kg(-1) DW, and 47.2% of the grain samples exceeded the maximum permissible concentration (MPC) for Cd in cereal grains. In addition, differences between genotypes over the two years were fairly consistent, and Beitalys and Shang 98-128 showed the lowest grain Cd concentration, being 97.5% lower than that in the two highest Cd accumulators E-barley 6 and Zhenong 8 in the second harvest year. The great genotypic differences in Cd concentrations indicated that it is possible to lower Cd content of barley through cultivar selection and breeding for use at sites where Cd concentration in grain exceeds the MPC. Significant genotypic difference was also found in microelement concentrations. Correlation analysis showed that only Mn accumulation is synergetic with Cd accumulation, despite slightly positive relationship between Cd and Zn, Cu, or Fe in accumulation in barley grains.  相似文献

The use of gaseous ozone as a fungicide to preserve stored barley was studied. The effects of the following operating parameters on the fungicidal efficacy of ozone were examined: 1) the applied ozone dose, 2) ozonation time, 3) water activity of barley, and 4) temperature of barley. The effect of ozonation on germination of barley was also investigated. The experimental results showed that ozone was very effective in inactivation of fungi associated with the barley regardless of whether the fungi were in the forms of spores or mycelia. However, the mycelia were less resistant to ozone. With 5 minutes of ozonation, 96% of inactivation were achieved for spores as well as for mixtures of spores and small amount of mycelia by applying 0.16 and 0.10 mg of ozone/(g barley) x min, respectively. In addition, for sealed storage silos, inactivation of fungi continued when the ozone-containing gas was held inside the silos following a continuous ozone supply. The experimental results also revealed that increases in water activity and temperature of barley enhanced the fungicidal efficacy of ozone. Results of this study also indicated that the inactivation processes could be controlled by simply monitoring the exit ozone from the reactor instead of performing the time-consuming microbial examination. This finding would make the application of ozone in the preservation of cereal grains easier, simpler, and more practically applicable. The experimental results demonstrated that although ozonation above certain strength may reduce barley germination, inactivation of fungi was achieved with ozonation strengths far below the critical point.  相似文献

Cylindrocladium quinqueseptatum is a pathogen on a wide range of hosts. It affects at least 20 species of eucalypts and is an important causal agent of leaf blight of Eucalyptus camaldulensis in central and southern Vietnam. Results from previous studies and observations of broadscale infection patterns in mainland South East Asia were used to derive simple rules (i.e. mean minimum temperature of coldest month > or =16 degrees C and mean annual precipitation > or =1400 mm) to identify locations which are likely to have a high risk of C. quinqueseptatum leaf blight (CqLB). Climatic mapping programs, which include interpolated climatic data estimated for numerous locations, were used to map these high risk areas in Africa, Australia and Latin America as well as in South East Asia. The predicted high-risk areas included several regions where CqLB has already been reported and the maps generated suggested other areas which may be at risk under present climatic conditions given the presence of C. quinqueseptatum and susceptible hosts. Some simple climate change scenarios were also used to suggest areas in mainland South East Asia which may become vulnerable to CqLB over the next 50 years. It is concluded that climatic mapping programs can assist the broadscale evaluation of risk of CqLB infections, although it is recognised that more detailed models and survey information are also needed.  相似文献

The effects of joint action of SO(2) and HF on the yield and quality of wheat and barley were studied by exposing them to combinations of <13,130 or 267 microg m(-3) SO(2) and 0.03 or 0.38 microg m(-3) HF in open top chambers for 90 days. At the concentrations used, SO(2) had greater effects than HF. All responses were marked by compensatory changes. The treatments had no effect on wheat yield, although SO(2) reduced shoot weight. SO(2) increased the growth and yield of barley, and HF or SO(2) increased the grain protein concentration of barley and wheat. The effects of mixtures of SO(2) and HF were complex, but often antagonistic, as the addition of HF counteracted the effect of SO(2) alone.  相似文献

Abstract

The total alkaloid content and individual alkaloid composition were determined by colorimetry and high performance liquid chromatography, respectively, for Canadian triticale and barley ergot (Claviceps purpurea). The total alkaloid content was highly variable between individual sclerotia from the same or different sources and ranged from 0.042 to 0.752% for triticale and from 0.082 to 1.04% for barley; average values were 0.239% for Ottawa triticale, 0.289% for Prairie triticale, and 0.264% for barley. Ergocristine and its isomer ergocristinine were the major constituents in both grains. On average, Canadian ergot pooled from rye, wheat, triticale, and barley consists of ergocristine (31%), ergocristinine (13%), ergocristinine (13%), ergotamine (17%), ergotaminine (8%), ergocryptine (5%), ergocryptinine (3%), ergo‐metrine (5%), ergometrinine (2%), ergosine (4%), ergosinine (2%), ergocornine (4%), ergocorninine (2%), and unidentified alkaloids (3%) and an average total alkaloid content of 0.236%. With the exception of rye and barley ergot from the maritime regions, Canadian rye, wheat, triticale, and barley ergot is fairly uniform in individual alkaloid composition.  相似文献

The current trend of global warming is expected to stimulate the expansion of harmful cyanobacteria blooms. Previously, the occidental type of barley straw has been used to control blooms in Europe and America, but very little is known about the antialgal abilities of its oriental relative. We tested the use of Tibetan hulless barley straw--the progenitor of oriental barley--to inhibit the growth of cyanobacterium Microcystis aeruginosa. Flow cytometry allowed assessment at single-cell level, with morphologic parameters (cell volume, cell membrane integrity) and physiological parameters (in vivo Chlorophyll a fluorescence, metabolic activity) used as endpoints. The reduction of cell densities together with integrated cell membranes suggests that Tibetan barley may act as an algistatic agent. Doses from 2.0 to 8.0 g L?1 of Tibetan barley straw efficiently inhibited the alga, but these doses were much higher than those of occidental barley. Such a large dosage introduced additional nutrients, which stimulated the intracellular metabolic activity and induced two physiological subpopulations in the acute term. After mid and long-term exposure, the growth inhibition effect exceeded the stimulation effect, so that the cells' metabolic activity and Chlorophyll a fluorescence decayed, simultaneously with shrinkage in the algal cell volume.  相似文献

A granular formulation of terbufos (Counter 15G) was added in-furrow at time of planting of wheat and barley. Foliage collected at several times was analyzed for total terbufos residues as terbufoxon sulfone. Maximum residues from application of 1.5 and 3.0 kg/ha were 7.4 and 10.6 ppm, respectively, in wheat foliage samples collected 10 days postseeding. Wheat foliage collected at 53 days postseeding had residues averaging 0.32 and 0.58 ppm from the 1.5 and 3.0 kg/ha applications, respectively. In 1985 residues in barley were consistently less than in wheat in 1985 with 4.4 and 7.0 ppm detected in foliage collected 10 days post application from the 1.5 and 3.0 kg/ha applications, respectively and 0.21 and 0.34 ppm detected at 53 days. Grain samples had 0.01 ppm or less residue at harvest. Straw samples had up to 0.75 ppm total terbufos residues at harvest.  相似文献

Ship-board air samples collected between The Netherlands and South Africa in January-February 2001 were analysed for polycyclic aromatic hydrocarbons (PAHs) and polychlorinated naphthalenes (PCNs). The highest PAH concentrations occurred in the European samples, and in samples close to West Africa and South Africa. Consistently low PAH concentrations were measured in the southern hemisphere open ocean samples (190-680 pg/m3). The highest PCN concentrations occurred in the European samples, but high values were also detected off the West African coast, and in the sample taken closest to South Africa. Data are presented for diurnal cycles taken in the remote South Atlantic. The day:night ratios of phenanthrene, 1-methylphenanthrene and fluoranthene were typically approximately 1.5-2.5:1. The mechanism(s) causing this observation is/are not understood at present, but dynamic environmental process(es) is/are implicated.  相似文献

The toxicity and bioavailability of metals were assessed to verify the efficiency of a new chemical leaching process (METIX-AC) to minimize the risk of metals found in municipal sewage sludge. For this purpose, sludge samples were spiked with cadmium, copper and/or zinc before being treated using METIX-AC. The sludge decontamination resulted in a removal of spiked metals (79-89%), in a decrease of the more labile fractions, and in a corresponding increase of the residual fraction. The toxicity observed after exposure of two plant species, barley (Hordeum vulgare) and ryegrass (Lolium perenne), and a terrestrial invertebrate, Eisenia andrei, to sludge-soil mixtures, disappeared after treatment, although the adverse effects were minor before treatment. The sludge treatment also significantly decreased the bioaccumulation of cadmium, copper, and zinc in the exposed species. For cadmium, maximum tissue concentrations of 0.45+/-0.08 mg/kg in barley, 0.79+/-0.27 mg/kg in ryegrass, and 21.82+/-1.85 mg/kg in earthworm exposed to sludge before treatment decreased after treatment to values similar to those observed with negative controls.  相似文献

The documeneed adverse health effects of soil Cd and Pb have led to public concern over soil contamination with metals. A 4-year field experiment was conducted to study the transfer of Cd, Pb, and Zn from soil contaminated by smelter flue-dust to crop plants grown in a rotation. The soil was amended with Pb?Zn smelter flue-dust (2-66.8 kg per 10 m(2) plot) to simulate the long-term effect that the smelting of non-ferrous metal ore has on arable soils. The treated soil became strongly contaminated with metals (Cd 3.2-106 mg/kg, Pb 146-3452 mg/kg, Zn 465-11 375 mg/kg). Concentrations of Cd, Pb, and Zn in barley grain, barley straw meadow bluegrass, red clover, and potatoes were generally low. The highest metal concentrations were found in potato tubers (intact), meadow bluegrass, and barley straw. The observed reduction in crop yield was probably the result of possible nutrient imbalances rather than of metal (Zn, Cu) phytotoxicities. Zn and Cd uptake by the plants can be described by the saturation (plateau) model (y = ax(b), b < 1). The relationship between Pb in the soil and plants was linear with an extremely low slope (0.0001-0.0003). No excessive dietary intake of Cd is expected when Cd concentrations in barley grain and potato tubers grown on the contaminated soil are not higher than 0.6 and 1.0 mg/kg, respectively. Based on the risk analysis and taking into account the saturation model of the soil-plant metal relationship, it was concluded that, under the conditions of this experiment (neutral soil pH), soil with Cd concentrations of up to 30 mg/kg is still safe for production of these crop plants.  相似文献

The entry of Pb into the food chain is of concern as it can cause chronic health problems. The concentration of Pb was determined in cereal grain samples collected representatively from British Cereal Quality Surveys in 1982 and 1998 (n = 176, 250 and 233 for wheat collected in 1982 and 1998, and barley in 1998, respectively). In addition, paired soil and grain samples were collected from 377 sites harvested across Britain in 1998-2000. Wheat grain Pb ranged from below the analytical detection limit (0.02 mg kg(-1) dry weight, DW) to 1.63 mg kg(-1) DW, and barley grain Pb from <0.02 to 0.48 mg kg(-1) DW. The vast majority of samples (>99% for both wheat and barley, excluding Scottish barley samples collected in 2000) were well below the newly introduced EU limit for the maximum permissible concentration of Pb in cereals (0.2 mg kg(-1) fresh weight, equivalent to 0.235 mg kg(-1) DW). There was a significant reduction in wheat grain Pb in the 1998 survey compared with the 1982 survey. However, 40 barley samples collected from Scotland in 2000 in the paired soil and crop survey showed anomalously high concentrations of Pb, with 10 samples exceeding the EU limit. Washing experiments demonstrated that surface contamination, introduced during grain harvest and/or storage, was the main reason for the high concentrations in these samples. In the paired soil and crop surveys, there were no significant correlations between grain Pb concentrations with total soil Pb and other soil properties, indicating low bioavailability of Pb in the soils and limited uptake and transport of Pb to grain. The Pb in cereal grain is likely to originate mainly from atmospheric deposition and other routes of surface contamination during harvest and storage.  相似文献

Silicon-based fertilizers and soil amendments can have direct and indirect positive influences on cultivated plants. The solid forms of Si-based substances, the most widespread in use, are efficient only at high application rates due to their low level of solubility. Several types of Si-based substances such as fumed silica, slags from the iron and steel industry, modified slags, and a Si-rich product were tested using barley and pea as silicon accumulative and non-accumulative plants, respectively, at two application rates. The plants were grown under toxic concentrations of heavy metals in a greenhouse. Si-rich materials high in water-soluble Si had a positive effect at both the low and high application rates, and for both plant species. This type of substance can be regarded as Si fertilizer, demonstrating greater efficiency at a low application rate and lessened efficiency at a high application rate for protection of the cultivated plants against accumulation of the heavy metals.

The activity of Cd-induced POD isozyme isolated from the surface of intact barley roots growing under some abiotic stress conditions (toxic metals: Al, Co, Cu, Hg; drought, NaCl, extreme temperatures: heat, cold) and compounds activating (2,4-D) or inhibiting (SHAM) POD activity as well as H(2)O(2) and H(2)O(2) scavenger (DTT) was characterized. Strong Cd concentration-dependent accumulation of one cationic POD isozyme was observed on PAGE, which correlated with Cd- and other stress induced root growth inhibition. This isoPOD is basic with isoelectric point about 9 and its localization is restricted only to the apical part of the barley root tip.  相似文献

Toxicity of gamma irradiated mycotoxins aflatoxin B1 (AFB1) and ochratoxin A (OTA) was investigated in vitro. AFB1 and OTA stock solutions (50?mM, in methanol) were gamma irradiated (5 and 10 kGy) and non-irradiated and irradiated mycotoxins solutions were tested for cytotoxicity on Pk15, HepG2 and SH-SY5Y cell lines (MTT assay, 1–500?μM concentration range; 24?h exposure). Degradation of mycotoxin molecules was examined by liquid chromatography tandem mass spectrometry (HPLC-MS/MS). AFB1 and OTA radiolytic products were less toxic than the parent mycotoxins to all of the tested cell lines. Gamma irradiation even at 5 kGy had effect on AFB1 and OTA molecules however, this effect was dependent on chemical structure of mycotoxin. Since gamma irradiation at low dose reduced initial level of both mycotoxins, and gamma irradiated mycotoxins had lower toxicity in comparison to non-irradiated mycotoxins, it can be concluded that gamma irradiation could be used as decontamination method.  相似文献

The present study determines the influence of three ionic liquids (ILs) containing cations with diversified structure on the growth and development of spring barley seedlings and common radish leaves. Increasing amounts of 1-butyl-1-methylpyrrolidinium hexafluorophosphate [Pyrrol][PF6], 1-butyl-1-methylpiperidinium hexafluorophosphate [Piper][PF6], and 1-butyl-4-methylpyridinium hexafluorophosphate [Pyrid][PF6] were added to the soil on which both plants were cultivated. The results of this studies showed that the applied ILs were highly toxic for plants, demonstrated by the inhibition of length of plant shoots and roots, decrease of fresh mass, and increase of dry weight content. Common radish turned out to be the plant with higher resistance to the used ILs. The differences in the cation structure did not influence phytotoxity of ILs for spring barley. Furthermore, all ILs led to a decrease of photosynthetic pigments, which was directly followed by decreased primary production in plants. Oxidative stress in plants occurred due to the presence of ILs in the soil, which was demonstrated by the increase of malondialdehyde (MDA) content, changes in the H2O2 level, and antioxidant enzymes such as superoxide dismutase (SOD), catalase (CAT), and peroxidase (POD). The changes in the chlorophyll contents and the increase of POD activity turned out to be the most significant oxidative stress biomarkers in spring barley and common radish. Both spring barley and radish exposed to ILs accumulated a large amount of fluoride ion.

Four non-filtered and four charcoal-filtered open-top chambers were employed to determine the effects of ambient levels of gaseous air pollutants at Braunschweig, FRG, on growth and yield of potted plants of winter and spring barley. During the exposure period (November 1985-August 1986) monthly mean values of gaseous air pollutants (microg m(-3)) ranged between 34 and 127 for SO(2), 34 and 52 for NO(2) and 12 and 33 for O(3) in winter (November-March), and 16 to 26 for SO(2), 20 to 33 for NO(2) and 42 to 53 for O(3) in spring-summer (April-August). Monthly 2% percentile values for these gases reached (microg m (-3)) 561 for SO(2), 140 for NO(2) and 170 for O(3). The filtering efficiencies of the charcoal filters used averaged 60% for SO(2), 50% for NO(2) and 70% for O(3). All plants of winter barley from the unchambered plot were killed by severe frost periods in winter, 1986. Little frost damage occurred on plants grown in the chambers. Air filtration resulted in higher numbers of plants of winter barley per pot, i.e. a higher number of individuals per area, and a higher dry weight of whole plants and ears compared to the non-filtered atmosphere. In the experiments with spring barley, fresh and dry weight of whole plants were lower and dry weight of leaves were higher in the filtered open-top chambers. These effects could not be observed at all harvests which were carried out during the growing season. Grain yield and sulphur content of the leaves of both barley cultivars were not affected by the air filtration. Production of biomass of spring barley grown in ambient air was higher than of that grown in open-top chambers.  相似文献

Groundwater is the main source of water in Mediterranean, water-scarce, semiarid regions of Tunisia, Africa. In this study of the Korba coastal aquifer, 17 water wells were studied to assess their suitability for irrigation and drinking purposes. Assessment parameters include pH, salinity, specific ion toxicity, sodium adsorption ratio, nutrients, trace metals pollutants, and fecal indicators and pathogens. Results indicate that salinity of groundwater varied between 0.36 dS/m and 17.4 dS/m; in addition, its degree of restriction is defined as "none", "slight to moderate", and "severe" for 18, 23, and 59% of the studied wells, respectively. To control salts brought in by irrigation waters, the question arises as to how much water should be used to reach crop and soil requirements. To answer this question, a new approach that calculates the optimum amount of irrigation water considering the electrical conductivity of well water (ECw), field crops, and the semiarid meteorological local conditions for evapotranspiration and rainfall is developed. This is applied to the authors' case study area; barley and lettuce were selected among the commonly grown crops because they are high- and low-salinity tolerant, respectively. Leaching requirements were found to be independent of the crop selected, and depend only on the season, that is, 250 to 260 mm/month in the driest season, with a minimum of 47 mm/month though all seasons. A high bacteriological contamination appears in almost all samples. However, if disinfected and corrected for pH, all the well waters can be used for animal farming (including livestock and poultry), although only 29% could be used for human consumption.  相似文献
