During cadmium uptake by the shore crab Carcinus maenas (L.), cadmium appears in both haemolymph and urine within 1 h, and comes into equilibrium with the haemolymp after some 60 h. The equilibrium concentration for haemolymph cadmium is always considerably less than that of the external medium, whether the latter is close to background level or at the artificially high concentration of 2.2 mg l-1. Despite earlier evidence that cadmium entering the haemolymph is mainly bound to protein, current results show most of this to be freely labile and quickly displaced by cadmium taken up subsequently. There is some evidence to suggest that cadmium is passed on from the haemolymph to the hepatopancreas. A large proportion of whole-body cadmium is adsorbed onto the exoskeleton and gills. The comparatively rapid efflux of 109Cd from loaded crabs over a period of 1 week is mainly due to desorption of this cadmium from the outer body surface.  相似文献

The bloodworm Glycera dibranchiata Ehlers, 1968 accumulates cadmium through the general body surface and the intestine. Absorption through the gut accounts for cadmium which rapidly binds to coelomic proteins. Intracoelomic injection of 109Cd demonstrates that cadmium binds readily to hemoglobin and other proteins. The degree of cadmium binding is pH-dependent. The apparent pK of binding sites in body wall and musculature homogenates is 5.39. Cadmium ions injected into the coelom at 7 g g-1 tissue increase proline incorporation rates into the positively charged hemoglobin (cathode fraction) by 15-fold in 3 days. A 3-fold increase of proline incorporation was observed in the anode hemoglobin fraction over the same time period. Radioactivity in various protein fractions decreases at different rates after injection of 109Cd. Comparisons between the function of mammalian metallothionein and the coelomic fluid proteins of G. dibranchiata as a detoxification mechanism are discussed.  相似文献

The crab Carcinus maenas (L.) was exposed to radioactively labelled cadmium dissolved in seawater at concentrations of 0.1, 1 and 10 ppm, the latter concentration being toxic to the crabs (50% mortality after 12.3 days). Net accumulation of cadmium from solution was proportional to the level and time period of cadmium exposure. Total absorbed cadmium levels reached 0.0043 and 0.0412 mg Cd g-1 dry weight after 40 days exposure to 0.1 and 1 ppm Cd, respectively, and 0.1115 mg Cd g-1 dry weight after 12.3 days average exposure to 10 ppm Cd. The highest tissue concentration was found in the midgut gland, reaching 0.786 mg Cd g-1 dry weight after 12.3 days average exposure to 10 ppm Cd. The midgut gland only contained about 10% of the total cadmium absorbed from solution, while the exoskeleton contained the bulk of obsorbed cadmium (59 to 80%) probably passively adsorbed onto the surface. When cadmium was absorbed by the crabs from a food source, the midgut gland contained 16.9% of the total absorbed cadmium whereas the exoskeleton now contained only 22.2%. Ten percent of the cadmium available in the food source (Artemia salina) was accumulated by the crabs. When placed in cadmium-free seawater, crabs that had accumulated cadmium from solution lost 69% of the absorbed cadmium in 10 days, mostly from the exoskeleton which lost 78% of its original absorbed cadmium concentration.  相似文献

The burrowing marine shrimp Callianassa australiensis (Dana) was collected from an uncontaminated area in Western Port, Victoria, Australia in 1977. The shrimp were exposed to cadmium-contaminated water and sediment for 8 wk. The concentrations ranged from 0.5 to 63 g Cd 1-1 for water and 0.5 to 63 g Cd g-1 for sediment. The shrimp accumulated cadmium from water at a rate commensurate with increases in the concentration of cadmium in water and the duration of the experiment. Although the cadmium concentration in the sediments was 103 times higher than that in water, it hat no effect on cadmium uptake by the shrimp. The concentration factors decreased with increasing concentration of cadmium in water but increased as the duration of exposure increased. The shrimp dry weight decreased with increasing concentration of cadmium in water and duration of exposure. As was the case with cadmium uptake by the shrimp, these two factors acted interactively on the shrimp dry weight, but the third factor, concentration of cadmium in sediment, had no effect.  相似文献

The artificially high soil cadmium (Cd) concentration screening method was used to screen Cd-hyperaccumulators from floricultural plants. Among the five species of floricultural plants screened, Cosmos bipinnata showed the characteristics of Cd-accumulators. A pot experiment was conducted to further study Cd accumulation characteristics of C. bipinnata. The results showed that the biomass, chlorophyll content, superoxide dismutase activity, peroxidase activity and soluble protein content of C. bipinnata first increased and later decreased with the increase in soil Cd concentration, but the carotenoid content and catalase activity of C. bipinnata reduced. Cd contents in roots, stems, leaves and shoots of C. bipinnata increased with increasing soil Cd concentration. When the soil Cd concentration was 50?mg?kg?1, the Cd content in shoots was up to 112.62?mg?kg?1, which was higher than the Cd-hyperaccumulator critical value. The root and shoot bioconcentration factors exceeded 1 in various Cd treatments, but the translocation factors were less than 1. When the soil Cd concentration was 50?mg?kg?1, the Cd accumulation in shoots achieved the maximum of 224.30?μg plant?1. Therefore, considering the tolerance and accumulation of Cd, C. bipinnata is a Cd-accumulator that could be used to remediate Cd-contaminated urban soil.  相似文献

We investigated the effect of Cd with different concentrations in soil (control, 0.5, 1.0, 1.5, and 2.0 mg kg?1) on Salvia miltiorrhiza. The growth of S. miltiorrhiza was examined at 90 and 120 d, while active components were tested at 0, 30, 60, 90, and 120 d; Cd uptake was measured at days 0, 90, and 120. The biomass data indicated that high Cd concentration can stimulate the accumulation of biomass after a long treatment. The Cd content in the above- and below-ground parts strongly corresponds to the bioavailable Cd extracted by ethylene diamine tetraacetic acid (EDTA) and acetic acid. The Cd content in the below-ground parts of control approached the Cd standards given by World Health Organization after 120 d. The Cd content in the below-ground parts of S. miltiorrhiza at a Cd concentration of 0.5 mg kg?1 exceeded the Cd standards after being grown for 90 and 120 days. The accumulation of hydrosoluble components in the above-ground parts appeared later than that in the below-ground parts. With longer treatment time, high Cd treatment promoted an accumulation of hydrosoluble and liposoluble components. The Cd content in the below-ground plant parts indicated that there was still a risk of exceeding the Cd limit for S. miltiorrhiza grown in the soil contaminated by low-concentration Cd after a long time.  相似文献

As much as 89, 176 and 292 g Cd g-1 dry weight were accumulated by adult Crassostrea virginica after treatment for 40 wk with 5, 10 and 15 g Cd kg-1, respectively, in flowing seawater at ambient salinity and temperature without mortalities. Cadmium accumulation increased with increased concentration of cadmium in seawater; greater amounts were accumulated during the summer months. Uptake patterns measured as cadmium content were similar among the total soft parts, gill, mantle and visceral mass. A continuous increase of cadmium concentration in the visceral mass was observed. This differed from the uptake patterns observed as cadmium concentration in gill, mantle and total soft parts. Although cadmium accumulation in the total soft parts and the tissues was curvilinear over the entire study period, significant linear relations between cadmium concentration and time indicated a general increasing trend. At seawater temperatures below 6°C, when oysters were not actively feeding, cadmium concentrations in the total soft parts varied significantly between treatments, but not within treatments. In the tissues, the rate of uptake expressed as cadmium concentrations was visceral mass>gillmantle. Cadmium concentration in the total soft parts varied inversely with dry weight, whereas cadmium concentration in the total soft parts increased, whereas the content decreased. Cadmium concentration decreased in mantle and gill but increased in the visceral mass during spawning, whereas cadmium content decreased in all tissues. Regression analyses indicated that during spawning dry weight decreased at the same rate in gill and mantle, but they lost less weight and lost it more slowly than visceral mass. Also, during spawning, cadmium content decreased in mantle and gill at the same rate but more slowly than in the visceral mass. In mid-August, Cd concentration decreased despite the continuous addition of cadmium to the seawater; however, Cd content increased, suggesting that organism weight was responsible for fluctuations in cadmium concentrations.  相似文献

Effects of Cadmium on Nutrient Uptake and Translocation by Indian Mustard   总被引:1,自引:0,他引:1  
Plants that hyperaccumulate metals are ideal subjects for studying the mechanisms of metal and mineral nutrient uptake in the plant kingdom. Indian Mustard (Brassica juncea) has been shown to accumulate moderate levels of Cd, Pb, Cr, Ni, Zn, and Cu. In this experiment, 10 levels of Cd concentration treatments were imposed by adding 10–190 mg Cd kg–1 to the soils as cadmium nitrate [Cd(NO3)2]. The effect of Cd on phosphorus (P), potassium (K), calcium (Ca), magnesium (Mg), and the micronutrients iron (Fe), manganese (Mn), copper (Cu), and zinc (Zn) in B. juncea was studied. Plant growth was affected negatively by Cd, root biomass decreased significantly at 170 mg Cd kg–1 dry weight soils treatment. Cadmium accumulation both in shoots and roots increased with increasing soil Cd treatments. The highest concentration of Cd was up to 300 mg kg–1 d.w. in the roots and 160 mg kg–1 d.w. in the shoots. The nutrients mainly affected by Cd were P, K, Ca, Fe, and Zn in the roots, and P, K, Ca, and Cu in the shoots. K and P concentrations in roots increased significantly when Cd was added at 170 mg kg–1, and this was almost the same level at which root growth was inhibited. Zn concentrations in roots decreased significantly when added Cd concentration was increased from 50 to 110 mg kg–1, then remained constant with Cd treatments from 110 to 190 mg kg–1. However, Zn concentrations in the shoots seemed less affected by Cd. It is possible that Zn uptake was affected by the Cd but not the translocation of Zn within the plant. Ca and Mg accumulation in roots and shoots showed similar trends. This result indicates that Ca and Mg uptake is a non-specific process.  相似文献

The study deals with the toxicological impact of cadmium nanoparticles (Cd NPs) on Bacillus subtilis as a model Gram-positive bacterium. Cadmium oxide (CdO) NPs (~22 nm) and cadmium sulfide (CdS) NPs (~3 nm) were used in this study. Both the NPs were found to inhibit the cell viability of B. subtilis when added to the culture at mid-log phase, the viable cell number declined with increasing concentration of Cd NPs. At mid-log phase, 15 mg L?1 CdO NPs inhibited growth by ~50%, whereas at 30 mg L?1 growth completely ceased. Under the same conditions, CdS NPs inhibited growth by ~50% at a concentration of 8 mg L?1, and at 20 mg L?1 growth was completely retarded. The cells changed their morphological features to a filamentous form with increasing Cd NPs exposure time, leading to associated with clumping. NPs treated cells when stained with 4′, 6-diamino-2-phenylindole, showed filamentous multinucleated bead structure, suggesting irregularities in cell division. Increasing intracellular oxidative stress due to Cd NPs exposure might be one of the reasons for the cell morphological changes and toxicity in B. subtilis.  相似文献

Laboratory soft bottom microcosms, containing sediment with natural populations of meio-and microfauna and the amphipod Pontoporeia affinis Lindström, were used to investigate chronic effects of cadmium on a typical Baltic Sea benthic community. Temperature, salinity, organic content of the sediment and the life cycles of the amphipods in laboratory control microcosms were in good agreement with conditions in the Baltic Sea. In Experiment 1, P. affinis was exposed to four assayed cadmium concentrations, 0 (control), 6.3, 41 and 127 g Cd·1-1, for 265 d. At the lowest cadmium concentration (6.3 g Cd·1-1), the sediment contained about 700 times, and P. affinis about 1 700 times, the water concentration at the end of the experiments. In microcosms without P. affinis, the sediments accumulated less cadmium. Effects on P. affinis embryogenesis were detected after 105 d of exposure in all tested cadmium concentrations. Mortality of juveniles was significantly increased after 265 d of exposure in mean assayed concentrations of 41 and 127 g Cd·1-1. In Experiment 2, P. affinis was exposed to cadmium in assayed concentrations, 0, 6.5 and 46 g Cd·1-1 for 460 d. At the lowest water concentration (6.5 g Cd·1-1), the prolonged exposure did not increase the sediment concentration, which indicates a steady state, while concentrations in P. affinis reached about 3 500 times the lowest water concentration after 460 d. Juvenile P. affinis matured sexually and reproduced normally in all microcosms, but their offspring died in all microcosms with added Cd. This occurred at water concentrations not uncommon in polluted marine areas. The effects are serious in comparison to those found in conventional short-term laboratory experiments with the same concentrations and demonstrate the importance of long-term experiments in ecotoxicology.  相似文献

Assimilation efficiencies (AEs) and physiological turnover-rate constants (k) of six trace elements (Ag, Am, Cd, Co, Se, Zn) in four marine bivalves (Crassostrea virginica Gmelin, Macoma balthica Linnaeus, Mercenaria mercenaria Linnaeus, and Mytilus edulis Linnaeus) were measured in radiotracer-depuration experiments. Egestion rates of unassimilated elements were highest during the first 24 h of depuration and declined thereafter. Significant egestion of unassimilated Co, however, continued for up to 5 d in Macoma balthica, Mercenaria mercenaria and Mytilus edulis. With the exception of the extremely low values for 110 mAg, 109Cd, and 65Zn in C. virginica, physiological turnover-rate constants (k) showed no general pattern of variation among elements, bivalve species or food types, and were relatively invariant. Values from  ≤0.001 to 0.1 d−1 were observed, but excluding those for Co, most values were  ≤0.04 d−1. In all four species, the AEs of Ag, Am, and Co were generally lower than those of Cd, Se, and Zn. The AEs of Ag, Cd, Se, and Zn in these bivalves are directly related to the proportion of each element in the cytoplasmic fraction of ingested phytoplankton, indicating that >80% of elements in a prey alga's cytoplasm was assimilated. C. virginica, Macoma balthica, and Mercenaria mercenaria assimilated ∼36% of the Ag and Cd associated with the non-cytoplasmic (membrane/organelle) fraction of ingested cells in addition to the cytoplasmic fraction. The ratio of AE:k, which is proportional to the consumer–prey trace-element bioaccumulation factor (concentration in consumer:concentration in prey) was generally greater for Cd, Se, and Zn than for Ag, Am, and Co. This ratio was lowest in Mytilus edulis, suggesting that this bivalve, the most widely employed organism in global biomonitoring, is relatively inefficient at accumulating important elements such as Ag, Cd, and Zn from ingested phytoplankton. Metallothionein (MT) has a great capacity of binding metal ions showing an interesting connection with metal toxicology, as a biochemical marker for environmental metal pollution.

To normal male Wistar rats (200±10 g) and other groups with ferropenic anemia, are administered 1 mg Cd/Kg/day, during 6 days, and MT labelled with the administration 2 h before sacrifice of 3 μCi 109CdCl2, also through intragastric catheter. The MT concentration in the intestinal mucosa is expressed in μg MT/g fresh tissue, being for control rats 1.4 ± 0.5; for rats administered with 6 doses of Cd 2.5 ± 0.6 (P<0.05); with ferropenic anemia 4.3±0.7 (P<0.001), and for anemic rats treated with 6 doses Cd 12 + 0,3 (P<0.001) μg MT/g fresh tissue respectively. PAGE 15% T, 2% CBis show for intestinal mucosa 2 MT peaks and for brain 3 MT peaks. Anemia induce MT accumulation and increases cadmium incorporation, being anemic subjects eligibles to be submitted first to control and detoxication than the rest of the population, and also MT should be studied as biochemical marker of the pollution.  相似文献

Metallothionein induction inMytilus edulis exposed to cadmium   总被引:1,自引:0,他引:1  
The exposure of mussels,Mytilus edulis, collected from Whitsand Bay, southwest England, in August 1988, to sublethal concentrations of cadmium (400µg l–1) for 65 d resulted in the induction of metallothionein (MT) synthesis in the soft tissues. In cadmium-exposed mussels, metallothionein concentrations, measured by differential pulse polarography, increased by a factor of three, from 2 to 3 mg g–1 to a maximum of 9 mg g–1 after 30 d. No significant changes could be detected in controls. Cadmium accumulated in the soft tissues of mussels correlated significantly with metallothionein concentrations and can be described by the relationship: MT (mg g–1)=0.045 Cd (µg g–1)+3.03 (r=0.803,P<0.001). Gel chromatography of heat-treated cytosolic extracts showed that the accumulated cadmium is bound principally to the newly formed metallothioneins. Copper and zinc were also analysed in the whole soft-tissues and in subcellular fractions of cadmium-exposed mussels. Although copper concentrations were not affected by cadmium-exposure, zinc levels were significantly reduced. The results demonstrate that the induction of metallothioneins inM. edulis is a quantifiable biological response to sublethal levels of cadmium exposure.  相似文献

The uptake of cadmium by the shoots ofZostera marina L. (eelgrass) was examined in the laboratory. Experiments were carried out in experimental chambers which allowed the separation of the leaves from the root-rhizome portions of intact shoots. Cadmium uptake by the root-rhizome portions over 24 h was directly related to substrate cadmium concentration (1.0, 5.0 and 10.0 g Cd ml-1) and varied from 6.5 to 30.0 g Cd g-1. Cadmium uptake by the root-rhizomes and the leaves in a substrate concentration of 1 g Cd ml-1 was also related to exposure times (24, 48 and 72 h). Maximum uptake by the root-rhizomes and the leaves was observed after 72 h at a substrate concentration of 1 g Cd ml-1 and was equivalent to 48 and 94 g Cd g-1, respectively. Translocation of cadmium from the leaves to the root-rhizomes was observed after 24 h, and at the end of 72 h was equivalent to 27% of the total leaf uptake. No cadmium movement from the root-rhizomes to the leaves was detected.  相似文献

通过温室盆栽实验,研究了镉在北京褐潮土中对玉米(品种郑单958)幼苗的毒性效应及其生物富集特性,并通过聚合酶链式反应-变性梯度凝胶电泳(PCR-DGGE)技术,探讨了镉对玉米根际微生物群落结构的影响。结果表明,高浓度(>100mg·kg-1)镉对玉米幼苗的生长发育有明显的抑制作用,北京褐潮土中镉引起玉米幼苗株高下降1/2的效应浓度(EC50)为654.6mg·kg-1,引起玉米根部和地上部干质量下降1/2的EC50分别为323.6和110.2mg·kg-1,玉米幼苗地上部生物量(干质量)可作为评价重金属生态毒性的适宜终点。玉米幼苗对镉有一定的吸收累积效应,镉在玉米幼苗各组织中的浓度分布为根>茎>叶,其中根部对镉有一定的富集作用(生物富集系数BAF>1)。镉污染可引起玉米根际微生物群落结构发生改变,高浓度(1000mg·kg-1)镉可导致部分微生物种群数量减少甚至完全消失,表明镉污染可对植物幼苗、植物根际微生物以及植物-微生物之间的相互作用造成重要的干扰和威胁。  相似文献

Accumulation and binding of cadmium in the tissues of the shore crabCarcinus maenas (L.) were investigated in a series of laboratory experiments. The relation between the physiological condition of individual crabs and their ability to transport cadmium from haemolymph to hepatopancreas was specifically addressed. Cadmium was removed from the haemolymph with a half-life of approximately 10 h, and half of the cadmium removed from the haemolymph was taken up in the hepatopancreas. The efficiency with which individual crabs transported cadmium from the haemolymph to the hepatopancreas was strongly related to physiological parameters, such as concentrations of calcium and magnesium in the hepatopancreas and haemolymph volume and haemolymph protein concentration. The transport of cadmium from the haemolymph to the hepatopancreas was saturated in crabs exposed to more than 2 to 4 mg Cd l–1 in the sea water. In the hepatopancreas of unexposed crabs, cadmium was bound mainly in the insoluble tissue fraction (40%) and in the protein fraction with a molecular weight of approximately 6000 D (50%). Exposure to 0.25 to 1.5 mg Cd l–1 for 2 w led to dose-dependent increases of the amounts of cadmium bound in the high-molecular weight protein fraction and in the insoluble tissue fraction. Modes of internal cadmium transport and accumulation in tissues and variability in physiological parameters are discussed.  相似文献

Pot experiments were conducted on cole (Brassica) grown in soils jointly treated with traces of two heavy metals cadmium (Cd) and zinc (Zn). As the concentration of heavy metals in the soil increased, the uptake of these metals by the plants rose. However, the ratio of heavy metal concentration in soil to uptake by plants increased at a slower rate. Bioavailability of heavy metals considered between the roots and soil using non-linear regressions was shown to be statistically significant. Similarly, the bioavailability of these two heavy metals between leaves and roots using a linear regression was also statistically significant. The bioconcentration factors (BCFs) for Cd and Zn were 0.282 and 4.289, respectively. Significant variation of BCF with the heavy metal bioavailability in soil was noted from non-linear models. The transfer factors (TFs) were 4.49 for Cd and 1.39 for Zn. The Zn concentration in leaves under all treatments did not exceed threshold set standards, but Cd levels exceeded these standards when the concentration of Cd in the soil was more than 1.92 mg kg?1 dry weight (dw). Data indicate that cole (Brassica) is not a suitable crop for oasis soils because of plant contamination with heavy metals, especially Cd.  相似文献

Recently, although dynamic approaches have been used increasingly to describe metal bioaccumulation in aquatic organisms, the validation of such laboratory-derived modelling is rarely assessed under environmental conditions. The present study aims to evaluate and verify statistical and mathematical models for the bioaccumulation of cadmium (Cd) and to evaluate the suitability of Gammarus pulex as a biomonitor for Cd. To optimise Cd2+ ion bioaccumulation conditions a Box–Behnken design was used and whether G. pulex can be used as an estimated tool to assess aquatic quality was investigated. A Box–Behnken experimental design combined with response surface modelling (RSM) was employed for Cd2+ bioaccumulation in G. pulex. Independent variables including temperature, contact time and Cd2+ ion concentration significance and their interactions were tested using analysis of variance. The selected variables’ optimum values were obtained by solving the quadratic regression model along with by analysing the response surface contour plots. Based on Box–Behnken design, the applied model is significant because model F-value and R2 were obtained as 5121.49, 0.9998, respectively. Bioaccumulation of Cd amount was analysed using electrothermal atomic absorption spectrophotometer. The obtained data suggest that G. pulex can be evaluated as Cd biomonitor.  相似文献

Ulmus tree leaves were successfully used as a novel and efficient biosorbent for removing cadmium, (Cd(II)), from aqueous solutions in a batch system. A multivariate strategy for optimization of removal efficiency conditions of Cd(II) was carried out. A 23 full factorial design with three center points (9 runs) was performed for screening the main variables and reducing the large number of experimental runs. Initial concentration of metal ion (C m), amount of sorbent (m), and pH were considered as the three main variables at two different levels. The maximum removal efficiency of Cd(II) was achieved within 1 h contact time. It was found that all the main factors and their interactions were significant at p < 0.05. Doehlert response surface methodology was utilized (13 runs) for finding a suitable mathematical model. The analysis of variance and some statistical tests such as lack-of-fit, coefficient of determination (R 2), and residual distribution plot confirmed the validity of the model. The optimum conditions for maximum removal of Cd(II) by Ulmus tree leaves were found as pH = 3.4, m (amount of sorbent) = 0.128 g, C m (initial concentration of metal ion) = 12.1 mg L?1.  相似文献

The uptake of cadmium by the Haptophyceae Hymenomonas elongata was studied as regards the energetic processes and Ca‐transport. For this purpose, experiments were carried out in the presence of an uncoupler of the phosphorylation : CCCP and of an inhibitor of calcium channels and also of Ca2+‐ATPase : verapamil. To reduce the number of assays, a factorial experimental design was used, in which all variables:— concentration of Cd in the medium—incubation time and—the presence of the inhibitor, are at two levels. In the absence of inhibitor, Cd uptake was found to vary as a function of metal concentration in the medium and of incubation time. CCCP significantly increased Cd uptake by H. elongata at sufficient incubation time. Therefore Cd uptake seemed linked to an energy‐dependent process, involving an ATPase. Verapamil immediately increased Cd uptake, implying an interaction between Cd and Ca.  相似文献
