Red morwong (Cheilodactylus fuscus; Cheilodactylidae: Perciformes), are large temperate rocky reef fish that are patchily distributed in local aggregations of 3 to >100 fish. The home ranges and aggregating behaviour of red morwong were investigated using external tags (n = 114, over 35 mo) and ultrasonic transmitters (n = 9 over 20 d) at coastal and estuarine sites in south-east Australia. Adult red morwong had a mean home range during the day of 1865 m2 (SE = 268). Night movement, determined by ultrasonic telemetry, indicated a significantly greater mean home range of 3639 m2 (SE = 416). Home range did not differ significantly with size or sex. Fish movements were greatest and aggregation sizes varied most during the non-reproductive mid-summer period. Tagged fish (N = 20) displaced 200 to 900 m returned in 1 to 3 d to the point of capture, often traversing open sand habitat and other aggregations. During the day, fish were commonly found on bouldered habitat. During the night, these site-specific aggregations fragmented as fish dispersed over a variety of substrata, with crepuscular peaks in activity. Investigations of factors affecting feeding success in fish larvae require knowledge of the scales of variability of the feeding process itself and the indices used to assess this variability. In this study, we measured short-term (diel) variability in feeding rates of wild haddock (Melanogrammus aeglifinus) larvae four times per day during a 10-d cruise in the northern North Sea. Feeding activity was evaluated using indices of gut fullness, prey digestive state and biochemical measurements (tryptic enzyme activity). The gut fullness and the enzyme activity indices indicated moderate to high rates of food consumption throughout the cruise. Time series analysis of the three indices showed significant diel variability in all indices and enabled identification of significant lags between food uptake and peak digestive enzyme activity. The typical pattern of food consumption and digestion was characterized by maximal ingestion of prey early in the evening (19:00 hrs) and peak digestive enzyme activity at 01:00  hrs. The time scale over which enzyme activities reacted to prey ingestion was ca. 6 h, and is consistent with expectations from controlled laboratory experiments with other larval fish species. Significant diel variability in tryptic enzyme activity suggests that attempts to relate this measure of feeding success to other variables (e.g. food concentrations) should take care to accommodate natural cycles in feeding activity before making statistical comparisons. Phenoloxidase (PO) from the colonial ascidian Botryllus schlosseri was purified using two different chromatographic strategies. A three-step purification was developed in order to maintain enzyme activity, whereas an easier purification procedure was adopted to obtain enough PO for the production of specific polyclonal antibodies. The enzyme showed optimal pH and temperature values of 7.0 to 7.5 and 35 °C, respectively, and a K m value of 4.62 ± 0.76 mM was estimated using l-DOPA as substrate. A molecular weight of 160 kDa was determined after SDS-PAGE under non-reducing conditions. The addition of the reducing agent β-mercaptoethanol caused the disappearance of the 160 kDa band and the appearance of a new band at 80 kDa, suggesting that active PO is a dimer and the two subunits are linked by disulphide bridges. Parma microlepis (Günther) were collected from Malabar, an urban location close to the centre of Sydney, Australia, and from Jervis Bay, a reference location 170 km south of the city centre. At each location, fish were collected from two sites separated by 100 to 200 m. The ultrastructure of normal liver tissue is described based on 20 female fish collected from Jervis Bay, where fish are known to be exposed to low levels of organochlorine contaminants. Alterations in the endoplasmic reticulum, mitochondria, lysosomes and nuclei of hepatocytes were identified and quantified in the liver tissue of fish from this location and compared to alterations in 20 female fish collected from Malabar, where fish are exposed to higher concentrations of organochlorine pesticides such as DDT compounds. There were significant differences in the percentage of hepatocytes with swollen mitochondria (F = 124.025, df = 2, 2, P = 0.008) and atypical nuclei (F = 22.198, df = 2, 2, P = 0.043) between sites (100 to 200 m apart), but there were no clear differences between the percentage of structural alterations in the hepatocytes of P. microlepis from Jervis Bay and Malabar. Associations between liver morphology and the organochlorines aldrin, dieldrin, DDE and chlordane were examined using a Pearson correlation matrix. Significant correlations were detected between the percentage of hepatocytes with dilated endoplasmic reticulum and the concentrations of the pesticide aldrin (r = 0.600, df = 11, r crit(α = 0.05) = 0.553). Significant associations were also detected between the percentage of hepatocytes with disorganised endoplasmic reticulum and the concentrations of dieldrin and DDE residues in fish (r = 0.576, r = 0.567, respectively, df = 13, r crit (α = 0.05) = 0.514). However, there was little evidence that ultrastructural alterations in fish responded to increasing concentrations of these pesticides in a consistent dose-response manner.  Antarctic krill Euphausia superba has a central role in the ecosystem of the Southern Ocean and knowledge of its growth rate is central to determining the factors influencing population dynamics. The length of Antarctic krill in the diet of Antarctic fur seals Arctocephalus gazella at South Georgia revealed a consistent increase in size between ca. 42 and ca. 54 mm over the period October–March, indicating growth rates much higher than predicted by existing models. Geographical variation in growth rate may result in 2-year-old krill at South Georgia attaining the same size as 3-year-old krill in the Antarctic Peninsula region. The effect of geographical variation in growth rate on the population structure of krill has important implications for comparing the fate of individual cohorts over large scales and in the interpretation of krill life-cycles. The aim of this study was to determine whether the composition of the demersal fish fauna in coastal marine waters in temperate Australia changes markedly with increasing water depth and distance from the shore and whether the composition of the fish fauna in water depths of 5 to 35 m undergoes cyclic, seasonal changes. Samples of demersal fishes were therefore collected by trawling over the predominantly sandy substrate at nine sites located in water depths of 5 to 15 m or 20 to 35 m and within 20 km of the shore in four regions along ∼200 km on the lower west coast of Australia. The sampling regime involved trawling for fishes at each site at night in seven consecutive seasons between the summer of 1990/1991 and winter of 1992. A total of 72 435 fishes, representing 77 families, 143 genera and 172 species was caught. The compositions of the fish faunas in offshore waters with depths of 5 to 35 m were shown to differ markedly from those previously recorded for nearshore marine waters in the same regions. However, as some species, such as Sillago burrus, S. vittata, S. bassensis and Rhabdosargus sarba, increase in size, they move out from their nursery areas in nearshore waters into deeper and more offshore waters, where spawning occurs. Ordination showed that, in each of the four regions, the composition of the fish fauna in depths of 5 to 15 m differs from that in depths of 20 to 35 m. This difference is attributable to the fact that some species, such as  S. burrus, S. vittata and Upeneichthys lineatus, are far more abundant in depths of 5 to 15 m, whereas other species, such as S. robusta, U. stotti and Lepidotrigla modesta, occur predominantly in depths of 20 to 35 m. However, the samples collected from the single site that was inshore but in deeper water demonstrate that the composition of the fish fauna is influenced by distance from shore as well as by water depth. The compositions of the fish faunas differed with latitude, largely due to the fact that some subtropical species, such as Polyspina piosae, S. burrus and  S. robusta, did not extend down into the more southern regions. Ordination also showed that the composition of the fish faunas at all but one of the nine sites underwent pronounced and consistent cyclic, seasonal changes. This seasonal cyclicity at the different sites was attributable to sequential patterns of immigrations and emigrations by a number of fish species during the course of the year. These seasonal migrations involved, inter alia (1) movements of certain species from their nursery areas into these deeper waters, e.g.  S. bassensis and Scobinichthys granulatus; (2) migrations into and off the sandy areas of the inner continental shelf, e.g. Arnoglossus muelleri; (3) migrations to spawning areas, e.g. Sillago robusta; and (4) movements into areas where detached macrophytes accumulate in winter, e.g. Cnidoglanis macrocephalus and Apogon rueppellii. An allozyme survey, using starch-gel electrophoresis, was carried out on eight populations of the Antarctic nemertean worm Parborlasia corrugatus (McIntosh, 1876) collected from locations around the South Orkney Islands, Antarctica. These populations were separated by distances in the order of tens of kilometres. Genetic variation was estimated over 22 enzyme loci for all populations examined, giving an observed heterozygosity of 0.142. This was much lower than the expected heterozygosity (H e  = 0.201), and it was found that there was a significant deficiency of het‐erozygotes across four enzyme loci ( p ≤ 0.01). A more detailed examination of this deficiency of heterozygotes was undertaken for the six populations and six variable enzyme loci for which the most complete data sets existed. A significant deficiency of heterozygotes was found at the enzyme locus Odh-1 for four of the six populations examined ( p ≤ 0.01). Mean F is (0.240) indicated a significant ( p ≤ 0.01) within-population component of the heterozygote deficiency estimated for the six populations sampled, and this was mainly due to the␣Ap-1, Odh-1 and Pgm-1 loci. The mean F st value (0.036) was also significant ( p ≤ 0.01), indicating a degree of genetic differentiation between populations. The observed levels of genetic differentiation between populations of P. corrugatus and the significant heterozygote deficiencies were unexpected, because this species has been reported to have a long-lived planktotrophic larva. It is hypothesised that recruitment of P. corrugatus in the South Orkney Islands originates from genetically distinct populations located in the Weddell Sea and to the west of the Antarctic Peninsula. Shifts in the relative position of the Weddell Sea Front, Weddell–Scotia Confluence and Scotia Front, relative to the South Orkney Islands, provide a mechanism for variation in the origin of recruits over time. Numerically and in biomass, the lanternfish Electrona antarctica is the dominant fish in the vast pelagic region of the Southern Ocean bounded on the north by the Antarctic Convergence and in the south by the Antarctic continental shelf. It is an important krill predator, and in turn is important in the diets of flighted and swimming seabirds. Further, it is the southernmost and coldest-dwelling representative of the globally distributed fish family Myctophidae. The present study was undertaken to estimate the species' growth rate and average life span, to incorporate the information in a basic energy budget, and to compare the growth of E. antarctica with more northerly confamilials. Fishes were aged using primary growth increments that were resolved on sagittal otoliths using three sequential techniques: thin-section grinding and polishing, etching, and scanning electron microscopy (SEM). Based on increment width (0.8 to 1.2 μm), continuity, and previous studies on confamilials, the microincrements were assumed to be deposited on a daily basis. Montages of SEM photomicrographs were constructed for each sagitta to allow the daily rings to be counted over the entire life span of 31 individuals representing the entire size range of the species. Results suggest a larval stage of 30 to 47 d and a maximum life span of 3.5 yr, with females growing faster than males in the last 1.5 yr of life and reaching a larger maximum size. Construction of a simple energy budget using the best information available suggests that a surplus of energy is available to support the observed growth rates (0.05 to 0.07 mm d−1). The results of the present study contrast markedly with previous estimates of an 8 to 11 yr maximum age for E. antarctica. These results provide important data addressing the ecology and population dynamics of the pelagic Antarctic ecosystem. E. antarctica is the end-member species in the continuum of vertically migrating myctophids that extend from the equator to the polar circle. Its growth rate is consonant with that of all other myctophid species examined using primary growth increments to determine age. The present study, in conjunction with earlier studies, suggests that growth rates of mesopelagic species are far higher than previously thought. The concentration of fluoride in the body parts of a range of Antarctic crustaceans from a variety of habits was examined with the aim of determining whether fluoride concentration is related to lifestyle or phylogenetic grouping. Euphausiids had the highest overall fluoride concentrations of a range of Antarctic marine crustaceans examined; levels of up to 5477 μg g−1 were found in the exoskeleton of Euphausia crystallorophias. Copepods had the lowest fluoride levels (0.87 μg g−1 whole-body); some amphipods and mysids also exhibited relatively high fluoride levels. There was no apparent relationship between the lifestyle of the crustaceans and their fluoride level; benthic and pelagic species exhibited both high and low fluoride levels. Fluoride was concentrated in the exoskeleton, but not evenly distributed through it; the exoskeleton of the head, carapace and abdomen contained the highest concentrations of fluoride, followed by the feeding basket and pleopods, and the eyes. The mouthparts of E.␣superba contained almost 13 000 μg F g−1 dry wt. Antarctic krill tail muscle had low levels of fluoride. The dynamics of infestation by parasitic juveniles of gnathiid isopods were investigated at Lizard Island in the summer of 1997/1998 to determine when, and at what rate, they infest fishes. Variation in gnathiid abundance on wild-caught fish (Hemigymnus melapterus) between dawn and sunset was examined, and unparasitized H. melapterus in cages were exposed to gnathiids in the field for 4 h (8 h for fish sampled at 06:00 h) at five different times of the day and night (10:00, 14:00, 18:00, 22:00, 06:00 hrs) on three reefs. To control for any potential effect of the cleaner-fish Labroides dimidiatus, which consumes large numbers of gnathiids each day, sampling was carried out on reefs from which all cleaner-fish had been removed. Gnathiid abundance decreased during the day. Standardized abundance per wild-caught fish was 1.9 times higher at dawn than at sunset. Gnathiids successfully infested fish in cages. Sixty-one percent of the fish in cages were infested with ≥1 gnathiids, with 51% of the fish having between 1 and 4 gnathiids after 4 h exposure. Gnathiids infested fish both day and night. The log10 (abundance +1) of gnathiids per caged fish varied significantly between time periods, with higher gnathiid numbers on fish sampled at 18:00, 22:00, and 06:00 hrs than on fish sampled at 10:00 and 14:00 hrs. In contrast to gnathiid abundance on wild-caught fish, the number of gnathiids on caged fish sampled at sunset was as high as that at dawn. The estimated mean (SE) cumulative number of gnathiids per caged fish [mean size = 13.7 cm (±0.25)] per day was 7.8 (1.1); this is similar to the estimated mean number of 7.3 gnathiids on similar-sized wild-caught fish at 6:00 hrs. The high infestation rate of gnathiids on caged fish in the late afternoon contrasted with the low numbers on wild-caught fish at this time, suggesting that factors other than infestation behaviour may be responsible for the low numbers of gnathiids on wild-caught fish in the afternoon. The euphausiid Euphausia crystallorophias Holt and Tattersall, 1906 is considered to be a neritic species. It has been found in greatest abundance along the Antarctic continental margins, often in association with regions of pack ice. Although E. crystallorophias has been observed at some islands to the west of the Antarctic Peninsula, the species has not previously been reported from islands of the maritime- or sub-Antarctic further north. During an oceanographic transect in November 1997 from South Georgia to the South Sandwich Islands, acoustic observations revealed a dense, discrete pelagic target at 50 m. The target was fished and was found to be an aggregation of small E. crystallorophias. The fishing location (54.48°S; 30.61°W) was >1500 km from the Antarctic continent, and >250 km from the nearest land, in water of several thousands of metres depth – clearly a non-neritic environment. Examination of hydrographic data revealed that the E. crystallorophias swarm had been located within a fast-flowing band of water that had characteristics of water found near the Antarctic Peninsula. This band was ≃150 km wide, and had a speed ranging from 9 to 22 km d−1 in a north-easterly direction. The possible origins of this E. crystallorophias swarm are explored in the light of the eddy-dominated current patterns prevalent in the Weddell–Scotia Confluence region, and with reference to published growth-rate estimates for the species. We discuss the potential for long-distance dispersal of E. crystallorophias and other neritic species in fast current jets, and examine how such oceanographic features could facilitate long-distance dispersal, colonization, and gene flow. We analysed growth of the Antarctic bryozoan Melicerita obliqua (Thornely, 1924) by x-ray photography and stable isotope analysis. M. obliqua colonies form one segment per year, thus attaining maximum length of about 200 mm within 50 years. In the Weddell and Lazarev Seas, annual production/biomass ratio of M. obliqua is 0.1 yr−1, which is in the range of other Antarctic benthic invertebrate populations. Production amounts to 3.34 mg Corg m−2 yr−1 and 90.6 mg ash m−2 yr−1 on the shelf (100 to 600 m water depth), and to 0.13 mg Corg m−2 yr−1 and 36.8 mg ash m−2 yr−1 on the slope (600 to 1250 m water depth). In the Antarctic krill Euphausia superba two forms of the chitinolytic enzyme N-acetyl-β-d-glucosaminidase (NAGase, EC 3.2.1.52) have been described, previously identified as NAGase B and NAGase C. Here, we demonstrate the organ-specific distribution and physiological relevance of both forms using a polyclonal antibody preparation which allows them to be distinguished immunologically. While NAGase B was localized in the integument and displayed a pattern of activity related to the moult cycle, the activity of NAGase C was independent of the moult cycle and was predominantly found in the gastrointestinal tract. Accordingly, NAGase B played a significant role in chitin degradation during the krill's moult, whereas NAGase C participated in the digestion of chitin-containing dietary components. Chromatographic elution profiles of isolated organs confirmed the immunological results by displaying characteristic organ-specific patterns in NAGase activity. The molecular characteristics of the moulting form, NAGase B, may further indicate a vesicular transport of moulting enzymes from the epidermis into the ecdysial space. Based on our results we develop a hypothesis explaining the concurrent processes of simultaneous chitin degradation and chitin synthesis occurring during moult. The influence of different N:P supply ratios on cell accumulation, chemical composition and toxicity of the marine haptophyte Chrysochromulina polylepis was examined in semi-continuous cultures. A non-axenic strain of C. polylepis was exposed to five different N:P supply ratios (N:P = 1:1, 4:1, 16:1, 80:1 and 160:1, by atoms), in order to create a range of N- and P-limited conditions. The toxicity per cell in C. polylepis was determined on four occasions at steady state cell density using the haemolytic activity of the cells expressed as saponin nanoequivalents. Haemolytic activity was demonstrated in all treatments, and increased in the algae when cell growth was nutrient limited (N:P = 1:1, 4:1, 80:1 and 160:1), compared to cells grown under non-limiting conditions (N:P = 16:1). This occurred regardless of the growth-limiting nutrient (N or P) and became more pronounced as nutrient limitation increased. In P-limited cultures the haemolytic activity per cell increased linearly with the cellular N:P ratio, whereas the N-limited cultures showed an opposite trend. The haemolytic activity per cell showed an inverse relationship with both cellular N and cellular P content. Cells limited by P showed a higher haemolytic activity than cells limited by N. The results suggest that toxicity in C. polylepis is strongly influenced by the physiological state of the algae. This may partially explain the large variability previously observed in the toxicity of C. polylepis blooms. The potential ecological significance of our findings is also discussed. We investigated the effects of addition of dissolved organic matter (DOM) on the growth and chemical composition of Nodularia spumigena cells under conditions of P or N deficiency. Semi-continuous monocultures of N. spumigena Mertens were established. The cells were exposed to the following treatments: nutrient-sufficient (+NP), nitrogen-deficient without DOM (−N) and with DOM (−NDOM), and phosphorus-deficient without DOM (−P) and with DOM (−PDOM). The −NDOM treatment had the highest N. spumigena cell density (6.1 × 107 cells l−1) during the experiment, whereas the P-deficient treatments had the lowest cell numbers (0.53 to 0.66 × 107 cells l−1). Both −P and −PDOM treatments yielded P-deficient N. spumigena cells. Dissolved organic nitrogen may provide N and increase N. spumigena yield under conditions of N deficiency. Symbiotic dinoflagellate algae (Symbiodinium sp.) isolated from the scleractinian coral Plesiastrea versipora and incubated in homogenized host tissue released 4 to 7 times as much glycerol (14 to 46 nmol glycerol/106 algae) as those incubated in seawater (3 to 6 nmol glycerol/106 algae) after 4 h incubation in the light. During this period, no release of triglycerides was detected. Intracellular glycerol increased 2- to 3-fold in algae incubated in host homogenate, but remained unchanged in algae incubated in seawater at a concentration of 0.82 ± 0.47 nmol glycerol/106 algae. In each incubation condition, intracellular triglyceride levels increased. However, in algae incubated in host homogenate, the intracellular levels of triglycerides reached only about 75% of the amount reached in algae incubated in seawater (max. 18.55 ± 2.40 nmol glycerol/106 cells). Host homogenate did not stimulate the release of glycerol from algae during dark incubation. These data show that the glycerol released by algae incubated in host-tissue homogenate was derived from increased synthesis of glycerol or from diversion of some glycerol or other photosynthetic intermediates from incorporation into algal triglyceride stores, and did not come from existing stores. Cytometric quantification of cellular fluorescence upon cleavage of fluorescein diacetate (FDA) is presented as a sensitive and rapid technique to assess phytoplankton metabolic activity during exposure to prolonged darkness of 10 to 12 d. Two distinct types of metabolic response to darkness are distinguished: Type I cells (Brachiomonas submarina, Pavlova lutheri, Chrysochromulina hirta) adapt to prolonged darkness by reducing their metabolism to a lower level of activity (∼10% of initial in P. lutheri, C. hirta, ∼0.5% in B. submarina) within few days, whereas Type II cells (Prymnesium parvum, Bacteriastrum sp., unidentified pennate diatom) continue with unchanged activity. Type I cells were able to maintain their initial cell abundance and commenced rapid cell growth upon re-illumination after 12 d of darkness. Among Type II cells, diatoms were able to maintain cell abundance and growth capacity as well, whereas P. parvum was not. Type I cells are expected to exhibit competitive advantages in environments with frequent or long dark periods. Bacterivory further supports dark survival in C. hirta. Samples of the Antarctic octopus Pareledone turqueti were taken from three locations on the Scotia Ridge in the Southern Ocean. The genetic homogeneity of these populations was investigated using isozyme electrophoresis. Whilst panmixia appeared to be maintained around South Georgia (F ST = 0) gene flow between this island and Shag Rocks, an island only 150 km away but separated by great depths, was extremely limited (F ST = 0.74). These results are examined with respect to the discontinuous distribution of P. turqueti throughout Antarctica. An estimate of effective population size was also calculated (N e = 3600).  Early development of the Antarctic sea urchin Sterechinus neumayeri was examined under two differ-ent culture regimes: one to simulate development near-bottom (“demersal development”) and the other to simulate the development of embryos in the water column (“pelagic development”). When embryos of both treatments reached the hatching blastula stage at 5 d post-fertilization (−1.5 °C), the blastulae that had undergone demersal development evidenced significant differences (by ANOVA or suitable non-parametric comparison) in the following: a thicker blastoderm layer (12%, P < 0.001), higher ash-free dry weights (19%, P < 0.01), lower mass-specific respiration rates (50%, P < 0.001), higher incorporation rates of 35S-methionine into protein (23%, P < 0.003), and a differential pattern of protein synthesis. When embryos developed demersally, they remained in the jelly-coat material released with the eggs at spawning. Quantitative isolation of this jelly-coat material in S. neumayeri demonstrated that it contained a significant amount of organic matter, 115 ng ash-free dry mass per egg, equivalent to 17% of the egg's initial organic mass. From September to November 1991, UV-absorbing mycosporine-like amino acids (MAAs) were monitored in a natural population of the sea urchin Sterechinusneumayeri from a coastal area of Anvers Island (Antarctic Peninsula). MAA concentrations were determined for specific tissues (gonad, digestive tract and body wall) from adults collected at four depths (intertidal, 8, 15 and 24 m). Four MAAs were identified: mycosporine-glycine, shinorine, porphyra-334 and paly-thine. Concentrations of MAAs among replicate individuals varied considerably. Ovaries had high concentrations of MAAs (84 to 1389 μg g−1 dry wt), while testes had non-detectable levels. The relative abundance of specific MAAs in ovaries appeared to be related to the spawning cycle. Digestive-tract samples had MAA concentrations as high as 3000 μg g−1 dry wt, but the mean MAA content in intertidal individuals decreased by 70% over 3 mo during spring. The body walls of sea urchins had very low amounts of MAAs (≤ 0.08 μg g−1 dry wt). There were significant depth differences in the␣total MAA content of the ovary ( p <0.001), ( p <0.015), digestive tract ( p <0.001), and body wall with organisms from the intertidal and 8 m depth having the highest concentrations of MAAs. Biological dosimetry indicated that UV-B (280 to 320 nm) wavelengths penetrated 3 to 7 m below the sea ice during the study period. The total MAA content in ovaries decreased with depth on all sample dates; however, the MAA content of the digestive tract and body wall did not exhibit a consistent pattern of change with depth. The MAA content of tissues did not change significantly with the temporal gradient of light exposure that was established by both ozone depletion and increasing photoperiod, except in the digestive tract sampled from intertidal specimens. Adult urchins are probably well-protected from UV exposure by the water column and a calcareous test; however, the results of this study suggest that, even under ice cover, depth of habitation is a determinant of MAA content in S. neumayeri. Large daily and seasonal fluctuations in the light regime, which are characteristic of Antarctic coastal environments, apparently do not provide reliable cues to elicit a detectable, temporal, biochemical response.
