Biochemical genetic variation at a leucine aminopeptidase (LAP) locus is related to salinity variation in several marine bivalve molluscs. This paper details an investigation of the Long Island Sound model of LAP selection (LAP genotype-dependent mortality occurring among newly settled Mytilus edulis mussels) carried out in 1997 among three M. galloprovincialis mussel populations along the salinity gradient of Wellington Harbour, New Zealand. Significant LAP genotypic heterogeneity was observed at the LAP locus between small (<25 mm shell length) and large (>25 mm shell length) M. galloprovincialis from Petone and Eastbourne (the two sites experiencing the greatest salinity variation), whereas genotypic heterogeneity was not significantly different between small and large mussels from Seatoun (the site experiencing the least salinity variation). The Lap 3 allele decreased in frequency and the Lap 4 allele increased in frequency at Petone and Eastbourne, whereas the Lap 3 and Lap 4 allele frequencies remained effectively constant at Seatoun. Both these findings are consistent with the Long Island Sound model of selection. At all three locations, the Lap 3,3 genotype decreased in frequency from small to large mussels, whereas the Lap 3,4 genotype increased in frequency from small to large mussels. All other LAP genotypes occurred at low frequencies (<0.10) at all three locations and showed no evidence of frequency change from small to large-size mussels nor evidence of clinal change among the three locations. These genotype frequency data possibly indicate that the Lap 3,3 genotype is at a selective disadvantage compared to the Lap 3,4 genotype at all three locations, and that this selective disadvantage is related to the extent of salinity variation which exists at each location. Further investigation is required before it can be determined if the Long Island Sound model of selection best describes the size-dependent and location-specific changes in LAP allele and genotype frequencies along this salinity gradient. Comparison of the population genetic structure at the LAP locus in 1995 and in 1997 revealed a profound change from heterozygote excesses to heterozygote deficiencies for all three M. galloprovincialis populations. The reason for the change is unknown, but the change indicates that population genetic structure at the LAP locus is highly variable in time, but consistent in space, among these M.␣galloprovincialis populations. Analysis of stable isotopes of oxygen and carbon in the otolith carbonate of pink snapper, Pagrus auratus, from several locations in Shark Bay, Western Australia, indicated that snapper are highly location specific. The hypersaline (36 to >60‰) Shark Bay, on the coast of Western Australia, generated strongly characteristic isotopic signatures in the otolith carbonate of snapper collected from the various locations indicating low levels of individual movement of the species. Oxygen isotopes showed enrichment in 18O in otolith carbonate with salinity (0.10: Δ δ18O/Δ salinity ‰) typical for the evaporation of seawater. The enrichment in 13C (up to 1.75‰) was attributed to the incorporation of metabolically derived CO2 from an enrichment of 13C in the food web within Shark Bay. This was possibly a result of lower concentrations of dissolved CO2 with increasing salinity causing a reduction in isotope fractionation during photosynthesis. Results complement recent genetic and tagging studies and provide further evidence of the complex nature of snapper stock structure in the Shark Bay region. Published online: 17 July 2002  相似文献

Samples representing the damselfishes (Perciformes: Pomacentridae) Stegastes nigricans, Chrysiptera biocellata, C. glauca, and C. leucopoma from North Male Atoll, Republic of Maldives, and Guam, Southern Marianas, were surveyed electrophoretically. A sample representing C. leucopoma from Ngemelis Island, Republic of Palau, was included in the analysis to provide a within-Micronesia comparison. On average, products of 23 presumptive protein-coding loci were examined in each sample. In comparisons of Maldivian and Guamanian samples, absolute or virtually fixed allelic differences were detected at: (1) ADA * and PEPS * for Stegastes nigricans; (2) ADA *, AAT-1 *, AAT-2 *, and mMDH * for C. biocellata, and; (3) ADA *, AAT-1 *, and AAT-2 * for C. glauca. At three polymorphic loci, ADA *, sMDH-1 *, and sMDH-2 *, similar allele frequencies were found in Palauan and Guamanian samples representing C. leucopoma, suggesting that these samples share a gene pool. Significantly different allele frequencies at ADA * and sMDH-1 * and a virtually fixed allelic difference at sMDH-2 * were detected between the Maldivian samples and each of the Micronesian samples representing C. leucopoma. The observed patterns of allele frequency differentiation suggest that Maldivian and Micronesian samples of each of the four study species represent separate demes.  相似文献

A survey of intraspecific allozymic variation among samples of the damselfishes Chrysiptera cyanea and Pomacentrus coelestis collected from Palau and Okinawa revealed levels of genetic divergence far in excess of estimates reported previously for populations of coral reef fishes. Absolute or nearly fixed differences in allele frequencies were detected at ADA *, sMDH-2 *, MEP-1 *, PEPB *, PEP-LT * and sSOD *, and at sAAT *, GPI-A *, LDH-1 *, and PEPB *, among the geographic samples of C. cyanea and P. coelestis, respectively. Examination of allele frequencies at most other loci (26 for C. cyanea; 24 for P. coelestis) revealed slight differentiation or identical fixation among the geographic samples. The observed patterns of allele frequency distribution suggest the existence of localized demes of these fishes: these demes may be cryptic and/or incipient species. Whether or not speciation is incipient, the observed patterns of allozymic and isozymic variability suggest that natural selection is a factor in the maintenance of population substructuring of the study species. Pronounced allelic differences were highly concentrated at a small number of loci: strongly bimodal frequency distributions of loci relative to their genetic identities (I) were observed for among-population comparisons in both study species. Allozymes encoded by the PEPB * locus in samples of the noncongeners from Palau exhibited identical electrophoretic mobilities. In Palauan P. coelestis, mSOD * is expressed but sSOD * apparently is not, whereas in Okinawan P. coelestis, both mSOD * and sSOD * are expressed.  相似文献

The tiger shark (Galeocerdo cuvier Peron and Lesueur 1822) is a widely distributed predator with a broad diet and the potential to affect marine community structure, yet information on local patterns of abundance for this species is lacking. Tiger shark catch data were gathered over 7 years of tag and release research fishing (1991–2000, 2002–2004) in Shark Bay, Western Australia (25°45′S, 113°44′E). Sharks were caught using drumlines deployed in six permanent zones (~3 km2 in area). Fishing effort was standardized across days and months, and catch rates on hooks were expressed as the number of sharks caught h−1. A total of 449 individual tiger sharks was captured; 29 were recaptured. Tiger shark catch rate showed seasonal periodicity, being higher during the warm season (Sep–May) than during the cold season (Jun–Aug), and was marked by inter-annual variability. The most striking feature of the catch data was a consistent pattern of slow, continuous variation within each year from a peak during the height of the warm season (February) to a trough in the cold season (July). Annual growth rates of recaptured individuals were generally consistent with estimates from other regions, but exceeded those for populations elsewhere for sharks >275 cm fork length (FL), perhaps because mature sharks in the study area rely heavily on large prey. The data suggest that (1) the threat of predation faced by animals consumed by tiger sharks fluctuates dramatically within and between years, and (2) efforts to monitor large shark abundance should be extensive enough to detect inter-annual variation and sufficiently intensive to account for intra-annual trends.  相似文献

Summary In a natural population, we document changes in the frequencies of alleles influencing body size and size-correlated alternative male reproductive tactics, and we examine the possible role of sexual selection in producing these changes. Male swordtails (Xiphophorus nigrensis, Rio Choy) exhibit three body size classes (small, intermediate, and large) that primarily derive from allelic variation (s, I, L) at the Y-linked pituitary (P) locus. Some XX individuals are male. They are small and can be fathered either by XX or XY males. We compared the frequencies of Y-linked P alleles across two generations in a natural population. There was a significant decrease in the Y-s genotype relative to Y-I and Y-L genotypes. Laboratory experiments suggest that the disadvantage of Y-s resulted, at least in part, from female preference for larger courting males relative to small chasing males. All larger courting males are of the Y-I or Y-L genotypes. Although the frequency of Y-linked P alleles changed across generations, there was no change in the distribution of male body size classes. The stability of the phenotypic distribution across generations, despite changes in P allele frequencies, was maintained by the production of small XX sons by Y-I and Y-L males. The Atlantic surfclam, Spisula solidissima (Dillwyn, 1817), is a dominant member of the benthic community on the continental shelf from Georges Bank to North Carolina, USA. This bivalve has supported a major fishery, primarily off New Jersey and the Delmarva Peninsula, since the 1960s. Early papers documented that these populations were at historical lows in the mid-1970s owing to commercial harvesting and a hypoxic event off New Jersey. It was also shown that major recruitment took place off New Jersey in 1976 and off the Delmarva Peninsula in 1977. Because the size frequencies of surfclams from federal surveys do not show distinct year classes, there has been uncertainty about the number of year classes in these populations throughout the 1980s and 1990s. The present study describes changes in population age- and size-structure from 1978 to 1997 in federal waters (≥5.5 km from shore) of the USA. Given the 30 to 35 year life span of S. solidissima, these populations could be composed of many year classes. Yet, these populations were composed of only two to three year classes in 1978. Through annual recruitment, the number of year classes increased over time, and populations off New Jersey and the Delmarva Peninsula contained at least 19 year classes in 1997. This major change in population structure over time was not evident from examination of available size-frequency data, and could only be inferred from data on age-composition. Throughout the 1980s and 1990s, the surfclam fishery was supported by multiple year classes. The mean and variance of recruitment to the New Jersey region, as indicated by the abundance of 4-year-olds over time, was greater than that off Delmarva, particularly between 1980 and 1986. The instantaneous rate of adult mortality, which includes the effect of harvesting, was approximately 0.26 yr−1 in each region. Received: 11 September 1998 / Accepted: 1 February 1999  相似文献

In 1986 we observed significant genetic heterogeneity among samples of bicolor damselfish (Stegastes partitus) from geographically proximate coral reefs in the Florida Keys. This result was in contrast to results of virtually all previous studies of coral reef fishes. To assess temporal stability of localized genetic differentiation, we resampled reefs in 1989. Within approximately two generations, allele frequencies at the most differentiated locus,ACO-1 *, had changed by as much as 0.65, resulting in almost complete homogeneity between previously differentiated subpopulations. Estimates of 10.78 migrants per generation suggested that high gene flow is the most likely factor responsible for the significant change in allele frequencies. We attribute the original genetic differentiation to a population bottleneck, possibly caused by environmental perturbations such as hurricanes and consequent genetic drift. For reef fishes with pelagic egg and/or larval stages, a growing body of data suggests that: (1) populations are geographically extensive gene pools, and (2) rates of dispersal appear to be high enough to allow for continual repopulation of isolated and/or perturbed coral reef communities.  相似文献

Six shells of the Gastropoda species Nassa mutabilis L. were collected in the Adriatic Sea at a depth of a few metres off the central Italian coast during 1996 and 1997. The oxygen and carbon isotope composition of spot samples collected from the shell apex to the top was measured according to well-established techniques. The results obtained allow the following conclusions: (1) the CaCO3 of the N. mutabilis shells is essentially formed by aragonite as proved by X-ray diffraction measurements; (2) the CaCO3 is precipitated under isotope equilibrium conditions or very close to equilibrium with the environmental water; (3) shell accretion takes place all year round, although the warm period is preferred since the accretion rates are higher during the warm half of the year and considerably reduced in winter; (4) the carbon isotope composition changes according to the age of the specimen, probably in relation to changes in its nutritional regimen; and (5) the alternation of warm and cold periods recorded by the shells allows the evaluation of their age and of the season of their birth and death. Colonies of Pocillopora spp. containing the crab Trapezia digitalis were collected from Hawaii in summer of 1979 and of 1980. Individuals of T. digitalis in Hawaii vary extensively in coloration of the carapace: crabs were separated into phenotypic classes by their degree of carapace reticulation. When presented with two females of different phenotype, males preferentially paired with the female which was similar to them in reticulation. The genetic population structure of T. digitalis was unusual by comparison with other species of Trapezia. Previously reported homozygote excesses at esterase-4, hexokinase-1, phosphoglucoseisomerase, and general protein-2 cannot be explained by the single-locus Wahlund effect, since there was not significant genetic differentiation among phenotypic classes, as measured by Wright's F ST statistic. There was significant variation among classes in allele frequencies at a fifth locus, hexokinase-2. Two pairs of loci were in significant gametic disequilibrium. Gametic disequilibrium was not detected in seven other species of Trapezia. It is suggested that multi-locus genetic analysis and an increased geographic sampling range are needed to determine the genetic population structure of T. digitalis in relation to observed variation in coloration.  相似文献

Latitudinal differences in host-specificity of marine Monogenea and digenea   总被引:1,自引:0,他引:1
Rohde  K. 《Marine Biology》1978,47(2):125-134
Data from 15 surveys of marine trematodes (average of 91 trematode species and more than 80 fish species per survey) and from 12 surveys of marine Monogenea (average of 52 Monogenea species and more than 49 fish species per survey) show that the degree of host specificity of marine digenetic trematodes increases from cold to warm seas; Monogenea do not show such a trend, and the trend is probably reversed in the Pacific Ocean. The difference between the two groups is explained in terms of r - and k - strategy. Monogenea tend to follow a k - strategy (great complexity of adult, few offspring), which results in a high degree of host- and sitespecificity to facilitate mating in low-density populations. Only one or a few related host species can be infected, and as more related host species are present in the warm Pacific, host-specificity there is reduced. Digenea tend to follow an r- strategy (simple strucure of adult, many offspring), part of which is to infect many ecologically suitable hosts. Host-specificity in cold-temperate seas is reduced because of the less patchy and ecologically less restricted distribution of hosts.  相似文献

Allozymes were examined in quantitative lunar monthly collections of larval recruits of the western rock lobster Panulirus cygnus George over three recruitment seasons at two sites nearly 350 km apart in Western Australia. At Alkimos, the southern site, recruitment occurs in a relatively narrow peak early in the spring, whereas at the northern Houtman Abrolhos Islands, recruitment extends into the summer months. In the 1995/1996 recruitment season, the frequency of the GPI * 100 allele increased from early to late in the season, but the frequencies were indistinguishable at the two sites in each monthly collection. The combination of this temporal variation in allelic frequencies with the contrasting patterns of recruitment at the Abrolhos Islands and Alkimos resulted in genetically different cohorts at the two sites. This pattern was ephemeral, as it was not repeated in the subsequent two years. Thus, ephemeral genetic patchiness in P. cygnus can be generated by the locally-specific genetic mix of recruits obtained from a common larval pool. This mechanism is the probable explanation of previously observed temporal and possible spatial genetic variation in adult P. cygnus, and highlights the importance of studying recruitment in order to understand the genetic structure of marine species. Samples were taken from a 2 900 m deep permanent station (centred on 54°40N; 12°16W) in the southern Rockall Trough, from November 1975 to September 1980, inclusive. Revision of juvenile size frequencies was necessary after removal of postlarvae of an annual non-viable settlement of Ophiocten gracilis (G. O. Sars) that were mistakenly included in a previous analysis. The present study of the revised data shows that settlement of Ophiura ljungmani (Lyman) occurs in summer when disk-size frequencies are dominated by the newly metamorphosed postlarvae. The likely age composition in larger sizes was suggested from the progression of modes along the size axis in juvenile size classes in consecutive were developed from Gaussian mixtures fitted by constrained computer search using, as starting values, component parameter values suggested from the frequency histograms. Choice between competing solutions was made on the basis both of goodness-of-fit and biological plausibility. However, identification of year-class structure amongst sizes >3 to 4 mm disk diameter remained conjectural owing to the low frequencies of adults in the samples. Comparison of the fitted component-means in the time series suggested a marked seasonal growth pattern, with maximal growth in spring. Probable age composition amongst adult sizes was extrapolated from von Bertalanffy growth parameters estimated from the means of juvenile year classes.Communicated by J. Mauchline, Oban  相似文献

We evaluated stock structure and demographic (population) history of red snapper (Lutjanus campechanus) in the northern Gulf of Mexico (Gulf) via analysis of mitochondrial (mt)DNA sequences from 360 individuals sampled from four cohorts (year classes) at three localities across the northern Gulf. Exact tests of genetic homogeneity and analysis of molecular variance both among cohorts within localities and among localities were non-significant. Nested clade analysis provided evidence of different temporal episodes of both range expansion and restricted gene flow due to isolation by distance. A mismatch distribution of pairwise differences among mtDNA haplotypes and a maximum-likelihood coalescence analysis indicated a population expansion phase that dated to the Pleistocene and probably represents (re)colonization of the continental shelf following glacial retreat. The spatial distribution of red snapper in the northern Gulf appears to have a complex history that likely reflects glacial advance/retreat, habitat availability and suitability, and hydrology. Habitat availability/suitability and hydrology may partially restrict gene flow among present-day red snapper in the northern Gulf and give rise to a metapopulation structure with variable demographic connectivity. This type of population structure may be difficult to detect with commonly used, selectively neutral genetic markers.Communicated by P.W. Sammarco, Chauvin  相似文献

Seafood mislabeling is common in both domestic and international markets. Studies on seafood fraud often report high rates of mislabeling (e.g., >70%), but these studies have been limited to a single sampling year, which means it is difficult to assess the impact of stricter governmental truth‐in‐labeling regulations. We used DNA barcoding to assess seafood labeling in 26 sushi restaurants in Los Angeles over 4 years. Seafood from 3 high‐end grocery stores were also sampled (n = 16) in 2014. We ordered 9 common sushi fish from menus, preserved tissue samples in 95% ethanol, extracted the genomic DNA, amplified and sequenced a portion of the mtDNA COI gene, and identified the resulting sequence to known fish sequences from the National Center for Biotechnology Information nucleotide database. We compared DNA results with the U.S. Food and Drug Administration (FDA) list of acceptable market names and retail names. We considered sushi‐sample labels that were inconsistent with FDA names mislabeled. Sushi restaurants had a consistently high percentage of mislabeling (47%; 151 of 323) from 2012 to 2015, yet mislabeling was not homogenous across species. Halibut, red snapper, yellowfin tuna, and yellowtail had consistently high (<77%) occurrences of mislabeling on menus, whereas mislabeling of salmon and mackerel were typically low (>15%). All sampled sushi restaurants had at least one case of mislabeling. Mislabeling of sushi‐grade fish from high‐end grocery stores was also identified in red snapper, yellowfin tuna, and yellowtail, but at a slightly lower frequency (42%) than sushi restaurants. Despite increased regulatory measures and media attention, we found seafood mislabeling continues to be prevalent.  相似文献

To determine the genetic population structure of blue crabs (Callinectes sapidus Rathbun), electrophoretic allozyme analysis was performed on 750 individuals collected from 16 nearshore locations ranging from New York to Texas, USA. Twenty enzymes and non-enzymatic proteins coded by 31 presumptive loci were examined. Twenty-two loci were either monomorphic or polymorphic at less than theP 95 level; alleles for these polymorphic loci were geographically dispersed. Allele frequencies for three of the remaining polymorphic loci were homogeneous over all populations, as were levels of polymorphism and heterozygosity. Phenograms generated by the UPGMA (unweighted pair-group method using arithmetic averages) and distance Wagner methods exhibited no geographic pattern in the clustering of populations. Estimates ofN em (effective number of migrants per generation between populations) indicated substantial gene flow, with aalues sufficiently high to infer panmixia between all blue crab populations from New York to Texas. However, despite this high level of gene flow, two striking patterns of geographic differentiation occurred: genetic patchiness and clinal variation. Allele frequencies atEST-2, GP-1, IDHP-2, DPEP-1, DPEP-2, andTPEP exhibited genetic patchiness on local and range-wide geographic scales, and allele frequencies atEST-2 varied temporally. Genetic patchiness in blue crabs is likely to be the result of the pre-settlement formation and subsequent settlement of genetically heterogeneous patches of larvae; allele frequencies of those larval patches may then be further modified through ontogeny by localized selection. In the Atlantic Ocean, a regional latitudinal cline ofEST-2 allele frequencies was superimposed on the range-wide genetic patchiness exhibited by that locus. This pattern against a background of high gene flow is highly likely to be maintained by selection. In estuaries along the Atlantic Ocean coast, a combination of low adult long-distance migration and a high retention rate of locally spawned larvae could serve to segregate populations and allow for the development of the geographic cline inEST-2. The Gulf of Mexico showed no apparent cline, perhaps due to long-distance migration of females in some regions of the Gulf, or to masking by genetic patchiness. These results emphasize the importance of both ecological and evolutionary time scales and structuring mechanisms in determining genetic population structure.  相似文献

The reproductive parameters of three species of barnacles common in warm-water regions,Balanus amphitrite Darwin,B. eburneus Gould andB. trigonus Darwin, cultured singly or in pairs for 2 1/2 yr, were compared. All specimens, collected from Tokyo Bay and studied between 1985 and 1987, were fed on newly hatchedArtemia sp. larvae (Tetra strain) and phytoplankton culture and were maintained under constant temperature (20°C) and controlled photoperiods (10 h light:14 h dark). The breeding frequencies of these three species were equally high, from 2 to 4 broods mo–1 in the first year; they were relatively high between October and February, averaging 3 broods mo–1.B. amphitrite, B. eburneus andB. trigonus produced 24, 21 and 11 broods yr–1, respectively. Brood size was quite variable among the three species examined, yielding 4000 embryos brood–1 inB. amphitrite andB. eburneus and 6000 inB. trigonus. Brood interval averaged 4 to 9 d. The moult occurred over short intervals of 4 to 10 d moult–1. Both breeding and moulting frequencies were influenced by low temperature and starvation. Self-fertilization was observed in all species and was more frequent inB. amphitrite than inB. eburneus andB. trigonus. Nutritional condition and temperature seemed to be the main external factors regulating both breeding and moulting processes in barnacles.  相似文献

Samples of southern bluefin tuna, Thunnus maccoyii (Castelnau), taken from off the coasts of South Africa, Western Australia, South Australia and Tasmania from 1992 to 1994 were analysed for six polymorphic allozyme loci (ADA *, GDA *, GPI-A *, MPI *, PGDH * and PGM-1 *, n = 595 to 733 per locus) and for mitochondrial DNA variants revealed by three restriction enzymes (Bam HI, Bcl I and Eco RI) detecting polymorphic cut sites (n = 555). No significant spatial heterogeneity was detected. There were no sex-related differences in allele or mtDNA haplotype frequencies. Juveniles (30 to 35 cm and 46 to 54 cm) from what are thought to be two temporally-separated spawning peaks showed no significant genetic differentiation. There were also no significant differences in allele or haplotype frequencies between fish smaller than 70 cm and those larger than 70 cm. These data are consistent with the null hypothesis of a single unit stock of southern bluefin tuna, with a single spawning area. This is located to the south of Java and off the north-west coast of Australia. Received: 28 August 1996 / Accepted: 30 September 1996  相似文献

This study investigated incorporation and metabolism of saturated [(1-14C) 16:0] and unsaturated [(1-14C) 18:26 and (1-14C) 18:33] fatty acids in adult eastern oysters,Crassostrea virginica Gmelin (spawned from parents obtained in 1986 from Mobjack Bay, Virginia, USA), and the influence of temperature on these processes. InC. virginica, incorporation of injected palmitic (16:0) and linolenic (18:33) acids was increased when oysters which had been grown in warm water (22 to 23°C) were transfered to cold water (5 to 7°C) for 8 to 18 d. Incorporation of linoleic acid (18:26) was unchanged under these conditions. The changes in concentration may have been linked to depression of metabolism in these oysters, in particular that of 16:0, which was reduced by 90%. Oxidation of incorporated fatty acids was much higher in warm than in cold water. Cold-temperature conditioning ofC. virginica altered the distribution of fatty acids among the neutral and polar lipid fractions. Long-term exposure to cold water increased the proportion of fatty acids in the polar fraction, which may be related to maintenance of membrane fluidity. Short-term exposure to cold water had the opposite effect, which may be due to increased energy requirements as the oyster adapts to new conditions. Reutilization of14C-acyl groups demonstrated de novo synthesis of 16:0 and 18:0 fatty acids. Only limited elongation and no desaturation of the administered fatty acids was observed.  相似文献

King mackerel (Scomberomorus cavalla Cuvier) collected in 1992 and 1993 from 13 localities along the Atlantic coast of the southeastern USA and in the northern Gulf of Mexico were surveyed for variation in mitochondrial (mt)DNA and a nuclear-encoded dipeptidase locus (PEPA-2). Both polymorphic and fixed mtDNA restriction sites were identified and mapped using conventional and polymerase chain-reaction (PCR)-based methods. Heterogeneity in mtDNA haplotype frequencies was found only in comparisons of pooled haplotypes from Atlantic localities versus pooled haplotypes from Gulf localities. This finding indicates weak genetic divergence between king mackerel from the Atlantic and those from the Gulf. Frequencies of two PEPA-2 alleles essentially paralleled previous findings: one allele (PEPA-2a) was common among samples from western Gulf localities, whereas the other allele (PEPA-2b) was common among samples from Atlantic and eastern Gulf localities. There was considerable variation in PEPA-2 allele frequencies within broadly-defined regions. Variation in mtDNA haplotypes and PEPA-2 genotypes was independent, as was variation in mtDNA haplotypes with sex or age of individuals. Variation in PEPA-2 genotypes was not independent of sex or age of individuals. The latter result suggests that frequencies of PEPA-2 alleles in samples of king mackerel may stem, in part, from sex and age distributions of individuals within samples, and indicates that caution should be exercised in using allelic variation at PEPA-2 as a measure of population (stock) structure in king mackerel. The discordance in spatial patterning of mtDNA haplotypes versus PEPA-2 alleles across the Gulf (i.e. homogeneity in mtDNA haplotype frequencies versus heterogeneity in PEPA-2 allele frequencies) may be due to either female excess at several localities, sex-biased migration, or both. Observed patterns of genetic variation also are consistent with the hypothesis that king mackerel in the western Atlantic may have been subdivided during Pleistocene glaciation, and that the current distribution of PEPA-2 alleles may be a historical artefact.
