Introduction and large-scale production of synthetic halogenated organic chemicals over the last fifty years has resulted in a group of contaminants that tend to persist in the environment and resist both biotic and abiotic degradation. The low solubility of these types of contaminants, along with their toxicity and tendency to accumulate in food chains, make them particularly relevant targets for remediation activities. Among the mechanisms that result in dehalogenation of some classes of organic contaminants are stimulation of metabolic sequences through introduction of electron donor and acceptor combinations; addition of nutrients to meet the needs of dehalogenating microorganisms; possible use of engineered microorganisms; and use of enzyme systems capable of catalyzing reductive dehalogenation. The current state of research and development in the area of reductive dehalogenation is discussed along with possible technological application of relevant processes and mechanisms to remediation of soil and groundwater contaminated with chlorinated organics. In addition, an overview of research needs is suggested, which might be of interest for development of in-situ systems to reduce the mass of halogenated organic contaminants in soil and groundwater.  相似文献

In situ bioremediation (ISB) melds an understanding of microbiology, chemistry, hydrogeology, and engineering into a strategy for planned and controlled microbial degradation of specific contaminants. ISB creates subsurface environmental conditions, typically through reduction oxidation manipulation, which induce the degradation of contaminants via microbial catalyzed biochemical reactions. In turn, the microbes produce enzymes that are utilized to derive energy and that are instrumental in the degradation of target chemicals. To accomplish this chain of events, the type of microorganisms, contaminant, and the geological conditions at the site must be considered. Since in situ conditions are manipulated by engineered means, the most important consideration is the ability to transmit and mix liquids in the subsurface. The Interstate Technology Regulatory Council (ITRC)–ISB Team has recently completed a guidance document that describes a systematic approach to ISB in groundwater. ITRC is a state‐led coalition of more than 40 states working together with industry and stakeholders to achieve regulatory acceptance of environmental technologies. © 2003 Wiley Periodicals, Inc.  相似文献

In response to an oxygenated gasoline release at a gas station site in New Hampshire, a temporary treatment system consisting of a single bedrock extraction well, a product recovery pump, an air stripper, and carbon polishing units was installed. However, this system was ineffective at removing tertiary butyl alcohol from groundwater. The subsequent remedial system design featured multiple bedrock extraction wells and an ex situ treatment system that included an air stripper, a fluidized bed bioreactor, and carbon polishing units. Treated effluent was initially discharged to surface water. Periodic evaluation of the remediation system performance led to system modifications, which included installing an additional extraction well to draw contaminated groundwater away from an on‐site water supply well, adding an iron and manganese pretreatment system, and discharge of treated effluent to an on‐site drywell. Later, the air stripper and carbon units were eliminated, and an infiltration gallery was installed to receive treated, oxygenated effluent in order to promote flushing of the smear zone and in situ bioremediation in the source area. This article discusses the design, operation, performance, and modifications to the remediation system over time, and provides recommendations for similar sites. © 2007 Wiley Periodicals, Inc.  相似文献

In situ remediation of aniline from soils and groundwater using biological and physical treatments was conducted at the BASF Corporation facility in Geismar, Louisiana. To mitigate the migration of aniline, remediate contaminated soil and groundwater, and determine concentrations, 24 immobilized microbe bioreactors were fixed in the subsoil, and a horizontal recovery well and 7 monitoring wells were installed. Soil and monitoring wells were sampled quarterly to assess bioplug impact on the aniline concentrations. The recovery well was sampled monthly to estimate the pounds of aniline removed from groundwater. Soil pH, composition, and microbial counts were used to estimate the fate and transport. Aniline levels were lowered significantly after remediation and total cancer risk was below levels for industrial sites, as established by State of Louisiana Risk Evaluation/Corrective Action Program guidelines. © 2010 Wiley Periodicals, Inc.  相似文献

Proving the viability of in situ bioremediation technologies and gathering data for its full‐scale implementation typically involves collecting multiple rounds of data and often completing microcosm studies. Collecting these data is cumbersome, time‐consuming, costly, and typically difficult to scale. A new method of completing microcosm studies in situ using an amendable sampling device deployed and incubated in groundwater monitoring wells provides actionable data to expedite site cleanup. The device, referred to as a Bio‐Trap® sampler, is designed to collect actively colonizing microbes and dissolved organic compounds from groundwater for analysis using conventional analytical techniques and advanced diagnostic tools that can answer very specific design and viability questions relating to bioremediation. Key data that can be provided by in situ microcosm studies using Bio‐Trap® samplers include definitively demonstrating contaminant destruction by using compound‐specific isotope analysis and providing data on the mechanism of the degradation by identifying the responsible microbes. Three case studies are presented that demonstrate the combined flexibility of Bio‐Trap® samplers and advanced site diagnostics. The applications include demonstrating natural attenuation of dissolved chlorinated solvents, demonstrating natural attenuation of dissolved petroleum compounds, and using multiple Bio‐Trap® samplers to comparatively assess the viability of bioaugmentation at a chlorinated solvent release site. At each of these sites, the in situ microcosm studies quickly and cost‐effectively answered key design and viability questions, allowing for regulatory approval and successful full‐scale implementation. © 2010 Wiley Periodicals, Inc.  相似文献

This article describes a design approach that has been developed for bioremediation of chlorinated volatile organic compound–impacted groundwater that is based upon experience gained during the past 17 years. The projects described in the article generally involve large‐scale enhanced anaerobic dechlorination (EAD) and combined aerobic/anaerobic bioremediation techniques. Our design approach is based on three primary objectives: (1) selecting and distributing the proper additives (including bioaugmentation) within the targeted treatment zone; (2) maintaining a neutral pH (and adding alkalinity when needed); and (3) sustaining the desired conditions for a sufficient period of time for the bioremediation process to be fully completed. This design approach can be applied to both anaerobic and aerobic bioremediation systems. Site‐specific conditions of hydraulic permeability, groundwater velocity, contaminant type and concentrations, and regulatory constraints will dictate the best remedial approach and design parameters for in situ bioremediation at each site. The biggest challenges to implementing anaerobic bioremediation processes are generally the selection and delivery of a suitable electron donor and the proper distribution of the donor throughout the targeted treatment zone. For aerobic bioremediation processes, complete distribution of adequate concentrations of a suitable electron acceptor, typically oxygen or oxygen‐yielding compounds such as hydrogen peroxide, is critical. These design approaches were developed based on understanding the biological processes involved and the mechanics of groundwater flow. They have evolved based on actual applications and results from numerous sites. An EAD treatment system, based on our current design approach, typically uses alcohol as a substrate, employs groundwater recirculation to distribute additives, and has an operational period of two to four years. An aerobic in situ treatment system based on our current design approach typically uses pure oxygen or hydrogen peroxide as an electron acceptor, may involve enhancements to groundwater flow for better distribution, and generally has an operational period of one to four years. Bioavailability is one main factor that influences the extent of biodegradation of hydrocarbons. They are very poorly soluble in water and easily adsorbed to clay or humus fractions, so they pass very slowly to the aqueous phase where they are metabolised by microorganisms. Cyclodextrins are natural compounds that form soluble inclusion complexes with hydrophobic molecules and increase degradation rate of hydrocarbons in vitro. In the perspective of an in situ application, we previously checked that -cyclodextrin does not increase eluviation of hydrocarbons through the soil and consequently does not increase the risk of groundwater pollution. The results of an in situ application of -cyclodextrin for bioremediation of a hydrocarbon polluted site are presented. We stated that the combination of bioaugmentation and enhanced bioavailability due to -cyclodextrin was effective for a full degradation.  相似文献

A study was conducted to evaluate the efficacy of PHOSter® technology for treating groundwater contaminated with trichloroethene (TCE) at Edwards Air Force Base, California. The technology consists of injecting a gaseous mixture of air, methane, and nutrients into groundwater with the objective of stimulating the growth of methanotrophs, a naturally occurring microbial group that is capable of catalyzing the aerobic degradation of chlorinated solvents into nontoxic products. Injection operations were performed at one well for a period of three months. Six monitoring wells were utilized for groundwater and wellhead vapor monitoring and for groundwater and microbial sampling. In the five monitoring wells located within 44 feet of the injection well, the following results were observed: dissolved oxygen concentrations increased to a range between 6 and 8 milligrams per liter (μg/L); the biomass of target microbial groups increased by one to five orders of magnitude; and TCE concentrations decreased by an average of 92 percent, and to below the California primary maximum contaminant level (MCL; 5 micrograms per liter [µg/L]) in the well closest to the injection well. © 2008 Wiley Periodicals, Inc. *

    The term environmental biotechnology has a certain air of modernity when in fact it has a long history of use, if one considers the underlying principles and not the appellation. However, as part of its complex meaning, there is a dynamic new definition and purpose in this discipline with regard to bioremediation. The ability to probe the environment at the molecular level with exquisite methods, to create a new awareness of fundamental biological processes therein, has created an important new paradigm in remediation engineering design and management. Further, biological lines of evidence made extremely robust through the merger of biotechnology and environmental science are poised to be incorporated into the very fabric of site evaluation and disposition at the regulatory level. At the operational level, the field of environmental biotechnology is driven by the “omics,” the common suffix for disciplines like genomics, proteomics, and metabolomics. An introduction to these elements of the process is followed by a review of how they are being used right now in a commercial framework, with the understanding that the entire process is still in the formative stages of its vast potential. © 2005 Wiley Periodicals, Inc.  相似文献

    After achieving remediation goals during only thirty-two months of operation, the first full-scale in-situ bioremediation (ISB) system in the state of Missouri was shut down in 1990. In addition to ISB, the system included a combination of soil venting and air stripping to remediate subsurface gasoline contamination at a large manufacturing facility. More than 84,000 pounds of gasoline were degraded or removed from the fractured limestone bedrock aquifer and overburden materials. The successful application of ISB in this complex geologic environment and the fact that this was the first such system to complete remediation in Missouri make this system unique.  相似文献

    This article is a critical analysis of the treatment potential of bioremediation technology to degrade eight major environmental pollutants, polycyclic aromatic hydrocarbons, phenols, pentachlorophenols, creosote, polychlorinated biphenyls, trichloroethylene, chlorobenzoates, and chlorophenols. The discussion includes information on transformation mechanisms, identification of intermediate metabolites, elucidation of partial or complete pathways, effects of environmental parameters, as well as current and future industrial application. Results indicate that bioremediation used in conjunction with other physical and chemical treatment methodologies can effectively transform most prevalent nonchlorinated organic contaminants and some chlorinated contaminants, such as creosote and pentachlorophenol, into innocuous materials. Successful biodegradation of several other chlorinated organic compounds, notably polychlorinated biphenyls and trichloroethylene, is currently possible only under controlled laboratory conditions. Future successful field applications, however, appear promising.  相似文献

    The potential application of enhanced in situ bioremediation (EISB) for treatment of a plume containing high concentrations of 1,2‐dichloroethane (1,2‐DCA), as well as lower concentrations of other chlorinated ethanes, chlorinated methanes, and chlorinated ethenes was evaluated through the implementation of four field trials. The field trials confirmed that EISB is an effective technology for treating multiple contaminants, with estimated mass removal rates on the order of several kilograms per day and groundwater concentration reductions nearing 100 percent of the initial concentrations. The field trials also demonstrated that engineering controls could be effectively used to overcome potential inhibitions related to high concentrations of 1,2‐DCA. © 2008 Wiley Periodicals, Inc.  相似文献

    The use of lignin degrading fungi for decomposition of a wide variety of xenobiotics has become an area of intensive research. One distinct advantage of lignin degrading fungi over bacteria is that they do not require preconditioning to a particular pollutant prior to transformation. This degradative ability has been attributed to a nonspecific and nonstereoselective extracellular lignin-degrading enzymatic system (ligninase) which is induced by the fungi under nitrogen or carbon-limiting conditions (Reid, 1979). Ligninases (lignin-peroxidases) are responsible for the initial oxidative attack on lignin and other complex molecules via formation of a free radical thereby leading to depolymerization of complex molecular structures. Potential degradative ability of peroxidases may extend to include (1) sorbed contaminants, (2) high molecular-weight, hydrophobic contaminants, and (3) complex mixtures of chemicals typical of a contaminated site.  相似文献

    针对百菌清具有在土壤中药效稳定、不易分解、代谢周期长、长期大量施加导致土壤严重污染等特点和问题,简要介绍了百菌清的毒性作用机制。总结了降解土壤中百菌清的物理法、化学法和生物法的原理及优缺点。重点阐述了生物修复百菌清污染土壤的主要降解菌株及其效果、降解途径以及降解产物及其毒性,分析了土壤性质、微生物种类、温度、土壤含水率等因素对百菌清降解效果的影响。指出今后的研究重点应为降解中间产物的毒性分析及其进一步的降解与转化问题,而复合菌制剂或多酶复合体系可实现百菌清的彻底降解和无害化。  相似文献

    Conventional vapor intrusion characterization efforts can be challenging due to background indoor air constituents, preferential subsurface migration pathways, sampling access, and collection method limitations. While it has been recognized that indoor air concentrations are dynamic, until recently it was assumed by many practitioners that subsurface concentrations did not vary widely over time. Newly developed continuous monitoring platforms have been deployed to monitor subsurface concentrations of methane, carbon dioxide, oxygen, hydrogen sulfide, total volatile organic constituents, and atmospheric pressure. These systems have been integrated with telemetry, geographical information systems, and geostatistical algorithms for automatically generating two‐ and three‐dimensional contour images and time‐stamped renderings and playback loops of sensor attributes, and multivariate analyses through a cloud‐based project management platform. The objectives at several selected sites included continuous monitoring of vapor concentrations and related physical parameters to understand explosion risks over space and time and to then design a long‐term risk reduction strategy. High‐frequency data collection, processing, and automated visualization have resulted in greater understanding of natural processes, such as dynamic contaminant vapor intrusion risk conditions potentially influenced by localized barometric pumping. For instance, contemporaneous changes in methane, oxygen, and atmospheric pressure values suggest there is interplay and that vapor intrusion risk may not be constant. As a result, conventional single‐event and composite assessment technologies may not be capable of determining worst‐case risk scenarios in all cases, possibly leading to misrepresentation of receptor and explosion risks. While dynamic risk levels have been observed in several initial continuous monitoring applications, questions remain regarding whether these situations represent special cases and how best to determine when continuous monitoring should be required.     Quasi‐passive in situ remediation technologies, such as the use of permeable reactive barriers to treat contaminated groundwater or applications of granular activated carbon to treat polychlorinated biphenyl (PCB)‐contaminated, near‐surface sediments, are proven or promising technologies that may be limited in application due to the traditional construction techniques normally used for placement in the environment. High‐pressure waterjets have traditionally been used to excavate material during mining operations or to cut rock or other durable material. Waterjets have the potential to place amendments in the subsurface at depths greater than those that can be obtained using traditional construction techniques. Likewise, waterjets may have less negative impact on near‐surface utilities and/or sensitive ecological systems. Laboratory experiments were performed to characterize the placement of two solid amendments in a simulated saturated aquifer. A second set of experiments was performed to characterize the effectiveness of waterjets for placing a third amendment in simulated intertidal sediments. The laboratory work focused on characterizing the nature of the waterjet penetration of the aquifer matrix and the saturated sediments, as well as the corresponding waterjet parameters of pressure, nozzle size, and injection time.     In 1995 the University of Tennessee's Waste Management Research and Education Institute and Canon Inc. began an analysis of the extent to which remediation firms and research centers have implemented bioremediation strategies, particularly for the cleanup of trichloroethylene (TCE) in soil and groundwater. The research involved the mailing of surveys to a select, representative group of environmental professionals involved in TCE cleanup activities. The survey was divided into two parts. Part I gathered cost information for TCE cleanup, using both bioremediation and “conventional” cleanup technologies. Part II asked the survey recipients to relate their opinions on the use of nonindigenous microorganisms for bioremediation, especially their assessment of the effectiveness, reliability, safety, and predictability of this approach. The results of this survey are discussed in this article.  相似文献

    Azo dyes, which are the most commonly used dyes in the textile industry, are aromatic compounds with N═N– groups. The treatment of these pollutants has been receiving considerable attention due to their persistence and release of dyes into the environment. The existing treatment approaches are not only expensive but also result in the production of concentrated sludge, which creates a secondary disposal issue. Under particular ecological conditions, a variety of microbes, including bacteria, fungi, algae, and yeasts, can not only decolorize numerous dyes but can also degrade them. In this respect, microbial degradation is a successful, cost-effective, biologically friendly, and ecologically sustainable treatment strategy. This review paper discusses research articles identified in the ScienceDirect bibliographic database for the last 10 years (from January 1, 2010 to June 29, 2022). Only the most appropriate research articles were included in the review process which was identified by searches with keyword phrases Azo-dye degradation with bacteria, fungi, algae, yeast, and microbial consortia. The review paper also emphasizes the constraints that persist and the future scope for the degradation of dyes via genetic engineering.  相似文献
