The mineralization of 14C‐ETU was measured by the evolution of 14CO2 and described with a mathematical model consisting of two terms — one term describing the immediate mineralization of 14C‐ETU and another term describing the first order degradation of humus and/or biomass, where 14C had been built in. The influence of pesticide concentration, depth of soil, and incubation temperature showed combined interaction effects on the amount of 14CO2 formed during the process and on the degradation rate of the pesticide. With the addition of soil extract, a combined effect between concentration and addition of organic extract was seen for the degradation rate, while a three‐way interaction between depth, concentration and organic extract was seen for the formation of 14CO2. Degradation of 14C‐ETU can thus not be described only through investigations of one single of the mentioned parameters.  相似文献

Measurements in situ were made of oxygen exchange and 14C fixation in calcareous sediments off the coast of Florida, USA, and at various sites in the Caribbean Sea at depths to 60 m. Sediment samples were analyzed for total organic carbon, nitrogen and photosynthetic pigments. In some cases, measurements were made of the pH of sediment interstitial water and its concentration of total CO2. Photosynthetic properties of the common red algaGracilaria conferta, collected from the eastern Mediterranean Sea were investigated in 1989, in order to begin evaluating its adaptative strategies with regard to the inorganic carbon composition of seawater, and to test whether the alleged C4 photosynthesis of anotherGracilaria species is common within the genus. Net photosynthetic rates ofG. conferta were, under ambient conditions of inorganic carbon (ca. 10µM, CO2 and 2.2 mM HCO 3 - ), not sensitive to O2 over the range 10 to 300µM, and the CO2 compensation point was low (ca. 0.005µM). Ribulose-1,5-bisphosphate carboxylase/oxygenase was the major carboxylating enzyme, with a crude extract activity of 175µmol CO2 g–1 fresh wt h–1 while phosphoenolpyruvate carboxylase and phosphoenolpyruvate carboxykinase were present at 70 and 20%, respectively, of that activity. No activities of the decarboxylases NAD-and NADP-malic enzyme could be detected. The14C pulse-chase incorporation pattern showed thatG. conferta fixes inorganic carbon via the photosynthetic carbon reduction cycle only, with no evidence for photosynthetic C4 acid metabolism. Photosynthesis at the natural seawater pH of 8.2 was, at 25°C and saturating light, saturated at the ambient inorganic carbon concentration of 2.5 mM. It is proposed that, under ambient inorganic carbon conditions, a CO2 concentrating system other than C4 metabolism provides an internal CO2 concentration sufficient to suppress the O2 effect on ribulose-1,5-bisphosphate carboxylase/oxygenase and, thus, on photorespiration, in a medium where the external free CO2 concentration is lower than theK m(CO2) of the carboxylating enzyme. Since inorganic carbon, under natural saturating light conditions, seems not to be a limiting factor for photosynthesis ofG. conferta, it likely follows that other nutrients limit the growth of this alga in nature.  相似文献

The purpose of this work is to study the co-cultivation of Chlorella sp. and wastewater wild algae under different cultivation conditions (i.e. CO2, light intensity, cultivation time, and inoculation ratio) for enhanced algal biomass and lipid productivity in wastewater medium using Response Surface Methodology (RSM). The results show that mixed cultures of Chlorella sp. and wastewater wild algae increase biomass and lipid yield. Additionally, findings indicate that CO2, light intensity and cultivation time significantly affect algal productivity. Furthermore, CO2 concentration and light intensity, and CO2 concentration and algal composition, have an interactive effect on biomass productivity. Under different cultivation conditions, the response of algal biomass, cell count, and lipid productivity ranges from 2.5 to 10.2 mg/mL, 1.1 × 106 to 8.2 × 108 cells/mL, and 1.1 × 1012 to 6.8 × 1012 total fluorescent units/mL, respectively. The optimum conditions for simultaneous biomass and lipid accumulation are 3.6% of CO2 (v/v), 160 µmol/m2/s of light intensity, 1.6/2.4 of inoculation ratio (wastewater-algae/Chlorella), and 8.3 days of cultivation time. The optimal productivity is 9.8 (g/L) for dry biomass, 8.6 E + 08 (cells/mL) for cell count, and 6.8 E + 12 (Total FL units per mL) for lipid yield, achieving up to four times, eight times, and seven times higher productivity compared to nonoptimized conditions. Provided is a supportive methodology to improve mixed algal culture for bioenergy feedstock generation and to optimize cultivation conditions in complex wastewater environments. This work is an important step forward in the development of sustainable large-scale algae cultivation for cost-efficient generation of biofuel.

The gene for the catalytic domain of thermostable endo-β-1,3-glucanase (laminarinase) LamA was cloned from Thermotoga maritima MSB8 and heterologously expressed in a bioengineered Synechococcus sp. PCC 7002. The mutant strain was cultured in a photobioreactor to assess biomass yield, recombinant laminarinase activity, and CO2 uptake. The maximum enzyme activity was observed at a pH of 8.0 and a temperature of 70°C. At a CO2 concentration of 5%, we obtained a maximum specific growth rate of 0.083 h–1, a biomass productivity of 0.42 g?L–1?d–1, a biomass concentration of 3.697 g?L–1, and a specific enzyme activity of the mutant strain of 4.325 U?mg–1 dry mass. All parameters decreased as CO2 concentration increased from 5% to 10% and further to 15% CO2, except enzyme activity, which increased from 5% to 10% CO2. However, the mutant culture still grew at 15% CO2 concentration, as reflected by the biomass productivity (0.26 g?L–1?d–1), biomass concentration (2.416 g?L–1), and specific enzyme activity (3.247 U?mg–1 dry mass).

Studies were carried out to determine optimum conditions for the investigation of symbiotic zooxanthellae in vitro and to gain insight into factors influencing release of photosynthate by the symbionts. Zooxanthellae isolated from the reef coral Agaricia agaricites and incubated with an homogenate of host tissue release twice as much photosynthate as controls in seawater. The animal homogenate retained its stimulatory activity for 3 h at room temperature (ca. 26°C). Release of photosynthate was markedly influenced by time after isolation of algae from the host, variation in homogenate concentration, and prolonged exposure to homogenate. Release was not influenced by cell concentration, light intensity, or glycerol in the incubation medium. If zooxanthellae are labelled in vitro with glucose 14C, the principle product released is alanine 14C. The mechanism of action of homogenate on zooxanthellae in vitro is discussed in terms of its effect on algal cell membrane permeability. A preliminary fractionation of host homogenate is described.  相似文献

 The physico-chemical microenvironment of larger benthic foraminifera was studied with microsensors for O2, CO2, pH, Ca2+ and scalar irradiance. Under saturating light conditions, the photosynthetic activity of the endosymbiotic algae increased the O2 up to 183% air saturation and a pH of up to 8.6 was measured at the foraminiferal shell surface. The photosynthetic CO2 fixation decreased the CO2 at the shell down to 4.7 μM. In the dark, the respiration of host and symbionts decreased the O2 level to 91% air saturation and the CO2 concentration reached up to 12 μM. pH was lowered relative to the ambient seawater pH of 8.2. The endosymbionts responded immediately to changing light conditions, resulting in dynamic changes of O2, CO2 and pH at the foraminiferal shell surface during experimentally imposed light–dark cycles. The dynamic concentration changes demonstrated for the first time a fast exchange of metabolic gases through the perforate, hyaline shell of Amphistegina lobifera. A diffusive boundary layer (DBL) limited the solute exchange between the foraminifera and the surrounding water. The DBL reached a thickness of 400–700 μm in stagnant water and was reduced to 100–300 μm under flow conditions. Gross photosynthesis rates were significantly higher under flow conditions (4.7 nmol O2 cm−3 s−1) than in stagnant water (1.6 nmol O2 cm −3 s−1), whereas net photosynthesis rates were unaffected by flow conditions. The Ca2+ microprofiles demonstrated a spatial variation in sites of calcium uptake over the foraminiferal shells. Ca2+ gradients at the shell surface showed total Ca2+ uptake rates of 0.6 to 4.2 nmol cm−2 h−1 in A. lobifera and 1.7 to 3.6 nmol cm−2 h−1 in Marginopora vertebralis. The scattering and reflection of the foraminiferal calcite shell increased the scalar irradiance at the surface up to 205% of the incident irradiance. Transmittance measurements across the calcite shell suggest that the symbionts are shielded from higher light levels, receiving approximately 30% of the incident light for photosynthesis. Decomposition of urea in seawater was studied in Mikawa Bay, a shallow eutrophic bay on the southern coast of central Japan. The urea concentration in seawater ranged from 1.3 to 5.9 μg-at. N/1 and comprised 12 to 40% of the dissolved organic nitrogen. Using 14C labelled urea, the rate of CO2 liberation from urea and the incorporation rate of urea carbon into the particulate organic matter were determined. For the surface samples, high rates of CO2 liberation from urea as well as the incorporation of urea carbon into the particulate organic matter were observed in the light, while much lower rates were obtained in the dark. Incubation experiments with exposure to different light intensities revealed that the rate of CO2 liberation from urea and the incorporation of urea carbon into particulate organic matter changed with light intensity, showing a pattern similar to that of photosynthesis. The highest liberation and incorporation rates were observed at 12,000 lux. Incubation in light and in dark produced marked decreases and increases, respectively, in urea and ammonia, while no appreciable changes were observed for nitrate and nitrite. It is suggested that urea decomposition associated with photosynthetic activity of phytoplankton is one of the major processes of urea decomposition, and that it plays a significant role in the nitrogen supply for phytoplankton in coastal waters.  相似文献

Calcification, photosynthesis and respiration of the scleractinian coral Astrangia danae were calculated from the changes in total alkalinity, pH, calculated total CO2, and oxygen concentration produced by colonies incubated in glass jars. A correction for changes in ammonia, nitrate and nitrite was taken into account and the method evaluated. The fluxes of oxygen and CO2 were highly correlated (r=0.99). The statistical error of alkalinity determinations was less than 10% of the changes observed in the slowest calcifying samples. Metabolism of polyparium alone was estimated by difference after removal of tissue and reincubation of bare corallum. Zooxanthellae concentration in the polyps was obtained from cell counts made on homogenates of polyp tissue. The calculated photosynthetic rate of the zooxanthellae in vivo was 25 mol O2 (108 cell)-1 h-1 at a light intensity of 120 Ein m-2 s-1. In corals having 0.5x109 zooxanthellae/dm2 of colony area up to 8% of the total photosynthesis was attributed to the corallum microcosm. Polyp respiration, photosynthesis, and CaCO3 uptake rates were all much higher than rates previously reported from A. danae, apparently because in these experiments the organisms were better fed. This increased photosynthesis in turn enhanced calcification still further. The symbiosis therefore appears to provide a growth advantage even to fed corals, under the conditions of these experiments.  相似文献

Measuring the amount of fossil fuel carbon stored in the vegetation is now crucial to understand the mechanisms ruling climate changes. In this respect, highly polluted areas such as major towns represent “natural” laboratories because fossil fuel CO2 (14C-free) is isotopically distinct from mean atmospheric CO2 (14C-labeled). Here, a14C study of urban grasses near a major highway in Paris, France, shows that plants store up to 13% of fossil fuel carbon. The 14C composition of urban grasses is thus a novel parameter to assess the fossil fuel CO2 pollution.  相似文献

Grazing effects on nitrogen fixation in coral reef algal turfs   总被引:2,自引:0,他引:2  
This study addressed whether grazing by the sea urchin Diadema antillarum influenced rates of nitrogen fixation by algal turf communities on Caribbean coral reefs. Because the turfs were nitrogen-limited, we also assessed whether newly-fixed nitrogen was important for supporting net primary productivity by the turfs. We measured acetylene reduction in turfs grown in treatments excluding or including D. antillarum in the presence of other herbivores at 3 m water depth on Tague Bay forereef, St. Croix, U.S. Virgin Islands. These were the first measurements of acetylene reduction on coral reefs under quasi-natural conditions of high water-flow and photosynthetic oxygen generation. Rates of acetylene reduction under these conditions were as high as any measured previously in coral reef communities (mean 7.6 nmol C2H4 cm−2 h−1). Algal turfs grazed by D. antillarum and other herbivores had chlorophyll-specific acetylene reduction rates up to three times higher than when D. antillarum was excluded. High rates of nitrogen fixation by the turfs were sufficient to meet <2% of the nitrogen required to support net chlorophyll-specific primary productivity over 24 h. Grazer-mediated increases in nitrogen fixation do not appear responsible for a parallel enhancement of net primary productivity. Tree-ring chronologies provide long-term records of growth in natural environmental conditions and may be used to evaluate the impacts of climatic change and [CO2] increase on forest productivity. This study focuses on 21 Pinus halepensis forest stands in calcareous Provence (in the south-east France). Changes in productivity are simulated using the global biogeochemistry model BIOME3, that we have adapted to run with chronological data. Tree-ring data (width and density) were used to estimate, for each stand, an observed series of changes in productivity. Simulated and observed productivity changes are then compared to validate the chronological biogeochemistry model BIOME3C. Variations in productivity were well reconstructed at 15 sites. After this validation, BIOME3C was used to simulate forest productivity changes for a 2×CO2 scenario. The 2×CO2 climate used as input was obtained using results from Météo-France’s ARPEGE atmospheric general circulation model (AGCM), downscaled to local meteorological stations. Productivity increases moderately for all stands (from 17 to 24%) when climatic changes alone were taken into account. The main factor responsible for this increase is a reduction in summer drought severity. Productivity increases highly for all stands (from 72 to 86%) when the physiological fertilising effect of the [CO2] increase is considered separately. When both climatic changes and the [CO2] increase were taken into account, productivity increases highly, from 107% (for Moustier) to 141% (for La Ciotat). The direct fertilising effect of [CO2] increase has a greater influence on the forest stands productivity than the indirect climatic changes effect. These results also exhibit the importance of the synergy between the effects of climate change and [CO2] increase, as the increase in productivity resulting from the combined effects are more than the sum of the individual CO2 and climate effects. Although the detected effects of global change during the 20th century were slight, acceleration of these changes is likely to lead to great changes in the future productivity of P. halepensis forests.  相似文献

Four colonies of Acropora formosa were incubate with Na2 14CO3 for separate 2 h periods within a 24 h period, and then returned to the reef from which they were collected. Terminal branches were collected at intervals over the following 5 d and analysed for radioactivity associated with the skeleton and certain organic pools. Colonies incubated at night showed little or no loss of fixed radioactivity during the 5 d on the reef. However, 50–60% of photosynthetically-fixed 14C was lost from the terminal branches during the first 40 h on the reef. This loss of radioactivity probably resulted from release of mucus and dissolved organic carbon from the coral tissues. Most of the loss of photosynthetically-fixed 14C was due to decrease in the radioactivity of lipids (80% of the total 14C loss) and methanol-water soluble compounds. Determination of any sequencing in metabolic compartments was made difficult by the rapidity with which 14C dissappeared from most of the metabolic pools measured. 14C was incorporated into the skeleton throughout the 5 d on the reef, although the rate of incorporation was very low in colonies which had been incubated with Na2 14CO3 at night.  相似文献

When symbiotic dinoflagellate algae (Symbiodinium sp., isolated from the coral Plesiastrea versipora) were incubated with NaH14CO3 in the light in seawater, they released 22.69±9.16 nmol carbon/106 algae. Release of photosynthetically fixed carbon was stimulated more than six-fold for algae incubated in host-tissue homogenate (148.54±97.03 nmol C/106 algae) and more than four-fold (102.00±49.16 nmol C/106 algae) for algae incubated in a low molecular weight fraction (≤1 000 M r ) prepared from host homogenate. Soluble released 14C-labelled products, as determined by chromatography and autoradiography, were the same when algae were incubated in either host homogenate or the low molecular weight fraction. After 4 h incubation in the light (300 mol photons m−2 s−1),␣intracellular␣glycerol increased in algae incubated with the low molecular weight fraction (an increase of 0.39 to␣0.67 nmol glycerol/106 algae) compared with little or no increase in algae incubated in seawater (0 to 0.12 nmol glycerol/106 algae). Partial inhibition of triglyceride synthesis (up to 51%) was also observed when algae were incubated in the low molecular weight fraction. All these effects are the same as those observed when algae were incubated in host homogenate. In an attempt to evaluate CO2 changes as an index of net ecosystem metabolism, CO2 and dissolved organic carbon (DOC) by infrared (IR) analysis and O2 by the Winkler method were followed over 12 diel cycles in a salt marsh, a simulated estuarine ecosystem, and the mixed layer of the northwestern Caribbean Sea. Each ecosystem exhibited replicable diel cycles, net production during the photoperiod, and a significant inverse correlation between CO2 and O2 changes. Daily rates of system production and respiration calculated from CO2, however, exceeded those from O2 by 1.5 to 3.5 fold in nearshore waters and by 2 to 6 fold in the open ocean. Net total system apparent production based on O2 and CO2, respectively, were 2 345 and 3 604 mg C m-3d-1 for the salt marsh, 348 and 625 mg C m-3d-1 for the estuarine ecosystem, and 53 and 306 mg C m-3d-1 for the oceanic ecosystem, both parameters exceeding 14C data in the literature for similar environments by one to two orders of magnitude. The IR CO2 productivity estimates are compatible with the diel cycles in DOC. In the marsh and Caribbean Sea, maxinium DOC concentrations were usually observed in the evening following a gradual accumulation during the photoperiod, while minimal values occurred in the early morning. In all ecosystems the average net release of DOC lagged CO2 uptake and O2 production and represented 22.7 and 43.3% of the carbon fixed as estimated from CO2 uptake and O2 production, respectively. If the consistently higher CO2 measurements are not a systematic error nor due to atmospheric diffusion, then diel variation in O2 and CO2 may not always be quantitatively coupled due in part to habitat-dependent factors such as the nonphotosynthetic incorporation of CO2 and chemosynthetic removal of O2.  相似文献

With global climate change, ocean warming and acidification occur concomitantly. In this study, we tested the hypothesis that increasing CO2 levels affect the acid–base balance and reduce the activity capacity of the Arctic spider crab Hyas araneus, especially at the limits of thermal tolerance. Crabs were acclimated to projected oceanic CO2 levels for 12 days (today: 380, towards the year 2100: 750 and 1,120 and beyond: 3,000 μatm) and at two temperatures (1 and 4 °C). Effects of these treatments on the righting response (RR) were determined (1) at acclimation temperatures followed by (2) righting when exposed to an additional acute (15 min) heat stress at 12 °C. Prior to (resting) and after the consecutive stresses of combined righting activity and heat exposure, acid–base status and lactate contents were measured in the haemolymph. Under resting conditions, CO2 caused a decrease in haemolymph pH and an increase in oxygen partial pressure. Despite some buffering via an accumulation of bicarbonate, the extracellular acidosis remained uncompensated at 1 °C, a trend exacerbated when animals were acclimated to 4 °C. The additional combined exposure to activity and heat had only a slight effect on blood gas and acid–base status. Righting activity in all crabs incubated at 1 and 4 °C was unaffected by elevated CO2 levels or acute heat stress but was significantly reduced when both stressors acted synergistically. This impact was much stronger in the group acclimated at 1 °C where some individuals acclimated to high CO2 levels stopped responding. Lactate only accumulated in the haemolymph after combined righting and heat stress. In the group acclimated to 1 °C, lactate content was highest under normocapnia and lowest at the highest CO2 level in line with the finding that RR was largely reduced. In crabs acclimated to 4 °C, the RR was less affected by CO2 such that activity caused lactate to increase with rising CO2 levels. In line with the concept of oxygen and capacity limited thermal tolerance, all animals exposed to temperature extremes displayed a reduction in scope for performance, a trend exacerbated by increasing CO2 levels. Additionally, the differences seen between cold- and warm-acclimated H. araneus after heat stress indicate that a small shift to higher acclimation temperatures also alleviates the response to temperature extremes, indicating a shift in the thermal tolerance window which reduces susceptibility to additional CO2 exposure.  相似文献

Pieces of branch from the staghorn coral Acropora acuminata were incubated with 45CaCl2 and NaH14CO3 under identical conditions in the light or in the dark. Specimens were then processed in different ways. All specimens were placed in N KOH to digest tissues. Some were placed in KOH immediately after incubation; others were placed in KOH after 2 h washing, or after 2 h extraction with methanol-chloroformwater. Specimens were washed in running fresh water or running seawater; some were killed in liquid N2 before washing. Radioactivity associated with skeleton and tissues was determined. The method of processing profoundly affected the results. In dark incubations, there was up to a four-fold difference in apparent skeletal incorporation of 45Ca++ between average values obtained for the different treatments. For 14C incorporation, there was a difference of up to 2.5 times. In light incubations, skeletal incorporation of both radioisotopes showed a two-fold difference between high and low average values obtained for the different treatments.  相似文献

During a cruise to the eastern Canadian Arctic (Northern Baffin Bay) in the summer of 1980, we took advantage of the 24-h photoperiod to conduct a 32-h time course experiment of 14C accumulation under natural solar radiation. The degree of non-linearity in the time course was judged against a time-dependent curve of radioactivity constructed by cumulatively adding the amount of 14C taken up in sequential short (2 h) incubations of plankton held in a replicate bottle but left unlabelled until removed for assay. Departure from linearity was due first to decreasing rates of 14C incorporation into polysaccharides and then into lipids. There was a close correspondence between 14C incorporation into proteins in the 32-h incubation and in the sequence of short incubations. These observations are consistent with patterns in utilization of photosynthetic end-products established from laboratory studies of unicellular algal cultures. Based on parallel or independent control experiments, it was judged that complicating factors such as diel light changes, nitrogenous nutrient exhaustion, bottle size effects or inhibitory conterminants in NaH14CO3 stock solutions would not seriously affect our interpretation that non-linearity resulted from catabolic loss of radiocarbon.  相似文献

Thallus segments of Fucus serratus L. and F. vesiculosus L. (Phaeophyceae, Fucales) were transferred into seawater media with a salinity range from 32.65 to 2.25 and maintained for at least 2 weeks. Several parameters of chemical composition as well as rates of photosynthetic and respiratory oxygen exchange, 14C-assimilate patterns, and release of 14C-assimilates into the culture medium have been investigated. Compared to controls, in both species dry weight, ash, chloride, and mannitol contents distinctly decline proportionally to reduction of salinity in the incubation media, whereas content of total N (in terms of protein content) remarkably increase. Respiratory O2-consumption is markedly increased at lower salinities, whereas rate of photosynthetic O2-evolution shows some depression. Relatively little effects of salinity changes are observed in distribution of photosynthetically assimilated 14C among the major groups of photosynthates. Release of 14C-assimilates into the incubation medium never exceeds 2% of total 14C-uptake, but is stimulated in media of reduced salt content. The results are discussed with emphasis on phenomena of long-term adaptation and osmoregulation in the marine fucoid species.  相似文献

Light-dependent 14CO2 fixation by the algae of Diplosoma virens (Hartmeyer) ranged between about 3 and 27 moles mg-1 chlorophyll h-1. The principal first products of 14C fixation were 3-phosphoglyceric acid and phosphorylated sugars, indicating that ribulose bisphosphate carboxylase was the primary carboxylation enzyme. The activity of this enzyme in crude extracts of the algae was 4 to 6 moles CO2 mg-1 chlorophyll h-1. The principal end product of 14C fixation by these algae in the ascidian host was a water-soluble oligosaccharide which was an -1,4-glucan. A maximum of 7% of the 14C fixed was found in insoluble materials of the algae or its host after 60 min 14CO2 fixation. Whether the -1,4-glucan is a product of algal or animal metabolism remains to be determined.  相似文献
