Efficacy of Goniothalamin a styryl-pyrone isolated from Goniothalamus wightii Hook.f. and Thoms, against beet armyworm, Spodoptera exigua (Hübner), populations was determined under laboratory condition. The experiments were carried out with concentrations of 5, 10, 15 and 30 ppm in an artificial diet and compared with control insects. Laboratory bioassay showed that the goniothalamin had a strong effect on food utilization, moulting and gut histology. The food consumption and conversion of ingested and digested food to body matter decreased with increasing pyrone concentration. The antifeedant activity was also observed in larvae of S. exigua. The treated third instar larvae exhibited mortality in a dose dependent manner. At 5, 10, 15 and 30 ppm/insect, the pyrone gave 23%, 45% 63% and 100% mortality respectively. The larvae of S. exigua gained significantly less weight until pupation in the 10 and 15 ppm pyrone concentrations. Duration of larval period was also affected after treatment with pyrone. The metamorphosis was delayed with additional moulting (7th instar) after treatment with 10 and 15 ppm of goniothalamin, the percentage of larvae successfully moulted into progressive instars was significantly decreased with an increase in pyrone concentrations. The effects of goniothalamin on midgut ultrastructure of third instar larvae of S. exigua were investigated by using light microscopy. Cross sections of the midgut showed that the epithelial cells were destroyed. Significant damage of the midgut epithelium was observed along with lysis.  相似文献

Effects of ozone fumigation of Asclepias curassavica L. and A. syriaca L. on feeding preference, growth, development, and nutritional indices of monarch larvae were investigated in conjunction with changes in specific leaf metabolites. While foliar chemistry was quite variable, fumigation generally decreased sugars and proteins, and increased amino acids and phenolics in A. curassavica. Effects were similar in A. syriaca except that sugars were generally increased while amino acids were usually not affected. On A. curassavica, 3rd instar larvae preferred ozone-treated leaves while 4th instars showed no preference; conversely, on A. syriaca, 3rd instars showed no preference while 4th instars preferred control leaves. Relative growth rate and relative consumption rate of 5th instars were greater on fumigated plants of both species, but other nutritional indices were unaffected. Larvae developed more rapidly on intact fumigated plants of both species than on the respective controls. The results suggest that enhanced feeding stimulation may be the primary cause of the altered behavior and performance on ozone-fumigated plants.  相似文献

Food utilization and growth of the 5th and 6th instar Spodoptera litura Fabricius larvae, and its effect on reproduction potential was evaluated by feeding larvae diets with different doses of Ni for 3 generations. Dose-dependent relationships between Ni levels and food consumption and growth were variable with different larval developmental period and Ni exposure duration. RCR, AD and RGR of the 6th instar larvae were much more affected by Ni exposure than those of 5th instar larvae, and the effects were strongest in the 3rd generation. It was found that RCR was significantly stimulated after 1 and 20 mg kg−1 Ni exposure, while AD was significantly inhibited after 1, 5, 10 and 40 mg kg−1 Ni exposure. However, lower levels of Ni (?5 mg kg−1) significantly increased and higher levels of Ni (?10 mg kg−1) significantly decreased RGR. In 3 successive generations, 10 mg kg−1 Ni significantly increased the ECI and ECD of the 5th instar larvae, and 5 mg kg−1 Ni significantly increased the ECD of the 6th instar larvae. However, ECD were all significantly inhibited with 20 mg kg−1 Ni exposure. Results also revealed that durations of larvae were shortened at low levels of Ni, but extended at high levels of Ni. Fecundity was inhibited by the highest Ni doses in each generation, while improved by low Ni doses in the 3rd generation. Hatching rates in all treatments were significantly decreased in a Ni dose-dependent manner. Study indicated that effects of Ni on these parameters were predominant with the increasing Ni exposure period.  相似文献

Abstract

In a series of recent studies, we evaluated the influence of delivery systems, drop size and application timing on the efficacy of aerially applied Bacillus thuringiensis against gypsy moth infestations. Use of different nozzle systems including Micronair, Flat Fan or Twin Jet, did not appear to result in significant differences in Bt coverage efficiency, foliage protection or population reduction. Nor was there any significant difference in population reduction when Bt was applied at two different drop sizes with volume median diameters of 110 and 163 μm. The efficacy of different formulations on larval populations were similar when used against the younger 1st and 2nd instar but differed when treatment was delayed until the population matured to the 3rd and 4th instar stages.  相似文献

Cadmium, like many other pollutants, is nondegradable and can be accumulated by Lymantria dispar at a level that affects fitness components, physiology, and development, which could indicate presence of environment pollution by heavy metals. The cadmium effect on fitness-related traits in the third, fourth, fifth, and sixth instar of L. dispar L. was determined. Furthermore, activities of the following antioxidative defense components after the larvae had been fed on the artificial cadmium-supplemented diet (50 μg Cd/g dry food) were assessed: superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APOX), total glutathione amount (GSH), glutathione-S-transferase (GST), glutathione reductase (GR), and the amount of free sulfhydryl (SH) groups. Statistically significant delay of development in the fourth, fifth, and sixth instar and decrease of the larval mass in the third and fourth instar were estimated after the exposure to cadmium through food in comparison to the control. There were no changes in SOD activity of cadmium-treated larvae. Significantly lower CAT, APOX, and GR activities were recorded in the third, fifth, and in the third instar, respectively. At the same time, higher activity was recorded in the sixth instar, while GST activity was higher in the third. GSH content was significantly lower during all instars after treatment but the amount of SH groups was higher in older larvae. The strategy of antioxidative defense and the adjustment or modulation of fitness-related traits in presence of cadmium was dependent on the age of larvae in L. dispar, which might be used in early metal risk assessment in Lepidoptera and other insects.  相似文献

This study was conducted to investigate the potential use of biomarkers in bioassays with chironomids to assess contamination by pesticides in temperate and tropical climates. Two species of midge were studied, the widespread Chironomus riparius and the tropical Kiefferulus calligaster (Kieffer, 1911). Preliminary studies included investigations of the effects of temperature on larval development and the influence of larval age on normal variability of cholinesterase (ChE) and glutathione S-transferase (GST) activities and protein content. In the second phase, the influence of two abiotic factors particularly important in tropical conditions (temperature and oxygen concentration) and of the organophosphorous (OP) insecticide dimethoate on biochemical and conventional endpoints was investigated. Results showed that K. calligaster is morphologically and physiologically similar to C. riparius and for both, the time of larval development decreases with the increase of temperature. Moreover, 3rd and 4th instars appeared to be the most suitable for biomarkers determinations. ChE activity seems to be valuable biomarker regarding temperature and dissolved oxygen (DO) variations, while some caution should be taken when using GST as an environmental biomarker, since it shows some dependence of these parameters. C. riparius was more sensitive to dimethoate than K. calligaster suggesting that the use of bioassays with the former species in tropical conditions may overestimate the toxicity of OP pesticides to autochthonous species. When testing sub-lethal effects of dimethoate to C. riparius, ChE activity showed to be a very sensitive parameter detecting significant effects at the lowest concentration that caused emergence delay of larvae, suggesting that it is an ecologically relevant parameter.  相似文献

Ingestion of the bacterial toxins Bacillus thuringiensis (Berliner) sub sp kurstaki, neem seed kernel extract and Vitex negundo L. (Lamiales:Verbenaceae) leaf extract to the rice leaffolder (RLF), Cnaphalocrocis medinalis (Guenée) (Lepidoptera: Pyralidae), resulted in altered leaf-folding behaviour and biology. In laboratory experiments, treatment with plant extracts and bacterial toxins suppressed leaf folding behaviour of C. medinalis. Larvae fed lower doses moulted into progressive instars, but in the late fourth to early fifth instars, spinning behaviour completely stopped, feeding was reduced and larvae regurgitated a sticky, semisolid substance that occluded the tunnel in which the pupae were formed. With the combination of Btk and botanicals, average leaf consumption was decreased by a factor of two even at reduced concentrations when compared with controls. Duration of larval and pupal stages, adult longevity and fecundity were more affected by treatment with the combination of bacterial toxins and both botanicals than by the treatment with the bacterial toxins or botanicals individually. All larvae died in the treatment of bacterial toxins and both botanicals combined.  相似文献

The goal of this study was to contribute to understanding of the mechanisms behind sensitivity differences between early and late instar larvae of Chironomus riparius and to address the influence of the differences in standard testing approaches on the toxicity evaluation. A 10-day contact sediment toxicity test was carried out to assess sensitivity to cadmium exposure in relation to different age and laboratory culture line origin of test organisms. Chironomid larvae of early (OECD 218 method) and late instar (US-EPA600/R-99/064 method) differed substantially in sensitivity of traditional endpoints (OECD: LOEC 50 and 10 μg Cd/g dry weight (dw); US-EPA: LOEC?>?1000 and 100 μg Cd/g dw for survival and growth, respectively). Bioaccumulated cadmium and metallothioneins (MTs) concentrations were analyzed to investigate the role of MTs in reduced sensitivity to cadmium in late instar larvae. Metallothioneins were induced after treatment to greater Cd concentrations, but their levels in relation to cadmium body burdens did not fully explain low sensitivity of late instars to cadmium, which indicates some other effective way of detoxification in late instars. This study brings new information related to the role of MTs in age-dependent toxicant sensitivity and discusses the implications of divergence in data generated by chironomid sediment toxicity tests by standardized methods using different instars.  相似文献

In laboratory experiments a cladoceran, Daphnia ambigua, was exposed for six instars to the insecticide carbaryl at five different concentrations (1, 2, 3, 4, 5 microg litre(-1)) or received short-term (10-h) exposure to 5 microg carbaryl litre(-1) at different life stages of the animal. In the long-term exposure experiments carbaryl at 1 microg litre(-1) did not have any detectable effects on the animals, whereas 2 microg litre(-1) reduced the survival, growth and reproduction of the animals, and higher concentrations killed all the animals examined before they grew to the fifth instar. These results indicate that the range of sublethal concentrations of carbaryl for D. ambigua is narrow. In the short-term exposure experiments the animals did not suffer any damage when they were exposed to the chemical during the egg stage. However, their growth and egg production were reduced when the short-term exposure was conducted during the life stages from the final embryo stage to the third instar. The effect was most marked when the animals were treated during the first instar, indicating that this is the stage when the animals are most sensitive to the chemical.  相似文献

Abstract

Final (sixth) instar larvae of Spodoptera litura were fed in increasing amounts of azadirachtin and the effects on feeding activity, dietary utilization and mid‐gut carboxylesterase activity were determined. The antifeedant activity of azadirachtin did not increase in a dose‐dependent manner as reflected by the decline in antifeedant effect at higher doses of 30 and 50 ppm. Dietary concentrations of azadirachtin although significantly lowered the efficiencies of conversion of both ingested (ECI) and digested (ECD) food, it failed to lower the approximate digestibility (AD). The latter however, increased significantly with increasing concentrations of azadirachtin. Feeding of higher doses (30 and 50 ppm) resulted in significant increase in midgut carboxylesterase activity. Thus insensitivity to antifeedant effects and induction of midgut carboxyesterase activity in the older larvae of S.litura In response to high doses of azadirachtin observed in this study can have adverse consequences unless particular attention is paid in developing application strategies for azadirachtin based biopesticides for use in pest control protocols.  相似文献

Meliaceae plant products have been shown to exert pesticidal properties against a variety of insect species. In agricultural pest control programs, such products may have the potential to be used successfully as botanical insecticides. The effect of the neem (Azadirachta indica) limonoids azadirachtin, salannin, deacetylgedunin, gedunin, 17-hydroxyazadiradione and deacetylnimbin on the biology and mortality of rice leaffolder larvae was investigated. In laboratory experiments, treatment with neem limonoids suppressed leaf folding behaviour of C. medinalis. Biological parameters (larval duration, pupal duration adult longevity and fecundity) were also affected by the treatment. Azadirachtin, salannin, and deacetylgedunin showed high bioactivity at all doses, while the rest of the neem limonoids were less active, and were only biologically active at high doses. Azadirachtin was most potent in all experiments and produced almost 100% larval mortality at 1 ppm concentration. These results indicate neem limonoids affect the larval behaviour. These effects are most pronounced in early instars.  相似文献

The effects of exposure of sugar maple (Acer saccharum Marsh.) to ozone on the entire larval stage of a native insect have not been previously investigated. This study reports the effects of sugar maple seedlings exposed to different ozone concentrations on the relative performance and the feeding preference of the forest tent caterpillar (Malacosoma disstria Hbn.). Three-year-old seedlings were set in nine open-top field chambers in the spring of 1992 and 1993. Three ozone concentrations were generated: charcoal-filtered ambient air (0x), ambient air (1x) and three times ambient air (3x). In 1992, female and male larval development time did not differ among ozone treatments. In 1993, female larvae reared on 3x developed faster than those on 0x and 1x, while male larvae were not affected. Ozone treatments did not influence pupal weights except for males in 1993 where pupae reared on 0x were heavier than 1x but did not differ from 3x. Larval and pupal survival rates were not affected by ozone in either year. Finally, 4th and 5th instar larvae showed a significant feeding preference for 3x foliage in 1993 but not in 1992. The response of the forest tent caterpillar to ozone exposed seedlings varied between years and could be more sensitive to annual climatic variations than ozone.  相似文献

Gene expression responses of paper birch (Betula papyrifera) leaves to elevated concentrations of CO2 and O3 were studied with microarray analyses from three time points during the summer of 2004 at Aspen FACE. Microarray data were analyzed with clustering techniques, self-organizing maps, K-means clustering and Sammon's mappings, to detect similar gene expression patterns within sampling times and treatments. Most of the alterations in gene expression were caused by O3, alone or in combination with CO2. O3 induced defensive reactions to oxidative stress and earlier leaf senescence, seen as decreased expression of photosynthesis- and carbon fixation-related genes, and increased expression of senescence-associated genes. The effects of elevated CO2 reflected surplus of carbon that was directed to synthesis of secondary compounds. The combined CO2 + O3 treatment resulted in differential gene expression than with individual gas treatments or in changes similar to O3 treatment, indicating that CO2 cannot totally alleviate the harmful effects of O3.  相似文献

Sustained cultivation of Bacillus thuringiensis (Bt) transgenic crops requires stable transgene expression under variable abiotic conditions. We studied the interactions of Bt toxin production and chronic ozone exposure in Bt cry1Ac-transgenic oilseed rape and found that the insect resistance trait is robust under ozone elevations. Bt Cry1Ac concentrations were higher in the leaves of Bt oilseed rape grown under elevated ozone compared to control treatment, measured either per leaf fresh weight or per total soluble protein of leaves. The mean relative growth rate of a Bt target herbivore, Plutella xylostella L. larvae was negative on Bt plants in all ozone treatments. On the non-transgenic plants, larval feeding damage was reduced under elevated ozone. Our results indicate the need for monitoring fluctuations in Bt toxin concentrations to reveal the potential of ozone exposure for altering dosing of Bt proteins to target and non-target herbivores in field environments experiencing increasing ozone pollution.  相似文献

The effect of copper on the activity of Balbiani rings (BR1 and BR2) and nucleolar organizing region (NOR) in chromosome IV of the salivary gland of the 4th instar larvae of Chironomus ninevah has been investigated. Sublethal concentrations, i.e. 0.02, 0.1, 0.15 and 0.2 mg litre(-1) suppress the activity of BR1, and are statistically significant. The same concentrations reduced the activity of BR2 and NOR, but without significant differences, except 0.1 mg litre(-1) which was significant. The purpose of this work was to evaluate the effect of chronic toxicity of Cu(2+) on the expression of gene activity of midge salivary gland chromosomes Although there may be many more aquatic organisms that are more sensitive to copper, this midge was selected as a biological indicator species because of its association with benthic sediments.  相似文献

Effect of active molluscicidal components of Sapindus mukorossi and Terminalia chebula on the acetylcholinesterase (AChE), acid and alkaline phosphatase (ACP/ALP) activity in the nervous tissue of freshwater snail Lymnaea acuminata were studied. In vivo and in vitro exposure of saponin (active component of S. mukorossi pericarp) and tannic acid (active component of T. chebula) significantly inhibited the AChE, ACP and ALP activity in the nervous tissue of L. acuminata. The inhibition kinetics of these enzymes indicate that saponin and tannic acid caused competitive and competitive-non-competitive inhibition of AChE, respectively. Saponin also caused competitive and competitive-non-competitive inhibition of ACP and ALP, respectively, whereas tannic acid caused competitive-non-competitive inhibition of ACP and ALP. Thus the inhibition of AChE, ACP and ALP by saponin and tannic acid in the nervous tissue of L. acuminata may be the cause of molluscicidal activity of S. mukorossi and T. chebula.  相似文献

Mosquitoes transmit dreadful diseases, causing millions of deaths every year. Therefore, screening for larvicidal and pupicidal activity of microbial extracts attributes could lead to development of new and improved mosquito control methods that are economical and safe for nontarget organisms and are ecofriendly. Synthetic chemical insecticides occupy predominant position in control strategies. These hazardous chemicals exert unwarranted toxicity and lethal effects on nontarget organisms, develop physiological resistance in target, and cause adverse environmental effect. For vector control, fungal-mediated natural products have been a priority in this area at present. In the current study, effective larvicidal and pupicidal effect of mycosynthesized silver nanoparticles (Ag NPs) using an entomopathogenic fungi Trichoderma harzianum against developmental stages of the dengue vector Aedes aegypti was investigated. An attractive possibility of green nanotechnology is to use microorganisms in the synthesis of nanosilver especially Ag NPs. The mycosynthesized Ag NPs were characterized to find their unique properties through UV-visible spectrophotometer, X-ray diffraction analysis, Fourier transform infrared, and surface characteristics by scanning electron microscopy. To analyze the bioefficacy, different test concentrations for extracellular filtrate (0.2, 0.4, 0.6, 0.8, and 1.0 %) and Ag NPs (0.05, 0.10, 0.15, 0.20, and 0.25 %) were prepared to a final volume of 200 mL using deionized water; 20 larvae of each instars (I–IV) and pupa were exposed to each test concentration separately which included a set of control (distilled water) group with five replicates. Characterization of the synthesized Ag NPs were about 10–20 nm without aggregation. Susceptibility of larval instars to synthesized Ag NPs was higher than the extracellular filtrate of T. harzianum alone after 24-h exposure, where the highest mortality was recorded as 92 and 96 % for first and second instars and 100 % for third, fourth instars, and pupa. Lethal concentration 50 values of 0.079, 0.084, 0.087, 0.068, and 0.026 % were recorded for I–IV instars and pupa, respectively, when exposed to Ag NPs at 0.25 % concentration. Toxicity was exhibited against first (1.076 %), second (0.912 %), third (0.770 %), fourth (0.914 %) instars larvae, and pupa (0.387 %) with extracellular filtrate at a concentration of 1 % that was three- to fourfold higher compared to Ag NPs; no mortality was observed in the control. The present study is the first report on effective larvicidal and pupicidal activity of Ag NPs synthesized from an entomopathogenic fungi T. harzianum extracellular filtrate and could be an ideal ecofriendly, single-step, and inexpensive approach for the control of A. aegypti.  相似文献
