Nickel has an adverse effect on some aspects of protein metabolism of the freshwater fish, Cyprinus carpio. The main changes observed were: (a) Decrease in soluble, structural and total proteins, AlAT and AAT activities with an increase in the levels of free amino acids, protease and GDH activities and ammonia in the gill and kidney at 1, 2, 3, and 4 days of exposure to a lethal concentration, 40 mg litre(-1) of nickel; (b) Increase in soluble, structural and total proteins, free amino acids and the activities of protease, AlAT, AAT and GDH with a decrease in ammonia and urea in these organs at 1, 5, 10 and 15 days of exposure to sublethal concentration, 8 mg litre(-1); (c) The magnitude in these changes increased over time with both concentrations of the metal, and was more marked in gill than in kidney.  相似文献

Freshwater fish, Cyprinus carpio were exposed to sublethal concentration (1.2 microg l(-1)) of cypermethrin for 6, 12, 24 and 48 h to analyze various parameters of protein metabolism in functionally different tissues. Total, structural and soluble proteins showed decrement; where as free amino acids and the activities of protease, aspartate aminotransferase and alanine aminotransferase significantly increased in cypermethrin exposed fish. Interestingly, ammonia content decreased but urea and glutamine increased at all periods of exposure. It was also observed that alterations steadily increased with the period of exposure and exhibited tissue specificity. Thus variation in the protein metabolism of the fish exposed to cypermethrin indicates its toxic effect on the cellular metabolism thereby leading to impaired protein synthetic machinery.  相似文献

Piscicidal activities of aqueous extracts of Euphorbia tirucalli were very well established, but their ultimate mode of action on fish metabolism was not yet known. Exposure of fishes over 24h or 96h to sub-lethal doses (40% and 80% of LC(50)) of aqueous extract of E. tirucalli stem-bark and latex, significantly (P<0.05) altered the level of total protein, total free amino acids, nucleic acids, glycogen, pyruvate, lactate and activity of protease, alanine aminotransferase, aspartate aminotransferase, acetylcholinesterase and cytochrome oxidase enzyme in liver and muscle tissues of freshwater fish Channa punctatus. The alterations in all these biochemical parameters were significantly (P<0.05) time- and dose-dependent. After 7d of withdrawal of treatment of 80% of LC(50) of E. tirucalli extracts shows that there was a partial recovery in the levels of glycogen but nearly complete recovery in total protein, total free amino acids, pyruvate, lactate, nucleic acids level and activity of protease, aspartate aminotransferase, alanine aminotransferase, acetylcholinesterase and cytochrome oxidase enzyme in both the tissues of fish. Thus aqueous extracts of E. tirucalli adversely affect respiratory pathway of fish and cause energy crisis during stress by suppressing ATP production. The reversibility of the action of the aqueous extracts would be an additional advantage in their use.  相似文献

Exposure of the freshwater field crab Barytelphusa guerini to a sublethal concentration of NaF (30 mg litre(-1)) caused significant alterations in protein metabolism. After an exposure time of 15 days, the crab was found to have a marked depletion of total protein and free amino acid content. A significant elevation in transaminases, Aspartate aminotransferase (AAT) and Alanine aminotransferase (ALAT), and Glutamate dehydrogenase (GDH) activities was reflected in the free amino acid levels of the tissues.  相似文献

Jundiá (Rhamdia quelen, Quoy and Gaimard), a South American teleostean fish, was exposed to sub-lethal concentrations of cypermethrin (30% and 45% of the 48-h LC(50) value of 0.265 ppm) for 2, 4 or 8 days. Serum biochemical and hematological values and behavioral changes were studied. The 30% LC(50), 0.08 ppm, produced significant increases in Mg(2+), P, K(+), creatinine, urea, glucose, cholesterol, aspartate aminotransferase and alkaline phosphatase levels, and reduction in total proteins and triglycerides in serum. The 45% LC(50), 0.12 ppm, produced significant increase in Na(+), Mg(2+), P, K(+), creatinine, urea, glucose, cholesterol, and alkaline phosphatase, and reduction in triglycerides and alanine aminotransferase levels in serum. At this concentration, the fish showed behavior changes such as hyper-excitability, asphyxia, and widening of mouth and operculum. The hematological values remained normal, except for hemoglobin concentrations and the mean corpuscular hemoglobin concentration, which increased with exposure to 0.08 ppm and 0.12 ppm cypermethrin. Results of the present work show that biochemical analysis of serum can be useful to detect incipient cypermethrin intoxication of the shoal.  相似文献

The diethyl ether, chloroform, acetone and methanol extract of Nerium indicum leaf were evaluated for their piscicidal activity against common freshwater air breathing predatory fish Channa punctatus. The rank of order of toxicity (LC50) of the leaf extract was, diethyl ether extract (17.34 mg/l)>acetone (40.01 mg/l)>chloroform (40.61 mg/l)>and methanol (106.37 mg/l). There was a significant negative correlation between LC50 values and exposure periods. Thus increase in exposure period, LC50 decreases from 17.34 mg/l (24 h) to >13.58 mg/l (96 h) in the diethyl ether extract. Similar trends were also observed in acetone, chloroform and methanol extracts. Exposure of sub-lethal doses (40% and 80% of LC50) of the diethyl ether extract of N. indicum leaf (which has maximum piscicidal activity) for 24 or 96 h caused significant alteration in the level of total protein, total free amino acid, nucleic acid, glycogen, pyruvate, lactate and activity of enzyme protease, phosphatases, alanine aminotransferase, aspartate aminotransferase and acetylcholinesterase in liver and muscle tissue. The alterations in all the above biochemical parameters were significantly (P<0.05) time and dose dependent. There was a significant recovery in all the above biochemical parameters, in both liver and muscle tissues of fish after the seventh day of the withdrawal of treatment. Thus, the leaf extracts of N. indicum have potent piscicidal activity against fish C. punctatus and also significantly affect both aerobic and anaerobic pathway of respiration in fish.  相似文献

Bisphenol A (BPA), which is ubiquitous in the environment, is an example of an endocrine-disrupting compound (EDC). Ammonium assimilation has an important function in plant growth and development. However, insufficient information on the potential effect of BPA on ammonium assimilation in plants is available. In this study, the effects of BPA on ammonium assimilation in roots of soybean seedlings were investigated. During the stress period, 1.5 mg L?1 of BPA improved glutamine synthetase (GS)/glutamate synthase (GOGAT) cycle and glutamate dehydrogenase (GDH) pathway in ammonium assimilation. The amino acid and the soluble protein contents increased in the soybeans. At 17.2 and 50.0 mg L?1 of BPA, the GS/GOGAT cycle was inhibited and the GDH pathway was promoted. The amino acid content increased and the soluble protein content decreased. During the recovery period, the GS/GOGAT cycle and the GDH pathway recovered at 1.5 and 17.2 mg L?1 of BPA but not at 50.0 mg L?1 of BPA. The amino acid content continuously increased and the soluble protein content decreased compared with those in the control treatment. In summary, BPA treatment could affect the contents of soluble protein and amino acid in the soybean roots by regulating ammonium assimilation.  相似文献

Chlorpyrifos (O,O-diethyl O-3,5,6-trichloro-2-pyridylphosphorothioate) is one of the organophosphate pesticides widely used in agricultural practices throughout world and irreversible inhibitor of cholinesterase in all animal species. Limited efforts have been made to study acute genotoxic effects of chlorpyrifos (CPF) in different tissues of fish using genotoxic biomarkers. Therefore, the present investigation was aimed to study the induction of DNA damage by CPF in freshwater teleost fish Channapunctatus using micronucleus assay (MN assay) and alkaline single-cell gel electrophoresis (comet assay). The value of LC(50) - 96 h of CPF was determined as 811.98 microgl(-1) for C. punctatus, in a semi-static system and on the basis of LC(50) value three acute concentrations viz., 203, 406 and 609 microgl(-1) were determined. The fishes were exposed to the different concentrations of CPF for 96 h and samplings were done at regular intervals for assessment of the MN frequencies and DNA damage. In general, significant effects (P<0.01) from both concentrations and time of exposure were observed in exposed fishes. It was found that the micronucleus induction was highest on 96 h at all concentrations in the peripheral blood. Similar trend was observed for the DNA damage measured in terms of the percentage of tail DNA in the lymphocyte and gill cells. This study explored the combined use of micronucleus assay and comet assay for in vivo laboratory studies using fresh water fish for screening the genotoxic potential of xenobiotics.  相似文献

Cypermethrin, a type II pyrethroid insecticide, is widely used throughout the world in agriculture, forestry, horticulture and homes. Though the neurotoxicity of cypermethrin has been thoroughly studied in adult rodents, little is so far available regarding the developmental toxicity of cypermethrin to fish in early life stages. To explore the potential developmental toxicity of cypermethrin, 4-h post-fertilization (hpf) zebrafish embryos were exposed to various concentrations of cypermethrin (0, 25, 50, 100, 200 and 400 μg L−1) until 96 h. Among a suite of morphological abnormalities, the unique phenotype curvature was observed at concentrations as low as 25 μg L−1. Studies revealed that 400 μg L−1 cypermethrin significantly increased malondialdehyde production. In addition, activity of antioxidative enzymes including superoxide dismutase and catalase were significantly induced in zebrafish larvae in a concentration-dependent manner. To further investigate the toxic effects of cypermethrin on fish, acridine orange (AO) staining was performed at 400 μg L−1 cypermethrin and the result showed notable signs of apoptosis mainly in the nervous system. Cypermethrin also down-regulated ogg1 and increased p53 gene expression as well as the caspase-3 activity. Our results demonstrate that cypermethrin was able to induce oxidative stress and produce apoptosis through the involvement of caspases in zebrafish embryos. In this study, we investigated the developmental toxicity of cypermethrin using zebrafish embryos, which could be helpful in fully understanding the potential mechanisms of cypermethrin exposure during embryogenesis and also suggested that zebrafish could serve as an ideal model for studying developmental toxicity of environmental contaminants.  相似文献

Wild and farmed mussels, Mytilus edulis, coexist with salmon farms in Scottish sea lochs. A synthetic pyrethroid, cypermethrin, is licensed for use on fish farms to control sea lice infestations as a formulation called Excis. In this study, uptake of cypermethrin from Excis exposure is investigated through the use of gas chromatography with mass-spectrometry. The effects of Excis on mussels are also examined by measuring the neutral red retention time of lysosomes, aerial survival and shell closure. The isomeric ratios of cis:trans cypermethrin measured in mussels are around 80:20; a marked increase from 40:60 to which the mussels were exposed. This is most likely due to preferential metabolism of trans-isomers, as the same response is seen in vertebrates. There is a pronounced behavioural effect of shell closure, where mussels exposed to 1000 microg/l cypermethrin shut their shells within an hour of exposure. Arguments are presented for this effect being either a voluntary response on recognition of cypermethrin, or an effect arising from an involuntary action of cypermethrin on the adductor muscle. Even at 1000 microg/l cypermethrin, neutral red retention time and aerial survival are not affected. The data suggest that the responses of mussels shown here are unlikely to occur in the field, even at the concentrations of cypermethrin used in fish cages, for the treatment of sea lice.  相似文献

Our study was designed to investigate the induction of oxidative stress by cypermethrin; a Type II pyrethroid in rat liver and kidney. In addition, the protective role of sesame oil against the toxicity of cypermethrin was investigated. Animals were divided into four equal groups; the first group used as control while groups 2, 3 and 4 were treated with sesame oil (5 mL/kg b.w), cypermethrin (12 mg/kg b.w) and the combination of both sesame oil (5 mL/kg b.w) plus cypermethrin (12 mg/kg b.w), respectively. Rats were daily administered with their respective doses for 30 days by gavage. Repeated oral administration of cypermethrin was found to reduce the level of glutathione (GSH) and the activities of the antioxidant enzymes. While, the level of TBARS was elevated indicating the presence of oxidative stress. The activities of LDH, AST and ALT were decreased in the liver extract while increased in the plasma of the cypermethrin-treated group. Also, the levels of urea and creatinine were significantly increased after treatment with cypermethrin. Liver and kidney injury was confirmed by the histological changes. In conclusion, the administration of sesame oil provided significant protection against cypermethrin-induced oxidative stress, biochemical changes, histopathological damage and genomic DNA fragmentation.  相似文献

The impact of biotech-soybean technology on freshwater ecosystems is being evaluated in the Rolling Pampas region, Argentina. The effect of cypermethrin, the main soybean insecticide, on low-order temperate-stream fish populations was investigated for two consecutive crop cycles under field-use conditions in biotech-soybean production. Cypermethrin was unable to induce mortality or behavioral effects on any of the fish species resident in a first-order stream across a crop field (pulsed acute exposure scenario) sprayed according to conventional practices. No spatially or temporally dependent effects were observed on population parameters (size-class structure, abundance, survival, sex and immature/mature ratio, condition factor) of resident or caged Cnesterodon decemmaculatus after spraying or rainfall events, not even one year after, at the beginning of the next crop cycle. Although cypermethrin was "very highly toxic" to C. decemmaculatus in laboratory water (96h-LC(50)=0.43microg/l), its toxicity was reduced in filtered (78%) and unfiltered (92%) stream water. Changes in LC(50) values were mainly correlated with the OC content of each water fraction (r(2)=0.99; p<0.01; n=9), showing that both DOC and TOC contributed proportionally to toxicity reduction. Protective effects of stream water (12-fold reduction LC(50) values) explained the lack of effects on fish populations in the field, despite cypermethrin water concentrations after spraying reached values comparable with the 96h-LC(50). Therefore, cypermethrin under field-use conditions in transgenic-soybean production represents a low risk of acute exposure for fish populations inhabiting low-order temperate-streams rich in TOC. The relationship between LC(50) and TOC could be a convenient way to improve risk estimation based on laboratory toxicity testing.  相似文献

Three widely used xenobiotics pentachlorophenol (PCP), 2,4-dichlorophenoxyacetic acid (2,4-D) and 2-chloro-2,6-diethyl-N-(butoxymethyl) acetanilide (Butachlor) are evaluated for acute toxicity and stress behavior on freshwater fish (Heteropneustes fossilis, Clarias batrachus, Channa punctatus) and mosquito larvae (Culex pipiens fatigans). The experiment was carried out by medium treatment using intermittent flow-through system. Median lethal concentrations (LC50) were calculated by probit analysis. The LC50 values and 95% confidence intervals showed variable range for tested chemicals. Mosquito larvae generally appeared resistant than fish, while H. fossilis was found to be most sensitive. Stress signs in the form of behavioral changes are also observed. Both types of organisms are recommended as good bioindicator for the risk assessment of aquatic environment due to chemicals tested.  相似文献

Limited toxicity data are available for estuarine and marine species and the widely used pyrethroid insecticide, permethrin. This study determined acute effects of permethrin on survival, lipid peroxidation, acetylcholinesterase activity, and splenocyte proliferation for two fish species found in South Carolina estuaries; juvenile red drum (Sciaenops ocellatus) and adult mummichog (Fundulus heteroclitus). Juvenile S. ocellatus were significantly more sensitive than adult F. heteroclitus to permethrin exposure, with a 96-h LC50 value of 8 μg/L determined for red drum compared to 23 μg/L for mummichog. Lipid peroxidation activity of the liver increased in permethrin-treated fish compared to control animals after 24 h and decreased after 96 h. Permethrin had no effect on acetylcholinesterase activity of the brain at the concentrations tested. Permethrin exposure significantly inhibited splenocyte proliferation, indicating an immunosuppressive effect. Most of the effects of permethrin on fish cellular stress enzymes and survival occurred at concentrations much higher than those typically measured in the environment. However, inhibition of splenocyte proliferation in juvenile red drum occurred at approximately twice that of measured permethrin concentrations in surface water. These findings may prove useful to the future management and regulation of pyrethroid insecticide use near estuarine habitats.  相似文献

The pyrethroid pesticides at lower concentrations resulted in a slight activation of alpha-chymotrypsin. Methidation inhibited the alpha-chymotrypsin activity by about 20%. These pesticides modified the lipase activity to a lesser extent; the highest inhibition was measured with cypermethrin. The carboxypeptidase A activity was inhibited by both pyrethroid pesticides and methidation. The results suggest that these pesticides might interact with the active conformation of the studied hydrolytic enzymes, resulting in changes in their activities.  相似文献

Mortality caused by the aqueous extracts of leaf and stem bark of four plants belonging to family Euphorbiaceae and Apocynaceae against freshwater fish Channa punctatus has been reported. It was found that dilute aqueous solutions of leaf and stem bark were active in killing the fishes. The toxic effect of stem bark of all the plants were time as well as dose dependent. There was significant negative correlation between LC50 and exposure periods. Thus, the LC50 values of stem bark extracts of Euphorbia royleana, Jatropha gossypifolia, Nerium indicum and Thevelia peruviana were decreased from 0.050 g/l (24 h) > to 0.020 g/l (96 h); 4.61 g/l (24 h) > to 4.34 g/l (96 h); 0.097 g/l (24 h) > to 0.041 g/l (96 h) and 4.05 g/l (24 h) > to 3.17 g/l (96 h), respectively. It has been suggested that these plant products cannot be used directly in freshwater bodies, without their detailed studies on long-term effects on non-target organism as well their structure activity relationship.  相似文献
