17β-estradiol (E2) is a natural estrogenic hormone found in animal manure and urine, which may cause endocrine disruption in sensitive organisms. 17β-Estradiol has been widely detected in the environment, and animal agriculture may be an important source. The objectives of this study were to investigate the potential sources and/or spatial and temporal characteristics contributing to detections of E2 at the farm-scale. Soil cores, segmented into 0.15 m increments, were taken down to the water table from four locations (three potential E2 sources and one control) at or around a swine (Sus scrofa domesticus) farm on five different dates between 2006 and 2007. Estradiol was quantified in the soil-water extracts using liquid chromatography-with tandem mass spectrometry (LC/MS/MS) analysis. Estradiol detections were widespread and found in 128 out of 345 extractions (37%). Concentrations ranged from 0 to 1910 ng L−1 (porewater equivalents). A location that received field application of manure had significantly lower E2 concentrations compared to other locations. Also, the spring 2007 E2 concentrations were significantly higher than all other sample dates, perhaps related to climatic and hydrological events. Results suggested E2 was not directly related to manure sources, but was widespread in this environment. Where E2 was detected, highest concentrations favored the upper profile, while the greatest frequency of detections was in the lower profile and near the water table. Detections of E2 were associated with high organic mater contents in the upper profile and high sand contents in the lower profile. The study suggests that E2 is widespread in some soil environments, raising questions as to the source and mechanisms that facilitate its transport and mobility.  相似文献

A study using 17β-14C4-estradiol (14C-E2) was performed to confirm and characterize the catalytic transformation of estrogens in the presence of a model vegetable matter (namely rabbit food) as a surrogate material for vegetable wastes found in sewage. Results corroborated the occurrence of an abiotic transformation. Unknown transformation byproduct(s) accounted, respectively, for 38% and 9% of the initial radioactivity in liquid and extractable solid phases after 72 h; on the other hand, only 15% and 7% of this radioactivity corresponded to 14C-E2 in those same matrices. Mass balance was closed including the radioactivity irreversibly bounded to the solid phase. Formation of 14C4-estrone was monitored by Liquid Chromatography with tandem Mass Spectrometry detection; negative results were found in all sampling events. This process could be harnessed to optimize sustainable technologies for the removal of phenolic microcontaminants from wastewater.  相似文献

The removal of 17β-estradiol (E2) by biodegradation and sorption onto humic acid (HA) was examined at various HA concentrations. Subsequently, estrogenicity associated with E2 removal was estimated using E-screen bioassay. Results showed that E2 biodegradation and its subsequent transformation to estrone (E1) were significantly reduced with increasing HA concentration. In addition, the presence of nutrients enhanced the biodegradation of E2. Overall, E2 biodegradation was the dominating contributor to its removal, which demonstrated a significantly negative correlation with E2 sorption at various HA concentrations. The sorption of E2 by HA was significantly enhanced with increasing HA concentration. Estrogenicity associated with residual E2 showed that there existed a significant difference among various HA concentrations, with the lowest value in the absence of HA. The findings suggest that the presence of HA and nutrients in natural waters should be considered in assessing estrogenicity of environmental samples due to complex sorption and biodegradation processes.  相似文献

Natural steroid estrogens (e.g., 17 β-estradiol, E2), synthetic steroid estrogens (e.g., 17 α-ethinylestradiol, EE2) and pharmaceutical antibiotics (e.g., ciprofloxacin) are chemicals detected in biosolids and sewage sludges because they partition into the solids fraction during the wastewater treatment process. This research utilized a three-way factorial design (six media × two estrogens × three antibiotic treatments) to quantify cumulative E2 and EE2 mineralization over 133 d (MAX) in a range of sewage sludge and biosolid samples in the presence (4 and 40 mg kg?1) and absence of ciprofloxacin. The same three-way factorial design was utilized to quantify the impact of the six media, E2 or EE2, and ciprofloxacin on cumulative soil respiration over 133 d (RESP). Minimal ciprofloxacin mineralization was observed (<0.05% over 133 d), but despite its persistence, ciprofloxacin had no significant effect on MAX of E2 or EE2, and, in general, no significant effect on RESP. MAX ranged from 38.38% to 48.44% for E2 but from only 0.72% to 24.27% for EE2 although RESP was relatively similar, ranging from 101.00 to 866.54 mg CO2 in the presence of E2 and from 69.55 to 893.95 mg CO2 in the presence of EE2. The sorption-limited bioavailability of EE2, which is inherently resistant to biodegradation due to chemical structure, as MAX and Freundlich sorption coefficients (Kf) were negatively correlated. As such, the Kf values of EE2 were largest in composted biosolids in which EE2 was particularly resistant to microbial degradation as the MAX of EE2 was <3%. In contrast, the MAX of E2 showed a positive association with the Kf values of E2 because some steps in the E2 transformation process have been found to occur in the sorbed phase. The MAX of E2 was significantly greater in the biosolid and composted biosolid media than in any other media, whereas the MAX of E2 decreased in the following order: secondary sewage sludge > primary sewage sludge > biosolid = composted biosolid. This suggests that sewage sludges in municipal lagoons and pre-treatment holding lagoons are a more favorable media for mineralization of EE2, whereas biosolids in post-treatment storage lagoons are a more favorable media for the mineralization of E2. The presence of ciprofloxacin will have no impact on the potential E2 or EE2 mineralization rates in these cases.  相似文献

The degradation of 17β-estradiol (E2) in sterile soil and Mn-free soil slurries was determined. In 0.075 g ml−1 soil slurry, E2 with an initial concentration of 0.0267 μmol g−1 was rapidly degraded and near equimolar estrone (E1) accumulated. A mass balance involving E2 and E1 existed throughout the reaction. The E2 degradation was thus an oxidation process and E1 was the only product. The concurrent release of Mn(II) during E2 oxidation and a lack of E2 oxidation in Mn-free soil slurry together demonstrated that soil manganese oxides were responsible for E2 oxidation. The degree of E2 oxidation was higher at high pH than at low pH, consistent with the fact that the reaction released protons. This study suggests that manganese oxides may be used as soil amendments to effectively oxidize E2 to less potent E1 in soil.  相似文献

In streams, chemicals such as 17β-estradiol (E2) are likely to occur in pulses. We investigated uptake and biomarker responses in juvenile brown trout (Salmo trutta) of 3- or 6-h pulses of concentrations up to 370 ng E2 L−1. Uptake by the fish was estimated from disappearance of E2 from tank water. A single 6-h pulse of 370 ng E2 L−1 increased the plasma vitellogenin concentration, liver Erα- and vitellogenin-mRNA. Exposure to 150-160 ng E2 L−1 for 6 h increased vitellogenin in one experiment but not in another. Two 6-h pulses had a larger effect one pulse. Brown trout in the size range 24-74 g took up E2 linearly with time and exposure concentration with a concentration ratio rate of 20.2 h−1. In conclusion, the threshold for induction of estrogenic effects in juvenile brown trout at short term pulse exposure appears to be in the range 150-200 ng E2 L−1.  相似文献

Natural steroidal estrogens, such as 17 β-estradiol (E2), as well as antimicrobials such as doxycycline and norfloxacin, are excreted by humans and hence detected in sewage sludge and biosolid. The disposal of human waste products on agricultural land results in estrogens and antibiotics being detected as mixtures in soils. The objective of this study was to examine microbial respiration and E2 mineralization in sewage sludge, biosolid, and soil in the presence and the absence of doxycycline and norfloxacin. The antimicrobials were applied to the media either alone or in combination at total rates of 4 and 40 mg kg?1, with the 4 mg kg?1 rate being an environmentally relevant concentration. The calculated time that half of the applied E2 was mineralized ranged from 294 to 418 days in sewage sludge, from 721 to 869 days in soil, and from 2,258 to 14,146 days in biosolid. E2 mineralization followed first-order and the presence of antimicrobials had no significant effect on mineralization half-lives, except for some antimicrobial applications to the human waste products. At 189 day, total E2 mineralization was significantly greater in sewage sludge (38 ±0.7%) > soil (23 ±0.7%) > biosolid (3 ±0.7%), while total respiration was significantly greater in biosolid (1,258 mg CO2) > sewage sludge (253 mg CO2) ≥ soil (131 mg CO2). Strong sorption of E2 to the organic fraction in biosolid may have resulted in reduced E2 mineralization despite the high microbial activity in this media. Total E2 mineralization at 189 day was not significantly influenced by the presence of doxycycline and/or norfloxacin in the media. Antimicrobial additions also did not significantly influence total respiration in media, except that total CO2 respiration at 189 day was significantly greater for biosolid with 40 mg kg?1 doxycycline added, relative to biosolid without antimicrobials. We conclude that it is unlikely for doxycycline and norfloxacin, or their mixtures, to have a significant effect on E2 mineralization in human waste products and soil. However, the potential for E2 to be persistent in biosolids, with and without the presence of antimicrobials, is posing a challenge for biosolid disposal to agricultural lands.  相似文献

This study characterizes the effect of oxygen in the abiotic transformation of estrogens when they are contacted with a surrogate of the vegetable wastes found in sewage. 17β-Estradiol (E2) and 17β-14C4-estradiol (14C-E2) were utilized as model compounds. Batch experiments were run under both oxic and anoxic conditions. In order to accomplish an accurate mass balance of the target estrogen, two analyses were performed simultaneously: first, radioactivity counting, and second, quantitation of E2 and 14C-E2, as well as their transformation product estrone and 14C4-estrone, by Liquid Chromatography tandem Mass Spectrometry. Under oxic conditions, the total concentration of 14C-E2 was found to decrease by 78% in 72 h (15% and 7% remained in the liquid and solid phases, respectively). Conversely, when the estrogens were contacted with the synthetic influent under anoxic conditions, E2 was quantitatively recovered after 72 h (70% and 22% in aqueous and solid matrices, correspondingly). These results suggest that when the concentration of dissolved oxygen is null or limited, catalysis through an oxidative coupling mechanism is halted. Moreover, it was confirmed that the catalytic reaction occurred solely in the presence of the solid phase of the model vegetable matter.  相似文献

Purpose

Hydroxylated polybrominated diphenyl ethers (OH-PBDEs) have emerged as contaminants of environmental concerns because they pose potential risks to human and animal health. The purpose of this study was to investigate the in vitro metabolism of OH-PBDEs and their potential inhibition against 17??-estradiol (E2) metabolism.

Methods

Rat liver microsomes were used as a source of P450 enzymes in an in vitro metabolism study of OH-PBDEs. Inhibition of E2 metabolism and kinetic study were performed by incubating with rat liver microsomes in the presence of OH-PBDEs.

Results

The obtained data clearly demonstrated that OH-PBDEs, especially those congeners with lower bromination, could be metabolized to bromophenol and diOH-PBDEs. The less metabolic rate of OH-PBDEs was observed with the increasing number of bromine substituents. OH-PBDEs with hydroxyl group and bromine adjacent to the ether bridge showed faster metabolic rates. In addition, the results showed non-competitive inhibition of E2 metabolism by OH-PBDEs with IC50 values in the range from 13.7 to 55.2???M. The most potent OH-PBDE inhibitor was found to be 3??-OH-BDE-100. The inhibitory potencies for OH-PBDEs were significantly higher than those of parent PBDE and methoxylated metabolites, providing the evidence that PBDEs exerted estrogenic activity in part by their hydroxylated metabolites.

Conclusions

OH-PBDEs exhibited large differences in their capacity to be metabolized and to inhibit E2 metabolism in rat liver microsomes. The finding might increase our understanding of healthy risk associated with PBDEs in human and wildlife.  相似文献

To examine the impact of long-term solid beef cattle (Bos Taurus) manure on soil properties and 17β-estradiol sorption and mineralization, this study utilized a soil that had received beef cattle manure over 35 years. The 17β-estradiol was strongly sorbed and sorption significantly increased (P < 0.05) with increasing soil organic carbon content (SOC) and with an increasing annual rate of beef cattle manure. The 17β-estradiol mineralization half-life was significantly negatively correlated, and the total amount of 17β-estradiol mineralized at 90 days (MAX) was significantly positively correlated with 17β-estradiol sorption. The long-term rate of manure application had no significant effect on MAX, but the addition of fresh beef cattle manure in the laboratory resulted in significantly (P < 0.05) smaller MAX values. Over the past decade, the environment has been polluted by a wide spectrum of exogenous chemicals and environmental analysis has become one of the most progressive parts of analytical research. The aim of this work was to determine the kinetics of natural degradation, and to identify the degradation products of the massively used estrogenic drug, 17-α-ethinylestradiol. The photodegradation, oxidation and thermostability conditions were selected according to ICH requirements for pharmaceutical stability testing. A simple 72-h photodegradation study in purified water exhibited significant first-order kinetics with the kinetic constant k = 0.0303 h?1, and degradation halftime 22.8 h. The basic halftime could be reduced to 17.1 h by the addition of sea salt, and increase in temperature. Monohydroxy, dihydroxy and dehydrogenated derivatives of ethinylestradiol with intact steroidal structure were identified as major degradation products resulting from simple photodegradation. The addition of an oxidative agent significantly accelerated the degradation rate; combined with higher temperature, the degradation halftime was reduced to 1.1 h with the first-order kinetic constant k = 0.632 h?1. TOC analysis showed a notable decrease of organic mass (18% in 3 days) during oxidation experiments, and confirmed the degradation of steroidal structure.

A laboratory incubation study was conducted to investigate the effect of agricultural antibiotics (sulfamethazine, tylosin, and chlortetracycline) on the persistence and transformation of 17β-estradiol in Sequatchie loam. We measured concentrations of 17β-estradiol and its primary metabolite (estrone) in soils spiked with antibiotics and 17β-estradiol. Dehydrogenase activity (DHA) was also measured as an indicator of the total microbial activity of the soils. The presence of antibiotics significantly decreased transformation of 17β-estradiol to estrone. There was a positive correlation between the DHA and the concentrations of estrone in soil spiked with 17β-estradiol only, implying that the reaction is mainly catalyzed by dehydrogenases. However, the positive correlation was weakened in soil spiked with 17β-estradiol and antibiotics together. We recommend that any study evaluating the fate and transport of estrogenic hormones in soil should include the effect of agricultural antibiotics because antibiotics and estrogenic hormones are commonly excreted together in environmental samples.  相似文献

The effect of ship emissions in the urban environment of Göteborg has been studied by multivariate analysis. The simultaneous measurements of relevant gases and sub-micron particles make identification of ship plumes possible. Increased concentrations of these species due to ship emissions are quantified for ships entering the inner part of the harbour. Annual depositions of SO2 and NO2 are estimated to be 220 and 115 kg km−2 yr−1, respectively. Exposure of transient particles (less than 0.1 μm in diameter) to this part of the harbour increased by a factor of 3 in number concentration when a ship plume was recorded. Ni, Pb, V and Zn are shown to have positive correlation with NO emissions from ships.  相似文献

Concentration of heavy metals and natural gross radioactivity were measured in the surface water and sediment of Hazar Lake (Elazi?, Turkey). Eight sampling sites were pre-defined in different locations of the lake. A preliminary study on heavy metals (Zn, Fe, Mn, Ni, Cu, Cr, Co and Pb), major elements (Na, K, Ca, Mg) concentrations and natural radioactivity related to 226Ra, gross-alpha and gross-beta radiations in the surface water and deep sediments were determined. The obtained results showed that, in general, the heavy metals (Zn, Fe, Mn, Ni, Cu and Pb) and major elements (Na, K, Ca, Mg) concentrations in water did not exceed WHO (World Health Organization, 1999), EC (Europe Community, 1998), EPA (Environment Protection Agency, 2002) and TSE-266 (Turkish Standard, 1997) guidelines. Generally, heavy metals and major elements concentration of the sediments were found decrease in sequence of Fe>Mg>Ca>Mn>Zn>Ni>Cr>Cu>Co>Pb. The results of this study indicated that a general absence of serious pollution in the Hazar Lake. The results obtained from the radioactivity determination indicate that the surface water radioactivity concentration of 226Ra, gross-alpha and gross-beta were ranging from 0.52+/-0.02 to 2.02+/-0.06 Bq/l and from 0.65+/-0.03 to 2.52+/-0.07 Bq/l and from 0.01+/-0.01 to 0.14+/-0.01 Bq/l, respectively. Deep sediment radioactivity concentrations of 226Ra is ranging from 0.07+/-0.03 to 0.32+/-0.07 Bq/g.  相似文献
