The toxicity of pentachlorophenol (PCP) on Chlorella pyrenidosa algae was investigated with specific attention given to possible variation of toxic effects with time. A concentration-effect relationship was observed in which there was significant inhibition of PCP on cell density and chlorophyll A content. The inhibition rate of PCP on cell density was dependent on exposure time. The IC50 values after exposure times of 2, 4 and 6 days for cell growth were 4.18 +/- 0.49, 3.49 +/- 0.40 and 3.30 +/- 0.26 mg/L, respectively. There was also inhibition of chlorophyll A production, which appeared to increase marginally with exposure time for a given concentration of PCP. The corresponding IC50 values on day 2, 4 and 6 were 2.30 +/- 0.12, 2.63 +/- 0.38 and 3.30 +/- 0.34 mg/L, respectively. The effect of PCP on nitrate reductase (NR), was first stimulation followed by an inhibition phase. It is postulated that the observed temporal changes in the activity of nitrate reductase (NR) may occur through the addition or loss of phosphorus in the NR protein.  相似文献

The acute toxicity was determined for soil algae Chlorella kesslerei and Anabaena inaequalis, exposed to pesticides lindane, pentachlorophenol (PCP), isoproturon (IPU), and methyl parathion (MP). Toxicity markers included growth inhibition, chlorophyll biosynthesis, and total carbohydrate content, as a function of dose and time. Concentration response functions (EC50) were estimated by probit data transformation and weighted linear regression analyses. Lindane's toxicity to Chlorella increased sharply with time (EC50 = 7490, 10.3, 0.09 mg L(-1); 24, 48, 72 h), but remained nearly constant through 72 h with Anabaena (8.7-6.7 mg L(-1); 24-72 h). PCP at low concentrations stimulated algal growth and chlorophyll a production, an effect reversed at higher doses. Anabaena was less tolerant of PCP and MP than was Chlorella. The 96-h static EC50 values for Chlorella were: 0.003, 34, 0.05, and 291 mg L(-1) for lindane, PCP, isoproturon, and MP, respectively; for Anabaena, these were 4.2, 0.13, 0.21, and 19 mg L(-1). Carbohydrate production responses were similar to those of cell density (growth) and chlorophyll biosynthesis, with MP having the lowest adverse impact. The overall relative toxicity among the four tested pesticides was: for Chlorella, lindane > IPU > PCP > MP; and for Anabaena, PCP > IPU > lindane > MP. The results confirm that toxicants such as these pesticides may affect individual (though related) species to significantly different degrees.  相似文献

We investigated changes in biomass, biochemical fingerprints, fatty acid methyl ester (FAME) profile and functional status of the natural aquatic microbial communities upon impact of an Atradex pulse. The Atradex was applied to microcosm tanks at concentrations ranging from 24.5microgL(-1) to 245mgL(-1). The biomass of all microbial communities declined to a minimum level on day 4 with the effect being more pronounced in treated groups. Similarity between microbial communities also decreased on day 4 with the greatest change occurring at a concentration of 245mgL(-1) Atradex. After 8 days exposure to Atradex, microbial communities in all treated groups (except tanks spiked with 245mgL(-1) Atradex) recovered and showed similar metabolic fingerprints and FAME profiles to those of controls. Our results indicate that exposure to an Atradex pulse at concentration above 245mgL(-1), may irreversibly change the structure and functional status of aquatic microbial communities.  相似文献

In the present study surfactant addition with the help of either a mechanical dispersion or a thermal treatment was applied in order to increase the solubility and the bioavailability of phenanthrene in aqueous media, and therefore to promote its biodegradation. Among four tested surfactants (Tween 80, Brij 30, sodium dodecyl sulphate and rhamnolipids), Brij 30 (0.5 gL(-1)) showed the best results allowing us to attain about 20 mgL(-1) of soluble phenanthrene. An additional thermal treatment at 60°C for 24h, 200 rpm permitted to increase the solubility of phenanthrene in the presence of Brij 30 (0.5 gL(-1)) to about 30 mgL(-1). Higher dispersions of phenanthrene particles as well as the reduction of their size were obtained using Ultra-Turrax and French press. The biodegradation of phenanthrene by Pseudomonas putida was then investigated. The reduction of size of phenanthrene particles by mechanical dispersion did not influence its biodegradation, suggesting that P. putida consumed only soluble phenanthrene. The addition of Brij 30 (0.5 gL(-1)) permitted to obtain more phenanthrene metabolized. The use of Brij 30 coupled with a transitory heating of phenanthrene-containing medium at 60°C led to an even more complete biodegradation. This might be a promising way to enhance biodegradation of PAHs.  相似文献

A toxicity test using spore release of the aquatic green alga, Ulva, was developed and evaluated by assessing the toxicity of different organic and inorganic chemicals and elutriates of sewage or waste sludge. The toxic ranking of four metals was: Cu (EC50 of 0.040mgL(-1))>Cd (0.095mgL(-1))>Pb (0.489mgL(-1))>Zn (0.572mgL(-1)). The EC50 for TBTO ranged from 24 to 63microgL(-1). The most toxic VOC was formalin (EC50 of 0.788microlL(-1)) and the least toxic was acetone. Spore release was significantly inhibited in all elutriates; the greatest and least toxic effects were for industrial sewage (3.29%) and filtration bed (10.08%), respectively. The bioassay is simple, inexpensive and sensitive. The cosmopolitan distribution of Ulva means that the test would have a potential application worldwide.  相似文献

Bioassays were conducted in 2001 and 2002 to estimate toxicities and dose-response relationships of 24 Bemisica tabaci Gennadius populations to pyriproxifen, acemitaprid, and diafenthiuron. LC50s ranging from 0.014 to 0.096 mgL(-1), 0.60 to 1.3 mgL(-1), and 3.5 to 6.7 mgL(-1) were observed respectively for pyriproxifen, acemitaprid, and diafenthiuron. These LC50s much lower than the field doses recommended for each compound. A fast increase in rates of mortality within a narrow range of lethal concentrations was observed for each compound. indicating that all three compounds were highly effective at killing whiteflies. In a separate experiment, pyriproxifen, acemitaprid, and diafenthiuron were tested in 2001 and 2002 to compare their effectiveness and assess their impact on parasitism in the field. In both years all three compounds significantly prevented B. tabaci populations from reaching economic injury levels in cotton and minimized adverse effects on parasitism. Our results provide for the first time baseline toxicological, field efficacy, and effect on parasitism data for pyriproxifen, acemitaprid, and diafenthiuron against B. tabaci in West Africa. These compounds should be included in a resistance management program of the cotton pest complex and their use should be restricted to prevent the building of resistance in B. tabaci populations.  相似文献

Lichens are an excellent model to study the bioaccumulation of heavy metals but limited information is available on the molecular mechanisms occurring during bioaccumulation. We investigated the changes of the lichen proteome during exposure to constant concentrations of mercury. We found that most of changes involves proteins of the photosynthetic pathway, such as the chloroplastic photosystem I reaction center subunit II, the oxygen-evolving protein and the chloroplastic ATP synthase β-subunit. This suggests that photosynthesis is a target of the toxic effects of mercury. These findings are also supported by changes in the content of photosynthetic pigments (chlorophyll a and b, and β-carotene). Alterations to the photosynthetic machinery also reflect on the structure of thylakoid membranes of algal cells. Response of lichens to mercury also involves stress-related proteins (such as Hsp70) but not cytoskeletal proteins. Results suggest that lichens adapt to mercury exposure by changing the metabolic production of energy.  相似文献

A series of lab and greenhouse studies were undertaken to understand how Cu and Zn toxicity influences Bromus carinatus (Hook and Arn.) growth, to what degree an organic amendment (yard waste compost) may reduce Cu and Zn bioavailability in Cu-Zn minespoil and promote plant growth in combination with fertilizer, and how the vertical distribution of compost in the minespoil influences rooting depth. Root Cu and Zn toxicity thresholds were determined to be 1 mgL(-1) and 10 mgL(-1) in solution, respectively. The compost amendment had exceptionally high Cu and Zn binding capacities (0.17 and 0.08 g metal g C(-1), for Cu and Zn, respectively) that were attributed to high compost humic and fulvic acid concentrations. Maximum plant biomass was achieved when minespoil was amended with compost and fertilizer in combination. Fertilizer alone had no effect on plant growth. Mixing compost into the minespoil was essential to promote adequate rooting depth.  相似文献

A red tide was detected in the inner parts of Tolo Harbour, Hong Kong, in November 2000. Water samples were collected from a fixed station at the centre of the red tide patch for microscopic analysis of phytoplankton community composition and high performance liquid chromatography (HPLC) analysis of phytoplankton pigments. At the peak of the red tide on 24 November 2000, phytoplankton was dominated by the dinoflagellate Scrippsiella trochoidea. The red tide began to decline at the end of November and, by 1 December 2000, the phytoplankton was dominated by diatoms. Chlorophylls and carotenoids in water samples were analysed using HPLC pigment separation technique. Dinoflagellates were indicated by the signature pigment peridinin. Significant correlation (r=0.999) was found between the peridinin concentration and dinoflagellate density. A decrease in peridinin and an increase in fucoxanthin, a major carotenoid in diatoms, marked the shift in phytoplankton composition at the end of the red tide. HPLC analysis also revealed the occurrence of minor phytoplankton groups that are difficult to identify by light microscopy. Red tide monitoring and study of red tide dynamics in Hong Kong have been based on cell counting and spectrophotometric or fluorometric measurement of chlorophyll a. HPLC pigment analysis provides an effective alternative for investigating phytoplankton dynamics during red tide and other algal blooms.  相似文献

The dependence of bats in Britain on houses as roosts may result in them being exposed to pesticides used in remedial timber treatments. Pentachlorophenol (PCP) and permethrin are used as a fungicide and an insecticide for timber treatment, respectively. The present study investigated toxicity and distribution in body tissues of these two pesticides in pipistrelle bats. Four groups of nine to ten bats were kept in separate outdoor flight enclosures and were provided with roost boxes treated with either PCP only, permethrin, PCP/permethrin mixture or solvent only (control). At the start of the experiment, mean (+/-SE) PCP and permethrin concentrations on the surface of wooden blocks that had been treated in the same way as roost boxes were 69.32+/-6.76 mg g(-1) (n=6) and 3.3+/-1.6 mg g(-1) (n=3), respectively. All bats exposed to PCP and PCP/permethrin treated boxes died within 24 and 120 h, respectively; nine out of the ten controls survived the 32 day experimental period (P<0.001; both groups compared with control). Bats exposed to permethrin treated boxes survived as well as controls. Mean (+/-SE) carcass PCP concentration (excluding deposits on fur) of bats exposed to PCP and PCP/permethrin treated boxes was 13.11+/-2.52 microg g(-1)BW (n=20). PCP burdens on fur were positively correlated with total weight of PCP in the carcass (P<0.001). PCP was present in fat depots, liver, kidney and the remainder of the body which, despite containing low PCP concentrations, was the main PCP reservoir (66.4+/-5.0% of carcass PCP load; n=20). Total PCP in the carcass was significantly correlated with lipid weight (P<0.005). Permethrin was not detectable in body washes and tissues of bats exposed to PCP/permethrin mixture or permethrin.  相似文献

The plants of Spinacia oleracea L. grown on contaminated soil (CS) and different amendments of tannery sludge (TS) have shown high accumulation of metals in its edible part. The accumulation of toxic metal (Cr) in the leaves of the plants grown on CS was recorded as 40.67 microgg(-1)dw. However, the leaves of the plants grown on 100% TS have accumulated about two times (70.80 microgg(-1)dw) higher Cr than the 10% TS (31.21 microgg(-1)dw). Among growth parameters, the root length was more affected at 90 d than the shoot length, number of leaves and leaf area. The study of scanning electron micrographs showed 29.31% increase in stomatal length in the leaves of the plants grown on CS as compared to garden soil (GS), which served as control, however it decreased in the plants grown on higher amendments of TS. The decrease in MDA content at initial period of exposure and lower amendment was recorded in the leaves, whereas, significant increase (>10% TS onward) was observed with increase in tannery sludge ratio at 90 d as compared to GS. A coordinated increase in all the studied antioxidants (cysteine, non-protein thiol, ascorbic acid, carotenoid contents) was found up to 75 d of growth. At 90 d, most of the antioxidant decreased as compared to 75 d causing oxidative stress as evidenced by increased level of lipid peroxidation and decreased chlorophyll and protein contents. Maximum increase of 181.43% in MDA content and maximum decrease of 53.69% in total chlorophyll content was recorded in the leaves of the plants grown on 100% TS after 90 d of growth. The plants grown on CS have shown an increase in shoot length, number of leaves, leaf area, photosynthetic pigments and protein contents and in all the studied antioxidants. Thus, these plants are able to combat stress involving defense mechanism, resulting in healthy growth of the plants. The results are well coordinated as there is no change in the MDA content as compared to the plants grown on GS. In view of high Cr accumulation in edible part of S. oleracea grown on CS after irrigation with tap water, it is not advisable to use these plants for edible purposes. Summing up, it is recommended that the level of metals in the edible part should be checked instead of healthy growth as deciding parameter for consumption. It is demonstrated through this study that metal enriched plants have detoxification mechanism and grow well on organic matter enriched contaminated soil.  相似文献

In 1990, the Gulf War caused an unprecedented environmental catastrophe. More than 700 oil wells were set on fire, emitting thousands of tons of oxides of nitrogen, sulfur, soot and oil mist at high temperatures. The blazing oil wells and lakes were the major cause of environmental pollution, damaging flora and fauna throughout Kuwait and the surrounding region. We set up this study in order to investigate whether Vicia faba could be used as a bioindicator of oil pollution. Growth parameters of Vicia faba, such as levels of photosynthetic pigments, proteins, free amino acids, phenols, sugars, biomass, moisture and fatty acids, were studied. Levels of total reducible sugars, phenols, proteins, free amino acids and proline were found to be higher in plants grown in hydrocarbon-polluted soil, compared to those grown in control soil. However, biomass, moisture, pheophytin, the 435/415 nm ratio for chlorophylls and the chlorophyll a/carotenoid ratio, were higher in the plants grown in control soil than those grown in polluted soil. Our results indicate that Vicia faba can be used as a bioindicator of pollution and a ratio of the fatty acid C(20:1)/C(18:1) is an important index of environmental oil pollution.  相似文献

Batch kinetic and isotherm studies were carried out to determine the adsorptive characteristics of peat and bentonite mixtures for pentachlorophenol, and to examine the hydraulic conductivity of peat-bentonite mixtures to determine if they are applicable for use as cutoff barriers. Batch kinetic studies showed that over 90% of PCP was removed from water spiked with approximately 1 mg/l of PCP using a peat-bentonite (5%) mixture. The equilibrium time was 8 hours. The optimum pH range for adsorption of PCP by the peat-bentonite mixture was found to be 3-3.5. Batch isotherm studies showed that the adsorption of PCP by the peat-bentonite mixture from aqueous solution was best described by the Freundlich isotherm equation. Batch adsorption studies using various ratios of bentonite in the mixture showed that the adsorption of PCP decreased linearly with increased amount of bentonite in the mixture, indicating that adsorption of PCP by the peat moss portion of the mixture was the dominant process. The inverse of the hydraulic conductivity was found to increase exponentially with an increase in the bentonite content of the mixture over the range studied. The minimum hydraulic conductivity observed was 3.3 x 10(-7) cm/s for a 50% peat-50% bentonite mixture. Peat-bentonite mixtures can be used to successfully remove PCP from aqueous media and can be used effectively as a barrier to attenuate the migration of PCP through soil and groundwater systems.  相似文献

Air pollution levels are increasing at an alarming rate in many developing countries, including India and causing a potential threat to crop production. Field experiments were conducted to examine the impact of urban air pollutants on biomass (yield) and some physiological and biochemical parameters of palak (Beta vulgaris L. var. All Green) that grew from germination to maturity at seven periurban sites of Allahabad city having different concentrations of air pollutants under different levels of nutrients. The 6h daily mean NO2, SO2 and O3 concentrations varied from 2.5 to 42.5, 10.6 to 65 and 3.5 to 30.8 microg m(-3), respectively at different locations. Levels of air pollution showed significant negative correlations with photosynthetic pigments, protein, ascorbic acid and starch contents and catalase activity of palak leaves. A significant negative correlation was found for total biomass with SO2 (r=-0.92), NO2 (r=-0.85) and O3 (r=-0.91) concentrations. The increased fertilizer application (N, P and K) over the recommended dose resulted in a positive response by reducing losses in photosynthetic pigments and total biomass. This study proved that ambient air pollution of Allahabad city is influencing negatively to the growth and yield of palak plants.  相似文献

An organic-aqueous two-liquid-phase partitioning system was developed to degrade high concentrations of pentachlorophenol (PCP). Dioctyl sebacate was selected among 12 non-aqueous phases as the most suitable solvent to control the delivery of PCP to the aqueous phase for being non-biodegradable and biocompatible. In shake-flask experiments, the two-phase system was able to support the removal of 1g PCP l(-1) of total liquid phase. The performance of the two-liquid phase partitioning system (TLPPS) in shake-flask was evaluated under different conditions. At the initial biomass concentrations of 7, 25, and 58 mg dry weight l(-1), the volumetric removal rates of PCP obtained were 25.7+/-0.5, 32.1+/-0.1, and 39.3+/-2.9 mg PCP l(-1)h(-1), respectively. Higher performance was observed at lower organic-aqueous phase ratios (16% and 28%) than higher ones (37% and 44%). In a 2-l TLPPS, the degradation of 10 g PCP was completed in less than 100 h at a total volumetric rate of 142 mg l(-1) h(-1). Kinetics study using Monod model showed that compared to monophasic systems, the biphasic system significantly enhanced the maximum specific growth rate and PCP removal rate. Results of this biphasic system showed no accumulation of unknown by-product(s) which has been reported for physical-pretreatment or high-performance biphasic systems of PCP degradation.  相似文献

Purpose

The objective of this study was to determine the removal of zinc and copper by two freshwater green microalgae Chlorella pyrenoidosa and Scenedesmus obliquus and to investigate changes of algal ultrastructure and photosynthetic pigment.

Methods

Algal cells were exposed for 8 days to different initial zinc or copper concentrations. Heavy metal concentrations were detected by an atomic absorption spectrophotometer. Algal growth, ultrastructure, and photosynthetic pigment were analyzed by a microplate reader, transmission electron microscope, and spectrophotometer, respectively.

Results

Low zinc and copper concentrations induced increase in algal growth, whereas application of high zinc and copper concentrations suppressed the growth of both algae. High metal concentrations also decreased the photosynthetic pigments and destroyed algal cell ultrastructure. The zinc removal efficiency by both algae increased rapidly during the first day and thereafter remained nearly constant throughout the experiment. The copper removal efficiency by both algae increased slowly during the whole experimental periods. In all cultures, the quantity of both metals removed intracellularly was much lower than the adsorbed quantity on the cell surface.

Conclusions

Both strains of the microalgae had proven effective in removing zinc and copper from aqueous solutions, with the highest removal efficiency being near 100%. In addition, C. pyrenoidosa appeared to be more efficient than S. obliquus for removing copper ions. On the contrary, S. obliquus appeared to be more efficient than C. pyrenoidosa for removing zinc ions.  相似文献

The sorption characteristics of pentachlorophenol (PCP) in the surficial sediments were investigated using a selective extraction procedure. The results show that the Gamma(max) of PCP sorption decreased from 1.60mumolg(-1) to 0.69mumolg(-1) by approximately 60% after selective removal of organic materials from the sediments. The sorption of PCP in the sediments after selective removal of Mn oxides increased nearly up to 600% (from 1.60mumolg(-1) to 11.11mumolg(-1)) and, to a less degree, the PCP sorption in the sediments after simultaneous removal of Fe/Mn oxides (Gamma(max)=3.53mumolg(-1)). The analysis of the data using an additional model indicates that the contribution of Mn oxides to PCP sorption was negative, and Fe oxides and organic materials both have greater potential for sorption of PCP with less contribution from residues including Mn and Fe oxides in the residual fractions determined by a sequential extraction procedure and clay and silicate minerals. The differences in the decreased degrees of PCP sorption with increasing of Cu suggest that competition between Cu and PCP for sorption sites mainly takes place on Fe oxides.  相似文献

A cohort of 2122 US pentachlorophenol (PCP) production workers from four plants in the National Institute for Occupational Safety and Health Dioxin Registry was exposed to PCP and to polychlorinated dibenzo-p-dioxin and dibenzofuran contaminants of PCP production. A subcohort of 720 was also exposed to 2,3,7,8-tetrachlorodibenzodioxin, a contaminant of trichlorophenol (TCP) while using TCP or a TCP derivative. PCP and several production contaminants have been implicated as animal carcinogens. A priori hypotheses were that the cohort would have elevated standardized mortality ratios (SMRs) for aplastic anemia, soft-tissue sarcoma, and non-Hodgkin lymphoma, as suggested by human studies, and for leukemia and liver, adrenal, thyroid, and parathyroid cancer, as suggested by animal studies. From 1940 to 2005 1165 deaths occurred with an overall SMR of 1.01 [95% confidence limits (CI), 0.95-1.07]. Overall cancer mortality (326 deaths, SMR 1.17, CI 1.05-1.31) was in statistically significant excess. There were excess deaths for trachea, bronchus and lung cancers (126 deaths, SMR 1.36, CI 1.13-1.62), non-Hodgkin lymphoma (17 deaths, SMR 1.77, CI 1.03-2.84), chronic obstructive pulmonary disease (63 deaths, SMR 1.38, CI 1.06-1.77), and medical complications (5 deaths, SMR 3.52, CI 1.14-8.22). In race- and sex-specific analyses, white males had increased non-Hodgkin lymphoma mortality (17 deaths, SMR 1.98, CI 1.15-3.17) and males of other races had increased leukemia mortality (four deaths, SMR 4.57, CI 1.25-11.7). The excess of cancers of a priori interest, non-Hodgkin lymphoma and leukemia, provide some support for the carcinogenicity of PCP, however, further studies with more detailed exposure assessment are needed.  相似文献

A glass house experiment was conducted to investigate the effect of soil arsenic on photosynthetic pigments, chlorophyll-a and -b, and their correlations with rice yield and growth. The experiment was designed with three replications of six arsenic treatments viz. control, 10, 20, 30, 60, 90 mg of As kg(-1) soil. Arsenic concentration in initial soil, to which the above mentioned concentrations of arsenic were added, was 6.44+/-0.24 mg kg(-1). Both chlorophyll-a and -b contents in rice leaf decreased significantly (p<0.05) with the increase of soil arsenic concentrations. No rice plant survived up to maturity stage in soil treated with 60 and 90 mg of As kg(-1). The highest chlorophyll-a and -b contents were observed in control treatment (2.62+/-0.24 and 2.07+/-0.14 mg g(-1) were the average values of chlorophyll-a and -b, respectively of the five rice varieties) while 1.50+/-0.20 and 1.04+/-0.08 mg g(-1) (average of five rice varieties) of chlorophyll-a and -b, respectively were the lowest. The content of photosynthetic pigments in these five rice varieties did not differ significantly (p>0.05) from each other in control treatment though they differed significantly (p<0.05) from each other in 30 mg of As kg(-1) soil treatment. Among the five rice varieties, chlorophyll content in BRRI dhan 35 was found to be mostly affected with the increase of soil arsenic concentration while BRRI hybrid dhan 1 was least affected. Well correlations were observed between chlorophyll content and rice growth and yield suggesting that arsenic toxicity affects the photosynthesis which ultimately results in the reduction of rice growth and yield.  相似文献
