逆胶束体系中纤维素酶解特性研究   总被引:3,自引:1,他引:3  
通过采用几种不同属性类型的表面活性剂构建逆胶束体系,以羧甲基纤维素为底物,研究了纤维素酶解的特性.通过电导率的测量,确定由CTAB、SDS、Tween-80和鼠李糖脂构造的逆胶束体系的最大增溶水量W0(W0=[H2O]/[SA])分别为15.2、20.1、2.3和40.3.在此条件下,考察了不同表面活性剂浓度和不同纤维素酶用量对逆胶束体系酶解行为的影响,并与水溶液体系的酶解反应进行对比.结果表明,单位底物条件下,纤维素酶用量为0.15 FPU/g,CTAB、SDS、Tween-80和鼠李糖脂浓度均为1 cmc时,逆胶束体系酶解产量最高,其中鼠李糖脂体系产量可达198.03 mg,分别比CTAB、SDS和Tween-80体系高了10.89%、31.09%和45.30%.CTAB、SDS、Tween-80和鼠李糖脂浓度为1 cmc时,其逆胶束体系酶解产量比水溶液体系分别高了34.36%、21.24%、11.44%和34.62%.在最佳表面活性剂浓度条件下单位底物酶用量为5 FPU时,逆胶束体系酶解反应从经济和糖产量方面考虑均为最适合.生物表面活性剂鼠李糖脂体系的产量高于3种化学表面活性剂体系,说明生物表面活性剂在构建逆胶束体系及增强逆胶束稳定性能上具有特殊的潜力.  相似文献

The influence of the nonionic surfactant Tween 80 on pentachlorophenol (PCP) oxidation catalyzed by horseradish peroxidase was studied. The surfactant was tested at concentrations below and above its critical micelle concentration (CMC). Enhancement of PCP removal was observed at sub-CMCs. The presence of Tween 80 in the reaction mixture reduced enzyme inactivation which occurred through a combination of free radical attack and sorption by precipitated products. A simple first-order model was able to simulate time profiles for enzyme inactivation in the presence or absence of Tween 80. At supra-CMCs, the surfactant caused noticeable reductions in PCP removal, presumably through micelle partitioning of PCP which precluded the hydrophobic PCP molecule from interacting with the enzyme.  相似文献

从降解菌YF11中提取了降解权衡利弊并测定了该降解酶对对硫磷,甲基对硫磷和杀螟松的降解特性。对硫磷和甲基对硫磷酶促降解的最适pH均为8.0,杀螟松的最适pH为8.5,最适温度为32.5℃,该降解酶对对硫磷,甲基对硫磷和杀螟松的米氏常数分别为187.2,205.5和212.4mmol/ml。  相似文献

黄土丘陵区撂荒农田土壤酶活性及酶化学计量变化特征   总被引:3,自引:6,他引:3  
明确土壤胞外酶活性及酶化学计量比在农田撂荒过程中的变化特征及驱动因素,对揭示土壤养分可利用性随植被恢复的变化规律和阐明生态系统养分循环机制有重要意义.本文以黄土丘陵区不同撂荒年限坡耕地(0、10、20和30 a)为研究对象,通过测定土壤酶活性及酶计量比、土壤理化性质、植物多样性和科组成,探讨农田撂荒后土壤微生物养分限制状况及酶活性和酶计量比的主控因子.结果表明,β-1,4-N-乙酰氨基葡萄糖苷酶(NAG)、亮氨酸氨基肽酶(LAP)和碱性磷酸酶(ALP)的活性均随撂荒年限增加而显著增加,而β-1,4-葡萄糖苷酶(BG)活性则相反.土壤BG:(NAG+LAP)和BG:ALP呈现出与土壤BG活性相同的随撂荒年限增加而减小的趋势;而(NAG+LAP):ALP呈先升后降趋势,在撂荒20 a时达到最高值.酶化学计量的向量长度随撂荒年限增加而显著减小,表明撂荒过程中土壤微生物受碳(C)限制程度的减弱.撂荒前10 a酶化学计量的向量角度小于45°,后20 a大于45°,说明撂荒前期微生物受氮(N)限制,后期受磷(P)限制.冗余分析结果揭示土壤有机碳含量、总氮含量、C:N、C:P、pH以及植被多样性是调控酶活性和酶计量比变化的主要因子.分类变异分析表明,土壤和植物因素可解释不同撂荒年限下土壤酶活性及酶计量比差异的62.0%,植被特征与土壤理化特性之间的交互作用是土壤酶活性及酶计量比变化的主控因子,解释度为37.1%.综上所述,黄土丘陵区农田撂荒过程中应考虑外源P的投入以缓解生态系统中有效P的不足,研究结果可为理解脆弱生境恢复生态系统中微生物介导的生物地球化学循环机制以及指导土壤养分管理和生态可持续发展提供理论依据.  相似文献

测定了降解菌Alcaligenes sp.YF 11粗酶液对拟除虫菊酯杀虫剂的降解性能,粗酶液降解杀灭菊酯、甲氰菊酯、氯氯菊酯、溴氰菊酯氯菊酯、三氟氯氰菊酯的最适pH分别为8.0、8.5、8.0、8.0、9.0、7.5;米氏常数分别为41.4、136.8、65.4、222.8、5.2、8.67nmol/ml;最适温度为32.5℃,该粗酶液在pH5.0-10.5均具有降解活性。  相似文献

以汽爆稻草秸秆为原料,通过将几个酶解罐串联来提高最终还原糖浓度.考察了酶浓度,酶解单元组成以及稀释率对汽爆稻草秸秆酶解的影响.结果表明,最佳条件为:20FPU/g,酶解单元由4个酶解罐组成,稀释率为0.075/h.在此条件下,酶解时间为24h时,与传统批次酶解相比,汽爆稻草秸秆总转化率从18%~21%提高到39.5%.与只有1个酶解罐的膜反应器相比,每g底物还原糖产量从0.25g提高到0.4g,最终所得还原糖的平均浓度从4.56g/L提高到27.23g/L.  相似文献

金属酶能显著提高甲烷菌的生长速度及产甲烷活性。通过小试和工程应用,其作用在产气率、气体甲烷含量、容积负荷和COD去除率的增加上都得到证实。试验得出:当TOC和COD分别约为1.0×10~4mg/L和2.7×10~4mg/L时,金属酶添加量为26.3ml/kg TOC较宜,此浓度下甲烷菌体增殖速度最快,活性最高。  相似文献

The purpose of this study was to examine the natural history and differential diagnosis of ultrasound-detected, isolated fetal ascites. Retrospective review of our patient data base, from 1989 to 1993, revealed 18 patients with fetal ascites diagnosed sonographically. Fetuses presenting with generalized hydrops were excluded. One of the 18 fetuses with ascites had a chromosomal abnormality (trisomy 21), four fetuses had intrauterine infections, seven had gastrointestinal processes, two had genitourinary tract abnormalities, and four were labelled as ‘idiopathic’ (all four resulting in normal neonates). Seventeen of 18 fetuses survived; there was one fetal demise secondary to active syphilis. One fetus with parvovirus infection required intrauterine transfusion and did well. Two infants are developmentally retarded, including one with trisomy 21 and one with microcephaly secondary to cytomegalovirus infection. Fourteen of 18 fetuses had documented in-utero resolution of the ascites. Eleven of the 18 were associated with polyhydramnios sometime during fetal life. None of the fetuses developed hydrops. In conclusion, fetal ascites can result from many different aetiologies, including gastrointestinal and genitourinary anomalies. Chromosomal abnormalities and viral aetiologies must also be considered. Fetuses who have isolated ascites can have a good outcome with resolution of the ascites antenatally.  相似文献

Natural and synthetic estrogens from sewage treatment systems are suspected to influence the reproductive health of the animals in the rivers.In this article we investigated the enzymatic treatment of three estrogens (estrone,17β-estradiol,and 17α-ethynyletstradiol) by a fungal laccase which oxidize phenolic compounds with dissolved oxygen.The elimination of the estrogenic activities by enzymatic oxidation was demonstrated by medaka vitellogenin assay.In addition,we developed an enzymatic treatment system comprised of β-D-glucuronidase and the laccase for 17β-estradiol 3-(β-D-glucuronide) degradation.The two enzymes eliminated 17β-estradiol 3-(β-D-glucuronide) and the intermediate,17β-estradiol,efficiently.  相似文献
