The novel Serratia proteamaculans isolated from a chromium-contaminated site was tolerant to a concentration of 500 mg Cr(VI)/l. The optimum pH and temperature for reduction of Cr(VI) by S. proteamaculans were found to be 7.0 and 30 °C, respectively. The Cr(VI) reduction rate decreased with the increase in Cr(VI) concentration from 100 to 400 mg/l, suggesting the enzymatic chromium reduction. Resting and permeabilised cell assays provided the better evidence that chromate reduction in S. proteamaculans is enzymatic. Reduction by cell-free filtrate shows no extracellular chromate-reducing activity, revealing that this activity may be associated to membrane fraction and/or cytosolic fraction. Assays conducted with cytosolic and particulate fraction of S. proteamaculans confirmed the role of membrane-bound proteins in Cr(VI) reduction. Furthermore, chromium reduced by heat-treated cells suggests that membrane-associated chromate reductase activity of S. proteamaculans is preceded by its adsorption on the cell surface. 相似文献

Environmental monitoring by means of biomarker parameters assessed in different species is a useful tool. It has the advantage of providing a quantitative response as well as valuable information of ecological relevance on the chronic adverse effects caused by water pollution. The aim of this study was to assess the response of biochemical and physiological parameters of Cnesterodon decemmaculatus, a native teleost, simultaneously caught in two sites of Reconquista river, a highly polluted peri-urban river. This study compared the measured parameters with that of specimens of the same species captured in an unpolluted body water, and correlated the detected changes with the physicochemical profile of the water at each site. A comparison was made of selected parameters of gill, brain and liver and of somatic indices of fish collected from polluted and reference sites. The main parameters whose changes allowed to discriminate between sampling sites were gill (Na(+),K(+))-ATPase, brain acetylcholinesterase (AChE) and liver aminotransaminases activities; tissues' protein content and liver somatic index (LSI) were also sensitive biomarkers in brain and liver, respectively. The results showed that the response of the measured biomarkers allowed for the differentiation of sampling sites according to their water quality and confirmed that Cnesterodon decemmaculatus may be a useful test organism for the biomonitoring of freshwater environments. In addition, the simultaneous measurement of the physicochemical parameters of the water samples showed a good correspondence between the biomarkers responses and the environmental chemical stress conditions. 相似文献

The impact of an industrial effluent containing high loads of calcium, cadmium, lead chloride and sulphate, on a river ecosystem was assessed using a combination of an effluent toxicity test, an ambient toxicity test and an ecological survey. Only this combination of techniques made it possible to discriminate between the effects of the discharge and those of the background pollution. Each of the individual techniques detected essential effects which the other failed to reveal. With the physical and chemical measurements, important increases of several components were measured at all sampling sites downstream of the discharge. With the ecological survey, however, no large changes in water quality could be determined at the sampling sites, due to the high degree of pollution present upstream of the discharge. Reproduction of Daphnia magna, exposed to sublethal effluent dilutions, was followed over two generations. The offspring of the first generation were shown to have an increased sensitivity to the effluent, compared to the first generation that was born from previously unexposed mothers. Besides the toxicity of the effluent, the acute and chronic toxicity of its main component, CaCl(2), was also determined. The results of the CaCl(2)-tests and toxicity data from literature for the suspected toxicants were transformed to Toxic Units (TU). Using the sum of the TUs we investigated the possibility of predicting effluent toxicity to Daphnia magna. Effluent toxicity was under-estimated by calculating the sum of the TUs of the individual components. Dilution of the effluent to a level at which the measured toxicant concentrations comply with European regulations still showed significant effects on Daphnia reproduction. 相似文献

Contaminated sediment in the river basin has become a source of pollution with increasing importance to the aquatic ecosystem downstream. To monitor the temporal changes of the sediment bound contaminants in the River Elbe and the River Dommel monthly toxicity tests were applied to layered sediment and river water samples over the course of 10 months. There is an indication that contaminated sediments upstream adversely affected sediments downstream, but this process did not cause a continuous increase of sediment toxicity. A clear decrease of toxic effects in water and upper layer sediment was observed at the River Elbe station in spring related to high water discharge and algal blooms. The less obvious variation of sediment toxicity in the River Dommel could be explained by stable hydrological conditions. Future monitoring programmes should promote a more frequent and intensive sampling regime during these particular events for ecotoxicological evaluation. 相似文献

Metalliferous soils cover a relatively large surface area in Morocco, and up to now no hyperaccumulating plants have been identified on these mining or these industrial sites. The aim of this work was to assess the extent of metal accumulation by plants found in three mining areas in southern Morocco with the ultimate goal of finding metal hyperaccumulating species by using the MetPAD biotest. The biotest helps to obtain information on the selective metal toxicity of aqueous extracts from the plants. A strong metal toxicity, as revealed by the biotest is an indication of a hyperaccumulating plant. Toxicity tests were run concurrently with chemicals analyses of metals in plants and their water extracts. The chemical analyses allow the determination of the hyperaccumulated metal(s). Specimens of the plant species mainly growing on and in the vicinity of the three mines were sampled with their corresponding soils. The results show that all plants analyzed had lower heavy metal content and toxicity despite the relatively very high soil concentrations. A comparison of our results with the criterion used to classify the hyperaccumulator plants indicates that plants we collected from mining sites were hypertolerant but not hyperaccumulators. This was confirmed by transfer factors generally lower than 1. Nevertheless, these tolerant plants species can be used as tools for revegetation for erosion control in metals-contaminated sites (phytostabilization). 相似文献

The establishment of an efficient surface water quality monitoring (WQM) network is a critical component in the assessment, restoration and protection of river water quality. A periodic evaluation of monitoring network is mandatory to ensure effective data collection and possible redesigning of existing network in a river catchment. In this study, the efficacy and appropriateness of existing water quality monitoring network in the Kabbini River basin of Kerala, India is presented. Significant multivariate statistical techniques like principal component analysis (PCA) and principal factor analysis (PFA) have been employed to evaluate the efficiency of the surface water quality monitoring network with monitoring stations as the evaluated variables for the interpretation of complex data matrix of the river basin. The main objective is to identify significant monitoring stations that must essentially be included in assessing annual and seasonal variations of river water quality. Moreover, the significance of seasonal redesign of the monitoring network was also investigated to capture valuable information on water quality from the network. Results identified few monitoring stations as insignificant in explaining the annual variance of the dataset. Moreover, the seasonal redesign of the monitoring network through a multivariate statistical framework was found to capture valuable information from the system, thus making the network more efficient. Cluster analysis (CA) classified the sampling sites into different groups based on similarity in water quality characteristics. The PCA/PFA identified significant latent factors standing for different pollution sources such as organic pollution, industrial pollution, diffuse pollution and faecal contamination. Thus, the present study illustrates that various multivariate statistical techniques can be effectively employed in sustainable management of water resources. Highlights ? In March 1988 the authors completed a report on the risk assessment of mixtures of halogenated dibenzo-p-dioxins and dibenzofurans at the request of the National Institute of Public Health and Environmental Protection in the Netherlands. Based on a study of the current knowledge of these compounds, a set of toxicity equivalency factors was proposed for use in such risk assessments. This paper presents the section of this report that deals with the literature on the effects of these compounds and the choice of toxicity equivalency factors. Consequent to the growing demand for alternative sources of energy, the seeds from Jatropha curcas remain to be the favorite for biodiesel production. However, a significant volume of the residual organic mass (seed cake) is produced during the extraction process, which raises concerns on safe waste disposal. In the present study, we assessed the toxicity of J. curcas seed cake using the zebrafish ( Danio rerio) embryotoxicity test. Within 1-h post-fertilization (hpf), the fertilized eggs were exposed to five mass concentrations of J. curcas seed cake and were followed through 24, 48, and 72 hpf. Toxicity was evaluated based on lethal endpoints induced on zebrafish embryos namely egg coagulation, non-formation of somites, and non-detachment of tail. The lowest concentration tested, 1 g/L, was not able to elicit toxicity on embryos whereas 100 % mortality (based also on lethal endpoints) was recorded at the highest concentration at 2.15 g/L. The computed LC50 for the J. curcas seed cake was 1.61 g/L. No further increase in mortality was observed in the succeeding time points (48 and 72 hpf) indicating that J. curcas seed cake exerted acute toxicity on zebrafish embryos. Sublethal endpoints (yolk sac and pericardial edema) were noted at 72 hpf in zebrafish embryos exposed to higher concentrations. The observed lethal endpoints induced on zebrafish embryos were discussed in relation to the active principles, notably, phorbol esters that have remained in the seed cake even after extraction. 相似文献

The potential applications of immobilized bitter gourd peroxidase in the treatment of model wastewater contaminated with phenols have been investigated. The synthetic water was treated with soluble and immobilized enzyme preparations under various experimental conditions. Maximum removal of phenols was found in the buffers of pH values 5.0-6.0 and at 40 degrees C in the presence of 0.75 mM H(2)O(2). Fourteen different phenols were independently treated with soluble and immobilized bitter gourd peroxidase in the buffer of pH 5.6 at 37 degrees C. Chlorinated phenols and native phenol were significantly removed while other substituted phenols were marginally removed by the treatment. Phloroglucinol and pyrogallol were recalcitrant to the action of bitter gourd peroxidase. Immobilized bitter gourd peroxidase preparation was capable of removing remarkably high percentage of phenols from the phenolic mixtures. Significantly higher level of total organic carbon was removed from the model wastewater containing individual phenol or complex mixture of phenols by immobilized bitter gourd peroxidase as compared to the soluble enzyme. 2,4-dichlorophenol and a phenolic mixture were also treated in a stirred batch reactor with fixed quantity of enzyme for longer duration. The soluble bitter gourd peroxidase ceased to function after 3h while the immobilized enzyme was active even after 6h of incubation with phenolic solutions. 相似文献

A new Aeromonas bioassay is described to assess the potential harmful effects of the glyphosate-based herbicide, Roundup ®, in the Albufera lake, a protected area near Valencia. Viability markers as membrane integrity, culturability and β-galactosidase production of Aeromonas caviae were studied to determine the influence of the herbicide in the bacterial cells. Data from the multifactor analysis of variance test showed no significant differences ( P > 0.05) between A. caviae counts of viability markers at the studied concentrations (0, 50 and 100 mg l −1 of glyphosate). The effects of Roundup® on microbial biota present in the lake were assessed by measuring the number of indigenous mesophilic Aeromonas in presence of different amounts of the herbicide at 0, 50 and 100 mg l−1 of glyphosate. In samples containing 50 and 100 mg l−1 of glyphosate a significant (P < 0.05) increase in Aeromonas spp. counts and accompanying flora was observed. The acute toxicity of Roundup® and of Roundup® diluted with Albufera lake water to Microtox® luminescent bacterium (Vibrio fischeri) also was determined. The EC50 values obtained were 36.4 mg l−1 and 64.0 mg l−1 of glyphosate respectively. The acidity (pH 4.5) of the herbicide formulation was the responsible of the observed toxicity. 相似文献

Ambient water toxicity estimations were conducted in Piles Creek as part of an ecological risk assessment study. This creek is part of a highly polluted estuarine system located near Linden, New Jersey. An upstream and a downstream site in the creek were evaluated for effects on survival, growth and sexual maturity of the estuarine mysid, Mysidopsis bahia. Manasquan Inlet, Manasquan, New Jersey, served as the reference site. Seven-day screening toxicity tests were conducted once a month over a three month period. Results indicate that survival of mysids was not affected in any toxicity test. Mysid growth was significantly reduced at both of the test sites, in two out of three trials. Sexual maturity was shown to be adversely affected in mysids exposed to ambient waters from both test sites throughout the duration of the study. 相似文献

In the present work, the influence of Ca 2+ concentration on the toxicity of single cut linear alkylbenzene sulfonate (LAS) homologues was studied. Precipitation boundary diagrams for each homologue were obtained, indicating turbid and clear zones depending on the LAS and Ca 2+ concentrations. The separation between transparent and turbid zones is given by the so-called precipitation line. LAS toxicity to Daphnia magna was determined at concentrations close to this precipitation line. It was observed that when Ca(LAS) 2 precipitation progresses, LAS bioavailability decreases for test animals, and the toxicity diminishes even at high nominal LAS concentrations. According to the “free ion activity model” (FIAM), the toxicity of a given chemical compound is mainly due to the ionic species (Ca 2+–LAS −) and not due to the precipitated molecule, Ca(LAS) 2. The significance of the present study is in connection with the assessment of LAS sorption/precipitation studies in soils and sediments, where in situ toxicity is strongly influenced by Ca 2+/Mg 2+ ions, according to the results presented in this work. 相似文献

Tropospheric ozone (O(3)) is considered one of the most important air pollutants affecting human health. The role of peri-urban vegetation in modifying O(3) concentrations has been analyzed in the Madrid region (Spain) using the V200603par-rc1 version of the CHIMERE air quality model. The 3.7 version of the MM5 meteorological model was used to provide meteorological input data to the CHIMERE. The emissions were derived from the EMEP database for 2003. Land use data and the stomatal conductance model included in CHIMERE were modified according to the latest information available for the study area. Two cases were considered for the period April-September 2003: (1) actual land use and (2) a fictitious scenario where El Pardo peri-urban forest was converted to bare-soil. The results show that El Pardo forest constitutes a sink of O(3) since removing this green area increased O(3) levels over the modified area and over down-wind surrounding areas. 相似文献

We investigated the effects of pollutants on two pine species (Pinus koraiensis and Pinus rigida) in an industrial region in Korea, using a physiological approach. The concentrations of fluorine (F) and chlorine (Cl) in the atmosphere, in precipitation and soil water at the damaged site were all significantly higher than at a control site. Moreover, the concentrations of F, Cl and Mn in pine needles were significantly higher, and essential elements and chlorophyll in needles were significantly lower at the damaged site than at the control site. The photosynthetic capacities, shoot length and survival statistics of needles of the two pines were all significantly reduced at the damaged site compared to the control site, especially P. rigida. Based on our comparison of photosynthetic responses and the concentrations of F, Cl and Mn in needles of the two pine species, P. koraiensis is more resistant to excess Mn in its needles than P. rigida. 相似文献

The freshwater quality criteria of acetonitrile were studied on the basis of the features of the aquatic biota in China, and with reference to US EPA's guidelines. Acute tests were performed on 13 different species to determine 48h-EC 50/96h-EC 50 (or 96h-LC 50) values for acetonitrile. 2ld survival-reproduction tests with Daphnia magna and growth inhibition tests with Lemna minor were also conducted to estimate lower chronic limit and upper chronic limit values. In the acute tests, Radix plicatula was the most sensitive species to acetonitrile followed by Chironomus sp., Cyprinus carpio and D. magna in turn. The final acute value of acetonitrile was 2290 mg/l. In the chronic tests, reproduction of daphnids was significantly reduced by acetonitrile at 320 mg/l. Acute-to-chronic ratios ranged from 2 to 15.8. A final chronic value of 413.9 mg/l was obtained and a final plant value was 1342 mg/I. A criterion maximum concentration (1145 mg/l) and a criterion continuous concentration (413.9 mg/l) were derived. 相似文献

Sea salts (seven brands from six commercially-available sources) were dissolved in water to develop 30 g/l solutions, and selected water quality characteristics were then monitored for 96 h. One or more water quality characteristics changed significantly during the 96 h period in six of the reconstituted sea salts. Measured characteristics of sea water diluted to 30 g/l demonstrated no changes during the observation period. The sea salts from different sources also demonstrated differences in absolute concentrations of some characteristics measured. Application of the Biotic Ligand Model to predict copper toxicity to the bivalve Mytilus edulis in solutions of the salts tested yielded 96-h median-lethal concentrations that ranged from 2 to 13 μg/l. Since water quality affects toxicity of many environmental pollutants, the source of the sea salt and equilibration time should be considered when planning toxicity tests. 相似文献
