复合垂直流人工湿地基质氧化还原酶活性研究   总被引:1,自引:0,他引:1  
对复合垂直流人工湿地系统(IVCW)基质中5种常见的氧化还原酶活性进行了研究.结果表明,氧化还原酶活性存在显著性季节差异,过氧化物酶在春、夏、秋三季的酶活性显著高于冬季的酶活性(P<0.05);多酚氧化酶和过氧化氢酶的活性最高在秋季;脱氢酶在夏、冬两季时酶活性显著高于秋季和春季的酶活性(P<0.05);硝酸盐还原酶在春、冬季时显著高于夏、秋季(P<0.01).空间分布上,除过氧化物酶外,湿地下行流池的酶活性显著高于上行流池,并且随着基质深度的增加递减;但基质硝酸盐还原酶各层之间差异不显著.图6表3参17  相似文献

选择攀枝花苏铁林下计划烧除试验后苏铁及其根系周围土壤(0~15 cm)为对象,研究火烧对攀枝花苏铁植株生长、叶片生理和苏铁根系周围土壤理化性质的影响,以期为区域攀枝花苏铁种群恢复和生态环境保护提供理论依据.结果表明:计划烧除区域苏铁成年树的株数及株高没有显著变化,新生叶片数目和幼苗株数相对未烧除区域显著增加,增幅分别达201.66%和317.7%;烧除区域苏铁植株新生叶片叶绿素a、叶绿素b、叶绿素a+b,可溶性糖、蛋白质含量,硝酸还原酶(NR)、谷氨酰胺合成酶(GS)活性,叶片碳(C)、氮(N)、磷(P)、钾(K)含量,C/N、N/P比与对照区域新生叶片相比无显著性差异,只有类胡萝卜素含量显著降低.计划烧除改变了苏铁根围土壤(0~15 cm)的理化性质,表现在烧除后的土壤含水量、pH值及总N、P、K含量显著降低;但是,火烧显著提高了土壤有机碳(TOC)、硝态氮(NO3--N)、铵态氮(NH4+-N)含量,微生物量C、N含量也略有升高;这说明火烧有利于增加苏铁林下土壤养分(N)的有效性,为烧除后苏铁快速恢复生长提供有利条件.  相似文献

A survey of the distribution of six enzymes of nitrogen and carbohydrate metabolism was carried out with eleven species of marine phytoplankton representing seven classes. The activities of some enzymes, e.g. NADPH-glutamate dchydrogenase, were absent in several organisms. The effects of nitrogen deficiency on enzyme activities in nitrogen-limited chemostat cultures ofChlorella stigmatophora were investigated. Increasing nitrogen deficiency was accompanied by increases in the activity of nitrate reductase, NADPH-glutamate dehydrogenase and glutamine synthetase, and decreases in photosynthesis, respiration, chlorophyll and total cell protein. In nitrogen-deficient algae, an increase in the activity of the nitrogen-assimilating enzymes may facilitate the efficient utilization of available nitrogen under these conditions.Present address: School of Biological Sciences, University of Nebraska-Lincoln, Lincoln, Nebraska 68588-0118, USA  相似文献

Influent and final effluent was collected from the CMM Ltd., Bethora, Ponda, Goa and were analysed for pH, DO, BOD, enzyme activity and chlorophyll content of the waste stabilization pond for over a period of two years of which the data for one year (pre monsoon, monsoon and post monsoon periods) is given. The study revealed that the DO was maximum during the pre-monsoon months and least during the monsoon. Maximum removal of BOD and phosphate was observed during the pre-monsoon periods. Enzymatic activity was at its peak during the monsoons than during the other months. Chlorophyll content was maximum during the pre-monsoon months due to increased growth of phytoplankton as the conditions were favourable for their growth. Also depending on the concentration of different chlorophyll pigments, one can come to know the different groups of algae inhabiting the stabilization ponds.  相似文献

In the present study species like Mangifera indica, Linn., Cassia fistula, Linn., and Eucalyptus hybrid were exposed to different air pollution load for short duration (active biomonitoring). Variation in biochemical parameters like chlorophyll, protein, soluble sugar free amino acid, ascorbic acid, nitrate reductase, superoxide dismutase and peroxidase in the leaves were found to be pollution load dependent. These variations can be used as indicators of air pollution for early diagnosis of stress or as a marker for physiological damage to trees prior to the onset of visible injury symptoms. Just by analyzing these biochemical indicators air quality can also be assessed.  相似文献

Epigeic earthworms (E. fetida) were cultured on variety of organic wastes amended with cattle manure to determine the influence of diets and the seasonal environmental factors on growth and reproduction. The results showed that growth and reproductive strategies of E. fetida varied with different diets and seasons. Growth and reproduction of worms in all wastes were significantly more in winter and monsoon than in summer season. Hence winter and monsoon seasons could be considered congenial for vermiculture. During all seasons, worm activities were more in cattle manure followed by amended Bengal gram grain husk and Mixed Organic waste by E. fetida. Parthenin containing diet had deleterious effects on cocoon production.  相似文献

Porphyra perforata J. Ag. was collected from a rocky land-fill site near Kitsilano Beach, Vancouver, British Columbia, Canada and was grown for 4 d in media with one of the following forms of inorganic nitrogen: NO 3 - , NH 4 + and NO 3 - plus NH 4 + and for 10 d in nitrogen-free media. Internal nitrogen accumulation (nitrate, ammonium, amino acids and soluble protein), nitrate and ammonium uptake rates, and nitrate reductase activity were measured daily. Short initial periods (10 to 20 min) of rapid ammonium uptake were common in nitrogen-deficient plants. In the case of nitrate uptake, initial uptake rates were low, increasing after 10 to 20 min. Ammonium inhibited nitrate uptake for only the first 10 to 20 min and then nitrate uptake rates were independent of ammonium concentration. Nitrogen starvation for 8 d overcame this initial suppression of nitrate uptake by ammonium. Nitrogen starvation also resulted in a decrease in soluble internal nitrate content and a transient increase in nitrate reductase activity. Little or no decrease was observed in internal ammonium, total amino acids and soluble protein. The cultures grown on nitrate only, maintained high ammonium uptake rates also. The rate of nitrate reduction may have limited the supply of nitrogen available for further assimilation. Internal nitrate concentrations were inversely correlated with nitrate uptake rates. Except for ammonium-grown cultures, internal total amino acids and soluble protein showed no correlation with uptake rates. Both internal pool concentrations and enzyme activities are required to interpret changes in uptake rate during growth.  相似文献

The effect of mercury (Hg) on the biochemical parameters of Lycopersicon esculentum Mill leaf was studied. Application of mercuric chloride in varying concentrations (0.5, 1.0, 1.5 and 2.0 mM HgCl2 kg(-1) sand) caused significant reduction that went up to 89% and 72% chlorophyll a and chlorophyll b contents respectively (at flowering stage), 69% in carotenoid content, 64% in total soluble protein content and 91% in nitrate reductase activity (all at post-flowering stage). The amounts of nitrate and proline increased maximally (151% and 143% respectively) at the flowering stage, whereas total soluble sugar enhanced by 57% at the post-flowering stage. Changes observed in most of the parameters, were concentration dependent. Such studies seem to be able to discover suitable bioindicators of heavy metal pollution.  相似文献

选取2种入侵植物(一年蓬Annual Fleabane及加拿大蓬Erigeron Canadensis)及本土植物(艾蒿Artemisia argyi)的根际土壤微生物种群为研究对象,以分析不同根际土壤微生物种群的数量及测定根际土壤微生物酶活的活性为切入点来探析入侵植物对土壤微生物的影响及其响应机制。结果显示:2种入侵植物显著增加了其根际土壤中的细菌的数量,而显著抑制了真菌与放线菌的数量。另外,入侵植物一年蓬显著抑制其根际土壤中纤维素酶的活性,而3种植物的根际土壤硝酸还原酶活性无显著差异。入侵植物对其他3种土壤酶(即转化酶、脲酶及酸性磷酸酶)活性的影响却呈现出截然相反的影响,即一年蓬显著增加了3种根际土壤酶的活性,而加拿大蓬却显著减少了3种根际土壤酶的活性。导致这种现象的原因可能是不同入侵植物的根系释放不同的化学物质进而对土壤微生物的数量和活性造成不同的影响。  相似文献

The aim of this study was to evaluate the effects of copper (Cu) stress on accumulation and transport of trace elements, nitrogen assimilation, and growth parameters of Verbascum olympicum. Eight-week-old seedlings were grown in Hoagland's solution and exposed to 0, 50, 250, or 500?µM CuSO4 for seven days in laboratory conditions. Bioaccumulation of trace elements (boron, bismuth, cobalt, Cu, iron, lithium, manganese, molybdenum, nickel, lead, zinc) in the roots and leaves was determined by inductively coupled plasma–mass spectrometry after one, three, and seven days. Chlorophyll content, nitrate reductase, and glutamine synthetase activities, soluble protein content, and biomass were determined. Copper accumulated in the roots and leaves (up to 19609.8 and 256.2?mg?kg?1 dry weight, respectively). Other trace elements accumulated to higher levels in the roots of Cu-treated plants compared with those of control plants. High Cu concentrations decreased nitrogen-assimilatory enzyme activities. Compared with control plants, those treated with high Cu concentrations showed lower chlorophyll contents, total protein contents, and biomass. Nitrogen assimilation and growth parameters of V. olympicum were negatively affected by Cu treatment but mineral nutrition was not severely disrupted. The results support the suitability of V. olympicum as a candidate for phytoremediation of Cu-contaminated soils.  相似文献

The southwest monsoon on the west coast of India brings about dynamic changes in estuaries and coastal waters. The response of the meiofauna to monsoonal rain is obvious, but the impact of such environmental changes on the community structure of harpacticoid copepod species and their seasonality in the estuaries influenced by the tropical monsoon is poorly understood. In this study the spatial and temporal variability in abundance and community structure of meiobenthic copepods was investigated over an annual cycle (June 1983 to June 1984), in an estuary influenced by the tropical monsoon. Total meiofaunal abundance showed wide variations in space and time. Minimum and maximum densities were observed in the monsoon and pre-monsoon seasons, respectively. Quick recovery of harpacticoid populations in the early post-monsoon season indicated their recuperative power under adverse conditions. Of the 25 species recorded from lower, middle and upper reaches, eight comprised over 70% of the total copepod population. The peak and low occurrence of dominant species displayed striking correlations with the summer and rainy seasons, respectively. While other species were restricted in distribution, Stenhelia longifurca was recorded from all salinity regimes. The harpacticoid community was greatly influenced by the onset of the monsoon period, and their spatial and temporal variabilities were related with physico-chemical parameters and the variability of these parameters in the estuary.  相似文献

The potential activity of nitrate reductase was determined in uni-algal cultures in the laboratory and in natural marine phytoplankton assemblages. In the laboratory bioassays, distinct differences in nitrate reductase activity were observed in iron replete versus depleted cultures for Emiliania huxleyi, Isochrysis galbana and Tetraselmis sp. Cells from iron-depleted cultures had 15 to 50 percent lower enzyme activity than those from iron-replete cultures. Upon addition of iron, nitrate reductase activity was enhanced in depleted cells up to levels comparable to those of the replete cells. Bioassays in the northern North Sea conducted in 1993, under low iron conditions, demonstrated similar results. Upon addition of 2.5 nM iron, a distinct enhancement, to a maximum of three times, of nitrate reductase activity was observed within 32 h after addition. Therefore, iron can stimulate nitrate reductase activity. In spite of the clean techniques used, some nitrate reductase activity was always observed. Iron deficiency was shown to impair nitrate reductase activity, but it is unlikely that nitrate reduction would cease completely.  相似文献

Nitrogen uptake, assimilation and accumulation were studied in three populations of Gracilaria pacifica Abbott in Bamfield Inlet, British Columbia, Canada, over three summers, 1979–1981. Two of these populations were in the intertidal one high and one low, and the third was a subtidal cultured population. Nitrate uptake rates, internal nitrate content and nitrate reductase activities were highest in the low intertidal population. Time-courses of uptake and uptake kinetics were studied. Both nitrate and ammonium were taken up simultaneously. Thalli from the high-intertidal population showed enhanced nitrate and ammonium uptake following mild desiccation, and greater tolerance to desiccation in terms of maintaining nitrogen uptake after severe desiccation. Transplants were made to determine the effect of intertidal height and geographic location on responses to desiccation, nitrogen uptake, assimilation and accumulation. Nitrate and ammonium uptake rates were dependent on intertidal height and geographic location. Transplanting up the intertidal increased nitrate uptake and nitrate reductase activity, but decreased the nitrate content of the thalli. There were few significant differences in ammonium uptake rates, and ammonium, amino acid, and soluble-protein content of the various populations. All high-intertidal populations, transplanted or natural, showed enhanced nitrate uptake rates following desiccation. Enhanced ammonium uptake rates following desiccation were restricted to the high-intertidal thalli in only one geographic location. Tolerance to higher levels of desiccation also appeared to be intertidal height-dependent, but required more than five weeks to fully develop or disappear.  相似文献

Back in 1992, the Gulf of Aqaba Environmental Action Plan (GAEAP), a collaboration between the Aqaba Region Authority (ARA), Jordan and the World Bank, gave considerable emphasis to the environmental protection of the Gulf of Aqaba [The World Bank. Gulf of Aqaba Environmental Action Plan. Report No. 12244JO (1993).]. The document recommended the establishment of a marine reserve and the long term monitoring of the coastal habitats' environmental quality. The combination of a dedicated follow up, the collaborative efforts of ARA and the Marine Science Station (MSS), and the founding of the Aqaba Special Economic Zone Authority (ASEZA) have turned the recommendations into reality. A comprehensive monitoring program of the Jordanian coastal habitats commenced in 1999. The first three years of the program were financed by a donation from The Global Environmental Facility (GEF). In return, Jordan has committed itself to the maintenance of the monitoring program as an ongoing tool for sustainable coastal management. The monitoring program includes observations on benthic habitat, fish communities, bottom sediments and seawater quality. This paper focuses on the results of seawater-quality monitoring in the first three years. Records of weather conditions, coastal currents, seawater temperature, transparency, salinity, density, pH, alkalinity, dissolved oxygen, ammonia, nitrate, nitrite, phosphate, silicate, particulate matter, chlorophyll a, zooplankton biomass, total coliform, fecal coliform, hydrocarbons and sedimentation rate have been generated monthly since January 1999 at six coastal stations, and one offshore reference station, in the Jordanian waters of the Gulf of Aqaba. The coastal stations are located at sites with different benthic habitats and are occupied by different human activities. Offshore records of density (thermohaline structure), nutrients and chlorophyll a depicted two well-defined seasons; a nutrient-/chlorophyll a-rich, mixed water winter from December to April and a nutrient-/chlorophyll a-poor, stratified water summer from June to October. Short transition seasons appeared in May and November. The mixing and stratification seasons were also clearly depicted in the coastal waters. Statistical analysis of the three-year data collected at the offshore station revealed no significant inter-annual differences in the upper 125 m of the water column with respect to any of the measured parameters. At coastal stations, the water quality at the two northernmost stations was significantly different in comparison to the upper 125 m at the offshore station and to the other coastal stations, with respect to the two key indicator parameters: inorganic nitrogen and chlorophyll a. The three-year findings of the monitoring program are employed to suggest standard codes of reference for the coastal water quality.  相似文献

Denaturing gradient gel electrophoresis (DGGE) was used to elucidate spatial and temporal variations in bacterial community composition (BCC) from four locations along the central west coast of India. DNA extracts from 36 water samples collected from surface, mid-depth (-10 m) and dose to bottom (-20 m) during premonsoon, postmonsoon, monsoon were analyzed by PCRfor amplifying variable region of 16S rRNAgene and subsequently through DGGE. Prominent bands were excised, cloned and sequenced indicated the preponderance of gammaproteobacteria, bacteroidetes and cyanobacteria. Non-metric dimensional scaling of the DGGE gels indicated that the spatial variations in BCC were prominent among the sampling locations. Temporal variations in the BCC appear to be influenced by monsoonal processes. The canonical correspondence analyses suggest that the concentration of chlorophyll a and nitrate are two important environmental factors for both spatial and temporal variations in BCC. Chlorophyll a seems to be impart a top-down control of BCC while nitrate, the bottom-up control. Our results also suggest that BCC can vary over a small geographic distance in highly dynamic, seasonally predisposed tropical coastal waters.  相似文献

采用大田小区试验,通过新型植物生长调节剂处理春玉米,研究了生长调节剂对玉米氮素代谢关键酶活性的影响,结果表明,喷施不同配方的生长调节剂能明显提高春玉米氮代谢过程中关键酶的活性。在生长调节剂PGR2处理下,玉米功能叶片中硝酸还原酶(NR)活性比对照提高了109.3%~138.3%,其它处理下硝酸还原酶活性也有不同程度的提高;无论在玉米功能叶片还是籽粒中,谷氨酰胺合成酶(GS)和谷氨酰胺转化酶活性在测定的各时期均高于对照;功能叶片中蛋白水解酶活性也均高于对照。在所有处理中,以PGR2处理效果最佳。  相似文献

Growth experiments in batch cultures indicated that the uptake of nitrate by the marine pseudomonad PL1 was inhibited in the presence of ammonia provided that the ammonia concentration was higher than 1 mM. At ammonia concentrations of less than about 1 mM, however, both nitrate and ammonia were utilised simultaneously. The saturation constants for nitrate and ammonia uptake were both 2.6x10-4 M, and similar to the Michaelis constants of nitrate reductase for nitrate (2.9x10-4 M) and glutamine synthetase for ammonia (2x10-4 M). Nitrate reductase activity linked to NADH was detected in chemostat-grown cultures with nitrate as nitrogen source, and in cultures containing limiting concentrations of nitrate and ammonia, ammonia or glutamate. Enzyme synthesis appeared to be repressed in cultures containing an excess of ammonia or glutamate. Chemostat cultures utilised ammonia or glutamate in preference to nitrate, while there was no marked preference between ammonia and glutamate.  相似文献
